Štíhlá výroba a její implementace / Lean Manufacturing and its Implementation

The diploma thesis deals with the problematics of lean production in a selected company. The analytical part includes the introduction of the company and the analysis of the current state with using lean manufacturing tools to optimize material flows. The thesis is divided into four parts. The first part describes the objectives and methodology, followed by the theoretical basis of the work and analysis of the current state of the company. The final part includes suggestions of possible solutions leading to elimination or improvement of identified problems.

Detta är en studie av artikelplaceringar på logistikföretaget som är placerade pålagerplatser som inte gynnar artiklarnas omsättning. Lagerplatser som är ogynnsamma avA-klassicerande artiklar har påverkat plockfrekvens negativt. Det har bidragit att ledtidenfrån att hämta en A- artikel tar längre tid än att hämta en annan A-artikel med lägreomsättning. Studien identifierar de negativa aspekter som påverkar plockfrekvens och vill bidra medförbättringsförslag för logistikföretaget. Studiens metod skapades genom att samla in datafrån både primär-och sekundärdata. Frågeställning var en del av strukturerad observationoch olika vetenskapliga artiklar relaterade till ämnesområdet samt litteratursökning varen del av primärdata. Studien har resulterat att orsakerna som är ABC-klassificering och Lean har en kopplingtill sambandet mellan artikelplacering och plocktid. Orsakerna som förekommer harbidragit negativ påverkan hos logistikföretaget. Vidare har studien kommit fram till ett resultat genom intervjuer och litteraturteorier därlogistikföretaget förlorar många produktionstimmar kring orsaken. Logistikföretagetbehöver implementera nya åtgärder för förbättring och utveckling. Slutsatsen iundersökningen redovisar vad logistikföretaget ska åtgärda de negativa aspekterna sompåverkar artikelplacering och plocktiden. • Minimera slöseri genom att förkorta gångavståndet. • Omsortera artiklarna. • Kompetensutveckling inom Lean och 5S för att jobba smartare. • Implementera olika standarden som hjälper till att förnya arbetssätt ochutveckling. • Upprätthålla ordning och reda genom att identifiera artiklar som behöverplaceras om och leverantörer. • Utöva mer av parametrar (försäljning, uttag och plockfrekvens). • Effektivisera lagerhanteringssystem (in-och utleveranser, plock, lagerplatser). Arbetet avgränsas till en del av artikeloptimering inom logistik och placering av Aklassificerade artiklar samt påverkan i plockfrekvensen. COVID-19 har varit understudiens tid ett hinder som begränsade studiens författare vistelse hos logistikföretaget. / The purpose of the survey is to investigate article placements at the logistics companythat are unproductively located. Unproductive places of A-classing articles have had anegative effect on picking frequency, which has contributed to the lead time to retrieveone A-article taking longer than retrieving another A-article with lower turnover. The purpose of the survey is to identify the negative aspects that affect the pickingfrequency in order to provide the logistics company with information about the aspectsthat indicate improvement. The method survey was created by collecting data throughboth primary and secondary data. Structured observation and questioning were part of a semi-structured interview andvarious scientific articles related to the subject area as well as literature search were partof secondary data. The investigation has resulted in the current reasons behind theconnection between article placement and picking time having been negatively affectedby the logistics company. Furthermore, research has come to a result where the logistics company loses manyproduction hours around the cause that need to implement new measures for improvementand development. The conclusion of the survey reports that the logistics company shouldaddress the negative aspects that affect item placement and picking time. • Minimize waste by shortening the walking distance. • Rearrange the items. • Skills development within Lean and 5S to work smarter. • Maintain order by identifying items that need to be repositioned and suppliers. • Exercise more parameters (sales, withdrawals and picking frequency). • Streamline inventory managementsystems (inbound and outbound deliveries,picks, warehouse locations). The work was limited to part of article optimization in logistics and placement of Aclassified articles and the impact on the picking frequency. During the study, COVID-19 has been an obstacle that limited the study's authors to stay longer with the logisticscompany.

The subject of this thesis is to propose a new inventory management system, that was found in particular company as one of the biggest shortages, which the company currently faces. The first part of thesis contains theoretical background focused on inventory management, expecially on function and types of supplies, ordering systems of supplies and on methods of their inventory. The second part focuses on the analysis of the current state in company processes. Following the theoretical part, there were suggested appropriate inventory management systems to simplify the work in warehousing and logistic, but also to bring economic benefits to the whole company. In addition to these proposals were included also possible solutions of less problematic processes.

Les plantes de la famille des légumineuses ont acquis la capacité d’accueillir au sein d’organes symbiotiques, les nodosités, des bactéries fixatrices d’azote appelées bactéroïdes. Cette symbiose permet aux plantes hôtes de satisfaire leurs besoins en azote. Certaines légumineuses produisent au sein des nodosités une grande famille de peptides antimicrobiens particuliers, les NCR (Nodule-specific Cysteine-Rich). Ils permettent à l’hôte de contrôler la population bactérienne intracellulaire via leurs activités antimicrobiennes, mais aussi d’imposer aux rhizobia une différenciation terminale des bactéroïdes. Durant ma thèse, j’ai participé à l’identification et à la caractérisation de la protéine bactérienne BclA. Ce transporteur ABC est nécessaire pour la formation de bactéroïdes différenciés lors de la symbiose Aeschynomene-Bradyrhizobium. Pour mieux comprendre sa fonction symbiotique, j’ai étudié la relation entre BclA et une enzyme de modification du peptidoglycane, la DD-carboxypeptidase 1. J’ai pu montrer que ces deux facteurs agissent de manière indépendante dans la mise en place de bactéroïdes différenciés. Une analyse fonctionnelle de BclA et une expérience d’évolution expérimentale avec le mutant bacA de Sinorhizobium, un orthologue de bclA, apportent une meilleure compréhension du rôle de ces transporteurs in vivo. L’ensemble des résultats obtenus pendant ma thèse suggère que BclA et BacA assurent la résistance bactérienne face aux NCR in planta, comme un prérequis pour la suite du processus, mais ne sont pas nécessaires à la différenciation per se. De plus, leurs activités d’import des NCR ne semblent pas être le mécanisme sous-jacent du processus de résistance. / Plants of the legume family have acquired the ability to host in specific symbiotic organs, the roots nodules, nitrogen fixing bacteria, called bacteroids. This symbiosis allows plants to fulfill all their nitrogen requirements. Some legume plants produce in their nodules a large family of antimicrobials peptides called the NCRs (Nodule-specific Cysteine-Rich). Their antimicrobial activities allow the host plant to control the intracellular bacterial population. NCRs peptides also govern terminal differentiation of the bacteroids. During my PhD work, I participated in the identification and characterization of BclA. This bacterial ABC transporter is involved in bacteroid differentiation during the Aeschynomene- Bradyrhizobium symbiosis. In order to better understand its symbiotic function, I studied the link between BclA and a peptidoglycan-modifying enzyme, the DD-carboxypeptidase 1. I was able to show that these two factors act in an independent manner in the establishment of bacteroid differentiation. A functional analysis of BclA and an experimental evolution on Sinorhizobium bacA mutant, an orthologue of bclA, conferred a better understanding of the in vivo role of these transporters. The results obtained during my thesis suggest that the BclA and BacA function is to ensure bacterial resistance to NCRs, as a prerequisite for the bacterial differentiation process, but is not needed for differentiation per se. Furthermore, their NCR uptake activities do not seem to be the mechanism underlying the resistance.

Récemment, la grande complexité des applications modernes, par exemple dans la génétique, l’informatique, la finance, les sciences du climat, etc. a conduit à la proposition des nouveaux modèles qui peuvent décrire la réalité. Dans ces cas,méthodes MCMC classiques ne parviennent pas à rapprocher la distribution a posteriori, parce qu’ils sont trop lents pour étudier le space complet du paramètre. Nouveaux algorithmes ont été proposés pour gérer ces situations, où la fonction de vraisemblance est indisponible. Nous allons étudier nombreuses caractéristiques des modèles complexes: comment éliminer les paramètres de nuisance de l’analyse et faire inférence sur les quantités d’intérêt,dans un cadre bayésienne et non bayésienne et comment construire une distribution a priori de référence. / Recently, the great complexity of modern applications, for instance in genetics,computer science, finance, climatic science etc., has led to the proposal of newmodels which may realistically describe the reality. In these cases, classical MCMCmethods fail to approximate the posterior distribution, because they are too slow toinvestigate the full parameter space. New algorithms have been proposed to handlethese situations, where the likelihood function is unavailable. We will investigatemany features of complex models: how to eliminate the nuisance parameters fromthe analysis and make inference on key quantities of interest, both in a Bayesianand not Bayesian setting, and how to build a reference prior.
