Tinkamas informacijos apie maistą pateikimas, įskaitant maisto produktų ženklinimą, yra svarbi priemonė skatinti sveikatą stiprinantį gyvenimo būdą ir užtikrinti saugų maisto produktų vartojimą. Lietuvoje atliktų tyrimų duomenys rodo, kad ligų profilaktikos tikslu maisto produktus renkasi tik nedidelė dalis gyventojų, tačiau trūksta duomenų apie tai, kokios yra gyventojų maisto produktų pasirinkimo sąsajos su informacija apie maistą. Šio darbo tikslas – ištirti ir įvertinti informacijos apie maistą sąsajas su Lietuvos suaugusių gyventojų maisto produktų pasirinkimu visuomenės sveikatos požiūriu. Tyrimo rezultatai rodo, kad daugumos Lietuvos suaugusių gyventojų nuomone, maisto produktų ženklinimo informacija nėra visiškai suprantama ir aiškiai įskaitoma. Lietuvos suaugę gyventojai kaip labiausiai maisto produktų pasirinkimui darančią įtaką ženklinimo informaciją nurodė maisto produktų tinkamumo vartoti terminą, kainą ir kilmės šalį. Daugiau kaip pusė jų pageidauja, kad ženklinant maisto produktus būtų pateikiama maistingumo informacija. Labiausiai pageidaujama informacijos apie maisto produkto energinę vertę, riebalus, cholesterolį ir vitaminus. Daugiau moterų negu vyrų, aukštesnį išsilavinimą negu žemesnį išsilavinimą turinčių gyventojų domisi informacija apie maisto produktų maistingumą. Développement d'une méthode de marquage protéique par fluorescence

Le marquage protéique par fluorescence est une méthode de choix permettant d’étudier l’évolution des protéines depuis leur synthèse cellulaire jusqu’à leur dégradation, en plus de rendre possible leur localisation ainsi que la visualisation des interactions entre protéines. De cet intérêt certain ont découlé différentes techniques de marquage, dont celle présentement développée dans le groupe Keillor. Le principe de celle-ci repose sur la réaction entre deux maléimides portés par un fluorogène et une séquence peptidique cible, laquelle contient deux résidus cystéines séparés par une distance appropriée. Suite à cette double addition de thiols du peptide sur les maléimides du fluorogène, la fluorescence latente de ce dernier est régénérée, menant au marquage covalent de la protéine d’intérêt. Afin d’optimiser la spécificité et la sensibilité de cette méthode de marquage, la synthèse de nouveaux fluorogènes et l’étude de l’efficacité de quench de la fluorescence par les maléimides est présentement en cours dans les laboratoires du groupe Keillor. / The fluorescent labelling of proteins is a powerful approach for following the dynamic process of their cellular synthesis and degradation, in addition to determining their localization and protein-protein interactions. We have developed a ‘small molecule’-based labelling technique that is complementary to existing methods. Our fluorogenic approach relies on the use of dimaleimide fluorogens that react with a target peptide sequence that presents appropriately spaced, solvent-exposed Cys residues. The thiol addition reaction between target sequence and dimaleimide fluorogen restores the latent fluorescence of the latter and results in the covalent fluorescent labelling of the protein of interest. Synthesis of new fluorogens and quench efficiency studies are presently taking place in the Keillor group laboratory in order to optimize the specificity and sensitivity of the labelling method.

Aim: To determine the use and interpretation of the nutrition information on the food label of selected fat spreads by female consumers aged 25 to 45 years, living in Pietermaritzburg. Objectives: This study set out to determine the following objectives regarding the food label: what the demographics of the consumers making use of the label were; how these consumers used the label and their motivation behind this label use and did the use of the label alter the purchase of that product? Method: One hundred and fifty women aged 25-45 years were chosen from an accidental, non-probability sample of consumers shopping at selected supermarkets within Pietermaritzburg. Respondents were presented with a four part questionnaire surrounding the purchase of selected fat spreads. Results: The greatest number of respondents were from the white population group (n = 65), followed by black respondents (n = 46), Indian respondents (n = 29) and then coloured respondents (n = 10). Results showed that the respondent who was most likely to use the nutrition information on the food label had a tertiary education; was a primary food purchaser, lived with other people, had more than R1000 a month to spend on food and was conscious of choosing the healthier option. Fifty five percent of this study sample (n = 82) claimed to use the nutrition information on the label to assist with purchases and 68% (n = 102) found the nutrition information important for purchasing a new product. Of the potential factors that have previously been found to impede the use of the nutrition information label:-inadequate print size, lack of education as well as lack of nutrition information on the food label were found to be factors restricting label use in this study. The most commonly used sources of nutrition information were the media as well as friends and family and the most trusted source was the Health Professional. Discussion: If the nutrition information is to be used both correctly and effectively, there must be a major educational campaign that sets out to meet the needs of the population that are most vulnerable, especially those with an inadequate education. Conclusion: The consumer most likely to use the nutrition information on the food label has a tertiary education, is a primary food purchaser with a large amount of money available for groceries and is conscious of choosing a healthier option. From Probes to Cell Surface Labelling: Towards the Development of New Chemical Biology Compounds and Methods

Chemical biology encompasses the study and manipulation of biological system using chemistry, often by virtue of small molecules or unnatural amino acids. Much insight has been gained into the mechanisms of biological processes with regards to protein structure and function, metabolic processes and changes between healthy and diseased states. As an ever expanding field, developing new tools to interact with and impact biological systems is an extremely valuable goal. Herein, work is described towards the synthesis of a small library of heterocyclic-containing small molecules and the mechanistic details regarding the interesting and unexpected chemical compounds that arose; an alternative set of non-toxic copper catalyzed azide-alkyne click conditions for in vivo metabolic labelling; and the synthesis of an unnatural amino acid for further chemical modification via [3+2] cycloadditions with nitrones upon incorporation into a peptide of interest. Altogether, these projects strive to supplement pre-existing methodology for the synthesis of small molecule libraries and tools for metabolic labelling, and thus provide further small molecules for understanding biological systems.

Avidentifierade jobbansökningar är en relativt ny metod som under de senaste åren fått utrymme på den svenska arbetsmarknaden samt på forskningsfältet. Uppmärksamheten av metoden kommer av den tidigare forskning som visar att betydelsen av faktorer som bland annat etnisk bakgrund, kön och ålder är ibland avgörande för vem som kallas till jobbintervju. Därför åsyftar denna studie att undersöka hur rekryterare värderar och använder denna metod samt vilken betydelse avidentifiering har för rekryteringsprocessens utfall.Studien utgår ifrån ett hermeneutistiskt perspektiv och har en kvalitativ ansats. Med hjälp av en komparativ undersökningsdesign och flerfallsstudie som analysmetod behandlas det empiriska materialet, vilket utgörs av fem semistrukturerade intervjuer. Dessa intervjuer skapar tillsammans skapar två fall inom IT och konsultbranschen.Analysen behandlas med hjälp av det teoretiska ramverket vilket består av stämplingsteori samt de analytiska begreppen homosocialitet och etnosocialitet. Detta för att öka förståelsen för hur en sorteringsmetod som baseras endast på formella kriterier, utbildning och arbetslivserfarenheter, fungerar i praktiken. Det har framkommit att vid avsaknaden av faktorerna läggs fokus istället på den individuella kompetensen. Informationen gällande individens kompetens och arbetslivserfarenhet är den del som får mest kritik för att vara för ospecifik. Inom IT branschen har det visat sig att faktorer som ålder och etnicitet är av betydelse vid anställning. Det uppstår även en svårighet då kundinflytandet spelar så stor roll vid rekrytering och skapar en ram av vad som efterfrågas av varje anställd. Det är inte endast avgörande vad rekryterare eller arbetsgivare värderar utan det finns flera aktörer som spelar in vid valet av medarbetare. / Anonymous job applications are a relatively new method that has gained space in the Swedish labor market and the research field in recent years. Research shows that personal attributes are often crucial for who gets called to a job interview. Therefore, this study refers to examine how recruiters evaluate and use this approach and demonstrates the importance that anonymity has to the outcome of the recruitment process.The study is based on a hermeneutic perspective with a qualitative approach and was conducted using a comparative study design, with analysis based on several case studies. The empirical material consists of five semi-structured interviews, which together creates two cases within the IT and consulting industry.During the analysis, a theoretical framework has been presented which consists of the labelling theory and the analytical concepts of homosociality and etnosociality. These three are used to increase the understanding of how a sorting method based only on formal criteria, training and work experience, is used in practice. This has led an employer to focus on individual competency instead of ethnic background, gender or age. However, a problem remains in the industry where age is a determining factor and ethnicity is linked to Swedish language skills. Difficulties also arise when customer influence plays a large role in the recruitment process and this creates a framework of what is expected of every employee. This enables consultants to be adapted depending on the criteria that are necessary for the specific company. It is not only vital what recruiters or employers value but also what the clients wants and expect of the consultants.

An Examination of the Population Health Implications of Voluntary Food Fortification and Nutrition-related Marketing Practices in Canada.

The 2004 Canadian Community Health Survey (CCHS) revealed many indicators of poor diet quality in Canada which, together with the high rates of obesity and diet-related chronic disease, suggest that shifts in dietary patterns are urgently needed. Given the widespread promotion of foods on the basis of nutrition and health, the aim of this work was to explore the population health implications of voluntary food fortification and nutrition-related marketing in Canada. Using the CCHS, the potential impact of a proposed discretionary food fortification policy on nutrient inadequacies and excesses was examined, in addition to the relationship between consumption of foods eligible to be fortified under this policy and indicators of dietary quality. To better understand the potential risk associated with liberal fortification practices, the National Health and Nutrition Examination Survey (2007-08) was used to examine potential for risk of excess associated with voluntarily fortified food consumption in the US, where these practices have long been permitted. The results suggest that proposed changes to voluntary fortification may reduce inadequacy and increase excess, and may reinforce poor diet patterns. Excessive nutrient intakes were also found to be associated with consumption of voluntarily fortified foods in the US, particularly among children. Therefore, there appears to be real potential for risk associated with voluntary fortification practices in Canada. The extent, nature, and population health implications of nutrition marketing in Canada was examined, using a survey of front-of-package nutrition-related marketing on foods within three large grocery stores in Toronto. Nutrition-related marketing was found on 41% of all foods surveyed, and was widely found on highly processed, often fortified foods. References to nutrients of public health concern (e.g. sodium, vitamin D) were infrequently found. Overall, this practice provides limited nutritional guidance. Current directions in nutrition policy in Canada should be re-evaluated, to ensure that they support healthy diet patterns.

Functional properties of the intact and compromised midbrain dopamine system

The midbrain dopamine system is involved in many aspects of purposeful behaviour and, when compromised, can have devastating effects on movement and cognition as seen in diseases like Parkinson's. In the healthy brain, dopamine neurons are thought to play particularly important roles in learning by signalling errors in reward prediction. The objective of this thesis was to investigate the diversity in the functional properties of the midbrain dopamine system, and how this is altered through genetic variation of relevance to Parkinson's and development of cell phenotype. This objective was addressed with a combination of behavioural experiments, in vivo single-cell recording and labelling (both in anaesthetised as well as awake rodents), immunofluorescence labelling, retrograde tracing and stereology. In a first set of experiments, it was demonstrated that chronic as well as acute genetic challenges can alter the firing patterns of midbrain dopamine neurons. Using a novel bacterial artificial chromosome-transgenic rat model, it was shown that the R1441C mutation in human leucine-rich repeat kinase 2, which is linked to Parkinson's, leads to motor deficits and an age-dependent reduction in the in vivo firing variability and burst firing of substantia nigra pars compacta (SNc) dopamine neurons. These findings help reveal processes of early, pre-degenerative dysfunction in dopamine neurons in Parkinson's. Similar effects on firing variability and burst firing of SNc dopamine neurons were found in a mouse model with conditional knock- out of the transcription factors Forkhead box A1 and A2 (FoxA1/2) in midbrain dopamine neurons. These findings indicate that FoxA1/2 are not only crucial for the early development of dopamine neurons, but also their function in the mature brain. In a second set of experiments in wildtype mice, it was demonstrated that midbrain dopamine neurons (located in SNc and ventral tegmental area) show diverse expression of the molecular markers Calbindin, Calretinin, Aldh1a1, Sox6, Girk2, SatB1 and Otx2. It was found that selective expression of these markers is of use for discriminating between midbrain dopamine neurons that project to dorsal striatum or nucleus accumbens. To elucidate whether the diverse molecular marker expression would map onto dopamine neurons whose firing correlates with distinct behavioural events, midbrain dopamine neurons were recorded and labelled in head-fixed awake mice either exposed to neutral sensory stimuli or performing a classical conditioning paradigm. The population activity of midbrain dopamine neurons was not modulated by neutral sensory stimuli. Interestingly, fewer than 50% of identified dopamine neurons showed phasic firing increases following reward- predicting cue and/or reward delivery, despite the common assumption that most (if not all) midbrain dopamine neurons signal reward prediction errors. Instead, firing was modulated by other explanatory factors, such as licking, or showed no modulation during the task. Response types of midbrain dopamine neurons were not correlated with their anatomical location nor the selective or combinatorial expression of the markers Aldh1a1, Calbindin and Sox6. In conclusion, the first set of experiments identified how different genetic burdens can alter the in vivo firing of midbrain dopamine neurons, and provide new insights into how circuits can change in pathological or compensatory ways at early disease stages in Parkinson's. The second set of experiments revealed striking heterogeneity of midbrain dopamine neurons in the intact system, and established further a functional diversity in the response types of identified midbrain dopamine neurons that is only partially consistent with canonical reward prediction error signalling.
