In the introduced diploma work, Controversial and survivor affairs of the lineage Thurn-Taxis on the estate Dobrovice-Loučeň, namely from the end of 18th century to the beginning of the 20th century, engages in the controversial and survivor affairs of the lineage Thurn-Taxis in the frame of the named estate in the delimitated period. The main aim of the work is to clarified the origin of the lineage Thurn-Taxis, to clear up the disputes of Fürstenberg about the inheritance of the estate in the end of 18th century and briefly to describe the development of the estate Dobrovice-Loučeň with the holders from the second line of the lineage Thurn- Taxis. The main purpose of this work is the content analysis and the analysis of the controversial affairs, for example last will or the list about the distributing of the inheritance, a few representatives of the lineage from the Czech line and of the princess Marie Josefa of Fürstenberg. The princess bequeathed the estates Dobrovice, Loučeň and the manor Vlkava for the Czech line. The suppluments added to the work complete the all pieces of knowledge in its text part. Chemosensory Responses in Azospirillum brasilense

The ability to swim and navigate the surrounding environment confers an advantage to motile bacteria, allowing the occupation of niches that are optimum for survival and growth. Bacteria are too small to sense their environment spatially, so they must sense the environment temporally by comparing the past and present environments and altering their motility accordingly. Chemotaxis systems coordinate flagellar motility responses with temporal sensing of the environment. Chemotaxis is proposed to be involved in plant root colonization by directing soil bacteria toward root exudates of various cereals, promoting growth. The nitrogen-fixing alpha-proteobacterium Azospirillum brasilense utilizes chemotaxis to navigate its environment by integrating various environmental signals into a chemotaxis signal transduction pathway. In chemotaxis, transducers receive environmental sensory information and transmit the signal to the histidine kinase CheA, which relays the signal to the response regulator CheY. A novel chemotaxis transducer, Tlp1, has been identified and characterized as an energy sensor by constructing a tlp1 mutant and performing behavioral and root colonization assays. In order to adapt to changing environmental conditions, chemotactic microorganisms must employ a molecular “memory” comparing present environmental conditions to ones previously experienced and resetting the chemotaxis transducer to a prestimulatory status. A recently identified chemotaxis operon revealed a methyltransferase CheR and methylesterase CheB, comprising an adaptation system, suggesting that A. brasilense undergoes methylation-dependent taxis responses, contrary to previous reports. Chemotaxis and methanol release assays suggest that adaptation by methylation in locomotor behavior involves the presence of other unknown methylation systems, and the contribution of CheR and CheB to chemotactic and aerotactic responses is complex. There is growing evidence that chemotaxis-like signal transduction pathways control a myriad of other cellular processes regulated in a temporal fashion. This would convey an advantage to cells by allowing modulation of cellular processes based on slight changes in environmental conditions and provide checkpoints for energetically consuming processes. Mutations in components of the chemotaxis-like signal transduction system revealed differences in cell size and exopolysaccharide production. This work shows that the signal transduction pathway of A. brasilense modulates cell length in response to changes in nutrient conditions, independently of growth rate.

Os moto-táxistas ao executar suas atividades ficam expostos a agentes agressores como ruído e vibração além de constantes perigos do trânsito (quedas e acidentes), que lhes causam prejuízo físico e mental. Portanto, é de grande preocupação a melhoria das condições de trabalho destes profissionais, se não controlando a fonte de ruído, mas, oferecendo alternativas que assegurarão uma boa qualidade de trabalho. Objetivo: Avaliar a exposição ao ruído e proteção auditiva em moto-táxistas atuantes na cidade de Balneário Camboriú. Como objetivos específicos buscou-se avaliar o posto de trabalho quanto ao ruído e outras condições de salubridade; averiguar a acuidade auditiva dos moto-táxistas; pesquisar o grau de percepção do profissional quanto ao ruído; apurar a atenuação sonora dos capacetes com medições no laboratório e em campo; propor um Programa de Prevenção Auditiva para esta classe de trabalhadores; averiguar a possibilidade do uso do capacete combinado com plugue pelos moto-táxistas. Métodos: O presente trabalho foi realizado em duas etapas: primeiramente em laboratório que consistiu em avaliar a atenuação de ruído de três modelos de capacetes através da norma ANSI 12.6/97A e, a segunda, em campo que teve como procedimento aplicar uma entrevista com roteiro semi-dirigido a 17 moto-táxista da cidade de Balneário Camboriú, quantificar a atenuação sonora dos três modelos de capacetes, analisar a dose de ruído, além de levantar o perfil audiométrico dos profissionais e a exposição dos mesmos. Resultados: Os moto-táxistas estão expostos a um nível de pressão sonora superiores a 82,1 dBA e a doses de ruído acima do permitido para seu tempo de trabalho diário, ou seja, 12 h/diárias. Comparando a dose de ruído de todos os moto-táxistas durante as 12 horas de trabalho diário e levando-se em consideração o capacete, possibilitando a verificação de que as doses de ruído ultrapassam 100% diariamente para todos os trabalhadores e nos três modelos de capacetes estudados. Dos moto-táxistas estudados, 8 apresentaram limiares auditivos dentro dos padrões de normalidade e 9 apresentaram alteração dos limiares tonais. Os baixos níveis de atenuação sonora dos capacetes, tanto no laboratório quanto em campo, enfocou a não efetividade dos mesmos como protetores auriculares. Ficou evidente o desconhecimento dos moto-táxistas sobre os efeitos do ruído no seu ambiente de trabalho, mas os motociclistas apresentaram queixas extra-auditivas e auditivas que poderiam ser relacionadas ao ruído intenso. Conclusão: na indisponibilidade de mudar o NPS no ambiente de trabalho medidas de minimização dos efeitos do ruído devem ser adotadas, entre elas a proposição de um Programa de Proteção Auditiva e alternativas de equipamentos de proteção individual, visando à melhoria da qualidade de vida dos profissionais.

El contexto de este trabajo de título subyace en el rubro del transporte privado, en particular dentro del marco operacional de la empresa One Radio Taxi , elegida como caso de estudio. Se investigó en profundidad acerca del mecanismo utilizado en el proceso de asignación de carreras que implementa esta compañía, con el fin de definir y analizar los factores de mayor incidencia; para así, lograr identificar los diversos aspectos mejorables de dicho proceso. En este proyecto se rediseñó el proceso de asignación, implementando dos mecanismos diferentes que realicen la asignación de cada solicitud de traslado, dependiendo de su naturaleza. Se definió que la característica más adecuada para categorizar las carreras es la diferencia de tiempo entre la hora en que ésta es solicitada y la hora en que se debe recoger al pasajero. Respecto al criterio anterior se definieron dos categorías de carrera, aquellas con ventanas superiores e inferiores a 12 horas. Por último, se estableció la distancia recorrida al día por la flota de la empresa, como el indicador primario de rendimiento. Este indicador posteriormente se utilizó para contrastar el desempeño de cada módulo de asignación implementado, con respecto a los resultados obtenidos en base al método utilizado por la empresa en cuestión. Culminado el proceso anterior, se definieron los diversos requisitos asociados al software de apoyo necesario para la implementación de cada rediseño planteado en relación a las categorías de solicitud de traslado. En base a estas se construyó un prototipo funcional asociado a cada mecanismo de asignación elaborado. Posterior a esto se procedió a diseñar y desarrollar diversas herramientas que permitieron llevar a cabo un proceso de simulación en base a información operacional de One Radio Taxi. Durante la etapa final del proyecto se observó que cada módulo implementado presentó rendimientos superiores en relación a la distancia promedio recorrida por la flota para atender a un mismo número de carreras. El trabajo concluye habiendo desarrollado efectivamente un rediseño de proceso, con el objetivo de mejorar el mecanismo de asignación de una flota de radio taxi. Si bien el periodo de simulación se llevó a cabo bajo diferentes supuestos, debido a que estos fueron considerados de igual forma para la evaluación de cada mecanismo de asignación; la comparación entre cada uno es significativa en términos relativos. Se estableció como trabajo futuro recomendado la puesta en marcha de cada módulo y la integración de un servicio externo de georreferencia con el objetivo de mejorar la precisión del proceso de simulación.

Macroscopic fundamental diagrams for Stockholm using FCD data

Macroscopic fundamental diagrams (MFD) reveal the relations among flow, speed and density in a large geographic region. After literature review on macroscopic analysis, the similar methodology is applied in this thesis. The purpose of the thesis is to find the evidence that is able to prove MFD existing in Stockholm urban region. Both floating car data (FCD) based on global positioning system (GPS) data from taxis travelling in Stockholm region and traffic data from fixed detectors data source are used to construct the fundamental diagrams. Geographically, the usage of data is extended from single link to multiple links, then to the entire study region. The temporal phase is restricted in one weekday and weekend. The diagram of flow vs. speed based on single detector is found disordered, by contrast, the diagrams of cumulative flow, speed and density for all detectors represent orderly. MFD diagrams proposed in Yokohama case study by Geroliminis and Daganzo are reproduced with cumulative data in this thesis.  Therefore, it can be proved that MFD exists when using data from multiple links. However, the cumulative data from fixed detectors only represents the traffic on links where they locate, not the entire region. To overcome it, GPS data from taxis, which covers the whole region, is analyzed with same method. Because full taxis travel in the same manner as normal vehicles, they are selected to approximate traffic in whole region. A neat curve of flow vs. speed is produced and it coincides with corresponding diagram in the reference paper. It enhances the conclusion that MFD exists in the entire study region. Moreover, based on the constant ratio between average link flow and region exit flow, a controlling density policy is discussed in aiming for maximizing trip completion.

El presente trabajo de investigación evalúa los programas de fidelización enfocado en el sector de servicios de taxi en Lima Metropolitana, los cuales se desarrollan a través de plataformas digitales como las aplicaciones móviles, adaptándose a las necesidades de los usuarios actuales y aprovechando la coyuntura actual del sistema de transporte que se presenta en la ciudad de Lima, la cual ha generado motivadores que han permitido desarrollar esta nueva estructura del rubro de taxis; es así que se ha investigado acerca de los programas de fidelización que surgieron en otros países y los factores que permitieron replicarlo en la nuestra ciudad. Asimismo, se indagó acerca de los perfiles del consumidor limeño respecto al uso de aplicaciones y el efecto de los programas de fidelización, con esa razón se realizaron entrevistas a expertos en marketing y digital, encuestas a los usuarios de taxi para validar el éxito o fracaso de los programas de fidelización en aplicaciones de taxi. Con ello se obtuvo que los factores más relevantes para los usuarios al momento de solicitar un taxi por aplicación son la comodidad, seguridad y participación en campañas. / This research evaluates the loyalty programs focused on the taxi service sector in Lima Metropolitan, which are developed through digital platforms such as mobile applications, adapting to the needs of current users and taking advantage of the current situation of the Transport system that is presented in the city of Lima, which has generated motivators that have allowed to develop this new structure of the category of taxis; so we have investigated the loyalty programs that have emerged in other countries and the factors that allowed them to replicate in our city. Likewise, we inquired about the profiles of the Lima consumer regarding the use of applications and the effect of loyalty programs, with that reason we conducted interviews with marketing and digital experts, we also did surveys to taxi users to validate success or failure of loyalty programs in taxi applications. Au cours des dernières décennies, la mobilité dans les villes camerounaises a été marquée par la croissance considérable du trafic des motos-taxis, qui constituent désormais le mode de déplacement dominant. La sédimentation actuelle des deux-roues dans la circulation génère des nuisances, des problèmes et des coûts pour la collectivité et les usagers. La conséquence la plus directe provoquée par cette mutation de la mobilité est l’augmentation croissante des accidents. Les motos sont désormais au coeur de violents conflits d’usage sur la voie publique. L’objectif de notre recherche est double, il s’agit d’abord d’améliorer la prise de conscience de l’importance des enjeux et des externalités et, ensuite, de proposer des pistes de réflexion pour favoriser le passage d’une cohabitation conflictuelle à une cohabitation pacifique entre les motos-taxis et les autres usagers de la route. Celle-ci nécessite une réponse urbanistique, pédagogique, sociologique et politique, elle implique une gouvernance et une gestion urbaine appropriée. / During the last decades, daily urban and rural displacements in Cameroonian towns have suffered from an increase of motorcycle taxis in traffic flow. Those taxis are now considered the major mode of urban transport. This current predominance of two-wheelers in the urban landscape generates an explosive growth of pollution, road traffic accidents and public spending. The most direct and profound impact of this mutation is the increase of motorcycle accidents. Motorcycles are consequently the cause of violent conflicts among road users on the public highway. Our research has two goals: the first one is improving people's awareness of what is at stake concerning the motorcycle system and his negative effect in urban mobility, and the second one is giving food for thought in order to improve relations between motorcycle taxis and the other road users. We also want to help find solutions to manage and integrate motorcycles and their commercial use in the city. To do so, we will focus on urban, instructive, sociological and political approaches and on appropriate urban planning.
