Um total de 38 tucunarés e 16 matrinxãs foram coletados, os quais foram levados para o laboratório de Helmintologia do Departamento de Parasitologia, IB, Unicamp, onde foi feita a biometria, determinados os sexos dos peixes e analisados os órgãos a procura de ecto e endoparasitos. No tucunaré, cestódeos adultos da espécie Proteocephalus microscopicus (Prevalência = 76,3%, Intensidade de infecção = 280,9 vermes/peixe, Abundância média = 214,5) e Bothriocephalus cuspidatus (P= 5,3%, I=2 vermes/peixe, A=0,10) apresentaram-se parasitando o intestino. Nos olhos foram encontrados metacercárias do gênero Diplostomum sp. (P=28,9%, I=4,36 larvas/peixe, A=1,26). Monogênea do gênero Notozothecium (P= 47,4%, I=2,33 vermes/peixe, A=1,10)foi encontrado parasitando as brânquias e o nematódeo adulto Procamallanus (Spirocamallanus) inopinatus no intestino, (P = 5,3%, I= 1 verme/peixe, A=0,05). Um típico padrão de distribuição agregada foi observado em P. microscopicus, além disso observou-se correlação negativa entre intensidade de infecção e comprimento dos peixes, mostrando que peixes menores albergaram maior número deste cestódeo. Ainda para P. microscopicus, a intensidade de infecção e a transparência da água apresentaram correlação positiva. Correlação negativa entre a prevalência da infecção e pH da água foi observado para Notozothecium sp., indicando maior presença de monogêneas em águas mais ácidas. A presença deste gênero de monogênea em Cichla monoculus pode evidenciar especificidade ecológica, pois é um parasita habitual de Serrasalmidae. A espécie Bothriocephalus cuspidatus ainda não foi descrita parasitando Cichla monoculus, sendo a primeira ocorrência deste parasita em tucunaré. No matrinxã foram encontrados nematódeos adultos de Procamallanus (S.) inopinatus no intestino (P= 56,3%, I = 1,44 vermes/peixe, A= 0,81) comumente encontrado nesta espécie e monogêneas não identificadas nas brânquias / Abstract: Samples of tucunaré (Cichla monoculus) and matrinxã (Brycon amazonicus) were collected biweekly in two lakes of Rio das Pedras farm, Campinas - SP, from June 2005 to September 2006 with the objective of researching the helminthological fauna of the Amazonian species introduced at these ponds. A total of 38 tucunarés and 16 matrinxãs were collected, taken to the Laboratory of Helminthology, Parasitology Department, Biology Institute, UNICAMP, where we made the biometry and sexed the fish and analyzed the organs, searching for ecto and endoparasites. In the tucunaré, adult cestodes of the species Proteocephalus microscopicus (Prevalence = 76.3%, Intensity of infection = 280.9 worms/fish, Mean Abundance = 214.5) and Bothriocephalus cuspidatus (P= 5.3%, I = 2 worms/fish, MA= 0.10) were found in the intestine. In the eyes we have found larvae of Diplostomum sp. (P= 28.9%, I = 4.36 larvae/fish, MA= 1.26). Monogenean Notozothecium sp. (P= 47.4%, I = 2.33, worms/fish, MA= 1.10) was found parasitizing the gills and the adult nematode Procamallanus (Spirocamallanus) inopinatus (P = 5.3%, I = 1 worms/fish, MA= 0.05) in the intestine. A typical pattern of aggregate distribution were observed in P. microscopicus; the infection intensity and host length were found to be negative correlated, showing that smaller fish housed larger number of P. microscopicus. Between infection intensity and transparency of the water, there was a positive correlation. Negative correlation were found between prevalence of the infection and the water pH for Notozothecium sp, indicating higher presence of monogeneans in more acid water, in Cichla monoculus this parasite can evidences ecological specificity, because this parasite is usual in the Serrasalmidae family. The species Bothriocephalus cuspidatus is not commonly found in the Cichla monoculus, been this one its first appearance. In the matrinxã we have found adult nematodes of the species Procamallanus (Spirocamallanus) inopinatus (P = 56.3%, I = 1.44 worms/fish, MA= 0.81) in the intestine, a commom nematode for this specie and unidentified monogeneans in the gills / Mestrado / Mestre em Parasitologia

Caracterização da fauna parasitária do pseudalopex gymnocercus (Graxaim-do-campo) e do cerdocyon thous (graxaim-do-mato) na região do Rio Grande do Sul

Com o objetivo de estudar a fauna parasitária de Cerdocyon thous e Pseudalopex gymnocercus foram estimadas a prevalência, a intensidade média de infecção e abundância de helmintos gastrintestinais e artrópodes nesses hospedeiros. Foram capturados 40 canídeos silvestres na Região Sul do Estado do Rio Grande do Sul, nos municípios de Pedro Osório e Pelotas. Os animais foram capturados usando-se armadilha tipo Live Trap. Logo após a captura, os animais eram sedados, momento em que se coletava sangue e buscava-se coletar artrópodes ectoparasitos mediante exame minucioso. Em seguida os animais eram transportados para o Laboratório de Parasitologia do Departamento de Microbiologia e Parasitologia do Instituto de Biologia da UFPel, onde eram sacrificados usando-se pentobarbital sódico. Após a necropsia, os compartimentos do trato digestório, respiratório e renal foram isolados e examinados na busca de parasitos. O crânio dos animais foi usado para identificação taxonômica dos canídeos, dos quais 45% eram C. thous e 55% P. gymnocercus, podendo-se inferir que ambos canídeos são encontrados habitando a região fisiográfica desse trabalho. A totalidade dos animais (100%) apresentou-se parasitado. Amblyomma aureolatum é o carrapato com maior prevalência em P. gymnocercus (23,7%) e C. thous (61,11%) na região sul do Rio Grande do Sul. Nos nematódeos, os parasitos mais freqüentes foram: Ancylostoma caninum (C. thous 22,22%e P. gymnocercus 45,45%), Molineus felineus (C. thous 5,56% e P. gymnocercus 9,90%), Strongyloides spp. (C. thous 16,67% e P. gymnocercus 22,73%), Trichuris spp (C. thous 11,11% e P. gymnocercus 13,64%), e Capillaria hepática (C. thous 5,56% e P. gymnocercus 13,64%). Nos trematódeos, os parasitos encontrados foram: Alaria alata (C. thous 50,00% e P. gymnocercus 36,36%) e Athesmia heterolecithodes (C. thous 5,56%). Nos cestódeos, os parasitos encontrados foram: Spirometra spp (C. thous 61,11% e P. gymnocercus 54,55%) e Diphillobotriidae (C. thous 77,78% e P. gymnocercus 81,82%). Nos Acanthocephala os parasitos encontrados foram: Centrorhynchus sp. (C. thous 5,56%). Nos protozoários, os 7 parasitos encontrados foram: Babesia spp (P. gymnocercus 13,64%), diagnosticada por exame direto. / With the objective of studying parasitic fauna of Cerdocyon thous and Pseudalopex gymnocercus the prevalence, mean intensity of infection and abundance of gastrintestinal helminths and arthropods were observed in those hosts. 40 wild canids were captured in the South Area of the State of Rio Grande do Sul, in the counties of Pedro Osório and Pelotas. The animals were captured being used trap type “Live Trap”. Therefore after the capture, the animals were anesthesied and in that moment, the blood was collected and the presence or not of ectoparasites was observed. Therefore the animals were transported for the Laboratory of Parasitology, Department of Microbiology and Parasitology, Institute of Biology, UFPel, where they were sacrificed being used sodic pentobarbitol. After this, the portions of the intestinal and respiratory tract and kidney were isolated and examined in the search of parasites. The skull of the animals was used for the taxonomic identification of the hosts. C. thous (45%) and P. gymnocercus (55%) where observed. All hosts (100%) were parazited by some parasite type. Two species of Amblyomma were identified: A. aureolatum and A. tigrinum. Among the nematoda, the most frequent parasites were: Ancylostoma caninum (C. thous 22,22%e P. gymnocercus 45,45%), Molineus felineus (C. thous 5,56% e P. gymnocercus 9,90%), Strongyloides spp. (C. thous 16,67% e P. gymnocercus 22,73%), Trichuris spp (C. thous 11,11% and P. gymnocercus 13,64%), and Capillaria hepática (C. thous 5,56% and P. gymnocercus 13,64%). These characteristics give the meat a great potential gastronomic, so the commercial breeding boars may be considered a promising market. This study aimed to identify the species of helminths that inhabit the digestive and respiratory tract of Sus scrofa scrofa from commercial breeding. Were collected and individually the digestive and respiratory tracts of 40 wild boars from commercial breeding during the slaughter process in the slaughterhouse. Of the 40 animals analyzed, 87.5% were parasitized by helminths, they being, with their respective prevalence, Trichuris suis (67,5%), Ascaris suum (47,5%), Trichostrongylus colubriformes (45%), which has first recorded parasitizing wild boars, and Oesophagostomum dentatum (5.0%). With respect to the parasites of the respiratory tract, the genus Metastrongylus was recorded in the bronchi and bronchioles in 60% of animals tested, with the occurrence of three species with their respective prevalence, M. apri (52,5%), M. salmi (20%) and M. pudendotectus (7,5%), registering the highest prevalence of Metastrongylus in wild boars from commercial breeding of so far. With the exception of O. dentatum, the other parasites have their first record parasitizing wild boar in a subtropical region. / A carne do javali sempre foi considerada nobre, possuindo um sabor característico e apresentando baixos teores de gordura e colesterol. Tais características conferem a essa carne um grande potencial gastronômico, logo, a criação comercial de javalis pode ser considerada um mercado promissor. O presente estudo teve por objetivo identificar as espécies de helmintos que habitam o trato gastrointestinal e respiratório de Sus scrofa scrofa provenientes de criatório comercial. Foram coletados e individualizados os tratos gastrointestinais e respiratórios de 40 javalis oriundos de criatório comercial durante o processo de abate em frigorífico. Dos 40 animais analisados, 87,5% estavam parasitados por helmintos gastrointestinais, sendo eles com suas respectivas prevalências, Trichuris suis (67,5%), Ascaris suum (47,5%), Trichostrongylus colubriformes (45%), o qual é registrado pela primeira vez parasitando javalis e Oesophagostomum dentatum (5,0%). Com relação aos parasitos do trato respiratório, o gênero Metastrongylus foi registrado nos brônquios e bronquíolos de 60% dos animais analisados, com ocorrência de três espécies com suas respectivas prevalências, M. apri (52,5%), M. salmi (20%) e M. pudendotectus (7,5%), registrando-se a maior prevalência de Metastrongylus em javalis oriundos de criatório comercial até o momento. Com exceção de O. dentatum, os demais parasitos têm seu primeiro registro parasitando javalis em uma região de clima subtropical.

The species is not in danger of extinction, but it is appreciated by the illegal trade in wild animals. His parasite fauna is little known and has been recorded only one Nematoda and three Phthiraptera. The diet is mainly based greasses seeds, but can consume small fruit and insects. The study aimed to identify helminths and arthropods associated with red-crested cardinal and for that were examined wild birds, captives and of undetermined origin, totaling 40 specimes. The wild birds and undetermined origin were the most parasitized. The helminthfauna was composed of Aproctella carinii, Dispharynx nasuta, Capillaria sp. Diplotriaena sp. (Nematoda); Tanaisia oviaspera, Tanaisia valida, Tanaisia sp., Prosthogonimus ovatus (Trematoda); Orthoskrjabinia sp. (Cestoda); Mediorhynchus sp. (Acanthocephala). Tanaisia sp. was most prevalent (10%) and more abundant (0.48), D. nasuta and Orthoskrjabinia sp. occurred with greater mean intensity of parasitism (5 helminths/host). The arthropodfauna was represented by Myrsidea coronatae, Philopterus sp. and Brueelia sp. (Phthiraptera); Ptilonyssus sairae and Sternostoma pirangae (Gamasida), where M. coronatae and P. sairae were the most prevalent with 65% and 50%, respectively. All helminths are reported for the first time in P. coronata, Orthoskrjabinia sp. is recorded for the first time in Brazil; A. carinii, Diplotriaena sp., T. valida, T. oviaspera and Mediorhynchus sp. are cited for the first time in the Rio Grande do Sul state. The helminthfauna found in P. coronata, indicates that the species is omnivorous eating habits. Among the arthropods, Philopterus sp., Brueelia sp., P. sairae and S. pirangae have your first record infecting P. coronata; M. coronatae is recorded for the first time in Brazil, this report marks the first occurrence of S. pirangae in Brazil and the first of P. sairae in the Rio Grande do Sul state. / Paroaria coronata (cardeal) ocorre apenas na América do Sul. A espécie não está em perigo de extinção, mas é visada pelo comércio ilegal de animais silvestres. Sua fauna parasitária é pouco conhecida, tendo sido registrado somente um Nematoda e três Phthiraptera. A dieta desta ave baseia-se principalmente em sementes de gramíneas, mas pode consumir pequenos frutos e insetos. O estudo teve por objetivo identificar os helmintos e artrópodes associados à P. coronata e para tal foram examinadas aves silvestres, cativas e de origem indeterminada, totalizando 40 espécimes. As aves silvestres e de origem indeterminada foram as mais parasitadas. A helmintofauna esteve composta por Aproctella carinii, Dispharynx nasuta, Capillaria sp. Diplotriaena sp. (Nematoda); Tanaisia oviaspera, Tanaisia valida, Tanaisia sp., Prosthogonimus ovatus (Trematoda); Orthoskrjabinia sp. (Cestoda); Mediorhynchus sp. (Acanthocephala). They are fed of herbaceous vegetables, they live in tunnels of grassy, being able many times to inhabit agricultural peridomiciliar regions, close to highways of the south region it Brazil, being many adopted times as animal of esteem. With the objective to know the diversity of parasites of this rodent, were examined 33 specimens of Cavia aperea that after abated according to Protective Association of the Animals, they had been brushed and washed for collection of ectoparasites, being after that necropsied and its organs examined in search of endoparasites. The ectoparasites after removed from the animal, were clarified in acetate of metila and mounted in slides with Canada balsam. For collection of the helminths it was examined and one washed separately each organ in sieve, nematodes had been clarified in lactofenol and the members of the Classes Trematoda and Cestoda were prepared according Amato s technique diaphaned in creosote and mounted in slides with Canada balsam. Of the total of examined animals, 100% were infested by lice of the species: Trimenopon hispidum, Gyropus ovalis, Gliricola lindolphoi and G. porcelli, with exception of this, all others are registered for the first time in the State Rio Grande do Sul. Of Siphonaptera, Polygenis (Neopolygenis) atopus it was registered prevalence of 6,1%. The helminthes were: of the Phylum Nematoda - Trichostrongylus sp., Paraspidodera uncinata, Vianella travassosi, Ackertia burgosi and Graphidioides subterraneus; Class Trematoda - Pseudoquinqueserialis caviae, Pseudohippocrepis suttonae n.g. n.sp., Taxorchis caviae and the Cestoda Monoecocestus parcitesticulatus, all cited for the first time in the state Rio Grande do Sul, with exception of Paraspidodera uncinata. / Cavia aperea, ou popularmente preá, é um roedor de hábito diurno, principalmente nas primeiras horas da manhã e nas ultimas da tarde. Alimentam-se de vegetais herbáceos, vivem em túneis de gramíneas, podendo muitas vezes habitar regiões peridomiciliares rurais, perto de rodovias da região sul do Brasil, sendo muitas vezes adotados como animais de estimação. Com o objetivo de conhecer a diversidade de parasitos deste roedor, examinou-se 33 exemplares de Cavia aperea que após eutanásiados segundo a Associação Protetora dos Animais, foram escovados e lavados para coleta de ectoparasitos, sendo em seguida necropsiado e seus órgãos examinados em busca de endoparasitos. Os ectoparasitos após removidos do animal foram clarificados em acetato de metila e montados em lâminas com bálsamo do Canadá. Para coleta dos helmintos examinou-se e lavou-se cada órgão em separado em tamis, os nematóides foram clarificados em lactofenol e os membros das classes Trematoda e Cestoda foram preparados segundo técnica de Amato e em seguida diafanizados em creosoto de Faia e montados em lâminas com bálsamo do Canadá. Dos dos animais examinados, 100% estavam infestados por piolhos das espécies: Trimenopon hispidum, Gyropus ovalis, Gliricola lindolphoi e G. porcelli, e com exceção deste último, todos os outros são registrados pela primeira vez no Rio Grande do Sul. De Siphonaptera encontrou-se Polygenis (Neopolygenis) atopus com prevalência de 6,1%. Os helmintos encontrados foram: do Filo Nematoda Trichostrongylus sp., Paraspidodera uncinata, Vianella travassosi, Ackertia burgosi e Graphidioides subterraneus; Classe Trematoda Pseudoquinqueserialis caviae, Pseudohippocrepis suttonae n.g. n.sp. e Taxorchis caviae e a Classe Cestoda - Monoecocestus parcitesticulatus, todos citados pela primeira vez no Rio Grande do Sul, com exceção de Paraspidodera uncinata.

Alternativas farmacológicas para a resistência parasitária múltipla em rebanhos de ruminantes: uma nova abordagem / Pharmacological alternatives for multiple parasitic resistance in ruminant livestock: a new approach

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Infections by gastrointestinal nematodes are among the main diseases of the ruminants around the world. These infections cause - in their clinical or subclinical courses a decrease of the productivity of the herds, thus, the economic viability of the ruminant farms is directly related to an adequate control of these parasites. Although the benefits of the use of several practices of alternative control of parasites are proven and, despite of that these practices have been improved through the years, in general, the use of efficacious antiparasitic drugs is imperative to control satisfactorily the helminths of the ruminants. However, the development of parasite resistance to these drugs has become common and, in some cases, parasitic populations are multiple resistant to several classes of anthelmintics and endectocides, and it makes very difficult to control the helminthosis adequately. In this study are shown some experiments conducted with the aim of to find ways to restore the efficacy of the treatments, for recovering the usefulness of active ingredients for which the gastrointestinal nematodes of ruminants became resistant. Efficacy tests of the drugs were made using Fecal Egg Count Reduction Tests (FECRT) and larvae cultures, for the identification of the genus of nematodes existing, before and after the treatments. We evaluated the anthelmintic action of high concentrated (long acting) drugs, overdoses (high dosages), commercial combinations of drugs and combinations of chemicals, formulated with basis in previous efficacy tests for each active ingredient alone. These combinations, done from drugs sold separately, gave us the best results. Based in these observations, we are proposing a new approach regarding helminth control in multi-resistant populations, by using combinations of drugs with different mechanisms of action on the parasites, and taking advantage of the efficacy tests, performed before the choose of drugs, and then, to prove the results obtained with the combinations done particularly for each herd. / As infecções por nematódeos do trato gastrintestinal estão entre as principais enfermidades de ruminantes ao redor do mundo. Essas infecções, clínicas ou subclínicas, impactam negativamente a produtividade dos rebanhos, e a viabilidade econômica dos mesmos está diretamente relacionada a um controle adequado desses parasitas. Embora estejam comprovados os benefícios de utilizarem-se diversas práticas de controle alternativo de parasitas, as quais vêm sendo aprimoradas ao longo dos anos, em geral, a utilização de fármacos antiparasitários eficazes sobre os helmintos que infectam os ruminantes ainda é essencial para que se obtenham resultados satisfatórios. No entanto, o desenvolvimento de resistência parasitária a tais fármacos tem se tornado cada vez mais comum e, em alguns casos, as populações de parasitas já apresentam resistência múltipla a várias classes de anti-helmínticos e endectocidas, comprometendo o adequado controle das helmintoses. Neste estudo apresentam-se experimentos que foram conduzidos com o objetivo de encontrarem-se meios de restabelecer a eficácia dos tratamentos, recuperando-se a utilidade de princípios ativos para os quais os nematódeos gastrintestinais de rebanhos de ruminantes já tenham desenvolvido resistência. Foram utilizados testes de eficácia com base na redução da contagem de ovos nas fezes dos animais após os tratamentos (FECRT), e em culturas de larvas para a identificação dos gêneros de nematódeos presentes nessas amostras. Avaliaram-se os efeitos anti-helmínticos de fármacos de alta concentração (longa ação), de superdosagens, de combinações comerciais e de combinações de drogas elaboradas com base em estudos prévios de eficácia das mesmas (isoladamente), utilizando-se para isso, fármacos comercializados separadamente. Os melhores resultados foram obtidos, justamente, com estas últimas. Propõe-se, a partir destas constatações, uma nova abordagem em relação ao controle antiparasitário em populações de helmintos multi-resistentes, utilizando-se a combinação de diferentes fármacos (com princípios ativos e mecanismos de ação distintos), e valendo-se de testes de eficácia para a escolha dos fármacos e para a comprovação dos resultados obtidos com suas combinações especificamente em cada rebanho.

In der vorliegenden Arbeit wurden 212 Kotproben von Chamäleons auf Parasitenstadien und 75 Tierkörper pathologisch sowie bei einem nachgewiesenen Parasitenbefall histopathologisch untersucht. Ziel war es, anhand dieser Untersuchungen das Vorkommen und die Schadwirkungen von Endoparasiten unter Berücksichtigung der Herkunft, des Alters, des Geschlechts und der Chamäleonart zu beschreiben. Von 212 Kotproben wiesen 55,2% Endoparasitenstadien auf. Bei 54,7% der 64 sezierten und auswertbaren Tiere wurden Endoparasiten nachgewiesen. Der Anteil positiver Proben zeigt zwischen Nachzuchten (55,5%) und Wildfängen (54,1%) keinen wesentlichen Unterschied. In Wildfängen konnten häufiger Endoparasiten mit einem indirekten Lebenszyklus ermittelt werden, Nachzuchten beherbergten dagegen öfter Parasiten mit einem direkten Entwicklungszyklus. In den untersuchten Chamäleons konnten regelmäßig Kokzidien der Gattung Isospora und Oxyuriden nachgewiesen werden. Zestoden konnten in der koproskopischen Untersuchung gar nicht aufgefunden werden, wohingegen sie in der pathologischen Untersuchung sporadisch im Darm diagnostiziert werden konnten. Die Häufigkeit des koproskopischen Nachweises von Parasitenstadien bezogen auf die Gesamtzahl der untersuchten Chamäleons stellte sich wie folgt dar: in 30,4% wurden Protozoon gefunden, 21,7% der Tiere waren mit Kokzidien infiziert (davon 78,3% Isospora spp., 13,0% Choleoeimeria spp., 6,5% Eimeria spp., 2,2% Mischinfektion Isospora spp./Choleoeimeria spp.) und 8,5% mit Flagellaten oder Ziliaten. Bei 83,3% der Tiere mit gastrointestinalen Symptomen konnte ein Befall mit Kokzidien der Gattung Isospora nachgewiesen werden. In 38,7% der koproskopischen Untersuchungen konnten Nematoden (65,9% Oxyuriden, 19,5% Askariden/ Heterakiden, 1,4% Rhabdias sp., 2,8% Strongyloides sp., je 0,5 % Spirurida, Heterakiden/Filarien, Oxyuriden/Strongyloiden) und in 2,8% Trematoden (Digenea) aufgefunden werden. Anamnestisch konnten in 35,8% aller Tiere klinische Symptome beobachtet werden, wovon bei 88,2% der erkrankten Tiere ein Endoparasitenbefall nachzuweisen war. Insgesamt 64,1% der sezierten Chamäleons waren mit Endoparasiten befallen, wovon 68,3% Mono- und 31,7% Mischinfektionen beherbergten. In 31,3% der sezierten Chamäleons wurden Nematoden gefunden und der Befall wurde in 55,0% dieser Fälle als hochgradig eingestuft. Es wurden Befallsraten von 25,0% für Strongyloides sp., 23,4% für Askariden/ Heterakiden,15,0% für Filarien, 5,0% für Rhabdias sp., 9,4% für Zestoden, 10,9% für Digenea registriert. In 11,3% der Fälle lagen Mischinfektionen vor. Damit ist ein Endoparasitenbefall bei Chamäleons häufig und kann zu Erkrankungen führen. Die Exposition unterscheidet sich bei Wildfängen und Nachzuchten aufgrund der unterschiedlichen Umgebungsbedingungen. Auch klinisch unauffällige Tiere waren zu 27,8% mit Parasiten befallen, so dass eine klinische Symptomatik nicht zwingend aus einem Parasitenbefall resultiert. Insgesamt betrachtet verdient der Endoparasitenbefall von Chamäleons das Augenmerk von Tierärzten und Tierhaltern und sollte bei augenscheinlich hohem Infektionsdruck zu Gegenmaßnahmen, insbesondere auch einer verbesserten Hygiene, Anlaß geben. / In the present study 212 chameleon fecal samples were examined for parasite stages and 75 carcasses were examined histopathologically and pathologically in a proven case of a parasite infestation. The basis of this study was to describe the occurrence and harmful effects of internal parasites considering the origin, age and sex of the chameleons. Of the 212 fecal samples 55.2% showed stages of endoparasites. Parasites were detected at 54.7% of 64 evaluated and dissected animals. The proportion of positive samples shows no significant difference between offspring (55.5%) and wild specimens (54.1%). In wild specimens common internal parasites could be determined with an indirect life cycle, however offspring harbored more parasites with a direct life cycle. In the studied chameleons coccidia as the genus Isospora and Oxyurids were regularly detected. In the coprological study Cestodes could not be found, while in the pathological examination they could be diagnosed sporadically in the intestine. Based on the total number of investigated chameleons the frequency of detection of parasite stages are presented as follows: Protozoa were found in 30.4%, 21.7% of the animals were infected with coccidia (of which 78,3% Isospora spp, 13,0% Choleoeimeria spp., 6.5% Eimeria spp., 2.2% polyinfections between Isospora spp./ Choleoeimeria spp.) and 8.5% with flagellates or ciliates. At 83.3% of the animals with gastrointestinal symptoms coccidia of the genus Isospora were detected. In 38.7% of the fecal examination nematodes were determined (65.9% Oxyurids, 19.5% Ascarids/ Heterakis, 1.4% Rhabdias sp., 2.8% Strongyloides sp., 0.5% Spirurida, Heterakids/ Filariae, Oxyurids/ Strongyloides sp.) and Trematodes in 2.8% (Digenea) were found. The anamnesis showed that clinical symptoms could be observed in 35.8% of all of the animals, whereas endoparasite infestation could be detected inn 88.2% of the affected animals. Overall, 64.1% of the dissected chameleons were infested with parasites, of which 68.3% harbored mono- and 31.7% polyinfections. In 31.3% of the dissected chameleons nematode infestations were found and 55.0% of these cases were classified as severe. Prevalences were registered: 25.0% for Strongyloides spp., 23.4% for Ascarids/ Heterakids, 15.0% for Filaria, 5.0% for Rhabdias sp., 9.4% for Cestodes, 10.9% for Digenea. In 11.3% of the cases mixed infections were reported. Thus, endoparasite infestation is common among chameleons and can lead to diseases. Exposure differs from wild-specimens and captive-bred due to the different environmental conditions. Also, 27.8% of clinically healthy animals were also infested with parasites, which means that clinical symptoms are not necessarily the result of a parasitic infestation. Overall, chameleon endoparasites deserve the attention of veterinarians and pet owners and should be treated promptly when there is a high likelihood of infection or hygiene is of concern.

Reducing the ‘Neglect’ in Neglected Tropical Diseases: A Review of the Debate surrounding the Effectiveness of Mass Deworming – A Case Study of Kenya –

Neglected tropical diseases are parasitic and bacterial diseases mainly prevalent in developing countries affecting people living in poverty. The World Health Organization’s human rights-based approach emphasizes the “prevention, control, elimination and eradication of neglected tropical diseases” through the use of preventative chemotherapy, such as the mass administration of deworming drugs to improve people’s health.This research paper will take a deeper look at how WHO has been communicating NTDs to make them less ‘neglected’ and how the NTD discourse has been shaping development organizations’ action. In addition, it aims to investigate how successful mass deworming has really been in terms of the recent debate.This study is using a combination of a discourse analysis and qualitative interviews in order to investigate how the NTD discourse and recent initiatives by international organizations have contributed to making NTDs less neglected. It deconstructs representations of the ‘Other’ – the superiority of the ‘West’ over the ‘Rest’ – in relation to the NTD discourse and its inherent power structures. Discourses are analyzed to identify power relations between governments, development organizations, pharmaceutical industries, and recipients of deworming drugs as part of Kenya’s 2013 deworming campaign.The results showed that the NTD discourse has helped raise awareness for NTDs. NTDs and their debilitating effect on populations have been better and more widely communicated, making them less ‘neglected’. WHO and other development organizations’ actions have contributed to making NTDs more visible and have given NTDs higher priority on the global health agenda. Findings from this research study revealed that the ongoing debate has not had a negative impact on international funding. More research and development of a vaccine against NTDs is needed to find more ways to tackle these devastating diseases.

Impact of helminths and helminth products on immune responses

Helmintheninfektionen induzieren in ihren Wirten Typ 2 (Th2) Immunantworten, welche parasitäre Larvenstadien hemmen und/oder zur Abstoßung von intestinalen Würmern führen. Th2-Antworten können auch gegenüber Umweltallergenen ausgebildet werden und allergische Reaktionen vermitteln. Mastzellen (MZ) spielen eine herausragende Rolle für die Wirtsantwort gegen Parasiten. Die Ergebnisse der vorliegenden Arbeit zeigen, dass eine Infektion von MZ-defizienten Mäusen mit Helminthen zu einer reduzierten Produktion von IL-25, IL-33 und TSLP, einer beeinträchtigten Etablierung von Th2 Immunantworten und einer erhöhten Wurmlast führt. Diese Parameter konnten allerdings nach einem erfolgreichen Transfer von Knochenmark aus Wildtypmäusen wiederhergestellt werden. Die vorliegende Arbeit beschreibt somit eine wichtige Funktion von Mastzellen für die Initialisierung von Th2 Immunantworten. Des Weiteren konnte gezeigt werden, dass ein Cystein-Protease-Inhibitor - AvCystatin sowohl Parameter von Atemwegsentzündung und -hyperreaktivität als auch Lieschgraspollen (Phl p 5b)-spezifische Immunantworten in einem klinisch relevanten Mausmodell für Atemwegsentzündung inhibiert. Gleichzeitig führte die Behandlung mit AvCystatin zu einem Anstieg von IL-10 und erhöhten Frequenzen von CD4+CD25+Foxp3+ T Zellen. Die in vivo Effekte von AvCystatin wurden unabhängig von der Protease-inhibitorischen Aktivität des Immunmodulators oder dessen Fähigkeit zur Oligomerisation vermittelt. AvCystatin unterdrückte zudem die Allergen-spezifische Produktion von IL-13 und induzierte in vitro unter Verwendung mononukleärer Zellen aus dem peripheren Blut (PBMCs) von Graspollen allergischen Patienten einen IFN-gamma vermittelten Th1 shift. Zusammenfassend tragen die Ergebnisse der Arbeit zu einem besseren Verständnis für die frühen Ereignisse von Th2 Immunantworten bei und zeigen, dass Helminthenmoleküle Bystandereffekte auf Immunantworten vermitteln können, die gegen andere Antigene gerichtet sind. / Helminth infections induce protective type 2 (Th2) immune responses in the host leading to arrested larval development and/or intestinal worm expulsion. Moreover, Th2 immune responses are initiated against harmless environmental allergens and mediate development of allergic disease. Among multiple mechanisms implicated in host responses to parasites and allergens, mast cells (MCs) play a pivotal role. The present study shows that MC-deficient mouse strains following infection with two gastrointestinal helminths had dramatically reduced early production of the tissue-derived cytokines IL-25, IL-33, and TSLP, which resulted in impaired induction of Th2 immune responses as well as increased parasite burden. These parameters were restored after transfer of WT bone marrow. These data reveal an important role for MCs in orchestrating type 2 immune responses. Parasites have developed various strategies to modulate the immune system via induction of a range of regulatory mechanisms. In this study AvCystatin, the filarial cysteine inhibitor, was found to inhibit airway inflammation and hyperreactivity induced by a clinically relevant allergen of timothy grass pollen (Phl p 5b). AvCystatin increased levels of the regulatory cytokine IL-10 and total numbers of CD4+CD25+Foxp3+ T cells. The immunomodulatory effect in vivo was found to be independent of AvCystatin’s protease inhibitor activity or oligomerization. Finally, AvCystatin suppressed allergen-specific production of IL-13 and created a shift towards Th1 immunity by increased levels IFN-gamma of human peripheral blood mononuclear cells (PBMCs) from grass pollen allergic patients. The findings contribute to a better understanding of the early events that dictate the priming of type 2 immune responses. Furthermore, helminth product-induced suppression may also have effects on bystander responses to unrelated antigens, thus, suggesting a promising preventive and therapeutic concept in the treatment of aberrant inflammations.

Introdução - A prática de reúso pela aplicação de esgotos tratados na agricultura atualmente é considerada parte integrante dos recursos hídricos por muitas nações. No entanto, oferece riscos à saúde da população pela presença de bactérias, cistos de protozoários, ovos de helmintos e vírus, capazes de sobreviver por longos períodos em ambientes desfavoráveis e apresentarem um curto tempo entre a infecção e o desenvolvimento da doença. Objetivo - Estabelecer medidas preventivas para o lançamento de efluentes nos ecossistemas aquáticos, controle sanitário na redução de patógenos transmitidos por águas contaminadas e a disposição de efluentes tratados em solos tropicais como ferramentas de auxílio do manejo integrado dos recursos hídricos em bacias hidrográficas, por meio do uso de metodologia de gerenciamento de risco com aplicação do Sistema APPCC, em um conjunto de tecnologias na produção de efluente tratado de esgoto de origem doméstica para avaliar o potencial de emprego em solos agrícolas. Métodos - A pesquisa foi desenvolvida na estação de tratamento de esgoto e no campo experimental para pesquisas de utilização de efluente na agricultura no município de Lins. Os indicadores biológicos estudados e os pontos de amostragem foram determinados através da utilização da metodologia de controle de qualidade Análise de Perigos e Pontos Críticos de Controle (APPCC). Foram coletadas amostras do solo irrigado e dos poços de monitoramento do lençol freático. As amostras foram submetidas a análises físico-químicas, microbiológicas e parasitológicas. Aos resultados obtidos foi aplicada a Avaliação Quantitativa do Risco Microbiológico, para a determinação do risco anual de infecção. Resultados - O efluente final apresentou concentrações médias de 8,13x105 NMP/100 mL de coliformes totais e 4,69x105 NMP/100 mL de Escherichia coli, não sendo observados ovos de helmintos. O solo apresentou maior concentração de coliformes totais e E. coli na camada superficial 1,01x106 e 8,70x103, diminuindo ao longo da profundidade. Os ovos de helmintos foram encontrados entre a superfície e os 15 cm de profundidade em concentrações entre 0,07 e 0,87 ovos/g de peso seco. Foi constatada a presença de adenovírus em concentração de 3,56x105 cópias genômicas/g entre os 5 e 10 cm de profundidade em 2015. Nos poços de monitoramento foram constatadas concentrações de coliformes totais variando entre < 1,00 e 1,01x103 NMP/100 mL e E. coli entre <1,00 e 1,00x100 NMP/100 mL. Não foi observada a presença de vírus entéricos. O risco anual de infecção estimado para E. coli no efluente final foi 1,02x10-2 pppa, no solo variou entre 2,68x10-3 e 7,59x10-3 pppa e nos poços de monitoramento variou entre 1,42x10-8e 4,38x10-9 pppa. Para os ovos de helmintos observados no efluente do tratamento primário o risco anual de infecção calculado foi 4,04x10-2 pppa e no solo variou entre 9,57x10-1 e 9,76x10-1 pppa. Conclusões No período da pesquisa, as amostras dos sistemas de tratamento de esgoto e de irrigação demonstraram atender as expectativas de remoção de patógenos condizentes aos seus parâmetros de projeto, havendo restrições para seu lançamento em corpos hídricos e aplicação na agricultura. O solo mostrou capacidade de retenção dos organismos patogênicos nos primeiros 60 cm de profundidade, reduzindo as suas concentrações a níveis inferiores a 1,00 NMP/100 mL nos aquíferos. / Introduction - Currently, the practice of reuse through the application of treated sewage in agriculture considers an integral part of water resources for many nations. However, threatens the health of the population by the presence of bacteria, protozoan cysts, helminth eggs and viruses able to survive for long periods in harsh environments and present a short time between infection and development of disease. Objective - Establish preventive measures for the release of effluents on aquatic ecosystems, sanitary control in the reduction of pathogens transmitted by contaminated water and the disposal of treated effluent in tropical soils as aid tools in integrated management of water resources in river basins. For this purpose it was employed as risk management methodology the Hazard Analysis and Critical Control Points system (HACCP) in a set of technologies in the treated effluent production of domestic sewage to assess the employment potential in agricultural soils. Methods - The study was conducted in the sewage treatment plant and in an adjoining experimental field situated at Lins city. The biological indicators and the sampling points were determined by using the quality control methodology Hazard Analysis and Critical Control Points (HACCP). Samples of irrigated soil and groundwater monitoring wells were collected. The samples were subjected to analysis, physical-chemical, microbiological and parasitological. The results obtained was applied to Quantitative Microbial Risk Assessment (QRMA), for determining the annual risk of infection. Results - The final effluent presented average concentrations of 8.13x105 MPN / 100 mL of total coliforms and 4.69x105 MPN / 100 mL of Escherichia coli, not being observed helminth eggs. The soil showed the highest concentration of total coliforms and E. coli in the surface layer and 1.01x106 8.70x103, decreasing along the depth. Eggs from helminths were found between the surface and 15 cm depth in concentrations from 0.07 to 0.87 eggs / g dry weight. It was found the presence of adenovirus in concentration 3.56x105 genomic copies / g between 5 and 10 cm depth in 2015. In monitoring wells total coliform concentrations were found ranging from <1.00 and 1.01x103 NMP / 100 mL and E. coli between <1.00 and 1.00x100 MPN / 100 mL. It was not observed the presence of enteric viruses. The estimated annual risk of infection to E. coli in the final effluent was 1.02x10-2 pppy, soil ranged from 2.68x10-3 and 7.59x10-3 pppy and monitoring wells ranged from 1.42x10-8 e 4.38x10-9 pppy. For helminth eggs observed in the effluent from the primary treatment, the annual risk of infection was calculated 4.04x10-2 pppy and soil ranged from 9.57x10-1 and 9.76x10-1 pppy. Conclusions - During the survey period, samples of sewage and irrigation treatment systems showed meet the expectation removal of pathogens consistent to its design parameters, with restrictions on its release in water and applied in agriculture bodies. The soil of pathogenic organisms showed retention capacity in the first 60 cm depth by reducing its concentration to levels below 1.00 MPN / 100 mL in aquifers.
