Avaliação da atividade de nucleotidases na hemostasia e parâmetros de estresse oxidativo em mulheres grávidas normais, com diabetes melito gestacional, pré-eclampsia e portadoras do vírus HIV

O período gestacional é bastante complexo e do ponto de vista vascular, ocorrem inúmeras alterações e adaptações no sistema circulatório, na funcionalidade plaquetária e no sistema endotelial com o objetivo de prevenir futuras complicações na mãe ou no concepto no momento do parto e após o nascimento. Além das alterações na circulação, nos processos vasculares e no sistema imune que são inerentes a este período, existem trabalhos demonstrando a geração de espécies reativas de oxigênio (EROs) e de nitrogênio (ERNs) as quais poderão desencadear um desequilíbrio do balanço redox. Os nucleotídeos de purina ATP, ADP, AMP, além do nucleosídeo adenosina participam da regulação vascular endotelial, de processos inflamatórios e do controle imunológico entre outras funções. O controle da produção e dos níveis intracelulares e extracelulares destes nucleotídeos e nucleosídeos são regulados por uma cascata enzimática, na qual, participam as enzimas E-NTPDases (Ecto-Nucleosídeo Trifosfato Difosfoidrolase), ENPPs (Ecto-Nucleotídeo Pirofosfatase Fosfodiesterase), 5`-Nucleotidase e adenosina desaminase. O objetivo deste trabalho foi investigar a atividade de enzimas envolvidas no processo hemostático de gestantes normais e com complicações, como a diabetes melito gestacional, a pré-eclampsia e contaminadas com o vírus HIV, além de alguns parâmetros de estresse oxidativo e a agregação plaquetária durante o terceiro trimestre gestacional. Os resultados obtidos demonstram um aumento na atividade das enzimas NTPDase e 5`-nucleotidase em plaquetas de gestantes normais e com diabetes melito gestacional e pré-eclampsia comparadas com mulheres não grávidas. Ocorreu um aumento na atividade da enzima adenosina desaminase no soro de gestantes normais e com diabetes melito gestacional, pré-eclampsia e contaminadas com o vírus HIV. Foi verificado, também, um aumento na atividade plaquetária, através da medida da agregação plaquetária, nas gestantes normais e com diabetes melito gestacional, pré-eclampsia e contaminadas com o vírus HIV. Por outro lado, foi verificado um aumento na atividade das enzimas superóxido dismutase, catalase, butirilcolinesterase e acetilcolinesterase juntamente com um aumento da peroxidação lipídica e uma redução dos níveis de ácido ascórbico e conteúdo sulfidrílico total. Além disso, não foi encontrada alteração significativa na análise de carbonilação protéica. Os resultados apresentados aqui sugerem que estas ectonucleotidases atuam na modulação de respostas vasculares, inflamatórias, imunes e circulatórias. Finalmente, foi detectado um aumento na atividade de enzimas antioxidantes e uma redução no conteúdo de moléculas antioxidantes, acompanhadas de um aumento no dano oxidativo aos lipídios de membrana o que remete à uma produção aumentada de espécies reativas e a uma tentativa do organismo em combater este desequilíbrio. / The pregnancy stage is very complex in the vascular point of view, occurring many changes and adjusts in the circulatory system, in the platelets functionality and in the endothelial system with the function of prevent future complications to the pregnant or to the newborn at the delivery and in the afterbirth. Besides the circulation changes, in the vascular processes and in the immune system that are inherent to this stage, there are studies that report the generation of reactive oxygen (ROS) and nitrogen species (RNS), which could trigger an imbalance in the redox status. The purine nucleotides ATP, ADP, AMP and the nucleoside adenosine participate in the endothelial vascular regulation, inflammatory events, immune regulation, among other functions. The control of the production, the intracellular and extracellular levels of these nucleotides and nucleosides are regulated by an enzymatic cascade, in which, participate the enzymes E-NTPDases (ectonucleotise triphosphate diphosphohydrolases), E-NPPs (ectonucleotide pyrophosphatase/phosphodiesterases), 5’-nucleotidase and adenosine deaminase. The purpose of this study was to investigate the activity of enzymes involved in the hemostatic dynamics from normal pregnant and those with complications, such as the gestational diabetes mellitus, the pre-eclampsia and infected with the HIV. We also investigated some oxidative stress parameters and the platelets aggregation during the third gestational trimester. The results demonstrated an increase in the enzymes NTPDase and 5’-nucleotidase in platelets from normal pregnant and those with gestational diabetes mellitus and pre-eclampsia compared with not-pregnant women. It was also observed an increase in the enzyme adenosine deaminase activity in the serum from normal pregnant and those with gestational diabetes mellitus, pre-eclampsia and infected with the HIV. It was also verified an increase in the platelets activity through the measure of platelets aggregation in the normal pregnant, those with gestational diabetes mellitus, pre-eclampsia and infected with the HIV. Otherwise, it was verified an increase in the activity of the enzymes superoxide dismutase, catalase, butyrylcholinesterase and acetylcholinesterase together with an increase in the lipid peroxidation and a decrease in the ascorbic acid and total sulphydryl groups content. Moreover, it was not observed any significant alteration in the protein carbonilation. The results showed here suggest that these ectonucleotidases participate in the modulation of vascular, inflammatory, immune and circulatory responses. Finally, it was detected an increase in the activity of antioxidant enzymes and a decrease in the antioxidant molecules content, together with an increase in the membrane lipids oxidative damage, which brings about an increased production of reactive species and a struggle of the body to combat this unbalance.

Diagnostic and Prognostic Capacity of Serum Glycan Nodes in Different Types of Cancer

abstract: Glycans are monosaccharide-based heteropolymers that are found covalently attached to many different proteins and lipids and are ubiquitously displayed on the exterior surfaces of cells. Serum glycan composition and structure are well known to be altered in many different types of cancer. In fact, glycans represent a promising but only marginally accessed source of cancer markers. The approach used in this dissertation, which is referred to as “glycan node analysis”, is a molecularly bottom-up approach to plasma/serum (P/S) glycomics based on glycan linkage analysis that captures features such as α2-6 sialylation, β1-6 branching, and core fucosylation as single analytical signals. The diagnostic utility of this approach as applied to lung cancer patients across all stages as well as prostate, serous ovarian, and pancreatic cancer patients compared to certifiably healthy individuals, nominally healthy individuals and/or risk-matched controls is reported. Markers for terminal fucosylation, α2-6 sialylation, β1-4 branching, β1-6 branching and outer-arm fucosylation were most able to differentiate cases from controls. These markers behaved in a stage-dependent manner in lung cancer as well as other types of cancer. Using a Cox proportional hazards regression model, the ability of these markers to predict progression and survival in lung cancer patients was assessed. In addition, the potential mechanistic role of aberrant P/S glycans in cancer progression is discussed. Plasma samples from former bladder cancer patients with currently no evidence of disease (NED), non-muscle invasive bladder cancer (NMIBC), and muscle invasive bladder cancer (MIBC) along with certifiably healthy controls were analyzed. Markers for α2-6 sialylation, β1-4 branching, β1-6 branching, and outer-arm fucosylation were able to separate current and former (NED) cases from controls; but NED, NMIBC, and MIBC were not distinguished from one another. Markers for α2-6 sialylation and β1-6 branching were able to predict recurrence from the NED state using a Cox proportional hazards regression model adjusted for age, gender, and time from cancer. These two glycan features were found to be correlated to the concentration of C-reactive protein, a known prognostic marker for bladder cancer, further strengthening the link between inflammation and abnormal plasma protein glycosylation. / Dissertation/Thesis / Doctoral Dissertation Chemistry 2018

Oxidative Stress and a High Fat Diet in Rats: An Intervention Study on the Effects of an Organometallic Compound on Enzyme Function, Inflammatory Markers, Endotoxins and Fasting Serum Glucose and Insulin Levels

abstract: Cardiovascular disease has reached epidemic proportions resulting in its ranking as the number one cause of mortality in the Western world. A key player in the pathophysiology of vascular disease is oxidative stress due to free radical accumulation. This intervention study was conducted to evaluate any potential mediation of oxidative stress using a soil-derived organometallic compound (OMC) with suspected antioxidant properties. A 10-week study was conducted in male Sprague-Dawley rats (n = 42) fed either a high-fat diet (HFD) consisting of 60% kcal from fat or a standard Chow diet containing only 6% kcals from fat. Rats from each diet group were then subdivided into 3 subgroups (n = 6-10 each) that received 0.0 mg/mL, 0.6 mg/mL or 3.0 mg/mL OMC. Neither the diet nor OMC significantly changed protein expression of inducible nitric oxide synthase (iNOS) in isolated aortas. Plasma levels of the inflammatory marker, tumor necrosis factor alpha (TNFα) were below detection after the 10-week trial. Superoxide dismutase (SOD), a scavenger of the free radical, superoxide, was not significantly different following HFD although levels of SOD were significantly higher in Chow rats treated with 0.6 mg/mL OMC compared to HFD rats treated with the same dose (p < 0.05). Lipopolysaccharides (LPS) were significantly increased following 10 weeks of high fat intake (p < 0.05). This increase in endotoxicity was prevented by the high dose of OMC. HFD significantly increased fasting serum glucose levels at both 6 weeks (p < 0.001) and 10 weeks (p < 0.025) compared to Chow controls. The high dose of OMC significantly prevented the hyperglycemic effects of the HFD in rats at 10 weeks (p = 0.021). HFD-fed rats developed hyperinsulinemia after 10 weeks of feeding (p = 0.009), which was not prevented by OMC. The results of this study indicate that OMC may be an effective strategy to help manage diet-induced hyperglycemia and endotoxemia. However, further research is needed to determine the mechanism by which OMC helps prevent hyperglycemia as measures of inflammation (TNFα) and vascular damage (iNOS) were inconclusive. / Dissertation/Thesis / Masters Thesis Nutrition 2018

Foi feita a caracterização eletroquímica do aço inoxidável austenítico UNS S31254 em meio de NaCl 0,11 mol L-1 na ausência e presença de soro albumina bovina (BSA) visando seu emprego em implantes ortopédicos. Foram empregadas como técnicas: medidas de potencial de circuito aberto, curvas de polarização, cronoamperometria, EIE, XPS, MEV, EDS e EEO. O comportamento eletroquímico do aço 254 foi comparado com o de outros aços empregados em implantes ortopédicos (ISO 5832-9, ASTM F138, e AISI 316L) na ausência e presença de BSA. O aço 254 se mostrou semelhante ao ISO 5832-9: encontra-se passivado desde o potencial de corrosão até o de transpassivação; a presença de inclusões de óxidos de cálcio e alumínio no aço 254 foi considerada a responsável por um potencial de transpassivação 100 mV menos positivo do que o observado com o aço ISO 5832-9. Foi detectada. além de óxido de Cr(III), a presença de Mo na forma Mo(VI) no filme passivo do aço 254. A ação da BSA, ora passivante ora catalisadora, depende de sua concentração, da natureza do substrato metálico, e do potencial na interfase metal-solução. A BSA modifica o mecanismo de oxidação do aço 254 e inibe seletivamente a dissolução dos seus elementos constituintes, em particular, níquel e cromo. / The electrochemical characterization of UNS S31254 has been made in 0.11 mol L-1 NaCl medium in the absence and presence of bovine serum albumin (BSA) in order to propose its application in orthopaedic implants. The techniques employed were: open circuit potential measurements, polarization curves, chronoamperometry, EIS, XPS, SEM, EDS and EEO. The electrochemical behavior of 254 SS was compared to that observed for ISO 5832-9, ASTM F138 and AISI 316L stainless steels, used in orthopedic implants, in the absence and presence of BSA. 254 SS is similar to ISO 5832-9 SS: it is passivated on the potential range between the corrosion and the transpassivation potentials; the presence of calcium and aluminum oxides can be responsible for the shift of about 100 mV to less positive potentials on the transpassivation potential when compared to ISO 5832-9 SS. The presence of Mo(VI) was detected beside Cr(III) as passivating film for 254 SS. BSA action depends on its concentration, the nature of the metallic substract and on the potential in the metal-solution interphase. BSA changes the oxidation mechanism of 254 SS and promotes the selective dissolution of the elements particularly nickel and chromium.

A coleta não-traumática é atrativa especialmente para crianças e quando repetidas coletas são requeridas. A desvantagem da saliva como uma ferramenta de diagnóstico é a variabilidade, que ocorre em decorrência dos fatores fisiológicos e do modo de coleta. Numa primeira etapa o trabalho teve por objetivo estudar as atividades das enzimas fosfatase alcalina (FAlc), fosfatase ácida total (FAT) e fosfatase ácida resistente ao tartarato (TRAP) na saliva total não estimulada de crianças, investigando as influências de fatores como sexo, faixa etária e horário de coleta, bem como a correlação das enzimas com a taxa de fluxo salivar. Nesta etapa, 120 crianças saudáveis de ambos os sexos, nas faixas etárias de 1-5 e 6-12 anos de idade tiveram as amostras de saliva coletada entre 8:00-10:00 horas ou entre 14:00-16:00 horas. A segunda parte do trabalho teve por objetivo avaliar a correlação das atividades enzimáticas da FAT, TRAP, proteína tirosina fosfatase de baixa massa molecular relativa (PTP-BMr) e FAlc na saliva e no soro de crianças, além de analisar a influência do sexo e da faixa etária na atividade das enzimas, no soro e na saliva total. Para tanto 32 crianças de ambos os sexos, nas faixas etárias de 1-5 anos e 6-12 anos de idade tiveram as amostras de saliva e sangue coletadas entre 8:00-10:00 horas. Os resultados da primeira etapa sugerem que as atividades da FAT, TRAP e FAlc são influenciadas, de formas distintas, pelos fatores sexo, faixa etária e horário de coleta e que não existe correlação entre as atividades das enzimas e a taxa de fluxo salivar. / Saliva can be collected by simple, non-invasive methods and is easily stored. The non-traumatic collection is specially appealing for children and when repeated collections are required. The main disadvantage of the saliva as a diagnosis tool is the variability that happens due to the physiologic factors and dependence on mode of the collection. In a first stage the work had for objective to study the activities of the enzymes alkaline phosphatase (ALP), total acid phosphatase (TAP) and tartrate-resistant acid phosphatase (TRAP) in the whole unstimulated saliva of children, investigating the influences of factors as sex, age group and time of collection, as well as the correlation of the enzymes with the salivary flow rate. In this stage, 120 healthy children of both sexes, in the age groups of 1-5 and 6-12 years of age had the saliva samples collected between 8:00-10:00 hours or between 14:00-16:00 hours. In a second stage, the work had for objective to evaluate the correlation of the enzymatic activities of TAP, TRAP, low molecular weight protein tyrosine phosphatase (LMW-PTP) and ALP in the saliva and in the children's serum, besides analyzing the influence of the sex and of the age group in the activity of the enzymes, in the serum and in the whole unstimulated saliva. For so much 32 children of both sexes, in the 1-5 year-old age groups and 6-12 years of age had the saliva samples and blood collected between 8:00-10:00 hours. The results of the first stage suggest that the activities of TAP, TRAP and ALP are influenced, in different ways, for the factors sex, age group and time of collection and that correlation doesn't exist between the activities of the enzymes and the salivary flow rate.

The serum is considered na important coproduct for the dairy industry, configuring about 85-90% of the total milk volume, its proteins are recognized by their biological properties, since they have an excellent composition and bioavailability of essential amino acids. The present study had as objective to characterize the serum protein profile of goat colostrum produced by goats Created in Paraíba and to evaluate possible biological activities. The serum (SR) was obtained by acid precipitation in pH 4.1 and fractionated by ammonium sulfate precipitation to obtain the fractions 0-30% (SF1), 30- 60% (SF2) and 60-90% (SF3). The analysis of the protein profile was verified by SDS-PAGE and total amino acids. The biological properties of the samples were verified with the analysis of antioxidant, DNA protection and antibacterial activities. It was verified that colostrum serum presents proteins with molecular masses between 12-160kDa and an excellent profile of amino acids. From the obtained results it is possible to affirm that all the fractions were able to sequester the radicals ABTS and DPPH. For the DNA protection test, the concentracion of 10μg of SF3 conferred a greater integrity to the molecule. The samples presented inhibitory activity for both gram-negative and gram-positive bacteria. Colostrum serum proteins have biological properties with potential biotechnological application and pharmaceutical interest / O colostro oriundo das primeiras secreções das glândulas mamárias é considerado um alimento essencial para o desenvolvimento dos recém-nascidos, devido seu elevado teor de proteínas, peptídeos antimicrobianos, macro e micronutrientes. O soro é considerado um coproduto importante para a indústria leiteira, configurando cerca de 85–90% do volume total do leite. Suas proteínas são reconhecidas pelas propriedades biológicas, uma vez que possuem uma excelente composição e biodisponibilidade de aminoácidos essenciais. O presente estudo teve como objetivo caracterizar o perfil proteico do soro de colostro caprino produzido por cabras criadas na Paraíba e avaliar possíveis atividades biológicas. O soro (SR) foi obtido por precipitação ácida em pH 4,1 e fracionado por precipitação com sulfato de amônio para obtenção das frações 0-30% (SF1), 30-60% (SF2) e 60-90% (SF3). A análise do perfil proteico foi verificado por SDS-PAGE e aminoácidos totais. As propriedades biológicas das amostras foram verificadas com as análises das atividades antioxidante, proteção ao DNA e antibacteriana. Verificou-se que o soro do colostro apresenta proteínas com massas moleculares entre 12-160kDa e um excelente perfil de aminoácidos. A partir dos resultados obtidos é possível afirmar que todas as frações foram capazes de sequestrar os radicais ABTS e DPPH. Para o teste de proteção ao DNA, a concentração de 10μg de SF3 conferiu uma maior integridade à molécula. As amostras apresentaram atividade inibitória tanto para as bactérias gramnegativas quanto para gram-positivas. As proteínas do soro do colostro apresentam propriedades biológicas com potencial aplicação biotecnológica e de interesse farmacêutico.

A dosagem do hCG é o parâmetro mais importante para detecção da persistência de trofloblasto ativo na parede do útero ou em outros locais do organismo. Objetivo: Comparar concentrações séricas do hormônio hCG em pacientes com DTG utilizando duas variações do método de quimioluminescência comercialmente disponíveis em nosso centro. Métodos: Este estudo incluiu pacientes com DTG avaliadas e acompanhadas no Centro de Doenças Trofoblásticas da Faculdade de Medicina de Botucatu (CDTB) – UNESP, de novembro de 2014 a outubro de 2015. Amostras de soro das pacientes foram testadas para dosagem de hCG em duas variações do método de quimioluminescência ARCHITECT® i2000 SR e IMMULITE® 2000 Xpi. Concentrações séricas de hCG foram comparadas contra a hipótese nula e a concordância clínica foi determinada em dois momentos: admissão da paciente e evolução pela curva de hCG de acordo com os valores de hCG entre os dois equipamentos. Resultados: 73 pacientes com DTG foram incluídas no estudo. Destas, 45 tinham MH e remissão espontânea, enquanto 28 tiveram NTG. Uma boa concordância nos valores médios de hCG entre IMMULITE® 2000 Xpi e ARCHITECT® i2000 SR quando hCG <100 mUI/mL. Para valores de hCG >100 mUI/mL, houve diferença significativa entre ensaios (p<0,05), com valores medidos pelo ARCHITECT® i2000 SR sendo maior que aqueles medidos pelo IMMULITE® 2000 Xpi em pacientes com MH e remissão espontânea ( Immulite = 0.79Architect ; p < 0,01 ) ; R2 = 90 % ) e NTG ( Immulite = 0,51Architect ; p < 0,01 ) ; R2 = 98 % ). As condutas clínicas no momento da admissão das pacientes foram concordantes em 100% dos casos [73/73(100%); kappa1; p<0,001] e pela evolução da curva de hCG, a concordância observada foi de 98% [71/72(98%); kappa 0,93; p<0,001].Conclusão: O uso do equipamento IMMULITE® é recomendado para monitoramento de pacientes com DTG, entretanto, ARCHITECT® apresentou desempenho analítico e coerência clínica em relação aos resultados de hCG, o que faz deste equipamento uma boa alternativa. / Introduction: Gestational trophoblastic disease (GTD) is a group of disorders spanning the conditions that range from hydatidiform mole (HM) to gestational trophoblastic neoplasia (GTN). Determining human chorionic gonadotrophin (hCG) serum levels is crucial for the early detection and optimal management of GTN. Objective: To compare serum hCG levels in patients with GTD using 2 commercially available hCG immunoassays. Methods: Serum samples were obtained from patients with GTD attending the Botucatu Trophoblastic Diseases (Botucatu Medical School, São Paulo State University- Unesp) from November 2014 to October 2015. Serum hCG levels were measured with both IMMULITE® 2000 XPi and ARCHITECT® i2000 chemiluminescence tests. Serum hCG levels were compared against the null hypothesis. Agreement with clinical management was determined by comparing baseline hCG measures as well as the hCG curves obtained with both assays. Results: Seventy-three patients with GTD were included in the analysis. Of these, 45 had HM and experienced spontaneous remission, while 28 had GTN. There was a perfect (zero difference) agreement in mean hCG levels between IMMULITE® 2000 XPi and ARCHITECT® i2000 when hCG < 100 mUI/mL. For hCG values greater than 100 mUI/mL, there was a significant difference between assays (p < 0.05), with levels measured via ARCHITECT® i2000 being higher than those measured with IMMULITE® 2000 XPi in patients with spontaneous remission (Immulite = 0.79Architect; p < 0.01); R2 = 90%) and GTN (Immulite = 0,51Architect; p < 0,01); R2 = 98%). Baseline test results agreed in all cases (73/73(100%); kappa1;p<0,001] and hCG curve agreement was 98% [71/72(98%); kappa0,93;p<0,001].Conclusion: IMMULITE® has been the assay recommended for diagnosing and monitoring patients with GTD. However, our results suggest that as ARCHITECT® and IMMULITE® show similar performance in measuring hCG levels and determining clinical management, ARCHITECT® may be used an alternative.
