A study of the molecular mechanism of progestin-induced regulation of IL-12 and IL-10 and implications for HIV pathogenesis

Thesis (PhD)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: Medroxyprogesterone acetate (MPA) and norethisterone (NET) and its derivatives (norethisterone enanthate (NET-EN); norethisterone acetate (NET-A)), designed to mimic the actions of the endogenous hormone progesterone (Prog), are extensively used by women as contraceptives and in hormone replacement therapy (HRT). A number of reports have indicated that these synthetic progestins affect immune function in the female genital tract thereby increasing the risk of acquiring sexual transmitted infections. Despite these findings, very little is known about their mechanism of action at the cellular level, in particular their steroid receptor-mediated effects on cytokine gene expression. In the first part of this thesis, the effect of Prog, MPA and NET-A on the expression of the endogenous pro-inflammatory cytokine gene, interleukin (IL)-12p40, and anti-inflammatory cytokine gene, IL-10, was investigated in a human ectocervical epithelial cell line, Ect1/E6E7. Quantitative realtime PCR (qPCR) showed that all three ligands significantly upregulated the tumor necrosis factor alpha (TNF )-induced IL-12p40 gene expression, while IL-10 gene expression was downregulated. Moreover, by reducing the glucocorticoid receptor (GR) levels with siRNA, these effects were shown to be mediated by the GR. A more detailed investigation into the molecular mechanism of the progestogen-induced upregulation of IL-12p40 gene expression, using chromatin immunoprecipitation (ChIP), siRNA, co-immunoprecipitation and re-ChIP analyses, showed that the progestogen-bound GR is recruited to the CCAAT enhancer binding protein (C/EBP)- regulatory element of the IL-12p40 promoter, most likely via an interaction with the transcription factor C/EBP . Similar experiments for the progestogen-induced downregulation of IL-10 gene expression showed that the progestogen-bound GR is recruited to the signal transducer and activator of transcription (STAT)-3 regulatory element of the IL-10 promoter, most likely via an interaction with the transcription factor STAT-3. The second part of this study elucidated the influence of the HIV-1 accessory viral protein R (Vpr) on progestogen-induced regulation of IL-12p40, IL-12p35 and IL-10 in the Ect1/E6E7 cell line. Results showed that in these cells, the overexpression of Vpr significantly modulated the effects of Prog, MPA and NET-A on the mRNA expression of IL- 12p40 and IL-10, while only the NET-A effect was modulated on IL-12p35. Moreover, reducing the GR protein levels by siRNA suggested that the GR is required by Vpr to mediate its effects. Taken together, these results show that Prog, MPA and NET-A promote the pro-inflammatory milieu in the ectocervical environment, and that during HIV-1 infections, this milieu is modulated. Furthermore, the results suggest that the use of MPA or NET in vivo may cause chronic inflammation of the ectocervical environment, which may have important implications for ectocervical immune function, and hence susceptibility to infections such as HIV-1. / AFRIKAANSE OPSOMMING: Medroksieprogesteroon asetaat (MPA), noretisteroon (NET) en derivate daarvan noretisteroon enantaat (NET-EN); noretisteroon asetaat (NET-A), ontwerp om die funksies van die natuurlike hormone progesteroon (Prog) na te boots, word wêreldwyd deur vroue as voorbehoedmiddels sowel as vir hormoon vervangingsterapie (HVT) gebruik. Daar is verskeie aanduidings dat hierdie sintetiese progestiene die immuunfunksie in die vroulike geslagskanaal kan beïnvloed en ook die moontlike vatbaarheid van seksueel oordraagbare infeksies kan verhoog. Ten spyte hiervan, is baie min bekend oor hulle meganisme van werking op ‘n molekulêre vlak, veral in die besonder hul effek op sitokinien geenuitdrukking. Die effek van Prog, MPA en NET-A op die geenuitdrukking van ’n endogene pro-inflammatoriese sitokinien, interleukin (IL)-12, en ’n anti-inflammatoriese sitokinien, IL-10, asook die onderliggend meganisme van werking, in ’n menslike ektoservikale sellyn, Ect1/E6E7, is in die eerste deel van hierdie studie ondersoek. Verder is gevind dat induksie van IL-12p40 en inhibisie van IL-10 deur Prog, MPA en NET-A deur die glukokortikoïed reseptor (GR) gedryf word, aangesien volledige opheffing van die effekte op hierdie sitokinien gene waargeneem is wanneer die GR proteïen vlakke deur middel van kort inmengende ribonukleïensuur (siRNS) verminder is. 'n Meer beskrywende ondersoek in die molekulêre meganisme is uitgevoer deur gebruik te maak van chromatien immunopresipitasie (ChIP), siRNS, mede-immunopresipitasie en her-ChIP analises. Hierdie resultate het voorgestel dat die progestogeen (Prog en die sintetiese progestiene)-gebonde GR tot die CCAAT verbeterende bindings protein (C/EBP)- regulatoriese element van die IL-12p40 promotor betrek word en dat die transkripsie faktor C/EBP benodig word om transkripsie van die IL-12p40 geen te aktiveer. Vermindering van die GR proteïen vlakke deur middel van siRNS het getoon dat Vpr die GR benodig om hierdie veranderinge mee te bring. In samevatting, die resultate van hierdie proefskrif stel voor dat Prog, MPA en NET-A die pro-inflammatoriese milieu in die ektoservikale omgewing bevorder, en dat hierdie milieu gedurende MIV-1 infeksies verander. La maladie d'Alzheimer (MA) est une maladie neurodégénérative irréversible et progressive entraînant des troubles cognitifs et comportementaux. L'inflammation est caractéristique de la MA. L'interleukine-10 (IL-10), une cytokine anti-inflammatoire a été associée à un risque plus faible de développer la MA. Cependant le lien entre l'IL-10 et la progression de la MA n'a jamais été étudié. Le but de cette thèse a été d'étudier le rôle de l'IL-10 dans le développement ainsi que dans la progression de la MA. Pour mener à bien cette étude, 31 sujets atteints par la MA et 20 sujets contrôles cognitivement intacts ont été recrutés. En fonction de la vitesse de diminution du test de Mini-Mental State Examination et de l’évolution des troubles cognitifs sur deux ans, les patients souffrant de la MA ont été divisés en deux sous-groupes: les patients avec une progression lente (MA lent) et ceux avec une progression rapide (MA rapide). Les analyses se sont portées sur la concentration d’IL-10 en périphérie (plasma, production par les cellules mononuclées du sang périphérique (PBMCs) après stimulation par les peptides Aβ) ainsi que le polymorphisme de son promoteur en position -592, -819 et -1082. En complément, d’autres cytokines impliquées dans l’inflammation ont été étudiées : l’IL-6 (sa concentration plasmatique, sa production par les PBMCs à la suite d’une stimulation par les peptides Aβ et son polymorphisme en position -174) et les polymorphismes du TGF-β1 (-10 et - 25), de l’IFN-γ (-874) et du TNF-α (-308) ainsi que le gène de l'apolipoprotéine E (ApoE). Une étude de la longueur des télomères, liée à l’inflammation, a été aussi réalisée. Les résultats ont montré une association entre le génotype AA et l’allèle A du polymorphisme de l’IFN-γ en position -874 avec la progression rapide de la MA. Une augmentation statistiquement significative de la production d'IL-10 après stimulation par les peptides Aβ a été montrée chez les patients atteints avec une progression lente (MA lent). Une longueur significativement plus courte des télomères a été aussi associée aux patients MA lent. L’ensemble de ces travaux suggère qu’un profil de forte production de l’IL-10 ainsi qu’un profil génétique d’IFN-γ (TT -874) pourrait ralentir la progression de la MA. Il est aussi apparu que la longueur des télomères pourrait être un marqueur du déficit cognitif. Il est clair que ces résultats préliminaires ont besoin d’être confirmés par une étude de plus grande envergure, avec un nombre de patients plus élevé. / Alzheimer’s disease (AD) is an irreversible and progressive neurodegenerative disorder leading to cognitive and behavioral impairment. Inflammation is hallmark of AD although the exact mechanisms involved and the roles of the different inflammatory components are far less clear. Interleukin-10 (IL-10) is a key anti-inflammatory cytokine and IL-10 -1082 A > G polymorphism has been associated with a lower risk of developing AD although the link between IL-10 and the AD progression have never been studied. The aim of this study is to study the role of IL-10 in the risk of developing AD and its role in AD progression. In order to complete successfully this study, 31 AD patients and 20 cognitively intact controls were recruited. Depending of the rate of decrease of mini-mental state test examination (MMSE) and evolution of cognitive disorders, AD patients were divided in two subgroups: patients with slow progression (AD slow) and those with fast progression (AD fast). Analysis were focused on periphery concentration of IL-10 (plasma and and its production by peripheral blood mononuclear cells (PBMCs) after Aβ peptides stimulation) as well as its promoter polymorphism in position -592, -819 and -1082. In addition, other cytokines involved in inflammation were studied: IL-6 (its plasma concentration, its production by PBMCs following stimulation with Aβ peptides and its polymorphism at position -174) and polymorphisms of TGF-β1 (-10 to - 25), IFN-γ (-874) and TNF-α (-308) as well as the gene polymorphisms of Apolipoprotein E (ApoE). A study of telomere length, link to inflammation, was also performed. Results showed IFNγ -874AA genotype and -874A allele was associated with AD fast progression. A statistically significant increase of IL-10 production by PBMCs stimulated with Aβ peptides was shown in AD slow patients. A significantly shorter telomere length was also associated with AD slow patients. All of this work suggests that a profile with high IL-10 production and high IFN-γ (-874 TT) genotype could confer a slower AD progression. It was also found that telomere length may be a marker of cognitive impairment. It is clear that these preliminary results need to be confirmed in a larger study with a larger number of patients.

Genetic mechanisms may modulate the response of an individual because they can interfere with gene expression of important mediators of inflammation. The objective of this study was to evaluate the association of the polymorphism of SNP-1082G / A gene promoter in interleukin-10 with periodontal disease in 36 cases and 30 controls. Strategy was used ARMS-PCR for allelic discrimination. Of individuals with periodontal disease, 16 (44%) had genotype AG, followed by 13 (36%) with genotype GG and 7 (20%) with genotype AA, highlighting a greater prevalence of heterozygotes for locus of IL-10, especially in individuals aged from 30 to 35 years in the control group, 13 (43%) of subjects had the genotype AG, 12 (40%) had GG and only 5 (17%) were classified as AA, highlighting a higher prevalence of heterozygotes for locus of IL-10, especially in individuals aged from 40 to 45 years. The data indicated that the study populations were in equilibrium according to Hardy-Weinberg Theorem. The analysis of the frequency of genotypes and allele frequencies concluded that no causal relationship found between the presence of genotype G or A and the development of periodontal disease in adults. The SNP-1082G / A IL-10 gene showed no predictive value for periodontal disease and therefore can not use it with prognostic value. / A periodontite é considerada uma desordem inflamatória de etiologia bacteriana que resulta em danos ao tecido periodontal, devido à complexa interação entre os periodontopatógenos e o sistema de defesa do hospedeiro. Mecanismos genéticos podem modular a resposta de um indivíduo uma vez que podem interferir na expressão gênica de importantes mediadores da inflamação. O objetivo deste estudo foi avaliar a associação do polimorfismo do SNP - 1082G/A no promotor do gene da interleucina-10 com a doença periodontal em 36 casos e 30 controles. Foi utilizada a estratégia de ARMS-PCR para a discriminação alélica. Dos indivíduos com doença periodontal, 16 (44%) apresentaram o genótipo AG, seguidos de 13 (36%) com o genótipo GG e 7 (20%) com o genótipo AA, destacando uma maior prevalência de heterozigotos para o loco da IL-10, principalmente nos indivíduos da faixa etária de 30 a 35 anos , no grupo controle, 13 (43%) dos indivíduos apresentaram o genótipo AG, 12 (40%) apresentaram GG e apenas 5 (17%) foram classificados como AA, destacando uma maior prevalência de heterozigotos para o loco da IL-10, principalmente nos indivíduos da faixa etária de 40 a 45 anos . Os dados indicaram que as populações estudadas encontravam-se em equilíbrio, segundo o Teorema de Hardy-Weinberg. A análise das frequência dos genótipos e das frequências alélicas permitiram concluir que não encontramos relação causal entre a presença do genótipo G ou A e o desenvolvimento da doença periodontal em adultos. O SNP -1082G/A do gene da IL-10 não mostrou valor preditivo para a doença periodontal e, portanto, não podemos usa-lo com valor prognóstico.

O desenvolvimento do câncer está associado a falhas no sistema imune. Investigamos como o microambiente tumoral de mama afetaria a diferenciação de monócitos. Observamos que a alta frequência de macrófagos (MΦ) CD163+ associou-se à baixa sobrevida das pacientes e a um baixo infiltrado de células T CD3+ nos tumores. Sobrenadantes obtidos da dilaceração des tumores (SNDil) induziram a diferenciação de monócitos para MΦ CD163highIL-10high, os quais suprimiram células T CD4+. O fenótipo CD163highIL-10high associou-se a altos níveis de M-CSF, TGF-β e VEGF nos tumores. Monócitos circulantes de pacientes falharam em diferenciar-se em M1-M Φ; deram origem à DCs capazes de induzir alta frequência de células T reguladoras (Tregs) e produziram altos níveis de citocinas anti-inflamatórias sob ativação por LPS. Em conclusão, o microambiente tumoral favorece a diferenciação de MΦ CD163highIL-10high supressivos e afeta sistemicamente o potencial de diferenciação de monócitos de pacientes. / Cancer development is associated with failures in the immune system. We investigated here if breast tumor microenvironment affect the differentiation of monocytes. We observed that the high frequency of macrophages (MΦ) CD163+ was associated with poor patients survival and a low infiltration of CD3+ T cells in tumors. Supernatants obtained from dilacerated tumors (SNDil) skewed the differentiation of monocytes into CD163highIL10high MΦ, which suppressed CD4+ T cells. The CD163highIL-10high phenotype was associated with high levels of M-CSF, TGF-β and VEGF in tumors. Circulating monocytes from patients failed to differentiate into M1-MΦ; gave rise to DCs able to induce high frequency of regulatory T cells (Tregs) and produced high levels of anti-inflammatory cytokines under LPS activation. In conclusion, the tumor microenvironment promotes the differentiation of suppressive CD163highIL10high MΦ and affects the potential of differentiation of patients\' monocytes systemically.

Die Rolle regulatorischer T-Zellen bei der Masernviruspathogenese / The role of regulatory T-cells in measles virus pathogenesis

Tregs dienen zur Aufrechterhaltung der Balance im Immunsystem. Die Infektion, Aktivierung oder Induktion von Tregs durch Pathogene kann diese Balance empfindlich stören, eine Immunsuppression zur Folge haben und zur Ausbildung von Autoimmunerkrankungen oder Persistenzen beitragen. Das MV verfügt nicht nur über vielfältige Mechanismen der Immunsuppression, während einer MV-Infektion herrschen zudem Bedingungen vor, welche die Zahl und Aktivität von Tregs beeinflussen könnten. Aufgrund der Expression von Reifungsmarkern auf Trn ist zudem eine präferenzielle Infektion dieser Zellpopulation denkbar. MV-Infektionen können sowohl die akute MV-Enzephalitis, eine Autoimmunerkrankung, nach sich ziehen, als auch die Persistenz SSPE ausbilden. Ob diese Komplikationen mit spezifischen Aberrationen in der Menge und Aktivität von Tregs im Zusammenhang stehen, war bisher nicht bekannt. In dieser Arbeit konnte gezeigt werden, dass auf unstimulierten Trn der Reifungsmarker und MV-Rezeptor CD150 exprimiert wird und es in Folge dessen in vitro zu einer präferentiellen nicht produktiven Infektion und Depletion von Trn kommt. Ex vivo ließ sich ein deutlicher Depletionseffekt während der frühen akuten MV-Enzephalitis nachweisen, der nach Vaczinierung eines gesunden Probanden und Challenge eines immunisierten Affen nicht auftrat. Ob dieser Depletionseffekt ursächlich für die Enzephalitis ist, oder es sich um einen Begleiteffekt handelt ließe sich an Modellorganismen durch mitogene Manipulation der Trn während einer MV-Infektion untersuchen. Auch bei SSPE kann es zu einer Depletion von Trn kommen, dies scheint jedoch nicht mit der Progression dieser Erkrankung im Zusammenhang zu stehen. Wahrscheinlich ist dagegen ein Zusammenhang mit der Induktion von Tregs. In MV-stimulierten Proben von SSPE-Patienten wurde im Mittel signifikant mehr IL-10 exprimiert als in den Kontrollen. In Proben seropositiver gesunder Spender wurde IL-10 in den ersten Stunden nach MV-Stimulation fast ausschließlich von induzierten Tregs exprimiert. Weitere Versuche sind nötig, um die Evidenz zu steigern und zu ermitteln, ob auch in Patientenproben die frühe IL-10 Expression nach MV-Stimulation von induzierten Tregs dominiert wird. Zusammenfassend kann gesagt werden, dass es sowohl bei der MV-Enzephalitis als auch bei SSPE zu signifikanten Wechselwirkungen mit Tregs kommt. Ob sich eine MV-Enzephalitis auch ohne Depletion von Trn ausbilden kann und ob die Ausbildung von SSPE erhöhte IL-10 Expression voraussetzt, werden weitere Untersuchungen ergründen müssen. / Treg are supposed to keep the balance throughout the immune system. Infection, activation and induction of Treg by pathogens can interrupt with this balance. Immunosuppression autoimmunity and viral persistence can result from this interference. MV does not only cause immunosuppression during infection by multifaceted mechanisms, but also effects circumstances known to interfere with the frequency and activity of Treg. Due to the expression of maturity markers on the surface of Trn a preferential infection of this cell population by MV seems possible. MV-infection can involve both, acute MV-encephalitis, an autoimmune disease, and the virus persistence SSPE. It was not investigated until now, if thous complications are associated to specific aberrations in the frequency and activity of Treg. In this paper it was demonstrated that CD150, which is both a maturity marker and a MV-receptor, is expressed on unstimulated Trn. Due to this expression the Trn are infected preferentially and non productively in vitro leading to apoptosis. Ex vivo a noticeable depletion effect was detected during the early acute MV-encephalitis. This effect did not accure after vaccination of a healthy proband, and the challenge of an immunised monkey. Mitogenic manipulation of Trn in model organisms during MV-infection could give advice, if this depletion effect is causal to MV-Encephalitis or rather a concomitant effect. Depletion of Trn can also arise in the presence of SSPE, but this seems not to interfere with the progression of disease. By contrast there is presumable an interrelation of SSPE with the induction of Tregs. In MV-stimulated probes from SSPE-patients the median IL-10 expression was significantly higher than in controls. Probes from seropositive healthy donators were used to identify the IL-10 expressing cells. During the first hours after MV-stimulation IL-10 was mainly expressed by induced Treg. Subsequent investigations are necessary to enhance the evidence and to determine if induced Treg also dominate the early IL-10 expression after MV-stimulation in patient probes. To give a resume you could claim that there are significant interrelations between MV-encephalitis respectively SSPE and Treg. If a depletion of Trn is necessary for the development of MV-encephalitis and if increased levels of IL-10 expression are required for the formation of SSPE has to be examined in further experiments.

New mechanisms modulating S100A8 gene expression

S100A8 is a highly-expressed calcium-binding protein in neutrophils and activated macrophages, and has proposed roles in myeloid cell differentiation and host defense. Functions of S100A8 are not fully understood, partly because of difficulties in generating S100A8 knockout mice. Attempts to silence S100A8 gene expression in activated macrophages and fibroblasts using RNA interference (RNAi) technology were unsuccessful. Despite establishing validated small interfering RNA (siRNA) systems, enzymaticallysynthesized siRNA targeted to S100A8 suppressed mRNA levels by only 40% in fibroblasts activated with FGF-2+heparin, whereas chemically-synthesized siRNAs suppressed S100A8 driven by an S100A8-expression vector by ~75% in fibroblasts. Suppression of the gene in activated macrophages/fibroblasts was low, and some enzymatically-synthesized siRNAs to S100A8, and unrelated siRNA to GAPDH, induced/enhanced S100A8 expression in macrophages. This indicated that S100A8 may be upregulated by type-1 interferon (IFN). IFN-β enhanced expression, but did not directly induce S100A8. Poly (I:C), a synthetic dsRNA, directly induced S100A8 through IL-10 and IFN-dependent pathways. Induction by dsRNA was dependent on RNA-dependent protein kinase (PKR), but not cyclooxygenase-2, suggesting divergent pathways in LPS- and dsRNA-induced responses. New mechanisms of S100A8 gene regulation are presented, that suggest functions in anti-viral defense. S100A8 expression was confirmed in lungs from influenza virus-infected mice and from a patient with severe acute respiratory syndrome (SARS). Multiple pathways via mitochondria mediated S100A8 induction in LPS-activated macrophages; Generation of reactive oxygen species via the mitochondrial electron transport chain and de novo synthesis of ATP may be involved. This pathway also regulated IL-10 production, possibly via PKR. Extracellular ATP and its metabolites enhanced S100A8 induction. Results support involvement of cell stress, such as transfection, in S100A8 expression. A breast tumor cell line (MCF-7) in which the S100A8 gene was silenced, was established using micro RNA technology; S100A8 induction by oncostatin M was reduced by >90% in stably-transfected cells. This did not alter MCF-7 growth. The new approach to investigate the role of S100A8 in a human tumor cell line may assist in exploring its functions and lead to new studies concerning its role in cancer.

Modulation of Dendritic Cells with the Interleukin-10 Gene on Polycation-Modified Polymeric Particles

Gene therapy has emerged as a field to modulate cell functions by introducing genes of interest to target cells. An emerging focus in this field is to employ non-viral vectors to deliver immunosuppressive cytokines to dendritic cells (DCs) to attenuate damaging immune responses. DCs serve as potential targets for suppression of T cell responses. In this work, we investigated the ability of polycation-modified polymeric particles complexed with interleukin-10 (IL-10) gene to modulate DCs. The delivery systems (designated as PSO10H6 and PLGAO10H6) were formed by coating cationic peptide O10H6 (O: ornithine; H: histidine) on the polystyrene (PS) and poly (lactic-co-glycolic acid) (PLGA) particulates. A mouse IL-10 encoding plasmid (pIL-10) was loaded on the surface of PSO10H6 and PLGAO10H6 via ionic interactions. Physical characterization of these particles revealed stable colloidal dispersions (diameters: 297.2±14nm in PLGAO10H6-pIL-10 and 126.0±8nm in PSO10H6-pIL-10). DNA molecules carried by PSO10H6 and PLGAO10H6 were protected from serum digestion. Results from in vitro gene transfection studies showed two-fold enhancement of IL-10 expression in bone marrow-derived DCs transfected with PSO10H6-pIL-10 and PLGAO10H6-pIL-10 compared to untransfected DCs. Their suppressive functions were evaluated in an in vitro mixed lymphocyte model. Results indicated that PSO10H6-pIL-10 and PLGAO10H6-pIL-10 modified DCs elicited weakest proliferation of allogeneic bulk T cells as well as CD4 and CD8 T cells among all the delivery modes. Using cell-embedded Matrigel as a surrogate graft, we showed that IL-10 gene-modified DCs suppressed host cell infiltration in vivo. These data suggested PSO10H6-pIL-10 and PLGAO10H6-pIL-10 deliver an overriding suppressive signal to T cells. Further studies revealed T cells stimulated by the IL-10 gene-modified DCs exhibited characteristics of regulatory T (Treg) cells, as evident by up-regulation of a Treg cell marker forkhead-type transcription factor 3 (Foxp3). This result was concomitant with an increase in of transforming growth factor beta (TGF-beta) production. <br>Taken together, this work demonstrated that PSO10H6 and PLGAO10H6 are effective in delivering pIL-10 to modulate DCs to suppress T cell responses. Therapeutic immunomodulation of allergic lung disease using regulatory dendritic cells in a mouse model of asthma

We report herein that IL-10-treated dendritic cells (DC) can be used effectively to reverse established severe asthma-like disease in a mouse model. Our lab had shown previously that allergen-presenting splenic CD8¦Á+ DCs could ¡Ö50% reduce airway hyper responsiveness (AHR), eosinophilia, and Th2 responses in asthma-phenotype mice, but only marginally reduce IgE/IgG1 levels. We now show that bone marrow-derived DCs that have been differentiated in the presence of IL-10 (DCIL-10) are effective in reversing the asthma phenotype. Co-culture of DCIL-10 with T memory (TM) cells from asthma-phenotype mice was associated with lack of Th2 responses, and this was partially reversed by IL-2. Immunostimulatory DC activated these Th2 cells. <i>In vivo</i>, delivery of allergen-pulsed DCIL-10, either into the airway or intraperitoneally abrogated AHR from weeks 3-10 post-treatment, and ameliorated lung eosinophilia and Th2 (IL-4, -5, -9, & -13, IgE) responses, as well as circulating allergen-specific IgE responses for at least 32 weeks following treatment. Repeated OVADCIL-10 treatments kept AHR normalized for 8 weeks as well as Th2 responses significantly low. In vivo, delivery of anti-IL-10R, but not anti-TGF-¦Â from day 12-21 after treatment had moderate effects on DCIL-10-driven tolerance, but 1-methyl tryptophan (inhibitor of indoleamine-2,3-dioxygenase) treatment had significant effects on Th2 responses. The mechanisms mediating tolerance in vivo are likely complex, but we speculate that infectious tolerance sustains this regulatory activity during the 32-week period in which we have observed tolerance to be in place.
