Previous issue date: 2016-02-26 / Capes / Promop / The hematology and serum biochemistry are great tools
used for early diagnosis in birds. However, the capture
stress and sedation can cause changes in blood count.
Heparinized plasma is often used to perform biochemical
tests, but most of the biochemical kits do not allow the
use of this type of sample. This study aimed to evaluate
the differences found in the blood count among guans
physically restrained and anesthetized with isoflurane.
There were also investigated possible differences in the
results of biochemical tests using serum and heparinized
plasma and EDTA as anticoagulants. It was used 32
free-living guans, which live in the IBAMA Park in the city
of Painel - SC and was captured with trapdoor. Blood
samples were obtained after physical restraint of the
birds by puncturing the ulnar vein and the obtained
sample was placed in heparinized tubes The birds were
anesthetized with isoflurane and after 30 minutes a new
blood collection took place, with the birds still
anesthetized, through puncture of the jugular vein and
samples were divided and placed into tubes containing
heparin or EDTA 3% and tube without anticoagulant.
Blood cells count was performed in times of physical and
12
chemical restraint with heparinized samples. Biochemical
analysis was performed only at the time of chemical
restraint in the samples containing serum and
heparinized plasma and EDTA 3%. Data from the
hematology results were analyzed by paired t-test and
the results of biochemical analyzes by ANOVA test, and
the differences submitted to Tukey test. There was a
decrease in the erythrocyte count, hemoglobin and PVC
measurement and increased MCHC in chemical restraint
moment. The TPP values, fibrinogen and thrombocytes
also decreased in the chemical restraint moment.
Regarding the analysis of differences between serum
and plasma with different anticoagulants, there was an
increase in albumin values and a decrease in globulin
values in heparinized plasma compared with the serum
and EDTA 3%. EDTA 3% plasma had lower GGT and
calcium values compared to serum and plasma with
heparin. It was concluded that isoflurane influences the
hematological parameters and should be taken into
account in interpreting the test. Also, the biochemical
parameters evaluated in this study from serum and
heparinized or EDTA 3% plasma, mostly showed no
differences and could be used in clinical biochemical
evaluation of these animals in such conditions / A hematologia e bioquímica sérica são grandes
ferramentas utilizadas para o diagnóstico precoce em
aves. No entanto, o estresse de captura e a sedação
podem causar alterações no hemograma. O plasma
amostra.
aumento no CHGM no momento contenção química. Os
valores de PPT, fibrinogênio e trombócitos também
diminuíram no momento contenção química.

January 2007 (has links)
Abstract: The laboratory tests are known as important diagnostic tools in veterinary medicine, regardering the diagnosis of many diseases, specially in dogs. Among ali these tests, the biochemical ones can be brought out, not only for their clinical importance, but also for their easy performance and because they are inserted as ordinary tests in the veterinary laboratories. Due to the lack of information as for the viability of blood components in dogs, and the need of blood storage under different conditions, the aim of this study was to check out the stability of some blood components under different temperatures and storage periods. Therefore, the blood samples of 10 healthy dogs were evaluated regardering sodium, potassium, calcium, phosphorus, colesterol, tryglicerides, total protein, albumine, creatinine, urea and enzyme activities of asparatate aminotransferase, alanine aminotransferase, alkaline phosphatase, gama glutamyl transferase and creatine quinasa stored under different temperature (25°C, 4°C, -4°C, -20°C e -70°C) and assessed in different intervals (just after collection, 12 hours, 24 hours, 36 hours, 7 days, 15 days, 30 days, 60 days and 90 days after collection). Among ali the results, it was inferred that the freezing temperature (-20°C) provided the best stability of the evaluated parameters, and 15 out of them showed to be stable during the whole protocol. / Mestre

The biological rhythm can be understood as an expression of a cyclic biological phenomenon. Physiological markers such as melatonin and corticosterone are under the influence of endogenous clocks and respond to different variations of light intensities may be involved in behavioral changes. Several reports have documented an increase in their concentrations in the presence of stressful situations. This work aimed to evaluate the effect of the temporal pattern of behavioral and biochemical parameters in response to stress by restraint in rats. We used male Wistar rats with 70 days (150-180g) were divided into four groups by Zeitgebers (ZT) 0, 6, 12 and 18, subdivided into: control, immediately, 6 and 24 hours after a session of stress restriction. For the behavioral assessment, we used the open field apparatus (OF) and Plus Maze (PM). We analyzed the serum levels: corticosterone, melatonin, glucose and activity of the enzymes NTPDase and 5'nucleotidase. Data were expressed as Mean + SEM, and analyzed using one-way ANOVA/SNK, P <0.05. Behavioral responses measured in the PM showed a temporal pattern in the number of PHD and NPHD, and time spent in open arms and closed. Data from OF showed only temporal pattern in the number of dung pats. Exposure to restraint stress was able to break the rhythm pattern observed in the behaviors evaluated in the PM and OF. In relation the systemic biomarkers was observed immediately after stress there is a loss of temporal pattern of corticosterone levels and 6 h after stress there is a phase delay of ZT0 to ZT18, whereas 24 hours after stress levels return to standard time normal; immediately after the stress showed a suppression of normal melatonin peak (ZT18), with the peak in the morning (ZT0), 6 and 24 hours after stress was a loss of temporal pattern. Glucose levels had the highest peak at ZT18 similar to melatonin. Observed a reversal of the rhythmic pattern associated with a decrease in ZT18, held until 6 hours after glucose levels immediately after stress, and the temporal pattern returned 24 hours after stress. In enzymatic activity, we observed that acute stress causes a decrease in activity of enzymes dependent nucleotidases time of day at which the animal is exposed to stress, and this effect seems to persist for at least 24 hours. The results of this dissertation show that the time of day which the experiment is performed biochemical and behavioral changes influencing the results, which may be a confounder in the interpretation of results.

The in vitro antimicrobial activity of advanced platelet rich fibrin (A-PRF) against microorganisms of the oral cavityBhamjee, Feheem January 2017 (has links)
Magister Chirurgiae Dentium - MChD (Oral Medicine and Periodontics) / In recent years, the development and use of autologous platelet rich concentrates (PC's) has
gained traction within the rapidly progressive, multidisciplinary field of regenerative
medicine. A PC subtype, marketed as advanced platelet rich fibrin (A- PRF), is a recent
advancement of the original PRF protocol and promoted as a "blood concentrate"
containing platelets, leukocytes, circulating stem cells and endothelial cells. A-PRF in the
form of membranes, plugs, or even shredded particulates are increasingly being used as
surgical adjuncts in areas of previous infection or left exposed within the microbial rich oral
environment. Although recent literature has noted the biologic benefits of this material
within the context of wound healing and regeneration, the antimicrobial potential of APRF
has remained unexplored. The aim of this investigation is to determine if A-PRF
displays antimicrobial activity against microbes of the oral cavity with a null hypothesis that
its activity is no different to a clot of unprocessed venous blood.
Methodology: A-PRF and whole blood samples were obtained from consenting individuals
and utilised to conduct an in-vitro agar disk diffusion investigation to determine their
antimicrobial activity. Standardised samples of A-PRF, unprocessed clotted blood and 0.2%
chlorhexidine gluconate (CHX) were tested against organisms cultured from fresh oral rinse
samples and pure cultures of candida albicans, streptococcus mutans, staphylococcus
aureus and enterococcus faecalis. The antimicrobial activity was assessed in accordance to
the established principles of the agar disk diffusion method and measurement of inhibition
zones.
Results: A-PRF displayed antimicrobial activity against all of the individual organisms
tested within this study following a 24 hour incubation period. However, no significant
differences were noted between A-PRF and a natural clot of blood when tested against
cultures of the oral rinse sample. Finally, the antimicrobial activity of A-PRF is significantly
inferior to an equal volume of the CHX preparation.
Conclusion: Although A-PRF displays antimicrobial activity; its strength, spectrum and
biologic activity within a polymicrobial environment requires further investigation.

The hemorrhagic syndrome caused by contact with Lonomia obliqua caterpillars has reached epidemic proportions in southern Brazil. This study aimed at assessing the efficacy of different dosages of an antilonomic serum (SALon) in the treatment of patients who had accidental contact with Lonomia obliqua caterpillars. A randomized, double-blinded, prospective clinical trial was conducted at São Vicente de Paulo Hospital, a tertiary hospital in Passo Fundo, Brazil. From January 2000 to April 2002, 44 patients with grade I or II hemorrhagic syndrome were randomly assigned to either group A (22 patients treated with 10,5mg total dose, 3 vials of SALon) or group B (22 patients treated with 17,5mg total dose, 5 vials of SALon). Treatment efficacy was assessed according to time necessary for blood coagulation to go back to normal, incidence of adverse reactions, and hospitalization time. There were no differences in clinical picture and laboratory findings between the two groups. Mean time for coagulation to go back to normal was 15.3±6.6 h and 19.1±8 h in groups A and B, respectively. Adverse reactions and hospitalization time were similar in both groups. SALon was effective and well tolerated. Results suggest that substantially smaller doses than those routinely prescribed may be sufficient for the treatment of envenoming by Lonomia caterpillars.

The pregnancy stage is very complex in the vascular point of view, occurring many changes and adjusts in the circulatory system, in the platelets functionality and in the endothelial system with the function of prevent future complications to the pregnant or to the newborn at the delivery and in the afterbirth. Besides the circulation changes, in the vascular processes and in the immune system that are inherent to this stage, there are studies that report the generation of reactive oxygen (ROS) and nitrogen species (RNS), which could trigger an imbalance in the redox status. The purine nucleotides ATP, ADP, AMP and the nucleoside adenosine participate in the endothelial vascular regulation, inflammatory events, immune regulation, among other functions. The control of the production, the intracellular and extracellular levels of these nucleotides and nucleosides are regulated by an enzymatic cascade, in which, participate the enzymes E-NTPDases (ectonucleotise triphosphate diphosphohydrolases), E-NPPs (ectonucleotide pyrophosphatase/phosphodiesterases), 5’-nucleotidase and adenosine deaminase. The purpose of this study was to investigate the activity of enzymes involved in the hemostatic dynamics from normal pregnant and those with complications, such as the gestational diabetes mellitus, the pre-eclampsia and infected with the HIV. We also investigated some oxidative stress parameters and the platelets aggregation during the third gestational trimester. The results demonstrated an increase in the enzymes NTPDase and 5’-nucleotidase in platelets from normal pregnant and those with gestational diabetes mellitus and pre-eclampsia compared with not-pregnant women. It was also observed an increase in the enzyme adenosine deaminase activity in the serum from normal pregnant and those with gestational diabetes mellitus, pre-eclampsia and infected with the HIV. It was also verified an increase in the platelets activity through the measure of platelets aggregation in the normal pregnant, those with gestational diabetes mellitus, pre-eclampsia and infected with the HIV. Otherwise, it was verified an increase in the activity of the enzymes superoxide dismutase, catalase, butyrylcholinesterase and acetylcholinesterase together with an increase in the lipid peroxidation and a decrease in the ascorbic acid and total sulphydryl groups content. Moreover, it was not observed any significant alteration in the protein carbonilation. The results showed here suggest that these ectonucleotidases participate in the modulation of vascular, inflammatory, immune and circulatory responses. Finally, it was detected an increase in the activity of antioxidant enzymes and a decrease in the antioxidant molecules content, together with an increase in the membrane lipids oxidative damage, which brings about an increased production of reactive species and a struggle of the body to combat this unbalance.

Concurrent strength and endurance training have been widely investigated in many populations. However, few studies have compared its effects with strength and endurance training separately. Thus, the aim of the present study was to investigate the effects of concurrent training on muscle strength and activation, endurance capacity and hormonal concentrations in elderly men. Twenty-three healthy men (65 ± 4 years) were matched in 3 groups: concurrent (CG, n=8), strength (SG, n=8) or aerobic training group (AG, n=7). Each one trained 3 times a week during 12 weeks strength, aerobic in cycle ergometer or both in the same session. Before and after training period, subjects were evaluated in parameters related to muscle strength, dynamic and isometric muscle activation, endurance capacity and serum hormones. There were significant increases on lower-body strength in all groups (P<0.05), with higher increases in SG (67%) than CG (41%) and both higher than AG (25%) (both P<0.01). Only SG and CG increased the upper-body strength (P<0.01). There were significantly increases in isometric strength and maximal muscle activation only in SG (P<0.05), as decreases in isometric submaximal muscle activation to the same load in SG (P <0.05) after training in the muscles evaluated. Indeed, only CG and AG have increases endurance capacity (P<0.05), with no differences between these groups, and both CG and AG decreased the dynamic muscle activation in rectus femoris to the same power after training. In addition, there were significant decreases on free testosterone in AG after training. The present results suggested that the different types of training resulting in different adaptations in performance variables, as like as neuromuscular and endocrine parameters in elderly subjects. The interference effect observed to concurrent training could be related with impairment of neural adaptations.
