The influence of coaching on entrepreneurial goal-setting behaviour

The purpose of this study is to collect data on the current practices of coaching as applied in an incubated environment to report on the influence of coaching on entrepreneurial goal-setting behaviour. Previous research has indicated that much criticism has been lobbied at government programmes aimed at providing entrepreneurship development and support, some recognition has been given to some of these government initiatives as well as the progress achieved by some. It is further stated in the Global Entrepreneurship Report (2012) that a number of national experts commend the existence of business support agencies (such as the Small Enterprise Development Agency). A number of significant findings materialized from the current study where it was found that coaching influences entrepreneurial behaviour and that a need exists for more in-depth coaching sessions specifically focused on entrepreneurship in the South African context. Further to this it was found that a need exists for entrepreneurs to be coached by other entrepreneurs and that the role of a coach should be clarified to set realistic expectations from the onset of the intervention. These findings are in agreement with some of the key recommendations from South African national experts as cited in the GEMS 2012 report. The current research adds to the body of research on coaching in general. It also contributes specifically entrepreneurial development programmes in the South African context. The outcome from this research has implications for business coaches, entrepreneurs and entrepreneurial development/support agencies as well as those who provide training and incubation programmes for entrepreneurs. / Industrial & Organisational Psychology / M. A transformação genética possibilita a introdução de genes de interesse nos genomas, podendo assim ser empregada na tentativa de melhorar características agronômicas e florestais. No entanto, para a obtenção de plantas transgênicas são necessários protocolos eficientes de regeneração de plantas in vitro. Em teca, dados sobre cultura de tecidos são escassos, havendo a necessidade de determinar condições ótimas para a mesma. Com isso, o trabalho teve por objetivos estudar a organogênese in vitro de teca visando desenvolver um método de regeneração eficiente, avaliar condições para o processo de transformação e testar a susceptibilidade da lagarta Hyblaea puera a toxinas produzidas pelo Bacillus thuringiensis. Foram avaliadas a influência de TDZ e BAP na indução da competência organogenética em hipocótilos, nó cotiledonar e cotilédones de teca. Os biorreguladores AIB, BAP, NAA e GA3 foram utilizados na regeneração de segmentos de hipocótilo, nó cotiledonar, raiz, epicótilo e cotilédone. Antibióticos supressores de Agrobacterium tumefaciens e a higromicina (seleção de células transgênicas), foram também avaliados. Finalmente, testes com o inseticida biológico DipelTM e esporos de B. thuringiensis crescidos em laboratório foram realizados com as lagartas de Hyblaea puera. Na aquisição de competência organogenética o TDZ proporcionou um aumento de 46% na regeneração e o BAP 26% quando comparados ao controle. Para a organogênese in vitro foi avaliado um máximo de 70% de regeneração em nó cotiledonares em meio MS adicionado de 1 mg.L-1 de BAP + 0,5 mg.L-1 de GA3. Entretanto, em outras concentrações dos meios de regeneração hipocótilos, raiz, cotilédones e epicótilos tiveram máximas frequências de regeneração em torno de 60%, 60%, 30% e 10%, respectivamente. Os antibióticos supressores da Agrobacterium tumefaciens tiveram efeitos diferentes para cada explante. Timentin e cefotaxima na concentração de 300 mg.L-1 aumentaram o número de brotos em hipocótilos e nó cotiledonar em 1,6 e 2,0 vezes, respectivamente. Em cotilédone esses antibióticos tiveram efeitos negativos no número de brotos. Carbenicilina em todas as doses influenciou negativamente a regeneração em todos os explantes utilizados. A higromicina a 2,5 mg.L-1 inibe em 100% a regeneração de cotilédones, nó cotiledonar e hipocótilo. Os ínstares mais novos de H. puera são susceptíveis tanto ao produto comercial DipelTM quanto aos esporos crescidos em laboratório, apresentando 100% de mortalidade a concentrações de 2x105 UFC após 24 horas de ingestão. Mostrando assim seu potencial na transgenia visando à expressão de genes de Bt para a resistência a insetos. Os resultados apresentados nesse trabalho contribuem para o ganho de informação sobre os fatores que influenciam a organogênese desta espécie, bem como, definir parâmetros que possam ser utilizados em experimentos futuros visando à transformação genética da espécie. / Genetic transformation allows the introduction of genes in host genomes and can therefore be used to improve forestry and agronomic traits like insect resistence. However, efficient plant regeneration protocols are necessary to obtain transgenic plants. Thus far, information about in vitro teak (Tectona grandis L. f) organogenesis is scarce. Therefore, the aims of this study were: develop an efficient protocol for in vitro organogenesis of teak, assess conditions for its genetic transformation and test the susceptibility of the caterpillar Hyblaea puera to toxins produced by Bacillus thuringiensis. We evaluated the influence of TDZ and BAP on the induction of organogenic competence in hypocotyl, cotyledonary nodes and cotyledons. Growth regulators IBA, BAP, NAA and GA3 were used in the regeneration of the hypocotyl, cotyledonary node, root, epicotyl and cotyledon. Antibiotics for suppression of Agrobacterium tumefacien (timentin, cefotaxime and carbenicillin) and for selection of transgenic cells (hygromycin) were also evaluated. Finally, tests with the biological insecticide DipelTM and spores of B. thuringiensis grown in laboratory were performed with the caterpillar of Hyblaea puera. TDZ increases 46% the regeneration frequency and BAP 26% when compared to controls. Cotyledonary nodes showed the best regeneration frequency (70%) growing on MS medium added of 1 mg.L-1 BAP + 0.5 mg.L-1 GA3. Hypocotyls, roots, cotyledons and epicotyls presented variable frequency of regeneration (60%, 60%, 30%, and 10% respectively) growing on distinct concentrations of grown regulators. We tested three antibiotics (timentin, cefotaxime, and carbenicillin) to suppress A. tumefaciens in vitro growth and they presented different effects on the organogenesis of each explants used in this study. Timentin and cefotaxime at concentration of 300 mg.L-1 increased the number of buds on hypocotyls and cotyledonary nodes. Conversely, these antibiotics had negative effects on the number of shoots of cotyledonary explants. Carbenicillin at all doses presented a negative influence on regeneration of all explants. Hygromycin at concentration of 2.5 mg.L-1 inhibits 100% of regeneration of cotyledons, cotyledonary nodes, and hypocotyls. The young instars of H. puera are susceptible to likely both commercial product DipelTM and spores grown in the laboratory, presented 100% mortality at concentrations of 2x105 CFU after 24 hours of ingestion. These findings suggest its potential to be used in teak transgenic approaches for insect resistance. Our results contribute to information about factors that influence the organogenesis of this specie, as well as define parameters that can be used in future experiments aimed at the genetic transformation of the specie.

Translocation and female subjectivities in four contemporary narratives : Kingston’s The woman warrior, Magona’s To my children’s children and Forced to grow and Hoffman’s Lost in translation

Thesis (MA (English Studies))--University of Stellenbosch, 2009. / ENGLISH ABSTRACT: Drawing on theories of gender and subjectivity, this thesis explores the way in which constructions of modernity as well as tradition are mapped onto geographical localities and thus expressed through gender acts. The female protagonists in Maxine Hong Kingston’s The Woman Warrior, Sindiwe Magona’s To My Children’s Children and Forced to Grow, as well as Eva Hoffman’s Lost in Translation undergo either transnational translocation or imagined translocation where they straddle multiple cultural contexts concurrently. The role of globalism and modernity amplifies the female’s ambiguous position and therefore challenges her gender identity as she takes on additional gender characteristics. This challenge, a result of translocation, causes both the individual and collective nature of the subject to be emphasised and placed in multiple cultures concurrently. The female’s subjectivity is under much tension as the cultures she immerses herself in interlace but also clash. As a result of this, her sense of self is constantly in flux as she attempts to achieve stability and coherence. This sense of a gendered, stable and located self will, I argue, both dissipate and transmutate upon undergoing physical or imagined translocation. In addition, this thesis examines the manner in which globalism allows for the dissolving of boundaries and explores the extent to which the ambiguous position these female protagonists occupy enables them to reformulate and refashion their gender identity as well as write themselves away from the marginalised positions they inhabit. I will further explore how female subjects are compelled to take on additional feminine or masculine attributes upon translocation, seeming to become androgynous in the reformulation of their gender identity for a certain period of time. I will argue that protagonists supplement their gender in order to obtain a sense of belonging in a specific cultural context which requires this alteration of gender, and argue that this is also a means by which they liberate themselves from the marginal positions they occupy in their ethnic culture where sexism and prejudice are prevalent. However, I will demonstrate that modernity does not only provide them with liberation and autonomy, but that simultaneously it is also restrictive on the subject’s gender identity. Finally, this thesis explores whether the female protagonists are able to use their ambiguous positioning strategically in order to generate coherence of the self yet, concurrently, maintain fluidity between multiple cultural boundaries of the self. / AFRIKAANSE OPSOMMING: Hierdie verhandeling gebruik geslags- en subjektiwiteitsteorieë om ondersoek in te stel na die maniere waarop konstruksies van moderniteit en tradisie uiting vind in geslagshandeling. Ek wil ook kyk na die maniere waarop die vroulike subjek genoop is om, as gevolg van translokasie, addisionele vroulike of manlike karaktertrekke aan te neem, met dié dat dit blyk dat die protagoniste vir 'n ruk lank androgene eienskappe in hul geslagsidentiteit toon. Ek argumenteer dat die protagoniste hul geslag aanvul, nie net sodat hul aanklank binne 'n spesifieke kulturele konteks kan vind nie, maar ook as 'n manier waarop hul hulself kan bevry van die marginale posisies waarin hulle hul in 'n etniese kultuur, waar seksisme en vooroordeel gedy, bevind. Nietemin wil ek ook aantoon dat moderniteit nie bloot net bevryding en selfstandigheid aan die vroulike protagoniste bied nie, maar dat dit ook tegelykertyd beperkings op die subjek se geslagsidentiteit plaas. Die uitkoms van hierdie tesis is om te bepaal of die vroulike protagoniste in staat is tot die strategiese gebruik van hul dubbelsinnige posisionering, wat koherensie van die self sal meebring, en tog terselfdertyd vloeibaarheid tussen verskillende kulture sal behou.

The thesis then points out the importance of the influence of a family, school and the environment where the children live and grow up. Primary school pupils, concretely second class pupils in the age of 7 to 8 were the subject of the research. The method of written questioning was used for the research. The research showed that children have positive stance to sport. Furthermore, parents help children develop healthy nutrition and healthy lifestyle habits and children understand these terms.

The aim of this graduation theses is finding out the children{\crq}s conception of health, nutrition, exercise and sport. The object of research is children{\crq}s relation to exercise and nutrition. This theses is also describing influence of family, school, television and surroundings for formation of this relation. It also describes cooperation with pupils. 115 children of the third classes of primary schools were researched. These children were in the age of 8 {--} 9 years. The used methods in this research were questionnaires and observations. The research showed that the majority of pupils take right view to health, nutrition and exercise and most of the children practise some sport. This theses showed the problems of the way living of pupils in the third classes of primary schools.

Pathogenic to men and animals is found naturally in the environment and has the ability to multiply on adverse conditions such as high salinity and chilling temperatures. However, their detection in food is difficult because it is laborious, time consuming and expensive. Therefore comes to searching for methods to detect simple and fast. Immunological methods are very promising in this regard, but the conditions under which the organism is grown should be ideal for maximizing the expression and subsequent detection of the target antigen. Internalin A protein (InlA) of L. monocytogenes is an excellent target for detection of this pathogen in immunological tests, but the ideal conditions to enhance its expression had not yet been described. Therefore thus study analyzed the expression of InlA in two strains of L. monocytogenes serotype 4b, a clínical and other non-clínical, in non selective enrichment broths Luria-Bertani (LB), Brain Heart Infusion (BHI), Tryptic Soy Broth (TSB) and selective broths Fraser Broth (FRA), Listeria Enrichment Broth (LEB), Listeria Enrichment Broth - University of Vermont Medium (UVM), to incubation at 29 and 37 °C, through the ELISA and real time RT-PCR. All data were statistically analyzed considering a significance level of 5% (p <0.05). In the ELISA it was found that expression of InlA is strain-specific, in other words, was influenced by the origin of the strain, because the strain non-clínical of L. monocytogenes showed higher InlA expression levels than the clínical strain, and the medium used directly interfere with the expression of this antigen, and the most appropriate medium of enrichment for use in detection methods, regardless of the origin of the strain,was FRA, TSB and LEB. It was also observed that there was no significant difference in the expression of InlA when strains were grown at 29 and 37 °C. The real-time RT-PCR data showed inconclusive, since there was no statistically significant difference between the conditions analyzed, requiring thus more study. In conclusion, it was found that InlA gene expression on influenced by origin of the strain and culture media. Regardless of the origin of the strain, the preferred media for use in InlA protein detection methods are FRA, TSB and LEB / Listeria monocytogenes é um microrganismo infeccioso causador de listeriose, uma doença de origem alimentar que acomete principalmente imunocomprometidos, gestantes, idosos e crianças. Considerado patogênico tanto para homens quanto para animais, está distribuído naturalmente no ambiente e possui a capacidade de multiplicar-se sobre condições adversas, como alta salinidade e temperaturas de refrigeração. Todavia, sua detecção em alimentos é dificultada por ser trabalhosa, demorada e dispendiosa. Por isso, vem-se buscando métodos de detecção mais simples e rápidos. Os métodos imunológicos são muito promissores neste sentido, mas as condições em que o microrganismo é cultivado devem ser ideais para maximizar a expressão e consequente detecção do antígeno alvo. A proteína internalina A (InlA) de L. monocytogenes é um excelente alvo para detecção desse patógeno em testes imunológicos, porém as condições ideais para potencializar sua expressão ainda não foram descritas. Portanto, nesse trabalho analisou-se a expressão de InlA em duas cepas de L.monocytogenes sorotipo 4b, uma de origem clínica e outra não-clínica, nos caldos de enriquecimento não seletivos Luria-Bertani (LB), Brain Heart Infusion (BHI) e Tryptic Soy Broth (TSB), assim como nos caldos seletivos Fraser Broth (FRA), Listeria Enrichment Broth (LEB) e Listeria Enrichment Broth - University of Vermont Medium (UVM), a 29 e 37 °C, utilizando ELISA indireto e RT-PCR em tempo real. Todos os dados obtidos foram submetidos a análises estatísticas considerando um nível de significância de 5% (p<0,05). Através do ELISA indireto verificou-se que a expressão da InlA é cepa-especifica, ou seja, foi influenciada pela origem da cepa, pois a L. monocytogenes não-clínica apresentou maiores níveis de expressão de InlA do que a cepa clínica, e que os meios utilizados interferem diretamente na expressão desse antígeno, sendo os meios de enriquecimento mais indicados para uso em métodos de detecção, independentemente da origem da cepa, o FRA, TSB e LEB. Observou-se também que não ocorreu diferença significativa na expressão de InlA quando as cepas foram cultivadas a 29 e 37°C. O RT-PCR em tempo real apresentou dados inconclusivos, visto que não houve diferença estatística significativa entre as condições analisadas, necessitando, dessa forma, de maiores estudos.

Growth regulators IBA, BAP, NAA and GA3 were used in the regeneration of the hypocotyl, cotyledonary node, root, epicotyl and cotyledon. Antibiotics for suppression of Agrobacterium tumefacien (timentin, cefotaxime and carbenicillin) and for selection of transgenic cells (hygromycin) were also evaluated. Finally, tests with the biological insecticide DipelTM and spores of B. thuringiensis grown in laboratory were performed with the caterpillar of Hyblaea puera. TDZ increases 46% the regeneration frequency and BAP 26% when compared to controls. Cotyledonary nodes showed the best regeneration frequency (70%) growing on MS medium added of 1 mg.L-1 BAP + 0.5 mg.L-1 GA3. Hypocotyls, roots, cotyledons and epicotyls presented variable frequency of regeneration (60%, 60%, 30%, and 10% respectively) growing on distinct concentrations of grown regulators. We tested three antibiotics (timentin, cefotaxime, and carbenicillin) to suppress A. tumefaciens in vitro growth and they presented different effects on the organogenesis of each explants used in this study. Timentin and cefotaxime at concentration of 300 mg.L-1 increased the number of buds on hypocotyls and cotyledonary nodes. Conversely, these antibiotics had negative effects on the number of shoots of cotyledonary explants. Carbenicillin at all doses presented a negative influence on regeneration of all explants. Hygromycin at concentration of 2.5 mg.L-1 inhibits 100% of regeneration of cotyledons, cotyledonary nodes, and hypocotyls. The young instars of H. puera are susceptible to likely both commercial product DipelTM and spores grown in the laboratory, presented 100% mortality at concentrations of 2x105 CFU after 24 hours of ingestion. These findings suggest its potential to be used in teak transgenic approaches for insect resistance. Our results contribute to information about factors that influence the organogenesis of this specie, as well as define parameters that can be used in future experiments aimed at the genetic transformation of the specie.

Alice’s Vacillation between Childhood and Adolescence in Lewis Carroll’s Alice’s Adventures in Wonderland.

In the novel Alice’s Adventures in Wonderland by Lewis Carroll, Alice, the protagonist, is supposed to be seven years of age. However, the reader can perceive her as older than that and get the impression that she has entered adolescence. Alice vacillates between being a child and striving to act like an adult in her various encounters in Wonderland. In this essay, I will examine Alice’s emotional and intellectual phases in her search for identity, and show the different levels according to developmental theory. Erik Erikson’s, Jean Piaget’s and John Dewey’s research together with other studies form the theoretical framework of this paper. I will demonstrate that while the book does not trace her development as such (i.e. it is not a typical Bildungsroman), it nevertheless highlights a child’s development by juxtaposing different developmental stages. The scientific and realistic functions of developmental theory may at first seem haphazard in the analysis of a literary character in a fantasy world. But, this essay illustrates Carroll’s professional familiarity with his child protagonist through the logic and consistency of his depiction of Alice.  Alice’s adventures in Wonderland reflect the child-adult conflict of Alice on her inner quest for identity. To her the first steps into adulthood, ie. adolescence, include not only psychological growth as in maturity but also physical growth; to grow is to grow up. Her dramatic alterations in size in Wonderland cause great turmoil and confusion as she senses an obligation to adapt her behavior.  Lewis Carroll knew his child protagonist well.
