"Dom skiter i vad du har gjort, dom vill bara att du ska lyckas" : En fallstudie om organisationen Process Kedjan / "They don't care what you did, they just want you to suceed" : A case study about the organization Process Kedjan

This thesis aims to examine the importance of the organization Process Kedjan regarding youth’s termination of delinquency and drug abuse. It also aims to examine in which extent the organization helps the youths in different ways than authorities, as well as how the organization can function as a complement to authorities. The thesis is designed as a case study and is based on the perspective of the organization’s members. The study was performed by using qualitative interviews with five boys who have received or still are receiving help from the organization. Three of them are still receiving support and two of them are now working in the organization. They are all in the ages between 18 and 25, and has been active in delinquency and drug abuse in the ages between approximately 12 and 20. The theory of age-graded informal social control by Laub and Sampson was applied in the analysis. In the processing of the empirical material it emerged that Process Kedjan has been of great importance for the youths in their desistance of delinquency and drug abuse. Other factors of significance has been: close relationships, daily routines and activities and a sense of guilt for the harm they have caused other human beings. Another finding in the study was that Process Kedjan has qualities that cannot be found in authorities. These qualities are summarized in four different terms: identification, reliance, accessibility and fellowship. By the result we could find that Process Kedjan is an important complement to the authorities work regarding to prevent relapse in delinquency and drug abuse among youths, but collaboration between the organization and different authorities is required to be able to help a youth. The organization has contributed to a better view on authorities among the youths, they now understand the importance of dealing with for instance social services. / Denna uppsats syftar till att undersöka vilken betydelse organisationen Process Kedjan har för ungdomars upphörande av kriminalitet och missbruk. Den syftar även till att undersöka i vilken utsträckning organisationen hjälper ungdomar på andra sätt än myndigheter samt hur organisationen kan fungera som ett komplement till myndigheter. Uppsatsen är utformad som en fallstudie och utgår från medlemmarna i Process Kedjans perspektiv. Empirin har insamlats genom kvalitativa intervjuer med fem unga män som får eller har fått hjälp av organisationen. Tre av dem tar fortfarande emot hjälp och två arbetar numera i organisationen. I analysen tillämpades Laub och Sampsons teori om upphörandeprocesser, vändpunkter och sociala band. Vid bearbetning av det empiriska materialet framkom det att Process Kedjan har varit av stor betydelse för att ungdomarna ska kunna upprätthålla ett liv utan brott och missbruk. Andra viktiga faktorer har varit: nära relationer framför allt med familj, dagliga rutiner och aktiviteter samt en skuldkänsla av att ha orsakat skada hos andra människor. Det framkom också att Process Kedjans verksamhet innehåller delar som inte går att finna hos myndigheter. Dessa har sammanfattats i fyra olika begrepp: identifikation, tillit, tillgänglighet och gemenskap. I resultatet går det att utläsa att Process Kedjan är ett viktigt komplement till det arbete som myndigheter utför angående att förebygga återfall i kriminalitet och missbruk bland ungdomar, men för att kunna hjälpa en ungdom krävs ett samarbete mellan organisationen och olika myndigheter. Med hjälp av stöd från organisationens personal har ungdomarna fått en förändrad syn på myndigheter och ser nu vikten av att ha en kontakt med exempelvis socialtjänsten.

Effect of CTCF and Cohesin on the dynamics of RNA polymerase II transcription and coupled pre-messenger RNA processing

The CCCTC-binding factor (CTCF) is a versatile, multifunctional zinc-finger protein involved in a broad spectrum of cellular functions. In mammalian cells, CTCF functions together with the Cohesin complex, an essential regulator of sister chromatid cohesion. Together, CTCF and Cohesin have been shown to regulate gene expression at a genome-wide level in mammalian cells. In the yeast Saccharomyces pombe, Cohesin has been implicated in transcription termination of convergently transcribed genes, in a cell cycle dependent manner. The aim of this thesis was to investigate the possibility of direct transcriptional involvement of CTCF and Cohesin in human cells. The first model system applied for this experimental purpose was the β-globin gene with introduced canonical CTCF-binding sites replacing the endogenous Co- Transcriptional Cleavage (CoTC) element downstream of β-globin. The results obtained indicate that recruitment of CTCF to the β-globin 3` flanking region does not prevent read-through transcription. However, CTCF-binding does mediate RNA Polymerase II (Pol II) pausing at the site of recruited CTCF. This results in more efficient pre-mRNA 3` end processing and therefore rescues β-globin mRNA to wild type levels. Cohesin was not detected at the introduced CTCF-binding sites. These results are a contribution to our understanding of the spatio-temporal requirements for cotranscriptional events like 3` end pre-mRNA processing and Pol II kinetics. The second part of my thesis presents an investigation on the involvement of CTCF and Cohesin in lipopolysaccharide (LPS)-induced Tumor Necrosis Factor α (TNFα) gene expression regulation in human monocytes and differentiated M1- and M2-type macrophages. These studies provide first evidence of Cohesin recruitment to the TNFα gene body and its regulatory NFκB-binding sites. Differences in the recruitment profiles obtained indicate potential regulatory differences of TNFα among the three cell types. Preliminary data provide an insight into the effects on TNFα mRNA levels upon down-regulation of Cohesin subunits.

A critical ethical assessment of the South African Termination of Pregnancy Bill

Thesis (MPhil)--University of Stellenbosch, 2001. / ENGLISH ABSTRACT: Perhaps one of the most talked about subjects worldwide and in South Africa these days is the abortion issue. A growing number of women admit to having had one. Basically there are two opposing views and values on the question of abortion. We normally hear people referring to the 'abortion issue'; my understanding of this is that there is a dialogue going on at the moment concerning abortion. In South Africa before the current Choice of Termination of Pregnancy (TOP) Bill, some activists' women and the ever-growing 'feminists' movements were lobbying and demanding that abortion be decriminalised. As we may all be aware, up until 1 February 1997, abortion or termination of pregnancy (TOP) in South Africa was conceivable under very restrictive atmosphere. Before the introduction of the current Termination of Pregnancy Bill, a majority of women had no access to abortion services in the country, hence the growing number of back-street job. By implication this means that most women given the choice, would not seek the experience of abortion, but if they do, it would be available to the in safe, legal, accessible and affordable service. Not only does the Act conceal that terminating pregnancy that occurred through criminal acts such as rape and incest is justifiable. The current liberal Termination of Pregnancy Bill also gives pregnant women the 'right' or 'freedom' to abort whenever and for whatever reason they deem fit. Part of the ethical dilemma of the abortion issue is that there are those who holds a view that always where there is a conflict of rights and interests, the foetus' rights must give way to, or that the foetus' rights must be overridden by those of a pregnant women. Pro-choice advocates maintain that a woman's choice to terminate her pregnancy is her own business and hers alone, in other words, this for them is a private decision. Well, I argue that this is not necessarily the case, ethically, the father of the unborn child should also be considered in such a decision. Given that virtually every abortion has risks, the parents of the aborting woman and to some extent the society at large are involved. Therefore, to solely talk of the 'mother's right to choose' is basically suggest that morality is "relative" and such relativism is conceived from the idea of privatisation of abortion and life in general. In the following pages I will look at the arguments in support of abortion and against it, and these are criticised. Also discussed are the ethical implications of the new South African Termination of Pregnancy Act. Broadly speaking, technology advancement has made it possible to detect the unborn baby's physical condition (sometimes even its mental state) while the mother is still pregnant. The ethical implications of this medical intervention are used to decide whether the unborn child should live or die. Given this, if the purpose of these prenatal diagnosis were for the destruction of the unborn, therefore, advocates of the movements such as 'the right to life', and 'pro-lifers' would argue that because of particularly twisted purpose, prenatal diagnosis must be abolished. Furthermore, I will acknowledge that the Termination of Pregnancy Bill as we have it, is appraised by feminists movements and others who are not necessarily feminists as allowing increased and unrestricted access to 'free' and 'safe' abortion in the government hospitals and clinics. However, I argue that this was rather prematurely introduced. I argue that a number of pregnant women claiming to be poor still present themselves to private doctors and private clinics for abortion and they pay anything between R 600- 800 or more depending where these services are provided. On the other hand, for one reason or another, other women still choose to terminate their pregnancies back street way although the risks are high in such servicing stations. In the light of these facts, one wonders whether it is appropriate to legislate for the termination of pregnancy or would it have been a worthwhile decision to delay the legislation of abortion for a while and thoroughly make a research and relevant preparation for it. I also argue that ideology plays an important part in the abortion debates. Besides, the abortion debate is also characterised by indoctrination, the purpose of which is to leave other confused. In both cases facts are misrepresented or false statements are made, and this for me is ethically unacceptable. I will also comment on the importance of linguistics, that is, the proper understanding of normal English terms and what I refer to as 'deceptive language' used by campaigners. Inthe last part of this thesis, I will outline some basic approaches to ethics and which belong to what is referred to as postmodernism. The Postmodern worldview deconstructs metanarratives so that no one particular belief is more believable than another. This worldview bring with it ethical relativism, which is a theory which holds that morality is relative to the individual. Three movements are given as an example of this move toward ethical relativism, they are: (a) Emotivism, (b) Subjectivism, and (c) Situationalism While I will argue that rape and incest are evil acts, and support abortion in cases involving such acts, however, I also believe that abortion is not the answer to the problem of rape and incest. I will propose a number of recommendations the South African government should have made before legislating for abortion. For instance, by creating separate abortion service facilities even in the hospital premises, with properly trained staff; so that people who came to seek advice for abortion are not intimidated by those who go to full terms with their pregnancy. Included in this thesis is a case study to demonstrate the complexity of the abortion issue to everyone involved. Some psychological and emotional symptoms following abortion will be outlined and this according to women who do share their abortion story is a reality they have to live with every day of their lives. / AFRIKAANSE OPSOMMING: Aborsie is moontlik een van die mees veelbesproke kwessies van ons tyd, sowel in Suid- Afrika as wêreldwyd. 'n Groeiende aantal vroue erken dat hulle al een gehad het. Basies is daar twee opponerende gesigspunte en waardes betrokke by die twispunt rondom aborsie. Aborsie was voor die aanvaarding van die jongste wetgewing (d.w.s. voor 1 Februarie 1997) in Suid-Afrika slegs moontlik onder streng beperkings. Voor die huidige wet ( die "Termination of Pregnancy Bill") in werking gekom het, het die meerderheid vroue geen toegang tot aborsie gehad in Suid-Afrika nie, wat gelei het tot 'n toename in agterstraat aborsies. Die nuwe wet gee nie slegs die reg om te aborteer aan vroue wat swanger is as gevolg van kriminele optrede soos verkragting of bloedskande nie. Die huidige, buitengewooon liberale wet gee ook vir alle praktiese doeleindes aan die vroue die reg om aborsie op versoek te ondergaan tot op 20 weke van swangerskap. Die doel van hierdie werkstuk is om hierdie nuwe liberale wet aan 'n krities ondersoek te onderwerp. Deel van die etiese dilemma rondom die kwessie van aborsie spruit voort uit die feit dat daar diegene is wat reken dat, indien daar enige konflik tussen regte en belange is, die regte van die fetus ondergeskik is aan die regte van die swanger vrou. Diegene ten gunste van aborsie voer aan dat die keuse gemaak moet word deur die betrokke vrou, en dat so 'n keuse uitsluitlik haar eie is. Ek argumenteer dat dit nie noodwendig die geval is nie. Die vader van die ongebore kind behoort ook 'n sê te hê in hierdie saak. Gegee dat elke aborsie sekere risiko's insluit, het die ouers van die betrokke vrou en die samelewing ook 'n belang by so ,'n situasie. Dus is om slegs te praat van die 'vrou se reg om te Ides' om te suggereer dat moraliteit "relatief' is, en sulke relativisme word afgelei van die idee van die privatisering van aborsie en die lewe in die algemeen. Maar ek wil argumenteer dat die wet te vroeg aangeneem is. Ek argumenteer dat 'n groot aantal verwagtende vroue voorgee dat hulle arm is en poog om 'n aborsie te kry by private dokters en klinieke, en dan tussen R600 - R800 of meer betaal vir so 'n diens, afhangende van waar dit geskied. Aan die ander kant, om een of ander rede, kies sommige vroue steeds om hulle swangerskappe te termineer deur agterstraat-aborsies, ten spyte van die risiko's. Gegewe hierdie feit, wonder mens of dit gepas was on 'n wet in te stel aangaande die terminasie van swangerskap, en of dit nie beter sou wees om die wetgewing uit te stel tot volledige navorsing gedoen is en voorbereiding getref is nie. Ek argumenteer ook dat ideologie 'n belangrike rol speel in die aborsie-debat. Die aborsie-debat word ook gekenmerk deur indoktrinasie ten einde mense te verwar. In beide gevalle is daar die wanvoorstelling van feite of word valse stellings gemaak, wat eties onaanvaarbaar is. Ek salook kommentaar lewer oor die belangrikheid van taal, dws die korrekte verstaan van normale (Engelse) terme en wat ek na verwys as die 'misleidende taal' wat gebruik word deur sekere kampvegters betrokke by die debat. In die laaste deel van die werkstuk sal ek sekere basiese benaderings tot etiek ondersoek, veral dié wat na verwys word as "postmodernisme". Die Postmoderne gesigspunt dekonstrueer metanarratiewe sodat geen spesifieke oordeel langer meer geloofwaardig is as 'n ander nie. Hierdie gesigspunt word dan ook vergesel deur etiese relativisme, wat huldig dat moraliteit relatief is tot die individu. Drie bewegings word genoem as voorbeelde van hierdie beweging na etiese relativisme, nl: (a) Emotivisme, (b) Subjektivisme, en (c) Situasie-etiek Alhoewel ek argumenteer dat verkragting en bloedskande morele verkeerd is, en alhoewel ek aborsie in sulke gevalle voorstaan, glo ek nie dat aborsie 'n antwoord bied op die probleem van verkragting en bloedskande nie. Ek sal 'n aantal voorstelle maak aangaande wat eintlik moes gebeur het voor die regering die huidige aborsiewet aanvaar het. Byvoorbeeld, dat aparte aborsie-fasiliteite, selfs by die hospitaal en met opgeleide personeel, geskep moes word ten einde te voorkom dat diegene wat advies vra aangaande aborsie nie geïntimideer word deur persone wat nie wil aborteer nie. Ingesluit in hierdie studie is 'n gevallestudie wat die kompleksiteit van die kwessie rondom aborsie, vir al die rolspelers, demonstreer. Sekere emosionele en sielkundige simptome, veroorsaak deurdat 'n persoon besluit het om te aborteer, sal geskets word. Vir vroue wat 'n aborsie ondergaan het is hierdie 'n realiteit waarmee hulle elke dag moet saamleef

Kinetik der konventionellen radikalischen und der RAFT-Polymerisation an Nanopartikeloberflächen / Kinetics of the conventional radical and the RAFT polymerization from nanoparticle surfaces

Diese Arbeit gibt neue Einblicke in die kinetischen und mechanistischen Aspekte der konventionellen radikalischen und der Reversible-Addition–Fragmentation-chain-Transfer- (RAFT-)Polymerisation von der Oberfläche von Silica-Nanopartikeln. Die Terminierungskinetik der oberflächeninitiierten Polymerisation von n-Butylmethacrylat (n-BMA) wurde mit Hilfe der Single-pulse–pulsed-laser-Polymerisation (SP–PLP) in Kombination mit der Elektronenspinresonanzspektroskopie (ESR-Spektroskopie) analysiert. Die Polymerisation wurde dabei durch den neu entwickelten oberflächengebundenen Photoinitiator 2,2-Dimethoxy-2-phenyl-1-(2-(2-(trimethoxysilyl)-ethyl)phenyl)-1-on (DMPTS) initiiert. Die erhaltenen Geschwindigkeitskoeffizienten der Terminierung kt, die ausschließlich der Terminierungsreaktion zwischen zwei oberflächengebundenen Radikalen zugeordnet werden konnten, waren signifikant kleiner als die Literaturwerte für das entsprechende homogene Polymerisationssystem. Erstaunlicherweise konnte außerdem keine Kettenlängenabhängigkeit von kt gefunden werden. Dies zusammen mit der Tatsache, dass bei temperaturabhängigen Untersuchungen eine Aktivierungsenergie für kt gefunden wurde, die in exzellenter Übereinstimmung mit der des Propagationsgeschwindigkeitskoeffizienten von n-BMA ist, lässt das Bild einer reaktionsdiffusionskontrollierten Terminierung oberflächengebundener Makroradikale entstehen. Die Entwicklung des oberflächengebundenen Photoinitiators DMPTS erlaubte weiterhin neue Einblicke in die Propagationskinetik – insbesondere deren Temperaturabhängigkeit betreffend – oberflächengebundener Makroradikale. Mittels der PLP von n-BMA in Kombination mit der Größenausschlusschromatographie (engl. size-exclusion chromatography, SEC) wurden Werte für den Geschwindigkeitskoeffizienten der Propagation kp und die entsprechende Aktivierungsenergie ermittelt, die exzellent mit den Daten der International Union of Pure and Applied Chemistry (IUPAC) für die Polymerisation von n-BMA in homogenem Reaktionsmedium übereinstimmen, sodass kein Einfluss der Oberflächenverankerung der Makroradikale auf die Propagationskinetik ersichtlich ist. Weiterhin wurde die Kinetik der RAFT-Polymerisation von n-Butylacrylat (BA) von Oberflächen bei –40 °C mit Hilfe verschiedener ESR-spektroskopischer Methoden für den R- und den Z-Gruppen-Ansatz unter Verwendung oberflächengebundener Trithiocarbonate als RAFT-Agenzien untersucht. Die kinetischen Analysen des R-Gruppen-Ansatzes mittels stationärer ESR-Experimente lieferten Gleichgewichtskonstanten Keq, die um eine Größenordnung kleiner sind als die Literaturwerte für entsprechende homogene Polymerisationssysteme, was darauf hindeutet, dass die Oberflächenverankerung des RAFT-Agens einen signifikanten Einfluss auf die Kinetik der RAFT-Polymerisation hat. Durch die Aufnahme zeitlicher Konzentrationsverläufe der Intermediatspezies und der propagierenden Radikale nach Einzelpulsinitiierung mittels der hochaufgelösten zeitabhängigen ESR-Spektroskopie (SP–PLP–ESR–RAFT) in Kombination mit PREDICI®-Simulationen wurden diese Ergebnisse bestätigt, da eine Abnahme des Geschwindigkeitskoeffizienten der Addition eines Makroradikals an die RAFT-Gruppe mit zunehmender Anzahl beteiligter oberflächengebundener Makromoleküle gefunden wurde. Dieser Befund ist im Einklang mit der Vorstellung, dass die Additionsreaktion bei Beteiligung verankerter Makromoleküle diffusionskontrolliert abläuft, was auf die unterdrückte Translations- und die eingeschränkte Segmentdiffusion oberflächengebundener Polymerketten zurückzuführen ist. Ein noch kleinerer Wert von Keq wurde für den Z-Gruppen-Ansatz gefunden und kann darauf zurückgeführt werden, dass die in Lösung wachsenden Ketten für die Additionsreaktion mit der RAFT-Gruppe, die sich über den gesamten Verlauf der Polymerisation in unmittelbarer Nähe zur Partikeloberfläche befindet, durch die Polymerschicht auf der Oberfläche diffundieren müssen. Die Wahrscheinlichkeit dafür, dass eine Additionsreaktion stattfindet, nimmt mit zunehmender Polymerschichtdicke ab, was durch einen kleineren Wert von Keq bei Verwendung von einem RAFT-Agens, das über die Z-Gruppe verankert war und über eine makromolekulare anstatt einer niedermolekularen R-Gruppe verfügte, gezeigt werden konnte. Die vorgestellten Ergebnisse geben einen umfangreichen Überblick über den Einfluss der Oberflächenverankerung von Komponenten, die am Prozess der radikalischen Polymerisation beteiligt sind, auf die Kinetik der oberflächeninitiierten radikalischen Polymerisation und ebnen den Weg für detaillierte, systematische Untersuchungen der Zusammenhänge zwischen der Kinetik von radikalischen Polymerisationen an Oberflächen und der molekularen Struktur der erhaltenen oberflächengebundenen Polymere.

Apribojimai nutraukti darbo sutartį reorganizuojant įmonę / Restrictions on the termination of an employment contract when restructuring an enterprise

Šio darbo pirmoje dalyje yra nagrinėjama Europos Bendrijų teisės aktuose nustatytos garantijos darbuotojams, taikomos įmonės reorganizavimo atveju. Tyrimo metu analizuojama Direktyva Nr. 2001/23/EB, remiantis Europos Teisingumo Teismo praktiką, aiškinamos direktyvos nuostatos įtvirtinančios individualią bei kolektyvinę darbuotojų interesų apsaugą. Antrojoje dalyje analizuojama reorganizavimo samprata civilinėje bei darbo teisėje, atskleidžiama reorganizavimo sąvoka, būdai, etapai. Pateikiamas reorganizavimo palyginimas su likvidavimo procedūra bei restruktūrizavimu. Taip pat analizuojamos Lietuvos Respublikos darbo kodekso normos, nustatančios apribojimus darbdaviui nutraukti darbo sutartį reorganizuojant įmonę. Tyrimo metu paaiškėja, kad nacionalinės teisės aktai nevisiškai atitinka Direktyvos Nr. 2001/23/EB tikslus. Nors reorganizavimas pats savaime nėra teisėta priežastis darbo santykių nutraukimui, tačiau darbuotojų apsauga nėra absoliuti. Reorganizavimo metu ir jam pasibaigus įmonėje susiklosto sudėtingi darbo santykiai, darbo santykių tęstinumas ne visada yra užtikrinamas. Todėl trečioje darbo dalyje yra kalbama apie darbdavio teisės nutraukti darbo sutartį įgyvendinimą bei jos apribojimus, aptariama darbuotojų perkėlimo galimybė, informavimo ir konsultavimo pareigos realizavimas. Darbo pabaigoje yra suformuluotos išvados remiantis atliktu teisės aktų bei Europos Teisingumo Teismo bei Lietuvos Aukščiausiojo Teismo praktikos tyrimu bei teisės doktrinos analize. / In the first part the main issue is analyzes of European Communities law and the Directive No 2001/23/EC witch laid down legal guarantees for the employees during the process of enterprises reorganization. The main provisions of the directive that include protection of individual and collective employee’s interests were explained according to The European Court of Justice practice. In the second part of the work was done analyzes of the concept of reorganization in civil and labor law, in purpose to find out definition of reorganization, means and stages of process. There are given a comparison of reorganization and liquidation, reorganization and restructuring. This part of work also includes analyzes of Labour Code norms, which laid down restrictions on employer who is unallowable to terminate the employment contract during reorganization of the enterprises. The study reveals that national legislation does not fully comply with purposes of Directive No 2001/23/EC. Title: Reasons for the termination competitive activity of tennis players Objectives: The primary objective of this thesis is to analyze the possible reasons for termination competitive activity of teenage tennis players, which are obtained by qualitative method of data collection. The secondary objective is to create proposal of categorization theoretical model. Methods: In this thesis we used the narrative interview with the elements of the interview and the interview method. Interview method was used in collecting data to create a theoretical model of categorization. The narrative interview method with the elements of the interview was used as the main method of collecting data on probands. Results: We found that the termination of adolescent competitive activity is caused by more reasons. One of the main reasons was the influence by parents (especially the father), the other reasons were performance decrease, lack of motivation, injury, bad training condition in the club. Categorization was split into five main groups: social, psychological, economic, training and health. The goal of this thesis is to describe the effective legislation of the cooperatives. Where appropriate this thesis compares the effective legislation to the previous legislation. The thesis tries to locate the problematic areas and to find solution to the given problems. This thesis is divided into 5 main chapters. The first chapter describes the history of cooperatives, because I think that without understanding the historic development no topic can be properly understood. Also this chapter describes the cooperative at a general level. The second chapter is formed of a general description of the membership in a cooperative with comparison to the previous legislation. This chapter covers also the basic principles of the membership in cooperative, as well as the content of the membership in cooperative. The third chapter describes the individual methods of the formation of the membership in cooperative pursuant to the effective legislation and where appropriate compares it to the previous legislation. Request for termination of life in Benelux countries from the perspective of civil law The master's thesis is about euthanasia which is known as one of the most controversial and discussed topic in the world of law and medicine, especially termination of life on request. The aim of this work is to innovatively interduce unique legislation of termination of life on request in Benelux countries from the perspective of civil law because I am convinced of its importance despite being overshadowed by the meaning of criminal perspective. In a first chapter the terminology, definition and theory dividing will be explained. The other terms which are connected with the things above are the content of this part as well. The human rights related to euthanasia are discussed in next part of this work. Benelux countries have their own euthanasia enactment - this is what third chapter is about. Next part shows in-depth analysis of due care criteria which are countained in second chapter of Termination of life on request and Assisted Suicide Act.
