Alterações morfológicas, apoptose e expressão de metaloproteinases no tecido laminar de equinos submetidos à obstrução intestinal experimental : efeitos da hidrocortisona /Laskoski, Luciane Maria. January 2009 (has links)

Sua patogenia este relacionada a ativação de metaloproteinases e degradação do colágeno e/ou hemidesmossomos. 0 objetivo deste estudo foi analisar alterações morfológicas, apoptose, metaloproteinases (MMP) 2 e 9 e infiltração leucocitária, pela pesquisa da lipocalina liberada por neutrófilos (NGAL), no tecido laminar de equinos submetidos a obstrução intestinal experimental. Os animais foram distribuídos nos grupos: controle (Gc), instrumentado (Gi), com procedimento de enterotomia sem obstrução; obstruído tratado com hidrocortisona (Gt) e obstruído não tratado (Gnt). 0 tecido laminar foi analisado pelas técnicas de microscopia em luz, imunohistoquímica e zimografia. Os animals dos grupos experimentais (Gi, Gt e Gnt) demonstraram alterações morfológicas, MMPs e substância formada pela M M P-9 e NGAL superiores aos resultados do Cc. A presença de apoptose não foi evidenciada pelo procedimento cirúrgico. 0 Gc não foi diferente quanto as alterações morfológicas quando comparado ao Gi, e também não diferiu do Gt em relação e MMP-9, principalmente associada a neutrófilos. Com estes resultados, conclui-se que: o modelo experimental pode causar alterações morfológicas associadas a laminite; a apoptose não participa da fase inicial de alterações laminares no córium laminar; a hidrocortisona pode ser utilizada para coibir os efeitos deletérios da ativação inflamatória associada a diversas doenças, sem comprometer o córium laminar. / Abstract: The laminitis is associated to systemic inflammatory activation that occur in several diseases, mainly gastrointestinal. The pathogeny of the laminitis is related to matrix metalloproteinases (MMPs) activation with collagen or hemidesmossomos degradation. The aim of this study is evaluated of the morphologic alterations, apoptosis, MMPs 2 and 9 and leukocyte infiltration, for research of MMP-9 related to neutrophil gelatinase associated to lipocalin (NGAL) in laminar corium of horses subjected to intestinal obstruction experimental. The animals were divided in groups: control (Cc), instrumented (Gi), with enterotomy without obstruction; obstruction treated by hydrocortisone (Gt) and without treatment (Gnt). The laminar tissue was analyzed by optical microscopy, immunohistochemistry and zymography. The animals showed morphologic alterations, MM Ps expression and MMP-9 associated to NGAL greater the Gc. The presence of apoptosis was not evidend by cirurgical procedure. However, the Gc was not different to Gi about morphologic alterations, and too Gt, about MMP-9, mainly the form conjugated to NGAL. The results indicate that the experimental model can cause morphologic alterations associated with laminitis. The apoptosis is not prerequisite for laminar lesion. The hydrocortisone can be administrated to reduce the deleterious effects of inflammatory activation in several diseases, without to affect the laminar corium. / Mestre

Estudo do polimorfismo dos genes KIR e HLA em pacientes com câncer de mama e grupo controleJobim, Maria Regina Sampaio Leite January 2014 (has links)
O presente estudo tem como objetivo investigar a frequência dos diversos polimorfismos dos genes KIR (Killer Immunoglobulin-like Receptors) e HLA C1 e C2 em um grupo de pacientes com câncer de mama e comparar com um grupo controle de indivíduos sadios. As células natural killer (NK) são linfócitos que diferem das células T e B e que fazem parte da imunidade natural, reconhecendo as moléculas HLA (Antígenos Leucocitários Humano) de classe I em células infectadas por vírus ou em células tumorais, através de seus receptores de membrana. Os principais receptores das células NK são conhecidos como receptores KIR, sendo codificados por genes localizados no cromossomo 19q13.4 e classificados em grupos funcionais supressores e ativadores. Neste estudo, analisamos 15 genes KIR e alelos do sistema HLA de classe I em 230 pacientes caucasóides e em 278 controles, usando a técnica de PCR com primers específicos (PCR-SSO e PCR-SSP). Nossos resultados demonstraram uma frequência maior do genótipo supressor 2DL2 (P<0,001) em pacientes com câncer de mama, quando comparados ao grupo controle. Os genes HLA-C2 e HLA-BW4 não apresentaram diferenças significantes entre os grupos. Contudo, o gene HLA-C1 foi observado em maior frequência nos pacientes com câncer de mama. Considerando que estes achados sugerem uma potencial associação entre o sistema de genes KIR, HLA classe I e o câncer de mama, estudos adicionais sobre este tema são necessários. / We investigated the frequency of various KIR (Killer Immunoglobulin-like Receptors) and HLA C1 and C2 gene polymorphisms in a group of patients with breast cancer and healthy controls. Natural Killer (NK) cells are lymphocytes that differ from T and B cells and are part of the innate immune system, recognizing class I Human Leukocyte Antigens (HLA) molecules on target cells (virus-infected as well as cancer cells), through specific cell surface receptors. KIR comprises the main class of NK receptors, being encoded by genes located in chromosome 19q13.4. They possess both suppressor and activating functional groups. Fifteen KIR genes and class I HLA alleles obtained from 230 Caucasians patients, as well as 278 controls were studied, using PCR techniques with specific primers (PCR-SSO and PCR-SSP). Our results showed a higher frequency of suppressor genotype 2DL2 (P<0,001) in patients with breast cancer as compared to controls. No significant difference between HLA-C2 and HLA-BW4 alleles were observed between the study groups. Notably, a higher frequency of HLA-C1 gene was noted in patients with breast cancer. Our results suggest a potential association between KIR genes, HLA class I and breast cancer, deserving further investigation.

Foi estudado o efeito da lactona sesquiterpênica licnofolido sobre o \"burst\" respiratório de leucócitos polimorfonucleares inflamatórios (PMN) estimulados por forbol (PMA), pelo peptídeo quimiotático fMLP ou zimozan opsonizado (OZ). O licnofolido inibiu de forma dose-dependente a liberação de O2•- pelos PMN, sem alteração do período \"Iag\" do complexo NADPH. oxidase. O efeito foi mais acentuado quando os PMN foram estimulados diretamente pela via de proteína quinase C. A adição de ditiotreitol ou glutationa reduzida (GSH) às suspensões celulares antes da incubação com licnofolido preveniu parcialmente o efeito inibitório. O tratamento dos PMN com a lactona determinou uma queda drástica dos níveis celulares de GSH livre, sem incremento de glutationa oxidada (GSSG). A reação direta entre GSH e licnofolido foi confirmada com a detecção de um aduto glutationil-licnofolido através de identificação por espectrometria de massa (ESI-MS/MS). A S-tiolação protéica induzida pelo PMA foi reduzida em PMN tratados com Iicnofo/ido, como detectado através de determinação de incorporação de [35S], sendo que 80% desses tióis foram identificados como GSH. Uma série de proteínas S-glutationadas foi detectada através de autoradiografias, sendo que aquelas correspondentes a 38 e 24 kDa tiveram essa modificação póstraducional suprimida pelo tratamento com dose de licnofolido capaz de suprimir o \"burst\" respiratório dos PMN. Estes resultados indicam que a depleção celular de GSH causada pelo licnofolido impede a sustentação do \"burst\" respiratório pelos PMN, em correlação direta com a diminuição de S-glutationação protéica. / An investigation was made into the action of the sesquiterpene lactone lychnopholide on the respiratory burst of inflammatory polymorphonuclear leukocytes. Lychnopholide determined concentration-related inhibition of the generation of phorbol 12-myristate 13-acetate-, chemotatic peptide-, and opsonized zymozan-induced superoxide anion with no effect on the lag time of the assembly of the NADPH oxidase complex, such action was greater on the protein kinase C pathway that on both membrane receptor dependent stimuli via. Subsequent additions of D-glucose, Ca2+, Mg2+, dithiothreitol ar reduced glutathione (GSH) did not reverse the inhibitory action. The addition of both thiols prior to the lychnopholide treatment partially hindered the inhibition rate. The endogenous level of GSH in leukocytes was drastically depleted under the lychnopholide treatment, without corresponding increases occurring in the oxidized form (GSSG). A direct reaction between glutathione and lychnopholide was confirmed from a glutathionyl-lychnopholide adduct detected by electrospray mass spectrometry analysis and identified by tandem mass analysis in cellular extracts. Protein S-thiolation induced by PMA stimulation was decreased in lactone-treated PMN as detected by [35S] scintillation count, which indicated that about 80% of the thiols were glutathione. A subset of S-glutathionylated proteins was identified through gel electrophoresis, which revealed that the modification of the phorbol-triggered protein sulfhydryl in the protein bands corresponding to 38 and 24 kDa was precluded by the lychnopholide treatment correlated with respiratory burst inhibition. These results show that GSH depletion determined by lychnopholide treatment renders PMN to sustain respiratory burst, whose action is proportional to protein S-glutahionylation decrease.

Psychotria myriantha müll arg. (rubiaceae) : caracterização dos alcalóides e avaliação das atividades antiquimiotáxica e sobre o sistema nervoso centralFarias, Fabiane Moreira January 2006 (has links)
O gênero Psychotria destaca-se na família RUBIACEAE pela produção de alcalóides bioativos e por sua taxonomia complexa, sendo muitas vezes relacionado aos gêneros Palicourea, Cephaelis, Calycodendron e Calycosia. A divisão de Psychotria nos subgêneros Psychotria, Tetramerae e Heteropsychotria foi proposta com o objetivo de auxiliar a classificação quimiotaxonômica do gênero. Estudos demonstram que o subgênero Psychotria (espécies pantropicais) produz alcalóides poliméricos, formados por duas ou mais unidades de triptamina; enquanto o subgênero Heteropsychotria parece estar envolvido com a produção de alcalóides indol monoterpênicos, de acordo com pesquisas realizadas com diferentes espécies coletadas no Sul do Brasil. O isolamento e purificação dos alcalóides estrictosamida, ácido estrictosidínico e miriantosina, a partir de Psychotria myriantha, corroboram com esta hipótese, permitindo a inclusão da espécie no subgênero Heteropsychotria. A literatura descreve várias atividades para extratos e alcalóides isolados de espécies de Psychotria, como antimicrobiana e analgésica, por exemplo. Neste trabalho, o extrato n-butanólico de alcalóides de P. myriantha, além de seus alcalóides isolados, apresentaram atividade inibidora da migração de leucócitos, sugerindo um efeito antiinflamatório, e capacidade de inibir a ação da enzima acetilcolinesterase. Extratos e alcalóides isolados da espécie foram avaliados quanto à atividade antioxidante em CCD, frente ao DPPH, apresentando resultado negativo. O extrato EBA e o alcalóide ácido estrictosidínico aumentaram o tempo de latência no teste da retirada da cauda frente ao estímulo térmico, indicando uma atividade analgésica do tipo opióide. A influência do ácido estrictosidínico, alcalóide isolado em maior quantidade em massa, sobre os níveis de DA, DOPAC, 3-MT, HVA, 5-HT e 5-HIAA em estruturas cerebrais de ratos foi verificada. Hipocampos de animais que receberam injeção intra-hipocampal bilateral de ácido estrictosidínico (20 μg/μL) apresentaram redução de 83,4 % nos níveis de serotonina, em comparação ao grupo controle, enquanto os córtices desses animais apresentaram redução nos níveis de DOPAC (35,9%), 3-MT (24,7%) e 5-HIAA (9%). Hipocampos e estriados de ratos tratados com injeção i.p. de ácido estrictosidínico (10 mg/kg) demonstraram diminuição de 63,4 e 28,7% nos níveis de 5-HT, respectivamente. As alterações nos níveis de aminas biogênicas nas estruturas avaliadas, além das atividades analgésica e inibidora da acetilcolinesterase, indicam que P. myriantha e espécies do subgênero Heteropsychotria constituem uma potencial fonte de substâncias bioativas no tratamento de distúrbios do sistema nervoso central. / Psychotria genus is an important in RUBIACEAE due to bioactive alkaloids production and complex taxonomy, being related to Palicourea, Cephaelis, Calycodendron and Calycosia genera. The division of Psychotria in Psychotria, Tetramerae and Heteropsychotria subgenera was proposed with the aim of aiding the genus chemotaxonomic classification. Studies demonstrate that Psychotria subgenus produce polyindoline alkaloids formed by two or more triptamine units; whereas Heteropsychotria subgenus seems to be involved with indole monoterpene alkaloids production, according to researches with different species collected in Southern Brazil. Isolation and purification of strictosamide, strictosidinic acid and miriantosine from Psychotria myriantha corroborated this hypothesis, allowing its inclusion into Heteropsychotria subgenus. The scientific literature describes several activities to Psychotria extracts and alkaloids, such as antimicrobial and analgesic, for example. In this work, P. myriantha n-butanolic alkaloid extract and isolated compounds inhibited the leukocyte migration, suggesting an antiinflammatory activity, and a weak ability to inhibit the action of acetylcholinesterase enzyme. Alkaloids and extracts from P. myriantha were evaluated as regards their antioxidant activity using DPPH, with no positive results. EBA and strictosidinic acid increased the latency in the tail flick model, indicating an opioid analgesic activity. The influence of strictosidinic acid on the levels of DA, DOPAC, 3-MT, HVA, 5-HT and 5-HIAA in brain structures of rats was verified. Hippocampus with intra-hippocampal injection of strictosidinic acid (20 μg/μL) displayed a decrease of 83.4% in serotonin levels, in comparison with control group; whereas the cortex showed a decrease in the levels of DOPAC (35.9%), 3-MT (24.7%) and 5-HIAA (9%). Hippocampus and striatum that received intraperitoneal injection of strictosidinic acid (10 mg/kg) showed 5-HT levels reduction of 63.4 e 28.7%, respectively. The biogenic amine levels alterations in the studied structures, associated with the analgesic and acetylcholinesterase inhibitor activities, suggest that P. myriantha and species from the Heteropsychotria subgenus constitute a font of bioactive compounds in the treatment of central nervous system disturbs.

associated with immune-modulation and suppression of the immune response by the
interaction with specific NK and T cell receptors. The HLA-E gene is considered the
most conserved locus in the human HLA; however, this low variability might be a
consequence of the scarce number of studies focusing this subject. In this mastering
thesis we assessed the HLA-E coding and 3’ untranslated region variability in a
group of individuals from Brazil and the results were evaluated together with data
from the 1000Genomes Consortium. Altogether, only 28 variation sites were found in
approximately 2724 bp evaluated. These variation sites were arranged into 33
haplotypes, most of them (98.2%) encoding one of the two HLA-E molecules found
worldwide, i.e., the molecules associated with the allele groups E*01:01 and E*01:03.
Interestingly, 85% of all haplotypes were represented by only three different
sequences, each of them associated with one of the main known HLA-E coding
alleles, E*01:01:01, E*01:03:01 and E*01:03:02, all of them found worldwide. This
phenomenon, together with the comparisons with other primate sequences, reveals
that these two main allele groups (and molecules) arose early before human
speciation, and indicates that E*01:03:01 might be the oldest allele. In addition, the
low nucleotide diversity found for the HLA-E coding and 3’UTR in worldwide
populations suggests that the HLA-E gene is in fact a conserved gene, which might
be a consequence of its key role in the modulation of the immune system. / O loco HLA-E é um gene do Complexo Principal de Histocompatibilidade
Humano (MHC), cujo produto está relacionado com a modulação e supressão da
resposta imunitária por meio da interação com receptores específicos das células
NK e linfócitos T. O gene HLA-E é considerado o loco menos polimórfico dos genes
do complexo HLA, no entanto, esta baixa variabilidade pode ser uma consequência
do pequeno número de estudos realizados sobre esse tema. No presente trabalho, a
variabilidade das regiões codificadoras e 3’ não traduzida do gene HLA-E foi
analisada em amostras brasileiras e os resultados foram comparados com dados
obtidos pelo projeto 1000Genomes. Considerando todas as populações avaliadas,
apenas 28 pontos de variação foram encontrados em uma região de
aproximadamente 2724-pb. Estes pontos de variação estão arranjados em 33
haplótipos diferentes, a maioria deles (98%) codificando uma das duas moléculas
HLA-E frequentemente encontradas, E*01:01 e E*01:03. Ainda, 85% dos haplótipos
encontrados foram representados por apenas três sequências diferentes, cada uma
deles associada a um dos principais alelos da região codificadora do gene HLA-E,
E*01:01:01, E*01:03:01 e E*01:03:02. Todas essas sequências foram encontradas
em todas as populações avaliadas. Este fenômeno, em conjunto com as
comparações envolvendo sequências de primatas, sugere que estes dois grupos de
alelos principais (e moléculas) surgiram antes da especiação e dispersão humana,
além de indicar que o alelo E*01:03:01 pode ser o mais antigo dentre os demais.
Ainda, a baixa diversidade nucleotídica encontrada para a região codificadora e 3'
NT do gene HLA-E em populações de todo o mundo sugere que este gene é, de
fato, bastante conservado, provavelmente devido ao seu papel chave na modulação
das respostas imunes.

and presumption of black ancestry. There are few studies and data about
health of these subjects, and between available study highlight higher
prevalence of overweight and related diseases. However, national survey
data also show that anemia and malnutrition are prevalent among black
women and children. Nevertheless, lacking most comprehensive studies to
understand the health profile of quilombo population from Brazil. The
objective of this research was to evaluate nutritional status, anemia
frequency and cardiometabolic risk factors in adults and elderly from
quilombolas communities of Goias state/Brazil. We carried out an analytical
cross-sectional study with 287 subjects. Were performed anthropometric
measurements (weight, height, waist circumference, and calculated BMI and
Waist to height ratio - WHtR) blood tests (complete blood count, serum
ferritin, fasting glucose and lipid profile), and socioeconomic and
demographic questionnaire. The first article discussed the rate of anemia and
iron deficiency and its relationship to overweight. Frequency of anemia and
iron deficiency was low (5,9% and 4,7% respectively) and wasn’t association
with overweight. Women had higher overweight than men, and men had
higher underweight. BMI was higher in urban communities and in adults.
Hemoglobin was lower in urban communities (p<0,05). The correlation
indicated that increasing BMI is accompanied by decreases in hemoglobin
and ferritin. RBC indices indicated 10,9% of microcytosis and 9,4% of
anisocytosis. The second article discusses the assessment of nutritional
status and association with cardiometabolic risk factors. Participants
presented 54,4% of overweight and 67,9% higher waist-to-height ratio. The
group with overweight differs from the eutrophic group in relation to HDL,
VLDL, TG, leukocytes, lymphocytes, and platelets. And 15.2% of the
population had three or more metabolic risk factors, moreover, in this group,
leukocytes and lymphocytes were significantly higher. Women had more
metabolic abnormalities than men. In conclusion, the main health problem in
these communities refers to overweight and related diseases. Metabolic
abnormalities increase the risk of developing cardiovascular disease, which
are the leading cause of death in Brazil. The low prevalence of anemia and
iron deficiency needs to be better investigated together food consumption.
It’s must also be investigated genetics anemia, such as thalassemia and
sickle cell due the abnormalities found in RBC indices without decreasing
hemoglobin and ferritin. / Comunidades quilombolas são grupos étnicos, definidos pelo critério de auto
atribuição que possuem presunção da ancestralidade negra. Existem poucos
estudos sobre a saúde desses sujeitos e, entre os existentes destacam-se
aqueles que indicam maior prevalência de excesso de peso corporal e
doenças associadas. Porém, dados de pesquisas nacionais apontam que a
anemia e o baixo peso são altamente prevalentes entre mulheres e crianças
negras. Apesar disso, faltam estudos mais abrangentes para compreender o
perfil de saúde especificamente da população quilombola. Portanto, o
objetivo dessa pesquisa foi avaliar o estado nutricional, frequência de
anemia e fatores cardiometabólicos de adultos e idosos de comunidades
quilombolas de Goiás. Realizou-se uma pesquisa transversal analítica com
287 sujeitos. Foram realizadas avaliações antropométricas (peso, estatura,
circunferência da cintura, cálculo do índice de massa corporal - IMC e da
relação cintura-estatura - RCE), exames de sangue (hemograma completo,
ferritina sérica, glicemia de jejum e lipidograma) e questionário
socioeconômico e demográfico. O primeiro artigo abordou o índice de
anemia e deficiência de ferro e sua relação com o excesso de peso. A
frequência de anemia e deficiência de ferro foi baixa (5,9% e 4,7
respectivamente) e não apresentou associação significativa com o IMC. A
frequência de excesso de peso foi elevada (52,0%), e as mulheres
apresentaram maior porcentagem de sobrepeso/obesidade do que os
homens. Na análise comparativa dos valores médios, observou-se que o
IMC foi maior nas comunidades urbanas e entre os adultos, e a hemoglobina
foi menor nas comunidades urbana (p<0,05). A correlação indicou que o

de anisocitose. O segundo artigo abordou a avaliação do estado nutricional e
a associação com fatores de risco cardiometabólicos. A frequência do
excesso de peso foi de 54,4%, e 67,9% dos sujeitos tinham RCE elevada.
As variáveis do perfil lipídico, glicêmico e de células brancas apresentaramse
alteradas nos grupos com excesso de peso e RCE aumentada. Porém, os
parâmetros metabólicos com diferença estatística entre os grupos foram o
CT, HDL, VLDL, Triglicerídeos, Leucócitos, Linfócitos e Basófilos. E 15,2%
dos participantes tinham três ou mais fatores de risco metabólico, sendo que
as mulheres apresentaram mais alterações metabólicas do que os homens.
Conclui-se que o principal agravo à saúde nas comunidades investigadas
refere-se ao excesso de peso e doenças associadas. As alterações
metabólicas aumentam o risco de desenvolvimento de doenças
cardiovasculares, que são a principal causa de óbito no Brasil. A baixa
prevalência de anemia e deficiência de ferro precisa ser melhor investigada
aliada ao consumo alimentar. Considera-se necessário investigar as
anemias de causa genética como as talassemias e a falciforme devido as
alterações que ocorreram nos índices hematimétricos sem diminuição da
hemoglobina e ferritina.

Factors determining the composition of a public cord blood stem cell bank including HLA diversityMellet, Juanita January 2013 (has links)
The human leukocyte antigen (HLA) is the most polymorphic region in the human genome and accounts for more than 10% of human diversity. This region plays an important role in matching donors and recipients for transplantation. The South African Bone Marrow Registry (SABMR) does not reflect the demographics of the South African population. The large number of polymorphisms resulting from HLA diversity in the Black South African population and their limited representation in the SABMR reduce the chances of finding adequate matches between donors and recipients in this group. Umbilical cord blood is an alternative to bone marrow for the treatment of fatal diseases. Less strict HLA matching is required due to the naive nature of the T cells in cord blood. A public umbilical cord blood bank is a necessity in trying to cater for the diverse population in South Africa. However, the ethnic diversity of the South African population poses a great challenge in constituting a public umbilical cord blood bank that is representative of the entire population. The Roche designed next generation sequencing (NGS) high resolution (HR) HLA typing kit enables sequencing of additional HLA exons and could improve the degree of matching between individuals to ultimately decrease adverse reactions. An extensive study of the literature was performed to establish the demographics, linguistics, and HLA diversity of the South African population to determine how a public cord blood bank should be constituted. In addition, HLA genotyping was performed by 454 NGS on 20 samples that had previously been HLA typed by conventional methods. The 454 NGS technique made use of a Roche designed medium and high resolution HLA typing kit to genotype the samples. It was possible to assign accurate genotypes to 95.5% of the loci of interest for the total number of 20 samples using the MR kit, compared with 98.5% using the HR kit. In conclusion, the present study indicates the extreme HLA diversity in the South African population, and therefore, recommends constituting the first public umbilical cord blood bank in Gauteng on the basis of race or major ethnic groupings. A minimum number of 10 000 cord blood units is needed to initiate the bank. Chronically Elevated Corticosterone Levels, via Cocoa Butter Injections of Corticosterone, Do Not Affect Stress Response, Immune Function, and Body Condition in Free-living Painted Turtles (Chrysemys picta)Juneau, Véronique January 2014 (has links)
Chronic stress can result in elevated circulating levels of glucocorticoid hormones in vertebrates, which can affect their stress response, their immune function, and eventually their fitness. I tested the effect of chronic corticosterone (CORT) elevation on the acute stress responsiveness, immune function, and body condition of free-living painted turtles (Chrysemys picta) in Gatineau Park, using slow-release exogenous CORT administration. While Silastic implants did not predictably elevate circulating CORT concentrations in painted turtles, injections of CORT-laden cocoa butter kept circulating levels elevated for up to 3 weeks, to concentrations likely physiologically and ecologically relevant for the species. I measured the acute CORT stress response, parasitaemia, heterophil-to-lymphocyte ratios, and total leukocyte counts after 1 week and 3 weeks, and determined body condition after 1 week, 3 weeks, and 1 year. Compared to sham and control turtles, I observed no effect of treatment on these hormonal, immune, and body condition metrics of stress, possibly because CORT mediates resource allocation only in the presence of additional immune or energy challenges, because of the masking effect of extrinsic factors, or because free, not total, CORT appears to be biologically active.

Estudo do polimorfismo dos genes KIR e HLA em pacientes com câncer de mama e grupo controleJobim, Maria Regina Sampaio Leite January 2014 (has links)
O presente estudo tem como objetivo investigar a frequência dos diversos polimorfismos dos genes KIR (Killer Immunoglobulin-like Receptors) e HLA C1 e C2 em um grupo de pacientes com câncer de mama e comparar com um grupo controle de indivíduos sadios. As células natural killer (NK) são linfócitos que diferem das células T e B e que fazem parte da imunidade natural, reconhecendo as moléculas HLA (Antígenos Leucocitários Humano) de classe I em células infectadas por vírus ou em células tumorais, através de seus receptores de membrana. Os principais receptores das células NK são conhecidos como receptores KIR, sendo codificados por genes localizados no cromossomo 19q13.4 e classificados em grupos funcionais supressores e ativadores. Neste estudo, analisamos 15 genes KIR e alelos do sistema HLA de classe I em 230 pacientes caucasóides e em 278 controles, usando a técnica de PCR com primers específicos (PCR-SSO e PCR-SSP). Nossos resultados demonstraram uma frequência maior do genótipo supressor 2DL2 (P<0,001) em pacientes com câncer de mama, quando comparados ao grupo controle. Os genes HLA-C2 e HLA-BW4 não apresentaram diferenças significantes entre os grupos. Contudo, o gene HLA-C1 foi observado em maior frequência nos pacientes com câncer de mama. Considerando que estes achados sugerem uma potencial associação entre o sistema de genes KIR, HLA classe I e o câncer de mama, estudos adicionais sobre este tema são necessários. / We investigated the frequency of various KIR (Killer Immunoglobulin-like Receptors) and HLA C1 and C2 gene polymorphisms in a group of patients with breast cancer and healthy controls. Natural Killer (NK) cells are lymphocytes that differ from T and B cells and are part of the innate immune system, recognizing class I Human Leukocyte Antigens (HLA) molecules on target cells (virus-infected as well as cancer cells), through specific cell surface receptors. KIR comprises the main class of NK receptors, being encoded by genes located in chromosome 19q13.4. They possess both suppressor and activating functional groups. Fifteen KIR genes and class I HLA alleles obtained from 230 Caucasians patients, as well as 278 controls were studied, using PCR techniques with specific primers (PCR-SSO and PCR-SSP). Our results showed a higher frequency of suppressor genotype 2DL2 (P<0,001) in patients with breast cancer as compared to controls. No significant difference between HLA-C2 and HLA-BW4 alleles were observed between the study groups. Notably, a higher frequency of HLA-C1 gene was noted in patients with breast cancer. Our results suggest a potential association between KIR genes, HLA class I and breast cancer, deserving further investigation.

In patients after cardiac surgery, there is an increase in the level of molecules with both pro-inflammatory and anti-inflammatory effects. This increase is influenced by the patient's clinical condition, but also by the nature of the operation itself, which uses conventional extracorporeal circulation. This technique leads to damage to blood elements by direct contact with air and parts of the extracorporeal circulation, as well as to ischemia-reperfusion injury. The specifics of cardiac surgery then affect possible postoperative complications such as multiorgan failure or septic shock. The diploma thesis is divided into a theoretical and a practical part. The theoretical part describes the principle and influence of cardiopulmonary bypass on the human body and the complications that are associated with its use. Furthermore, IFN-γ is described herein as a potential marker of septic conditions that could reflect the clinical postoperative condition of patients. The practical part of the diploma thesis monitored the percentage change of selected cell populations and the production of IFN-γ in the peripheral blood of patients before and after cardiac surgery.
