Intoxicação cúprica experimental em ovinos : aspectos clínicos e laboratoriais /Pereira, Wanderson Adriano Biscola. January 2008 (has links)

Quando a capacidade de armazenagem hepática se esgota, o cobre é liberado para o sangue causando sinais clínicos da intoxicação. Para verificar as alterações presentes na bioquímica sérica, hemograma, morfologia eritrocitária, perfil de proteínas séricas e concentração de cobre sérico, durante as fases pré-hemolítica e hemolítica da intoxicação crônica por cobre, foram utilizados seis ovinos distribuídos aleatoriamente em dois grupos: G-1 (controle) e G-2 (experimentalmente intoxicados). Os três ovinos do G-2, além da dieta diária receberam 3mg de CuSO4. 5H2O/Kg PV, seguido de aumentos semanais de 3mg de CuSO4. 5H2O/ Kg PV na dose diária. Diariamente realizou-se o exame físico dos ovinos de ambos os grupos. Para obtenção do hemograma e dos componentes bioquímicos sanguíneos foram colhidas amostras com antes (M0 - M3), durante (M4) e após (M5 e M6) a crise hemolítica. Foram realizadas necropsias dos ovinos do G-2 que morreram e dos ovinos do G-1, submetidos à eutanásia ao fim do experimento e fragmentos do fígado desses animais foram colhidos para avaliação histopatológica. Durante a fase pré-hemolítica evidenciaram-se poucas alterações, entretanto durante a crise hemolítica os ovinos do G-2 apresentaram anemia macrocítica normocrômica, predomínio de hemácias com morfologia de acantócitos, leucocitose por neutrofilia, elevação nas atividades séricas de AST,GGT, CK e hipercupremia; Os teores séricos de ceruloplasmina apresentaram-se diminuídos e os teores de transferrina, proteína de 35.000 Da e IgG de cadeia leve apresentaramse aumentados no M2; os animais intoxicados apresentaram anorexia, membranas mucosas ictéricas, fezes amolecidas de coloração verde-escura e hemoglobinúria. À necropsia observou-se mucosas, fígado e demais tecidos de coloração amarelada... When the liver storage capacity is exhausted, copper is released into the blood causing clinical signs of poisoning. Six lambs fed a basal diet were randomly assigned to 2 groups: G - 1 (control) and G-2 (experimentally intoxicated). The three sheep of the G-2, were drenched initially with 3 mg of CuSO4. 5H2O/ kg bw daily for a week. Every week an additional dose of 3mg CuSO4. 5H2O/ kg bw was included in the drench until signs of copper poisoning appeared. Every day, the lambs were monitored clinically. before (M0 - M3), during (M4) and after (M5 and M6) to hemolytic crisis, sample blood were obtained for biochemical analysis of AST, GGT and CK, blood count, serum proteins and serum concentration of copper. The lambs of the G-2 that died in the course of the experiment and the lambs of the G-1 euthanized at the end, were necropsied and fragments of liver, were obtained for histopathology. During the pre-hemolytic phase few changes were evidenced, however thought the hemolytic crisis the sheep of the G-2 were presented macrocytic normochromic anemia, a predominance of erythrocytes with acantocytes morphology, leukocytosis by neutrophilia, elevation in serum activities of AST, GGT and CK and hipercupremia; Serum levels of ceruloplasmin was presented reduced and the levels of transferrin, 35,000 of protein and IgG- light chain were increased in M2; the lambs of G-2 exhibited anorexia, jaundiced mucous membranes, soften and dark-green feces and hemoglobinuria. Gross examination of poisoned lambs revealed mucous membranes, liver and other tissues yellowish, dark kidneys and brownish urine. Histopathology of the liver showed megalocitose of hepatocytes, the disorganization of strands of hepatocytes, cholestasis and inflammatory infiltrate lymphocytic peri-portal. / Doutor

The purpose of this study was to evaluate the effects of an antimicrobial photodynamic therapy (aPDT) employing the photosensitizer methylene blue dissolved in ethanol on the systemic oxidative status likewise on the gingival collagen content of rats with periodontitis. Male Wistar rats were randomly divided into two main groups: NC (negative control; no periodontitis) and the remaining animals were submitted to periodontitis induction group. In the last group, the cotton ligature was placed at the first right mandibular molar of each animal in a submarginal position to induce experimental periodontitis. The animals with periodontitis were subdivided into groups according to the periodontal treatment as follow: SRP group (scaling and root planing), aPDT I group (SRP+aPDT+MB dissolved in water), and aPDT II group (SRP+aPDT+MB dissolved in ethanol). After 7 days, the ligature was removed and periodontal treatments were performed. At 7, 15 and 30 days, rats were euthanized and gingival tissue was removed for morphometric analysis. The erythrocytes were used to evaluate systemic oxidative status. Besides that, it indicated a protective influence of aPDT II in erythrocytes already at 15 days observed by the elevation in levels of systemic antioxidant defense. The morphometric findings showed that aPDT II group was the only one that restored the percentage of total collagen area also in 15 days, as well as, aPDT II group recovered the type I collagen area at the same time-point. According to this study we could suggest that aPDT employed as an adjunct to the standard treatment of periodontitis (SRP) increases systemic protective response against oxidative stress periodontitis-induced facilitating and accelerating the periodontal healing particularly when methylene blue is dissolved in ethanol. / O objetivo deste estudo foi avaliar os efeitos da terapia fotodinâmica antimicrobiana (TFDa) usando o fotossensibilizador azul de metileno (AM) dissolvido em etanol sobre o status oxidativo sistêmico, bem como sobre o conteúdo de colágeno gengival de ratos com periodontite. Ratos machos Wistar foram divididos aleatoriamente em dois grupos principais: CN (controle negativo; sem periodontite) e os animais restantes foram o grupo submetido a indução de periodontite. No último grupo, a ligadura de algodão foi colocada no primeiro molar inferior direito de cada animal em uma posição subgengival para induzir a periodontite experimental. Os animais com periodontite foram subdivididos em grupos de acordo com o tratamento periodontal, como segue: grupo RAR (raspagem e alisamento radicular), TFDa I grupo (RAR + TFDa + AM dissolvido em água), e grupo TFDa II (RAR + TFDa + AM dissolvido em etanol). Após 7 dias, a ligadura foi removida e foram realizados os tratamentos periodontais. Aos 7, 15 e 30 dias, os ratos foram submetidos à eutanásia e foi removido o tecido gengival para análise morfométrica. Os eritrócitos foram usados para avaliar o status oxidativo sistêmico. O status oxidativo demostrou maiores níveis de peroxidação lipídica no grupo CP em 7, 15 e 30 dias, e indicou uma influência protetora da TFDa II, nos eritrócitos, já em 15 dias, observada a partir da elevação dos níveis de defesa antioxidante sistêmica. Os achados morfométricos mostraram que o grupo TFDa II restabeleceu o percentual de área total de colágeno também em 15 dias, bem como recuperou a área de colágeno tipo I no mesmo tempo. A partir deste estudo podemos sugerir que TFDa utilizada como um adjuvante ao tratamento padrão periodontal (RAR) aumenta a resposta protetora sistêmica contra o estresse oxidativo induzido pela periodontite, facilitando e acelerando a cicatrização periodontal, particularmente quando o azul de metileno é solubilizado em etanol.

Krvetvorba u mihule mořské / Haematopoiesis in Sea lampreyKovář, Martin January 2017 (has links)
To find out if the haematopoietic system is common feature of vertebrates, we decided to examine haematopoiesis in a sea lamprey (Petromyzon marinus). All blood cells arises from the haematopoietic stem cells in higher vertebrates. We assume that this is common for the higher vertebrates and a jawless vertebrates, but nobody was interested in the jawless haematopoiesis since 1970. Using a reverse genetic, we identify homologues of important hematopoietic of higher vertebrates in transcriptome of the sea lamprey with emphasis on important receptors or transcription factors, because they can be used as the specific markers of different blood cells and their progenitors. Then we use those sequences for cloning, expression measurements and other work. We picked up sea lamprey as model organism because its unique phylogenetic position, important foe evo-devo studies, but also because lack of elementary knowledge about sea lamprey haematopoiesis. Towards Development Of Low Cost Electrochemical Biosensor For Detecting Percentage Glycated HemoglobinSiva Rama Krishna, V 01 1900 (has links) (PDF)
There is an ever growing demand for low cost biosensors in medical diagnostics. A well known commercially successful example is glucose biosensors which are used to diagonize and monitor diabetes. These biosensors use electrochemical analysis (electro analysis) as transduction mechanism. Electro analytical techniques involve application of electrical stimulus to the chemical/biochemical system under consideration and measurement of electrical response due to the oxidation and reduction reactions that occur because of the stimulus. They offer a lot of advantages in terms of sensitivity, selectivity, cost effectiveness and compatibility towards integration with electronics. Besides glucose, there are several biomolecules of significance for which electro analysis can potentially be used to develop low cost, rapid, easy to use biosensors. One such biomolecule is Glycated Hemoglobin (GHb). It is a post translational, non-enzymatic modification of hemoglobin with glucose and is a very good biomarker that indicates the average value of blood glucose over the past 120 days. It is always expresses as a percentage of total hemoglobin present in blood. Monitoring diabetes based on the value of percentage Glycated hemoglobin is advantageous as it gives an average value of glucose unlike plasma glucose values which vary a lot on a day to day basis depending on the dietary habits and the stress levels of the individual. This thesis is focused on the development of a low coat, easy to use, disposable sensor for measuring percentage Glycated hemoglobin.
The first challenge in developing such a sensor is isolation of hemoglobin. Unlike glucose which is present in blood plasma (liquid content of blood), hemoglobin resides inside red blood cells also known as erythrocytes. O isolate hemoglobin, these cells have to be broken or lysed. All the existing approaches rely on mixing blood with lysing reagents to lyse erythrocytes. Ideal biosensors should be devoid of liquid reagents. Keeping this in perspective, in this thesis, this challenge is addressed by developing two entirely buffer/reagentless techniques to lyse erythrocytes and isolate hemoglobin. In the first technique, cellulose acetate membranes are embedded with lysing reagents and are used for lysing reagents and are used for lysing application. In the second techniques, commercially available nylon mesh nets are modified with lysing reagents to lyse and isolate hemoglobin. These membranes or mesh nets can be easily integrated on top of a disposable strip.
After isolating hemoglobin, the next challenge is to selectively detect Glycated hemoglobin. Boronic acid conjugates are known to bind Glycated hemoglobin. Using this principle, a new composite is sysnthesized to specifically detect glc\ycated hemoglobin. The composite (GO-APBA) is a result of functionalization of Graphene Oxide (GO) with 3-aminophenylboronic acide (APBA). Detection of Glycated hemoglobin is achieved by modifying screen printed electrode strips with the synthesized compound, thus taking a step forwards achieving the objective.
Since Glycated hemoglobin is always expressed as a percentage of hemoglobin, the next challenge is to detect total hemoglobin. In this thesis a low cost way of detecting hemoglobin is achieved by using GO modified or surfactant modified screen printed electrode strips. Furthermore, the potential interferences that blood plasma can cause in these measurements are eliminated with the help of permselective coatings.
Thus using the technologies developed in this thesis, measurements of percentage Glycated hemoglobin can be potentially made on handheld electronic devices akin to glucose meters by using just a drop of blood.

Previous issue date: 2006 / Resumo: Surfatantes ou detergentes são compostos anfifílicos que, na presença de água, têm a característica de formar agregados micelares. Surfatantes induzem a desestruturação de outros agregados como bicamadas sendo, por isso, usados para ruptura celular ou solubilização de lipídios e proteínas de membrana. A capacidade lítica dos surfatantes resulta de sua estrutura química, que determina o modo de interação dos mesmos com as membranas. Em concentrações mais altas (acima da concentração micelar crítica), os surfatantes desestabilizam as bicamadas lipídicas, levando à formação de micelas-mistas. Ensaios hemolíticos são bons modelos para estudo do efeito lítico de surfatantes em biomembranas. Aplicando em eritrócitos humanos o tratamento quantitativo proposto por Lichtenberg (1985) para estudo da solubilização de bicamadas lipídicas mensurou-se, neste trabalho, as concentrações para início (Csat) e 100% de hemólise (Csol), induzidas por 25 surfatantes clássicos, pertencentes a cinco diferentes famílias. A variação dos valores de Csat, Csol determinada com diferentes hematócritos permitiu o cálculo da constante de ligação surfatante/membrana e da razão surfatante/lipídio de membrana (Re) para início e 100% de hemólise. O parâmetro Re foi usado para classificar os detergentes como fortes, médios ou fracos agentes solubilizantes, com boa correlação com dados da literatura o que nos permitiu propor seu uso para descrever o efeito lítico de surfatantes, como uma alternativa simples e aplicável as membranas biológicas. As transições durante o processo hemolítico foram acompanhadas pela técnica de Ressonância Paramagnética Eletrônica, com uso marcador de spin 5 doxil-estearato (incorporado a 1 mol% nas membranas de eritrócito) e lise induzida pelo surfatante não iônico Triton X100. Concomitante ao aparecimento de hemoglobina e fosfato livres no sobrenadante - indicadores da ruptura da membrana, medidas do parâmetro de ordem daquele marcador de spin permitiram estudar as transições que acontecem durante (membrana:membrana mista) e após (membrana mista:micela mista) a hemólise / Abstract: Surfactants or detergents are amphiphilic compounds that form micellar aggregates in the presence of excess water. Surfactants are able to induce disruption of lamellar aggregates, justifying their use for cell lysis or in the extraction of membrane constituents such as lipids and proteins. The lytic capacity of a given surfactant is determined by its chemical structure, that rules its interaction with the membranes. At high concentration (above the critic micelle concentration), surfactants destabilize lipid bilayer leading to mixed micelle formation. Hemolytic assays are a good model to study the lytic effect of surfactants on biomembranes. In this study we have applied to human erythrocytes the quantitative treatment proposed by Lichtenberg (1985) to describe the solubilization of model lipid membranes. The concentration for onset (Csat) and complete (Csol) hemolysis induced by 25 classic surfactants from five different families were measured. Changes in Csat and Csol values at different hematocrits allowed the determination of the surfactant/membrane lipid molar ratio (Re) for beginning and 100% lysis. The Re arameter was used to classify the surfactants as strong, medium or weak membrane solubilizers. The classification was in good correlation with data in the literature, allowing us to recommend the use of Re parameter to describe the lyric effect of surfactants on biomembranes. The transitions in the hemolytic process were accompanied by Electron Paramagnetic Resonance, using the 5-doxyl-stearate spin-probe (1 mol%, incorporated in the erythrocyte membrane) and the non-ionic surfactant Triton X100. Background and Aims: During positive energy balance, excess lipid storage in subcutaneous adipose tissue (SAT) is associated with increased lipolysis. Elevated circulating fatty acid (FA) concentrations from both SAT lipolysis and dietary fat intake may result in visceral adipose tissue (VAT) accumulation, impairment of glucose metabolism, altogether increasing obesity-associated metabolic risks. We aimed to test the hypothesis that FA composition of red blood cell total phospholipids (RBC-TPL) and SAT is associated with body fat centralisation (VAT/SAT ratio) and insulin sensitivity (SI) in black South African women with obesity. Methods: Participants’ (n = 41) body fat composition and distribution, SI, and RBC-TPL, abdominal and gluteal SAT (gSAT) FA composition (gas-liquid chromatography) were measured. Results: RBC-TPL contained higher proportions of saturated fatty acids (SFAs) than SAT (p < 0.001), which were associated with lower SI (p < 0.05). Mono-unsaturated fatty acids (MUFAs) and stearoyl-CoA desaturase-1 (SCD1)-16 were lower, while poly-unsaturated fatty acids (PUFAs), and delta-5 and delta-6 desaturase indices were higher in RBC-TPL than SAT (p < 0.001). Interestingly, FA profiles differed between SAT depots with higher SFAs and lower MUFAs, SCD1-16 and SCD1-18 indices in abdominal compared to gluteal SAT (p < 0.01). In both SAT depots, higher SFAs and lower PUFAs (n-3 and n-6) correlated with lower VAT/SAT ratio; and lower PUFAs (n-3 and n-6) and higher total MUFA correlated with higher SI. Conclusion: Our findings confirm the relationships between the FA composition of RBC-TPL and SAT and metabolic risk in black women with obesity, which are dependent on both the FA class, and the tissue type/blood compartment in which they are distributed.

Transfusions de globules rouges en néonatologie et syndrome de défaillance multiviscérale aiguëVilleneuve, Andréanne 04 1900 (has links)
Le niveau d’hémoglobine (Hb) d’un nouveau-né diminue dans les premiers mois de vie. Cette
anémie dite physiologique est plus sévère chez les nourrissons admis aux soins intensifs
néonataux (SIN), et ceux-ci nécessitent souvent une transfusion sanguine. En néonatalogie, les
indications de transfuser sont controversées et les pratiques transfusionnelles sont très
variables. Pour mieux comprendre ces pratiques, nous avons mené l’étude prospective
«Epidemiology and determinants of red blood cells transfusion in a neonatal intensive care
unit: a cohort study». 13.4% des patients consécutifs admis aux SIN pendant l’étude ont reçu
au moins une transfusion sanguine. Les prématurés nés à moins de 28 semaines d’âge
gestationnel ont reçu la majorité des transfusions (62.2%) mais les nourrissons à terme admis
aux SIN sont aussi fréquemment transfusés (4.9% des transfusions). Les principales
justifications évoquées par les cliniciens prescrivant des transfusions sont un niveau bas d’Hb,
la maladie de base et le désir d’améliorer l’oxygénation des organes. Notre étude a confirmé
une grande variabilité du seuil d’Hb justifiant une transfusion, s’étendant de 62 à 137 g/L.
Le syndrome de défaillance multiviscérale (SDMV), défini par l’observation simultanée d’au
moins deux dysfonctions d’organes, est un facteur important de mortalité-morbidité chez les
enfants traités en soins intensifs pédiatriques. L’association entre SDMV et transfusions est
bien décrite dans cette population. Deux listes de critères diagnostiques du SDMV pédiatrique
sont utilisées dans la littérature médicale : celles de Proulx et de Goldstein. Nous avons
entrepris l’étude de cohorte prospective «Multiple organ dysfunction syndrome in critically ill
children : clinical value of two lists of diagnostic criteria» dans le but de valider et de
comparer leur valeur diagnostique respective. Nos résultats ont démontré que l’épidémiologie
du SDMV varie selon la définition utilisée : l’incidence était de 21.4% vs. 37.3% selon les
critères de Proulx et de Goldstein respectivement. Les deux listes de critères diagnostiques
ont une bonne reproductibilité inter- et intra-observateur; celle de Proulx est cependant
associée à une plus haute mortalité à 90 jours (17.8% vs. 11.5%, p = 0.038). Le SDMV a été
décrit chez les nouveau-nés en SIN en utilisant le NEOMOD, un score adapté à cette
population. Avec une meilleure caractérisation, le SDMV deviendrait un critère de jugement
intéressant pour les essais cliniques randomisés en médecine transfusionnelle en néonatologie. / In the first few months of life, the level of hemoglobin (Hb) in the newborn normally
decreases. This physiological anemia is more severe in neonates admitted to a neonatal
intensive care unit (NICU), who frequently require a red blood cells (RBC) transfusion. In
neonatal medicine, the indications for transfusion are controversial and practices are highly
variable. To better understand those practices, we conducted the prospective study:
«Epidemiology and determinants of red blood cells transfusion in a neonatal intensive care
unit: a cohort study». Among the patients consecutively admitted to NICU during the study
period, 13.4% received at least one RBC transfusion. Although premature babies born at less
than 28 weeks gestation received the majority of transfusions (62.2%), term neonates admitted
to NICU were also frequently transfused (4.9% of transfusions). The main justifications for
giving a RBC transfusion were: low Hb level, underlying illness and to improve oxygen
delivery. We also observed a wide range of Hb thresholds that triggered a decision to transfuse
(from 62 to 137 g/L).
The multiple organ dysfunction syndrome (MODS), which is defined as the simultaneous
dysfunction of at least two organs or systems, is highly associated with mortality and
morbidity in critically ill children. The association between MODS and transfusions is also
well described in this population. Two sets of criteria of pediatric MODS are currently used in
the medical literature: one by Proulx, and another by Goldstein. We did the prospective cohort
study «Multiple organ dysfunction syndrome in critically ill children : clinical value of two
lists of diagnostic criteria» to validate and compare the diagnostic value of those two
definitions of MODS. We observed that the epidemiology of MODS varies according to which
list of criteria is used: the incidence was 21.4% vs. 37.3% with Proulx and Goldstein criteria,
respectively. Both sets of criteria have a good inter- and intra-rater reproducibility. The
diagnostic of MODS according to Proulx criteria is associated with higher 90-days mortality
(17.8% vs. 11.5%, p = 0.038). MODS is also described in neonates, using a score adapted to
this population, the NEOMOD. Neonatal MODS represents an interesting outcome measure in
clinical trials in neonatal transfusion medicine. However, prior to that, it needs to be better

Évaluation des effets de l'administration de fer intramusculaire sur l'anémie chez les oiseaux de proieDubé, Catherine 04 1900 (has links)
L’administration de fer dextran à 10 mg/kg intramusculaire (IM) est un
traitement empirique couramment recommandé en médecine aviaire lors
d’hémorragie ou d’anémie. L’objectif principal de cette étude était d’évaluer
les effets de ce traitement sur l’anémie chez les oiseaux de proie. Deux types
d’individus ont été utilisés : des crécerelles d’Amérique (Falco sparverius) où
une anémie par perte de sang externe aiguë a été créée (deux phlébotomies
de 20-40 % du volume sanguin total à un intervalle de 6 h) et des oiseaux de
proie sauvages de différentes espèces souffrant d’anémies diverses.
L’ensemble des oiseaux a été subdivisé aléatoirement en groupe traitement

pas d’effet sur l’érythropoïèse des rapaces souffrant d’anémie par perte de
sang externe aiguë, qu’elle provoque une légère inflammation au site
d’injection et qu’elle n’influence pas les réserves de fer. Le comptage des
réticulocytes en anneau et des polychromatophiles semble être deux

empirical treatment recommended in avian medicine for hemorrhage and
anemia. The purpose of this study was to evaluate the effects of this
treatment on anemia in birds of prey. Two kinds of specimen were used: the
American kestrel (Falco sparverius) where an acute external blood loss
anemia was created (with two phlebotomies of 20-40 % of the total blood
volume at 6 hours interval) and other various species of wild birds of prey
suffering from different types of anemia. All subjects were randomized into a
treatment (iron dextran 10 mg/kg IM) or a control (NaCl 0,9 % IM) group.
Monitoring was carried out to evaluate the evolution of the anemia, presence
of side effects and impact of an iron administration on their iron reserve. No
significant differences were observed between the two treatment groups for
clinical signs, packed cell volume, the percentage of
reticulocytes/polychromatophilic erythrocytes, bone marrow cellularity and
iron, plasmatic iron and creatine kinase. Most kestrels had a myositis at the
iron injection site. Our results suggest that an IM injection of 10 mg/kg iron
dextran has no effect on raptor erythropoiesis after an acute external blood
loss anemia, that it has no effect on iron reserve, and that it can cause mild
inflammation at the injection site. The polychromatophilic erythrocytes and the
reticulocytes ring form count were two equivalent methods.

L’administration de fer dextran à 10 mg/kg intramusculaire (IM) est un
traitement empirique couramment recommandé en médecine aviaire lors
d’hémorragie ou d’anémie. L’objectif principal de cette étude était d’évaluer
les effets de ce traitement sur l’anémie chez les oiseaux de proie. Deux types
d’individus ont été utilisés : des crécerelles d’Amérique (Falco sparverius) où
une anémie par perte de sang externe aiguë a été créée (deux phlébotomies
de 20-40 % du volume sanguin total à un intervalle de 6 h) et des oiseaux de
proie sauvages de différentes espèces souffrant d’anémies diverses.
L’ensemble des oiseaux a été subdivisé aléatoirement en groupe traitement
