Aspects influencing the release and establishment of the flowerbud weevil, Anthonomus santacruzi Hustache (Coleoptera : Curculionidae), a biological control agent for Solanum mauritianum scopoli (Solanaceae) in South Africa.

Solanum mauritianum (bugweed, woolly nightshade) is a perennial tree native to South America that has invaded many countries including South Africa and New Zealand. In South Africa, after 143 years of naturalization, the plant is ranked as the country‟s sixth worst weed and has invaded 1.76 million ha. Invaded areas include agricultural lands, forest plantations, water courses and conservation areas, especially in the eastern higher rainfall regions. The success of the spread of this weed is due to its production of very high numbers of bird-dispersed seeds. Since conventional control methods are unsustainable in the long term, the weed has been targeted for classical biological control since 1984. Following exploration work in its native range, biological control experts recommended that agents that are able to limit the weed‟s reproductive potential would help to manage the spread and invasiveness of this weed. Anthonomous santacruzi, a flower-feeding weevil found throughout the native range of the weed, was imported and tested between 1998 and 2002. Following approval for its release in South Africa in 2007, a new colony was imported and propagated at the University of KwaZulu-Natal Pietermaritzburg. This study was initiated to investigate aspects that could influence the release and establishment of this agent. Three aspects were investigated namely: (1) reassessing the weevil's host range to confirm that the new colony is not different from the colony tested originally and to assess the risks associated with the release of the weevil in New Zealand; (2) surveying the arthropods associated with S. mauritianum in the field to identify groups of predators that could interfere with the establishment of the weevils as well as to investigate, through laboratory-based trials using spiders as surrogate, the impact of these predators on the survival and proliferation of the weevils; and (3) propagation and release of the weevil and monitoring of its establishment. Host-specificity tests revealed that the host range of new colony is not different from that of the originally tested culture. In no-choice trials, the weevils fed and reproduced on some non-target Solanaceae species but reverted back to S. mauritianum in the choice tests. Although the risks for releasing the weevils in New Zealand were calculated to be very low, additional evidence is needed to demonstrate this conclusively. Future research to provide this evidence includes open-field trials complemented with a chemical ecology study, to resolve the case of two species, a New Zealand native and South African native, which have shown higher risks in comparison to the other tested species. For arthropods associated with S. mauritianum in the field, Araneae (especially Thomisidae), Thysanoptera, Hemiptera (especially Miridae) and Hymenoptera (especially Formicidae) were identified as generalist predators that could interfere with the establishment of A. santacruzi. However, their numbers in the field appear to be too low to provide a major threat. Also, laboratory trials using spiders as a surrogate suggested that A. santacruzi populations can survive and reproduce in the presence of such predators. The weevils were released at four sites in KwaZulu-Natal and monitoring of three of these has confirmed establishment at the warmest site along the South Coast but not at the coldest site in the Midlands. Titânio via foliar no metabolismo, absorção de nutrientes e produtividade de batata / Titanium foliar spray in metabolism, nutrient uptake and potato productivity

The potato (Solanum tuberosum L.) has great importance in human nutrition where it occupies the fourth place among the most consumed foods in the world. It is a highly demanding culture technology especially in the nutritional point of view. Despite the titanium (Ti) is not considered a nutrient, studies have shown beneficial effects of this element when applied to plants. The application of Ti leaf can be an alternative for this element is slightly movable in the soil and is generally present in insoluble forms. Thus, the objective was to evaluate the influence of application rates of Ti foliar in the potato crop. We conducted an experiment in the period from August to November 2014, installed at the experimental station Udi Pesquisa e Development in Uberlândia-MG, with kind Ágata. Variables related to metabolism were evaluated: SPAD chlorophyll content, nitrate reductase activity (NRA), lipid peroxidation (LP), urease, proline, catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD). Nutritional factors such as levels of nutrients nitrogen (N), copper (Cu), iron (Fe), manganese (Mn) and zinc (Zn) present in leaves and tubers were measured stages of growth, tuber and tuber filling. At the end of the crop cycle was quantified the average mass, diameter, commercial classification and productivity of tubers. The design was a randomized block design with four replications and six treatments. The supply of the Ti compound fertilizer was through for 5% of magnesium oxide, 10% sulfur trioxide and 0,85% titanium complex and the treatments was: T1 - without the application of Ti, T2 -10.2; T3 - 15.3; T4 - 20.4; T5 - 22.9 and T6 - 25.5 g Ti ha-1 divided into three equal applications during the growth phase, tuber and tuber filling. It was concluded that in the growth phase the foliar application of Ti reduces the Mn content in the leaves and there is increased activity of POD and ANR and reduced urease activity. In the tuberization phase the Fe absorption, SOD activity and POD response to increasing Ti dose. In tubers filling stage foliar application of Ti increases chlorophyll levels (Spad value). The application of Ti in the leaf growth stages, tuber and tuber filler results in a lower Zn content in the tubers. The average tuber weight, the average diameter of tubers and the total productivity of tubers are affected by foliar application of Ti. The dose Ti foliar applied for further tuber yield is 5.74 g Ti ha-1. / A batata (Solanum tuberosum L.) tem grande importância na alimentação humana onde ocupa o quarto lugar entre os alimentos mais consumido no mundo. É uma cultura altamente exigente em tecnologias especialmente sob o ponto de vista nutricional. Apesar do titânio (Ti) não ser considerado um nutriente, trabalhos demonstram efeitos benéficos deste elemento quando aplicado às plantas. A aplicação de Ti foliar pode ser uma alternativa pois este elemento é pouco móvel no solo e geralmente está presente em formas insolúveis. Desta forma, objetivou-se avaliar a influência da aplicação de doses de Ti via foliar na cultura da batata. Realizou-se um experimento no período de agosto a novembro de 2014, instalado na estação experimental Udi Pesquisa e Desenvolvimento em Uberlândia-MG, com utilização da cultivar Ágata. As variáveis relacionadas ao metabolismo avaliadas foram: teor clorofila SPAD, atividade da nitrato redutase (ANR), peroxidação lipídica (PL), urease, prolina, catalase (CAT), superóxido desmutase (SOD) e peroxidase (POD). Aspectos nutricionais como os teores dos nutrientes nitrogênio (N), cobre (Cu), ferro (Fe), manganês (Mn) e zinco (Zn) presentes em folhas e tubérculos foram mensurados fases de crescimento, tuberização e enchimento de tubérculos. Ao final do ciclo da cultura foi quantificada a massa média, diâmetro médio, classificação comercial e a produtividade dos tubérculos. O delineamento foi o de blocos casualizados, com quatro repetições e seis tratamentos. O fornecimento do Ti foi através de fertilizante composto por 5% de óxido de magnésio, 10% de trioxido enxofre e 0,85% de complexo de titânio com os seguintes tratamentos: T1 sem aplicação de Ti, T2 -10,2; T3 -15,3; T4 - 20,4; T5 - 22,9 e T6 - 25,5 g de Ti ha-1 divididos em três aplicações iguais durante a fase de crescimento, tuberização e enchimento de tubérculos. Concluiu-se que os teores de Cu não são influenciados pelas doses de Ti nas fases avaliadas. Na fase de crescimento a aplicação foliar de Ti reduz o teor de Mn nas folhas e há aumento da atividade da POD e ANR e redução da atividade da urease. Na fase de tuberização a absorção de Fe e atividade de enzimas SOD e POD tem resposta ao aumento da dose de Ti. Na fase de enchimento de tubérculos a aplicação foliar de Ti incrementa os teores de clorofila (valor Spad). A aplicação de Ti foliar nas fases de crescimento, tuberização e enchimento de tubérculos resulta em menor teor de Zn nos tubérculos. A massa média de tubérculos, o diâmetro médio de tubérculos e a produtividade total de tubérculos são influenciados pela aplicação foliar de Ti. A dose de Ti aplicada via foliar para maior produtividade de tubérculos é de 5,74 g de Ti ha-1. / Mestre em Agronomia

Para avaliação do híbrido Débora Victory, os tratamentos foram constituídos por nove épocas de amostragem, aos 2, 32, 46, 60, 77, 91, 109, 122 e 137 dias após o transplante (DAT) realizado em 01/04/2014. As plantas atingiram acúmulo estimado de massa seca e fresca total na última coleta de 550,2 e 9.528,2 g planta-1, respectivamente. A ordem decrescente do acúmulo total de nutrientes foi K > N > Ca > P > S > Mg, com valores estimados de 22,6; 10,4; 5,0; 2,3; 2,1 e 1,6 g planta-1, respectivamente. Na última coleta, o maior acúmulo de N, P, K ocorreu nos frutos e de Ca, S e Mg na parte vegetativa (caule+folhas+ inflorescências). O estudo da composição dos biofertilizantes foi constituído por seis biofertilizantes, elaborados com a mesma base para a fermentação (composta de 1 kg de farelo de arroz, 250 mL de melaço, 250 mL de microorganismos, marca EMBIOTIC, misturados em 15 litros de água), diferindo entre si pelo acréscimo de um ingrediente: T1 = torta de mamona (2 kg); T2 = farinha de sangue (2 kg); T3 = pó de casco e chifres (2 kg); T4 = farinha de ossos (2 kg); T5 = farinha de sangue (1 kg); T6 = farinha de ossos (1 kg) e cinco momentos de avaliação, aos 3, 7, 15, 22 e 30 dias após o preparo (DAP) dos biofertilizantes. Em cada momento foram analisados os teores de N, P e K e a condutividade elétrica. O biofertilizante a base de pó de casco e chifres apresentou maior teor de N do que os outros, alcançando o teor de 6,8 g L-1 aos 30 DAP. Os biofertilizantes a base de torta de mamona e pó de casco e chifres apresentaram maior teor de P, com médias de 3,2 e 2,6 g L-1, respectivamente, do que os de farinha de ossos (1 e 2 kg). O biofertilizante a base de torta de mamona apresentou maior teor de K, média de 2,5 g L-1, do que todos os demais, que foram iguais entre si. Independente do tipo de biofertilizante, os teores de K não se alteraram, os de N e a condutividade elétrica aumentaram e os de P reduziram ao longo do tempo de fermentação até os 30 DAP. / The objective of this research was to study the growth curve and absorption of nutrients in tomato 'Debora Victory', grown in organic system field in Itápolis-SP and to study the contents of N, P and K of six biofertilizers during the process fermentation. To evaluate the Débora Victory hybrid, the treatments consisted of nine sampling times at 2, 32, 46, 60, 77, 91, 109, 122 and 137 days after transplanting (DAT) held on 01.04.2014. The plants had estimated total dry and fresh weight, in the last sampling, of 550.2 and 9528.2 g plant-1, respectively. The descending order of total nutrient accumulation was K> N> Ca> P> S> Mg, with estimates of 22.6; 10.4; 5.0; 2.3; 2.1 and 1.6 g plant-1, respectively. In the last sampling, the greater accumulation of N, P, K occurred in fruits and Ca, Mg and S in the vegetative parts (stem + leaves + inflorescences). The experiment consisted of six biofertilizers, prepared on the same basis for fermentation (composed of 1 kg of rice bran, 250 ml of molasses, 250 ml of microorganisms, brand EMBIOTIC, mixed in 15 liters of water), differing from each other by an ingredient addition: T1 = castor bean cake (2 kg), T2 = blood meal (2 kg); T3 = hoof and horn powder (2 kg), T4 = bone meal (2 kg), T5 = blood meal (1 kg); T6 = bone meal (1 kg)) and five dates of evaluation: 3, 7, 15, 22 and 30 days after preparation (DAP) of biofertilizers. The following variables were analyzed: contents of N, P and K and the electrical conductivity. The hoof and horn powder biofertilizer showed higher N content than the others, reaching a maximum of 6.8 g L-1 at 30 DAP. The castor bean cake and hoof and horn powder, showed higher P contents, with average of 3.2 and 2.6 g L-1, respectively, than bone meal (1 and 2 kg) biofertilizer. The castor bean cake biofertilizer showed higher K content, average of 2.5 g L-1, than the others that did not differ each other. Regardless of the type of biofertilizer, the K content did not change, the N content and the electrical conductivity increased and the P content reduced during the time of fermentation until 30 DAP.

O reúso planejado é vastamente utilizado em todo o mundo, entretanto, no Brasil, mesmo possuindo regiões com escassez hídrica, essa prática ainda é pouco difundida. A berinjela (Solanum melongena L.) é uma hortaliça que possui grande importância no mercado de olerícolas no Brasil e no mundo. Objetivou-se com esta pesquisa avaliar a viabilidade técnica do uso de nitrogênio e fósforo juntamente com a irrigação utilizando efluente doméstico pós-tratado em filtro de areia com fluxo intermitente sob o crescimento, produção de fitomassa e floração de plantas de berinjela cultivada em ambiente protegido na região semiárida paraibana. O experimento foi realizado em ambiente protegido, no Centro de Ciências e Tecnologia Agroalimentar (CCTA/UFCG), Campus de Pombal – PB, e cujos tratamentos resultaram na combinação de dois fatores: quatro doses de adubação nitrogenada (N1 - 3,55; N2 - 6,2; N3 - 8,9; N4 - 11,55 g de N/vaso) e quatro doses de adubação fosfatada (P1 - 15,28; P2 - 26,74; P3 - 38,2; P4 - 49,66 g de P/vaso) correspondendo respectivamente a 40; 70; 100; 130% conforme indicação de adubação para a cultura da berinjela quando cultivada em vasos, irrigada com água residuária pós-tratada em filtro de areia intermitente (AR). Adicionou-se um tratamento com 100% da adubação com N e P e cujas plantas eram irrigadas com água de abastecimento (AA). O delineamento experimental adotado foi o de blocos inteiramente casualizados, com os tratamentos arranjados em esquema fatorial 4 x 4 + 1, com quatro repetições. Conclui-se é possível utilizar água residuária na irrigação de plantas de berinjela, nas condições edafoclimáticas do semiárido. Doses de 3,55 g de N e 15,28 g de P por planta quando se utiliza água residuária na irrigação de plantas de berinjela são suficientes para suprir as necessidades nutricionais da cultura. A massa fresca da folha de plantas de berinjela decresceu linearmente com o incremente das doses de N aplicadas. A produção de massa fresca e seca da parte aérea de plantas de berinjela irrigadas com água residuária com 40% da adubação com N e P não diferiu das plantas que receberam 100% da indicação de N e P quando irrigadas com água de abastecimento e de reúso. A floração não foi afetada pelas doses de nitrogênio e fósforo, nem pelos diferentes tipos de águas utilizadas na irrigação. Não houve efeitos significativos da interação dos fatores estudados sobre as variáveis avaliadas em nenhuma das épocas de estudo. / The use without control of water by man for various purposes has promoted scarcity of this natural resource especially in arid and semi-arid regions and consequently limiting the social and economic development of regions. The planned reuse is widely used around the world, however, Brazil, even having regions with water scarcity, this practice is not yet widespread. The eggplant (Solanum melongena L.) is a vegetable that has great importance in vegetable crops market in Brazil and worldwide. The objective of this research was to evaluate the technical feasibility of the use of nitrogen and phosphorus along with irrigation using post- treated wastewater in sand filter with intermittent flow in growth, biomass production and flowering eggplants grown in greenhouse in Paraíba semiarid region. The experiment was conducted in a greenhouse, in the Science and Technology Center Agrifood (CCTA / UFCG), Campus de Pombal - PB, and the treatments resulted in the combination of two factors: four doses of nitrogen (N1 - 3.55; N2 - 6.2; N3 - 8.9; N4 - 11.55 g N / pot) and four doses of phosphorus fertilization (P1 - 15.28; P2 - 26.74; P3 - 38.2; P4 - 49 66 g P / pot) corresponding respectively to 40; 70; 100; 130% as fertilizer indication for eggplants when grown in pots irrigated with treated wastewater post-intermittent sand filter (AR). It was added to a treatment with 100% application of N and P and whose plants were irrigated with water supply (AA). The experimental design was a completely randomized design, with treatments arranged in a factorial 4 x 4 + 1, with four replications. We conclude It follows you can use wastewater to irrigate eggplants, at conditions of semiarid. Doses of 3.55 g of N and 15.28 g P per plant when using wastewater in irrigation eggplants are sufficient to meet the nutritional needs of the crop. The fresh weight of eggplants sheet decreased linearly with the increment of N application. The fresh mass production and shoot dry eggplant plants irrigated with wastewater with 40 % application of N and P did not differ from plants receiving 100 % of the N and P indication when irrigated with water supply and reuse. The flowering was not affected by nitrogen and phosphorus doses, or by different types of water used in irrigation. There was no significant interaction of the factors studied on the variables evaluated in any of the study periods.

Entretanto, uma das formas de evitar tais perdas em solos de elevada CTC, é inibir o processo de nitrificação. Desta forma o fornecimento adequado de N associados a produtos que retardem esse processo podem reduzir os impactos negativos causados por tais perdas. Neste trabalho objetivou-se avaliar o efeito de extratos e folhas de Nim sobre a mineralização e nitrificação do nitrogênio no solo, bem como seu efeito no crescimento e nutrição nitrogenada da berinjela e rúcula. Três experimentos, em delineamento inteiramente casualizado, foram conduzidos com amostras de um solo de textura franco arenosa. No primeiro experimento, os tratamentos foram constituídos por um arranjo fatorial 4 x 2, compreendendo 4 tratamentos referentes a inibição da nitrificação à base de nim (extratos de folhas; extrato de sementes; folhas secas e sem inibidor) e duas doses de N (0 e 320 mg dm-3), com cinco repetições. No primeiro experimento, durante 28 dias, a cada sete dias, foram avaliados, os teores de nitrato (NO3-), amônio (NH4+), N-mineral e a relação amônio/nitrato presente no solo. O segundo, foi realizado com plantas de berinjela, da cultivar “Embú”, onde os tratamentos foram constituídos por um arranjo fatorial 4 x 4, sendo quatro tratamentos da inibição da nitrificação (extratos de folhas; extrato de sementes; folhas secas e sem inibidor) e quatro doses de N (20; 120; 220 e 320 mg dm-3), com três repetições. Na fase de préflorescimento, foram determinados as variáveis de crescimento; os teores foliares e os acúmulos das de nitrogênio. No terceiro experimento, utilizou-se plantas de rúcula, da cultivar “rúcula cultivada” onde os tratamentos foram constituídos por um arranjo fatorial 4 x 4, compreendendo quatro tratamentos da inibição da nitrificação (extratos de folhas; extrato de sementes; folhas secas e sem inibidor) e quatro doses de N (10; 120; 220 e 320 mg dm-3), com três repetições. No ponto de colheita da rúcula, avaliou-se o crescimento; teores foliares e acúmulos das frações de nitrogênio. Durante os 28 dias de incubação, os produtos à base de nim não inibiram claramente o processo de mineração ou nitrificação do N aplicado ao solo, houve apenas uma retardação da mineração deste N quando esteve associado a extratos de folhas ou de sementes. A adubação nitrogenada influenciou no crescimento da berinjela e da rúcula, proporcionando incremento na produção e nos teores e acúmulos de N nas folhas. A resposta da rúcula a adubação nitrogenada, foi dependente dos produtos à base de nim. / The eggplant and arugula are demanding vegetables in nitrogen (N) to a suitable production, making the N meet as the nutrient usually applied in agriculture in the form of mineral fertilizer and which has lower efficiency, because their losses various processes. However, one way to avoid such loss in high CTC soils isinhibit nitrification. Thus, the adequate supply of N associated with products that retard this process can reduce the negative impacts caused by such losses. This work aimed to evaluate the effect of extracts and leaves of neem on the mineralization and nitrification of nitrogen in the soil and its effect on growth and nitrogen nutrition eggplant and arugula. Three experiments in completely randomized design were conducted with samples of soil sandy loam texture. In the first experiment, the treatments were factorial arrangement 4 x 2, comprising 4 treatments related to nitrification inhibition based on neem (leaf extracts; seed extract, dried leaves and without inhibitor) and two N rates (0 and 320 mg dm-3), with five repetitions. In the first experiment for 28 days, every seven days, were evaluated the levels of nitrate (NO3-), ammonium (NH4+), N-mineral and ammonium relationship/nitrate in the soil. The second was made with eggplants, cultivar "Embu" where treatments were factorial arrangement 4 x 4, with four treatments of nitrification inhibition (leaf extracts; seed extract, dried leaves and without inhibitor) and four nitrogen rates (20, 120, 220 and 320 mg dm-3) with three replications. In the pre-flowering stage, they were determined growth variables; leaf contents and accumulations of nitrogen. In the third experiment, used rocket plants, cultivar "grown arugula" where treatments were factorial arrangement 4 x 4, comprising four treatments of nitrification inhibition (leaf extracts; seed extract, dried leaves and without inhibitor) and four nitrogen rates (10, 120, 220 and 320 mg dm-3) with three replications. At the point of picking arugula, was evaluated growth; foliar and accumulations of nitrogen fractions. During the 28 days of incubation, the neem-based products not clearly inhibited the process of mining or nitrification of N applied to the soil, there was only a deceleration of mining this when N was associated with extracts of leaves or seeds. Nitrogen fertilization influenced the growth of eggplant and arugula, providing an increase in production and content and accumulation of N in the leaves. Nesse sentido o uso de estratégias que viabilizem o seu cultivo, notadamente em regiões semiáridas, onde há potencial de cultivo e limitação de recursos hídricos. Assim, objetivou-se estudar o efeito de doses de fósforo como atenuante do estresse hídrico ocasionado por lâminas de irrigação no crescimento e na produção de frutos de berinjela. O experimento foi conduzido no Centro de Ciências e Tecnologia Agroalimentar da Universidade Federal de Campina Grande, no período de maio a setembro de 2016, usando-se o delineamento experimental de blocos ao acaso, onde se estudou cinco doses de fósforo, estabelecidas com base na recomendação para cultura (0,0%, 50%, 100%, 150% e 200% da recomendação) e duas lâminas de irrigação (50% e 100% ETr), perfazendo um esquema fatorial 2 x 5, totalizando 10 tratamentos, que foram repetidos em quatro bloco, com 12 plantas por parcela. As mudas de berinjela, cultivar Embú foram transplantadas aos 45 dias após a semeadura, em campo, usando-se o espaçamento de 1,0 x 0,60 m. A lâmina de 100% foi obtida por meio de estimativa da evapotranspiração usando a equação de Penman Monteith – FAO, com dados obtidos da estação agrometeorológica de São Gonçalo – Sousa – PB. Para aplicação das lâminas de água, usou-se um sistema de irrigação por gotejamento, com emissores de 2,6 l h-1 de vazão, e espaçamento de 30 cm entre emissores, colocando-se uma fita gotejadora por linha de cultivo. Durante o período de cultivo, realizou-se o monitoramento da umidade do solo, por meio de tensiômetros instalados em cada tratamento a uma profundidade de 15 cm. Avaliou-se o crescimento por meio da altura de planta, do diâmetro do caule e do número de folhas; variáveis fisiológicas por meio das trocas gasosas e a produção das plantas. Adubação acima da recomendação não proporciona melhorias nas características avaliadas. A adubação próxima a 100% da recomendação teve as melhores respostas para a lâmina de 50% da ETr, onde níveis inferiores ou superiores de P interferiram negativamente na maioria das características avaliadas. Não foi observado interação entre os fatores avaliados (lâminas e doses), porém em condições de déficit hídrico, a adubação com 100% da recomendação proporcionou efeito atenuante do estresse. Em condições de déficit hídrico onde a água é um fator limitante para irrigação, recomenda-se uma adubação fosfatada adequada à cultura, no entanto a falta de água não pode ser totalmente contornada com a adubação fosfatada. / Water is one of the limiting factors for agricultural development, both for lack and for excess, affecting growth, the development and production of plants, especially those considered sensitive. Such as eggplant, and it is important to use strategies that enable their cultivation, especially in semi-arid regions, where there is potential for cultivation and limitation of water resources. The objective of this study was to study the effect of phosphorus doses as attenuating water stress caused by irrigation slides on growth and physiology. The experiment was conducted at the AgroFood Science and Technology Center of the Federal University of Campina Grande, In the period from May to September 2016, using a randomized block design, where five phosphorus doses were studied, based on the recommendation for culture (0.0%, 50%, 100%, 150% and 200% of the recommendation) and two irrigation blades (50% and 100% ETr), making a 2 x 5 factorial scheme, totaling 10 treatments, which were repeated in four blocks, with 12 plants per plot. The eggplant seedlings, to grow Embú, were transplanted at 45 days after sowing, In the field, using the spacing of 1.0 m between rows and 0.60 m between plants, with the plot covering an area of 0.60 m2, being conducted up to 130 days after sowing, when the last Fruit harvest. As a 100% blade was obtained by estimating evapotranspiration using an equation of Penman Monteith - FAO, with data obtained from the São Gonçalo - Sousa - PB agrometeorological station. For application of the water slides, a drip irrigation system was used, with emitters of 2.6 l h-1 of flow, and spacing of 30 cm between emitters, placing a drip tape per culture line. During the cultivation period, the soil moisture was monitored by tensiometers installed in each treatment at a depth of 15 cm. Growth was evaluated by plant height, stem diameter and number of leaves; Physiological variables through gaseous changes, fruit quality variables, through physical and chemical analyzes of the fruits and the production of the plants. Fertilization above the recommendation does not provide improvements in the evaluated characteristics. Fertilization close to 100% of the recommendation had the best responses for the 50% ETr slide, where lower or higher levels of P interfered negatively in most of the characteristics evaluated. No interaction was observed between the factors evaluated (slides and doses), but under conditions of water deficit, fertilization with 100% of the recommendation provided an attenuating effect of stress. In conditions of water deficit where water is a limiting factor for irrigation, it is recommended a phosphate fertilization appropriate to the crop, however the lack of water cannot be completely bypassed with phosphate fertilization.

Oxydation et dégradation de l'ascorbate chez la tomate et impact sur la croissance et le métabolisme / Oxidation and degradation of ascorbate in tomato and impact on growth and metabolism

Contrôle de l'oxydation et de la dégradation du pool de vitamine C chez la tomate et impact sur la qualité du fruit et la tolérance au stress. Le métabolisme de l’ascorbate et plus principalement le statut redox du pool d’ascorbate sont impliqués dans la tolérance au stress et dans les processus primaires de croissance et de développement de la plante. La teneur et le statut redox de l’ascorbate chez les plantes sont régulés par (i) ses voies de biosynthèse, (ii) par le cycle ascorbate-glutathion permettant le recyclage des formes semi-oxydées et oxydées de l’ascorbate et (iii) par sa dégradation, l’ensemble de ces processus étant sous le contrôle de l’environnement. Au cours de ce travail de thèse, des méthodes de transgénèse nous ont permis d’identifier, chez différents génotypes de tomate à petit et gros fruits, les bouleversements physiologiques et métaboliques permettant de compenser des modifications de l’activité des enzymes monodéhydroascorbate réductase (impliqué dans le cycle ascorbate-glutathion) et ascorbate oxydase. Nous avons observé d’importantes modifications phénotypiques altérant le rendement en fruits de la plante sous conditions de culture pouvant générer un stress et également en condition normale de culture. Des liens entre l’activité des enzymes précités avec le métabolisme des sucres, la photosynthèse et la conductance stomatique sont révélés. Le déséquilibre entre les activités oxydantes et réductrices de ces enzymes constitue la première étape vers une dégradation de l’ascorbate. Le taux de dégradation se révèle très faible à la lumière, tandis qu’à l’obscurité une forte accumulation des produits de dégradation l’oxalate, le thréonate ainsi que l’oxalyl-thréonate est observé dans les feuilles de tomate. Enfin, l’activité de l’enzyme MDHAR est corrélée au taux de dégradation à l’obscurité. Les travaux de cette thèse mettent en avant l’importance du statut redox du couple ascorbate / monodéhydroascorbate dans les processus de croissance cellulaire et entre dans la régulation du rendement chez la tomate, et influe la dégradation de l’ascorbate. / Ascorbate metabolism and particularly ascorbate redox status are involved in stress tolerance and growth processes of plant cells. The concentration of ascorbate and its redox status are under control of (i) its biosynthetic pathways, (ii) the ascorbate-glutathione cycle allowing recycling of semi-oxidized and oxidized forms of ascorbate and (iii) its degradation rate. These processes are under environmental control. Transgenic lines modified for the activity of monodehydroascorbate reductase (involved in ascorbate-glutathione cycle) and ascorbate oxidase were generated in cherry and large-fruited genotypes of tomato. Physiological and metabolic modifications related to the modification of these enzyme activities were studied. We observed large phenotypic alterations that affected fruit yield under both stress conditions and normal growth conditions. Links between ascorbate recycling and sugar metabolism, photosynthesis and stomatal conductance were also revealed. An imbalance between the oxidizing and reducing activities of these enzymes is the first step leading to ascorbate degradation. We have shown that the degradation rate was very low under light, whereas under darkness the degradation compounds oxalate, threonate and oxalyl-threonate accumulated in tomato leaves. Also, the degradation rate is correlated with MDHAR activity. These results highlight the crucial role of the redox status of the ascorbate / monodehydroascorbate couple in growth processes and yield stability in tomato, and the impact on ascorbate degradation.

Nuevas aportaciones al metabolismo secundario del tomate. Identificación y estudio de moléculas implicadas en la respuesta a la infección con pseudomonas syrinagae pv. tomato

Los fenilpropanoides constituyen un grupo de metabolitos secundarios producidos y utilizados por las plantas como parte de la respuesta defensiva tanto constitutiva como inducible. Un gran número de ellos están implicados en la resistencia frente a la enfermedad a diferentes niveles: señalización (ácido salicílico), agentes antimicrobianos (fitoalexinas), y endurecimiento de la pared celular (lignina). Las amidas derivadas del ácido hidroxicinámico (HCAAs) son un conjunto de metabolitos, pertenecientes al grupo de los fenilpropanoides, que desempeñan un importante papel en la defensa de las plantas frente a patógenos y predadores. Las HCAAs se forman a partir de la condensación de tioésteres de hidroxicinamoil-CoA con feniletilaminas, tales como la tiramina. El último paso en la biosíntesis de las HCAAs está catalizado por el enzima tiramina hidroxicinamoil transferasa (THT). En la presente tesis se muestra la identificación y el estudio de cuatro HCAAs, p-cumaroildopamina, feruloildopamina, p-cumaroiltiramina y feruloiltiramina, asociadas a la infección de tomate con la bacteria Pseudomonas syringae pv. tomato. Su identificación y caracterización estructural se han llevado a cabo mediante técnicas de cromatografía líquida de alta resolución y espectrometría de masas (HPLC-MS). Se ha analizado la posible implicación del ácido salicílico y del etileno en la inducción patogénica de dichas moléculas y del enzima responsable de su biosíntesis (THT). Además, se ha estudiado la actividad antioxidante y antibacteriana in vitro de las cuatro HCAAs identificadas. Por último, se han obtenido líneas transgénicas de Arabidopsis thaliana y de tomate que sobreexpresan el gen de la THT, y se han analizado los perfiles cromatográficos de dichas líneas. / Zacarés Sanmartín, L. This work was carried out in collaboration with the Department of Adaptive Biotechnologies, Global Change Research Centre AS CR. It deals with the quantitative analysis of photosynthetic cell cultures. It uses images captured by a confocal fluorescent microscope to the automatic determining the number of cells in the sample. The work consists of a theoretical analysis, which briefly describes fluorescence and confocal microscopy. It also concisely introduces a microscope Leica TCS SP8 X, which I used to scan data. One capture is devoted to the theory of digital image processing. The second part deskribes the development of algorithm for processing 3D data and simplified algorithm for processing 2D data and its program implementations in a programming environment MATLAB R2013b. Grafical user interface is explained in detail. Done measurement are presented at the conclusion. It mentions compiled sample preparation protocol. The results of the program are compared with manual counting. Number of cells per 1 ml are determined by created program in samples of cell cultures Chenopodium rubrum (Cr) and Solanum lycopersicum (To).
