FBXO44-MEDIATED DEGRADATION OF RGS2

<p>  G Protein Coupled Receptor (GPCR) signaling plays a key role in intercellular communication and regulates many physiological processes relevant to disease. Approximately 30-40% of all FDA approved drugs target GPCR pathways, but limitations and off-target side effects remain obstacles. Regulator of G protein Signaling (RGS) proteins negatively modulate GPCR signaling by accelerating deactivation of the Gα subunit and thus represent a novel alternative to current approaches. While research on RGS proteins and how they are regulated has expanded rapidly, there are still gaps in knowledge for some members of the RGS family. One example is RGS2, which is selective for Gαq signaling. Lowered RGS2 levels are implicated in numerous diseases, and while the E3 ligase responsible for facilitating degradation of RGS2 has been identified more work needs to be done to viably drug it to enhance RGS2 protein levels. In this thesis, I explore how FBXO44, an E3 ligase substrate recognition component responsible for RGS2 degradation, interacts with RGS2 to explore approaches to inhibit degradation.</p> <p><br></p> <p>While the FBXO44-RGS2 interaction has been demonstrated previously, the degron sequence of RGS2 remained unknown. We hypothesized that FBXO44 binds RGS2 at its Nterminus and investigated this using N-terminally truncated RGS2 constructs. Our results indicated that FBXO44 binds between residues 5 and 16 of RGS2, as removal of these stabilized RGS2 against proteasomal degradation. Based on these results we designed a peptide microarray to identify important residues and properties for FBXO44 in vitro and found that Cys13 is essential for FBXO44 binding.</p> <p><br></p> <p>We also developed and optimized a high-throughput split luciferase screen to identify potential inhibitors of the FBXO44-RGS2 interaction. After forming a cell-line stably expressing tagged FBXO44 and RGS2 and optimizing assay condition, we achieved a robust assay for screening as determined by Z’-factor. <br>  </p>

[pt] Cotas de tela estão em vigor no Brasil, em sua forma atual, desde 2001. Por lei, cinemas devem exibir uma certa quantidade de dias de filmes brasileiros por ano. Embora duas décadas tenham se passado desde a instituição das cotas, são poucas as avaliações quantitativas dos seus efeitos. Além disso, a política expirou ao final de 2021 e não foi renovada. A reintrodução de cotas é certamente um assunto legislativo a ser debatido. Para investigar os efeitos da política, primeiro executamos um conjunto de regressões em forma reduzida, usando a variação exógena nas cotas de acordo com o tamanho dos complexos de exibição. Em seguida, nós construímos e estimamos um modelo de escolha discreta dinâmico das decisões de programação dos exibidores. As regressões em forma reduzida apontam para efeitos negativos das cotas de tela na bilheteria e no público de filmes estrangeiros e em geral, mas o impacto na receita e no público de filmes brasileiros é próximo a zero. Apesar disso, as cotas parecem induzir cinemas a exibirem mais filmes brasileiros. / [en] Screen quotas in Brazil have been in effect, in their present form, since 2001. Legislation requires movie theaters to screen Brazilian movies for a number of days on a yearly basis. Even though two decades have passed since its inception, quantitative analyses of the policy s effects have been scarce. Furthermore, the policy expired by the end of 2021. The reintroduction of quotas is certainly a matter of legislative relevance in the upcoming years. To investigate policy effects, we first run a set of reduced-form regressions, using exogenous variation in the movie theater quotas per viewing room. Next, we build and estimate a dynamic discrete choice model of exhibitor choice. Reduced-form regressions point to negative effects of screen quotas on overall and foreign films box-office and ticket sales, but impact on Brazilian movie revenue or public is either zero or very small. Nevertheless, quotas do seem to prompt movie theaters to screen more Brazilian movies.

Purpose: The purpose of this study is to investigate and analyze whether the current platform elements can handle the additional loads from the introduction of platform screen doors at various subway stations. The specific stations involved in the study were Norsborg, Alby, Fridhemsplan, Globen and Alvik. Method: By introducing the platform screen doors, the number of potential accidents is minimized, and the safety of passengers is increased. In order to see if the platforms would be capable of handling the extra load from the platform doors, engineers have measured the thickness of the current platforms to see whether they would be able to withstand the additional pressure that comes with the installation of platform screen doors. Results: Based on the calculations of the different platform types, the results show that platform type 1 can not handle loads from the platform doors and has to be reinforced to minimize the risks of breakage and tilting. Platform types 2 and 3 can manage the weight from the platform doors but should also be reinforced since there is a risk that they will break in the future. In order to achieve safety for people on the platform, the results show that a half-high platform door is the best choice. Conclusions: The available manufacturers on the market offer some different models of platform screen doors, both full and half-height. Since stations and their platform elements vary in design and quality, it is not the optimal choice to have the same type of doors on all stations. The best choice for indoor stations is to install full height screen doors where it is possible. Although, half-height screen doors can be installed on all stations. The half-height screen doors result in the most consistent performance while maintaining the safety of people on the platform.

I ett allt mer digitaliserat samhälle har även utbildningen blivit genomsyrad av denna digitalisering. Det har lett till en ökning i produktion av videoföreläsningar som studenter, på högskolenivå främst, allt oftare använder sig av i inlärningssyfte. E-lärande har många fördelar eftersom det bland annat leder till ökad studieflexibilitet. Det finns ett rikt utbud av tidigare forskning som kretsar kring områdena inlärningsbeteenden, skärmstorlekar och videoföreläsningar. Tidigare studier visar både på att det kan skilja sig i inlärning beroende på skärmstorlek men det finns också studier som tyder på motsatsen. Detta kandidatarbete undersöker om skärmstorlek har någon påverkan på hur mycket man lär sig av att se på en videoföreläsning. Det gjordes genom att gruppera 34 försöksdeltagare i tre grupper där alla fick se en vald föreläsningsvideo på tre olika grupper av skärmstorlekar: liten (4-7 tum), medel (13-17 tum) eller stor (27 tum). Några dagar innan och direkt efter undersökningen besvarade deltagarna även en enkät med sakfrågor om innehållet i den valda videon. Utifrån de resultat som framkom så visar det sig att olikheter mellan dessa skärmstorlekar inte ger en signifikant påverkan på hur mycket man lär sig av en föreläsningsvideo av intervju-typ. Det innebär alltså att man för sådana föreläsningvideor inte behöver ta hänsyn till skärmstorleken, varken som student eller som videoproducent. / In a more and more digitized society, the education has also been steeped in this digitization. This has lead to a rise in the production of video lectures which students, mostly at degree level, more often use for learning purposes. E-learning has many advantages since it, among other things, leads to an increased flexibility when studying. There are many previous studies on learning behaviours, screen sizes and video lectures. Earlier studies show that learning can differ depending on screen size but there are also studies showing the opposite. This bachelor’s thesis examines if screen size has an effect on how much one learns from watching a video lecture. This was done by grouping 34 test participants into three groups where they all watched a video lecture on three groups of screen sizes: small (4”-7”), medium (13”-17”) or large (27”). A few days before and directly after the test the contestants answered a survey with questions regarding the content in the specific video. The results are that differences between these screen sizes do not have a significant impact on how much one learns from a video lecture of interview type. This therefore means that one does not have to consider the screen size for that type of video lecture, either as a student or a video producer.

Numerical Investigation of Sloshing Motion Inside Tuned Liquid Dampers With And Without Submerged Screens

<p> A numerical algorithm has been developed to solve the sloshing motion of liquid in a Tuned Liquid Damper (TLD) outfitted by slat screens under large and random amplitude of excitation. It is based on the finite-difference method. The free surface has been reconstructed using volume of fluid method. Donor-acceptor technique has been used for tracking the volume fraction field. The effect of slat screen has been included and modeled using the partial cell treatment method. </p> <p> The algorithm is an integrated fluid-structure model where the response of the structure is determined considering the effects of TLD. The structure is assumed as a single degree of freedom system (SDOF) and its response is calculated using the Duhamel integral method. </p> <p> The algorithm has been validated against experimental data for the cases with and without screens. An excellent agreement was obtained between numerical and experimental results. </p> <p> An extensive parametric study has been carried out investigating the effect of slat screens and screen pattern on the TLD performance and on the structure response. A new parameter termed as slat ratio was introduced to characterize the slat screens based on their pattern. Results indicated that screen pattern has a significant effect on the TLD performance and it could lead up to 33 % reduction in structure response. It was found that decreasing the slat ratio will increase the damping effect of a TLD outfitted by slat screen. </p> <p> The validity of the most commonly used approach, Baines and Peterson model, to calculate pressure drop of slat screens has been investigated. A conelation factor as a function of Reynolds number and solidity ratio of screen has been proposed to improve the results of this model. A new concept termed as effective solidity ratio has been proposed to account for the physical significant of screen pattern on TLD performance. </p> / Thesis / Doctor of Philosophy (PhD)

Автором также доказано, что мифотворческие аспекты в экранной культуре являются важной формой конструирования новой виртуальной реальности, влияющей на общественное и индивидуальное сознание, на процесс социализации личности. В проектной части диссертации представлена научно-методическая разработка спецкурса для вуза «Мифологические аспекты современной экранной культуры» (онлайн-курс). / The dissertation is devoted to theoretical analyzes of myth-creating aspects in the development of on-screen culture (cinema, TV, video, computer channels, etc.) as a new communication paradigm. In the dissertation, relying on domestic and foreign sources, the screen culture is studied as a concept of the information era, the dialectics of the relationship in the development of the screen culture and the mythologization of reality as a dynamic system is established. The author also proved that the myth-creating aspects in the screen culture are an important form of constructing a new virtual reality that affects the social and individual consciousness, the process of personality socialization. In the design part of the dissertation presents the scientific and methodological development of a special course for the university "Mythological aspects of modern screen culture" (online course).

Entwicklung und Charakterisierung piezokeramischer Dickschichten mit hohem aktorischen Potenzial für aktive Mikrosysteme

Siebgedruckte piezokeramische Dickschichten eignen sich für Sensoren, Aktoren oder Ultraschallwandler. Zur Auslegung von Dickschicht-Aktoren werden grundlegende Kenntnisse über die elektromechanischen Eigenschaften benötigt. Diese Arbeit beschreibt die elektromechanischen Eigenschaften piezokeramischer Dickschichten unter Klein- und Großsignalbedingungen. Durch gezielte Variationen der Biegerlänge und Materialien werden Einflüsse auf die Gefügeentwicklung, das Arbeitsvermögen und weitere elektromechanische Kennwerte beschrieben und optimale Herstellungsparameter identifiziert. Interdigitalelektroden (IDE) ermöglichen die Polarisation und Ansteuerung der piezokeramischen Schicht in der Ebene (in-plane). Aufgrund des d 33 -Effektes kommt es zur Dehnung in der Ebene, woraus eine Verdopplung der Biegung in entgegengesetzter Richtung zu through-thickness-Aktoren resultiert. In der Arbeit wird das Arbeitsvermögen siebgedruckter in-plane-Biegeaktoren in Abhängigkeit vom Substratmaterial, von der Biegerlänge und vom piezokeramischen Material beschrieben und diskutiert. An zwei Mikropositioniersystemen wird die Eignung piezokeramischer Dickschichtaktoren für adaptive optische Systeme gezeigt.

Thin Indium Tin Oxide Layer Development for Crystalline Silicon/Perovskite Two Terminal Tandem Solar Cell

ITO is widely regarded as the optimal TCO for serving as front window layer in PSK/c-Si tandem solar cells. It is known to effectively mitigate several stability issues present in perovskite solar cells while demonstrating excellent lateral conductivities and optical transparency across the entire solar spectrum. However, due to the damaging effects of traditional magnetron sputtering methods on the underlying cell precursor and the limited range of annealing temperatures viable for maintaining the stability of Perovskite Solar cells, realizing the full capability of ITO layer is constrained. This investigation focuses on developing and optimizing the front Indium Tin Oxide (ITO) layer properties for high-efficiency monolithic Perovskite/PERC tandem solarcells. The study employs two widely employed industrial techniques, Magnetron Sputtering and Screen Printing for the deposition of ITO thin-films and subsequent metallization of Ag front contacts. The sputtering process parameters, namely the carrier speed, O2 : Ar ratio, and the sputter power were varied to obtain an optimized ITO layer, which exhibited a thickness of 53nm, Rsheet of 107 ohm/□, mobility of 37 cm2/V s, and 90 % average optical transparency between 400−1200nm. A low contact resistivity of 5.4mΩ·cm2 was achieved between the ITO and metal contacts which is the lowest reported value for ITO annealed at low temperature (140 °C). Champion cells, featuring Perovskite on Ohmic substrate and 2T perovskite/PERC tandem cells, exhibited high VOC values of 1.116 V and 1.75 V on 0.97 cm2 cell aperture areas and cell efficiencies of 17.2 % and 23.85 %. Additionally, a large area (158.7 cm2) tandem cell was also fabricated which demonstrated an excellent VOC of 1.75 V . The results of this investigation demonstrates the versatility of ITO layer properties achievable at low-temperatures through Magnetron sputtering and underscores the potential of existing commercialized technologies for the fabrication of high-efficiency tandem solar cells. / ITO anses allmänt vara den optimala TCO för användning som frontfönsterskikt i PSK/c-Si tandemsolceller. [ES] Los objetivos de este estudio son en primer lugar un análisis previo del proceso de introducción y asentamiento del sistema de estampación permeográfico conocido con el nombre de serigrafía como medio de expresión artística. Para identificar cuál fue el momento y las razones de su introducción en España, y más particularmente en Valencia, será necesario llevar a cabo una serie de acciones como son: puntualizar cuáles han sido los diferentes pasos que han marcado el desarrollo y evolución dentro del contexto valenciano, lo que nos conduce necesariamente al estudio de los principales artífices polarizados en tres núcleos: artistas, serígrafos y editores. También señalar cuáles fueron los principales artistas, grupos y tendencias y las causas de que este nuevo sistema fuera utilizado como medio técnico para estampar sus creaciones artísticas, profundizando en las características técnicas y estilísticas de estas obras y su relación tanto con el conjunto nacional como internacional. Se debería analizar, así mismo, la forma en que se produjo la asimilación del nuevo procedimiento por la sociedad española, en relación a otros artistas, estilos, editores, etc.; teniendo en cuenta varios puntos: que su uso artístico es muy reciente, que los primeros en utilizarla fueron grupos y movimientos de vanguardia, y las connotaciones industriales y publicitarias con que este arrancó.
