Efeito da laserfototerapia na reparação de osso alveolar humano: análise microtomográfica e histomorfométrica / Effect of laserphototherapy on human alveolar bone repair: microtomographic and histomorphometrical analysis

A instalação imediata de implante dental na região de molares é crítica, devido a grande perda óssea e da discrepância entre a espessura da crista alveolar e a plataforma do implante. Laserfototerapia (LFT) auxilia na reparação óssea, portanto pode acelerar a instalação do implante. Vinte pacientes foram selecionados para o estudo. Dez pacientes foram submetidos à LFT com laser de diodo de GaAlAs (808nm) no transcirúrgico de exodontia de molares, imediatamente após, e em 24h, 48h, 72h, 96h, 7 e 15 dias. As irradiações foram aplicadas no modo contínuo, em contato e pontual (100mW, 0,04cm2, 075,0J/cm2, 30s por ponto, 3J por ponto, em 5 pontos). O grupo controle (n=10) recebeu o mesmo tratamento, no entanto, o equipamento estava desligado. Quarenta dias depois, espécimes do tecido formado no interior dos alvéolos foram coletados para posterior análise por microtomografia (microCT) e histomorfometria. Dados de ambos os grupos foram comparados pelo test t de Student, enquanto aqueles das diferentes avaliações microtomográficas foram comparados pelo teste de correlação de Pearson (p<0.05). O volume relativo do osso, bem como a área relativa do osso foram significativamente maiores (p<0.001) no grupo laser do que no controle. No grupo controle houve correlação negativa significativa entre número e espessura de trabéculas, e entre número e separação entre trabéculas (p<0.01); entre espessura e separação das trabéculas a correlação foi positiva (p<0.01). O grupo laser mostrou correlação negativa significante entre número e espessura de trabéculas (p<0.01). A Laserfototerapia acelerou o reparo ósseo. Baseado na correlação de Pearson foi possível inferir que o grupo laser apresentou uma distribuição trabecular mais homogênea, com trabéculas menos espessas e mais numerosas, o que pode propiciar a redução do tempo para a instalação do implante. / The immediate dental implant placement in the molars region is critical, because of the high amount of bone loss and the discrepancy between the alveolar crest thickness and the dental implant platform. Laserphototherapy (LPT) improves bone repair thus could accelerate the implant placement. Twenty patients were selected for the study. Ten patients were submitted to LPT with GaAlAs diode laser (808nm) during molar extraction, immediately after, 24h, 48h, 72h, 96h, 7 and 15 days. The irradiations were applied in continuous wave, in contact and punctual mode (100mW, 0.04cm2, 075,0J/cm2, 30s per point, 3J per point, in 5 points). The control group (n=10) received the same treatment; however with the power of the laser off. Forty days later samples of the tissue formed inside the sockets were obtained for further microtomography (microCT) and histomorphometry analyses. Data of both groups were compared by the Student t test, whereas those from the different microCT parameters were compared by the Pearson correlation test (p<0.05). The relative bone volume, as well as the relative area were significantly higher (p<0.001) in the lased than the control group. In the control group there were negative correlations between number and thickness, and between number and separation of trabecula (p<0.01); between thickness and separation of trabecula the correlation was positive (p<0.01). The laser group showed significant negative correlation between the number and the thickness of trabecula (p<0.01). Laserphototherapy accelerated bone repair. By the Pearson correlation test it was possible to infer that the lased group presented a more homogeneous trabecula configuration, with thin and numerous trabecula, which would facilite the reduction of time for the installation of the implant.

A fim de cumprir os objetivos propostos, a presente pesquisa subdivide-se em três capítulos, os quais se concentram, por sua vez, em três escritos: o primeiro investiga a teoria inicial da religião nas Reden (1799); os dois últimos se ocupam do pensamento tardio, a saber: a construção filosófico-científica na Vorlesungen über die Dialektik (1811/31) e o desenvolvimento teológico do conceito de piedade na Introdução da Glaubenslehre (1830/31). / The following research aims at investigating the principles of the Dialectics and the Dogmatics of the German thinker Friedrich D. E. Schleiermacher (1768-1834). Conceived in its widest perspective, Schleiermacher’s work involves the question regarding the relationship between philosophy and theology as well as their proper place within his system of sciences. Insofar as Schleiermacher’s theory of feeling constitutes an analysis of the ultimate ground of the immediate self-consciousness’ structure, by means of which both the unity of the subjectivity and the general principles of knowing are articulated, our study defends the theory of feeling as a possible point of inflection in the relationship between philosophy and theology. In order to achieve the intentions proposed here we will be subdividing this research in three chapters, each one of which is based on three different works by Schleiermacher. The first one analyses the early theory of religion as developed by Schleiermacher in his Reden (1799). The two following chapters are focused on his mature thought, namely, his Vorlesungen über die Dialektik (1811/31), where he develops a philosophical system of sciences as such, and his Introduction to the Glaubenslehre, where the theological development of the concept of piety takes place.

Evaluation of the effect of stabilization time in eventually consistent systems

The effect of using the eventual consistency model is evaluated, compared to the effect of immediate consistency, by increasing the stabilization time in both models and using the immediate consistent system as a baseline for evaluations. The immediate consistent system performs better if the information and the decisions are replicated adequately fast throughout the system. When the stabilization time increases the effectiveness of eventual consistency emerges, which is most obvious when time constraints make it difficult to propagate information and decisions. By using a simulation to extract data for evaluations, it is verified in this research that as the stabilization time between different parts of a system increases, the eventual consistency will always outperform the immediate consistent system. This statement is valid in all situations where consistency models are useful. Of secondary importance in the research, by adding more decision layers to the eventual consistency model, the performance output is increased significantly, as swift and less well calculated decisions can be thwarted and corrected using the second decision layer.

EGR3 Immediate Early Gene and the Brain-Derived Neurotrophic Factor in Bipolar Disorder

Bipolar disorder (BD) is a severe psychiatric illness with a consistent genetic influence, involving complex interactions between numerous genes and environmental factors. Immediate early genes (IEGs) are activated in the brain in response to environmental stimuli, such as stress. The potential to translate environmental stimuli into long-term changes in brain has led to increased interest in a potential role for these genes influencing risk for psychiatric disorders. Our recent finding using network-based approach has shown that the regulatory unit of early growth response gene 3 (EGR3) of IEGs family was robustly repressed in postmortem prefrontal cortex of BD patients. As a central transcription factor, EGR3 regulates an array of target genes that mediate critical neurobiological processes such as synaptic plasticity, memory and cognition. Considering that EGR3 expression is induced by brain-derived neurotrophic factor (BDNF) that has been consistently related to BD pathophysiology, we suggest a link between BDNF and EGR3 and their potential role in BD. A growing body of data from our group and others has shown that peripheral BDNF levels are reduced during mood episodes and also with illness progression. In this same vein, BDNF has been proposed as an important growth factor in the impaired cellular resilience related to BD. Taken together with the fact that EGR3 regulates the expression of the neurotrophin receptor p75NTR and may also indirectly induce BDNF expression, here we propose a feed-forward gene regulatory network involving EGR3 and BDNF and its potential role in BD.

Le post-partum immédiat recèle les fondamentaux de la situation anthropologique, tant pour le bébé que pour la mère et le père et pour le socius. La première matrice de la vie psychique subordonnée à « l’espace dyadique fondamental » procède de la complexe alchimie entre la tessiture de l’investissement maternel et le tempérament et les capacités de régulation du bébé, sans négliger la structure en abyme qui encadre le maternage. A partir d’une clinique étendue dans le champ de la périnatalité cette recherche s’intéresse avec une double méthodologie qualitative clinique (Chapitre 1) et quantitative « outillée » (Chapitre 2 et 3), à la construction d’un espace dyadique, indispensable à la croissance du bébé et à l’investissement maternel, aux vicissitudes de cette construction, à la fonction régulatrice des rituels (relayés par les dispositifs de soins périnataux actuels) qui offrent une scansion au travail de l’enfantement. La communication dyadique entre mère est bébé, dont nous repérons le point d’orgue précoce vers deux mois avec les premières protoconversations, est étudié comme une co-génése transmodale et asymétrique complexe, tant par la psychanalyse que par la théorie de l’attachement, l’approche développementale et les neurosciences. Cet espace unique et original, renouvelé à chaque nouvelle naissance se trame à partir des « les formants de l’investissement maternel », constituée par les vecteurs de la vie psychique et leur combinatoire qui fournissent l’économie nécessaire à la situation maternante. A partir des trois grands courants de pensée conceptuelle et de traitement thérapeutique des troubles du lien précoce nous recensons le formant « en transformation » celui de « la transmission » le formant « en séduction ». Ces avancées théorico-cliniques ont un triple objectif, construire une lecture nouvelle de l’intersubjectivité primaire, de l’édification de la conscience de soi chez le bébé, des mécanisme normaux et pathologiques des identifications et de l’empathie, soutenir le principe des soins, l’édification d’une sémiologie dyadique de la psychopathologie précoce et ouvrir sur un modèle incluant la dimension du précoce et de ses formes de symbolisation dans les cures d’adultes et la prise en charge institutionnelle des populations précaires ou limites. / The immediate postpartum period contains the basics of the anthropological situation, for the baby as well as for the mother and the father and the socius. Psychic life first matrix subject to “the basic dyadic space” comes from the complex alchemy between the range of maternal investment and disposition and the baby’s original control abilities, without ignoring the play within a play which surrounds mothering. From an extended study in the perinatal field, this research takes an interest, with a double methodology of qualitative study (chapter 1) and quantitative “equipped” one (chapter 2 and 3), in the construction of a dyadic space, essential for the baby’s development and for the maternal investment, to this construction ups and downs, for regulatory functions of these rituals (relayed by current perinatal cares) which gives a scansion to childbirth work. Dyadic communication between mother and baby, with a climax around two months old with the first protoconversations, is studied as an asymmetrical transmodal complex co-genesis by both psychoanalysis and attachment theory, by developmental approaches and neurosciences. This unique and original space, which is renewed after every birth, is woven from “the forming of maternal investment” composed from psychic life vectors and their combination which provides enough energy for the mothering situation. From the three major schools of conceptual thoughts and from therapeutic treatments of early relationship disorder, we identify the forming “in transformation”, the “transmission” one and the forming “in seduction”. These breakthroughs have a triple purpose: to built a new reading of primary intersubjectivity, the building of baby’s self-awareness, normal and pathological mechanism of identification and empathy, to support the principle of cares and the edification of a dyadic semiology of the early psychopathology and to lead to a model that will include the aspect of the “early” and its symbolism form in adult therapy and the institutional support of fragile or borderline population.

Le lien d’appariement unissant un mâle et une femelle a été peu exploré chez les Oiseaux, alors que la plupart sont monogames. De plus, l’étude de la monogamie s’est souvent cantonnée à l’exploration des comportements liés à la reproduction et au choix du partenaire. L’objectif de cette thèse est d’identifier les comportements sociaux caractéristiques du lien d’appariement et leurs supports cérébraux chez un passereau social et monogame, le Diamant mandarin (Taeniopygia guttata). Grâce aux concepts de la théorie des réseaux appliqués à l’étude en laboratoire des interactions sociales, cette thèse s’intéresse à la valeur sociale du lien d’appariement. Elle montre qu’il représente un véritable partenariat social, suffisamment favorable pour que se forment des couples homosexuels en cas d’indisponibilité d’oiseaux du sexe opposé. Par l’étude en milieu naturel des communications acoustiques entre partenaires, ce lien est apparu comme le cadre d’échanges vocaux particuliers : de discrets duos de cris émis dans l’intimité du nid. En s’intéressant au rythme d’émission des vocalisations à l’échelle du groupe, cette thèse jette également un nouveau regard sur l’étude des communications acoustiques. La dynamique du réseau acoustique que représente le groupe est influencée par la proportion d’oiseaux appariés. Enfin, les causes proximales des liens sociaux sont abordées : l’activité du réseau cérébral impliqué dans l’expression des comportements sociaux, le « Social Behavior Network » (SBN), varie avec la possibilité de créer des liens chez les mâles. Cette thèse participe donc à définir le lien d’appariement comme un lien social complexe dont les avantages ne se limiteraient pas à la reproduction. Elle révèle ses conséquences tant sur le réseau social, que sur le réseau acoustique du groupe de diamants mandarins, et soulève l’implication potentielle du réseau cérébral SBN dans les mécanismes de sa formation et de son maintien / Although most birds are monogamous, the pair-bond between a male and a female remains relatively unexplored in these species. Moreover, the study of monogamy mainly focused on behaviours linked to reproduction and partner choice. The aim of this thesis is to identify the social behaviours and the neurophysiological mechanisms of pair-bonding in a social and monogamous passerine, the Zebra finch (Teaniopygia guttata). Using concepts of network theory to study social interactions in the laboratory, this thesis explores the social value of the pair-bond. We showed that the pair-bond is a genuine partnership, whose social value might explain why birds establish same-sex pair-bonds when partners of the opposite sex are scarce. The study of acoustic communication between mates in the field revealed that partners perform specific vocal displays : they use private duets of calls in the intimacy of their nest. By investigating the rhythm of emission of vocalizations at group scale, this thesis takes a fresh look at the study of acoustic communication. The proportion of paired birds in a group influences the dynamic of the acoustic network constituted by the social group. Finally, we tackled the proximal explanations of social bonding : in males, social relationships affect the activity of the brain network implicated in social behaviours, the Social Behavior Network (SBN). This thesis underlines the social complexity of the pair-bond that might bring benefits beyond reproduction. It reveals the consequences of pair-bonding on both social network and acoustic network, and points towards a role of the SBN in pair-bond establishment and maintenance

Obsessive-compulsive disorder (OCD) is a serious psychiatric condition manifested by repeated thoughts followed by stereotypic compulsive behavior. Alterations to cortico-thalamo-striato- cortical circuits are most often implicated in the pathophysiology of OCD. However, many studies have also found a changed volume, shape and activity of the hippocampus in OCD patients. This work focused on the activity of hippocampal CA1 cells during stereotypical checking behavior and on cognitive flexibility in a quinpirole (QNP) sensitization model of OCD. The activity of CA1 hippocampal cells during stereotypical checking was assessed in an enriched open-field test in QNP sensitized rats. Arc+ (activity-regulated cytoskeletal associated protein, or Arg 3.1) mRNA expression profiles were determined in CA1 coronal hippocampal sections following stereotypical checking. After the establishment of stereotypical checking (10 sessions), rats were exposed to the arena and sacrificed after 5 minutes. QNP sensitized animals visited the same objects with the same frequency as during previous sessions, while control rats did not. Locomotor activity was comparable between QNP treated rats and controls. Following sacrifice, rat brains were flash frozen and sliced to 20 µm thick sections. Returning to Society: Daily-life stressors in the immediate prison release phase

This paper is the third part of an ongoing research project by Andersson et al. 2014 using Interactive Voice Response, an innovative automatic telephone assessment to study Swedish paroled offenders in the first 30 days after prison. Repeated measures of qualitative reports on daily most stressful events (stressors) and quantitative severity ratings (stress) were used to study the perception of stress in the immediate prison release phase. Adding to the knowledge about prisoners’ reentry by exploring paroled offenders’ perception on daily stressful events and the stress intensity associated with these was the main purpose of this essay. Following a phenomenological approach, daily stressful events could be categorized into social, psychological and physical stressors and an insight in the everyday complexities through the reports of paroled offenders could be provided. While social stressors build the largest category, physical stressors are on average perceived as most severe. Overall stress severity shows an increase over the duration of the study period. The findings further support the feasibility of daily automated telephone assessments in the context of the Swedish Prison and Probation Service.
