Charakteristisch für Perlcellulose als Regenerat vom Typ Cellulose II sind sphärisch geformte, poröse Partikel mit einer hohen spezifischen Oberfläche und einer guten Bioverträglichkeit. Aufgrund ihrer Eigenschaften sind diese Cellulosemikropartikel besonders gut für medizinische Anwendungen geeignet. Im Mittelpunkt der Arbeit standen Herstellung, Charakterisierung und Modifizierung von Perlcellulosen mit Partikelgrößen von etwa 1 bis10 µm.
Im Rahmen der Arbeit wurden zunächst sieben technische Cellulose-2,5-acetate mit vergleichbaren molekularen Eigenschaften auf ihre Eignung zur Herstellung von Perlcellulose nach dem in EP0750007 beschriebenen Acetatverfahren untersucht. Dabei erfolgte der Vergleich verschiedener Eigenschaften. Aus allen untersuchten Celluloseacetaten können Perlcellulosen synthetisiert werden. Als besonders geeignet erwies sich ein Produkt mit einer Molmasse von über 100.000, einem Verhältnis der Molmasse zur numerischen Molmasse von etwa 1,5 und einer guten Löslichkeit in Ethylacetat / Methanol (100:17,5). Die hergestellte Perlcellulose hat eine geringe Partikelgröße und eine relativ enge Größenverteilung. Damit erfüllt dieses Cellulose-2,5-acetat alle Anforderungen für die Synthese von Perlcellulose.
Der entscheidende Verfahrensschritt zur Herstellung von Perlcellulose ist das Dispergieren der Emulsion mittels Inline-Ultraturrax. Die Partikelgrößenverteilung hängt im Wesentlichen von den Bedingungen während des Dispergierens ab. Im Rahmen der Arbeit gelang die reproduzierbare Herstellung von Cellulosemicrospheres mit einer Partikelgröße unter 5 µm.
Für die Herstellung von Cellulosemikropartikeln mit definierten Eigenschaften ist neben den Synthesebedingungen auch die Charakterisierung der Perlcellulosen von entscheidender Bedeutung. Dafür wurden zunächst etablierte Verfahren verwendet (Partikelgrößenmessung, REM und Quecksilberporosimetrie). Parallel dazu erfolgte die Entwicklung bzw. Einführung neuer Methoden. Im Vordergrund stand die Untersuchung des Sedimentationsverhaltens der Perlcellulosen durch analytisches Zentrifugieren. Davon ausgehend konnte ein Verfahren zur Berechnung der Porosität aus dem Sedimentationsvolumen entwickelt werden. Zum Nachweis der kompletten Deacetylierung der Proben wurde die Ramanspektroskopie genutzt. Durch die Anwendung vorhandener und die Entwicklung neuer Methoden wird die genaue Einstellung von Eigenschaften der Perlcellulosen (z. B. Partikelgröße und deren Verteilung, Morphologie sowie Porosität) ermöglicht und deren Reproduzierbarkeit gewährleistet.
Weitere Versuche hatten die Entwicklung von Endotoxinadsorbermaterial auf Basis von Perlcellulose und Polymyxin-B-sulfat (PMB) zum Ziel. Die Kopplung des PMB erfolgte meist nach Aktivierung der Proben mit Epichlorhydrin. Zunächst wurde die eingesetzte Epichlorhydrinmenge variiert, um das Optimum für die Aktivierung der Perlcellulosen zu finden. Weiterhin wurden unterschiedliche Mengen PMB angebunden und die Anbindung an nicht aktivierte Proben untersucht. Die Planung aller Versuche erfolgte jeweils nach Auswertung der an der Donau-Universität Krems durchgeführten Limulus- Amöbocyten- Lysat (LAL)-Tests. Mittels dieser Batchtests wurde die Wirksamkeit des Endotoxinadsorbermaterials sowohl im Vergleich zu unbehandeltem Blutplasma und als auch zu kommerziell erhältlichen Adsorbern auf Polystyrenbasis getestet. Endotoxinadsorber, die bei diesen Tests besonders gut bewertet wurden, konnten in einem up-scale- Versuch erstmals in größeren Mengen synthetisiert werden. Auch die direkte Herstellung von Endotoxinadsorbermaterial aus Perlcelluloseacetat konnte realisiert werden. Bei diesem neu entwickelten Verfahren erfolgen Deacetylierung und Aktivierung in einem Schritt. Damit kann die Herstellung vereinfacht werden.
Zur Gewährleistung der Erstfehlersicherheit in extrakorporalen Blutreinigungssystemen sollen magnetisierte Perlcellulosepartikel als bioverträgliche Marker eingesetzt werden. Versuche zur Magnetisierung von Cellulosemikropartikeln während des Herstellungsprozesses zeigten, dass die Einbindung von Magnetit bei Erhalt der sphärischen Partikelstruktur prinzipiell auch auf diesem homogenen Syntheseweg möglich ist. / Bead cellulose is regenerated cellulose II characterized by spherically shaped, porous particles with a high specific surface and a good biocompatibility. Because of their properties these cellulose microspheres are especially suited for medical applications. The focus of this work was the synthesis, characterization and modification of bead cellulose with particle sizes between 1 to 10 µm.
In the frame of this work seven technical cellulose-2.5-acetates were investigated with regard to their suitability for making bead cellulose according to the process described in EP0750007. These cellulose acetates have comparable molecular characteristics. Different properties were compared. Bead celluloses can be synthesized from all investigated cellulose acetates. A product with a molecular weight of more than 100,000 and with a ratio between molecular weight and numeric molecular weight of about 1.5 is special suited. This cellulose-2.5-acetate has a good solubility in ethyl acetate / methanol (100:17.5). The bead cellulose made from it has a low particle size and a relative narrow size distribution. Thus this cellulose acetate complies with the requirements for making bead cellulose.
The most important process step for making bead cellulose is the dispersing of the emulsion using an inline-ultraturrax. The distribution of particle size depends mainly on the conditions during dispersing. A reproducible synthesis of cellulose microspheres with a particle size range below 5 µm was successfully achieved.
In addition to determining conditions for manufacturing bead cellulose the characterization of the microspheres is essential to obtain bead cellulose with well defined properties. At first well-established methods of characterization were used (particle size measurement, SEM and mercury porosimetry). In parallel new methods were developed and implemented. The main focus was the investigation of sedimentation behaviour of bead cellulose using analytical centrifugation. Based on this knowledge of the sedimentation volume a new method to calculate the porosity was designed. Raman spectroscopy was used for detecting the complete deacetylation of the samples. By using well-established and newly developed methods properties of bead cellulose such as particle size and distribution, morphology and porosity can be accurately adjusted. In this way the reproducible synthesis of cellulose microspheres can be ensured.
The aim of further experiments was to develop an endotoxin adsorber material based on a coupling of bead cellulose with Polymyxin B sulfate (PMB). The coupling with PMB was carried out after activation of the samples by using epichlorohydrin. At first the added epichlorohydrin amount was diversified in order to find the optimum for the activation of bead cellulose. Later the coupling of different amounts of PMB took place and the linking of PMB to non activated samples was investigated too. The planning of all experiments occurred after evaluation of Limulus amebocyte lysate (LAL) tests at Danube-University Krems. Using these batch tests the effectiveness of the endotoxin adsorber material was tested compared to untreated blood plasma as well as commercial available adsorbers based on polystyrene. Endotoxin adsorbers showing the best adsorption rate were then synthesized for the first time in larger quantities. Also the direct synthesis of endotoxin adsorber material based on bead cellulose acetate could be realized. Using this newly developed method, deacetylation and activation occur during the same step. This means manufacturing process can be simplified.
Using magnetized bead cellulose as biocompatible marker particles is planned to achieve first fault safety in case of a membrane rupture during extracorporeal blood purification. Initial tests have shown that the magnetization of cellulose microspheres is possible during the manufacturing process. The incorporation of magnetite can be realized while keeping the spherical shape of the particles using this homogenous synthesis pathway.

Il a été démontrè que dans les deux cas Geq de manière non ambigus sèpare l’état fluide (Geq = 0) de l’état vitreux (Geq > 0). Nous avons aussi insisté sur le fait que Geq pour la couche mince dépend de lépaisseur du film h mais aussi de de la pression tangentielle qui est un résultante de la procédure de préparation de la couche mince / Focusing on the equilibrium shear modulus Geq of well-known glass-forming polymer model system (sampled by means of MD), we have addressed the general question of how the mechanical properties of the thin polymer films differs from the bulk. Using ”stress fluctuation” formalism we obtained Geq(T) for the bulk and films. It has been demonstrated that in both cases Geq unambiguously separates the fluid state (Geq = 0) from the glass (Geq > 0). We also stressed that Geq for the film does not only depend on film thickness h, but also on tangential pressure that is a consequence of the film preparation procedure.

Since ancient times, indigenous plants have been used by traditional healers for treating
various ailments. Sutherlandia frutescens is one of the most commonly used medicinal plants
of southern Africa. This widely distributed plant has been traditionally used to treat cancer
and HIV patients; however scientific validation is still in high demand. This research aimed to
phytochemically characterise the various extracts prepared and to determine if any
chemotypes were present. Subsequent biological characterisation was carried out to
preliminary ascertain whether this medicinal plant could have anti-cancer and/or immunemodulating
properties and which compounds might be responsible for these actions.
Various traditional and organic extracts were prepared. Extracts, fractions and compounds
generated were analysed and chemical profiles obtained. Column chromatographic techniques
were used to isolate and purify compounds and structure elucidation was carried out using
various analytical techniques. Sulforhodamine B and cytometric bead array assays were

À l'approche de la transition vitreuse les fondus de polymères montrent une augmentation importante de la viscosité de plusieurs ordres de grandeur. Le but de cette étude est d'acquérir une compréhension plus profonde des propriétés viscoélastiques des fondus de polymères vitrifiables. Les polymères sont modélisés comme des chaînes flexibles en utilisant un modèle de bille-ressort. Les propriétés dynamiques sont analysées dans le cadre de la théorie de couplage de mode idéale. Nous constatons que la température critique de couplage de mode varie avec l'inverse de la longueur de chaîne. En étudiant la fonction de relaxation de cisaillement, nous constatons que les processus de relaxation polymériques, ne sont pas modifiés, mais décalés vers des temps plus importants en approchant la transition vitreuse. / Polymer melts show a remarkable increase of their viscosity by many orders of magnitude on approaching the glass transition. The aim of this study is to gain a deeper insight into the viscoelastic properties of glass forming polymer melts. The polymers are modeled as flexible chains using a bead-spring model. The dynamic properties are analyzed in the framework of the ideal mode-coupling theory. We find that the critical temperature of the ideal mode-coupling theory scales with the reciprocal chain length. By studying the shear relaxation function we find that the polymer relaxation processes are not altered but shifted to later times in the approach of the glass transition.

Les particules soumises à un écoulement peuvent manifester des orientations préférées et une variété de déformations en fonction de leur géométrie et élasticité et du champ de vitesse de l'écoulement. A l’inverse, les flux peuvent être modifiés lorsque les contraintes des particules sont non négligeables. Cette thèse présente des résultats théoriques et numériques sur cette relation bilatérale en deux parties. La Ière partie commence par une analyse de stabilité et des simulations numériques qui montrent une particule brownienne semi-flexible dans un écoulement élongationnel effectuant un retournement, un phénomène associé aux flux de cisaillement. Le Chap. 2 étend les outils analytiques dédiés aux modèles simples ou aux flux indépendante du temps pour les modèles perle-barre-bond généraux dans les flux aléatoires. En partant des résultats des chapitres précédents, Le Chap. 3 aboutit à l'étude d'un degré de liberté lagrangien inexploré dans un écoulement turbulent : la flexion. Une particule semi-flexible courbe différemment dans les flux aléatoires bidimensionnels et tridimensionnels. La IIème partie concerne la turbulence élastique, un régime chaotique créé dans un écoulement de faibles forces inertielles par l'ajout de polymères élastiques. Le Chap. 4 estime le nombre de degrés de liberté d'une solution de ce régime via la dimension Lyapunov de l'attracteur du modèle Oldroyd-B bidimensionnel, un modèle connu de reproduire la turbulence élastique dans les simulations numériques. Le Chap. 5 pose des questions sur la nécessité d'élasticité pour produire un régime chaotique et conclut qu'une solution de polymère de barres peut créer un régime comparable à la turbulence élastique. / Particles, when subjected into a flow, may display preferred orientations and a variety of deformations depending on their geometry and elasticity and the flow velocity field. Flows can conversely be modified when the particle stresses are sufficiently large. This thesis presents theoretical and numerical results on this two-way relationship between particles and flows in two parts. Part I starts with a stability analysis and numerical simulations that show a simple semiflexible Brownian particle in an extensional flow undergo tumbling, a phenomenon normally associated to shear flows. Chapter 2 extends analytical tools available only for elementary polymer models or for steady flows to general bead-rod-spring models in random flows. By building on the results from the previous chapters, Chap. 3 culminates with the study of an unexplored Lagrangian degree of freedom in a turbulent flow: bending. A semiflexible particle is shown to display different bending behaviours in two- and three-dimensional random flows. This prediction is confirmed via direct numerical simulations of the particle in a turbulent flow. Part II concerns “elastic turbulence", a chaotic regime created in a flow with low inertial forces by the addition of elastic polymers. Chapter 4 provides an estimate for the number of degrees of freedom of a solution of this chaotic system via the Lyapunov dimension of the attractor of the two-dimensional Oldroyd-B model, a model known to reproduce elastic turbulence in numerical simulations. Chapter 5 questions the necessity of elasticity in producing a chaotic regime and concludes that a rodlike polymer solution can create a regime similar to elastic turbulence.

It uses the author’s experience as a skilled bead maker to investigate processes of bead production and mechanics of bead collection current in Scandinavia and Anglo-Saxon England in the period of 6th to 9th century AD. In the study the bead collections of seven graves are examined from the perspective of their production techniques, materials, and damage from wear and cremation. The results point to beads being acquired in different numbers and often worn for long periods of time before being buried.

Juvenil idiopatisk artrit (JIA) är en autoimmun reumatisk sjukdom som drabbar barn före 16 års ålder. Den vanligaste varianten är oligoartikulär JIA, vilket innebär att den påverkar två till fyra leder med symptom så som svullnad, ömhet, stelhet, rodnad eller nedsatt motorik. Oligoartikulär JIA är även associerad med uveit, vilket är en autoimmun ögonsjukdom som förstör synen. Sjukdomen är i nuläget dåligt förstådd och forskning behövs för att kunna identifiera markörer som kan användas för diagnostik. Enzymkopplad immunadsorberande analys (ELISA) är inom vårdens laborativa arbete en mycket vanlig metod för att undersöka patienters serum och identifiera specifika markörer som tyder på sjukdom. En ELISA kan utföras på olika sätt beroende på om den letar efter antigen eller antikroppar. Syftet med denna studie var att optimera en indirekt ELISA för detektion av autoantikroppar och genomföra en metodjämförelse med en redan väl undersökt kulbaserad autoantikroppsmetod. Studien omfattar 14 serumprov från olika patienter och använder sig av rekombinant framställda protein epitope signature tag (PrEST)-antigener för att identifiera autoantikroppar. PrEST-antigener består av ett humant variabelt proteinsegment och en protein-tag som består av ett albuminbindande protein (ABP) med en N-terminal HIS-tag (His6ABP). För att förhindra ospecifik inbindning till His6ABP-delen av PrEST-antigenerna så framställdes His6ABP, med vilket serum inkuberades innan de undersöktes med ELISA. Då resultatet tolkades binärt kunde stor likhet ses mellan de två olika metoderna för samtliga PrEST-antigener som undersöktes, vilket innebär att metoden kan fortsätta utforskas. Resultatet visade också att His6ABP-delen skulle kunna orsaka korsreaktivitet, vilket dock inte är något negativt för metodens validitet. / Juvenile idiopathic arthritis (JIA) is an autoimmune rheumatic disease that affects children before the age of 16. The most common variant is oligoarticular JIA, which means it affects two to four joints with symptoms such as swelling, tenderness, stiffness, redness, or reduced mobility. Oligoarticular JIA is also associated with uveitis, an autoimmune eye disease which damage eyesight. The disease is currently poorly understood, and research is needed to identify markers that can be used for diagnostics. Enzyme-linked immunosorbent assay (ELISA) is a very common method in healthcare laboratory work to examine patients' serum and identify specific markers that indicate disease. An ELISA method can be performed in different ways depending on whether it is looking for antigen or antibodies. The aim of this study was to optimize an indirect ELISA for detection of autoantibodies and conduct a method comparison with an already well established bead-based autoantibody array method. The study included 14 sera samples from different patients and used recombinantly produced protein epitope signature tag (PrEST) antigens to identify autoantibodies. PrEST antigens consist of a human variable protein segment and a protein tag consisting of albumin binding protein (ABP) with an N-terminal HIS-tag (His6ABP). To prevent non-specific binding to the His6ABP portion of the PrEST antigens, His6ABP were recombinantly produced and used for pre-absorption of serum before being examined by ELISA. When the result was interpreted binary, great similarity could be seen between the two different methods for all PrEST antigens examined, which calls for further exploring of the method. The result also showed that the His6ABP part could possibly cause cross-reactivity, which is interesting but doesn’t affect the validity of the method.

Novel Circulating and Tissue Biomarkers for Small Intestine Neuroendocrine Tumors and Lung Carcinoids / 小肠神经内分泌肿瘤及肺类癌患者体液和组织中新的生物标记物Cui, Tao January 2013 (has links)
Small intestine neuroendocrine tumors (SI-NETs) and lung carcinoids (LCs) are relatively indolent tumors, which originate from neuroendocrine (NE) cells of the diffuse NE system. Metastases can spread before diagnosis. Thus, potential cures become unavailable, which entitles new biomarker development. Indeed, we aimed at developing Ma2 autoantibodies and olfactory receptor 51E1 (OR51E1) as potential novel biomarkers and exploring other candidate protein markers in patients’ serum. First, we established a sensitive, specific and reliable anti-Ma2 indirect ELISA to distinguish SI-NET patients from healthy controls. We detected longer progression-free and recurrence-free survivals in patients expressing low anti-Ma2 titers. Moreover, a high anti-Ma2 titer was more sensitive than chromogranin A for the risk of recurrence after radical operation of SI-NET patients. We then investigated OR51E1 expression in SI-NETs and LCs. OR51E1 mRNA expression, analyzed by quantitative real-time PCR, was high in microdissected SI-NET cells, in LC cell lines and in frozen LC specimens. Immunohistochemistry (IHC) showed abundant OR51E1 protein expression in SI-NETs. OR51E1 co-expressed with vesicular-monoamine-transporter-1 in the majority of normal and neoplastic enterochromaffin cells. Furthermore, the study on LCs revealed that OR51E1, somatostatin receptor (SSTR) 2, SSTR3, and SSTR5 are expressed in 85%, 71%, 25% and 39% of typical carcinoids (TCs), whereas in 86%, 79%, 43% and 36% of atypical carcinoids (ACs). Based on the proposed IHC scoring system, in the LC cases, where all SSTR subtypes were absent, membrane OR51E1 expression was detected in 10 out of 17 TCs and 1 out of 2 ACs. Moreover, higher OR51E1 scores were detected in 5 out of 6 OctreoScan-negative LC lesions. In addition, the last presented study used a novel suspension bead array, which targeted 124 unique proteins, by using Human Protein Atlas antibodies, to profile biotinylated serum samples from SI-NET patients and healthy controls. We showed 9 proteins, IGFBP2, IGF1, SHKBP1, ETS1, IL1α, STX2, MAML3, EGR3 and XIAP as significant contributors to tumor classification.
