QUALIDADE DE SOBRECOXAS DE FRANGOS SUBMETIDAS À RADIAÇÃO UV-C / CHICKEN DRUMSTICKS QUALITY SUBJECTED TO UV-C RADIATION

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / During the chicken slaughter line, some steps are considered critical, due to the possibility of cross-contamination among carcasses, which directly influence in the shelf-life of the carcasses. New technologies that reduce the microbial counts, leading to an increase of the quality and shelf-life are very welcome. The ultraviolet-C (UV-C) radiation appears as an alternative, because is a non-thermal method, of low cost and of easy implementation, which has already proved to be lethal to many microorganisms in food surfaces. Thus, this work aimed to evaluate the use of doses of UV-C radiation of 5,4 and 9,46 kJ/m2, applied at two temperatures near the slaughter steps (5 and 25 °C), on the physicochemical parameters (color, pH, conjugated dienes content, peroxide value, thiobarbituric acid reactive substances and fatty acid profile) and microbiological (total aerobic mesophilic bacteria, psychrotrophic, yeasts and molds) of chicken drumsticks. The analyses were performed immediately after the doses were applied (zero day) and every 3 days, during 12 days of storage at 5 °C, but the fatty acid profile was analyzed after the doses were applied, and on sixth and twelfth days. The number of colonies of total aerobic mesophilic bacteria had significant reductions (p < 0,05) on the third, sixth and ninth days of storage, at both temperatures of application and after the doses were applied (zero day) for the application at 25 °C. Regarding the total aerobic mesophilic bacteria, application of UV-C radiation at 5 °C, provided shelf-life of carcasses of 9 days of storage, whereas at 25 °C, the shelf-life was 6 days. The number of colonies of psychrotrophic bacteria had significant decreases (p < 0,05) in all days of storage when applied the dose of 9,46 kJ/m2 at 5 °C, and only on the third and on the twelfth days when applied at 25 °C. At the sixth day, the number of colonies of psychrotrophic bacteria was characteristic of deterioration, regardless the treatment applied. The molds and yeasts were not affected by UV-C radiation. Generally, in a same temperature of application, the UV-C radiation doses didn t cause significant changes (p > 0,05) compared to the samples from the control group (without irradiation) in any of the physicochemical parameters, independent of the day of storage. The application of UV-C radiation at 25 °C compared to 5 °C, promoted higher formation of conjugated dienes and lower amount of omega-6 and omega-3 polyunsaturated fatty acids. Thus, the UV-C radiation could be applied in industry after the step of cooling carcass (at 5 °C) due to the non-acceleration of initiation of lipid oxidation, the non-diminishing of essential fatty acids and being effective on the reduction of aerobic mesophilic and psychrotrophic bacteria. / Durante a linha de abate de frangos, algumas etapas são consideradas críticas, devido à possibilidade de contaminação cruzada entre as carcaças, o que influencia diretamente na vida útil das mesmas. Novas tecnologias que venham a reduzir as contagens microbianas, levando ao aumento da qualidade e vida útil das carcaças são muito bem-vindas. A radiação ultravioleta-C (UV-C) aparece como uma alternativa, pois é um método não térmico, de baixo custo e de fácil aplicação, que já se mostrou letal para vários micro-organismos em superfícies de alimentos. Desta forma, o presente trabalho teve como objetivo avaliar o uso de doses de radiação UV-C de 5,4 e 9,46 kJ/m2, aplicadas em duas temperaturas próximas as de etapas do abate (5 e 25 °C), sobre os parâmetros físico-químicos (cor, pH, teor de dienos conjugados, índice de peróxidos, substâncias reativas ao ácido tiobarbitúrico e perfil de ácidos graxos) e microbiológicos (bactérias mesófilas aeróbias totais, psicrotróficas e bolores e leveduras) em sobrecoxas de frangos. As análises foram realizadas logo depois de aplicadas as doses (zero dia) e de 3 em 3 dias, durante 12 dias de armazenamento a 5 °C, sendo que o perfil de ácidos graxos foi analisado logo depois de aplicadas as doses e no 6º e 12º dias. O número de colônias das bactérias mesófilas aeróbias totais teve reduções significativas (p < 0,05) no 3º, 6º e 9º dias de armazenamento, em ambas as temperaturas de aplicação e logo depois de aplicadas as doses (zero dia) para a aplicação a 25 °C. Em relação às bactérias mesófilas aeróbias totais, a aplicação da radiação UV-C a 5 °C proporcionou tempo de vida útil das carcaças de 9 dias de armazenamento, enquanto que na aplicação a 25 °C, o tempo de vida útil foi de 6 dias. O número de colônias de bactérias psicrotróficas teve reduções significativas (p < 0,05) em todos os dias de armazenamento quando aplicada a dose de 9,46 kJ/m2 a 5 °C, e somente no 3° e 12° dias, quando aplicada a 25 °C. A partir do 6° dia de armazenamento, o número de colônias de bactérias psicrotróficas foi característico de deterioração, independente do tratamento aplicado. Os bolores e leveduras não foram afetados pela radiação UV-C. De maneira geral, em uma mesma temperatura de aplicação, as doses de radiação UV-C não causaram modificações significativas (p > 0,05) em relação às amostras pertencentes ao grupo controle (sem irradiação) em nenhum dos parâmetros físico-químicos, independente do dia de armazenamento. A aplicação da radiação UV-C a 25 °C em relação a 5 °C, promoveu maior formação de dienos conjugados e um menor teor de ácidos graxos poli-insaturados ômega-6 e ômega-3. Dessa forma, a radiação UV-C poderia ser aplicada na indústria após a etapa de resfriamento das carcaças (a 5 °C), devido a não acelerar o início da oxidação lipídica, não diminuir os teores dos ácidos graxos essenciais, e sim apresentar eficiência na redução de bactérias mesófilas aeróbias e psicrotróficas.

Chemical changes in Brazil nuts and co-products: characterisation and strategies of control and monitoring / Alterações químicas em castanha do Brasil e coprodutos: caracterização e estratégias de controle e monitoramento

Brazil nuts (Bertholletia excelsa, H.B.K.) are seeds of high nutritional value collected from South American rainforests and its productive chain is one of the most important non-timber economic activities in Brazilian amazon. The main objectives of this research were: 1) characterize the occurrence of chemical changes in Brazil nut kernels (BNK), cold-pressed Brazil nut oil (BNO) and Brazil nut flour obtained by water extraction (BNF); and 2) investigate strategies of control and monitoring these changes during storage. For this, consolidated techniques, such as spectrophotometry and chromatography, and a relatively new analytical technique, the electron spin resonance (ESR) spectroscopy, were employed. As major results, it was found that different combinations of storage temperatures and atmosphere packages have differently affected the tendency of radical formation and off-flavor volatile aroma compounds generation in BNK, and that the combination of refrigeration with vacuum packing was able to keep BNK at their best. It was demonstrated that a spin-trapping ESR spectroscopy method would be suitable to monitor oxidative changes in BNO with known history stored either in clear or in brown glass bottles under retail conditions. For BNF, it was demonstrated that minor variations on water activity (aw) might significantly affect the rates of both lipid oxidation and nonenzymatic browning reactions during storage. There was indication that for BNF with initial aw of 0.196, but not for BNF with initial aw of 0.101, under the studied conditions, secondary products from lipid oxidation might be substrates for nonenzymatic browning products formation. As a conclusion, these results may help to better understand chemical deteriorative processes in BNK and its co-products, according to the storage conditions, and that the use of less sample-demanding, fast and solvent-free analytical method to monitor these changes in BNO is feasible. / A castanha do Brasil (Bertholletia excelsa, H.B.K.) é uma semente de boa qualidade nutricional coletada em florestas tropicais da América do Sul, cuja cadeia produtiva é uma das mais importantes atividades econômicas não madeireiras da Amazônia brasileira. Os principais objetivos desta pesquisa foram: 1) caracterizar a ocorrência de alterações químicas em castanhas do Brasil (CB), óleo de castanha do Brasil obtido por prensagem a frio (OCB) e farinha de castanha do Brasil obtida por extração aquosa (FCB); e 2) investigar estratégias para controlar e monitorar essas alterações ao longo do armazenamento. Para isso, técnicas consolidadas como a espectrofotometria e a cromatografia, e uma técnica relativamente recente, a espectroscopia de ressonância de spin eletrônico (RSE), foram empregadas. Dentre os principais resultados obtidos, foi possível constatar o efeito de diferentes combinações de temperaturas e atmosferas de embalagem sobre a tendência de formação de radicais e sobre a geração de compostos voláteis de aroma relacionados a odor indesejável em CB, e que a temperatura de refrigeração combinada com a embalagem a vácuo foi a mais eficiente na preservação da qualidade da CB. Demonstrou-se que o uso de um método de aprisionamento de spins de RSE pode ser eficiente para monitorar alterações químicas em OCB com histórico conhecido embalado em frascos de vidro transparente ou marrons sob condições de armazenamento comercial. Para FCB, foi demonstrado que pequenas variações na atividade de água (aw) podem afetar significativamente as taxas de oxidação lipídica e de reações de escurecimento não enzimático durante armazenamento. Obteve-se indicação de que para FCB com aw inicial de 0,196, mas não para FCB com aw inicial de 0,101, produtos secundários da oxidação lipídica podem ser substratos para a formação de produtos do escurecimento não enzimático. Como conclusão geral, os resultados obtidos podem ajudar a explicar melhor os processos de deterioração química em CB e seus coprodutos, conforme as condições de armazenamento, e que o uso de um método que requer menor quantidade de amostras, é rápido e não usa solventes é viável para o monitoramento da qualidade de OCB.

ESTABILIDADE LIPÍDICA DE FILÉS DE JUNDIÁ (Rhamdia quelen) / LIPID STABILITY OF SILVER CATFISH FILLETS (Rhamdia quelen)

This work was aimed at studying lipid stability of silver catfish (Rhamdia quelen) fillets. The influence of the inclusion (5%) of soybean or rice oil in silver catfish diet and vacuum packaging on lipid and color stability of raw silver catfish fillets was evaluated during frozen storage (18 months). Besides, the influence of seven cooking methods (boiling, conventional baking, microwave baking, grilling, deep frying in soybean oil, canola oil, or partially hydrogenated vegetable oil) on the oxidation, proximate, and fatty acid composition of fillets was also evaluated. The different diets had no effect on the proximate composition of the fish fillets. The content of thiobarbituric acid reactive substances (TBARS, lipid oxidation index) of fillets from both diets increased after 12 months of storage with or without vacuum, but this increase was higher in fish fed with soybean oil. Color parameters were not affected by the vacuum storage or diet, but most parameters (a*, b*, chroma, and Hue values) were affected by the time of storage. Hue value showed that raw fillets tended to yellowness and after 18 months of frozen storage became greener. The content of various fatty acids in fillets was influenced by the type of vegetable oil used in the diet, but n-3/n-6 polyunsaturated fatty acids ratio and unsaturated/saturated ratio did not differ between diets. All cooking methods evaluated reduced moisture and increased protein content. Fat content was higher in the fried fillets. The free fatty acid content of fillets was significantly reduced by the different cooking methods, while conjugated dienes and peroxide values decreased for all fried samples, but remained constant in the samples submitted to the other cooking methods. Boiling and baking increased TBARS, while grilling and frying did not change TBARS. Boiling, baking, and grilling did not affect the silver catfish fillets fatty acid composition. Frying in canola oil increased n-3/n-6 ratio, in soybean oil increased general polyunsaturated fatty acid content, and in hydrogenated vegetable oil incorporated trans fatty acids in the fillets. Results indicated that fillets from silver catfish fed diets with soybean or rice oil have different lipid profile and lipid stability during frozen storage. Frying silver catfish fillets in canola oil could increase the low n-3/n-6 polyunsaturated fatty acids ratio of raw silver catfish fillets. / O presente trabalho teve por objetivo estudar a estabilidade lipídica de filés de jundiá (Rhamdia quelen). Foi avaliada a influência da inclusão (5%) de óleo de soja ou arroz na dieta do jundiá, e a embalagem a vácuo, sobre a estabilidade lipídica e a cor de filés crus congelados (18 meses). Avaliou-se também a influência de sete métodos de cocção (cozido em água, assado em forno convencional ou microondas, grelhado, ou frito em óleo de soja, arroz ou gordura vegetal hidrogenada) sobre a oxidação, a composição centesimal e de ácidos graxos de filés de jundiá. Não houve diferença na composição centesimal dos filés entre as dietas. O conteúdo de substâncias reativas ao ácido tiobarbitúrico (TBARS, índice de lipoperoxidação) aumentou nos filés dos peixes de ambas as dietas, após 12 meses de armazenamento congelado, tanto nos filés embalados com ou sem vácuo. Esse aumento foi mais acentuado nos filés dos peixes alimentados com dietas contendo óleo de soja. Os parâmetros de cor não foram afetados pela embalagem a vácuo ou pela dieta, mas a maioria dos parâmetros (a*, b*, croma e ângulo de matiz), foi afetada pelo tempo de estocagem. O valor de ângulo de matiz mostrou que os filés crus tendiam ao amarelo, e após 18 meses de armazenamento congelado tenderam ao verde. O conteúdo de vários ácidos graxos nos filés foi influenciado pelo tipo de óleo vegetal utilizado na dieta, porém a razão dos ácidos graxos poliinsaturados n-3/n-6 e insaturados/saturados não diferiu entre as dietas. Todos os métodos de processamento térmico avaliados reduziram a umidade e aumentaram o conteúdo de proteína dos filés. O teor de gorduras foi maior nos filés fritos. O conteúdo de ácidos graxos livres dos filés foi significativamente reduzido pelos diferentes métodos de processamento térmico, enquanto os teores de dienos conjugados e peróxidos diminuíram em todas as amostras submetidas à fritura, mas permaneceram constantes nas amostras submetidas aos demais métodos de processamento térmico. Nos filés cozidos e assados observou-se aumento dos teores de TBARS, enquanto os grelhados e fritos não sofreram alteração neste parâmetro. A composição de ácidos graxos dos filés de jundiá cozidos, assados e grelhados não foi alterada pelo tratamento térmico. A fritura em óleo de canola aumentou a razão n-3/n-6, enquanto que a fritura em óleo de soja aumentou o conteúdo total de ácidos graxos poliinsaturados, e a fritura em gordura vegetal hidrogenada incorporou ácidos graxos trans aos filés. Os resultados indicam que os filés de jundiá alimentados com dietas contendo óleo de soja ou de arroz tiveram perfil lipídico e estabilidade lipídica diferente durante o armazenamento congelado. A fritura dos filés de jundiá em óleo de canola poderia aumentar a razão de ácidos graxos poliinsaturados n-3/n-6, que é baixa nos filés de jundiá crus.

SUBSTITUIÇÃO DA CARNE MECANICAMENTE SEPARADA POR DIFERENTES CONCENTRAÇÕES DE HIDROLISADO PROTEICO EM MORTADELA / REPLACEMENT OF THE MECHANICALLY DEBONED CHICKEN MEAT FOR DIFFERENT CONCENTRATIONS OF PROTEIN HYDROLYSATE IN COOKED MEAT SAUSAGE.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The objective of this study was to develop a liquid protein hydrolysate from Mechanically Deboned Chicken Meat (MDCM) by enzymatic hydrolysis and to evaluate the addition of different concentrations (10, 20 and 30%) in a reduced-fat mortadella-type sausage. First, it was evaluated the addition of BHT antioxidant (0.01%) and curing salts (0.25%) in the production of protein hydrolysate at different hydrolysis times (5, 15, 30 and 60 minutes) in order to reduce lipid oxidation and brownish color formation. There were evaluated the degree of hydrolysis, pH, proximate composition, lipid oxidation and instrumental color. It was observed that the addition of BHT antioxidant and curing salts did not affect the degree of hydrolysis at 60 minutes. The pH was similar to the MDCM and the proximate composition was not affected by the hydrolysis process or by the addition of BHT antioxidant and curing salts. However, there was a synergistic action between the BHT antioxidant and the curing salts in lipid oxidation prevention of the hydrolysates during the whole period of hydrolysis. Also, there was observed a reduction in the formation of a brownish color mainly by the use of curing salts, evidenced by high levels of values of a* (redness). Subsequently, there was evaluated the addition of different concentrations protein hydrolysate containing BHT antioxidant and curing salts in a reduced-fat mortadella-type sausage. Thus, four treatments were made containing 0, 10, 20 and 30% of MDCM protein hydrolysate. There were evaluated the proximate composition, pH, lipid oxidation, colorimetric parameters, texture profile and microbiological and sensory characteristics during 60 days of storage at 4 ºC. The proximate composition, pH and microbiological characteristics were considered normal for this kind of product. The values of lipid oxidation of products containing MDCM protein hydrolysate increased up to 30th day of storage with subsequent decrease until the end of the storage period (60 days). The products containing MDCM protein hydrolysate had low values of L* (lightness) and a* (redness), higher lipid oxidation and soft textured, evidenced both by the panelists and by instrumental texture profile. The addition of up to 10% protein hydrolysate proved to be viable in the production of reduced-fat mortadella-type sausage, showing quality characteristics closer to the control treatment. / O objetivo deste trabalho foi desenvolver um hidrolisado proteico líquido de carne mecanicamente separada (CMS) de frango, através de hidrólise enzimática e avaliar a adição de diferentes concentrações (10, 20 e 30%) em mortadela com teor de gordura reduzido. Primeiramente, foi avaliada a adição de antioxidante BHT (0,01%) e sal de cura (0,25%) na produção do hidrolisado proteico em diferentes tempos de hidrólise (5, 15, 30 e 60 minutos) com o objetivo de reduzir a oxidação lipídica e a formação da coloração amarronzada. Avaliou-se o grau de hidrólise, pH, composição centesimal, oxidação lipídica e cor instrumental. Foi possível observar que a adição de antioxidante BHT e sal de cura não afetou o grau de hidrólise ao final de 60 minutos. O pH foi similar ao encontrado na CMS original e a composição centesimal não foi afetada pelo processo de hidrólise nem pela adição de antioxidante BHT e sal de cura. Entretanto, houve uma ação sinergística entre o antioxidante BHT e o sal de cura na prevenção da oxidação lipídica dos hidrolisados durante todo o período de hidrólise. Também, foi observada uma redução na formação da coloração amarronzada pelo uso principalmente do sal de cura, evidenciada pelos maiores teores de vermelho (a*). Posteriormente, foi avaliada a adição do hidrolisado proteico com antioxidante BHT e sais de cura, em mortadela com teor de gordura reduzido, contendo 0, 10, 20 e 30% de hidrolisado proteico de CMS. Foram avaliadas a composição centesimal, pH, oxidação lipídica, parâmetros de cor e as características microbiológicas, sensoriais e perfil de textura durante 60 dias em armazenamento a 4 ºC. A composição centesimal, pH e as características microbiológicas foram consideradas normais para o produto em questão. Os valores de oxidação lipídica dos produtos contendo hidrolisado proteico de CMS aumentaram até o 30º dia de armazenamento com posterior decréscimo até o final do período de armazenamento (60 dias). Os produtos contendo hidrolisado proteico de CMS apresentaram valores de luminosidade (L*) e valores de a* menores e textura mais amolecida, evidenciada tanto pelos provadores quanto pelo perfil de textura instrumental. A adição de até 10% do hidrolisado proteico se mostrou viável na fabricação de mortadela com teor de gordura reduzido, pois apresentou características de qualidade mais próximas ao tratamento controle.

Influência do processamento, de antioxidantes e da estocagem sobre a estabilidade oxidativa lipídica do ovo / Influence of processing, antioxidants and storage on oxidative stability of egg lipids

O ovo é consumido in natura ou processado, e usado como matéria-prima em diversos produtos. A pasteurização e a atomização são os principais processamentos aplicados ao ovo. Diferentes condições de processamento, embalagem e estocagem podem afetar a estabilidade oxidativa lipídica e reduzir a proteção antioxidante natural do ovo. O uso de embalagens onde não haja contato do ovo com a luz e oxigênio, estocagem em temperaturas baixas e uso de antioxidantes podem prevenir a oxidação lipídica. Atualmente, os antioxidantes sintéticos são cada vez mais substituídos por antioxidantes naturais ou feita uma associação entre eles. O alecrim (Rosmarinus officinalis L.) e o chá verde (Camelia sinensis L) constam entre os vegetais com grande potencial antioxidante. Com base no exposto, os objetivos do trabalho foram: a) Padronizar o método do fosfomolibdênio para avaliação da capacidade antioxidante total da fração lipídica (CATL) do ovo; b) Investigar o efeito da pasteurização e da atomização do ovo no que se refere à capacidade antioxidante e à estabilidade oxidativa da fração lipídica do ovo; c) Avaliar a estabilidade oxidativa de ácidos graxos e a capacidade antioxidante do ovo integral pasteurizado atomizado, estocado a 5ºC, por 90 dias; d) Verificar por meio de teste de concentração de antioxidantes, a estabilidade oxidativa lipídica do ovo integral pasteurizado atomizado adicionado da mistura de extrato de alecrim, extrato de chá verde e BHA (butilato de hidroxianisol); e) Otimizar a concentração da mistura de extrato de alecrim, extrato de chá verde e BHA a ser adicionada ao ovo líquido integral pasteurizado, posteriormente atomizado, pelo modelo matemático proposto pela metodologia de superfície de resposta; f) Investigar o efeito da mistura otimizada de extrato de alecrim, extrato de chá verde e BHA na estabilidade oxidativa lipídica do ovo integral pasteurizado atomizado, estocado sob as temperaturas de 5°C e 25ºC, por 90 dias. O método do fosfomolidênio para medir a CATL do ovo apresentou adequação analítica, indicada pela equação de regressão (y = 13,705x + 0,0808), coeficiente de determinação (R2 = 0,9931), limite de detecção (0,005mg &#945;-tocoferol/ mL), limite de quantificação (0,017mg &#945;-tocoferol/ mL), coeficiente de correlação (r = 0,9965), sendo que a precisão não indicou dispersão ao redor da média. A CATL diminuiu com o progresso do processamento e o inverso foi observado quanto aos lipídios, 7-CETO (7-cetocolesterol) e TBARS (substâncias reativas ao ácido tiobarbitúrico). O ovo integral pasteurizado atomizado (OIPA) mantido sob condições de estocagem consideradas ideais permaneceu estável em relação à hidratação, a CATL e as TBARS. Pelo teste de concentração de antioxidantes, foram ensaiadas dez misturas (alecrim, chá verde e BHA) no OIPA e refletiu na sua estabilidade oxidativa lipídica, verificada pelas TBARS, CATL, CT-F (capacidade redutora pelo reativo de Folin - Ciocalteau), AGL (ácidos graxos livres) e AS-233 (substâncias absorvidas a 233 nm). Os resultados mostraram que a metodologia de superfície de resposta (RSM) foi adequada para descrever a formação dos AGL e AS-233 no OIPA, podendo serem usadas para fins preditivos. A otimização da mistura de antioxidantes baseou-se no modelo matemático obtido com as AS-233, onde foi proposto como sistema antioxidante 150ppm de BHA, 600ppm de alecrim e 300ppm de chá verde. As condições de estocagem adotadas e a adição dos antioxidantes foram efetivas para manter estável o OIPA, sendo mais efetivo quando estocado a 5°C. Concluíu-se que o método do fosfomolibdênio apresentou adequação analítica. A pasteurização não afetou os parâmetros analisados (lipídios, TBARS, CATL e 7-CETO), mas a atomização provocou diminuição significativa da CATL, e elevação dos lipídios, TBARS e 7-CETO. Foi mantida a hidratação e a estabilidade oxidativa do OIPA estocado por 90 dias a 5°C, indicando que as condições de embalagem e estocagem foram efetivas. Pelo modelo matemático proposto pela RSM foi constatado que apenas os AGL e AS-233 podem ser usados para fins preditivos, optando-se pela otimização da concentração da mistura de antioxidantes, usando apenas o modelo matemático obtido nas AS-233. Com a presença ou não de antioxidantes, o OIPA estocado a 5°C, ao longo dos 90 dias, apresentou-se mais estável quanto à hidratação e à oxidação lipídica. / Egg is consumed in natura or processed and is used as raw material in various food products. The pasteurization and spray-drying processes are the main processing applied to the egg. Different processing conditions, packaging and storage can affect the lipid oxidative stability and may reduce the natural antioxidant protection of this product. The use of packages in which the egg has no contact with light and oxygen, storage at low temperatures, and the use of antioxidants can prevent lipid oxidation. Currently, synthetic antioxidants have been increasingly replaced by natural antioxidants or an association between synthetic and natural antioxidants is adopted by food companies. Rosemary (Rosmarinus officinalis L.) and green tea (Camellia sinensis L) are among the vegetables that present the highest antioxidant potential. Based on the above-mentioned considerations, the objectives of this research were: a) To standardize the method of phosphomolybdenum to evaluate the total antioxidant capacity of lipid fraction (CATL) egg; b)To investigate the effect of pasteurization and spray-drying on the antioxidant capacity and oxidative stability of the lipid fraction of egg; c) To assess the oxidative stability of fatty acids and the antioxidant capacity of whole egg subjected to pasteurization followed by spray-drying, stored at 5ºC for 90 days, d) To evaluate the by testing the concentration of antioxidants, oxidative stability of the lipid fraction of egg subjected to pasteurization and spray drying, added with a mixture of rosemary and green tea extracts and BHA (butylated hydroxyanisole); e) To optimize the concentration of the mixture composed of rosemary and green tea extracts and BHA to be added to pasteurized liquid egg (and further subjected to spray-drying) by using Response Surface Methodology; f) To investigate the effect of the addition of this optimized mixture of antioxidants on of the lipid fraction of spray-dried egg, stored at temperatures of 5 and 25°C for 90 days. The phosphomolybdenum method CATL of egg presented analytical suitability once it presented a high coefficient of determination (R2 = 0,9931), a regression equation expressed as y = 13.705 + 0.0808 x, a limit of detection of 0.005 mg &#945;- tocopherol/mL, a limit of quantitation of 0.017 mg &#945;-tocopherol/mL, a significant and high correlation coefficient (r = 0.9965), and the accuracy did not indicate dispersion around the mean. The CATL decreased with the progress of processing and the reverse was observed for the lipids, 7-CETO (7-ketocholesterol) and TBARS (thiobarbituric acid reactive substances). The pasteurized spray-dried egg (OIPA), which was kept under ideal storage conditions, remained stable in relation to moisture content, CATL and TBARS. In order to test the concentration of antioxidants to be added to OIPA, a total of 10 mixtures (rosemary, green tea and BHA) were assayed. The addition of antioxidants resulted in a higher oxidative stability of the lipid fraction as measured by TBARS, CATL, CT-F (reduction capacity by using the Folin-Ciocalteau reagent), AGL (free fat acids) and AS-233 (substances that absorb at 233 nm). The results showed that the response surface methodology (RSM) was adequate to describe the development of free fat acids and AS-233 in OIPA, and the RSM model proposed for AS-233 can be used for prediction purposes. The optimization of the antioxidant mixture based on the mathematical model proposed for AS-233 indicated that the mixture of 150ppm of BHA, 600ppm of rosemary extract and 300ppm of green tea extract was the best combination of antioxidants. The adopted storage conditions and the addition of antioxidants to OIPA were effective to maintain the response variables stable, and the stability was higher when OIPA was stored at 5°C. It was concluded that the method of phosphomolybdenum suitability presented analytically. The pasteurization did not affect the analyzed parameters (lipids, TBARS, CATL and 7-CETO), but the spray-drying caused a significant decrease CATL, and an increase in lipids, TBARS and 7-CETO. The hydration and the oxidative stability of OIPA remained stable for 90 days at 5°C, indicating that the packaging and storage conditions were effective. The RSM models proposed for free fat acids and AS-233 could be used for predictive purposes; however, the optimization procedure was performed taken into account only the AS-233 model. The OIPA stored at 5°C for 90 days, with or without the addition of the antioxidant mixture, was more stable in relation to hydration and lipid oxidation.

É muito elevado o potencial de utilização do pescado na forma de minced fish - MF, servindo de matéria-prima para outros produtos. O MF de sardinha foi avaliado quanto ao rendimento, ao efeito da lavagem sobre a melhoria da cor, à caracterização química e quanto à estabilidade química e sensorial durante estocagem sob congelamento de -17 a -18°C. O rendimento do MF de sardinha lavado com água foi de 55% em relação ao peixe inteiro e próximo do rendimento teórico da parte comestível. A lavagem do MF com água ou água + bicarbonato de sódio proporcionou a obtenção de um produto com cor vantajosamente mais clara e apreciável. Com relação à composição química básica o MF de sardinha lavado com água apresentou maior hidratação e menor teor de cinzas em relação ao controle e semelhantes quantidades de proteína e lípides. Comparado ao controle, o MF de sardinha lavado com água, não apresentou diferença quanto ao perfil de ácidos graxos e pH, o que não ocorreu com o nitrogênio das bases voláteis - N-BVT, que diminuiu, e às substâncias reativas ao ácido tiobarbitúrico - TBARS, que aumentaram. O MF de sardinha lavado com água, contendo ou não antioxidantes artificialmente adicionados, apresentou estabilidade química e sensorial quando mantido sob congelamento, com base nos parâmetros pH, N-BVT, TBARS, odor a ranço e cor, em até 26 semanas de estocagem. Os resultados do presente trabalho são favoráveis à possibilidade de obtenção de MF de sardinha lavado com água e seu emprego como matéria-prima para outros produtos pesqueiros. / The potentiality of fish\'s utilization as minced fish - MF is very high, actually turning into a raw material to other products. The sardine\'s MF was evaluated according to its yield, the washing effect improving the color, its chemical characteristic and chemical and sensorial stability, during frozen storage under -17,-18°C. The yield of sardine\'s MF washed in with water, was 55% related to the whole fish and close to the theoretic yield of eatable portion. The MF\'s washing in water or in water plus sodium bicarbonate provide a product with clearer and more desirable color. Related to the chemical basic composition, sardine\'s MF washed in water, presented better hydration and lower ashes contents than its control, and similar quantity of protein and lipids. Compared to the control, the sardine\'s MF washed in water, had not presented any difference related to the fatty acids profile and pH, that had not happened with the nitrogen of volatile bases - N-BVT, which had been decreased, and the thiobarbituric acid reactant substances - TBARS, that had been increased. The sardine\'s MF washed in water, containing or not added antioxidants, was chemical and sensorial stable when maintained frozen, based on parameters as pH, N-BVT, TBARS, rancidity odor, and color, stored for 26 weeks. The results of this work are propitious to the possibility of achieving sardine\'s MF washed in water, and your utilization as a raw material to others seafoods.

Interactive Effects of Nutrition, Environment, and Processing on Fresh Pork Quality, Intestinal Biomarkers of Heat Stress in Swine, and Career Success Factors for Agricultural Students

Heat stress (HS) induced changes in energy metabolism, proteolysis, lipogenesis, and oxidative balance have meat quality ramifications for livestock. However, several knowledge gaps exist in understanding heat stressed finishing pig physiology and pork quality characteristics and how dietary zinc may ameliorate undesirable outcomes. Research was completed to determine zinc supplementation effects on carcass composition, meat quality, and oxidative stability of fresh and processed pork from pigs subjected to a chronic, cyclic heat stress using a 2×2×2 factorial arrangement of treatments with main effects of environment (HS vs. thermoneutral; TN), added zinc level (50 vs. 130 mg kg<sup>-1</sup> available zinc), and zinc source (inorganic vs. organic). Commercial crossbred mixed-sex pigs (initially 72.0 kg) were group-housed under either TN (18.9-16.7°C) or cycling HS conditions with chronic diurnal heat (30-29°C/26-27°C for 12h:12h) on days 24-71 with acute heat waves (32-33°C/29-30°C for 12h:12h) on days 21-24, 42-45, and 63-65. One representative pig (<i>n</i>=80) per pen was slaughtered on day 64. The HS pigs were lighter bodyweight (<i>P</i>=0.039), yielded lighter carcasses (<i>P</i>=0.011), less last rib backfat (<i>P</i>=0.032), tended to have smaller loin eye area (<i>P</i>=0.062) but similar percent lean in belly center slices (<i>P</i>>0.10). Compared to TN, HS carcasses had higher 24-h pH (<i>P</i>=0.001) and decreased drip loss (<i>P</i>=0.034). Shifts in individual fatty acid profile of sausage product derived from HS carcasses were observed but were of insufficient magnitude to affect iodine value. Initially, sausage from HS carcasses tended (day 0, <i>P</i>=0.071) to have less thiobarbituric acid reactive substances than TN but over a 10-day simulated retail display, no treatment induced lipid oxidation differences (<i>P</i>>0.05) were observed in either sausage or displayed loin chops. Consistent treatment differences in CIE L*a*b* of products throughout the 10-day display were not observed. The relationships between physiological changes in pigs receiving supplemental zinc and their body and ambient temperatures were also investigated. A representative gilt (<i>n</i>=96) was selected for thermal monitoring from each pen of the 2×2×2 treatments plus 4 additional treatments representing 2 intermediate levels of Zn in both environments. Core body temperatures (T<sub>core</sub>) during the day 42-45 acute heat wave were continuously recorded via indwelling vaginal thermometers and infrared thermal imaging was used to measure skin temperatures at 12-hour intervals. From a 64-gilt subset of the 2×2×2 treatments, jejunum and ileum samples were collected on day 64 for analysis of villus height, crypt depth, and jejunal gene expression of heat shock proteins (27, 70, 90), occludin, and mucin (MUC2). The HS model induced thermoregulatory changes and increases in T<sub>core</sub> (<i>P</i><0.05). Day 42-45 ambient temperature was negatively correlated with expression of HSP-27 (r=-0.42, <i>P</i>=0.047), HSP-90 (r=-0.49, <i>P</i>=0.014), and occludin (r=-0.69, <i>P</i><0.001) in HS pigs. For the organic Zn supplemented pigs, ambient temperature was positively correlated with expression of HSP-27 (r=0.42, <i>P</i>=0.034) and MUC2 (r=0.45, <i>P</i>=0.017) and negatively correlated with villus height in jejunum (r=-0.42, <i>P</i>=0.027) and ileum (r=-0.38, <i>P</i>=0.048). Thermal Circulation Index (measure of heat dissipation) of HS pigs was negatively correlated with their ileum villus height (r=-0.51, <i>P</i>=0.015) and positively correlated with HSP-70 expression (r=0.46, <i>P</i>=0.041). The T<sub>core</sub> lacked correlation with most variables. This research demonstrates cyclic HS affects carcass composition and quality but does not appear to reduce display shelf-life of pork as indicated by lack of differences in lipid oxidation and color stability. In this HS model, zinc level or source imparted negligible benefits and thermal correlations with gut integrity characteristics existed for organic zinc supplemented and HS pigs. The degree of heat dissipation by heat stressed pigs appeared to be associated with classic HS damage and intestinal responses which may be useful indicators of HS in the grow-finish pig. Another agricultural challenge is maintaining higher education programming which establishes a successful career trajectory for agricultural students amid generational shifts in attitudes and background experiences. Undergraduates studying Animal Science and/or Agricultural Economics were surveyed to understand their perception of how collegiate curricular, co-curricular, and extracurricular experiences (coursework, club participation, relevant work experience, international experience, advising/mentoring, college life, and professional networking) contribute to their anticipated career success. A best-worst scaling experiment was used to force respondents (<i>n</i>=487) to make unbiased tradeoffs between the collegiate experience attributes. Responses were then related back to additional demographical and experience/perception characteristics of respondents. Students indicated relevant work experience was overwhelmingly the most critical of the 7 factors (57% preference share), followed by professional networking (19%), and coursework (14%). Students solely in a pre-veterinary Animal Science curriculum represented a distinct category of students regarding their beliefs and experiences. Further research is needed to investigate possible disconnects between student perceptions and reality in higher education and agricultural careers.

Le fromage fondu est issu de la seconde transformation du lait. Sa fabrication implique le mélange, le chauffage et la texturation de produits laitiers (fromage, beurre et poudres de lait) et non laitiers (agents émulsifiants, acide citrique et chlorure de sodium). Le résultat est un produit homogène, généralement tartinable et à la durée de conservation longue, souvent supérieure à 6 mois. Au cours de la fabrication et du stockage, les réactions d’oxydation des lipides, de caramélisation et de Maillard forment des composés odorants dont certains sont potentiellement indésirables pour la flaveur du produit. Dans le cadre du projet ANR-06-PNRA-023 Réactial, une démarche méthodologique a été mise en place afin : (i) d’identifier les marqueurs réactionnels précurseurs ou responsables des défauts de flaveur observés, (ii) de suivre l’évolution de ces marqueurs au cours des traitements thermiques appliqués, (iii) d’établir un schéma réactionnel observable en vue (iv) de la modélisation et de la prédiction de l’évolution des marqueurs réactionnels et indirectement de l’apparition de défauts de flaveur. Ceci a nécessité la mise au point d’une formulation de matrice de fromage fondu modèle ainsi que l’élaboration d’une cellule de traitement thermique. Différents couplages de chromatographie en phase gazeuse à l’olfactométrie ont été utilisés afin d’identifier les composés odorants. La chromatographie bidimensionnelle systématique couplée à la spectrométrie de masse à temps de vol a permis une semi-quantification des composés traces et ultra-traces tandis que les précurseurs ont été quantifiés par chromatographie liquide haute performance. Les données ainsi obtenues ont permis l’écriture d’un schéma réactionnel observable qui a donné lieu à une modélisation stoechio-cinétique multi-réponses. Un ajustement des données expérimentales par le modèle a pu être réalisé malgré une quantification partielle des différents constituants. / Processed cheese derives from a secondary milk processing step that involves mixing and heating dairy (cheese, butter and milk powders) and non-dairy products (emulsifiers). This processing yields a homogeneous product, usually spreadable, with a shelf-life often longer than 6 months. During processing and storage, lipid oxidation, caramelization and Maillard reactions occur and produce odour-active compounds. Some of them are potentialy involved in the development of odour defects. As part of the ANR-06-PNRA-023 Réactial project, a methodological approach was used in order to (i) identify key compounds responsible for the flavor defects observed, (ii) monitor the evolution of these markers during the heat treatment applied to the cheese matrix, (iii) establish an observable reaction scheme, (iv) model and predict the evolution of these compounds during thermal operations. To do this, a model cheese and its cooking cell were elaborated. Various couplings of gas chromatography with olfactometry were used to identify odorous compounds. Two-dimensional comprehensive chromatography allowed a semi-quantitation of trace and ultra-trace compounds, while precursors were quantitated by high performance liquid chromatography. An observable reaction scheme was extracted from these data and make the multi-response modelling step possible despite a partial quantitation of the volatile compounds.

High pressure processing is a food preservation technique which can be an alternative to heat treatment. Pressurization, in theory, produces changes in organoleptic properties and antioxidant compounds to a lesser extent. The objective of the present dissertation has been to study the effect of high pressure processing on oxidative processes and their relation to the organoleptic qualities of diced swede and packaged sliced ham. The results have pointed to pressurization at 600 MPa, and produce an effective decontamination is that which causes less loss of compounds or antioxidant properties. For swede, this pressure level also caused a minor modification of the organoleptic qualities. This dissertation has also shown that depending on feed composition, the effect of pressurization on the organoleptic qualities can be diametrically opposite. / La aplicación de altas presiones hidrostáticas es una técnica de conservación de alimentos que puede ser una alternativa al tratamiento térmico. La presurización, en teoría, produce una menor modificación de las propiedades organolépticas y de los compuestos antioxidantes. En el presente trabajo se ha estudiado el efecto de las altas presiones hidrostáticas en los procesos oxidativos y su relación con las cualidades organolépticas del nabo troceado y del jamón curado empaquetado loncheado. Los resultados obtenidos han apuntado a que la presurización a 600 MPa, además de producir una descontaminación efectiva, es la que causa una menor pérdida de compuestos o propiedades antioxidantes. En el caso del nabo, esta presión también es la que ocasiona una menor modificación de las cualidades organolépticas. En el presente trabajo también se ha observado que en función de la composición del alimento, el efecto de la presurización en las cualidades organolépticas puede ser diametralmente opuesto.
