[pt] A apresentação de aplicações hipermídia dinâmicas pode ser vista em
alguns casos como um processo de autoria recursivo, no qual aplicações são
recriadas em tempo de apresentação quando modificações no conteúdo são
desencadeadas por interações entre a máquina de apresentação e outras entidades
(tais como usuários, objetos imperativos, aplicações externas etc.). Em alguns
cenários de aplicações hipermídia dinâmicas, é possível identificar um padrão de
composição hipermídia que se mantém mesmo após o documento ter sido
recriado. Aplicações desse tipo são comuns, por exemplo, em um ambiente de
TV Digital Interativa. A existência de um padrão durante a apresentação de uma
aplicação para TV Digital Interativa permite estabelecer uma analogia entre os
processos de recriar documentos dinamicamente e de especificar aplicações pelo
método orientado a templates. Nesse último método, o autor especifica uma
aplicação preenchendo lacunas deixadas por um template que representa o
padrão de composição hipermídia da aplicação. Analogamente, na recriação
dinâmica de documentos o módulo que processa atualizações no documento faz
o papel de preencher as lacunas de templates. O objetivo principal do presente
trabalho é definir uma arquitetura, inspirada nessa analogia, para estruturar
aplicações NCL que podem ser refatoradas dinamicamente e que se mantêm em
conformidade com seus respectivos templates. Nested Context Language (NCL)
é a linguagem para autoria de aplicações do Sistema Brasileiro de TV Digital
terrestre. Para validar a solução proposta, foram desenvolvidas uma aplicação
que captura um cenário real de uso e uma ferramenta de autoria que permite
especificar graficamente o preenchimento de templates. / [en] The presentation of dynamic hypermedia applications may be seen as a
recursive authoring process, in which applications are recreated during
presentation time, whenever content changes are triggered by interactions
between the presentation engine and other entities such as users, imperative
objects, external applications, etc. In some scenarios of dynamic hypermedia
applications, it is possible to identify a hypermedia composition pattern that
remains consistent even after the document is recreated. This kind of
applications is common, for instance, in an Interactive Digital Television
environment. The presence of such a pattern throughout the presentation of an
application for Interactive Digital Television allows the establishment of an
analogy between recreating documents dynamically and authoring applications
through a template-driven authoring method. Using the latter, the authoring
process is conducted by filling gaps left by a template that represents the
hypermedia composition pattern of an application. Analogously, in the dynamic
document re-creation, the module that processes document updates fulfills the
role of filling the templates gaps. The main goal of the present work is to
define an architecture, inspired by this analogy, to structure NCL applications
that can be dynamically refactored and that remain conform to their respective
templates. Nested Context Language (NCL) is the language of Brazilian System
of Digital Terrestrial Television applications. In order to validate the proposal,
an application that captures a real scenario and an authoring tool for specify
graphically document filling was developed.

Previous issue date: 2016-10-07 / As linguagens declarativas normalmente atuam no desenvolvimento de aplicações multimídia, pois suas características permitem suportar adequadamente a natureza assíncrona e descritiva dessas aplicações. Neste cenário, a robustez surge como um fator determinante para a qualidade dessas aplicações, dada a vasta quantidade de plataformas e dispositivos usuais, que, ocasionalmente, apresentam problemas de execução. Neste contexto, as especificações dessas linguagens são de grande importância para o processo de desenvolvimento, pois além de direcionar a codificação, definindo as restrições léxicas, sintáticas e semânticas, também formalizam como os conteúdos devem ser executados pelos interpretadores. Este trabalho tem por objetivo investigar a viabilidade de se aplicar análises estruturais e comportamentais orientadas a conformidade para o desenvolvimento de aplicações multimídia, a fim de eliminar a dependência de interpretadores para atestar sua corretude. Para tal, a linguagem NCL foi utilizada como alvo do estudo, devido à sua representatividade para o problema. / Declarative languages are typically used in the development of multimedia applications, as its features allow to properly support the asynchronous and descriptive nature of these applications. In such a scenario, robustness appears as a determining factor in the quality of these applications, given the vast amount of platforms and devices that occasionally have implementation problems. In this context, specifications of these languages are of great importance to the development process, as they impose lexical, syntactic and semantic restrictions, and also formalize how content must be interpreted. This work aims at investigating the feasibility of applying structural and behavioral analysis oriented to compliance to the development of multimedia applications in order to eliminate dependence on interpreters to prove its correctness. To this end, NCL was used as a target of study due to its problem representativeness.

[pt] Ginga-NCL e a linguagem NCL foram recomendadas como ambiente de
multimídia interativo e linguagem para IPTV, pela União Internacional
das Telecomunicações (ITU do Inglês: International Telecommunication
Union). Para promover o uso dessas tecnologias, é necessária a criação de
ferramentas que ajudem a desenvolver aplicações usando NCL. Atualmente,
o suporte de ferramentas para o desenvolvimento em NCL é um pouco
limitado. Existem apenas poucos sistemas que auxiliam o desenvolvimento
de aplicações NCL. Esses programas não ajudam muito os desenvolvedores
em avaliar se o código NCL está bem escrito. Neste trabalho apresentamos
o NCL-Inspector, um sistema de críticas para código NCL, que objetiva
aumentar a capacidade do desenvolvedor em detectar aplicações NCL
suscetíveis a erros. Também foram levantados requisitos desejáveis em
sistemas de críticas para código. A forma como esses requisitos foram
apresentados possibilita que sejam aplicados em qualquer sistema de crítica
de código fonte. / [en] Ginga-NCL and the NCL language were selected as a recommendation as
the interactive multimedia environment and language for IPTV by the
International Telecommunication Union (ITU). In order to promote the
use of these technologies, it is necessary to create tools to help developing
applications using NCL. At present, the support provided by tools for NCL
development is quite limited. There are only few available systems that can
create NCL applications. These softwares do not help developers in assessing
the quality of the written NCL code. In this dissertation, we propose NCLInspector,
a critique system of the NCL code, which aims at leveraging
the developer s skills in detecting error-prone NCL applications. Also, we
specified the requirements for critiquing systems for source code inspection.

[pt] Em 2010, o Ministro das Comunicações instituiu o Sistema Brasileiro de Rádio Digital (SBRD). No entanto, um modelo de referência para o sistema ainda não foi definido. Este trabalho apresenta uma análise da radiodifusão sonora terrestre no Brasil e a partir desta são inferidos alguns requisitos para um rádio digital interativo. Esses requisitos são então utilizados para nortear a pesquisa. A relevância e importância do uso da NCL e do Ginga no rádio digital brasileiro, assim como acontece no Sistema Brasileiro de TV Digital (SBTVD), são discutidas, e a definição da forma como deve ser feito o transporte de aplicações NCL nos dois sistemas de rádio digital sendo considerados para adoção pelo país é realizada. Um novo perfil da linguagem NCL para o rádio digital é definido. Esse novo perfil recebeu o nome de perfil Digital Radio, ou simplesmente DR. O Ginga também é definido para uso em receptores de rádio digital, sendo que novos exibidores de mídia e recursos adaptados ao contexto do rádio digital são introduzidos. A implementação do Ginga da PUC-Rio é apresentada com as modificações necessárias para o rádio digital. Adicionalmente, um ambiente para validar a interatividade no rádio digital com o Ginga foi montado e testes exercitando toda a cadeia de transmissão e recepção de rádio digital com aplicações NCL embutidas foram realizados. As definições e conclusões que resultaram da pesquisa visam contribuir para a definição de um Sistema Brasileiro de Rádio Digital que seja poderoso em termos comunicacionais e agregue os recursos mais relevantes para o meio na era digital. / [en] In 2010, the Minister of Communications instituted the Brazilian Digital Radio System (SBRD), however a reference model for the system has not yet been set. This text presents an analysis of the terrestrial radio broadcasting in Brazil and presents some requirements for an interactive digital radio. These requirements are then used to guide the research. The relevance and importance of the use of NCL and Ginga in the Brazilian digital radio as in Brazilian Digital TV System (SBTVD), are discussed, and the definition of how the transport of NCL applications should be done in the two digital radio systems being considered for adoption by the country is performed. A new profile of NCL for use in digital radio is defined. This new profile was named Digital Radio Profile, or just DR. Ginga is also defined for use in digital radio receivers, and new media players and features adapted to the digital radio context are introduced. PUC-Rio s implementation of the Ginga middleware is presented with the necessary modifications for digital radio use. In addition, an environment to validate the interactivity in digital radio with the Ginga was assembled and tests exercising the entire digital radio chain of transmission and reception with embedded NCL applications were performed. The definitions and conclusions that resulted from research activities are expected to contribute to the definition of a Brazilian Digital Radio System that is powerful in communicational terms and aggregates the most relevant technologies for the medium in the digital age.

Cell biological defects in juvenile neuronal ceroid lipofuscinosisSchultz, Mark 01 December 2013 (has links)
Mutations in the CLN3 gene cause Juvenile Neuronal Ceroid Lipofuscinosis (JNCL), a form of Batten disease that is grouped within the broad class of lysosomal storage diseases. JNCL displays a primary central nervous system phenotype characterized by rapid onset blindness, wide spread brain atrophy and reversal of learned abilities with death occurring 10-20 years after symptom onset. The mechanisms underlying these phenotypes are not known. CLN3 encodes CLN3, a protein with no known molecular function. CLN3 is expressed at very low levels natively in most cells, and is highly hydrophobic.
Similar to other lysosomal storage diseases, it is difficult to ascertain the primary versus the secondary defects when the protein functions along the endosomal-lysosomal pathway. In JNCL one common finding among several labs, in various cellular systems, is a fluid-phase endocytotic defect. I took this commonality as a key to CLN3 function, and pursued cell biological pathways required for fluid-phase endocytosis. Fluid-phase endocytosis is regulated by cycling of the small GTPase Cdc42 and I discovered increased Cdc42-GTP in CLN3-null mouse brain endothelial cells. In mouse brain endothelial cells enhanced Cdc42-GTP increased Cdc42 dependent signaling, filopodial formation, and retarded cell migration. I also found reduced plasma membrane association of ARHGAP21, a known negative regulator of Cdc42. My data supports a model where loss of CLN3 reduces ARHGAP21 plasma membrane recruitment, and causes aberrant Cdc42 activation. Thus irregular Cdc42 activation underlies the commonly reported fluid-phase endocytic defects in JNCL.
Therapeutic development for JNCL has been hampered in part from the varying phenotypes ascribed to CLN3 deficiency. My discovery that the fluid-phase endocytic defects result from Cdc42 pathway aberrations, which in turn contributed to multiple downstream phenotypes, opened the door to novel JNCL therapeutics. Here I present work showing that a Cdc42 inhibitor corrects the Cdc42 dependent defects in vitro and multiple defects in a JNCL mouse model.

Aplicações Sensíveis ao Contexto utilizam informações contextuais do usuário para acionar serviços de acordo com a necessidade ou a situação atual do usuário, e podem, portanto, potencialmente enriquecer a experiência do usuário ao assistir TV. O objetivo desta dissertação é propor uma metodologia orientada a modelos para auxiliar o desenvolvimento dessas aplicações no ambiente de TV Digital (TVD), desde a fase de modelagem à realização, tendo como alvo de implementação a plataforma Ginga, que é o middleware definido pelo SBTVD para desenvolvimento de aplicações interativas. A metodologia oferece suporte à modelagem do universo de discurso da aplicação através de modelos de contexto e situação, bem como à definição de comportamentos reativos de aplicações usando uma abordagem baseada em regras. Para isso, este trabalho propõe uma linguagem baseada em regras denominada ECA-DL TVD, que permite a especificação de comportamentos reativos através de eventos, condições e ações. Este trabalho também define uma arquitetura conceitual, que visa estruturar o Gerenciador de Contexto, um dos componentes do Ginga. Uma das principais contribuiçoes deste trabalho volta-se para a geração automática de código NCL referente a (parte) dessa arquitetura conceitual. O trabalho propõe a utilização de técnicas da área de desenvolvimento orientado a modelos (MDD) e frameworks de transformação para que, a partir de modelos de contexto, situações e regras ECA-DL TVD, seja gerado código NCL. Finalmente, são apresentados dois cenários de aplicação que visam demonstrar a validade da metodologia proposta.

[pt] Um sistema de TV Digital (TVD) é definido por um conjunto de especificações que determinam as tecnologias envolvidas na codificação, transmissão, recepção e apresentação de conteúdos, incluindo a especificação da aplicação (programa não-linear), seus vários objetos de mídia e metadados relacionados. Nesse cenário, o suporte a aplicações é realizado por uma camada intermediária de software, ou middleware, posicionada, no ambiente de recepção, entre o código das aplicações e a infra-estrutura de execução (plataforma de hardware e sistema operacional). O projeto e implementação de um middleware
para receptores de sistemas de TVD trazem uma série de desafios. Entre eles estão: a eficiência na utilização de recursos, usualmente escassos no dispositivo receptor; o suporte à evolução dinâmica das funcionalidades do middleware; o suporte à recuperação de falhas em tempo de execução; os mecanismos para gerência de localização de recursos, permitindo o uso da mesma sintaxe de autoria em ambientes distintos de recepção; o suporte a edição ao vivo de programas não lineares, ou seja, das aplicações; a definição de uma infra-estrutura de transporte assíncrono de aplicações interativas e comandos de controle e; o controle do ciclo de vida das aplicações interativas, permitindo que aplicações possam ser iniciadas,
pausadas e retomadas em qualquer ponto de sua duração, sem perder o histórico de sua evolução. As soluções da maioria dessas questões presentes nos sistemas existentes apresentam algumas limitações importantes, e em alguns casos nem mesmo existem, sendo o problema apenas contornado. O presente trabalho propõe soluções alternativas para as questões levantadas, e as incorpora na especificação do middleware declarativo Ginga-NCL e em sua implementação de referência. O middleware Ginga-NCL e sua linguagem declarativa NCL foram adotados pelo SBTVD-T em 2007. No início de 2009, Ginga-NCL e NCL se tornaram parte dos padrões ISDB-TB e parte da recomendação ITU-R BT 1699. Ainda no início de 2009, Ginga-NCL e NCL tornaram-se a Recomendação ITU-T H.761 para serviços IPTV. / [en] Digital TV (DTV) systems are defined by a set of specifications that establish the technologies involved in the content encoding, transmission, reception and presentation, including the specification of applications (non-linear programs), their various related media objects and metadata. In this scenario, support to applications is accomplished through an intermediary software layer, or middleware, positioned, in the receiving environment, between the application code and the execution infrastructure (hardware platform and operating system). The middleware design and implementation bring a number of challenging issues.
Among them are: efficient resource management, since resources are usually scarce in DTV receiver devices; support to dynamic evolution of the middleware functionalities; support to fault recovery at runtime; the mechanisms for resource location management, allowing using the same syntax used in the authoring environment the different receiver environments; support to live editing of nonlinear programs (i.e. applications); the infrastructure definition for the asynchronous transport of interactive applications and control commands; and the life cycle control of interactive applications, allowing applications to be started, paused and resumed at any point in their life duration, without losing their evolution history. Most of these issues are addressed in the existing systems, however with important limitations; some of them are not even addressed, being only treated with workaround tricks. This work proposes alternative solutions to the mentioned issues and incorporates these solutions in the Ginga-NCL declarative middleware specification and in its reference implementation. Ginga- NCL and its declarative NCL language were adopted by SBTVD-T in 2007. In early 2009, Ginga-NCL and NCL have become part of the ISDB-TB standard and part of the ITU-R BT 1699 Recommendation. Even in early 2009, Ginga-NCL and NCL have become the ITU-T H.761 Recommendation for IPTV services.

Les peptides Cα-thioester jouent un rôle central dans la synthèse de protéines par voie chimique : ils sont des partenaires clés dans la ligation chimique native (NCL), réaction qui a révolutionné ce domaine. L’accès à ces peptides par Fmoc-SPPS, méthode largement utilisée dans les laboratoires, est encore restreint à des experts. Cette limitation représente à l’heure actuelle un frein à la démocratisation de la synthèse de protéines par NCL. Le premier volet de cette thèse décrit la conception et le développement d’une nouvelle méthode bioinspirée de synthèse de précurseurs de peptides thioester, fondée sur un dispositif de thioestérification intramoléculaire de type N-(2-hydroxy-5-nitrobenzyl)-cystéine (N-Hnb-Cys). La synthèse des peptides portant ce dispositif a été totalement automatisée à partir de réactifs peu couteux et ne requière pas d’étapes post-SPPS avant la NCL. La conception biomimétique du dispositif –ce dernier opérant via un réarrangement par transfert d’acyle N→S–, résulte en des cinétiques de NCL rapides à pH neutre. Un large éventail de précurseurs de thioester a été synthétisé, ceci nous a permis d’explorer le potentiel et les limites de la méthodologie, tout en soulignant son efficacité même pour des « séquences difficiles » et de longs peptides. Cette approche a été appliquée à la synthèse par ligation d’une gamme représentative de peptides naturels riches en cystéines, issus du venin d’un serpent, de mollusques, de primates ou encore de plantes. En particulier, nous avons décrit la première synthèse d’une Big-défensine (93 acides aminés), issue de l’huitre et dont l’activité biologique est en cours d’évaluation. Par ailleurs, une voie synthétique originale pour accéder à des peptides comprenant une cystéine C-terminale a été proposée. Elle repose sur l’introduction de cet acide aminé par NCL, évitant les réactions secondaires inhérentes aux approches existantes. Nous avons également appliqué notre approche à la synthèse par NCL intramoléculaire du squelette peptidique cyclique de plusieurs produits naturels, avec des rendements remarquables. L’ensemble de nos résultats est extrêmement encourageant pour la généralisation de notre approche. / Peptide Cα-thioesters play a prominent role in the chemical synthesis of proteins: they are key partners in native chemical ligation (NCL), a reaction that has revolutionized the field. Nonetheless, access to such peptides via the widely used Fmoc-SPPS is still limited to experts. This limitation is currently an obstacle to further popularization of NCL-based protein synthesis. The first part of this thesis describes the design and optimization of a new bio-inspired methodology for the synthesis of peptide thioester precursors, based on an N-(2-hydroxy-5-nitrobenzyl)-cysteine intramolecular thioesterification device (N-Hnb-Cys). Synthesis of peptides bearing this device was fully automated, starting from inexpensive materials. Importantly, no post-SPPS steps are required prior to NCL. The biomimetic design of the device –that operates through an N→S acyl shift–, results in fast NCL kinetics at neutral pH. A broad range of thioester precursors has been synthesized; this allowed us to explore the scope and limitation of the methodology, while stressing its efficiency even for demanding sequences and long peptides. This approach has been applied to the ligation-based synthesis of a representative variety of naturally occurring disulfide-rich peptides (DRP) sequences, from snake venom, molluscs, primates and plants. In particular, we have described the first synthesis of a Big-defensin (93 amino acids), discovered in oyster and whose biological activity is currently under evaluation. Furthermore, an original method for the synthesis of C-terminal cysteine-containing DRP has been proposed. It is based on the introduction of this amino acid through NCL, avoiding side reactions inherent to existing approaches. We have also applied our approach to the intramolecular NCL-based synthesis of the cyclic backbone of several DRP, with remarkable yields. Altogether, our results are extremely encouraging for the generalization of this methodology.

production of new genres, sub-genres of narrative in television products. It shows that
television, from computational systems, becomes from a connected device to a
transmission system in a continuous line for a semi-open system of processing events,
linking more closely the production system of the content to the user viewer. Presents a
description of the technical and conceptual patterns of Digital TV in the world and in Brazil,
its architecture and, in particular, the software tools for developing of interactive
applications for the T-SBTVD (Sistema Brasileiro de TV Digital-Terrestrial). Analyzes their
technological possibilities, towards a methodological analysis, which involves the narrative
structures (properties and classifications), showing how these can be considered in an
interactive narrative inside the current systems of TVDi. Discusses types of interactive
narratives already tried, which are taken as references, showing how the Complex
Narratives, the Narratives in Expanded Cinema, Hypermedia and last in Games (Digital
Games)., currently insert on the context of planning current and future TVDi. From the
obtained results, presents the development of a prototype of application of an Interactive
Narrative, using current technology standards within the SBTVD-T. Concludes in favor of a
broader discussion of the issue to contribute to the adequacy of narrative forms currently
used in television concencional to the new system in its emerging phase, focusing on the
essential aspects of interactivity with the user-viewer, a glimpse toward the new way to
watch TV without losing the current characteristics, but differentiates itself from other
medias / A presente pesquisa relaciona o conceito de TV Digital Interativa, em seus aspectos
técnico-computacionais, relacionando-os com as novas formas de expressão de conteúdo
no sentido de possibilitar a produção de novos gêneros, subgêneros narrativos dentro de
produtos televisivos. Mostra que a televisão, a partir dos sistemas computacionais se
transforma, de um aparelho conectado a um sistema de transmissão em linha contínua
para um sistema semi-aberto de processamento de eventos, associando mais fortemente
o sistema produtivo do conteúdo ao usuário-telespectador. Apresenta a descrição dos
padrões técnico-conceituais da TV Digital no mundo e no Brasil, sua arquitetura e, em
particular, as ferramentas de software para o desenvolvimento de aplicações interativas
para o SBTVD-T (Sistema Brasileiro de TV Digital-Terrestre). Analisa as suas
possibilidades tecnológicas, na direção de uma análise metodológica, a qual envolve as
estruturas narrativas (unidades e classificações), refletindo como estas podem ser
consideradas em uma narrativa interativa dentro dos atuais sistemas da TVDi. Discute
tipos de narrativas interativas já experimentadas, que são tomadas como referências,
mostrando como as Narrativas Complexas, as Narrativas no Cinema Expandido, na
Hipermídia e por último nos Games(Jogos Digitais)., se inserem atualmente no contexto
do planejamento da atual e futura TVDi. A partir dos resultados obtidos, apresenta o
desenvolvimento de um protótipo de aplicação de uma Narrativa Interativa, utilizando as
tecnologias padrões atuais dentro do SBTVD-T. Conclui em favor de uma discussão mais
ampla da questão no sentido de contribuir para a adequação das formas narrativas
atualmente utilizadas na televisão concencional ao novo sistema em sua fase emergente,
enfocando os aspectos essenciais da interatividade com o usuário-telespectador, na
direção do vislumbrar uma nova maneira de se ver TV, sem perder as atuais
características, porém diferenciando-se dos demais meios
