Kinetics and structure-guided characterisation and engineering of aldehyde deformylating oxygenase (ADO) for a renewable microbial biofuel platform

The increased demand for an alternative form of fuel has raised a great interest towards exploring various metabolic pathways and enzymes in several microbial species for hydrocarbon production. In recent years, cyanobacteria have emerged as an attractive microbial host and cyanobacterial metabolic pathways were targeted for engineering to produce "drop in" fuels such as propane and butane. Whilst appealing, practicalities for producing biofuels in cyanobacteria remain challenging, requiring the identification and engineering of natural biocatalysts and their integration into metabolic processes. Cyanobacterial hydrocarbon biosynthesis arises from fatty acid metabolism involving a potential enzyme, aldehyde deformylating oxygenase (ADO), which catalyses the decarbonylation of long-chain fatty aldehydes to alkanes, mainly in the conversion of octadecanal (C17H35CHO) to heptadecane (C17H36) and formate. The substrate specificity and preferences for long-chain aldehyde by ADO necessitates a detailed kinetic and structural characterisation in order to optimise/engineer this enzyme for future biotechnological applications. Thus, the main objective was to identify a potential ADO enzyme that can be optimised for shorter chain alkane production. By studying the substrate specificity and reaction kinetics of different ADO enzymes, it was found that ADO from Prochlorococcus marinus MIT 9313 (PmADO) is a potential target for short chain alkane production. The crystal structural of PmADO was solved and further GC-MS analysis was carried out to identify the chemical origin of a mixture of long-chain fatty acid in the active site, originated from E. coli cells during recombinant over-expression and purification. It was suggested that the structure-guided protein engineering for short-chain alkane production should be carried out along with the removal of this adventitious ligand from the active site in order to increase the alkane production. Four important residues present at the entrance of the ligand-binding cavity were targeted and saturated mutagenesis was performed on PmADO to identify variants that excluded the long fatty acid ligands from the active site but have specificity and higher conversion rates for shorter chain aldehydes. This identified two variants, V41Y and A134F, with the A134F variant that not only exhibiting an improved activity and turnover value of PmADO by four-fold but also improved binding affinity for butyraldehyde by 2 times. Finally the improved variants were incorporated in a host organism (E. coli) and the possibilities for the development of a microbial platform for renewable propane synthesis based on a fermentative clostridial butanol pathway were explored. Four pathways were designed namely atoB-adhE2, atoB-TPC7, nphT7-adhE2 and nphT7-TPC7 routes, which utilise CoA intermediates selected to incorporate ADO as the terminal enzyme. When PmADO was co-expressed with these pathways, the engineered E. coli host produced propane. The atoB-TPC7-ADO pathway was the most effective in producing propane (220 ± 3 μg/L). By (i) deleting competing pathways, (ii) including a previously designed A134F variant ofPmADO with an enhanced specificity towards short-chain substrates, and (iii) including a ferredoxin-based electron supply system, the propane titre was increased up to 3.40 ± 0.19 mg/L. It was also shown that the best propane producing pathways are scalable in a 250 mL flask and in a large-scale (up to 30 L) fermentor setup. This thesis focuses on the detailed kinetics and structure-guided characterisation and engineering studies on the ADO enzyme for the development of a renewable microbial biofuel platform.

Com a finalidade de avaliar empiricamente os resultados decorrentes da utilização desse instituto desde 1988, procedeu-se ao mapeamento dos padrões decisórios manifestados pelo Supremo Tribunal Federal nas ações dessa espécie. Essa análise tornou explícitos fatores que influenciaram os julgamentos proferidos pelo tribunal mencionado, especialmente quanto aos tipos de interesses e direitos que foram tutelados por meio da ADO. Os dados levantados durante a pesquisa possibilitaram que se avaliasse concretamente a efetividade da ADO, ou seja, se referido instituto realmente tem se mostrado apto a atingir as finalidades que ensejaram sua criação. Para tanto, tornou-se necessária investigação prévia a respeito dos objetivos a cuja consecução a ADO é voltada, os quais foram buscados nos trabalhos desenvolvidos pela Assembleia Nacional Constituinte de 1987-1988, na jurisprudência do STF e na doutrina jurídica brasileira. Os resultados decorrentes dessa investigação prévia foram, então, confrontados com os elementos empíricos relacionados à ADO, os quais revelam que sua utilização tem servido, de modo relevante, para provocar a atuação estatal nos casos de omissão inconstitucional, bem como apontam para uma diferenciada receptividade do STF às demandas de interesse corporativo. Constatou-se, portanto, que a concepção doutrinária segundo a qual a ADO seria incapaz de concretizar os fins a que se destina em razão da ausência de atribuição de poderes mais intensos ou ativos ao STF não encontra respaldo nos dados empíricos relativos ao instituto. Trata-se de entendimento baseado em considerações meramente teóricas que, além de incompatíveis com os dados empíricos referidos, também se opõem a elementos teóricos extraídos do debate entre Kelsen e Schmitt acerca de quem deve ser o guardião da Constituição. / The object of the present dissertation is the Direct Action of Unconstitutionality by Omission (ADO), which is a procedural modality created by the Constitution of 1988 to address the issue of violation of constitutional norms that demand the State’s positive action. With the objective of empirically evaluating the outcomes resulting from this institute since 1988, a survey was undertaken with regards to the ruling patterns by the Federal Supreme Court concerning these types of judicial claims. This analysis rendered explicit factors that have influenced the decisions ruled by the abovementioned Court, especially in regards to the types of interests and rights that have been secured by means of the ADO. The data that has been raised throughout the research has enabled the correct evaluation of the effectiveness of the ADO, i.e., whether said institute has effectively been apt to reach the intentions that resulted in its creation. For such purposes, a previous investigation concerning the objectives for which the ADO is aimed was fundamental, which were pursued in the works developed by the National Constitutional Assembly of 1987-1988, the Federal Supreme Court jurisprudence, and the Brazilian juridical doctrine. The results from this previous investigation were then confronted with the empirical elements related to the ADO, which reveal that its usage has served, in a relevant manner, to provoke the State’s action in cases of unconstitutional omission, and have also indicated a differentiated receptivity by the Federal Supreme Court in regards to judicial actions of corporate interest. It was therefore verified that the doctrinal conception according to which the ADO would be incapable of realizing its intended purposes because of the absence of more intense or active powers from the Federal Supreme Court does not find support in the empirical data related to the institute. The latter is an understanding based merely on theoretical considerations that, not only are incompatible with the abovementioned empirical data, but also contradict theoretical elements extracted from the debate between Kelsen and Schmitt concerning who should be the guardian of the Constitution.

Os dados apontam para a import?ncia da aten??o psicol?gica ao n?cleo familiar em processos de ado??o tardia. / Adoption establishes a filiation status, resulting from a legal act, which attributes to the child and parents the rights and obligations associated with such condition, being legally irrevocable. Nevertheless, in practice there are adoptions that do not concretize and the child returns to justice during or even after the legal process is closed. Late adoption is the denomination of the adoption of children over two years and it is still permeated by myths and stigmas, leading to a frequent return of the child to justice in these cases. The late adoption involves a process of building a unique relationship with a child whose backstory is commonly marked by the dissolution of the relationship with the family of origin, due to violation of rights and, in some cases, the experience of institutional care. Given such a scenario, this research, based on the Existential Analytic proposed by Martin Heidegger, seeks to understand the experience of mothers and children in the process of late adoption, in order to obtain subsidies to psychological attention in this context. This is a qualitative, phenomenological study with a comprehensive focus. The participants were two mothers and two children who have gone through late adoption for about two years. The procedures of data generation contemplated narrative interviews with mothers and individual meetings with children, in which ludic resources were used as mediators of expression (free drawings, unfinished children's story and "Story-Drawings" on late adoption). The procedures were audiotaped and transcribed. Data analysis was grounded in Heidegger's hermeneutics. The late adoption process, permeated by historical, social and cultural determinants and the web of meanings that create the historical singularity of each person involved have proved to be complex as seen in the narratives. The construction of the meanings of parenthood and filiation has been developing in the families in the study, from the experience of being-with-the-other, caring and dwelling in their peculiar modes of expression. The family of origin and the adoptive family mingle and differentiate by means of the experience of children, especially because of the existence of biological siblings. Data point to the importance of psychological care to family core in late adoption processes

This research aimed to seize and analyze the experience of exercising homoparenthood via adoption in the already said city and identify possible difficulties in the adoption process and daily life of the families surveyed, seeking also to analyze the process of social visibility, inclusion and family and community life of homo-affective family with children / the adoptive / as, especially in within the family, work and formal education / the kids / the. Following a qualitative approach, we used as a strategy for obtaining and building data, interviews with 04 families who realized Homoaffectives adoption in the city of Natal / RN, in a period of time well stablished. The interviews were compounds by a list of questions that sought to contemplate the adoption process, the difficulties / biases in the adoption process, experience as parents / mothers gays / lesbians and the rights of LGBT people in Brazil. The choice for the interview was due to the possibility of rapprochement with the daily lives of these families, in an attempt to identify difficulties and possibilities in homoparental exercise. The questioning of these themes in the interviews and systematized and analyzed responses reveal trends: statement that the choice to adopt as a means of family formation was due to the existence of a prior desire to be father and mother; full support of / family, both in relation to adoption, and in relation to sexual orientation; the assertion of the absence of any kind of difficulty and / or prejudice during the adoption processes and daily activities, both at work and in family and school / the kids / the while, paradoxically, some narratives have signaled clear situations of prejudice and discrimination, whether racial-ethnic or sexual orientation; non-participation and withdrawal from social movements and LGBT movements.

A aprecia??o dos dados mostra que o ambiente organizacional fornece press?o em como a tecnologia vem sendo adotada nesses estabelecimentos. Ficou evidenciado que os hoteleiros buscam adotar tecnologia para parecerem leg?timos perante o mercado, principalmente entre os consumidores. Para isso adotam padr?es, imitam a concorr?ncia, e buscam serem bem avaliados no ambiente virtual em sites como TripAdvisor, Booking, Decolar, entre outros, para que desse modo sejam aceitos pelo campo em que operam e prolonguem sua sobreviv?ncia no mercado. Conclui-se que o isomorfismo institucional est? presente na ado??o de tecnologia no setor hoteleiro analisado e que, a partir da pesquisa, ? poss?vel obter uma perspectiva de como essas for?as atuam nas organiza??es. / As the tourism sector incorporates technologies reconfiguring the distribution and use of products and services, Information and Communication Technology (ICT) becomes a subject of tourism research in view of such transformations in the sector. When looking at the studies that discuss the adoption of technology by tourism operators, these are focused on identifying the diversity or factors that differentiate organizations and their patterns from others in relation to the adoption of technology. Institutional theory establishes a macro view of structural change, defined as isomorphism in organizations. The general objective of the study is to analyze how this isomorphic pressure, based on institutional theory, is present in Natal / RN hotels in the adoption of information and communication technologies. For this, the research conducted interviews with 20 hotels in the city. The data collection instrument consisted of a semi-structured script, which was applied in person during the period from September to November 2017. For the evaluation of the data, the content analysis technique was used, with the use of the qualitative and mixed analyzes Software Nvivo, in its version 11.0. The results shows that the organizational environment provides pressure on how technology has been adopted in these establishments. It was evidenced that hoteliers adopt technology to appear legitimate to the market, especially among consumers. For this they adopt standards, imitate the competition, and seek to be well evaluated in the virtual environment in sites like TripAdvisor, Booking, Decolar, among others, so that they are accepted by the field in which they operate and prolong their survival in the market. It is concluded that the institutional isomorphism is present in the adoption of technology in the analyzed hotel sector, from the research, it is possible to obtain a perspective of how these forces act in the organizations.

Revelatory deceptions in selected plays by William Shakespeare

This dissertation is concerned with the paradox of revelatory deception a form of 'lying' which reveals truth instead of concealing it in four Shakespearean plays: Much Ado About Nothing, As You Like It, Hamlet, and King Lear. Through close analysis, I show that revelatory deceptions in these plays are metatheatrical, and read them as responding to contemporary writers who attacked the theatre for being inherently deceitful. This reading leads to the identification of parallels in the description of theatre in antitheatrical texts and the descriptions of revelatory deceptions in the plays. I suggest that correlations in phrasing and imagery might undermine antitheatrical rhetoric: for example, the plays portray certain theatrical, revelatory deceptions as traps which free their victims instead of killing them. Such 'lies' are differentiated from actual deceits by their potentially relational characteristics: deceptions which reveal the truth require audiences to put aside their self-interest and certainty to consider alternative realities which might reflect, reconfigure, and expand their understanding of the world and of themselves. The resulting truths lead either to the creation or renewal of relationships, as in Much Ado About Nothing and As You Like It, or offer glimpses at the possibility of renewal, which is ultimately denied, as in Hamlet and King Lear. A popular perception persists that the Franciscan friars of Romeo and Juliet and Much Ado About Nothing bear heavy blame for the results of the play, adversely for Friar Lawrence and positively for Friar Francis. The friars do formulate similar plans, but their roles vary significantly. I contrast their approaches using Michel Foucault's definition of pastoral power, with Friar Lawrence as an overly manipulative friar controlling the lovers in spiritual matters, and Friar Francis as a humble military friar returning from the Wars of Religion to share his authority with others. This distinction--especially with Friar Lawrence appearing chronologically first--demonstrates Shakespeare as more fluid in religious themes, contrary to a significant body of scholarship that asserts Shakespeare's pro-Catholic sympathies.

Long-Time Response With Ado-Trastuzumab Emtansine in a Recurrent Metastatic Breast Cancer

Breast cancer is the most common cancer in a woman with a five-year survival of patients with metastatic disease is estimated at 23%. Ado-trastuzumab emtansine (T-DM1) is a HER2-antibody drug conjugate currently approved for the treatment of HER2-positive pre-treated metastatic breast cancer (BC). We report a case of recurrent metastatic breast cancer with unusually lengthy progression-free survival (PFS) on T-DM1 chemotherapy. She was diagnosed with Triple Positive Stage IIIC multifocal invasive ductal carcinoma of the left breast. After completing neoadjuvant chemotherapy, she underwent a bilateral mastectomy. Final pathology showed partial response. Postoperatively, she received adjuvant chemotherapy and radiation therapy. She was started on Q21 days trastuzumab following completion of adjuvant chemotherapy. Systemic imaging showed liver lesions and biopsy confirmed recurrence. She was started on T-DM1, endocrine therapy with anastrozole was continued. She is currently status post 45 cycles. T-DM1 was approved for the treatment (single-agent) of HER2-positive, metastatic BC based on phase III data from the EMILIA and TH3RESA study. Median PFS in the T-DM1 arm was 9.6 months. Herein, we present a case of a woman with recurrent triple positive metastatic BC with a lengthy progression-free survival on T-DM1 chemotherapy.

It is understood the existence of these periods due to the predominance and repetition of certain behaviors in the social life of the 19 th , 20 th and early 21 st centuries, enabling the authors used to build the bibliography in this work to use such nomenclature. However, it is necessary to keep two aspects in mind: time is continuous and does not admit intersections; and the behaviors in each moment are not tight and the only verifiable ones in that time. Based on this, the study highlights, in each of the three moments, the most frequent attitudes mentioned in several books, as able to bring special characterization as well as a differentiated definition. After that, it verifies the relation between these moments and the formation and development of Rio Grande do Sul Ado Malagoli Museum of Art, electing three dates: the year of its official opening in 1957, pointing out characteristics from Modernity; the year it was moved to the building at Alfândega Square in 1978, highlighting traits of Post-Modernity; and the year of the exposition Art in France 1860-1960: The Realism , in 2009, stressing aspects of Hypermodernity. The crossing of the selected dates, by simple sampling, and characteristics of each of the periods previously mentioned are dealt with in the study, showing how social behaviors towards the museum may reflect characteristics of the chosen moments of culture. In order to develop the theme, the concept of social memory coined by Maurice Halbwachs is used along with an analysis of photographies, news reports, documents, interviews, exposition catalogues and attending sheets, and bibliography review which support the proposed analysis / O trabalho analisa pontos característicos de três momentos da cultura: Modernidade, Pós-Modernidade e Hipermodernidade. Estabelece-se e aceita-se, como ponto inicial, a partir do qual será desenvolvido o presente estudo, o entendimento de que existem os períodos acima nominados, tendo em vista a preponderância e repetição de determinados comportamentos na vida social do séculos XIX, XX e início do século XXI, autorizando, dessa forma, a nomenclatura adotada pelos autores consultados ao longo da elaboração do presente texto. Lembra-se, no entanto, dois pontos: o tempo é contínuo e não admite intersecções, e os comportamentos existentes em cada momento apontado não são estanques e únicos naquele espaço temporal. A partir disso, o estudo irá ressaltar, dentro de cada um dos três momentos, as atitudes mais ocorrentes, citadas pela literatura, como capazes de trazer uma caracterização especial, assim como uma definição diferenciada. A seguir, verificar-se-á a relação desses períodos com a formação e o desenvolvimento do Museu de Arte do Rio Grande do Sul Ado Malagoli, elegendo três datas emblemáticas: o ano da sua inauguração oficial, em 1957, destacando características encontradas na Modernidade; o ano da sua mudança para o prédio da Praça da Alfândega em 1978, ressaltando traços relacionados com a Pós-Modernidade; e o ano da realização da exposição Arte na França 1860-1960: O Realismo , em 2009, salientando pontos que trazem referências à Hipermodernidade. O entrelaçamento das datas eleitas e as características de cada um dos períodos supracitados são trabalhados no estudo, demonstrando como os comportamentos sociais em relação ao museu podem refletir as características dos momentos da cultura escolhidos.
