Bcr-Abl é a tirosina quinase (TK) responsável por causar a Leucemia Mielóide Crônica (LMC). Os últimos estudos de follow-up mostram que apenas 50% dos pacientes tratados com a segunda geração de inibidores de TK atinge a remissão completa, o que significa que metade desses pacientes necessita de um algo melhor do que está disponível. Wiskott Aldrich Syndrome Protein (WASP) é um gene essencial para o bom desenvolvimento e função das células hematopoiéticas. Ante esse contexto, decidimos investigar se WASP poderia ter algum papel ou relevância na LMC. Em conclusão, Bcr-Abl suprime a expressão WASP por um mecanismo epigenético. A re-expressão de WASP torna as células mais suscetíveis à apoptose em resposta ao Imatinib. Sugerimos que a recuperação da expressão WASP deve ser discutida como estratégia para a terapia da LMC. / Bcr-Abl is the tyrosine kinase (TK) responsible for causing Chronic Myeloid Leukemia (CML). This fusion protein up- and down-regulates several genes and pathways, producing a strong resistance to apoptosis and a blockage of cell maturation in the hematopoietic compartment. The last follow-up studies provided that only 50% of the patients treated with second generation achieve complete remission, what means that one-half of these patients needs something better. Wiskott Aldrich Syndrome Protein (WASP) is an essential gene for the proper development and function of the hematopoietic cells. In the light of this background, we decided to investigate if WASP could have some role or relevance in the CML context. In conclusion, Bcr-Abl suppresses WASP expression by an epigenetic mechanism. The re-expression of WASP makes the CML cells more susceptible to apoptosis and contribute to respond to Imatinib. We suggest that recovery of WASP expression should be discussed as a new and additional strategy for CML therapy.

Caracterização de subpopulações de Leucemia Mielóide Aguda portadora do rearranjo MLL quanto à resposta diferencial ao tratamento em longo prazo com Citarabina / Characterization of subpopulations of Acute Myeloid Leukemia harboring MLL rearrangements according to differential response to the long-term treatment with CytarabineLarissa Oliveira Guimarães 23 October 2015 (has links)
A natureza heterogênea da Leucemia Mielóide Aguda (LMA) tornou-se um desafio para o sucesso da quimioterapia convencional com o agente Citarabina (Ara-C), especialmente em leucemias com prognóstico desfavorável, como aquelas portadoras do rearranjo MLL. Visto que as células de LMA-MLL são consideradas sensíveis ao Ara-C quando comparadas às leucemias que não apresentam o rearranjo, mas a recaída à doença é frequente, a presente tese propôs estudar a relação entre características biológicas relacionadas às bases da resistêmcia ao Ara-C em LMA-MLL. A abordagem proposta foi a seleção de subpopulações de linhagens celulares portadoras do rearranjo MLL submetidas ao tratamento em longo prazo com Ara-C, comparando-as com as linhagens não expostas à droga. As células foram caracterizadas quanto: 1) ao potencial proliferativo na presença ou ausência de Ara-C; 2) a distribuição das células no ciclo celular; 3) a distribuição de marcadores clássicos de superfície de células-tronco hematopoiéticas, CD34 e CD38; e 4) o perfil de expressão global dos RNAs transcritos. O tratamento em longo prazo selecionou células mais resistentes ao Ara-C que as células parentais. Além disso, quanto ao ciclo celular, as células selecionadas com Ara-C apresentaram apoptose reduzida (fase sub-G1), acúmulo na fase de síntese (fase S) e aumento da capacidade proliferativa após reexposição à droga (fase G2-M). Quanto à análise de marcadores de células-tronco hematopoiéticas, observou-se que após o tratamento em longo com Ara-C, uma das linhagens celulares apresentou distribuição bimodal do marcador CD38. Quando separadas por sorting em citometria de fluxo, observou-se que as subpopulações com níveis distintos de expressão de CD38, denominadas MV-4-11 CD38High e MV-4-11 CD38Low apresentaram resposta distinta ao tratamento com Ara-C. Quando avaliadas quanto ao perfil global de expressão gênica, constatou-se que MV-4-11 CD38High eram mais semelhantes às células parentais, e que MV-4-11 CD38Low formavam um grupo isolado, distinto das outras duas populações celulares. A análise de ontologia gênica (GO) evidenciou que entre as categorias mais representativas de processos biológicos estavam atividades associadas à capacidade proliferativa, ao desenvolvimento e a resposta a estímulos. As análises de agrupamentos hierárquicos mostraram que: 1) o cluster de genes do desenvolvimento HOXA estava mais expresso nas células MV-4-11 CD38Low do que em MV-4-11 CD38High, que apresentaram expressão mais elevada do cluster HOXB; 2) o gene HOX mais diferencialmente expresso foi HOXA13, associado na literatura com prognóstico desfavorável em outros tipos de câncer; 3) dos genes associados a resposta a estímulos, o único relacionado à via de metabolização do Ara-C diferencialmente expresso entre as linhagens foi NME1; 4) aqueles que participam das vias de reparo de pareamento incorreto, reparo por excisão de bases e por excisão de nucleotídeos encontraram-se mais expressos nas células MV-4-11 CD38High que em MV-4-11 CD38Low. Além disso, diversas quinases dependentes de ciclinas (CDKs) também estiveram diferencialmente expressas entre MV-4-11 CD38High e MV-4-11 CD38Low. Sugere-se por fim, que o modelo in vitro proposto neste estudo para simular a situação de resistência ao Ara-C em subpopulações de LMA-MLL, demonstrou que os mecanismos de resposta à Citarabina nesta doença, vão além de alterações na detoxificação e metabolização da droga, e parecem mais associados a vantagens proliferativas e do desenvolvimento das células leucêmicas. Estas vias devem ser exploradas como alvos potenciais na terapia combinada ao Ara-C. / The heterogeneity of Acute Myeloid Leukemia (AML) became a challenge for the success of the conventional chemotherapy agent Cytarabine (Ara-C), especially in leukemias with poor prognosis, as those harboring MLL rearrangement. Since AML-MLL cells are considered sensitive to Ara-C when compared with leukemias that do not carry the rearrangement, but relapse is frequent, the present dissertation proposed to study the relationship between biological characteristics related to the basis of chemoresistance to Ara-C in AML-MLL. We proposed an approach based on the selection of subpopulations of cell lines bearing MLL rearrangement submitted to the long-term treatment with Ara-C, comparing them with the cell lines that were not previously exposed to the drug. The cells were characterized according to: 1) the proliferative potential in the presence and absence of Ara-C; 2) the distribution of the cells in the cell cycle; 3) distribution of hematopoietic stem cell classic surface markers, CD34 and CD38; and, 4) global expression profile of transcribed RNAs. The long-term treatment selected cells that are more resistant to Ara-C than the cells that were not previously treated (parental cells). Besides, according to cell cycle, the cells selected by Ara-C treatment present decreased apoptosis (sub-G1 phase), accumulation in the synthesis phase (S-phase) and increase in the proliferative capability after re-exposition to the drug (G2-M phase). Regarding the hematopoietic stem cell markers, we observed that after Ara-C long-term treatment, one of the cell lines exhibited a bimodal distribution of the CD38 marker. When sorted by flow cytometry, we observed that both subpopulations with distinct levels of CD38 expression, called MV-4-11 CD38High and MV-4-11 CD38Low also showed distinct response to Ara-C. When evaluated regarding to their global gene expression profiles, we verified that MV-4-11 CD38High were more closely related to the parental cells, and MV-4-11 CD38Low made up an isolated group, distinct of the other cell populations. Gene ontology (GO) analysis revealed that among the most representative categories of biological processes, activities associated with proliferative capability, development and response to stimuli were included. The hierarchical clustering analysis showed that: 1) the cluster HOXA of genes of development was more expressed in the MV-4-11 CD38Low than in the MV-4-11 CD38High cells, that presented increased expression of HOXB cluster; 2) the most differentially expressed HOX gene was HOXA13, which according to the literature is associated with poor prognosis in other types of cancer; 3) among the genes associated with response to stimuli, the only one related to Ara-C-metabolizing pathway that was differentially expressed between the cell lines was NME1; 4) those genes that take part in the mismatch repair, base excision repair and nucleotide excision repair pathways were more expressed in the MV-4-11 CD38High than in the MV-4-11 CD38Low cells. Additionally, several cyclin-dependent kinases (CDKs) were also differentially expressed between MV-4-11 CD38High and MV-4-11 CD38Low. Finally, we suggest that the in vitro model proposed in this study to mimic the situation of chemoresistance to Ara-C in subpopulations of AML-MLL, showed that the mechanisms of Ara-C response in this disease, go beyond changes in drug detoxification and metabolization, and seem more associated to proliferative and development advantages of the leukemic cells. These pathways should be explored as potential targets to Ara-C combination therapies.

Sabe-se que TRAIL é capaz de matar células tumorais de forma seletiva e que TRAIL tem sua expressão reduzida em diversos tumores, porém pouco se sabe sobre os mecanismos responsáveis pela sua inibição. Tendo em vista que a expressão de TRAIL pode ser regulada pelo Ácido Retinóico; que PRAME é capaz de inibir a via do ácido retinóico através da proteína EZH2 e que nós observamos anteriormente que a expressão de TRAIL esta diminuída em pacientes com LMC, nós decidimos investigar a associação entre PRAME, EZH2 e TRAIL na LMC. Nós demonstramos que PRAME, mas não EZH2, tem sua expressão aumentada em células BCR-ABL+ e sua expressão está associada com a progressão da LMC. Alem disto, existe uma correlação positiva entre PRAME e BCR-ABL e negativa entre PRAME e TRAIL nestes pacientes. A inibição da expressão de PRAME ou EZH2 por RNAi induziu um aumento da expressão de TRAIL. Estes dados revelam um novo mecanismo de regulação responsável por diminuir a expressão de TRAIL, e geram novos possíveis alvos para a terapia da LMC e, possivelmente, também para outros tumores. / TRAIL was shown to selectively kill tumor cells. Not surprisingly, TRAIL is down-regulated in a variety of tumor cells, but the mechanism responsible for TRAIL inhibition remains elusive. Because TRAIL can be regulate by retinoic acid; PRAME was shown to inhibit transcription of retinoic acid receptor target genes through the polycomb protein EZH2; and we have found that TRAIL is inversely correlated with BCR-ABL in CML patients, we decided to investigate the association of PRAME, EZH2 and TRAIL in BCR-ABL-positive leukemia. Here, we demonstrate that PRAME, but not EZH2, is up-regulated in BCR-ABL cells and is associated with the progression of disease in CML patients. In addition, PRAME expression is positively correlated with BCR-ABL and negatively with TRAIL in these patients. Importantly, knocking down of PRAME or EZH2 by RNA interference restores TRAIL expression. Our data reveal a novel regulatory mechanism responsible for lowering TRAIL expression and provide the basis of alternative targets for combined therapeutic strategies for CML.

Foram analisados os resultados retrospectivos do transplante de medula óssea alogênico em 100 pacientes portadores de leucemia mielóide crônica, divididos em dois grupos: Grupo I, de 49 pacientes que recebeu alta hospitalar após a pega medular, e grupo II, de 51 pacientes, que recebeu alta precocemente, antes da pega medular, para acompanhamento ambulatorial. Foram comparados os dias de ocupação de leitos hospitalares, a morbidade e a mortalidade entre os grupos de alta até o dia 100 pós transplante. Verificou-se que, no grupo de alta precoce, houve, significativamente, menos dias de ocupação de leitos hospitalares, sem aumento da morbidade e da mortalidade / We analyzed the results of a retrospective study of 100 patients with chronic myelogenous leukemia submitted to allogeneic stem cell transplantation in one of two settings: Group I, with 49 patients, transplanted in the traditional inpatient and group II, with 51 patients, in partial outpatient. We compared the median number of days spent in hospital, morbidity and mortality within 100 after bone marrow transplantation. We concluded that there was a significant reduction in the median of hospital length of stay in the partial outpatient group, without increasing morbidity and mortality

Relação entre o oncogene BCR-ABL e os receptores de tipo TOLL (TLR). / Relationship between the oncogene BCR-ABL and Toll-like receptors (TLR).Zenteno, María Emilia 17 November 2010 (has links)
Recentemente, a expressão gênica dos receptores TLR foi encontrada em diversos tipos de células tumorais. A sua participação na biologia do câncer é controversa já que foram descritas ações pró e anti-tumorais após a ativação de sua sinalização. Na Leucemia Mielóide Crônica (LMC) nada se tem demonstrado. BCR-ABL é uma oncoproteína quimérica cujo sítio tirosina quinasa constitutivamente ativado promove inúmeras vias de sinalizações que desencadeia a transformação celular. Este trabalho se inicia com a hipótese de existir uma relação entre o oncogene BCR-ABL e a expressão dos receptores TLRs. Nós verificamos em células murinas TonB210.1 com expressão de BCR-ABL induzível por doxiciclina que Tlr1 e Tlr2 tem sua expressão gênica relativa aumentada na presença da oncoproteína. A regulação positiva de Tlr1 é dependente da ação tirosina quinasa de BCR-ABL. Também mostramos que as vias p38 e JNK estão reprimindo a expressão de Tlr1 induzida por BCR-ABL enquanto que a via ERK é utilizada pelo BCR-ABL para promovê-la. Por outro lado, observamos que a ligação de TLR1/TLR2 com seu agonista sintético Pam3CSK4 em células TonB210.1 BCR-ABL positivas induz um aumento da produção de IL-6 e leva ao aumento da resistência a morte quando induzida pelas drogas Ara-C e VP16. Em conclusão, estes resultados indicam que BCR-ABL esta regulando a expressão gênica de alguns TLRs. Por tanto esses dados contribuem para a compreensão sobre o comportamento de células tumorais BCR-ABL positivas em um contexto de infecção e por conseqüência, dão margem ao estudo de novos alvos de fator de risco para a LMC. / Recently, the gene expression of TLR receptors have been described in several kinds of tumour cells. Its participation in cancer biology is controversial because roles were already been described in pro and anti-tumoral activities after their signaling activation. In Chronic Myeloid Leukemia (CML) there are no published data. BCR-ABL is a quimeric protein and its tyrosine-kinase site is activated constitutively. Thus, many signaling pathways are activated and several cell processes are altered thereby resulting in cellular transformation. This work has started with the hypothesis that a putative relationship between the oncogene BCR-ABL and the expression of TLR receptors could exists. We verified in murine cells TonB210.1 BCR-ABL expression inducible by doxycicline that Tlr1 and Tlr2 have their relative gene expression up-regulated in the presence of the oncoprotein. Therefore the Tlr1 regulation is dependent of BCR-ABL tyrosine kinase action. Using MAPK inhibitors we showed that p38 and JNK pathways are suppressing the TLR1 induction by BCR-ABL while ERK pathway is used by the oncoprotein for promote it. On the other hand, we observed in TonB210.1 BCR-ABL positive cells that the binding of TLR1/TLR2 heterodimer to their synthetic agonist Pam3CSK4 induced an increased production of IL-6 and when these cells were induced by Ara-C and VP-16 drugs the apoptosis resistance increased. In conclusion, these results indicate that the oncoprotein regulates the gene expression of some TLRs. Therefore, this fact gives us data about the behavior of BCR-ABL positive tumor cells in the context of infection and in consequence the study of new risk factor targets for CML.

Previous issue date: 2015-05-29 / Conselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPq / Despite advances in research oncology field, has been an increased incidence and
mortality caused by cancer, consisting in a major public health problem. Literature
reports, that Morita-Baylis Hillman adducts (MBHA) show promising biological
activities, such as: antiparasitic, against Leishmania, Plasmodium sp. and
Trypanosomes, as well as effect on sea urchin embryo development. To assess
anticancer and anti-inflammatory potential of three AMBH: A2CN, and A3CN, A4CN,
we used various biological models. In MTT assay, the adducts were potentially toxic
to eight cancer cell lines tested (HL-60, MOLT-4, K562, K562-Lucena, MCF-7, HT-
29, L929 and B16F10), however A2CN was the most cytotoxic, with IC50 lower for
most cells. Acute myeloid leukemia cells, HL-60 and MOLT-4, were more sensitive
and A2CN had IC50 22 and 21 μM in these cells. In chronic myelogenic leukemia
cells, K562 and Lucena, A2CN presented IC50 of 58 and 60 μM, respectively. A2CN
has been less cytotoxic to normal cells in peripheral blood and normal human
fibroblasts (FN1), and IC50 was 78 and 126 μM, respectively. The action mechanism
induced by A2CN in K562 cells was studied at 15, 30 and 60 μM. The cell viability
has not altered when analyzed with propidium iodide, using flow cytometry, showing
that A2CN not induced necrosis in these cells. A2CN increased mitochondrial
membrane depolarization by 20 % at highest concentration tested. Cytotoxicity of
A2CN in K562 cells were related to cell cycle arrest in G1-phase at 15 μM, and Sphase
at highest concentration (60 uM) and ROS production increased at the highest
concentration tested. Cell cycle arrest in S-phase was related to high expression
mRNA of p21, p27 and p53, together with decreased expression of cyclin D1. A2CN
also augmented expression of Kv1.3 and Kv3.1 genes in K562 cells treated with 60
μM. In electrophysiological assays, using the whole-cell "patch clamp", A2CN (120
μM) promoted increase total K+ current in K562, and conductance K+ ion was
elevated. The potassium channel blocker, 4-aminopyridine (4-AP) (1 mM) decreased
the A2CN cytotoxicity in K562 cells analyzed by MTT reduction, indicating that K+
channels have been involved in its cytotoxicity. The in vitro anti-inflammatory activity
of A2CN, A3CN and A4CN was evaluated in LPS-stimulated RAW 264.7
macrophages. AMBHs did not reduced cell viability to concentration 20 μM, however,
inhibited NO and ROS production induced by LPS. IL-1 β and IL-6 production was
completely inhibited by 10 μM of AMBH, but no change in TNF-α levels. Expression
of IL-1β and IL-6 genes were also altered by A2CN, A3CN and A4CN, but only A2CN
was able to inhibit gene expression of cyclooxygenase-2 (COX-2). Therefore, it can
be concluded that A2CN showed be potent anticancer activity, acting on molecular
targets that are objects preclinical and clinical studies in oncology, such as p53, p21,
p27 and cyclin D1, and proved to be a potent activator of potassium channels.
Furthermore, adducts showed remarkable anti-inflammatory potential, a reduction of
proinflammatory cytokines such as IL-1 β and IL-6 and COX-2 expression by A2CN. / Apesar dos avanços das pesquisas no campo da oncologia, existe um aumento na
incidência do câncer e na mortalidade por esta doença, consistindo em um dos
principais problemas de saúde pública. A literatura relata que os Adutos de Morita
Baylis-Hillman (AMBH) apresentam atividades biológicas promissoras, tais como:
antiparasitária, contra Leishmania sp., Plasmodium sp., e Trypanosoma cruzi, bem
como, efeito no desenvolvimento de células embrionárias de ouriço-do-mar. Para
avaliar o potencial anticâncer e anti-inflamatório de três AMBH: A2CN, A3CN e
A4CN, utilizou-se vários modelos biológicos. Os resultados mostraram que no teste
de redução do MTT, os adutos foram potencialmente citotóxicos para as oito
linhagens cancerígenas testadas (HL-60, MOLT-4, K562, K562-Lucena, MCF-7, HT-
29, L929, B16F10), contudo A2CN foi o AMBH mais citotóxico, apresentando menor
CI50 para a maioria das linhagens. As linhagens de leucemia mielóide aguda HL-60 e
MOLT-4 foram as mais sensíveis, e A2CN apresentou CI50 de 22 e 21 μM nestas
células. Nas células de leucemia mielóide crônica K562 e Lucena, A2CN apresentou
CI50 de 58 e 60 μM, respectivamente. A2CN foi menos citotóxico para as células
normais do sangue periférico e para a linhagem de fibroblastos humano normal
(FN1), cuja CI50 foi de 78 e 126 μM, respectivamente. O mecanismo de ação de
A2CN foi estudado nas células K562, nas concentrações de 15, 30 e 60 μM. A
viabilidade da membrana celular não foi alterada, quando analisada com iodeto de
propídeo em citômetro de fluxo, mostrando que, a molécula não induziu necrose
nestas células. A2CN promoveu um aumento da despolarização da membrana
mitocondrial em 20 %, na maior concentração testada, caracterizando um
envolvimento da via intrínseca da apoptose. A atividade citotóxica de A2CN em
K562 está relacionada à parada no ciclo celular na fase G1, a partir de 15 μM, e na
fase S na maior concentração testada de 60 μM. A produção de ROS foi aumentada
na maior concentração testada. A parada no ciclo celular na fase S está relacionada
com o aumento na expressão do RNAm de p21, p27 e p53 e diminuição na
expressão de ciclina D1. A expressão dos genes para Kv1.3 e Kv3.1 também foi
aumentada nas células K562, tratadas com 60 μM. Nos ensaios eletrofisiológicos
usando a técnica de whole-cell, “patch clamp”, A2CN (120 μM) promoveu um
aumento na corrente total de K+ em K562, bem como, aumentou a condutância ao
íon K+. O bloqueador de canal de potássio, 4-aminopiridina (4-AP) (1 mM) reduziu a
citotoxicidade de A2CN nas células K562, analisadas pela redução do MTT,
indicando que os canais de K+ estão envolvidos na citotoxicidade desta molécula. A
atividade anti-inflamatória de A2CN, A3CN e A4CN, foi avaliada in vitro na linhagem
de macrófagos Raw 264.7, estimulada com (1 μg/ml) de LPS. Os AMBH não
reduziram a viabilidade das células até a concentração de 20 μM, contudo inibiram a
produção de NO e a produção de ROS induzida pelo LPS a partir da menor
concentração dos AMBH, de 2,5 μM. A produção das citocinas IL-1 e IL-6 foi
completamente inibida por 10 μM dos AMBH, mas não houve alteração nos níveis
de TNF-α. A expressão nos genes das citocinas IL-1 e IL-6, também foram alteradas
por A2CN, A3CN e A4CN, porém, apenas A2CN foi capaz de inibir a expressão do
gene da Ciclo-oxigenase-2 (COX-2). Isto posto, pode-se concluir que A2CN
apresentou potente atividade anticâncer, atuando em alvos moleculares que são
objetos de estudos pré-clínicos e clínicos na área oncológica, como p53, p21, p27 e
ciclina D1, bem como demonstrou ser um potente ativador de canais de potássio.
Além disso, os adutos apresentaram notável potencial anti-inflamatório, com
diminuição de citocinas pró-inflamatórias como IL-1 e IL-6, e inibição da expressão
de COX-2 por A2CN.

attention in recent decades for their chemotherapeutic potentials. Specifically,
the interest in molecules containing the 1,2,3-triazole as chemotherapeutic
agents for various diseases has increased, because they are known to exhibit a
wide range of biological activities, such as anti-proliferative and anti-neoplastic.
The aim of this study was to evaluate the potencial anti-cancer effect of new
triazole derivatives from 1,4-naphthoquinone, elucidating their cytotoxicity and
cell death mechanisms involved. From five cancer cell lines tested, leukemic
cells (HL-60 and K562) were the most sensitive, and between the eight triazole
compounds tested (called C1 to C8), compounds C2 and C3 showed the best
IC50 of 14 μM and 41 μM for HL-60 cells and 24 μM and 81 μM for K562 cells,
respectively. However, these two compounds showed very little cytotoxic effect
towards PBMC normal cells with IC50 superior to 80 μM. Investigating the type
of cell death induced by compounds C2 and C3, it was showed that compounds
induced apoptosis in HL-60 cells and cell cycle arrest in the S-phase, and
necrosis in K562 ones. Cell cycle arrest in S phase was related to the
expression of the p21 gene in K562. Results revealed a reduced expression of
Bcl-2 protein and an increased expression of BAX protein in HL-60 cells.
Moreover, it was found cytochrome c release, suggesting involvement of the
intrinsic pathway in apoptosis induction in these cells. Activation of this pathway
may be through inhibition of ERK phosphorylation. Our results also showed that
pre-treatment of HL-60 cells with N-acetyl cysteine (1 mM) for 1 hour reduced
the cytotoxic effect of compounds C2 and C3 for 30,83% and 26,47%
respectively; indicating that the induction of apoptosis in HL-60 is mediated by
increased production of intracellular ROS. Thus, it can be concluded that C2
and C3 compounds showed cytotoxic effects on HL-60 and K562 cells, so, can
be considered as prototype anti-leukemic molecules. / O núcleo triazol e seus derivados têm atraído uma atenção considerável
nessas últimas décadas devido ao seu potencial quimioterápico. Mais
especificamente, tem se verificado o interesse em moléculas que contêm o
grupo 1,2,3-triazol, isto porque esta classe de heterocíclicos é conhecida por
exibir uma vasta gama de atividades biológicas, tais como, anti-proliferativo e
anti-neoplásico. O objetivo desse trabalho foi o de avaliar o potencial
anticancerígeno de novos triazóis derivados do 1,4-naftoquinona, elucidando as
suas citotoxicidades e o mecanismo de morte envolvido. Os resultados obtidos
revelaram que, das cinco linhagens cancerígenas testadas, as linhagens
leucêmicas HL-60 e K562 foram as mais sensíveis, e dentre os oito compostos
(denominados C1 a C8) testados, C2 e C3 foram os mais citotóxicos,
apresentando CI50 de 14 μM e 41 μM, em HL-60 e de 24 μM e 81 μM em K562,
respectivamente. Entretanto, os compostos foram menos citotóxicos nas
células normais do sangue periférico humano com CI50 acima 80 μM.
Investigando o tipo de morte induzido pelos compostos nas duas linhagens, foi
demonstrado que os compostos C2 e C3 induziram apoptose em HL-60. Já nas
células K562, este dois derivados provocaram a parada do ciclo celular na fase
S e necrose. A parada do ciclo celular na fase S em K562 foi relacionada com a
expressão do gene p21. Foi revelado a diminuição da expressão da proteína
Bcl-2 e o aumento da expressão da proteína BAX nas células HL-60, e ainda
verificou-se a liberação do citocromo c sugerindo a participação da via
intrínseca na indução da apoptose em HL-60. A ativação dessa via pode ser via
inibição da fosforilação da proteína ERK. Os resultados mostraram também
que durante uma hora de pré-tratamento das células HL-60 com o N-acetil
cisteina (1 mM), houve a redução da citotoxicidade dos compostos C2 e C3 de
30,83% e 26,47% respectivamente; indicando que a indução da apoptose em
HL-60 é mediada pelo aumento da produção das espécies reativas de oxigênio
intracelulares. Dessa forma, pode-se concluir que os compostos C2 e C3
apresentaram efeitos citotóxicos frente às linhagens HL-60 e K562, então,
podem ser considerados como protótipo de moléculas anti-leucêmicas.

Previous issue date: 2015-04-10 / Fundação de Amparo à Pesquisa do Estado de Goiás - FAPEG / The treatment of chronic myeloid leukemia (CML) has changed dramatically with the
advent of imatinib mesylate (IM). Besides the convenience of oral use, other benefits
were achieved with the new drug, with faster therapeutic responses and increased
survival, giving the CML similar characteristics as chronic diseases. In this scenario,
there was another challenge, drug compliance, since a significant proportion of patients
fail to ingest all the prescribed doses of imatinib. The concern was to optimize the
adherence of CML patients, the hematology ambulatory at the Clinical Hospital of the
Federal University of Goias (HC-UFG), led the authoress to create a film cartoon, as a
pro-adhesion educational intervention model. To investigate the effectiveness of this new
educational material, we used in 65 patients three adherence measures, two indirect
(Morisky Test and Molecular Response [MR]) and direct (plasma dosage of IM), before
and after the screening of film. In univariate analysis, from the Morisky Test, the film
was striking, with increased adherent patients, which increased from 15 (23.1%) to 43
(66.1%). The results of MR showed an improvement trend after the movie, because the
positive molecular response (major MR or complete MR) increased from 81.5% to
86.1%. Regarding the serum levels of IM, with daily doses of 400-800 mg IM, the premovie
samples showed higher average than the post-movie (2473.16 ± 1049.55 ng/ml
versus 1414.72 ± 715 73 ng/ml), with a variation coefficients interpatients of 43.4% and
50.6%, respectively. This high dispersion index found has been reported by other
authors. By multivariate analysis, patients were divided into three groups. The first
brought together compliant patients before and after the film with a good therapeutic
response (major MR) after the intervention. It was: patients over 53 years old, females,
with associated diseases before and after the treatment of CML that use more than two
drugs in addition to imatinib. The second group was marked by the change of not
adherence pre to adherence post-film. Its features were younger than or equal to 53, the
absence of other disease before the CML, the use of less than two drugs and complete
molecular response after the film. In the third group, we observed patients without
molecular response before and after the educational intervention and no medication
adherence after the film. They had in common their age (less than or equal to 53 years),
and drug discontinuation due to adverse reactions. The last represents the set of patients
resistant to the educational film, drawing attention to the fact that only one pro-adhesion
method may be insufficient for all individuals. It is concluded that medication adherence
was higher among patients older than 53 years, the educational film is an effective proadhesion
assistance and continuing education, if combined with another method, it could
help maintain or enhance the benefits achieved in this work. / O tratamento da leucemia mielóide crônica (LMC) mudou radicalmente com o advento
do mesilato de imatinibe (MI). Além da comodidade do uso oral, outros benefícios foram
alcançados com o novo fármaco, como respostas terapêuticas mais rápidas e aumento da
sobrevida, dando `a LMC características semelhantes `as de doenças crônicas. Neste
cenário, surgiu outro desafio, a adesão medicamentosa, pois uma proporção significativa
de pacientes deixa de ingerir a dose prescrita de imatinibe. A preocupação em otimizar a
adesão dos pacientes com LMC, do Ambulatório de Hematologia do Hospital das
Clínicas da Universidade Federal de Goiás (HC-UFG), motivou a autora a criar um filme
em desenho animado, como modelo de intervenção educativa pró-adesão. Para investigar
a eficácia deste novo material educativo, empregou-se, em 65 pacientes, três medidas de
adesão, duas indiretas (Teste de Morisky e Resposta Molecular [RM]) e uma direta
(dosagem plasmática do MI), antes e depois da exibição do filme. Em análise univariada,
pelo teste de Morisky, o filme foi impactante, com aumento dos pacientes aderentes, que
passaram de 15 (23,1%) para 43 (66,1%). Os resultados da RM indicaram uma tendência
de melhora após o filme, pois a resposta molecular positiva (RM maior ou RM completa)
passou de 81,5% para 86,1%. Em relação `a dosagem sérica do MI, com doses diárias
entre 400-800 mg de MI, as amostras pré-filme apresentaram média superior `as do pósfilme
(2.473,16 ± 1.049,55 ng/ml versus 1.414,72 ± 715,73 ng/ml), com coeficientes de
variação interpaciente de 43,4% e 50,6%, respectivamente. Este elevado índice de
dispersão encontrado tem sido relatado por outros autores. Pela análise multivariada, os
pacientes foram separados em três grupos. O primeiro, reuniu os pacientes aderentes
antes e após o filme e com boa resposta terapêutica (RM maior) após a intervenção.
Foram eles: os doentes com mais de 53 anos, do gênero feminino, com doenças
associadas antes e após o tratamento da LMC e que usam mais de dois medicamentos
além do imatinibe. O segundo grupo foi marcado pela mudança de não adesão pré para
adesão pós-filme. Suas características foram idade menor ou igual a 53, ausência de outra
doença antes da LMC, uso de menos de dois medicamentos e resposta molecular
completa pós-filme. No terceiro grupo, observou-se pacientes sem resposta molecular
antes e depois da intervenção educativa, bem como não adesão medicamentosa após o
filme. Eles tinham em comum a idade, menor ou igual a 53 anos, e suspensão do
medicamento por reação adversa. Estes últimos representam o conjunto de pacientes
resistentes ao filme educacional, chamando atenção para o fato de que somente um
método pró-adesão pode ser insuficiente para todos os indivíduos. Conclui-se que a
adesão medicamentosa foi maior entre os pacientes maiores de 53 anos, que o filme
educativo é uma intervenção pró-adesão eficaz e que a educação continuada, aliada a
outro método, poderia ajudar a manter ou ampliar os benefícios conquistados neste
trabalho.

Sabe-se que TRAIL é capaz de matar células tumorais de forma seletiva e que TRAIL tem sua expressão reduzida em diversos tumores, porém pouco se sabe sobre os mecanismos responsáveis pela sua inibição. Tendo em vista que a expressão de TRAIL pode ser regulada pelo Ácido Retinóico; que PRAME é capaz de inibir a via do ácido retinóico através da proteína EZH2 e que nós observamos anteriormente que a expressão de TRAIL esta diminuída em pacientes com LMC, nós decidimos investigar a associação entre PRAME, EZH2 e TRAIL na LMC. Nós demonstramos que PRAME, mas não EZH2, tem sua expressão aumentada em células BCR-ABL+ e sua expressão está associada com a progressão da LMC. Alem disto, existe uma correlação positiva entre PRAME e BCR-ABL e negativa entre PRAME e TRAIL nestes pacientes. A inibição da expressão de PRAME ou EZH2 por RNAi induziu um aumento da expressão de TRAIL. Estes dados revelam um novo mecanismo de regulação responsável por diminuir a expressão de TRAIL, e geram novos possíveis alvos para a terapia da LMC e, possivelmente, também para outros tumores. / TRAIL was shown to selectively kill tumor cells. Not surprisingly, TRAIL is down-regulated in a variety of tumor cells, but the mechanism responsible for TRAIL inhibition remains elusive. Because TRAIL can be regulate by retinoic acid; PRAME was shown to inhibit transcription of retinoic acid receptor target genes through the polycomb protein EZH2; and we have found that TRAIL is inversely correlated with BCR-ABL in CML patients, we decided to investigate the association of PRAME, EZH2 and TRAIL in BCR-ABL-positive leukemia. Here, we demonstrate that PRAME, but not EZH2, is up-regulated in BCR-ABL cells and is associated with the progression of disease in CML patients. In addition, PRAME expression is positively correlated with BCR-ABL and negatively with TRAIL in these patients. Importantly, knocking down of PRAME or EZH2 by RNA interference restores TRAIL expression. Our data reveal a novel regulatory mechanism responsible for lowering TRAIL expression and provide the basis of alternative targets for combined therapeutic strategies for CML.

Relação entre o oncogene BCR-ABL e os receptores de tipo TOLL (TLR). / Relationship between the oncogene BCR-ABL and Toll-like receptors (TLR).María Emilia Zenteno 17 November 2010 (has links)
Recentemente, a expressão gênica dos receptores TLR foi encontrada em diversos tipos de células tumorais. A sua participação na biologia do câncer é controversa já que foram descritas ações pró e anti-tumorais após a ativação de sua sinalização. Na Leucemia Mielóide Crônica (LMC) nada se tem demonstrado. BCR-ABL é uma oncoproteína quimérica cujo sítio tirosina quinasa constitutivamente ativado promove inúmeras vias de sinalizações que desencadeia a transformação celular. Este trabalho se inicia com a hipótese de existir uma relação entre o oncogene BCR-ABL e a expressão dos receptores TLRs. Nós verificamos em células murinas TonB210.1 com expressão de BCR-ABL induzível por doxiciclina que Tlr1 e Tlr2 tem sua expressão gênica relativa aumentada na presença da oncoproteína. A regulação positiva de Tlr1 é dependente da ação tirosina quinasa de BCR-ABL. Também mostramos que as vias p38 e JNK estão reprimindo a expressão de Tlr1 induzida por BCR-ABL enquanto que a via ERK é utilizada pelo BCR-ABL para promovê-la. Por outro lado, observamos que a ligação de TLR1/TLR2 com seu agonista sintético Pam3CSK4 em células TonB210.1 BCR-ABL positivas induz um aumento da produção de IL-6 e leva ao aumento da resistência a morte quando induzida pelas drogas Ara-C e VP16. Em conclusão, estes resultados indicam que BCR-ABL esta regulando a expressão gênica de alguns TLRs. Por tanto esses dados contribuem para a compreensão sobre o comportamento de células tumorais BCR-ABL positivas em um contexto de infecção e por conseqüência, dão margem ao estudo de novos alvos de fator de risco para a LMC. / Recently, the gene expression of TLR receptors have been described in several kinds of tumour cells. Its participation in cancer biology is controversial because roles were already been described in pro and anti-tumoral activities after their signaling activation. In Chronic Myeloid Leukemia (CML) there are no published data. BCR-ABL is a quimeric protein and its tyrosine-kinase site is activated constitutively. Thus, many signaling pathways are activated and several cell processes are altered thereby resulting in cellular transformation. This work has started with the hypothesis that a putative relationship between the oncogene BCR-ABL and the expression of TLR receptors could exists. We verified in murine cells TonB210.1 BCR-ABL expression inducible by doxycicline that Tlr1 and Tlr2 have their relative gene expression up-regulated in the presence of the oncoprotein. Therefore the Tlr1 regulation is dependent of BCR-ABL tyrosine kinase action. Using MAPK inhibitors we showed that p38 and JNK pathways are suppressing the TLR1 induction by BCR-ABL while ERK pathway is used by the oncoprotein for promote it. On the other hand, we observed in TonB210.1 BCR-ABL positive cells that the binding of TLR1/TLR2 heterodimer to their synthetic agonist Pam3CSK4 induced an increased production of IL-6 and when these cells were induced by Ara-C and VP-16 drugs the apoptosis resistance increased. In conclusion, these results indicate that the oncoprotein regulates the gene expression of some TLRs. Therefore, this fact gives us data about the behavior of BCR-ABL positive tumor cells in the context of infection and in consequence the study of new risk factor targets for CML.
