Hibridinių baltymų gamybos, panaudojant Saccharomyces cerevisiae raiškos sistemą, tyrimas / Production of hybrid proteins using saccharomyces cerevisiae expression systemKolendaitė, Rasa 25 November 2010 (has links)
Saccharomyces cerevisiae mielės yra vienas plačiausiai rekombinantinių baltymų gamybai, todėl šio darbo metu buvo kuriamos mielių S. cerevisiae raiškos sistemos, skirtos sintetinti tioredoksino geną arba gliukozės dehidrogenazės geną sujungtą su Abeta peptidu. Atrinkus stabilius hibridinių baltymų producentus, jie buvo auginami įvairiose kultyvavimo terpėse, keičiant temperatūrą bei indukcijos laiką. Tokiu būdu parenkamos optimalios kultūrų auginimo sąlygos, užtikrinančios didžiausias gaminamo baltymo kiekius. Šio darbo metu sukonstruoti mielių raiškos vektoriai, kuriuose hibridinių baltymų - tioredoksino sujungto su Abeta peptidu (Trx-ab) bei ir gliukozės dehidrogenazės sujungtos su Abeta peptidu (GDH-ab) - raiška yra reguliuojama galaktoze indukuojamu GAL-CYC1 promotoriumi. Atlikus sėkmingą Saccharomyces cerevisiae 21PMR mielių kamieno transformaciją rekombinantinėmis plazmidėmis, atrinkti optimalūs heterologinių baltymų gamintojai. Ištyrus sukonstruotas DNR plazmides, nustatytas didelis (90-95%) auksotrofinių markerių stabilumas ir tik apie 50% Trx-ab arba GDH-ab genų išsilaikymas mielėse. Sukonstruotos viduląstelinės ir ekstraląstelinės mielių ekspresijos sistemos, užtikrinančios tioredoksino sujungto su Abeta peptidu sintezę. Optimizuotos mielių auginimo sąlygos bei baltymų gryninimo procedūros ir nustatyta, kad didžiausias hibridinio darinio lygis stebimas viduląstelinio gamintojo citoplazminėje frakcijoje. [toliau žr. visą tekstą] / The principal advantages of S. cerevisiae yeast to express recombinant proteins was used to study expression peculiarities of the fused bacterial thioredoxin or glucose dehydrogenase genes with Abeta peptide sequence. In the next step of this work we investigated the growth conditions of stabily persisting yeast transformants in order to achieve a maximal production of the hybrid proteins. In the present work yeast expression vectors bearing either thioredoxin-encoding gene fused with Abeta peptide (Trx-ab) or its mutant sequence or glucose dehydrogenase gene fused with Abeta (GDH-ab) under the control of galactose-inducible GAL-CYC1 promoter were constructed. After successful transformation of yeast Saccharomyces cerevisiae strain 21PMR the eukaryotic producents of the hybrid proteins were isolated. Analysis of stability of heterologous expression plasmids indicated 90-95% maintenance of auxotrofic markers and only 50% - that of Trx-ab or GDH-ab genes. Intracellular and extracellular yeast expression system were designed to ensure synthesis of tioredoxin fused with Abeta peptide. Emocinių ir elgesio problemų turinčių paauglių pykčio raiškos būdai / The Ways of Anger Expression of Adolescents with Emotional and Behavioral ProblemsPaulauskienė, Gražina 22 June 2006 (has links)
The aim of the present work has been to analyze the ways of adolescents’ anger expression and to estimate the relations between emotional and behavioral problems. There was collected data about 198 11-18 years old adolescents. In the research there were used YSR 11/18: Youth self – report (Achenbach, 1991), TRI - Interpersonal Reactivity Index (Davis, 1980) and STAXI – C/A– State - Trait Anger Expression Inventory – 2 For Children and Adolescents (Spielberger, 1999).
The results of the research show that there is a positive correlation between anger expression index and internal behavioral problems (value of p is 0.00) and external behavioral problems (value of p is 0.00). Internal behavioral problems also correlate with anger as a state (value of p is 0.00), anger as a trait (value of p is 0.00), internal (value of p is 0.00) and external (value of p is 0.00) anger expression, internal anger control (value of p is 0.02). External behavioral problems correlate with anger as a state (value of p is 0.00), anger as a trait (value of p is 0.00) and external anger expression (value of p is 0.00). There was estimated a negative correlation between adolescents’ anger as a state and their competence (value of p is 0.02). A positive correlation estimated between external anger expression and social activity (value of p is 0.01). Every human being in close contact with his family, friends and society. Ties with these people are based on personal qualities and social habits which have been started since early childhood. The formation of social habits is especially important to children growing in partronage institutions. Social skills are an ability: - to get on well with other people and groups, to make and develop interpersonal ties, to look for compromises, to find the rational ways of solving conflicts; - to learn, to work and create together with the others, to achieve the common goals; - to participate in cultural, civil and political life of the community, to respect democratic principles and values of the society, to take care of the environment. Social upbringing is a process, the aim of which is to form important personal qualities of a child: responsibility, independence, tolerance, dutifulness and others which play a great role in their socialization. Sociological researches and practical works show that inmates of Children‘s Home form poor social skills due to objective and subjective reasons. That is why the expression of social shills in the Children‘s Home was chosen as the object of the research. The main purpose of this work is to give the light to the expression of social skills and possibilities of their formation in the children growing in orphanages. The contemporary society has set certain standards or norms defining what a human being should be and how he should behave. Other individuals, especially with some kind of incapacity, often become unacceptable or even unnecessary. Such an attitude towards an incapacitated person must be undisputedly changed. “The role of art in spreading cognitive powers of the intellect is recognized as one of the most important factors in development of the individual” (Matonis, 2000, :18-19).
A research has been conducted attempting to assess a potential for self-expression of teenagers with mental disorder and optimization of the socialization process (while them taking part in a social and art project).
The research in the social and art project “Katedra” involved 10 young persons with mental disorder and 5 professional artists. Also by means of filling in a questionnaire 123 assessors – spectators representing the viewpoint of the society on the results of the project, i.e. a performance with participation of disabled and healthy artists, were interviewed. The research revealed changes in the attitude of the society towards people with intellectual problems and showed growing possibilities of social optimization for the disabled persons. It was also concluded that participation in art projects helps young disabled people easier integrate into the society and offers them equal chances to play a role in the life of the society. In this work "Musical competence's features of the fourth year accordion players" is analyzed the features of the musical competency. The subject of research is a musical competence's feature. The purpose of work is to rate musical competence's features of the fourth year accordion players.
The object of musical education is musical culture of person, his artistic, intellectual, moral quality and spiritual wealth. This attitude forms adequate theoretical basics, maximizes pedagogical responsibility for the quality of education content, realization logistics, cultural development. So the objects of musical education, system of assignments, practical activity must be regularized accordingly.
Musical education must comprehensively influence culture; improve not only qualification, but competence too. This leverage is thinkable when details of competence are actualized all in one. The quality, complexion, influence of music subject are educed in this context.

Temos aktualumas ir problema. Mokymo institucijose mokant dailės, meno istorijos tai nesusiejama su vizualine raiška. Tai vieni kitus sąlygojantys dalykai. Aktualu išsiaiškinti ugdytinių žinias apie tapybos raiškos būdus ir jų raidą.
Tyrimo tikslas – atskleisti tapybos raiškos būdų raidos ypatumus.
Tyrimo objektas – tapybos raiškos būdų raida.
Tyrimo hipotezė – tapybos raiškos būdų bei raidos pažinimas daro įtaką studentų vizualinės raiškos lavinimui.Tyrimo uždaviniai: 1. Remiantis filosofine, menotyrine ir kita literatūra, atskleisti tapybos sampratą ir jos raiškos būdų raidą veikiančius veiksnius. 2. Išanalizuoti tapybos raiškos būdus ir aptarti jų raidą. 3. Ištirti gimnazijos II–IV klasių mokinių ir dailės specialybės I–II kursų studentų požiūrį į jų žinias apie tapybos raiškos būdus ir jų raidą.

This master’s work presents the analysis of the theoretical aspects of the organizational climate of Lithuanian and foreign authors in consideration of the peculiarities of financial mediation company, and the particularity of its activity. There is analyzed the concept of “organizational climate” and is presented the factors influencing the organizational climate in this work. With reference to scientific theoretical literature, a research of organizational climate has been carried out in SC bank “Snoras”, using a standardized “Test of Organizational Climate Crisis”, and evaluating the peculiarities and assumptions of the expression of organizational climate. This research together with the written report about it constitutes the main part of this master’s work. 51 respondents of SC bank “Snoras” Šiauliai branch have been examined in pre-test and 43 respondents representing 7 departments – in first and second retests. The data have been processed using the typical SPSS programme. It emerged that positive organizational climate dominates in SC bank “Snoras” Šiauliai branch, but critical dimensions of the expression of organizational climate have also been identified.

Mokytojas yra svarbiausias švietimo paslaugų teikėjas. Nuo jo kvalifikacijos, sociokultūrinio pasirengimo priklauso ne tik teikiamų paslaugų kokybė, bet ir konkrečios švietimo institucijos realios galimybės deramai atlikti savo funkcijas. Niekas kitas taip gerai nejaučia ir nemato, kuo gyvena, kaip auga, vystosi ir bręsta mokinys mokykloje, kaip jo mokytojas. Todėl mokytojo vaidmuo šiuo atveju yra labai svarbus. Deja, gyvenime yra ir kita pusė, kuri tiesiog šaukte šaukia, kad šiandien mokytojai nesijaučia saugūs dirbdami mokykloje.
Šio baigiamojo magistrinio darbo tikslas buvo išsiaiškinti smurto prieš mokytojus ypatumus šiuolaikinėje mokykloje, išnagrinėti kaip ir kodėl pasireiškia smurtas prieš mokytojus.
Tikslui pasiekti buvo iškelti tokie uždaviniai: pateikti smurto sampratą ir išanalizuoti sociologines nusikalstamo elgesio teorijas, aptarti smurto pasireiškimo formas, aptarti smurtą prieš mokytojus skatinančius veiksnius, aptarti smurto prieš mokytojus atsiradimo ir gyvavimo prielaidas švietimo institucijoje bei remiantis atlikto tyrimo duomenimis nustatyti mokinių, tėvų ir mokytojų požiūrį į smurtą prieš mokytojus ir galimas prevencines priemones.
Darbe buvo iškeltos tokios hipotezės: 1. Mokinių smurtas prieš mokytojus yra realiai egzistuojanti socialinė problema. 2. Quality of such services and actual abilities of particular educational institution to function properly depend on teacher’s qualification and social-cultural readiness. No one else can feel and see what makes schoolchildren tick or how they grow, develop and mature at school but their teacher. Hence the role of teacher is of extreme importance in this case. However, there is a dark side of life, which rants and raves that teachers do not feel safe working at school today.
The aim of the MA thesis was to find out peculiarities of violence against teachers in modern schools, analyze how violence against teachers expresses itself and why it is so.
The following tasks were put in order to achieve the abovementioned goals: to present a conception of violence and analyze sociological theories of criminal behaviour, discuss expression forms of violence, subject factors stimulating violence against teachers, scrutinize premises of origin and existence of violence against teachers in educational institutions and, based on survey data, gathered by the author of the thesis, ascertain attitudes of schoolchildren, their parents and teachers towards violence against teachers and possible preventive measures to be taken.
