Obregón mansilla, Alexandra J. I. January 2004 (has links)
The knowledge of protein structure constitutes one of the main approaches towards understanding the molecular basis of human disease, in particular, protein structure can be used to rationalize the effects of many disease-causing mutations, which often are associated with changes of protein stability. In this work, the BRCT domain is studied, an evolutionary protein module with autonomous folding, of approximately 95 aminoacids, found in numerous proteins involved in DNA repair, recombination and cell cycle control. One of the conserved features of the BRCT fold is a two helical bundle located against one side of the central four-stranded ß sheet, which features two of the best known mutations in breast cancer resulting from introducing a distabilizing charge within the interface of the BRCT-BRCT complex in BRCA1. We have chosen to probe the stability of the BRCT of DNLJ ligase in a thermophilic bacteria by removing the surface charge "R21" from this bundle. This is a first step to test whether this surface charge contributes significantly to the stability of this thermophilic domain, to gain insight about the possible role of BRCT stability on the breast cancer disease, and most of all, to determine the basis for the stability of this frequently found and biologically relevant structural domain. Our preliminar results suggest that the stability of the BRCT of the DNLJ ligasa of Thermus thermophilus BRCT domain is tolerant to the surface charge removal, but is affected by hydrophobicity. Although these mutations do not show significant variations in the structure and stability of the domain itself, they may affect their ability to interact with other protein modules.

Gonzalez Castillo, Gloria Carolina 06 July 2018 (has links)
The aim of this paper was to explore the relationship between the coping strategies and the perceived social support in the recent diagnosis stage. In a group of 56 women with breast cancer, whose ages fluctuate between 27 and 80 (M=53.71, DE= 12.87). The diagnosis time ranges from 1 to 8 months and they were treated in a prevention centre in Lima. The Questionnaire for Estimating Coping (COPE) (Crespo & Cruzado, 1997) and the MOS Questionnaire for Social Support in Primary Care (De la Revilla et al., 2005) were used. The results evidence the significant and direct relationships between the perceived support indices and the strategies: positive reinterpretation, planning, becoming religious; humor using and an inverse relationship with denial. Emotional/Informational support and affective support are directly related to the strategies search for support for emotional reasons and acceptance. Affective support is directly related to the strategies search for support for instrumental reasons. Age is positively related to becoming religious and the instrumental support; and, having a partner has a positive relationship with affective support. In conclusion: in the recent diagnosis phase, there is a relationship between perceived social support and strategies focused on emotion; the patients make use of different strategies; and, social support affects the use of strategies considered as adaptive. This information could be used in intervention for the recently diagnosis in order to achieve a better adjustment to cancer. / Tesis

Lostaunau Calero, Ana Vanessa, Torrejón Salmón, Claudia Sofía 25 March 2014 (has links)
Breast cancer might be considered a life-threatening event as it demands changes that could exceed the patient’s abilities to cope with their illness, therefore, affect HRQoL in a negative way. This study analyzes the contribution that traumatic stress, perceived stress, coping, migration and type of treatment have on the variability of physical and metal components of the HRQoL of 53 mastectomized women with breast cancer. The instruments used include the Spanish adaptation of the SF-36 health survey (Alonso, Prieto & Antó, 1995), the COPE Inventory (Carver, Scheier & Weintraub, 1989), the Perceived Stress Scale PSS (Cohen, Kamarck & Mermelstein, 1983) and the Peruvian adaptation of the traumatic stress scale (Gargurevich, 2006). The analysis found medium and high levels of the HRQoL. Physical component was more affected than mental component, especially in migrant patients. The model of the physical component was comprised by migration, treatment type and traumatic stress, which explained 30% of its variability. On the other hand, two models were obtained from the mental component; the first was comprised by traumatic stress and perceived stress, which explained 39% of its variability, while the second model, comprised by traumatic stress, perceived stress an emotion-oriented stress, explained 42% of its variability / Tesis

Sánchez Rupay, Jenny Natalíe 30 September 2015 (has links)
The objective of this investigation is to describe and compare the construct known as psychological well-being and its dimensions in women with breast cancer who had surgery in contrast to healthy women who share the same demographic characteristics. The sample consisted of 28 breast cancer patients, between the ages of 36 and 75 years old (M=56.11, SD= 11.93). The instrument used was the adaptation made by Diaz et al. (2006) of Ryff's Psychological Well- Being Scales (1988), which dimensions are: self-acceptance, autonomy, purpose in life, environmental mastery, personal growth and positive relations with others. No significant differences were found between both groups at the level of the global scale of psychological well-being and its dimensions. In response to the specific objectives, we found positive correlation between age and the dimensions of autonomy and self-acceptance. Negative correlation was found between the variable "number of surgeries" and global psychological well-being, self-acceptance, purpose in life and autonomy. The variable "type of surgery" presented significant differences between the group of patients that had a tumorectomy only and the group that went under two surgeries (tumorectomy and mastectomy). The first group reported higher levels of global psychological well-being, autonomy and self-acceptance. / Tesis

Maschio, Larissa Bazela. January 2015 (has links)
Abstract: Mammary tumors are the most common tumors among women and female dogs. Fibroblasts associated with cancer (CAFS) are the most abundant cells in the tumor microenvironment, able to mediate inflammatory responses acting in the differentiation and tumor progression. Melatonin is associated with mechanisms of action with oncostatics effects and oncoprotectors, and works to reduce the formation of surrounding fibroblasts in breast cancer. The objectives of this study were to assess the action of melatonin on mammary tumor cell lines human and canine metastatic (MDA-MB-231 and CF-41) and in co-culture with primary CAFS on cell viability and expression of proteins involved in the processes of angiogenesis, inflammation and apoptosis searching for the understanding of proteins interaction in the main signaling pathways active in the tumorigenic process. Cell viability was assessed by MTT assay and the semi-quantitative protein analysis by Membrane Antibody Array after treatment with melatonin. It was found that 1 mM of Melatonin reduced cell viability in both cell lines, human and canine CAFS and their co-culture (p <0.05). The semi-quantitative protein analysis after melatonin treatment showed a reduction of the expression of seven pro-angiogenic proteins and increased TIMP-1 and IL-6 protein in MDA-MB-231 cells and reduced IL-6 and MCP-1 and increased leptin, TGF-β, VEGF and thrombopoietin in the co-culture (p <0.05). Nineteen pro-inflammatory cytokine protein were reduced and I-309 increased in MDA-MB-231 cells (p <0.05). In the co-culture, six pro-inflammatory proteins were underexpressed and the IL-4, MCP-3 and SCF proteins were increased the expression (p <0.05). Twelve pro-apoptotic proteins increased and decreased expression of caspase-3 in MDA-MB-231 cells (p <0.05). In the co-culture, an increase of BIM, HPS27 and IGF-1R proteins and TNF-RII reduction was observed (p <0.05)... / Mestre

The mammary tumor study in female dogs is revealed as an excellent model for the investigation of breast tumors in humans. On both species, the immune system can interfere on neoplasia progression. The aim of this study was to evaluate the immunolocalization and quantification of CD4, CD8, CD4+CD25+ (Foxp3), CD28, CD152 and NK in tumour lymphocyte infiltration, to evaluate the efficacy of stimulation and immune system defense in canine mammary carcinoma. The expression of these markers using immunohistochemistry was made by streptoavidin-biotin peroxidase method. The tissues were collected from twenty mammary carcinomas, subdivided in ten simple carcinomas and ten complex carcinomas of twenty female dogs. This study identified no significant differences on quantification of positive cellular staining in simple carcinoma compared with complex carcinoma. The samples of simple carcinoma resulted significant differences on positive staining between the antibodies CD28 (p=0,03) and Foxp3 (p=0,01) and these markers revealed less quantitative staining compared with the others markers (CD4, CD8, CD152 and NK). The samples of complex carcinoma resulted significant differences on cellular staining quantification by the antibody CD152 (p<0,05) compared with the others markers (CD4, Foxp3, CD28, and NK) with exception when compared with CD8. The immune quantification of positive cellular staining about the markers CD28, CD56 and Foxp3 were smaller than CD8 (p=0,01), and the Foxp3 was smallest than those others markers. It was concluded that exist a negative control of immune system to complex and simple mammary carcinoma, but, this negative control was more significant in female dogs with complex mammary carcinoma / Mestre
