Développement de l'imagerie de perfusion cérébrale par marquage des spins artériels / Development of brain perfusion imaging using arterial spin labeling

Ce travail de thèse, principalement méthodologique, s'est intéressé aux techniques d'imagerie par résonance magnétique (IRM) permettant de mesurer le flux sanguin cérébral (CBF) et plus particulièrement aux techniques de marquage de spins artériels (ASL), qui utilisent les protons de l'eau du sang comme marqueur. Nous avons mis en place la séquence ASL de marquage pseudo-continu (pCASL) et évalué sa réponse à un stimulus hypercapnique. Nous avons évalué différentes stratégies pour optimiser l'efficacité d'inversion. Pour cela, nous avons également mis en place des outils de simulations numériques des approches ASL. Nos résultats démontrent que l'efficacité d'inversion est influencée par l'homogénéité du champ magnétique dans la région de marquage, ce qui pose un problème à haut champ magnétique. Le protocole pCASL optimisé a ensuite été évalué chez le rat à trois champ magnétiques (4.7, 7, et 11.7T) et comparé avec une approche en ASL continu classique (CASL). Cette comparaison a montré une excellente reproductibilité inter-animal et inter-champ de la méthode développée. Dans une seconde partie, nous nous sommes également intéressés à l'influence du temps de relaxation longitudinal (T1) du tissu cérébral sur les valeurs du CBF calculées. Pour cela, nous avons modifié le T1 du tissu par une injection intra-cérébrale de manganèse. Cette étude a montré la difficulté de prendre en compte le changement de T1 du tissu. Dans une troisième partie, nous avons évalué l'apport d'une antenne de marquage spécifique pour l'approche CASL en comparant les mesures de CBF obtenues avec celles de la pCASL. Nous avons observé une bonne concordance entre ces deux méthodes à travers les coupes. Nos résultats illustrent également l'importante contribution du transfert d'aimantation dans les séquences de CASL. Les outils développés au cours de cette thèse sont en cours d'application dans des protocoles d'étude de modèles de tumeurs cérébrales, d'accident vasculaire cérébral et de traumatisme crânien. / This PhD work, mainly methodological, focused on the techniques of magnetic resonance imaging (MRI) to measure cerebral blood flow (CBF) and more particularly on arterial spin labeling (ASL), which uses water protons from the blood as markers. We implemented the pseudo-continuous ASL labeling sequence (pCASL) and evaluated its response to a hypercapnic stimulus. We evaluated different strategies to maximize the labeling inversion effeciency. For this, we implemented numerical simulation tools of ASL approaches. Our results demonstrated that the inversion efficiency is influenced by the homogeneity of the magnetic field in the labeling region, which becomes a problem at high magnetic field. The optimized pCASL protocol was then evaluated in rats at three magnetic fields (4.7, 7, and 11.7T) and compared with a conventional continuous ASL approach (CASL). This comparison showed excellent inter-animal and inter-field reproducibility of the developed method. In a second part, we evaluated the influence of the longitudinal relaxation time (T1) of brain tissue on the final CBF values. For this, we modified the tissue T1 by an intracerebral injection of manganese. This study demonstrated the difficulty of taking into account the change in T1 of the tissue. In a third part, we evaluated the contribution of a specific labeling coil for the CASL approach by comparing measurements of CBF obtained with that of pCASL. We found a good agreement between these two methods for all slices. Our results also illustrate the significant contribution of magnetization transfer effects in CASL sequence. The tools developed during this thesis were also used in several preclinical studies including brain tumors, stroke and head trauma models.

Ska vi lägga pennan åt sidan? : en studie om för- och nackdelar med att skriva sig till läsning med dator utifrån sex lärares perspektiv

Syftet med undersökningen är att framhäva pedagogers perspektiv på för- och nackdelar med arbetet kring strategin att skriva sig till läsning. Metoden vi valt är kvalitativa intervjuer. Intervjuerna genomfördes med sex verksamma pedagoger som alla arbetar med att skriva sig till läsning med hjälp av dator. Pedagogerna fick inte några frågor innan intervjutillfället. Forskningsbakgrunden tar upp ett historiskt perspektiv på läs- och skrivinlärningen, verksamma lärare som arbetar med pedagogiken samt hur användandet av tekniken i strategin fungerar. Styrdokumentens syn på informations- och kommunikationsteknik belyses liksom pedagogernas roll i arbetet med att skriva sig till läsning. Studien visar att fördelarna med att skriva sig till läsning framförallt är att:  Undervisningen är lätt att anpassa efter individens behov och intresse.  Elever med läs- och skrivsvårigheter kan lyckas på lika förutsättningar som övriga elever.  Sambandet mellan skriv- och läslärandet tydliggörs i arbetet med att skriva sig till läsning. Största nackdelen är när informations- och kommunikationstekniken inte fungerar. Vår slutsats är att strategin är aktuell i dagens samhälle men att det fortfarande har bedrivits för lite forskning kring området. Ämnesord: ASL, IKT, skola, samspel, pedagogik

ASL- att skriva sig till läsning : Datorn som skrivdidaktisk verktyg / ASL reading througt writing : The computer as a writing didactic tool

Studien syftar till att undersöka hur fyra lärare utformar det tidiga läs- och skrivlärandet samt i vilken utsträckning datorn inkluderas i lärandet. Studien utgår från det sociokulturella perspektivet, via en kvalitativ ansats med intervjuer och observationer har lärarna beskrivit och åskådliggjort hur de använder datorn i deras undervisning. Studiens resultat visar att datorn ger ett mervärde i undervisningen då den bland annat bidrar till ett lugnare klassrumsklimat, att fler behov kan mötas samt att det är enklare att individanpassa och variera undervisningen. Parallellt med detta framgår att variationen är stor när det handlar om i vilken utsträckning datorn används och att grunden till stor del beror på lärarens digitala kompetens. Studiens resultat visar även att datorn inte bara medför möjligheter, utan även utmaningar i gestalt av tekniska komplikationer vilket kräver en god teknisk infrastruktur för att undervisningen med datorn ska fungera.

Deaf Bilingual Education: A Return to Our ASL Roots

The purpose of this thesis is to show the historical discrepancies in literacy skills of deaf students in the United States and how they correlate to language philosophies and the resulting educational practices. These philosophies are described chronologically beginning with the first formal educational institution, in 1817, followed by the Oralism Movement, the philosophy of Total Communication, and the Bilingual-Bicultural Approach. All three language approaches can be seen today in schools for the deaf across America, but only the bilingual-bicultural approach builds a foundation in a natural L1 (American Sign Language) in an effort to develop an L2 (English).

I denna undersökning fokuseras undervisningsmetoden Att skriva sig till läsning (ASL).Utifrån Arne Trageton (2014) beskrivs hur arbetet med metoden kan gå till i förskoleklass och årskurserna 1-2. Sedan presenteras tidigare forskning och övrig litteratur genom att belysa fördelar med metoden, varför metoden fungerar och nackdelar med metoden. Tidigare forskning är av delad mening vad det gäller om metoden är något att arbeta med i undervisningen eller inte och denna undersökning ska försöka svara på frågor som tidigare forskningar efterfrågar.Syftet med studien är att undersöka lärares uppfattningar av metoden Att skriva sig till läsning(ASL). Vilka för- och nackdelar har metoden för elevers lärande? Hur anser lärarna att metoden stöttar elever med svårigheter? Hur bidrar ASL- metoden till elevers lärande i jämförelse med mer traditionella undervisningsmetoder?Undersökningen grundar sig i en kvalitativ metod i form av intervju. Då undersökningen syftar till att utforska lärares uppfattningar och deras tolkningar av verkligheten gör sig metoden rättvis i denna undersökning. Informanterna i undersökningen är lärare som undervisar i förskoleklass eller årskurs 1-2 och som använder sig av metoden ASL.Resultatet i undersökningen visar stora fördelar med ASL. Metoden upplevs av lärarna att gynna alla elever och då särskilt de elever med läs- och skrivsvårigheter. Lärarna ser att eleverna presterar bättre i arbete med metoden i jämförelse med mer traditionella undervisningsmetoder.

Emergent Literacy Development: Case Studies of Four Deaf ASL-English Bilinguals

The research is clear; given the opportunity to do so, children begin transacting with print at very young ages (Ferreiro & Teberosky, 1982). Deaf children with full access to language from birth frequently experience higher success rates in literacy acquisition (Kuntze, 1998). However, there remains a paucity of studies on how young Deaf children whose success with literacy development can be reasonably predicted, begin their journeys toward literacy. With the understanding that early literacy experiences significantly impact all children's literacy development (Bus, Van Ijzendoorn, & Pelligrini, 1995), it is important to have a clearer understanding of how Deaf children develop emergent literacy skills.This dissertation presents a year-long case study on four young Deaf children from native-ASL families who were immersed in literacy-rich environments and how they developed literacy skills in school and at home. In order to provide the fullest possible picture, parents, teachers and children were interviewed and observed. As literacy development does not happen in isolation; this dissertation provides information about the children's sociocultural context that included the literacy experiences and beliefs of the adult participants and the children's own experiences at home and in school. Artifacts including writing samples and data from an early literacy checklist were also collected to provide information about each child's individual written language development.The data were organized and analyzed based on salient themes and framed by socio-psycholinguistic studies on hearing children by researchers such as Dyson (1993), Ferreiro & Teberosky (1982), and Goodman (1996). Results show that with full access to language and opportunities to develop reading and writing abilities, Deaf children's emergent literacy development is highly similar to that of monolingual and bilingual hearing children with some characteristics unique to Deaf ASL-English bilinguals. The results of this dissertation study adds to the general body of knowledge of how children develop literacy abilities even when they do not have face-to-face communication in their literate language. The results also inform current practices in Deaf education and provide researchers, educators, and parents with a framework for understanding the critical role that language and communication play on Deaf children's literacy development.

Exploring the benefits of a separate course in ASL fingerspelling and numbering to develop students’ receptive competency

In Canada, the field of study related to teaching American Sign Language fingerspelling and numbering is very new. This study focused its examination on the potential educational benefits when incorporating fingerspelling and numbering within an ASL course as opposed to the potential educational benefits when creating a separate fingerspelling and numbering course for second language learners. The study was conducted by administering surveys, pre-tests, and post-tests with students from two Canadian Deaf Studies Programs, as well as interviewing instructors. The participants’ receptive skill acquisition and complex rule comprehension of fingerspelling and numbering was examined in both instances (with and without a separate course) but no distinct differences were found. Data analysis of instructor responses indicated a strong inclination for advanced studies of fingerspelling and numbering for students who continue on to higher levels of education, specifically in American Sign Language-English Interpretation Programs. Additional research is needed, as this study was limited due to the small number of participants. In general, the results of the study confirm that fingerspelling and numbering are challenging practices for second language learners of ASL and that more curricular materials, focused on teaching these skills in natural conversational contexts, are needed. / October 2015
