I den här studien intervjuas tretton kristna hbt-personer om kristna normers roll i ”komma ut”-processer. Syftet med studien är att söka en förståelse för hur sexuell identitet kan skapasunder ungdomsår utifrån de normer och värderingar som kan finnas i kristna kontexter.Slutsatsen är att det i kristna kontexter ofta finns en stark heteronorm som gör det konfliktfylltför kristna hbt-personer att leva ut sina sexualiteter.För att hälsa ska kunna uppnås för kristna hbt-personer krävs en kristen kultur som inte baraaccepterar, utan även tar ställning för att all kärlek i trohet och respekt är lika värdefull ochlika rikligt välsignad som den heterosexuella.

När konsumenten får tillgång till ny teknologi möjliggörs nya sätt att se på tv och film, detta nya sätt att bestämma över reklamintag har ändrat sättet som konsumenten utsätts för reklam. När konsumtionsmarknaden ändras måste teknologin och marknadsföringstekniker följa efter. Produktplacering är ett sätt att integrera marknadsföring i kontext med kända aktörer och få konsumentens medvetenhet för produkter att öka. Produktplacering är inte ett nytt sätt att marknadsföra produkter men har blivit mer centralt då marknadens intolerans för reklam ökar. Undersökningen grundar sig på att beskriva hur filmindustrin använder sig av produktplacering inom svenskproducerade filmer. Genom att deskriptivt förklara vad produktplacering är och hur produkterna placeras genom produkttyp, tid, position i bild, aktörer och hur det genererar positiva och negativa effekter kan undersökningen skapa en bild över hur produktplaceringen i filmer används i Sverige. Produktplacering undersöks genom observation av två svenskproducerade filmer, Hundraåringen som klev ut genom fönstret och försvann samt Skumtimmen. Filmerna fungerar som en generalisering av den svenska filmindustrin genom att beskriva hur den såg ut år 2013. Företag kan gynnas av kunskapen inom produktplacering och använda teknikerna rätt för att generera avkastning, användningen av felaktiga tekniker kan i stället leda till förluster. I den svenska filmindustrin används vanligtvis produktplaceringstekniker genom att placera produkter medvetet genom visuell synlighet, i mitten av bilden och genom att integrerera produkterna med handlingen. Produkter som gör sig synliga i svenska produktioner varierar från stora produktioner till små, stora produkter har oftast en större variation av produkter än små produktioner men delar samma stora användning av produktplacering av livsmedel och dagligvaror. Mycket av de olikheter som stora och små produktioner har grundar sig på den tillgänglighet som filmen kommer nå ut i men även vilken form av genre filmen har. Det måste finnas en balans mellan filmindustrin och företagen för att största möjliga vinst skall kunna uppnås. Med denna undersökning hoppas jag som skribent kunna möjliggöra ett enklare sätt att undersöka produktplacering för att se att det används på det mest effektiva sättet.

Tidigare forskning visar att HBTQ-personer uppfattar stress och lider av psykisk ohälsa i samband med komma ut-processen. Syftet med studien är att utforska hur HBTQ- personer uppfattar förväntningar i komma utprocessen och undersöka vilka effekter dessa förväntningar har på komma utprocessen. Studien är kvalitativ och genomförd via semistrukturerade intervjuer och studiens urval är 6 homo- och bisexuella personer i varierande åldrar från 18 år. Studiens empiri analyseras via av en kvalitativ analysmetod. Tidigare forskning och empiri från intervjuer ligger till grund för resultat och analys. Studiens resultat visar på att det bland intervjupersonerna inte finns några mönster på hur förväntningarna uppfattas i komma ut-processen. Resultatet visar att intervjupersonerna uppfattar både tilldragande och frånskjutande faktorer som försvårar och möjliggör komma utprocessen. Trygga miljöer, positiv respons och normalisering av intervjupersonernas egna sexuella läggning möjliggör komma ut processen. Rädsla, oro och utsatthet är faktorer som försvårar. Resultatet visar också att stigmatisering, heteronormer och sammanhang problematiserar komma utprocessen. Behovet för intervjupersonerna att komma ut påverkas av sammanhang, normer och attityder. Intervjupersonerna uttrycker både negativa och positiva uppfattningar om förväntningar i samband med att komma ut.

La surexpression de récepteurs couplés aux protéines G (RCPG) dans certains cancers est mise à profit en médecine nucléaire pour développer des radioligands capables de diagnostiquer la présence de tumeurs. A titre d’exemple, des analogues de la somatostatine marqués à l’indium-111 (111In-OctreoScan) sont utilisés pour le diagnostic de tumeurs neuroendocrines. L’urotensine II (UII), qui présente des homologies structurales avec la somatostatine, est considérée comme le neuropeptide vasoactif le plus puissant découvert à ce jour. L’UII interagit avec un unique RCPG de très haute affinité appelé UT, classiquement couplé à la voie Gαq/PLC/IP3/Ca2+. L’UII exerce notamment des activités pro-mitotiques et chémoattractantes et une expression élevée de l'UT a été rapportée dans plusieurs types de tumeurs solides humaines provenant des poumons, de l'intestin, de la prostate ou du sein. Ces données suggèrent que l'UT pourrait être une cible prometteuse pour concevoir des analogues urotensinergiques radiomarqués à finalités diagnostiques voire thérapeutiques. Deux analogues urotensinergiques capables de lier des isotopes radioactifs (le DOTA-UII et le DOTA-urantide) ont été synthétisés et radiomarqués avec succès avec l’111In. L'incubation de l’111In-DOTA-UII dans du plasma humain a révélé que seulement 30% du radioligand étaient dégradés après 3 heures d’incubation. L'administration de concentrations croissantes de DOTA-UII et de DOTA-urantide sur des cellules HEK-293 exprimant l'UT induit une augmentation dose-dépendante de la concentration cytosolique de calcium, avec une puissance et une efficacité similaires à celles obtenues avec l'UII (EC50: 1,26 10-8 M et 2,09 10-8 M, UII et DOTA-UII, respectivement) et urantide (EC50: 1,82 10-8 M et 1,52 10-8 M, urantide et DOTA-urantide, respectivement). Alors que la fixation sur l’UT du DOTA-UII ou l’UII entraîne l'internalisation du complexe ligand-récepteur (ELISA et immunocytochimie) dans les cellules HEK-293 exprimant l'UT, l’urantide et le DOTA-urantide restent inactifs. L'injection intraveineuse de l’111In-DOTA-UII chez des souris C57BL/6 a révélé un léger signal principalement restreint dans les reins, indiquant une clairance rapide du peptide. Des résultats similaires ont été obtenus avec des souris dont le gène codant l’UT a été invalidé (mUTS2R-/-) ou des souris exprimant constitutivement la forme humaine de l’UT (mUTS2R-/- hUTS2R+/+). Enfin, l’111In-DOTA-UII a été injecté chez des souris Nudes porteuses de xénogreffes hétérotopiques de cellules humaines A549 (adénocarcinome pulmonaire) ou DLD-1 (adénocarcinome colorectal), exprimant fonctionnellement l’UT, comme nous l’avons préalablement vérifié par analyses western blot, par immunohistochimie et par des tests de migration/prolifération cellulaire. Dans les deux cas, l'imagerie TEMP/TDM n'a toutefois pas révélé de signal exploitable dans les tumeurs, suggérant que la clairance du radioligand est trop importante pour permettre l'accumulation du radiotraceur et la détection des tumeurs. L’ensemble de nos résultats démontre que la conjugaison de DOTA dans les analogues urotensinergiques n'altère pas l'activation de l'UT. Cependant, d'autres investigations sont nécessaires pour diminuer la clairance rénale et améliorer l'imagerie tumorale et ainsi permettre, à terme, de concevoir des radioligands urotensinergiques à finalités diagnostiques voire théranostiques. / Overexpression of G protein-coupled receptors (GPCRs) in tumor is widely used to develop GPCR-targeting radioligands for solid tumor imaging. For example, somatostatin analogue labeled with 111Indium (111In-OctreoScan) is used for the diagnosis of neuroendocrine tumors. The vasoactive neuropeptide urotensin II (UII), which shares structural analogies with somatostatin, interacts with a single high affinity GPCR named UT. High expression of UT has been reported in several types of human solid tumors from lung, gut, prostate or breast, suggesting that UT is a valuable target to design radiolabeled UII analogues for cancer diagnosis. Two urotensinergic analogues (DOTA-UII and DOTA-urantide) both containing the DOTA chelating group capable of complexing radioactive metal isotopes have been synthetized and radiolabeled with 111Indium. Incubation of 111In-DOTA-UII in human plasma revealed that only 30% of the radioligand was degraded after a 3h incubation period. Administration of graded concentrations of both DOTA-UII and DOTA-urantide in the vicinity of HEK293 cells expressing UT induced a dose-dependent increase in cytosolic calcium concentration, with similar potency and efficacy to that obtained with UII and urantide. These results demonstrated that conjugation of DOTA in urotensinergic analogues did not affect UT activation. DOTA-UII was also able to promote UT internalization in HEK293 cells expressing UT, while DOTA-urantide was ineffective. Intravenous injection of 111In-DOTA-UII in C57BL/6 mice revealed a slight signal mostly restricted in kidney, and similar results were obtained with knock-out mice or constitutively expressing human UT mice. Finally, 111In-DOTA-UII was injected into nude mice bearing heterotopic xenografts of human A549 cells (lung adenocarcinoma) or DLD-1 cells (colorectal adenocarcinoma) both expressing functional UT. In both cases, SPECT-CT imaging showed the absence of tumor uptake and significant renal and bladder uptakes, suggesting fast tracer clearance from the organism. However, further investigations will be necessary to decrease renal clearance and to improve tumor imaging.

Die Untersuchung vergangener Klimavariabilität ist ein einzigartiger Schlüssel zum Verständnis zukünftigen Verhaltens des Erdsystems unter anthropogener Einwirkung. Dies ist von besonderer Wichtigkeit, da es die einzige Realisierung des „Erdsystemexperiments“ ist, die für uns zugänglich ist. Paleoklimaarchive, wie Bäume, Stalagmiten oder Gletscher stellen in ihrer Struktur und Zusammensetzung zeitabhängige Aufzeichungen früherer Klimavariabilität dar. Die statistische Analyse von Zusammenhängen zwischen solchen Zeitreihen kann helfen, die den Paläoklimaproxies zugrundeliegenden Klimaprozesse und, letztlich, der Erdsystemdynamik zu verstehen. Drei Hauptherausforderungen müssen gemeistert werden, um dies möglich zu machen: die Zeitreihen sind unregelmäßig aufgelöst in (i) Zeit, (ii) Raum und die Zeit selbst ist eine Variable die rekonstruiert werden muss, was (iii) zusätzliche Unsicherheiten mit sich bringt. Dazu habe ich den Paläoklimanetzwerkansatz entwickelt, inspiriert von der zunehmenden Anwendung von Methoden aus dem Bereich der komplexen Netzwerke in der Klimatologie. Ich habe Schätzer für Pearson-Korrelation, Transinformation (Mutual Information) und Ereignissynchronisation (Event Synchronization) eingeführt, die keine Zeitreihen mit regelmäßigen Beobachtungsintervallen benötigen. Der Einfluß von Altersunsicherheiten auf Schätzungen solcher Ähnlichkeitsmaße wird numerisch durch Ensembles von möglichen Akkumulationsverläufen abgeschätzt. Ein einfaches Modell für Informationsflüsse im Asiatischen Sommermonsun (ASM) ermöglicht den Test der Fähigkeiten von (Paläoklima-)Netzwerkmaßen, räumlich-zeitliche Klimaänderungen von Zeitreihen räumlich heterogen verteilter Orte zu detektieren. / Investigating past climate changes offers a unique key to understanding the future behavior of the Earth system under anthropogenic perturbation, because it is the only realization of the “Earth system experiment” accessible. Paleoclimate archives such as trees, stalagmites, or glacial deposits provide in their structure and composition time-dependent records of earlier climatic variability. Statistical analysis of dependencies amongst such time series helps to infer on the climatic processes reflected in the paleoclimate proxy data and then, ultimately, on the dynamics of the Earth system. Three inherent technical challenges need to be met: the datasets are heterogeneously sampled in (i) time and (ii) space, and time itself is a variable that needs to be reconstructed which (iii) introduces additional uncertainties. To address these issues I developed the paleoclimate network framework, inspired by the increasing application of complex networks methodology in climate. I introduced estimators for Pearson correlation, mutual information and event synchronization that do not require time series sampled at regular intervals. The impacts of age uncertainty on such similarity estimates was assessed numerically, using ensembles of possible accumulation histories. A simple model for information flow in the Asian summer monsoon (ASM) was used to test the ability of (paleoclimate) network measures to detect spatio-temporal transitions from time series observed at heterogeneous locations in space.

Esta pesquisa tem por objetivo investigar alguns aspectos que permitam sistematizar uma teoria da arte na Veneza do século XVI, a partir de dois tratados artísticos: o Dialogo di Pittura, de Paolo Pino (1548) e o Dialogo della Pittura intitolato LAretino, de Lodovico Dolce (1557). Para isto, enfoca cinco problemas: o conceito de mimesis; a estrutura dos tratados em sua relação com a Arte Retórica; Pino e Dolce como representantes da doutrina do ut pictura poesis; o ideal de beleza e a disputa pela primazia da cor sobre o desenho. / This research deals with some aspects that enable to systematize an artistic theory in Venice from sixteenth century, from two artistic treatises: Paolo Pinos Dialogo di Pittura (1548) and Lodovico Dolces Dialogo della Pittura intitolato LAretino (1557). In order to get over this objective we will discuss five problems: mimesis notion; the treatises structure in your relation with Rhetoric; Pino and Dolce as ut pictura poesis doctrine members; the ideal of beauty and the debate for the primacy of colour over design.
