Produção de cordeiros em pastagem tropical : dos aspectos bioquímicos da pastagem ao comportamento ingestivo animal / Lamb production in tropical pasture : from the biochemical aspects of pasture to animal ingestive behavior

As pastagens que cobrem uma importante área no território mundial apresentam elevado potencial de produtividade e suas características bioquímicas podem gerar relevante impacto na produção de ruminantes. Porém, poucos trabalhos relatam a concentração de compostos bioquímicos em pastagens tropicais comumente usadas na produção animal. Diante deste fato, o capítulo II desta tese teve como objetivo elucidar e determinar a concentração de taninos condensados e tocoferol, bem como avaliar a produção e os valores nutritivos de espécies forrageiras tropicais normalmente usadas em sistemas de pastejo. Com este estudo foi possível constatar que algumas espécies de gramíneas e leguminosas se destacam não apenas pela sua capacidade de produção de biomassa e qualidade nutricional, mas também pela concentração de compostos bioquímicos, como a gramínea tifton que teve elevada concentração de α-tocoferol (202.3 ± 116.5 mg/kg matéria verde, MV). As leguminosas amendoim forrageiro (15.7 ± 5.2 g/kg matéria seca, MS) e feijão guandu (8.7 ± 0.8 g/kg MS) apresentaram boa concentração de taninos condensados, mas o grande destaque foi para a leguminosa nativa pega-pega (66.5 ± 13.8 g/kg MS) que além da alta concentração de taninos, esses apresentaram potencial para atividade biológica (PPF = 60.1 g/kg MS) Aliado as características bioquímicas, as espécies que irão compor o sistema de produção podem influenciar na biologia de vida livre dos nematoides parasitas do trato gastrintestinal (NGI) e no comportamento ingestivo dos animais. O capítulo III e IV avaliam três diferentes sistemas de alimentação de cordeiros em pastagens tropicais. O capítulo III traz como objetivo avaliar a distribuição de larvas infectantes (L3) em diferentes perfis de pastagem tropical (gramíneas e leguminosas) e o impacto da contaminação da pastagem na carga parasitária e desempenho de cordeiros. Onde, a inclusão da leguminosa tropical alterou a concentração de larvas infectantes na pastagem e a contaminação dos animais. No capítulo IV é apresentado o trabalho que teve como objetivo avaliar o comportamento ingestivo de cordeiros em condições de pastejo contínuo submetidos a diferentes sistemas de alimentação com a presença de leguminosa tropical contendo taninos condensados. Apesar da diferença estrutural e concentração de taninos condensados, de modo geral essas características da pastagem não alteraram o comportamento ingestivo dos cordeiros. / Pastures covering an important area in the world territory present high production potential and their biochemical characteristics can generate a significant impact on the production of ruminants. However, few studies report the concentration of biochemical compounds in tropical pastures commonly used in animal production. Facing this fact, chapter II of this thesis had as objective to elucidate and to determine the concentration of condensed tannins and tocopherol, as well as to evaluate the production and nutritional values of tropical forage species normally used in grazing systems. In this study, it was possible to verify that some species of grasses and legumes stand out not only for their mass production capacity and nutritional quality, but also for the concentration of secondary compounds, such as tifton grass, which had a high α-tocopherol concentration (202.3 ± 116.5 mg / kg green matter, GM). The forage peanut legumes (15.7 ± 5.2 g / kg dry matter, DM) and pigeon pea (8.7 ± 0.8 g / kg DM) had an excellent concentration of condensed tannins, but the main highlight was the native legume Desmodium incunum (66.5 ± 13.8 g / kg DM) that in addition to the high concentration of tannins, these had potential for biological activity (PPP = 60.1 g / kg DM) In addition to the biochemical characteristics, the species that will make up the production system can influence the ecology of the free-living stages of endoparasite of the gastrointestinal tract (NGI) and the ingestive behavior of the animals. Chapter IV presents the work that aimed to evaluate the ingestive behavior of lambs under continuous grazing conditions submitted to different feeding systems with the presence of tropical legume containing condensed tannins. Despite the structural difference and concentration of condensed tannins, in general these pasture characteristics did not alter the ingestive behavior of the lambs.

Thesis deals with the analysis of vitamin E in cereal samples, namely barley and wheat and malt produced from them. Selected samples were treated with low-temperature plasma, fungicide and biological treatment to elimination fungus and mycotoxins. The aim of the thesis was to find out the influence of the treatment, the combination of several types of treatment on vitamin E content in grain and malt. If the content of health-positive substances is not reduced and grain can be used for food, malt, or for feed purposes. The theoretical part focuses on the properties, occurrence, biochemistry and importance of vitamin E for living organisms. It also focuses on the properties of cereals that are a significant source of vitamin E. Malt production and sample treatment are also described. The experimental part describes the field experiment, individual types of treatment, extraction of vitamin E from the samples and determination of vitamin E content by high performance liquid chromatography with fluorescence detection. The results are compared with the current literature. It was found that the treatment did not have a significant negative effect on vitamin E content.

Tocopherol (vitamin E) content in invasive browse species on underutilized Appalachian farmland

The tocopherol (Vitamin E) content of forage from three invasive shrub species was measured to assess the value of the shrubs as a source of vitamin E for goats browsing on overgrown Appalachian pastures. Plant leaf clusters were collected from multiflora rose (Rosa multiflora Thunb.), autumn olive (Elaeagnus umbellata Thunb.), and Morrow's honeysuckle (Lonicera morowii Gray) in replicated plots at a site in southern West Virginia during the 1999 growing season. Alpha-, beta-, gamma-, and delta-tocopherol were extracted with hexane, separated by high performance liquid chromatography on a normal-phase diol column, and quantified. Significant differences (P<0.001) in concentration were found among species for all forms of tocopherol. Alpha-tocopherol predominated, accounting for more than 90% of the total tocopherols in all three species. Alpha-tocopherol levels increased in all species with maturity; however, the magnitude of the increase was not the same in all species. At the end of the growing season, autumn olive had the highest levels of alpha-tocopherol (1270 ± 55 ppm dry matter [DM]), followed by Morrow's honeysuckle (840 ± 55 ppm DM), and multiflora rose (610 ± 55 ppm DM). Goats grazing on mature browse may obtain adequate intake of vitamin E. High nutritive value and/or low concentrations of antiquality factors may not coincide with the high levels of vitamin E found in mature tissue, and the actual vitamin E intake will depend on the feeding behavior of the goat. / Master of Science

Les patrons de variation au sein des espèces d'arbres résultent d'évolutions complexes dont certaines sont particulièrement liées à la condition d'arbre. Si les facteurs expliquant cette variabilité ont été abordés pour les espèces des zones tempérées, ils restent encore peu connus pour des espèces fruitières des zones tropicales sèches offrant des particularités sur le plan adaptatif et relevant d'un processus de domestication ancien. Pour répondre à cet enjeu, l'objectif scientifique de cette thèse est de mieux comprendre l'impact des facteurs évolutifs sur la variabilité au sein d'une espèce d'arbre en zone soudano-sahélienne. Nous nous sommes intéressés à Vitellaria paradoxa (karité), une espèce jouant un rôle économique majeur par la production de beurre à partir des graines, dont l'aire de distribution s'étend du Sénégal à l'Ouganda sous la forme de peuplements naturels et agroforestiers. Dans une approche phylogéographique basée sur l'analyse du polymorphisme de marqueurs moléculaires supposés neutres (microsatellites chloroplastiques et nucléaires, et régions intergéniques du chloroplaste) et sur la modélisation des niches écologiques du Karité à différentes périodes du Quaternaire, nous mettons en évidence l'impact des perturbations liées aux dernières glaciations (il y a 20000 ans) sur la diversité génétique de l'espèce. Dans une seconde approche méthodologique, nous étudions l'impact des déterminants environnementaux sur la variabilité des constituants chimiques des graines de Karité, permettant d'énoncer de nouvelles hypothèses. Enfin, nous nous intéressons à l'expression du polymorphisme de gènes candidats, codant les enzymes stéaroyl-ACP-désaturase (SAD) et homogentisate phytyltransférase (HPT), en relation avec la variabilité de la composition chimique en acides gras et en tocophérols des graines. Les résultats obtenus permettent notamment de discuter de l'impact de la sélection naturelle, de la domestication et de la dérive génétique, et d'apporter des hypothèses évolutives et fonctionnelles potentielles expliquant la variabilité observée. / Patterns of variation within tree species are the result of complex evolutionary changes, some of which are particularly related to the condition of trees. If the factors explaining this variability were discussed for species in temperate zones, they remain little known for fruit tree species of dry tropics, which show special adaptive features and result from an old domestication process. To meet this challenge, the scientific purpose of this thesis is to improve our understanding of evolutionary factors affecting the variability within a tree species in the Sudano-Sahelian zone. We got interested in Vitellaria paradoxa (Shea tree), a species which plays a major economic role for the production of butter from seeds and whose distribution range extends from Senegal to Uganda. In a phylogeographic approach based on the analysis of polymorphism supposedly neutral molecular markers (chloroplast and nuclear microsatellites and chloroplast intergenic regions) and based on ecological niche modelling of Shea tree at different periods of the Quaternary, we shape the role of last glacial maximum (20.000 YBP) on the genetic diversity of the species. In a second methodological approach, we study the impact of environmental determinants on the variability of Shea nuts chemical constituents, stating novel hypotheses. Finally, we investigate the polymorphism of candidate genes, encoding enzymes stearoyl-ACP-desaturase (SAD) and homogentisate phytyltransférase (HPT), in connection with the variability of relative fatty acids compositions and tocopherols contents in seeds. Through the results obtained, we discuss the impact of natural selection, domestication and genetic drift, and provide evolutionary and functional hypotheses that potentially explain the varia bility observed.

Antioxidants as Risk Factors for Gingival Bleeding

Background: Studies of gingival bleeding and the effects of antioxidants on extracellular matrix and immunologic and inflammatory responses provide a rationale for hypothesizing that antioxidants reduce the risk for gingival bleeding.Methods: This study evaluated the role of antioxidants as contributing risk factors for gingival bleeding utilizing the Third National Health and Nutrition Examination Survey (NAHNES III). A sample of 18,825 adults (20 to ≥ 90 years of age), with dental measurement and assessment of serum levels of antioxidants were included in the study. Gingival bleeding was defined as those who had more than 30 percent of gingival bleeding in 28 sites examined. SPSS version 11.0 software and Epi-info 2000 were used to perform the statistical analysis.Results: Using multiple logistic regression in five separate antioxidants, the study showed an association between increased plasma levels of vitamin C (ascorbic acid) and decreased risk for gingival bleeding (OR= 0.33; 95% CI 0.15 to 0.72). An inverse relationship was also found between gingival bleeding and serum levels of beta carotene (OR=1.93; 95% CI 1.05 to 3.54). However, negative association was found between gingival bleeding and vitamin A (OR=2.60; 95% CI 1.04 to 6.50). No statistically significant association was observed between gingival bleeding and serum levels in vitamin E (alpha tocopherol) and selenium.Conclusion: Antioxidants, vitamin C, vitamin A, and beta carotene, were significant risk factors for gingival bleeding. This should be emphasized for improving the oral health of the U.S. adult population.

Estado nutricional relativo ao selênio de pacientes na fase de pós-tratamento da leucemia linfóide aguda e sua relação com o estresse oxidativo / Nutritional status of selenium in patients in post-treatment of acute lymphoblastic leukemia and its relationship with oxidative stress

Este estudo teve como objetivo avaliar o estado nutricional relativo ao selênio (Se) de pacientes na fase de pós-tratamento da leucemia linfóide aguda (LLA) e sua relação com o estresse oxidativo. Foram selecionados 24 pacientes no pós-tratamento da LLA (9,2 ± 1,9 anos) atendidos no Instituto de Oncologia Pediátrica da Universidade Federal de São Paulo e 60 indivíduos saudáveis (9,5 ± 1,3 anos) da Escola de Aplicação da Universidade de São Paulo. Foram coletados 10 mL de sangue venoso para análise de Se plasmático e eritrocitário, glutationa peroxidase (GPx), superóxido dismutase (SOD), &#945;- tocoferol, malondialdeído (MDA) e 8-oxo-desoxiguanosina (8-oxo-dGuo). A urina de 24 horas foi coletada para análise da excreção de Se, e três recordatórios de consumo alimentar de 24 horas para avaliação do Se ingerido. Os resultados obtidos quanto aos parâmetros bioquímicos de avaliação de Se não apresentaram diferença significativa entre os grupos de pacientes e controles, e foram respectivamente: Se plasmático, 44,4 ± 9,0 &#181;g/L e 48,7 ± 12,0 &#181;g/L (p = 0,122); Se eritrocitário, 49,9 ± 15,9 &#181;g/L e 45,0 ± 15,9 &#181;g/L (p = 0,202); Se urinário, 19,6 ± 14,8 &#181;g Se/g de creatinina e 18,6 ± 9,6 &#181;g Se/g de creatinina (p = 0,820). O consumo médio de Se foi de 27,4 ± 8,7 &#181;g/dia e 28,0 ± 1,5 &#181;g/dia (p = 0,756), respectivamente. Os grupos estudados foram considerados deficientes em Se, considerando os pontos de corte adotados. A atividade da GPx foi significativamente menor nos pacientes do que nos controles (33,3 ± 11,1 U/g Hb e 76,9 ± 25,9 U/g Hb) (p = 0,000), e a atividade da SOD não diferiu entre pacientes e controles (1796,9 ± 257,8 U/g Hb e 1915,9 ± 473,9 U/g Hb) (p = 0,145), assim como as concentrações de MDA (1,7 ± 0,3 &#181;mol/L e 1,8 ± 0,4 &#181;mol/L) (p = 0,053). A concentração de &#945;-tocoferol foi estatisticamente maior nos pacientes que nos controles (17,7 ± 4,7 &#181;mol/L e 10,6 ± 3,2 &#181;mol/L) (p =0,000), bem como a concentração de 8-oxo-dGuo (43,6 ± 28,0 8-oxo/106 dG e 21,3 ± 22,9 8-oxo/106 dG) (p = 0,014). Os resultados apresentados apontam que os participantes deste estudo estão deficientes em Se e, em especial os pacientes no pós-tratamento da LLA estão sujeitos a um aumento do estado de estresse oxidativo, pois apesar das concentrações de MDA serem semelhantes entre os pacientes e os controles, a atividade da GPx dos pacientes foi reduzida e a concentração de 8-oxo-dGuo e &#945;-tocoferol estavam aumentadas em relação aos controles. / This study aimed to evaluate the nutritional status of selenium (Se) in patients in post-treatment of acute lymphoblastic leukemia (ALL) and its relationship with oxidative stress. We selected 24 patients in post-treatment of ALL (9.2 ± 1.9 years) at the Pediatric Oncology Institute of Federal University of São Paulo and 60 healthy individuals (9.5 ± 1.3 years) of the School of Application at the University of São Paulo. We collected 10 mL of venous blood for analysis of Se in plasma and erythrocytes, glutathione peroxidase (GPx), superoxide dismutase (SOD), &#945;-tocopherol, malondialdehyde (MDA) and 8-oxo-deoxyguanosine (8-oxo-dGuo). The results regarding biochemical evaluation of Se did not differ significantly between patients in post-treatment of ALL and controls, and the results were respectively: Se in plasma, 44.4 ± 9.0 &#181;g/L and 48.7 ± 12.0 &#181;g/L (p = 0.122); Se in erythrocytes, 49.9 ± 15.9 &#181;g/L and 45.0 ± 15.9 &#181;g/L (p = 0.202); Se in urine, 19.6 ± 14.8 &#181;g Se/g creatinine and 18.6 ± 9.6 &#181;g Se/g creatinine (p = 0.820). The average intake of selenium was 27.4 ± 8.7 mg/day and 28.0 ± 1.5 mg/day (p = 0.756), respectively. Both groups were considered deficient in selenium, according to the cut-off points adopted. The GPx activity was significantly lower in patients than in controls (33.3 ± 11.1 U/g Hb and 76.9 ± 25.9 U/g Hb) (p = 0.000), and no difference in SOD activity was observed between groups (1796.9 ± 257.8 U/g Hb and 1915.9 ± 473.9 U/g Hb) (p = 0.145). MDA concentrations were not different between patients and controls (1.7 ± 0.3 &#181;mol/L and 1.8 ± 0.4 &#181;mol/L) (p = 0.053) and &#945;-tocopherol concentration was statistically higher in patients (17.7 ± 4.7 &#181;mol/L and 10.6 ± 3.2 &#181;mol/L) (p = 0.000), as well the concentration of 8-oxo-dGuo (43.6 ± 28.0 8-oxo/106 dG and 21.3 ± 22.9 8-oxo/106 dG) (p = 0.014). These results indicate that the participants in this study are deficient in Se mainly those who are in post-treatment of ALL are exposed to an increased state of oxidative stress, because although the concentrations of MDA were similar between patients and controls, the GPx activity of the patients was reduced and the concentration of 8-oxo-dGuo and &#945;-tocopherol were increased compared to controls.

Radiation-induced fibrosis : Characterization of the anti-fibrotic mechanisms displayed by pentoxifylline/vitamin E / Fibrose radio-induite : Mécanismes moléculaires impliqués dans l’action anti-fibrosante exercée par l’association pentoxifylline-vitamine E

La fibrose radio-induite est une complication sévère et tardive de la radiothérapie. Plusieurs études cliniques ont montré que la combinaison pentoxifylline-vitamine E est un traitement sûr et efficace contre la fibrose. Cependant, les mécanismes moléculaires de son efficacité restent inexplorés. Nous avons montré l’efficacité de la combinaison pentoxifylline-vitamine E dans l’entéropathie radique dans une faisabilité clinique. En parallèle, en utilisant un modèle unique, in vitro, de cellules musculaires lisses intestinales primaires isolées des personnes atteintes de l’entéropathie radique, nous avons montré une synergie entre la pentoxifylline et l’analogue hydrophile de vitamine E (trolox) qui permet à l’association d’inhiber l’expression de TGF-β1 au niveau de l’ARN messager et de la protéine. Cette action inhibitrice intervient au niveau transcriptionnel et conduit à une inhibition conséquente des cibles de la voie de signalisation TGF-β1/Smad (Col Iα1, FN1, PAI-1, CTGF), alors qu’elle semble sans effet sur la voie de signalisation Rho/ROCK. Pour la première fois, dans ces cellules issues de l’entéropathie radique, nous avons montré une surexpression de miR-210 ; microRNA induit par l’hypoxie. L’association pentoxifylline-trolox inverse la surexpression de miR-210 aussi bien dans les conditions normoxique que dans les conditions hypoxiques. L’implication de miR-210 dans l’entéropathie radique n’a pas été préalablement étudiée, néanmoins nous avons montré qu’un inhibiteur de miR-210 diminue l’expression de Col Iα1 dans ce modèle. L’effet anti-fibrosant exercé par l’association pentoxifylline-vitamine E est partiellement induit par l’inhibition de la cascade TGF-β1. L’inhibition de miR-210 est un deuxième mécanisme potentiel nécessitant d’autres investigations. Cette étude renforce les essais clinique antérieurs en montrant in vitro une synergie entre pentoxifylline et vitamine E et permettant de proposer cette association en première ligne thérapeutique dans la fibrose radio-induite. De plus, miR-210 est proposé comme une possible cible thérapeutique pour traiter la fibrose radio-induite. / Radiation-induced fibrosis is a serious late complication of radiotherapy. Pentoxifylline-vitamin E has proven effective and safe in clinical trials as treatment of fibrosis, while the molecular mechanism of its activity is yet unexplored. We showed efficacy of Pentoxifylline-vitamin E combination in radiation-induced enteropathy in a small clinical study. In parallel, using a unique in vitro model of primary smooth muscle cells isolated from intestinal samples isolated from humans with radiation enteropathy we showed that pentoxifylline and the hydrophilic analogous of vitamin E (trolox) synergize to inhibit TGF-β1 protein and mRNA expression. This inhibitory action is mediated at the transcriptional level and leads to subsequent inhibition of TGF-β1/Smad targets (Col Iα1, FN1, PAI-1, CTGF), while it has no effect on the Rho/Rock pathway. We have also demonstrated, for the first time, an overexpression of the hypoxia-induced microRNA miR-210 in the fibrotic cells. Pentoxifylline-trolox combination could reverse this miR-210 overexpression in normoxic and hypoxic conditions. While miR-210 has not been previously shown to be involved in radiation-induced enteropathy, we showed that miR-210 inhibitor could reduce mRNA expression of Col Iα1. The anti-fibrotic effect of combined pentoxifylline-vitamin E is at least in part mediated by inhibition of the TGF-β1 cascade. MiR-210 inhibition is another mechanism which needs further investigations. This study strengthens previous clinical data showing pentoxifylline-vitamin E synergy and supports its use as a first-line treatment of radiation-induced fibrosis. Also, it suggests miR-210 as a new potential therapeutic target for the treatment of this complication.

A lesão por balão cateterismo é um procedimento comumente utilizado para o estudo dos mecanismos de restenose. O estresse mecânico produzido pela passagem do balão promove alterações tanto na artéria lesada quanto na artéria contra-lateral. Observou-se um aumento da densidade de neurônios que contêm neuropeptídeos como a Substância P (SP) e o Peptídeo Relacionado com o Gene da Calcitonina (CGRP) na artéria contra-lateral à lesão, o que evidencia a ocorrência de um processo neurocompensatório. Observou-se ainda um aumento da reatividade desta artéria à fenilefrina (Phe) e à angiotensina II (Ang II) após 4 e 15 dias da lesão, respectivamente. O objetivo do presente trabalho foi estudar as conseqüências dos tratamentos com celecoxibe (inibidor seletivo da enzima COX-2), -tocoferol (Vitamina E, antioxidante natural) e losartan (antagonista dos receptores AT1), sobre a reatividade à Phe, Cloreto de Potássio (KCl), Acetilcolina (ACh), Ang II de artérias ipsilaterais e contra-laterais em relação a artérias controles. Em animais tratados com celecoxibe (10 mg/Kg - 2 vezes ao dia) ou -tocoferol (400 mg/Kg/dia) por 7 dias. Os resultados mostram que os dois tratamentos normalizam os valores de efeito máximo (Emax) da Phe nas artérias contra-laterais com endotélio aos níveis de artérias controle. Em animais tratados com celecoxibe, a artéria ipsilateral não respondeu à Phe, enquanto no tratamento com -tocoferol mostrou valores de Emax reduzidos em relação a animais controle. Os valores de Emax da ACh em artérias de animais controle e tratados com celecoxibe ou -tocoferol, são idênticos. Entretanto, ambos os tratamentos promoveram redução na potência da ACh em artérias controle quando comparadas com as de animais não tratados. Nos animais controles tratados com losartan a potência desse peptídeo foi menor do que controle e contra-lateral sem tratamento e contra-lateral tratada. Na artéria contra-lateral o Emax do KCL estava diminuído em relação ao controle e o tratamento com losartan não modificou este parâmetro. O Emax da ACh em artérias controle e contra-laterais não foi alterado pelo tratamento com losartan, entretanto houve aumento da potência deste agonista em artérias controles e contra-laterais em relação a artérias de animais controles. Os resultados obtidos no presente estudo indicam que a produção de espécies reativas de oxigênio, prostanóides vasocontritores e Ang II levam a alterações na reatividade aos agonistas estudados na artéria contra-lateral à lesão por cateter balão. / The injury by balloon catheter is a procedure commonly used to study the mechanisms of restenosis. The mechanical stress produced by the passage of the balloon promotes changes both in the injured artery in the contralateral artery. There was an increased density of neurons containing neuropeptides such as substance P (SP) and the Gene Related Peptide of Calcitonin (CGRP) in the contralateral artery to the lesion, which shows the occurrence of a process neurocompensatory. There was also an increase in the artery reactivity to phenylephrine (Phe) and angiotensin II (Ang II) after 4 and 15 days of injury, respectively. Efeitos da associação do beta-caroteno, alfa-tocoferol e do fumo no câncer de pulmão.

Some studies have shown that larger plasmatic indexes of beta-carotene or a great ingestion of foods rich in carotenoids diminish the risk of developing this neoplasia. On the other hand, studies suggest that the smokers' lung can alter the metabolism of beta-carotene, that is, producing greater risk of cell alterations and neoplasias. It is probable that the alpha-tocopherol has beneficial effect on this association. Objective: To evaluate the participation of beta-carotene and alpha-tocopherol in the development of lung cancer induced by urethane and its relationship with cigarette exposition. Material and Method: Balb C mice were divided into 10 groups: G1(cigarette), G2 (cigarette and urethane), G3 (only urethane), G4, G5 and G6 (cigarette, urethane and beta-carotene to 0.25, 0.05 and 0.005%, respectively), G8 (urethane and beta-carotene to 0.25%), G7, G10 and G11 (cigarette, urethane and beta-carotene to 0.25 and alpha-tocopherol 0.25; 0.05 and 0.005%, respectively).The exposition to the cigarette was for 10 minutes, twice a day, 5 days a week, during 16 weeks. For induction of tumors, urethane was administered intraperitonealy, in the dose of 3.0mg/Kg. Nodules and hyperplasias were quantified; morphometric analyses of the nodules were performed. Kruskal-Wallis and of Mann-Whitney tests were used for statistical analysis. Results: Group G3 presented greater number of nodules (P=0.001), smaller stromal fraction (P=0.011) and greater sum of the tumor area (P=0.047) compared to group G2. Group G8 showed smaller number of nodules (P=0.013) and hyperplasias (P=0.05) compared to group G3. Both smaller doses of beta-carotene (G5 and G6) were statistically similar, although with smaller number of nodules when compared to group G4 (P=0.04). Group G4 presented smaller number of hyperplasias than G8, however, the number of nodules did not alter (P=0.045%). Stromal fraction of groups G3 and G4 was similar, although smaller than G2 and G5 (P=0.011). According to the alpha-tocopherol, the stromal fraction of group G7 was greater than the one of the groups G2, G3, G4 and G10 (P=0.011). The 0.25%-beta-carotene diet increased the area of the nodules, demonstrated by the largest area (P=0.03), smaller area (P=0.03), sum of the areas (P=0.018) and average of the areas (P=0.006) in group G4 when compared to G2. Conclusion: According to these results, it was concluded that passive tobacco can be a protecting factor in the evolution of tumors induced by urethane in mice, however, there is no evidence that this could be dose-dependent. The supplementation of beta-carotene in 0.25%-dose can also be a protecting factor, however, associated to passive tobacco it has smaller effect than in lower doses (0.05 and 0.005%). The exposition to cigarette smoke does not alter the number of nodules induced by urethane in mice when in the presence of 0.25%-beta-carotene diet; however the hyperplasia number with the presence of the cigarette diminished. The association of the alpha-tocopherol to the 0.25%- beta-carotene and the passive tobacco produces a protecting factor, mainly in the dose of 0.25%. / O hábito de fumar cigarros é responsável pela morte de milhões de pessoas por ano em todo o mundo, estando diretamente relacionado ao câncer de pulmão. Estudos demonstraram que maiores índices plasmáticos de beta-caroteno ou a grande ingestão de alimentos ricos em carotenóides diminuem o risco de desenvolver esta neoplasia. Por outro lado, existem estudos que sugerem que o pulmão de fumantes pode alterar o metabolismo do beta-caroteno, gerando maior risco de alterações celulares e neoplasias. É provável que o alfa-tocoferol tenha efeito benéfico sobre esta associação. Objetivo: Avaliar a participação do beta caroteno e do alfa-tocoferol no desenvolvimento de câncer de pulmão induzido por uretana e sua relação com a exposição ao cigarro. Material e Método: Camundongos Balb C foram divididos em 10 grupos: G1(cigarro), G2(cigarro e uretana), G3 (uretana), G4, G5 e G6 (cigarro, uretana e beta-caroteno a 0,25; 0,05 e 0,005% respectivamente), G8 (uretana e Beta-caroteno a 0,25%), G7, G10 e G11 (cigarro, uretana e beta-caroteno a 0,25 e alfa-tocoferol a 0,25; 0,05 e 0,005% respectivamente). A exposição ao cigarro ocorreu durante 10 minutos, 2 vezes por dia, 5 dias por semana, por 16 semanas. Para a indução de tumores, foi administrada uretana por via intraperitoneal na dose de 3,0mg/Kg. Foram quantificados nódulos e hiperplasias, realizada análise morfométrica dos nódulos e submetidos aos testes de Kruskal-Wallis e de Mann-Whitney para análise estatística. Resultados: O grupo G3 apresentou maior número de nódulos (P=0,001), menor fração estromal (P=0,011) e maior soma da área de tumor (P=0,047) comparado ao grupo G2. O grupo G8, mostrou-se com menor número de nódulos (P=0,013) e hiperplasias (P=0,05) comparados ao grupo G3. As duas doses menores de beta-caroteno (G5 e G6) se mostraram estatisticamente semelhantes, porém com menor número de nódulos quando comparados ao grupo G4 (P=0,04). O grupo G4 apresentou menor número de hiperplasias que G8, porém o número de nódulos não se alterou (P=0,045). A fração estromal dos grupos G3 e G4 foram semelhantes, porém menores que G2 e G5 (P=0,011). Quanto ao alfa-tocoferol, a fração estromal do grupo G7 foi maior que a dos grupos G2, G3, G4 e G10 (P=0,011). A dieta com beta-caroteno a 0,25% aumentou a área dos nódulos, demonstrada pela maior área (P=0,03), menor área (P=0,03), soma das áreas (P=0,018) e média das áreas (P=0,006) no grupo G4 comparada com o grupo G2. Conclusão: Com estes resultados, concluiu-se que na evolução de tumores induzidos por uretana em camundongos o fumo passivo se comporta como um fator protetor, porém não há evidência de que seja dose-dependente. A suplementação de beta-caroteno na dose de 0,25% também age como protetor, porém associada ao fumo passivo tem efeito menor do que em doses mais baixas (0,05 e 0,005%). A exposição à fumaça do cigarro não altera o número de nódulos quando na presença de dieta com beta-caroteno a 0,25%, porém o número de hiperplasias com a presença do cigarro diminui. A associação do alfa-tocoferol ao beta-caroteno 0,25% e ao fumo passivo gera um fator protetor, principalmente na dose 0,25%.
