Structural study of mRNA translation in kinetoplastids by Cryo-electron microscopy / Etude structurale de la traduction de ARNm chez les kinétoplastides par cryomicroscopie électronique

Les kinétoplastides sont un groupe de protozoaires, et qui menace plus de 400 millions de personnes dans le monde entier. Ils possèdent des segments d'expansion d'ARNr (SE) inhabituellement plus larges dans les sous-unités 40S. Ici, nous avons purifié à partir de lysats de cellules de T. cruzi des complexes d'initiation natifs (48S IC) et des sous-unités de 40S natives que nous avons ensuite analysées par cryo-ME. La structure des 48S IC révèle certains des aspects spécifiques de la traduction aux kinétoplastides, tels qu'un réseau d’interaction complexe entre eIF3 et SEs. En outre, notre structure met en évidence le rôle de DDX60 dans l'initiation de la traduction chez les kinétoplastides. La structure d'une sous-unité 40S native révèle l'existence d'un facteur non caractérisé (appelé ηF). Le site de liaison de ηF suggère un rôle dans le contrôle de la traduction. De plus, nous avons rapporté́ la structure d’une nouvelle protéine ribosomale (-r) spécifique des kinétoplastides (KSRP). Notre travail pose les premières bases structurales des aspects spécifiques de l'initiation de la traduction chez les kinétoplastides. / Kinetoplastid is a group of flagellated protozoans, which threatens more than 400 million people world-wide. They possess unusual large rRNA expansion segments (ES) in the 40S, such as ES6S, ES7S and ES9S and their location suggests an involvement in the initiation process. Furthermore, all mature mRNAs possess a conserved 5’ spliced-leader. Here, we purified from T. cruzi cell lysates native initiation complexes and native 40S subunits that we then analysed by cryo-EM. The structure of native initiation complexes reveals several kinetoplastid-specific aspects of translation, such as an intricate interaction network between eIF3 and ES6S and ES7S. Furthermore, it reveals the role of DDX60 in translation initiation in kinetoplastids. The structure of native 40S subunits reveals the existence of an uncharacterized factor (termed ηF) bound at platform of the 40S. The binding site of ηF suggests a role in translational control. Moreover, we reported a novel kinetoplastid-specific ribosomal (r-) protein (KSRP) bound to the 40S subunit. Our work represents the first structural characterization of kinetoplastids-specific aspects of translation initiation.

Narratives of parents living with a child affected by chronic fatigue syndrome/myalgic encephalomyelitis

Background and Aims: Chronic Fatigue Syndrome/Myalgic Encephalomyelitis (CFS/ME) remains a poorly understood condition, shrouded by debate, stigma, and uncertainty. Unsurprisingly, the little available research suggests that caring for a Child or Young Person (CYP) affected by the condition can be extremely challenging. While the majority of available literature is quantitative in nature, there is some qualitative research examining the impact of having a CYP with CFS/ME on parents. However, there currently appear to be no studies examining the narratives of parents living with a CYP with CFS/ME. Therefore, this research aimed to hear how parents narrate their experiences of living with a CYP affected by CFS/ME, paying attention to how they construct their identity, and the contested condition. Methodology: This research drew on a qualitative approach that explored the narratives of the participants. A purposive sample of five parents of CYP affected by CFS/ME (5 mothers) was recruited for a single semi-structured interview. The interviews were audio-recorded, transcribed, and analysed using a narrative approach to explore what participants said and how they narrated their accounts. This was then situated within the social and cultural contexts that shaped them. Analysis and Findings: Multiple readings of the narratives allowed me to develop a summary of each individual's narrative account. These were presented, after which similarities and differences across narratives were considered. Analysis identified six areas of collective focus: 'stories of onset and diagnosis', 'stories of battle', 'stories of finding the person/people who can help', 'stories of impact', 'stories of seeking social support', and 'stories of coping and adjustment'. Participants' narratives were heavily influenced by dominant societal discourses surrounding CFS/ME and motherhood, and could be seen as a response to these narratives. Consequently, participants offered particular constructions of the condition, themselves, their CYP, and others that they had come into contact with. These findings are discussed with reference to their potential bearing for clinical practice, strengths and limitations of the methodology, and directions for future research.

To do so, first presents its production in the 1970s and their reflections in experimental films and anarchic developed throughout his career, such as Superoutro (1989). In the second part, we examine images and speeches addressed in the film under study which are reflected in the characteristic elements of the counterculture movement that was titled, based on the reflections of Marcos Alexandre Capellari, Armando Ferreira de Almeida, Ken Goffman and Dan Joy, Celso Favaretto, Gonçalo Júnior, Heloisa Buarque de Hollanda, Raymond Leo Matos, among others. New on-line mass spectrometric tools for studying urban organic aerosol sources

Atmospheric aerosols have been shown to have a significant impact on air quality and health in urban environments. Organic aerosols (OA) are one of the main constituents of submicron particulate matter. They are composed of thousands of different chemical species, which makes it challenging to identify and quantify their sources. OA sources have been previously studied; however quantitative knowledge of aerosol composition and their processes in urban environments is still limited. The results presented here investigate OA, their chemical composition and sources as well as their interaction with gases. On-line measurements of species in the particle and the gas phase were performed both from field-based and laboratory studies. Aerosol Mass Spectrometers (AMS) were used together with the Chemical Ionisation Mass Spectrometer (CIMS) and the Filter Inlet for Gases and AEROsols (FIGAERO). Two ambient datasets were analysed to develop methods for source apportionment, using the Multilinear Engine (ME-2), in order to gain new insights into aerosol sources in Manchester and London. Long-term measurements in London allowed the opportunity to perform seasonal analysis of OA sources and look into the relationship of hydrogen-like OA (HOA) and heavy- and light-duty diesel emissions. The seasonal analysis provided information about OA sources that was not possible to observe on the long-term analysis. During Bonfire Night in Manchester, with high aerosol concentrations, particularly biomass burning OA (BBOA), it was possible to identify particulate organic oxides of nitrogen (PON), with further identification of primary and secondary PON and their light absorbing properties. Through laboratory work, new insights into cooking organic aerosols (COA) were gained, a higher relative ion efficiency (RIEOA) value of around 3.3 for OA-AMS compared with the typical RIEOA of 1.4 was determined, which implies COA concentrations are overestimated when using the RIEOA value of 1.4. Dilution showed to have a significant effect on food cooking experiments, increasing both the gas/particle ratios and the O:C ratios. The data generated in this work, OA-AMS mass spectra and markers from both gas and particle phase identified with FIGAERO-CIMS, provide significant information that will contribute to the improvement of source apportionment in future studies. This work investigates OA, with a focus on primary organic aerosols originated from anthropogenic activities. These scientific findings increase our understanding of OA sources and can help to improve inventories and models as well as to develop plans and policies to mitigate the air pollution in urban environments.

Uppsatsen handlar om att pröva en teori som menar att en hög internetanvändning kan leda till ökad polarisering och därmed bli ett hot mot demokratin genom att ta bort debatt, diskussion och konsensus. Polariseringen ska framkomma genom att internetanvändarna har större möjlighet att förtränga information och åsikter som strider mot ens egna. Samtidigt ska dessa individer även ha större möjlighet att gå med i grupper som endast innehar likasinnade människor. Genom the daily me skapar människor även sig ett ramverk över hur deras vardag ska se ut gällande vilken information som de tar del av. Uppsatsen prövar huruvida samhällets åsikter är mer extrema i Norge och Polen år 2010 och år 2016 bland dem som använder sig av internet ofta kontra sällan, vilket det enligt teorin borde vara. Metoden för att testa detta utgår från en kvantitativ statistisk analys där variabler ställs mot varandra för att identifiera ett samband. Resultaten av uppsatsen visar på att det finns ytterst små mönster av en ökad polarisering bland de som använder internet ofta. Jag argumenterar för att resultaten visar inte på en tillräckligt stor polariseringstrend bland de som använder internet ofta. Värt och nämna är att på grund av att en stor del av kapitel 5 (resultat och analys) består av diagram och tabeller så kommer det att finnas en del större mellanrum mellan vissa delar. Utan dessa mellanrum hade till exempel en halva av en tabell hamnat på en sida och andra halvan på den andra.

Making productive land : utility, encounter, and oil sands reclamation in northeastern Alberta, Canada

This thesis is a landscape ethnography examining conflicting epistemologies and land use values in the Athabasca region of subarctic Canada. Based on 18 months of fieldwork in Fort McMurray, Alberta with the Fort McMurray Métis community and peatland scientists, I analyse oil sands reclamation as a site of encounter between Indigenous and settler peoples. I show how reclamation, a process of reconstructing ecological integrity of a post-extractive landscape for future use, is a historically contingent activity that centres on settler colonial ideologies of productivity. I argue that this ideology spurred a process that I call 'making productive land' which seeks to 'improve' upon or transform the Athabasca region from Indigenous homeland into settler territory or 'useful' land. Weaving archival documents with experiential material from ethnographic fieldwork, I analyse the Athabasca region as a multilocal and multivocal place to demonstrate that Indigenous communities such as the McMurray Métis hold their own definitions of land use that exist alongside, beyond, against, and entangled with settler notions of productivity. I conclude that, for Métis community members, reclaimed areas in fact represent a diminished experience of place or an unproductive landscape. For reclamation to be successful for Métis community members, I contend that reclamation policy and practice must expand beyond purely scientific and resource-based utility narratives to involve a relational element of healing and Métis notions of use that transcend settler colonial ideals of 'productive' land.
