Étude du transport de charges dans le niobate de lithium massif et réalisation de fonctions électro-optiques dans le niobate de lithium périodiquement polarisé / Study of charge transport in bulk lithium niobate and realization of electro-optical functions in periodically poled lithium niobate

Le premier volet de cette thèse est consacré à la modélisation des phénomènes de transport dans le LN. Partant d'une analyse critique des modèles de bande usuels, nous montrons leur inadéquation dans le cas du LN et nous proposons un modèle de saut basé sur la théorie des petits polarons. Nous étudions d'abord par simulation Monte-Carlo la décroissance d'une population de polarons liés NbLi4+ relaxant vers des pièges profonds FeLi3+. Nous montrons que les pièges FeLi3+ ont des rayons effectifs particulièrement grands, rayons qui augmentent encore à température décroissante, et limitent considérablement les longueurs de diffusion des polarons. Les résultats de simulations sont ensuite confrontés aux résultats expérimentaux obtenus par différentes techniques ; Absorption photo-induite, Raman, Enregistrement holographique et Pompe-sonde. Le deuxième volet de cette thèse est consacré aux applications électro-optiques dans le LN périodiquement polarisé (PPLN). Sous l’effet d’une tension électrique, l’indice de réfraction du PPLN est périodiquement diminué et augmenté, formant ainsi un réseau d’indice activable électriquement. Un premier composant utilisant l’effet électro-optique dans du PPLN a été développé et démontré expérimentalement. Dans ce composant, la lumière est défléchie sous l’effet de la tension électrique par le réseau d’indice. Ce déflecteur de Bragg atteint une efficacité de diffraction proche de 100% avec une faible tension de commande de l’ordre de 5 V. Un deuxième composant a également été proposé, où la lumière se propage perpendiculairement aux parois de domaines du PPLN. Dans cette configuration un réflecteur de Bragg électro-optique peut être réalisé / The first part of this thesis is devoted to the modeling of transport phenomena in the LN. From a critical analysis of the usual band models, we show their inadequacy in the case of LN and we propose a hopping model based on the theory of small polarons. We first study by Monte-Carlo simulation the population decay of bound polarons NbLi4+ in deep traps FeLi3+. We show that the traps (FeLi3+) have particularly large effective radii, which increase further at decreasing temperature, and considerably limit the diffusion lengths of the polarons. The results of simulations are then compared with experimental results obtained by different techniques; Light-induced absorption, Raman, Holographic storage and Pump-Probe. The second part of this thesis is devoted to electro-optical applications in the periodically poled LN (PPLN). Under the effect of an electrical voltage, the refractive index of the PPLN is periodically decreased and increased, thus forming an electrically activatable index grating. A first component using the electro-optical effect in PPLN has been developed and demonstrated experimentally. In this component, the light is deflected under the effect of the electrical voltage by the index grating. This Bragg deflector achieves a diffraction efficiency of close to 100% with a low drive voltage of the order of 5 V. A second component has also been proposed, where light propagates perpendicularly to the domain walls of the PPLN. In this configuration an electro-optic Bragg reflector can be realized

Insights into the Chemistry of Iron Complexes as Imaging and Photocytotoxic Agents

The current thesis addresses the various facets of the chemistry of photocytotoxic iron complexes including their syntheses, characterization, evaluation of the anti-proliferative activities in various cancer cell lines upon photo-exposure, mechanism of cell death, the cellular uptake, localization inside cells, the interaction with double stranded DNA and their ability to induce DNA photocleavage. Chapter I presents a general introduction to cancer and the anticancer agents. It covers various procedures available for cancer treatment and different aspects of chemotherapy are discussed in details. The mechanism of action of several chemotherapeutic agents, the DNA cleavage pathways and the anticancer activity of bleomycins are delineated. Photo-chemotherapy or photodynamic therapy which has emerged as an alternative treatment modality is described. It also contains a brief description of ideal photosensitizers and the ones that are currently approved. The potential of transition metal complexes as photo-chemotherapeutic agents is discussed based on the recent literature reports on the prospective photocytotoxic metal complexes, the photo-release of cytotoxic molecules from metal complexes, the DNA cleavage activities and their cytotoxicities. The biochemistry of iron and its medical utility which prompted the development of iron based cytotoxins has been presented. The objective of the present investigation is also defined in this chapter. Chapter II describes the syntheses, characterization, evaluation of visible light induced cytotoxicity and interaction with DNA of a series of iron(II) bis-terpyridine complexes. Some interesting redox behaviour observed for two of the complexes has been described in details and rationalized from theoretical calculations. The DNA binding affinities of the complexes and their ability to induce DNA photocleavage in green light are discussed. The importance of this work lies in the remarkable photocytotoxic behaviour of the iron(II) complexes with visible light which was not reported earlier. Chapter III addresses the syntheses of a series of iron(III) catecholate complexes which upon irradiation with red light can initiate photoreactions to generate cytotoxic species and induce death in HeLa, HaCaT, MCF-7 and A549 cells. The mechanisms of cell death, effect of the complexes on the cell cycle under various conditions, the uptake inside cells and the cellular localization of the complexes are studied. The DNA binding affinities of the five complexes and their ability to induce DNA photocleavage in red light are also presented here. These are the first iron based complexes to show red light induced photocytotoxicity. Chapter IV addresses the drawbacks associated with the aforementioned iron(III) catecholates and their modification with a mitochondria targeting triphenylphosphonium unit. The synthesis, characterization, photocytotoxicities in HeLa, HaCaT, MCF-7 and A549, cell death mechanisms and cellular uptake and localization of four iron(III) complexes are discussed. Chapter V describes the syntheses, characterization and the biological activities of carbohydrate appended iron(III) complexes and their non-glucose analogues. The selective and faster internalization of the glyco-conjugated complexes in HeLa cells has been studied using various spectroscopic and microscopic techniques. The red light induced cytotoxicities of the complexes, their effect on the progression of the cell cycle with and without irradiation and the mechanisms of cell death are explored. DNA binding abilities and photocleavage of DNA are also discussed. Chapter VI presents the syntheses, characterization of a series of iron(III) complexes of a pyridoxal derivative and their salicyldehyde analogues for exploring their differential photocytotoxicity and cellular uptake in cancer cells compared to normal cells. The visible light induced cytotoxicities of the complexes in HeLa, HaCaT, MCF-7 A549 cells and HPL1D cells, their effect on the progression of the cell cycle in dark and light, the mechanisms of cell death and the localization of the complexes inside the cells are explored. The references have been compiled at the end of each chapter and given as superscripts in the text. The complexes presented in this thesis are indicated by bold-faced numbers. Crystallography data of the complexes that are structurally characterized by single crystal X-ray crystallography are given in CIF format in the enclosed CD (Appendix-I). Due acknowledgements have been made wherever the work described is based on the findings of other investigators. Any unintentional omission that might have happened due to oversight is regretted. Studies on Near-IR Light Photocytotoxic Oxovanadium Complexes

The present thesis deals with different aspects of the chemistry of oxovanadium(IV) complexes, their interaction with double stranded DNA, photo-induced DNA cleavage, photo-enhanced cytotoxicity in visible light and red light and localisation and cellular uptake to understand the mechanism of cell death. Chapter I presents a general introduction on potential of transition metal complexes as photochemotherapeutic agents. A brief introduction about Photodynamic Therapy (PDT) as a new alternative to chemotherapy for treating cancer has been made. Various modes of interaction of small molecules with duplex DNA are described. Recent reports on metal-based photocytotoxicity, photo-induced DNA cleavage activity and cellular localization are presented in detail. Objective of the present investigation is also dealt in this Chapter. Chapter II of the thesis deals with the synthesis, characterization, DNA binding and photo-induced DNA cleavage activity of ternary oxovanadium(IV) complexes of ONO-donor 2-(2-hydroxybenzylideneamino)phenol (salamp) and phenanthroline bases to explore the photo-induced DNA cleavage activity in UV-A light of 365 nm and photocytotoxicity in visible light. Chapter III deals with the photo-induced DNA cleavage and photocytotoxicity of ternary oxovanadium(IV) complexes containing ONN-donor N-2-pyridylmethylidine-2-hydroxyphenylamine (Hpyamp) Schiff bases and phenanthroline bases. The objective of this work is to investigate the photo-induced DNA cleavage activity in near-IR light. Photocytotoxicity and cell cycle arrest have been studied in HeLa cancer cells. Chapter IV deals serendipitous discovery of planar triazinuim cationic species by vanadyl-assisted novel ring cyclization reaction. The compounds are synthesised, characterized and their DNA binding and anaerobic photoinduced DNA cleavage activity are presented. The importance of the thiazole moiety in the triazinuim species in cellular uptake has been investigated. Photocytotoxicity, localization and cell death mechanism have been studied in HeLa and MCF-7 cells. Chapter V describes the synthesis, characterization, DNA binding, photo-induced DNA cleavage activity and photocytotoxicity of oxovanadium(IV) complexes containing 2-(1H-benzimidazol-2-yl)-N-(pyridin-2-ylmethylene)ethaneamine (Hpy-aebmz) and curcumin as photosensitizer. The effect of conjugating naphthalimide on Hpy-aebmz on photoinduced DNA cleavage and photocytotoxicity has been studied. Cellular uptake, localization and mechanism of cell death induced by complexes have been investigated. Chapter VI presents ternary oxovanadium(IV) complexes having, 2-((1H-benzimidazol-2-yl)methylimino-methyl)phenol (Hsal-ambmz) and phenanthroline bases. The complexes were synthesized, characterized and their DNA binding property studied. Photo-induced DNA cleavage activity and photocytotoxicity in red light has been discussed. Anthracene has been conjugated to a tridentate ligand to investigate cellular uptake, localization and cell death mechanism. Mitochondria targeting property of the complexes having dipeptide has been studied and compared with clinically used drug Photofrin®. The references have been compiled at the end of each chapter and indicated as superscript numbers in the text. The complexes presented in this thesis are represented by bold-faced numbers. Crystallographic data of the complexes, characterized structurally by single crystal X-ray crystallography, are given in CIF format in the enclosed CD (Appendix-I). Longitudinal assessment of caries lesions, especially under the opaque preventive sealant, would be of utmost importance to the dental community. OBJECTIVES: The aim of this two-part in-vitro study is to evaluate the performance of multiple detection methods: The International Caries Detection and Assessment System (ICDAS); two quantitative light-induced fluorescence systems QLF; Inspektor™ Pro and QLF-D Biluminator™2 (Inspektor Research Systems B.V.; Amsterdam, The Netherlands); and photothermal radiometry and modulated luminescence (PTR/LUM) of The Canary System® (Quantum Dental Technologies; Toronto, Canada). All these are to be evaluated on their detection of caries on posterior human permanent teeth for 1) of primary occlusal lesions, and 2) under the sealant of primary occlusal lesions. METHODS: One hundred and twenty (N = 120) human posterior permanent teeth, selected in compliance with IU-IRB “Institutional Review Board” standards, with non-cavitated occlusal lesions ICDAS (scores 0 to 4) were divided into two equal groups. The second group (N = 60) received an opaque resin dental sealant (Delton® Light-Curing Pit and Fissure Sealant Opaque, Dentsply, York, PA). All lesions were assessed with each detection method twice in a random order except for ICDAS, which was not used following the placement of the sealant. Histological validation was used to compare methods in regard to sensitivity, specificity, % correct, and the area under receiver- operating characteristic curve (AUC). Intra-examiner repeatability and inter-examiner agreement were measured using intraclass correlation coefficient (ICC). RESULTS: 1) Of primary occlusal lesions, sensitivity, specificity, and AUC values were respectively: 0.82, 0.86 and 0.87 (ICDAS); 0.89, 0.60 and 0.90 (Inspektor Pro); 0.96, 0.57 and 0.94 (QLF-D Biluminator 2); and 0.85, 0.43 and 0.79 (The Canary System). Intra-examiner repeatability and inter-examiner agreement were respectively: 0.81 to 0.87: 0.72 (ICDAS); 0.49 to 0.97: 0.73 (Inspektor Pro); 0.96 to 0.99: 0.96 (QLF-D Biluminator 2); and 0.33 to 0.63: 0.48 (The Canary System). 2) Of primary occlusal lesions under the opaque dental sealants, sensitivity, specificity, and AUC values were respectively: 0.99, 0.03 and 0.67 (Inspektor Pro); 1.00, 0.00 and 0.70 (QLF-D Biluminator 2); and 0.54, 0.50 and 0.58 (The Canary System). Intra-examiner repeatability and inter-examiner agreement were respectively: 0.24 to 0.37: 0.29 (Inspektor Pro); 0.80 to 0.84: 0.74 (QLF-D Biluminator 2); and 0.22 to 0.47: 0.01 (The Canary System). CONCLUSION: Limited to these in-vitro conditions, 1) ICDAS remains the method of choice for detection of early caries lesion due to its adequately high accuracy and repeatability. QLF systems demonstrate potential in longitudinal monitoring due to an almost perfect repeatability of QLF-D Biluminator 2. The Canary System performance and repeatability were not acceptable as a valid method of early caries detection. 2) None of the methods demonstrated acceptable ability in detecting of occlusal caries under the opaque sealant. However, QLF-D Biluminator 2, with limitation to these in-vitro conditions and Delton opaque sealant, demonstrated a fair accuracy AUC (0.70) in detecting of caries under sealants at an experimental threshold of 12.5% ΔF.

In der vorliegenden Arbeit wird die neuartige Mikroskopiemethode der Ptychographie für die Untersuchung photorefraktiver Materialien genutzt. Photorefraktive Materialien zeichnen sich durch die Generation lichtinduzierter Brechungsindexänderungen aus. Die Ptychographie bietet die Möglichkeit, neben der generierten Brechungsindexänderung im photorefraktiven Material auch die für die Generation genutzte Intensitätsverteilung des Laserstrahls zu bestimmen. Es wird sowohl die Abhängigkeit der Brechungsindexänderung von der Zeit der Generation als auch die Abhängigkeit von der Polarisation des Lasers gemessen. Durch den Vergleich der gewonnenen Werte mit einer numerischen Simulation des photorefraktiven Effekts werden mikroskopische Parameter der lichtinduzierten Ladungswanderung ermittelt. Zudem wird aus der polarisationsabhängigen ptychographischen Messung das Raumladungsfeld und die korrespondierende Ladungsdichte im Material berechnet. Die Ptychographie liefert damit einen neuen Zugang zum quantitativen Verständnis der Photorefraktivität. / In the present thesis the novel microscopy technique of ptychography is applied to the investigation of photorefractive materials. Photorefractive materials exhibit a change of the refractive index due to the exposure to light. The method of ptychography determines the refractive index change of the material together with the intensity distribution of the laser beam that was used for its generation. In one part of the experiment the time dependence of the refractive index change versus the generation time is investigated, in the other part of the experiment the dependence of the refractive index change to the polarisation of the laser beam is examined. Microscopic parameters of the photorefractive charge migration are determined with the utilisation of a numerical simulation of the photorefractive effect and its comparison with the measurement. Finally, the whole space charge field with the corresponding space charge density is calculated from a set of ptychographic measurements of one refractive index change with different polarisation directions of the laser. The presented experiments and their evaluation show, that the method of ptychography opens a new possibility for a quantitative understanding of the photorefractive effect.
