Reducing image interpretation errors - Do communication strategies undermine this?Snaith, Beverly, Hardy, Maryann L., Lewis, Emily F. 08 1900 (has links)
No / Errors in the interpretation of diagnostic images in the emergency department are a persistent problem internationally. To address this issue, a number of risk reduction strategies have been suggested but only radiographer abnormality detection schemes (RADS) have been widely implemented in the UK. This study considers the variation in RADS operation and communication in light of technological advances and changes in service operation.
A postal survey of all NHS hospitals operating either an Emergency Department or Minor Injury Unit and a diagnostic imaging (radiology) department (n = 510) was undertaken between July and August 2011. The questionnaire was designed to elicit information on emergency service provision and details of RADS.
325 questionnaires were returned (n = 325/510; 63.7%). The majority of sites (n = 288/325; 88.6%) operated a RADS with the majority (n = 227/288; 78.8%) employing a visual ‘flagging’ system as the only method of communication although symbols used were inconsistent and contradictory across sites. 61 sites communicated radiographer findings through a written proforma (paper or electronic) but this was run in conjunction with a flagging system at 50 sites. The majority of sites did not have guidance on the scope or operation of the ‘flagging’ or written communication system in use.
RADS is an established clinical intervention to reduce errors in diagnostic image interpretation within the emergency setting. The lack of standardisation in communication processes and practices alongside the rapid adoption of technology has increased the potential for error and miscommunication.

Développement de bionansondes en biofonctionnalisant des boîtes quantiques (quantum dots) par des anticorpsRousserie, Gilles 30 November 2012 (has links)
Ce travail porte sur différentes méthodes de détection de protéines par des anticorps (Acs) marqués par des boîtes quantiques (QD, « quantum dots »). La première partie de la thèse porte sur le développement et l'optimisation de méthodes de conjugaison d'Acs polyclonaux à des QDs. Une étude de 2007 avait démontré que les conjugués anticorps-quantum dots (Acs-QDs) commerciaux ne présentent que de très rares anticorps fonctionnels à leur surface [Pathak et al., 2007]. Nous avons donc : (i) développé des conditions de réduction ménagée des ponts disulfures des Acs en utilisant soit du dithiothreitol, soit du 2-mercapotethanolamine, (ii) purifié les fragments d'Acs fonctionnels grâce à une colonne d'affinité, (iii) conjugué ces fragments fonctionnels d'Acs aux QDs en utilisant l'agent de liaison amine-thiol SMCC et (iv) développé une méthode de purification des conjugués sur gel d'agarose afin d'éliminer les fragments d'Acs non conjugués et les QDs libres. Des tests en cytométrie en flux ont permis de déterminer l'efficacité des conjugués pour détecter l'expression de la molécule CD4 à la surface de lymphocytes T humains. Un marquage de CD4 réalisé avec des conjugués préparés selon notre méthode s'avère être cinq fois plus sensible qu'en utilisant des conjugués réalisés selon les recommandations commerciales. Une autre méthode de préparation des Acs pour la conjugaison aux QDs a été testée : l'ajout de groupements fonctionnels sulfhydriles sur des amines primaires grâce au N-succinimidyl S-acetylthioacetate (SATA). Cependant, cette méthode ne permet pas d'avoir un contrôle précis du nombre ni de l'emplacement des groupements fonctionnels sulfhydriles ainsi ajoutés. Les tests de conjugaison aux QDs qui ont été réalisés avec ces Acs-SH, en utilisant l'agent de liaison SMCC, a entraîné la formation d'agrégats de taille variable. Cette méthode a donc été abandonnée.La seconde partie de la thèse porte sur la démonstration de la faisabilité et l'optimisation d'un marquage de l'antigène carcino-embryonnaire (CEA, « carcinoembryonic antigen ») humain à la surface de lignées cellulaires murines avec un anticorps simple domaine (sdAb) anti-CEA biotinylé détecté grâce à des conjugués streptavidine-QDs commerciaux et analysé par cytométrie en flux. Ces sdAbs ont été biotinylés selon deux approches : (i) avec des agents de biotinylation chimique qui permettent d'ajouter une biotine sur les amines primaires (biotinylation in vitro) et (ii) par une enzyme lors de leur production par E. coli (biotinylation in vivo). La détection du CEA humain à la surface des 100 000 cellules (lignées cellulaires murine MC38 et MC38-CEA) par ces sdAbs biotinylés a été testée en cytométrie de flux puis comparée à celle obtenue par un anticorps monoclonal biotinylé in vitro. Les résultats ont démontré que les sdAbs biotinylés ont une sensibilité de détection similaire que la biotinylation soit réalisée in vitro ou in vivo. Par contre, la sensibilité de la détection du CEA est environ cinquante fois meilleure en utilisant des sdAbs biotinylés (0,6 fmol d'anticorps sont nécessaires pour détecter le CEA) qu'avec des anticorps monoclonaux biotinylés (33 fmol). / We have been working on different ways to detect proteins with antibodies (Abs) labeled by quantum dots (QDs). The first part of his work is to develop and optimize the conjugation of polyclonal antibodies to quantum dot. A study has reported that commercial antibody-quantum dots conjugates (Abs-QDs) present very few functional Ab fragments at the surface of the conjugate [Pathak et al., 2007]. We had: (i) developed an advanced procedure of antibody reduction using dithiothreitol (DTT) or 2-mercaptoethanolamine (2-MEA) (to prevent the loss of antibody functions), (ii) purified the active fragments by affinity purification on column, (iii) conjugated the active reduced antibody fragments to QDs using the amine-thiol crosslinker SMCC for SH coupling, and (iv) developed the purification of the conjugates on agarose gel to remove free QDs and unconjugated antibody fragments. Our conjugates present about a five times better ability to detect CD4 by flow cytometry on 500 000 isolated human lymphocyte T cells than those made after the commercial procedure. Another procedure for antibody preparation was addition of sulfhydryl groups on primary amines using N-succinimidyl S-acetylthioacetate (SATA). However this procedure presents some variable yield and the number and the localization of sulfhydryl groups added on antibody cannot be fully controlled. Thereby, the conjugation of these Abs-SH to QDs using SMCC chemistry leads to the creation of aggregates. This Ab preparation in preparation for conjugation to QDs was abandoned.The second part of our work focusses on testing and comparing the ability to detect carcinoembryonic antigen (CEA) by flow cytometry with anti-CEA single domain antibodies (sdAbs) labeled by QDs. The sdAbs were biotinylated by using two methods: (i) in vitro chemical biotinylation reagent which adds biotins on primary amines, and (ii) enzymatic in vivo biotinylation during their production in E. coli. These anti-CEA sdAb biotinylation methods were compared to in vitro biotinylated monoclonal Abs against CEA for their respective ability to detect human CEA on 100 000 mice cells (MC38 and MC38-CEA cell lines) using flow cytometry. The results show that the limit detection for biotinylated sdAbs is similar between in vitro and in vivo biotinylation. Furthermore the limit detection with biotinylated sdAb (0.6 fmol are required to detect CEA) is about fifty times better than with biotinylated monoclonal Abs (33 fmol).

Prevalence analysis of putative periodontal pathogens in patients with aggressive periodontitis and healthy elderlyEdesi-Neuss, Lilian 21 November 2005 (has links)
Marginale Parodontitis, die multikausale Erkrankung des Parodonts ist eine Infektionskrankheit, modifiziert durch Wirtsfaktoren und äußere Einflüße. Die als pathogene Mischflora bezeichnete Kombination kommensaler Mikroorganismen spielt die primäre Rolle in der Ätiopathogenese der Parodontitis. In der Aufstellung des Studienziels wurden einzelne Bakterienarten (T. forsythensis, P. gingivalis, A. actinomycetemcomitans, C. rectus, F. nucleatum, Fusobacterium spp., P. intermedia, E. corrodens, V. parvula und C. ochracea) ausgewählt, die eventuell als "Markerkeime" in der aggressiven Form der Parodontitis betrachtet werden können. Dazu wurde eine Kontrollgruppe untersucht, die eine gesunde parodontale Flora besitzt. Die angewandte Nachweismethode basiert auf der PCR-Amplifikation von 16S rDNA und darauffolgender dot-blot Hybridisierung mit Oligonukleotidsonden. Die entsprechenden Sonden wurden hergestellt, optimiert und evaluiert. Für die epidemiologische Untersuchung wurde subgingivale Plaque von vier Parodontaltaschen und einer Kontrollstelle von 45 Patienten mit aggressiver Parodontitis, sowie an fünf Stellen von 21 Senioren entnommen. Die Prävalenz der einzelnen Bakterienarten wurde mit Hilfe des Chi-Quadrat Test verglichen. Obgleich eine hohe interindividuelle Variabilität der Kolonisationsmuster zu beobachten war, konnten T. forsythensis, P. gingivalis, C. rectus und F. nucleatum signifikant häufiger in den Parodontaltaschen als an den gesunden Stellen nachgewiesen werden und können deswegen als "Leitkeime" der aggressiven Parodontitis angesehen werden. A. actinomycetemcomitans konnte nur bei einzelnen Patienten mit aggressiver Parodontitis festgestellt werden. Die Ergebnisse für P. intermedia und E. corrodens ließen keine eindeutige Assoziation sowohl mit der aggressiven Parodontitis als auch mit dem gesunden Parodontalzustand zu. Bei Senioren wurde C. ochracea besonders häufig nachgewiesen. Die Ergebnisse dieser Studie bewiesen die erfolgreiche Einsetzbarkeit der hergestellten Oligonukleotidsonden. / A multifactorial risk pattern of periodontitis has been recognized, where in addition to host and environmental factors, a pathogenic microbiota plays a primary role. The purpose of the current research was to analyze the prevalence of periodontitis-associated microorganisms in patients with aggressive periodontitis and periodontally healthy elders by using molecular-biologic detection methods like eubacterial PCR-amplification of 16S rDNA in combination with dot-blot hybridization. The oligonucleotide probes for the detection of T. forsythensis, P. gingivalis, A. actinomycetemcomitans, C. rectus, F. nucleatum, Fusobacterium spp., P. intermedia, E. corrodens, V. parvula and C. ochracea were designed and evaluated. The PCR products of 42 cultivated target and closely related bacteria were used for the optimization of hybridization conditions. For the epidemiological study, subgingival plaque was sampled from four pockets and one healthy site of 45 aggressive periodontitis patients as well as from five sites of 21 elderly. The differences in the prevalence of bacterial species were analyzed by the chi-square test. The data revealed frequent colonization by T. forsythensis, P. gingivalis, F. nucleatum and C. rectus in patients with aggressive periodontitis, however individual variations were obvious. These species could be predominantly identified in periodontal pockets, but were significantly less common in the healthy sites of the periodontitis patients and in the elderly. These putative pathogens can be conclusively determined as the key-bacteria in patients with aggressive periodontitis. No direct association for P. intermedia and E. corrodens with aggressive periodontitis or periodontal health could be seen. A. actinomycetemcomitans could be detected in only a few patients, reducing its suspected importance in the etiology of aggressive periodontitis. C. ochracea was highly prevalent in the well-maintained elderly, suggesting its association with healthy flora. The results of the study confirmed the reliability of the oligonucleotide probes in a specific and sensitive detection of the respective oral species.

(ABT). Em primeiro lugar, um estudo te?rico fornece uma vis?o hist?rica da pesquisa
psicol?gica da aten??o, desde os fundadores da Psicologia moderna at? a pesquisa
neuropsicol?gica integrativa atual e os modelos orientados empiricamente. Esta revis?o
busca esclarecer conceitos da aten??o e diferenciar esses conceitos dos de outros

Dot-Probe (DPT) e a Tarefa Stroop Emocional (EST). Na EST, o desenho da tarefa foi
alterado para levar em conta considera??es te?ricas importantes e para melhor adaptar a
tarefa ? medida de ABV. Na DPT, uma integra??o surpreendentemente rara de tempos
de rea??o e medidas de rastreamento ocular ? estabelecida, e novos ?ndices para calcular
o ABT e o ABV s?o propostos. A confiabilidade e validade dos ?ndices em ambas as

do estudo, incluindo recomenda??es para futuras adapta??es dessas tarefas / Attention is a multifaceted construct, one that has been at the center of
discussions across several moments in the history of philosophy and psychology. The
characteristic of attention to influence and regulate many other psychological process
(e.g., consciousness, memory, decision-making) stresses its importance, and logically
results in a hardship in segregating its theoretical boundaries and clearly defining this
phenomenon. In a current empirical field of research on attention, biases of attentional
orientation to threatening stimuli are investigated. However, models generated from
empirical findings lack sustentation on well-established theoretical models of attention,
and confusion exists across published experimental studies. Furthermore, experimental
tasks to assess biases towards threat require integration with new operationalization and
analysis strategies, which can provide better sensitivity, validity and measurement
reliability, such as eye tracking and the novel index of attentional bias variability
(ABV).
This dissertation is included in the subarea number 7.07.02.03-9 of the Conselho
Nacional de Desenvolvimento Cient?fico e Tecnol?gico (CNPq) ? Experimental
Psychology (Attentional and Cognitive Processes) ? which integrates the broader area
of Psychology. Two studies are presented to answer to the need of advancing research
about attention, attentional control (AC) and attentional bias relative to threat (ABT).
Firstly, a theoretical study provides a historical overview of psychological research on
attention, from the founders of modern Psychology to current neuropsychological
integrative research and empirically-oriented models. This review is expected to clarify
constructs of attention and to differentiate these constructs from those of other
Psychological domains. Instead of segregating research fields, this is likely to promote a

dialogue between fields that research the same phenomena ? but measure them
differently and attribute to them different names.
Following this theoretical review, an empirical study is presented, which
proposes two adaptations of classical experimental tasks to measure ABT: the DotProbe
Task (DPT) and the Emotional Stroop Task (EST). On the EST, task design is
altered to account for important theoretical considerations and to better adapt the task to
the measurement of ABV. On the DPT, a surprisingly rare integration of reaction times
and eye tracking measures is established, and novel indices to calculate ABT and ABV
are proposed. The reliability and validity of indices in both tasks is investigated with
university students and through the differentiation of such indices between groups of
high vs. low symptoms of anxiety and posttraumatic stress. The importance of pursuing
the improvement of psychometric qualities of experimental tasks is discussed in depth
upon the findings of the study, including recommendations to future experimental
designs.

Padronização da reação de Immuno-dot para detecção de Pet em sobrenadante de cultura de Escherichia coli enteroagregativa / Imuno-dot reaction standartization for pet detection in supernatant of enteroagregative Escherichia coli cultureAndréa Bernardes Vilhena Costa 07 December 2004 (has links)
Escherichia coli enteroagregativa (EAEC), destaca-se como um importante patógeno emergente causador de diarréia persistente em países em desenvolvimento e de diarréia aguda em países desenvolvidos. A grande heterogeneidade dos fatores de virulência caracteriza esta categoria, porém não foi estabelecido um marcador genético comum a todas as amostras de EAEC. O padrão de adesão agregativa (AA) em células HEp-2 e HeLa é a forma de caracterização e diagnóstico mais precisos desta categoria. Uma das toxinas envolvidas na patogênese é Plasmid-encoded toxin (Pet) pertencente à classe das proteínas autotransportadoras com características de uma serino protease denominada SPATEs. Iniciou-se este estudo com a determinação do padrão de adesão de 164 amostras EAEC, previamente caracterizadas como sonda pCVD432 ou onda AA positiva. Assim, 141 (86%) amostras, que apresentaram padrão de adesão agregativo, foram caracterizadas como EAEC. Face aos resultados obtidos, confirmou-se a baixa especificidade da sonda AA. A pesquisa do gene pet, por meio de ensaio de PCR, resultou na positividade de 12 (8,5%) amostras. Prosseguiu-se esse estudo com a padronização da reação de immuno-dot. Utilizando-se 300 µL do sobrenadante bacteriano, soro policlonal anti-Pet e o conjugado nas diluições 1/50 e 1/2.500, respectivamente, resultados bastante reprodutíveis foram obtidos. O método foi mais sensível que a detecção do gene por PCR. Por esse ensaio, detectou-se a toxina Pet em 16 (11,3%) das 141 amostras EAEC. Nenhuma das amostras controle negativo foi reconhecida pelo soro anti-Pet, assim como as amostras de E. coli produtoras das mais diversas toxinas. Apesar da baixa prevalência de amostras de EAEC produtoras da toxina Pet, neste estudo padronizou-se um método rápido, sensível, específico e de baixo custo para pesquisa desta toxina mostrando o potencial diagnóstico deste ensaio para uso em inquéritos epidemiológicos, o que poderá permitir determinar o papel da Pet no desenvolvimento de diarréia aquosa. / Enteroaggregative Escherichia coli (EAggEC) is an emerging diarrheal pathogen, whose pathogenesis is thought to comprise colonization of the intestinal mucosa with the release of secretogenic toxins. One of the toxin involved is the plasmid-encoded toxin (Pet), which is secreted by the autotransporter mechanism and belongs to a growing class of Enterobacteriaceae autotransporter proteins. Since the characteristic aggregative adherence pattern of EAggEC is associated with the presence of a large plasmid called pCVD432, DNA probes and PCR primers derived from this plasmid have been recommended as a screening method for EAggEC in the clinicai laboratory. In this study 164 E. coli isolates positive for the pCVD432 probe were tested for adherence to HEp-2 cells in which 141 isolates showed aggregative pattern, 12 isolates from them amplify a 1037-bp DNA fragment corresponding to pet gene by PCR. Using this samples we standardized an immuno-dot assay for EAggEC detection through Pet toxin as target antigen. 300 µl of bacterial supernatant were applied in a PVDF membrane, and using a rabbit polyclonal sera anti-Pet the expression of the toxin by immuno-dot was in the same isolates in which the gene was detected. Besides no negative controls reacted with Pet antisera, in which we included 40 isolates with no virulence markers for diarrheagenic E. coli and E. coli expressing toxins other than Pet. This method proves to be rapid, sensitive, specific and low cost, demonstrating this potential as diagnosis for Pet expression and its association with diarrhea.

Using this samples we standardized an immuno-dot assay for EAggEC detection through Pet toxin as target antigen. 300 µl of bacterial supernatant were applied in a PVDF membrane, and using a rabbit polyclonal sera anti-Pet the expression of the toxin by immuno-dot was in the same isolates in which the gene was detected. Besides no negative controls reacted with Pet antisera, in which we included 40 isolates with no virulence markers for diarrheagenic E. coli and E. coli expressing toxins other than Pet. This method proves to be rapid, sensitive, specific and low cost, demonstrating this potential as diagnosis for Pet expression and its association with diarrhea.

On the role of the electron-electron interaction in two-dimensional quantum dots and ringsWaltersson, Erik January 2010 (has links)
Many-Body Perturbation Theory is put to test as a method for reliable calculations of the electron-electron interaction in two-dimensional quantum dots. We show that second order correlation gives qualitative agreement with experiments on a level which was not found within the Hartree-Fock description. For weaker confinements, the second order correction is shown to be insufficient and higher order contributions must be taken into account. We demonstrate that all order Many-Body Perturbation Theory in the form of the Coupled Cluster Singles and Doubles method yields very reliable results for confinements close to those estimated from experimental data. The possibility to use very large basis sets is shown to be a major advantage compared to Full Configuration Interaction approaches, especially for more than five confined electrons. Also, the possibility to utilize two-electron correlation in combination with tailor made potentials to achieve useful properties is explored. In the case of a two-dimensional quantum dot molecule we vary the interdot distance, and in the case of a two-dimensional quantum ring we vary the ring radius, in order to alter the spectra. In the latter case we demonstrate that correlation in combination with electromagnetic pulses can be used for the realization of quantum logical gates. / At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 5: Manuscript.

Spatial confinement of electrons and their interactions as well as confinement of the spin dimensionality often yield drastic changes of the electronic and magnetic properties of solids. Novel quantum transport and optical phenomena, involving electronic spin degrees of freedom in semiconductor heterostructures, as well as a rich variety of exotic quantum ground states and magnetic excitations in complex transition metal oxides that arise upon such confinements, belong therefore to topical problems of contemporary condensed matter physics.
In this work electron spin systems in reduced dimensions are studied with Electron Spin Resonance (ESR) spectroscopy, a method which can provide important information on the energy spectrum of the spin states, spin dynamics, and magnetic correlations. The studied systems include quasi onedimensional spin chain materials based on transition metals Cu and Ni. Another class of materials are semiconductor heterostructures made of Si and Ge.
Part I deals with the theoretical background of ESR and the description of the experimental ESR setups used which have been optimized for the purposes of the present work. In particular, the development and implementation of axial and transverse cylindrical resonant cavities for high-field highfrequency ESR experiments is discussed. The high quality factors of these cavities allow for sensitive measurements on μm-sized samples. They are used for the investigations on the spin-chain materials. The implementation and characterization of a setup for electrical detected magnetic resonance is presented.
In Part II ESR studies and complementary results of other experimental techniques on two spin chain materials are presented. The Cu-based material Linarite is investigated in the paramagnetic regime above T > 2.8 K. This natural crystal constitutes a highly frustrated spin 1/2 Heisenberg chain with ferromagnetic nearest-neighbor and antiferromagnetic next-nearestneighbor interactions. The ESR data reveals that the significant magnetic anisotropy is due to anisotropy of the g-factor. Quantitative analysis of the critical broadening of the linewidth suggest appreciable interchain and interlayer spin correlations well above the ordering temperature. The Ni-based system is an organic-anorganic hybrid material where the Ni2+ ions possessing the integer spin S = 1 are magnetically coupled along one spatial direction. Indeed, the ESR study reveals an isotropic spin-1 Heisenberg chain in this system which unlike the Cu half integer spin-1/2 chain is expected to possess a qualitatively different non-magnetic singlet ground state separated from an excited magnetic state by a so-called Haldane gap. Surprisingly, in contrast to the expected Haldane behavior a competition between a magnetically ordered ground state and a potentially gapped state is revealed.
In Part III investigations on SiGe/Si quantum dot structures are presented. The ESR investigations reveal narrowlines close to the free electron g-factor associated with electrons on the quantum dots. Their dephasing and relaxation times are determined. Manipulations with sub-bandgap light allow to change the relative population between the observed states. On the basis of extensive characterizations, strain, electronic structure and confined states on the Si-based structures are modeled with the program nextnano3. A qualitative model, explaining the energy spectrum of the spin states is proposed.

Optical and Transport Properties of Quantum Dots in Dot-In-A-Well Systems and Graphene-Like MaterialsChaganti, Venkata 17 December 2015 (has links)
Quantum dots exhibit strongly size-dependent optical and electrical properties. The ability to join the dots into complex assemblies creates many opportunities for scientific discovery. This motivated our present research work on QDIPs, DWELLs, and graphene like QDs. The intention of this research was to study the size dependent achievements of QDIPs, DWELLs, and graphene like QDs with those of competitive technologies, with the emphasis on the material properties, device structure, and their impact on the device performance.
In this dissertation four research studies pertaining to optical properties of quantum dot and dot-in-a-well infrared photodetectors, I-V characteristics of graphene quantum dots, and energy and spin texture of germanene quantum dots are presented. Improving self-assembled QD is a key issue in the increasing the absorption and improving the performance. In the present research work, an ideal self-assembled QD structure is analyzed theoretically with twenty-hole levels (Intraband optical transitions within the valence band) and twenty-electron energy levels (DWELL). Continuing the efforts to study self-assembled QDs we extended our work to graphene like quantum dots (graphene and germanene) to study the electronic transport properties.
We study numerically the intraband optical transitions within the valence band of InxGa1-xAs/GaAs pyramidal quantum dots. We analyze the possibility of tuning of corresponding absorption spectra by varying the size and composition of the dots. Both ‘x ’ and the size of the quantum dot base are varied. We have found that the absorption spectra of such quantum dots are more sensitive to the in-plane incident light.
We present numerically obtained absorption optical spectra of n-doped InAs/In0.15Ga0.85As/GaAs quantum dot-in-a-well systems. The absorption spectra are mainly determined by the size of the quantum dot and have weak dependence on the thickness of the quantum well and position of the dot in a well. The dot-in-a-well system is sensitive to both in-plane and out-of-plane polarizations of the incident light with much stronger absorption intensities for the in-plane-polarized light.
We also present theoretically obtained I-V characteristics of graphene quantum dots, which are realized as a small piece of monolayer graphene. We describe graphene within the nearest-neighbor tight-binding model. The current versus the bias voltage has typical step-like shape, which is due to discrete energy spectrum of the quantum dot. The current through the dot system also depends on the position of the electrodes relative to the quantum dot.
In relation to graphene quantum dots, we present our study of buckled graphene-like materials, like germanene and silicene. We consider theoretically germanene quantum dot, consisting of 13, 27, and 35 germanium atoms. Due to strong spin-orbit interaction and buckled structure of the germanene layer, the direction of the spin of an electron in the quantum dot depends on both the electron energy and external perpendicular electric field. With variation of energy, the direction of spin changes by approximately 4.50. Application of external electric field results in rotation of electron spin by approximately 0.50, where the direction of rotation depends on the electron energy.

In this thesis, the strength of the LBIC system is enhanced in different aspects that includes its feasibility as a non-destructive characterization tool, the signal analysis and development of analytical solution to have better understanding on the defects and inhomogeneities in the quantum dot based hetero-structures for device applications, finally understanding its limits due to the size of the laser beam and interpretation of artefacts in the signal appearance due to the presence of co-devices.
Chapter#1 provides the introduction and literature survey of the LBIC system. It covers the importance and area of application of the LBIC.
Chapter#2 various tools and instrumentation are discussed briefly for the systems that are developed in the lab as well as standard tools utilised for the material characterization. A LBIC instrumentation a novel colloidal quantum dots (CQD) thin film deposition system is discussed. In the last part along with the standard characterization systems a software tool (semiconductor device simulator) is discussed, which is used to visualize and understand the LBIC profile that is obtained experimentally.
Chapter#3 provides the information of colloidal synthesis of PbS and HgxCd1-xTe quantum dots. Device fabrication process is explained step by step for the following devices. p-n junction silicon diodes, PbS-CQD/Si hetero-structures, ITO/PbS-CQD/Al crossbar structures and HgCdTe-CQD/Si hetero-structures.
Chapter#4 deals with the major constraints imposed on the LBIC due to the need of Ohmic contacts. To overcome this major limitation, in this work, the origin of the signal is studied with the remote contact geometry for silicon p-n junction devices. It was observed that the signals can be collected with the capacitively coupled remote contacts, where LBIC was ultimately demonstrated as contactless measurement tool without any compromise on the measurements and thus obtained physical parameters. The effect of finite laser beam size is also described, which was found to have effect on the actual dimensions measured with the LBIC images. LBIC utility is further enhanced with the Si/CQD based hetero-structure devices, which are the potential candidates in the evolving device technology to be utilized in various modular systems such as PDs and LED applications.
Chapter#5 discusses the origin and possible mechanisms for lateral photo-voltage which is closely monitored in the PbS-CQD/Si hetero-junction device systems. Interestingly, it is observed that there are two different line profiles for n and p type Si substrates. Different mechanisms that give rise to this kind of profiles were found to be distinct and are related to the band alignment of the CQD/Si hetero-structure. It lead to the revelation of an interesting phenomenon and believed to be universally observed irrespective of the materials involved in the formation of hetero-junction. Simulations and experimental results are quite consistent and in agreement with each other, which confirm the underlying physical mechanism that connects the LBIC anomalies with the band alignment.
Chapter#6 deals with the spatial variations in the transverse photocurrent in the PbS-CQD film which is studied as a function of applied bias. Analytical equation is setup for the photocurrent in the CQD film under applied bias with the help of available transport mechanism and equations from the literature. The spatial non-uniformity that exists in the photocurrent proved to be the result of spatial inhomoginities in the physical parameters. By correlating the spatial data to the analytical equation, it is shown that the inhomoginities can be predicted. This approach is important for the devices, where monolithic detectors are fabricated by depositing CQD film on Read-Out-Integrated-Circuit (ROIC), where the manifestation of non-uniformity can be understood and probably fixed.
Chapter#7 HgCdTe CQD based devices are studied for the purpose of photo-detector applications in MWIR (3 5 μm) region. HgxCd1-xTe Colloidal quantum dots are technologically important due to their wide absorption range that covers different regions of the atmospheric window. HgxCd1-xTe are successfully synthesised, which covers the absorption edge up to ~6.25 m in the IR region. Absorption and photo-response studies are carried out on HgxCd1-xTe/Si hetero-junctions under incident IR radiation. It is observed that the band gap of the quantum dots can be tuned easily by controlling the growth time as a parameter, thus moulded HgxCd1-xTe CQD/Si hetero-structures were found to have good photo-response.
Chapter#8 the summary and the future direction and scope of the work is discussed. This includes the interesting observations during this thesis work which are not reported here in details.
