A cana-de-açúcar é uma das principais culturas agroindustriais do Brasil. Como outras culturas de importância econômica, é também afetada por diversos patógenos como fungos, bactérias, vírus e fitoplasmas que podem limitar sua produção. A bactéria fastidiosa Leifsonia xyli subsp. xyli (Lxx) é o agente causal do raquitismo-da-soqueira da cana-de-açúcar (RSD). Esta doença não apresenta sintomas externos característicos de fácil reconhecimento. Dessa forma, a incidência do RSD pode não ser perceptível durante inspeções visuais de campo e, assim, ser facilmente disseminado de uma região para outra. A bactéria restrita ao xilema é transmitida mecanicamente de plantas infectadas para plantas saudáveis por meio da seiva presente em ferramentas de corte e em outros equipamentos durante o plantio, o cultivo e a colheita da cultura. Em razão da importância da cana-de-açúcar e do dano causado por Lxx, este trabalho visou avaliar a termoterapia e o cultivo in vitro em dois experimentos direcionados à obtenção de plantas de cana-de-açúcar da variedade SP80-3280 livres de Leifsonia xyli subsp. xyli e, assim, fornecer subsídio para o aprimoramento das medidas atuais de controle. O primeiro experimento combinou a termoterapia de toletes de uma gema (52 °C por 30 minutos) com o cultivo de meristemas apicais de três tamanhos (0,5 mm, 1,0 mm e 1,5 mm); o segundo combinou três tempos de termoterapia (1 hora, 2 horas e 3 horas) a 50 °C em gemas laterais imaturas isoladas do tolete com o cultivo in vitro. No primeiro experimento, os resultados de RT-PCR das amostras coletadas aos 15 dias na casa de vegetação mostraram que os três tamanhos de meristemas produziram plantas com títulos bacterianos em média 25 vezes mais baixos que suas genitoras, mas não as isentaram completamente de Lxx, com exceção de 8 plantas que não apresentaram Lxx com 90 dias de cultivo (4 plantas provenientes do cultivo de meristemas de 0,5 mm, 1 planta de meristema de 1,0 mm e 3 plantas de meristemas de 1,5 mm). Além disso, os meristemas de 0,5 mm não necessariamente regeneraram plantas com títulos mais baixos da bactéria quando comparados com os meristemas de 1,0 mm e 1,5 mm, mostrando que estes tamanhos testados não apresentaram correlação direta com o nível de infecção da planta regenerada por eles. Em relação ao segundo experimento, os resultados de RT-PCR mostraram que o incremento no tempo de termoterapia proporcionou maior redução nos títulos bacterianos, sendo os tratamentos de 2 horas e 3 horas os mais bem sucedidos com média de redução de 80,08% e 81,73%, respectivamente, mas não foi capaz de eliminar a bactéria. Portanto, estes experimentos demonstram que a termoterapia associada ao cultivo de tecidos é uma metodologia promissora, pois apresentaram redução da presença da bactéria Lxx em plantas de cana-de-açúcar, mas por não terem sido totalmente efetivos na eliminação da mesma, reforça a necessidade de estudos complementares. / Sugarcane is one of the major agro-industrial crops of Brazil. Like other crops of economic importance, it\'s also affected by several pathogens such as fungus, bacteria, viruses and phytoplasmas that may limit its production. The fastidious bacterium Leifsonia xyli subsp. xyli (Lxx) is the causal agent of ratoon stunting disease in sugarcane (RSD). This disease shows no external symptoms characteristic of easy recognition. Thus, the incidence of RSD may not be noticeable during visual inspection in the field and be easily spread from one region to another. The xylem-limited bacterium is transmitted mechanically from infected plants to healthy plants by the sap present in cutting tools and others equipments for planting, growing and harvesting the crop. Due to the importance of sugarcane and the damage caused by Lxx, this work aimed to evaluate thermotherapy and in vitro culture in two experiments in order to obtain sugarcane plants of variety SP80-3280 free from Leifsonia xyli subsp. xyli and thus provide subsidy for the improvement of current control methods. The first experiment combined thermotherapy on the individual bud (52 °C for 30 minutes) and the cultivation of apical meristems of three sizes (0,5 mm, 1,0 mm and 1,5 mm); the second combined three period of thermotherapy (1 hour, 2 hours and 3 hours) at 50 °C in immature lateral buds totally isolated from the tissue source followed by in vitro culture. In the first experiment, the results of RT-PCR samples collected at 15 days in the greenhouse showed that the three sizes of meristems produced plants with bacterial titers on average 25 times lower than their mother plants, but not completely exempted them from Lxx, except 8 plants that showed no Lxx after 90 days of cultivation (4 plants were from cultivation of meristems with 0,5 mm, 1 plant from 1,0 mm meristem and 3 plants from 1,5 mm meristems size). Furthermore, the meristems of 0,5 mm does not necessarily regenerate plants with lower title of the bacteria compared with the meristems of 1.0 mm and 1.5 mm, showing that these sizes tested did not present direct correlation with the level of infection on the plants regenerated from them. Regarding the second experiment, RT-PCR results showed that the increase in time of thermotherapy provided greater reduction in bacterial titers, and the treatments of 2 and 3 hours were the most successful with average reduction of 80,08 % and 81,73%, respectively, but was not able to eliminate the bacteria. Therefore, these experiments showed that thermotherapy associated with tissue culture is a promising methodology because it reduced titer of Lxx in sugarcane, but were not fully effective in eliminating the same, reinforcing the need for complementary studies.

Functional identification and initial characterization of a fish co-receptor involved in aversive signaling

Chemoreception plays an important role in predator-prey interactions and feeding dynamics. While the chemoreception of attractant or pleasant tasting compounds has been well studied, aversive chemoreceptive signaling has been difficult to investigate behaviorally in an ecological context because these interactions are species- and context- specific and deterrent compounds vary among prey. Using the coral reef system, this thesis explores on a molecular level the deterrent mechanism underlying detection by fish predators of an aversive compound, in order to gain a greater understanding of predator-prey interactions in this community. Like other organisms that are sessile or slow-moving, marine sponges have special mechanisms for defense from predation, commonly containing aversive-tasting compounds that defend these organisms from predation. To this end, we sought to identify and characterize a fish chemoreceptor that detects one or more of these compounds. We isolated a single cDNA clone encoding RAMP-like triterpene glycoside receptor (RL-TGR), a novel co-receptor involved in the signaling of triterpene glycosides. This co-receptor appears to be structurally and functionally related to receptor activity-modifying proteins (RAMPs), a family of co-receptors that physically associate with and modify the activity of G protein-coupled receptors (GPCRs). Expression in Xenopus oocytes showed that it responds to triterpene glycosides in a receptor-mediated manner and requires co-expression of a GPCR to enable signaling in oocytes; both of these receptors may be components of a larger signaling complex. A 40 bp portion of the gene is conserved across multiple fish species, but is not found in any other organism with a sequenced genome, suggesting that the expression of this receptor is limited to fish species. RL-TGR is the first identified gene encoding a co-receptor that responds to a chemical defense. This finding may lead the way for the identification of many other receptors that mediate chemical defense signaling in both marine and terrestrial environments, as this protein has the potential to represent the first of an entire family of co-receptors that respond to aversive compounds.

A língua de lagartos é estrutura componente do sistema sensorial fundamental para o forrageio. Desta forma, o presente estudo buscou investigar a estrutura morfo-histológica da língua de cinco espécies de lagartos brasileiros (Ameiva ameiva, Hemidactylus mabouia, Aspronema dorsivittatum, Ophiodes striatus e Tropidurus torquatus) e sua relação com os tipos de forrageio. Foram analisados o formato e comprimento de língua e confeccionadas lâminas histológicas coradas com HE e PAS, de exemplares das cinco espécies. As espécies estudadas apresentaram revestimento de epitélio pavimentoso estratificado queratinizado, com variações topográficas na espessura da camada córnea. Ameiva ameiva apresentou a língua mais especializada e restrita ao forrageio ativo, enquanto Hemidactylus mabouia e Tropidurus torquatus apresentaram língua com estrutura menos favorável ao “tongue-flicking” e, portanto, mais próximo de forrageio de emboscada. As espécies Ophiodes striatus e Aspronema dorsivittatum apresentaram resultados característicos tanto de forrageio ativo quanto de emboscada, demonstrando uma provável plasticidade entre estes extremos. Esta flutuação entre tipos de forrageio já foi observada dentro do antigo gênero Mabuya a partir de estudos prévios que indicam que fatores como disponibilidade de alimento e alterações de habitat são capazes de alterar a dinâmica de forrageio de algumas espécies. No presente estudo foi possível relacionar aspectos como tipos de papilas, formato de língua, arranjo muscular, camada de queratina, presença de botões gustativos, entre outros, com a provável dinâmica de forrageio das cinco espécies estudadas. O arranjo muscular encontrado nos forrageadores ativos foi considerado mais compacto e direcionado ao “tongue-flicking”, enquanto o dos forrageadores de emboscada o arranjo muscular se mostra menos restrito. A análise das características externas e microscópicas da língua trouxeram grandes contribuições para o entendimento de como cada espécie, por exemplo, otimiza a sua percepção do ambiente, como percebe o tempo e seu gasto energético. Além disso, contribui com maiores informações a cerca da morfologia e ecologia de espécies ocorrentes no Brasil. / The Squamata reptiles have a sophisticated sensory system, frequently adapted to the environment in which they live and the activities performed in their foraging. The tongue lizards is a fundamental structure of the sensory system for foraging. Thus, this study sought investigate the morphological and histological tongue structure of five species of Brazilian lizards (Ameiva ameiva, Hemidactylus maboiua, Aspronema dorsivittatum, Ophiodes striatus e Tropidurus torquatus) and its relationship with ambush and active foraging. For that, were analyzed the shape and elongation and made histological slides of language specimens of the five species and colored by HE and PAS. The tongue of all species showed stratified squamous keratinized epithelium, with the keratin layer varying in thickness and position in the tongue. Ameiva ameiva showed the most specialized tongue and restricted to wile foraging, while H. mabouia and T. torquatus presented tongue with less favorable to “tongue-flicking” and therefore closest to ambush foraging structure. The O. striatus and A. dorsivittatum specie showed characteristics of both foraging types, showing a probable plasticity between the extremes of wile and ambush foraging. This fluctuation between types of foraging has been observed within the old genus Mabuya from previous studies that indicate that factors such as food availability and habitat changes are able to alter the dynamics of some species foraging. In the present study it was possible to relate aspects such as types of papillae, tongue shape, muscular arrangement, keratin layer, among others, with the possible dynamics of foraging the five species. The muscle arrangement found in active foragers was considered more compact and focused for the “tongue-flicking”, while the ambush foragers muscle arrangement shown less restricted. And stand out from other factors such as the presence of taste buds. The analysis of external and microscopic characteristics of the tongue provided great contributions to the understanding of how the lizard optimizes its environment perception, as realize the time and its energy expenditure. Also contributes with more information about the morphology and ecology of species occurring in Brazil.

Effect of number of axillary buds on stem cuttings of sweet potato (pomoea batatas) CV. 'Blesbok' productivity using sack gardening technology

(Horticulture)) --University of Limpopo, 2022 / Worldwide, water scarcity, land degradation and unavailability of agricultural lands have posed serious threats on food security and income generation. Hunger, micronutrient deficiencies and starvation are likely to go up in Africa due to increasing urbanisation and rise in global food prices. As a result, it is important to identify innovative vegetable growing-based strategies, which can increase access to food by utilising the limited space available in rural and urban areas. Therefore, sack gardening technology could be a solution to both the lack of arable land and water scarcity in urban and rural areas. The objective of the study was to determine whether axillary bud number will have an effect on agronomic parameters and yield of sweet potato cv. 'Blesbok' produced using the sack gardening technology. Treatments, namely, 1, 2, 3, 4, and 5 number of axillary buds per stem cutting, were arranged in randomised complete block design, with 10 replicates. One bud (1) was used as a control. Different stem cuttings measuring from 10 to 30 cm long with approximately one (1) to six (6) axillary buds were collected from mature sweet potato plantation at a certified nursery from cv. 'Blesbok'. A total of eight (8) stem cuttings were planted in sacks consisting of growing media mixture of steam pasteurised loam soil and compost (3:1 v/v), 4 on top side of the sack and the remaining four on the side-holes of the sack. Insect pests were scouted and monitored weekly, while diseases were managed using spraying programmes as in commercial sweet potato production systems. Two weeks after transplanting, each plant was fertilised with 5 g nitrogen (N), phosphorus (P), and potassium (K) 2:3:2 (26) + 0.5% Zinc (Zn) + 5% sulphur (S) and 5% calcium (Ca). Number of axillary buds had significant effects on vine length, fresh shoot mass, number of shoots and dry shoot mass contributing 59%, 58%, 45% and 58% in total treatment variation (TTV), respectively, whereas axillary bud numbers had no significant effects on fresh tuberous root mass, chlorophyll content, vine stem diameter, tuberous root diameter, tuberous root length, dry tuberous root mass and number of tuberous roots. Relative to control (one bud), two (2), three (3), four (4), five (5) number of axillary buds decreased vine length, fresh shoot mass and dry shoot mass by 8-37, 30-55 and 26-55%, respectively. However, relative to control (one bud), number of axillary buds increased number of shoots by 8-17%. The quadratic relationship models were explained by 95, 92, 96 and 98% of vine length (VL), number of shoots (NS), fresh shoot mass (FSM) and dry shoot mass (DSM), respectively. In conclusion, findings of the study demonstrated that a single (1) or two (2) axillary bud(s) on stem cuttings of sweet potato CV. „Blesbok‟ showed better results for both yield and growth parameters as compared to three (3), four (4) and five (5) buds. / AgriSETA

The Relation of Growth Ring Widths in American Beech and White Oak to Variations in Climate

An analysis is made of beech from Ohio and white oak from Illinois using a stepwise multiple regression technique to evaluate ring growth and climatic relationships. Ring widths for beech are directly related to the moisture supply during August and to temperatures for May-July of the preceding year. They are equally related to moisture during June-August of the current year and somewhat dependent upon the precipitation of the previous winter. The earlywood width of white oak is directly related to available moisture during the preceding September and to moisture during the current June, and inversely related in slight degree to the temperature of April. Latewood width of oak is primarily dependent upon the availability of moisture during the current June and July but is somewhat related to the moisture during May of the previous year. Serial correlation is prominent from growth layer to growth layer in both species. The physiological relationships which may produce these results are discussed. Some relationships involving considerable lag in the growth response are possible controls of bud formation and food accumulation, while others involving more immediate response are primarily the effect of moisture stress within the tree.

A podridão floral dos citros (PFC) é uma doença causada pelo fungo Colletotrichum acutatum responsável por causar grandes danos à produção de citros no Brasil. A doença surge apenas em botões florais com 8mm de comprimento ou maiores, levando a formação de lesões alaranjadas nas pétalas, lesões necróticas no estigma, promove a queda prematura dos frutos e a retenção do cálice e pedúnculo, sendo este último sintoma denominado estrelinha. Este trabalho tem por objetivo: observar o modo de penetração do fungo no hospedeiro Citrus sinensis Valência e os estágios posteriores da colonização, verificar se há fatores estruturais e químicos pré-formados que expliquem o porquê do fungo não conseguir infectar botões florais com menos de 8mm, caracterizar anatomicamente o sintoma estrelinha e estigmas lesionados, investigar ultraestruturalmente pétalas inoculadas, analisar se há o estabelecimento de uma infecção quiescente nos tecidos foliares, analisar grãos de pólen após a inoculação in vivo e in vitro com o fungo. Botões florais sadios, pétalas e estigmas com e sem lesões, foram submetidos às técnicas convencionais de microscopia de luz e eletrônica. Folhas e grãos de pólen foram inoculados e analisados. Foi desenvolvida uma nova técnica de coloração para tecidos vegetais infectados por fungos. A resistência dos botões florais menores que 8mm pode estar associada às barreiras químicas e estruturais pré-formadas. O ápice, nesses botões, apresenta papilas entremeadas, cristais de oxalato de cálcio no mesofilo e câmara subestomática e cavidades de óleo localizadas muito próximas umas das outras. Botões com 8mm e 12mm possuem, no ápice, papilas com arranjo frouxo, ausência de cristais e maior distanciamento entre as cavidades de óleo. No ápice da pétala, verificou-se que as células papilosas são osmóforos. No sintoma estrelinha, nota-se sob a região de abscisão do ovário a instalação de um meristema de cicatrização. A lignificação das paredes das células da medula do receptáculo e do pedúnculo floral está associada à retenção destas estruturas na planta. Nas pétalas infectadas, o C. acutatum pode penetrar intra, intercelularmente e via estômato. O fungo pode crescer de modo subcuticular e intramural e coloniza todos os tecidos da pétala. A nova técnica de coloração se mostrou muito útil nas análises histopatológicas. O fungo associa-se aos tecidos vasculares. Acérvulos ocorrem em ambas as faces das pétalas. A cutícula nos estágios mais avançados da lesão apresenta-se alterada, ou seja, ocorre a perda da ornamentação estriada e maior deposição de material lipofílico. A síntese de materiais lipofílicos envolve o retículo endoplasmático liso e rugoso e plastídios. Vesículas provenientes de dictiossomos e de corpos multivesiculares são observadas ao longo da parede celular e estão associadas ao depósito de material lipofílico na cutícula. No estigma lesionado há a formação de uma camada de proteção. O fungo apresenta quimiotropismo e cresce em direção aos grãos de pólen infectando-os 24 horas após a inoculação. Sugere-se que C. acutatum pode utilizar grãos de pólen para a sua dispersão. Após 48 horas da inoculação as folhas apresentam conídios germinados com apressórios. / The postbloom fruit drop (PFD) is a disease caused by Colletotrichum acutatum responsible for causing great damage to citrus crops in Brazil. The disease appears only in flower buds 8 mm in length or greater, leading to orange lesions in petals, necrotic lesions on the stigma, promoting the young fruit drop and the retention of the calyx and peduncle, which is called buttons. In this context, this study aimed to: observe the fungus penetration mode into the host Citrus sinensis \'Valência\' and the later stages of colonization; study the presence of preformed structural and chemical factors to explain why the fungus cannot infect floral buds with less than 8 mm in length; characterize anatomically the symptom \"buttons\" and injured stigmas; investigate the ultrastructural changes in tissues of inoculated petals; analyze whether there is the establishment of a quiescent infection in leaf tissues, analyze pollen grains after inoculation in vivo and in vitro with the fungus. Healthy buds, petals and stigmas with and without lesions, were processed and analyzed using conventional light and electron microscopy techniques. Leaves and pollen grains were also inoculated and analyzed with light microscopy. It was developed a new staining method for fungal-infected plant tissues. The resistance of flower buds smaller than 8mm may be associated with preformed structural and chemical barriers. These buttons display the apex with interspersed papillae, with crystals in mesophyll and substomatic chamber and oil cavities, which are located very close to each other on the abaxial surface. In 8mm and 12mm flower buds, the papilas in the apex become weakly interspaced, the crystals are not observed and there is the increase of the distance between the oil cavities. The papillose cells are osmophores. In the symptom \"button\", it is noted in the abscission of the ovary, an installation of wound meristem. There is also the lignification in the pith of receptacle and pedicel that can be associated with the retention of these structures in the plant. In infected petals, it was found that C. acutatum can penetrate intra and intercelullar or via stomata. The fungus may grow subcuticular and intramural and colonize all tissues of petal. The new staining technique developed has proved very useful for histopathological analysis. The fungus is closely associated with vascular tissues. The acervulli occur on both surfaces of petals. The cuticle in the later stages of the lesion is altered, i.e., there is loss of striated ornamentation and increased deposition of lipophilic material. The synthesis of lipophilic materials involves rough and smooth endoplasmic reticulum and plastids. Vesicles from dictyosomes and multivesicular bodies were observed throughout the cell wall and are associated with the deposit of lipophilic material in the cuticle. There is the formation of protective layer over the stigma damaged area. The fungus shows chemotropism and grows toward the pollen infecting it 24 hours after inoculation. It is suggested that C. acutatum can use pollen grains for dispersal. After 48 hours of inoculation, the leaves have germinated conidia with appressoria.

Embora desfrute de inegável importância econômica, os citros estão sujeitos a muitos problemas sanitários sendo alguns, limitantes para o cultivo como é o caso das doenças causadas por vírus. A morte súbita dos citros é uma doença relacionada à combinação copa/ porta-enxerto e manifesta sintomas na região da enxertia sobre porta-enxertos intolerantes. Embora sua etiologia não tenha sido determinada, há indicações que a causa da MSC esteja relacionada a uma estirpe do vírus da tristeza dos citros (CTV), a um vírus do gênero Marafivirus, ou a uma associação entre eles. Uma vez que a transformação genética têm sido considerada como uma ferramenta auxiliar a programas de melhoramento de citros, o objetivo deste trabalho foi obter plantas transgênicas de limão \'Cravo\' e laranja \'Valência\' contendo o gene da replicase do Marafivirus e estudar a regeneração e obtenção de plantas in vitro de laranja azeda, via organogênese, visando futuros trabalhos de transformação genética. Experimentos para indução da organogênese in vitro foram realizados avaliando-se citocininas (BAP, TDZ e CIN), em diferentes concentrações, isoladamente ou em combinação com ANA, condições de luminosidade (fotoperíodo de 16 h e escuro por 30 dias), meios de cultivo e explantes (provenientes de plantas germinadas in vitro e de plantas mantidas em estufa). Além disso, avaliou-se o enraizamento dos brotos regenerados. Para a transformação genética, explantes de limão \'Cravo\' e laranja \'Valência\' foram inoculados e co-cultivados com a estirpe EHA 105 de Agrobacterium tumefaciens contendo o gene da replicase do Marafivirus (em seqüência sense e antisense interligadas por um íntron). A construção gênica foi elaborada a partir do plasmídeo pCAMBIA 2201, dirigidas pelo promotor 35S e terminador NOS, contendo ainda o gene de seleção nptII. A transformação foi confirmada por análises de PCR e \'Southern blot\'. A transcrição do gene foi avaliada por RT-PCR e \'northern blot\'. A adição de BAP, combinada ou não com ANA, e em combinações com CIN ao meio de cultivo, assegurou maior formação de gemas adventícias em segmentos de epicótilo de laranja azeda. Entretanto, TDZ não se mostrou favorável a essa resposta, que também é afetada pela ausência de luz. Os explantes provenientes do cultivo in vitro mostraram-se mais favoráveis à resposta organogênica. O enraizamento das brotações de laranja azeda regeneradas foi obtido no meio MT com metade da concentração de sais, sem ou com auxinas. Foi possível obter plantas transgênicas de limão \'Cravo\' e de laranja \'Valência\' contendo o gene da replicase do Marafivirus utilizando-se segmentos internodais como explantes. A análise de \'Southern blot\' confirmou a integração de um a quatro eventos de inserção do transgene no genoma das plantas. A transcrição do gene da replicase do Marafivirus e do gene nptII foi observada por RT-PCR. / In spite of great economic importance, the citrus industry is affected by many phytopathological problems some, limiting its cultivation such as virus-caused diseases. The citrus sudden death disease is related to scion/rootstock combinations and manifests symptoms in the grafting area of intolerant rootstocks. Although its etiology has not been determined, there are indications that the cause of MSC might be related to a strain of the Citrus tristeza virus (CTV), to a virus of the Marafivirus group, or to an association of both viruses. Since the genetic transformation has been considered as an auxiliary tool to programs of citrus improvement, the objectives of this work were to obtain transgenic plants of the Rangpur lime and Valencia sweet orange containing the Marafivirus replicase gene and study the in vitro regeneration of sour orange plants through organogenesis, aiming for future work in genetic transformation. Experiments for induction of in vitro organogenesis were carried out evaluating citocinins (BAP, TDZ and KIN), in different concentrations, separately or in combination with NAA, lighting conditions (photoperiod of 16 hours and darkness for 30 days), cultivation media and explants (coming from in vitro germinated plants and from green house cultivated plants). Besides this, rooting of the regenerated shoots was evaluated. For the genetic transformation, Rangpur lime and Valencia sweet orange explants were inoculated and co-cultivated with the EHA-105 Agrobacterium tumefaciens strain containing the Marafivirus replicase gene (in sense and antisense sequence linked by an intron). The genetic construct used derived from the pCAMBIA 2201 plasmid, driven by the 35S promoter and NOS terminator, containing the selection nptII gene. The genetic transformation was confirmed by PCR and Southern blot analysis. The gene transcription was evaluated by RT-PCR and northern blot. The addition of BAP to the culture medium, combined or not with NAA, and in combinations with KIN, assured a greater formation of adventitious buds in sour orange epicotyl segments. However, TDZ was not favorable to this response, that is also affected by the absence of light. Explants coming from in vitro cultivation were more favorable to the organogenic response. Rooting of sour orange regenerated shoots was obtained in MT medium with half the salt concentration, with or without auxin. It was possible to obtain transgenic Rangpur lime and Valencia sweet orange plants containing the Marafivirus replicase gene using internodal segments as explants. The Southern blot analysis confirmed the integration of one to four copies of the transgene in the plant genome. The transcription of the Marafivirus replicase gene and the nptII gene was observed by RT-PCR.
