Insulin Resistance and Muscle Insulin Receptor Substrate-1 Serine Hyperphosphorylation

Insulin resistance in metabolic syndrome subjects is profound in spite of muscle insulin receptor and insulin-responsive glucose transporter (GLUT4) expression being nearly normal. Insulin receptor tyrosine kinase phosphorylation of insulin receptor substrate-1 (IRS-1) at Tyr896 is a necessary step in insulin stimulation of translocation of GLUT4 to the cell surface. Serine phosphorylation of IRS-1 by some kinases diminishes insulin action in mice. We evaluated the phosphorylation status of muscle IRS-1 in 33 subjects with the metabolic syndrome and seventeen lean controls. Each underwent euglycemic insulin clamps and a thigh muscle biopsy before and after 8 weeks of either strength or endurance training. Muscle IRS-1 phosphorylation at six sites was quantified by immunoblots. Metabolic syndrome muscle IRS-1 had excess phosphorylation at Ser337 and Ser636 but not at Ser307, Ser789, or Ser1101. Ser337 is a target for phosphorylation by glycogen synthase kinase 3 (GSK3) and Ser636 is phosphorylated by c-Jun N-terminal kinase 1 (JNK1). Exercise training without weight loss did not change the IRS-1 serine phosphorylation. These data suggest that baseline hyperphosphorylation of at least two key serines within muscle IRS-1 diminishes the transmission of the insulin signal and thereby decreases the insulin-stimulated translocation of GLUT4. Excess fasting phosphorylation of muscle IRS-1 at Ser636 may be a major cause of the insulin resistance seen in obesity and might prevent improvement in insulin responsiveness when exercise training is not accompanied by weight loss.

Effect of a 10 Day Decrease in Physical Activity on Circulating Angiogenic Cells

Circulating angiogenic cells (CACs) are early predictors of cardiovascular health and are inversely proportional to related outcomes. Increased number and function of CACs is seen in healthy individuals compared with individuals with cardiovascular disease (CVD). Exercise increases CAC number and function in CVD populations, through a nitric oxide-mediated mechanism. Inactivity is a growing concern in industrialized nations; it is an independent risk factor for CVD and is linked to increased mortality. The purpose of this study was to understand the effect of reduced physical activity (rPA) on two CAC populations (CFU-Hill and CD34+) in highly active individuals. We examined the mechanisms underlying changes in CAC function as a result of rPA with maintained energy balance. The two sub-populations of CACs responded differently to rPA. CFU-Hill CACs, decreased in number and amount of intracellular nitric oxide while CD34+ cells, did not change. Gene expression analyses indicated that oxidative stress- related genes did not change in CFU-Hill cells with rPA. However, correlations between CFU-Hill cell numbers, intracellular nitric oxide, and genes that are related to nitric oxide were observed. We concluded that rPA caused the observed decrease in CFU-Hill number and intracellular nitric oxide through a decrease in nitric oxide cellular availability, not oxidative stress.

Associations Between Fat Free Mass Percentage and Relative Force Production in Two Strength Tests

Background: Knowledge of the relationship between fat free mass (FFM) and strength capacity is not only useful in athlete coaching, but may also be important in developing clinical strategies for evaluating health in general populations. Sufficient skeletal muscle mass and strength are important factors for health and physical function. Bioelectrical impedance analysis (BIA) is a popular and clinically useful tool for assessing body composition. However, being able to simultaneously estimate whole body muscle strength without having to perform additional tests may prove useful in evaluating an individual’s health and physical function. Aim: The primary purpose of this study was to quantify the association between total body fat free mass percentage (TBFFM%) and relative strength in the isometric midthigh pull test (IMTP) and grip strength (GS) test in a healthy general young Swedish population. A secondary purpose was to investigate if there are any sex differences.  Methods: The design for this study was cross-sectional. The studied sample represented a general Swedish population of healthy young adults. Participants were asked to participate in testing on one occasion. Three tests were used for collecting data: BIA for data on FFM and both IMTP and hand dynamometer grip strength test for data on maximal force production. All tests were performed in the movement lab on Halmstad University campus. Statistical analyses were done using Pearson’s correlation and linear regression analysis. Results: Participants for this study consisted of healthy females (n=18) and males (n=17) aged 19-39. The primary findings of this study are that there are strong correlations between TBFFM% and relative strength (r = 0.621) in the IMTP test as well as relative GS (r = 0.705) on a whole-group level. The correlation between TBFFM% and relative GS was slightly stronger. Categorized by sex, we found strong positive correlations between TBFFM% and relative IMTP (r = 0.551) and relative GS (r = 0.596) for the female group and moderately strong correlations for the male group (relative IMTP r = 0.411; relative GS  r = 0.422).On a whole-group level, every unit increase in TBFFM% increases relative strength in the IMTP by 0.538N/kgBW (β = 0.538) and relative GS by 0.015 kg/kgBW (β = 0.015). Conclusion: TBFFM% was strongly associated with both relative IMTP strength and relative GS where the association was stronger for females than for males. TBFFM% values may be indicative of an individual's strength, which could be of value in a research setting.

As small molecules, preparadas por meio de sucessivas formações de organo-estananas e reações de Stille, também foram sintetizadas com sucesso e em rendimentos apreciáveis. A espécie J2 não foi preparada com sucesso, obtendo-se, em contrapartida, um análogo dessa molécula, que foi devidamente isolado e caracterizado. No que tange à aplicação dos materiais em dispositivos fotovoltaicos, os resultados até então obtidos mostraram-se inconclusivos, sendo digno de nota, no entanto, que o estudo continua sendo conduzido por colaboradores. O trabalho de aplicação dos materiais em dispositivos magnetorresistivos apresentou resultados interessantes para os oligômeros FBF, BFB, FFF e FF, visto que dispositivos fabricados a partir das referidas espécies mostraram variação em seus valores de corrente elétrica quando submetidos a campo magnético de 200 mT. Por fim, foram desenvolvidos narizes eletrônicos com duas finalidades distintas: (a) identificação de diferentes espécies de madeira de interesse ambiental e (b) estudo da influência da atividade física no indivíduo por meio da análise do suor. Os sensores de gás foram preparados por meio da deposição de finos filmes de materiais orgânicos conjugados dopados sobre a superfície de eletrodos interdigitados. No estudo acerca da identificação de espécies de madeira, a análise de leave-one-out revelou 100 % de taxa de acerto na diferenciação entre as espécies angelim, cedro-rosa, imbuia e perobinha. The polymers and oligomers are all poli(p-phenylenevinylene)s derivatives containing 2,1,3-benzothiadiazole and/or substituted fluorene units in the main chain. Those materials\' preparation comprises a coupling Wittig reaction as the key step, through which the vinylenic bonds are formed. The small molecules mentioned above were synthesized by successive organostannane preparations followed by cross-coupling Stille reactions, leading to the target-compounds in considerably high yields. Although the route is a very useful methodology for synthesizing organic conjugated small molecules, J2 was not successfully prepared. The reaction led to a J2 analogue instead, that was properly isolated and characterized. The application of the cited materials in organic photovoltaics (OPVs) is still in progress, since the study has shown inconclusive results so far. The study of the organic magnetoresistive properties of the synthesized materials has led to interesting results for the oligomers FBF, BFB, FFF and FF. The devices containing those compounds in their active layers presented considerable variations in their electric current values when submitted to a magnetic field of 150 mT magnitude. Finally, electronic noses for two different applications were developed: (a) the identification of wood species that can be easily mistaken and (b) the study of exercise physiology through analyses of sweat samples. The gas sensors were prepared by the deposition of doped organic conjugated materials thin films onto the surface of intedigitated electrodes. The study on the differentiation of several species of wood showed 100 % rate of hits determined by leave-one-out statistics analysis. The study on the exercise physiology was also promising since it allowed the identification of the three different stages of the physical exercise with a 93 % rate of hits, also determined by leave-one-out methods. Thus, both studies suggest that the electronic nose can be a powerful tool to study many different targets in which the release of volatile compounds is involved

Expressão e seleção de microRNAs no músculo esquelético de homens saudáveis submetidos ao treinamento físico aeróbio\" / MicroRNA-206 skeletal muscle-specific modulates pattern of expression in myogenesis in response to endurance training in human

O treinamento físico aeróbio (TFA) foi estabelecido como uma conduta importante capaz de alterar a musculatura esquelética humana. Os microRNAs (miRs) surgiram como importantes reguladores de processos biológicos, modulando a expressão de genes pós-transcricionalmente. Os myomiRs são miRs específico do músculo esquelético, em especial o miR-206, que é necessário para uma eficiente regeneração das fibras musculares esqueléticas. No entanto, a expressão do miR-206 em resposta ao TFA, não é completamente comprendida. O objetivo do presente estudo foi determinar os padrões de expressão dos myomiRs na musculatura esquelética humana. Doze voluntários saudáveis foram biopsiados pré e pós-treinamento físico. As expressões gênicas e proteicas envolvidas na miogênese foram observadas, incluindo; PAX-7, MYF5, MYOD, MRF4, MYOG, CD31 e FSTL. Além disso, a freqüência cardíaca (FC), pressão arterial média (PAM), consumo máximo de oxigênio (VO2max), fluxo sanguineo no antebraço (FSA) e condutância vascular no antebraço (CVA), foram avaliados. Ademais, os myomiRs foram analisados por PCR em tempo real. O treinamento físico aeróbio foi realizado durante 16 semanas. Todas as variáveis foram reavaliadas após o treinamento. Os indivíduos apresentaram um aumento nas expressões dos myomiRs, em especial do miRs-206 de 93%. Estas alterações foram acompanhadas por aumento nas expressões dos genes; PAX-7, MYOD, MYF5, MFR4, MYOG e FSTL, respectivamente. No entanto, quando analisamos as expressões proteicas, houve redução na FSTL e PAX-7, de 24%, 29%, respectivamente. Além disso, em MYOD, CD31, MYOG e MHC houve aumentos de 21%, 41%, 79% e 94%, respectivamente. Ademais, houve uma diminuição na frequência cardíaca de reposuso de 12,5% e aumentos no VO2pico, FSA e CVA de 14,1%, 68%, 63%, respectivamente. Estes resultados sugerem que em indivíduos saudáveis o miRs-206 é altamente expresso após o treinamento físico aeróbio, dessa forma, modulando localmente processos miogênicos regenerativos em homens saudáveis / Endurance training (ET) has been established as an important phenotype capable of altering the human skeletal muscle. MicroRNAs (miRs) have emerged as important regulators of numerous biological processes by modulating gene expression at the post-transcriptional level. The myomiRs are particulars miRs of muscles, in special skeletal muscle-specific miR-206 that is required for efficient regeneration muscle fiber. However, the expression of myomiRs and in special miR-206 in response to ET in human skeletal muscle is not completely understood. Twelve healthy volunteers were biopsied pre and post period endurance training. Most of the biological processes involved in the transcriptional regulation were observed, including PAX-7, MYF5, MYOD, MRF4, MYOG, CD31 and FSTL, analyzed by real time PCR. Moreover, heart rate (HR), mean blood pressure (MBP), maximal exercise capacity (VO2peak) forearm blood flow (FBF) and forearm vascular conductance (FVC) were evaluated. The myomiRs levels analyzed by real-time PCR. Endurance training was performed for 16 weeks. All variables were re-assessed following completion of the training period. After endurance training, the individuals showed an increase in myomiRs, in special of 93% in human skeletal muscle in miRNA-206 levels. These alterations were accompanied by increase in PAX-7, MYOD, MYF5, MFR4, MYOG and FSTL gene expression, respectively. However, when analyzed by western blot comparing pre and post period there were reduction in FSTL of 24% and PAX-7 of 29% in protein levels, but in MYOD, CD31, MYOG and MHC there were increase of 21%, 41%, 79% and 94% in protein levels, respectively. In addition, there was a decrease in hear rate of 12.5% and increases in VO2peak of 14.1%, FBF of 68% and FVC of 63%.These results suggest that in healthy individuals the miR-206 is highly expressed after endurance training, thus modulating locally important parts in myogenic processes in humans

O estudo da influência da atividade física no indivíduo também se mostrou promissor na medida em que identificou os diferentes estágios do exercício físico com taxa de acerto de 93 %, também definida pela técnica estatística de leave-one-out. / The present work consists on the synthesis of two polymers - poly(2,1,3- benzothiadiazole-4,7-ylenevinylene-alt-9,9-n-dioctyl-2,7-fluorenylenevinylene) (PBTDV-alt-PDO27FV) and poly[4,7-(2,1,3-benzothiadiazole-ylene)-alt-(2,5-dioctyloxy-1,4-phenylenevinylene)] (PBTDV-alt-PDOPPV) - five oligomers - 4,7-bis(2-(9,9-n-dioctyl- 9H-fluorene-2-yl)vinyl)2,1,3-benzothiadiazole (FBF); 2,7-bis(2-(2,1,3- benzothiadiazole-4-yl)vinyl)9,9-n-dioctyl-9H-fluorene (BFB); 4-(2-(9,9-n-dioctyl-9H-fluorene-2-yl)vinyl)2,1,3-benzothiadiazole (FB); 1,2-bis(9,9-n-dioctyl-9H-fluorene)2- vinylene (FF) and 2,2\'-(2,2\'-(9,9-n-dioctyl-9H-fluorene)2,7-bis(vinylene)bis(9,9-n-dioctyl-9H-fluorene) (FFF) - and three small molecules - 7,7\'-(4,4-bis(2-ethyl-hexyl)-4H-silolo[3,2-b-:4,5-b\']dithiophene-2,6-diyl)bis(6-fluoro-4-(5\'-hexyl-[2\'-thiophene-2\'- thiazole]-5-yl)benzo[c][1,2,5]thiadiazole (G37FBT); 7,7\'-(4,4-bis(2-ethyl-hexyl)-4H-silolo[3,2-b-:4,5-b\']dithiophene-2,6-diyl)bis(6-fluoro-4-(2\'-isobutyl-[5\',2\'-dithiazole]-5-yl)benzo[c][1,2,5]thiadiazole (J1) and 7,7\'-(4,4-bis(2-ethyl-hexyl)-4H-silolo[3,2-b-:4,5-b\']dithiophene-2,6-diyl)bis(5\'-hexyl-[2,2-bithiophene]-5-bis(yl)benzo[c][1,2,5]thiadiazole) (J2) - followed by the application of such materials in organic photovoltaics (OPV), magnetoresistive devices and gas sensors. The polymers and oligomers are all poli(p-phenylenevinylene)s derivatives containing 2,1,3-benzothiadiazole and/or substituted fluorene units in the main chain. Those materials\' preparation comprises a coupling Wittig reaction as the key step, through which the vinylenic bonds are formed. The small molecules mentioned above were synthesized by successive organostannane preparations followed by cross-coupling Stille reactions, leading to the target-compounds in considerably high yields. Although the route is a very useful methodology for synthesizing organic conjugated small molecules, J2 was not successfully prepared. The reaction led to a J2 analogue instead, that was properly isolated and characterized. The application of the cited materials in organic photovoltaics (OPVs) is still in progress, since the study has shown inconclusive results so far. The study of the organic magnetoresistive properties of the synthesized materials has led to interesting results for the oligomers FBF, BFB, FFF and FF. The devices containing those compounds in their active layers presented considerable variations in their electric current values when submitted to a magnetic field of 150 mT magnitude. Finally, electronic noses for two different applications were developed: (a) the identification of wood species that can be easily mistaken and (b) the study of exercise physiology through analyses of sweat samples. The gas sensors were prepared by the deposition of doped organic conjugated materials thin films onto the surface of intedigitated electrodes. The study on the differentiation of several species of wood showed 100 % rate of hits determined by leave-one-out statistics analysis. The study on the exercise physiology was also promising since it allowed the identification of the three different stages of the physical exercise with a 93 % rate of hits, also determined by leave-one-out methods. Thus, both studies suggest that the electronic nose can be a powerful tool to study many different targets in which the release of volatile compounds is involved

Expressão e seleção de microRNAs no músculo esquelético de homens saudáveis submetidos ao treinamento físico aeróbio\" / MicroRNA-206 skeletal muscle-specific modulates pattern of expression in myogenesis in response to endurance training in human

O treinamento físico aeróbio (TFA) foi estabelecido como uma conduta importante capaz de alterar a musculatura esquelética humana. Os microRNAs (miRs) surgiram como importantes reguladores de processos biológicos, modulando a expressão de genes pós-transcricionalmente. Os myomiRs são miRs específico do músculo esquelético, em especial o miR-206, que é necessário para uma eficiente regeneração das fibras musculares esqueléticas. No entanto, a expressão do miR-206 em resposta ao TFA, não é completamente comprendida. O objetivo do presente estudo foi determinar os padrões de expressão dos myomiRs na musculatura esquelética humana. Doze voluntários saudáveis foram biopsiados pré e pós-treinamento físico. As expressões gênicas e proteicas envolvidas na miogênese foram observadas, incluindo; PAX-7, MYF5, MYOD, MRF4, MYOG, CD31 e FSTL. Além disso, a freqüência cardíaca (FC), pressão arterial média (PAM), consumo máximo de oxigênio (VO2max), fluxo sanguineo no antebraço (FSA) e condutância vascular no antebraço (CVA), foram avaliados. Ademais, os myomiRs foram analisados por PCR em tempo real. O treinamento físico aeróbio foi realizado durante 16 semanas. Todas as variáveis foram reavaliadas após o treinamento. Os indivíduos apresentaram um aumento nas expressões dos myomiRs, em especial do miRs-206 de 93%. Estas alterações foram acompanhadas por aumento nas expressões dos genes; PAX-7, MYOD, MYF5, MFR4, MYOG e FSTL, respectivamente. No entanto, quando analisamos as expressões proteicas, houve redução na FSTL e PAX-7, de 24%, 29%, respectivamente. Além disso, em MYOD, CD31, MYOG e MHC houve aumentos de 21%, 41%, 79% e 94%, respectivamente. Ademais, houve uma diminuição na frequência cardíaca de reposuso de 12,5% e aumentos no VO2pico, FSA e CVA de 14,1%, 68%, 63%, respectivamente. Estes resultados sugerem que em indivíduos saudáveis o miRs-206 é altamente expresso após o treinamento físico aeróbio, dessa forma, modulando localmente processos miogênicos regenerativos em homens saudáveis / Endurance training (ET) has been established as an important phenotype capable of altering the human skeletal muscle. MicroRNAs (miRs) have emerged as important regulators of numerous biological processes by modulating gene expression at the post-transcriptional level. The myomiRs are particulars miRs of muscles, in special skeletal muscle-specific miR-206 that is required for efficient regeneration muscle fiber. However, the expression of myomiRs and in special miR-206 in response to ET in human skeletal muscle is not completely understood. Twelve healthy volunteers were biopsied pre and post period endurance training. Most of the biological processes involved in the transcriptional regulation were observed, including PAX-7, MYF5, MYOD, MRF4, MYOG, CD31 and FSTL, analyzed by real time PCR. Moreover, heart rate (HR), mean blood pressure (MBP), maximal exercise capacity (VO2peak) forearm blood flow (FBF) and forearm vascular conductance (FVC) were evaluated. The myomiRs levels analyzed by real-time PCR. Endurance training was performed for 16 weeks. All variables were re-assessed following completion of the training period. After endurance training, the individuals showed an increase in myomiRs, in special of 93% in human skeletal muscle in miRNA-206 levels. These alterations were accompanied by increase in PAX-7, MYOD, MYF5, MFR4, MYOG and FSTL gene expression, respectively. However, when analyzed by western blot comparing pre and post period there were reduction in FSTL of 24% and PAX-7 of 29% in protein levels, but in MYOD, CD31, MYOG and MHC there were increase of 21%, 41%, 79% and 94% in protein levels, respectively. In addition, there was a decrease in hear rate of 12.5% and increases in VO2peak of 14.1%, FBF of 68% and FVC of 63%.These results suggest that in healthy individuals the miR-206 is highly expressed after endurance training, thus modulating locally important parts in myogenic processes in humans

Les facteurs de risque maternels, la santé pondérale et cardiométabolique des jeunes, et l’importance des habitudes de vie

La santé maternelle et la santé pédiatrique font partie des priorités de santé publique mondiale. La présente thèse a pour objectif d’élargir les connaissances portant sur l’influence de la santé maternelle durant la grossesse sur la santé pondérale et cardiométabolique de la population pédiatrique. Une première étude a montré qu’une exposition intra-utérine à une ou plusieurs conditions gestationnelles sous-optimales (diabète gestationnel, désordres hypertensifs de la grossesse, tabagisme maternel durant la grossesse) avait un effet délétère sur la santé pondérale et cardiométabolique de la population pédiatrique et que cet effet était spécifique au sexe. Une seconde étude a montré que l’efficacité mécanique et l’oxydation des substrats énergétiques (lipides et glucides) au repos et durant l’exercice ne sont pas altérées dans cette population en comparaison avec les enfants non exposés. Ainsi, ce résultat suggère que d’autres mécanismes seraient à l’origine des effets délétères des conditions gestationnelles sous-optimales sur la santé pondérale et cardiométabolique de cette population. Enfin, la troisième étude a montré que seule l’activité physique d’intensité légère réduisait l’effet délétère d’une exposition intra-utérine à plus d’une condition gestationnelle sous-optimale sur le taux de cholestérol à lipoprotéine de haute densité chez les garçons. De plus, une durée de sommeil correspondant aux recommandations canadiennes actuelles n’avait pas d’effet protecteur vis-à-vis du risque d’obésité abdominale (enfants) et d’obésité (filles) dans le contexte d’une exposition au tabagisme maternel durant la grossesse. Par ailleurs, les autres saines habitudes de vie telles que l’activité physique d’intensité moyenne à élevée et une alimentation riche en fruits et légumes, en produits laitiers et en produits céréaliers n’ont pas contrecarré les effets délétères d’une exposition intra-utérine à une ou plusieurs conditions gestationnelles sous-optimales sur la santé pondérale et cardiométabolique des enfants. L’ensemble de ces résultats souligne l’importance de la santé maternelle durant la grossesse pour la santé pondérale et cardiométabolique de la population pédiatrique. Par ailleurs, étant donné que ces effets délétères ne sont que peu contrecarrés par les habitudes de vie durant l’enfance, il reste important de continuer les efforts de prévention et de prise en charge des conditions gestationnelles sous-optimales auprès des femmes enceintes qui les présentent. En terminant, il est important de continuer à explorer les mécanismes sous-jacents à ces effets délétères et à déterminer si des interventions en habitudes de vie peuvent prévenir l’obésité et les facteurs de risque cardiométaboliques dans cette population. / Maternal and children’s health are major worldwide public health concerns. The current thesis aimed to explore the impact of maternal health during pregnancy on children’s and adolescents’ health and included three original research projects. The first study found that a prenatal exposure to independent or combined suboptimal gestational factors (SGF : gestational diabetes mellitus, hypertensive disorders during pregnancy, maternal smoking during pregnancy) was positively associated with obesity and cardiometabolic risk factors in children around puberty, and these associations were sex-dependent. The second study found that children exposed to one SGF display a similar physiological response in terms of mechanical efficiency and substrate oxidation at rest and during exercise (submaximal and maximal) in comparison with non-exposed children. The third study of the thesis found that light-intensity physical activity reduced the adverse impact of prenatal exposure to combined SGF on high density lipoprotein-cholestrol levels in boys. A sleep duration within the range of the current Canadian recommendations for sleep did not mitigate the risk of high waist circumference (children) and obesity (girls) in a context of exposure to maternal smoking during pregnancy. Furthermore, other lifestyle factors such as moderate-to-vigorous physical activity and a healthy diet (intakes of: fruits and vegetables, grains, and dairy products), did not mitigate the adverse impact of a prenatal exposure to independent or combined SGF on children’s risk of obesity and cardiomeatbolic outcomes. Taken together, these results suggest that it is important to continue maternal SGF prevention and management programs to provide optimal health for children. Moreover, future studies should also develop and evaluate the impact of lifestyle habits interventions to design future prevention strategies during childhood and adolescence to reduce obesity and cardiometabolic risk factors in this population.
