Fats in Mind : Effects of Omega-3 Fatty Acids on Cognition and Behaviour in Childhood / Fats in Mind : Effekter av omega-3 fettsyror på kognition och beteende under barndomen

The aim of this thesis was to examine possible effects of omega-3 fatty acids on children’s cognition and behavior. Longitudinal as well as cross-sectional comparisons were made among children with typical development and children with ADHD /at risk developing ADHD. The specific purposes were to examine (1) breast-feeding in relation to cognition; (2) relation between long chain poly unsaturated fatty acids (LCPUFAs) in mothers breast-milk and children´s cognition; (3) effects of EPA supplementation on cognition and behavior in children with ADHD; (4) if LCPUFAs have differential effects on working memory, inhibition, problem-solving and theory of mind (ToM). The main conclusions were as follows; (1) duration of breast-feeding was positively correlated to children levels of intelligence (IQ); (2) LCPUFAs in breast-milk was related to children’s ToM and IQ, the quotient DHA/AA, together with length of breastfeeding and gestation week explained 76% of the variance of total IQ; (3) subtypes of children with ADHD responded to EPA supplementation with significant reductions in symptoms, but there were no effects in the whole group with ADHD; (4) ToM ability was related to LCPUFAs, but not to any other cognitive measures as working memory, inhibition and problem-solving. To conclude, these results indicate that fatty acid status in breast-milk at birth affect general cognitive function in children at 6.5 years of age, including ToM. Short-term intervention with omega-3 fatty acids does not affect cognition in children with ADHD, but improves clinical symptoms as assessed by means of teacher ratings. These results further indicate that hot executive function and social cognition may be an area of interest for future research. / Syftet med denna avhandling var att undersöka möjliga effekter av omega-3 fettsyror för barns kognition och beteende. Både kortsiktiga och långsiktiga effekter undersöktes och studierna involverade både barn med typisk utveckling och barn med ADHD. De specifika syftena var att undersöka; (1) amning i relation till barns kognitiva utveckling; (2), relationen mellan långa fleromättade fettsyror (LCPUFA) i mammans bröstmjölk och barns kognition; (3) effekter på kognition och beteenden hos barn med ADHD av ett EPA tillskott; (4) om LCPUFA hade några differentiella effekter på arbetsminne, inhibering eller Theory of Mind (ToM). De huvudsakliga slutsatserna var följande; (1) amningslängden var positivt korrelerad med barnens intelligenskvot (IK); (2) LCPUFA i bröstmjölken var associerad med barnens ToM och IK, kvoten DHA/AA, tillsammans med amningslängd och graviditetslängd förklarade 76% av variansen i totala IK; (3) subgrupper av barn med ADHD svarade på EPA behandlingen med signifikant reducerade symptom, men för hela gruppen med ADHD hittades inga effekter; (4) ToM var relaterat till LCPUFA, men inga andra kognitiva mått som arbetsminne, inhibering eller problemlösning. Sammanfattningsvis indikerar dessa resultat att fettsyrestatus i bröstmjölk påverkar generell kognitiv förmåga, samt ToM, hos barn 6,5 år gamla. Korttidsintervention med omega-3 fetter påverkar inte kognition hos skolbarn med ADHD, men minskar kliniska symptom skattade i lärarskattningsskalor. Vidare indikerar resultaten att ”heta” exekutiva funktioner och social kognition kan vara av intresse för framtida forskning.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The objective of this study was to evaluate carcass characteristics, the non carcass components, the lipid profile and the sensorial characteristics of meat from steers finished in feedlot fed diets containing different levels of inclusion of soybean soapstock. We used 30 Nellore and Charolais-Nellore crossbred steers with initial mean age of 20 months and initial mean weight of 20 months and 328.3 kg (±18.8 kg) were allotted to five dietary treatments: 0, 30, 60, 90 and 120 g of soybean soapstock/kg dry matter (DM) of diet . The diet consisted of forage(corn silage):concentrate ratio of 39:61 (DM basis), and was offered "ad libitum" during 84 days. A inclusão de até 120 g de borra de soja/kg de MS na dieta não alterou o peso de abate e os rendimentos de carcaça quente e fria, assim como os componentes teciduais músculo, gordura e osso. The inclusion up to 120 g of soybean soapstock/kg DM in the diet did not alter the slaughter weight and hot and cold carcass yields, as well as the muscle, fat and bone tissues. It also did not alter the empty body weight and yield, and hot and cold carcass yields adjusted for the empty body weight. The relative weight of the blood decreased with the addition of up to 55 g of soybean soapstock/kg DM in the diet, but the inclusion of up to 120g/kg DM in the diet promoted a linear increase in the relative weight of the lung and spleen, a linear reduction of the relative weight of the omasum and rumen fat. The kidney fat deposition, when adjusted for empty body weight, increased with the inclusion of up to 50 g of soybean soapstock/kg DM. The total weight of external components, vital, gastrointestinal and internal fat did not differ. With respect to meat quality, including between 83 to 91 g soybean soapstock/kg of diet DM promoted the highest levels of Vacenic (C18: 1 trans -11) and CLA (C18: 2 cis-9 trans-11) fatty acids, however, promoted also the highest levels of trans fats in the steers meat. The levels of fatty acids of the omega-3 group decreased linearly with the inclusion of dietary soybean soapstock , increasing the omega-6:omega-3 ratio in the steers meat. The lipid content increased linearly with the addition of soybean soapstock , but the content of cholesterol remained similar. The inclusion of dietary soybean soapstock did not affect meat marbling, color, palatability, juiciness, tenderness, and losses of the thawing and cooking liquid. / Objetivou-se nesse estudo avaliar as características da carcaça, dos componentes não integrantes da carcaça e do perfil lipídico e sensorial da carne de novilhos terminados em confinamento, alimentados com dietas contendo diferentes níveis de inclusão de borra de soja. Foram usados 30 novilhos Nelore e cruzas Charolês-Nelore com idade média inicial de 20 meses e peso médio inicial de 328,3 kg (±18,8 kg), distribuídos em cinco tratamentos alimentares: 0; 30; 60; 90 e 120 g de borra de soja/kg de matéria seca (MS) da dieta. A dieta foi constituída de volumoso (silagem de milho):concentrado com relação de 39:61 (com base na MS), e ofertada à vontade durante 84 dias. A inclusão de até 120 g de borra de soja/kg de MS na dieta não alterou o peso de abate e os rendimentos de carcaça quente e fria, assim como os componentes teciduais músculo, gordura e osso. A inclusão de até 120 g de borra de soja/kg de MS na dieta também não alterou o peso e rendimento de corpo vazio, e rendimentos de carcaça quente e fria, ajustada ao peso do corpo vazio. O peso relativo do sangue reduziu com a inclusão de até 55 g de borra de soja/kg de MS na dieta, porém a inclusão de até 120g/kg de MS na dieta promoveu aumento linear no peso relativo do pulmão e baço, redução linear no peso relativo do omaso e da gordura ruminal. A deposição de gordura renal, quando ajustada ao peso de corpo vazio, aumentou com a inclusão de até 50 g de borra de soja/ kg de MS. Os pesos totais dos componentes externos, vitais, gastrintestinais e das gorduras internas não diferiram. Com relação a qualidade da carne, a inclusão entre 83 a 91 g de borra de soja/kg de MS da dieta promoveu os maiores teores dos ácidos graxos Vacênico (C18:1 trans-11) e CLA (C18:2 cis-9 trans-11), entretanto, promoveu também os maiores teores de gorduras trans na carne dos novilhos. Os teores de ácidos graxos da família ômega-3 reduziu linearmente com a inclusão da borra de soja na dieta, refletindo com isso, no acréscimo da relação ômega-6:ômega-3 na carne de novilhos. O teor de lipídios aumentou linearmente com a inclusão da borra de soja, porém o de teor de colesterol se manteve semelhante. A inclusão da borra de soja na dieta não alterou a marmorização, cor, palatabilidade, suculência, maciez, e perdas de líquidos ao descongelamento e cocção da carne.

Importância da relação dos ácidos graxos omega-6/omega-3 na alimentação / Importance of the relation of the fatty acids Omega-6/Omega-3 in the feeding

O experimento foi realizado no Biotério Experimental da USP em Pirassununga - SP. Na primeira fase foram utilizados 42 coelhos machos em crescimento da raça Nova Zelândia mantidos individualmente durante todo o experimento. O ensaio biológico totalizou 150 dias. Na Fase 1 os animais receberam dietas com indução de hipercolesterolemia (0,5% de colesterol), com desequilíbrio na proporção de Omega-6/Omega-3 (n-6:n-3) de 15:1 e vice versa e balanço na proporção de n-6:n-3 de 4:1 (controle), totalizando 6 ensaios, os quais foram divididos aleatoriamente em 7 grupos de 6 coelhos cada. Óleos de girassol e de peixe foram utilizados como fontes de ácidos graxos poli-insaturados n-6 e n-3, respectivamente. Na Fase 2, os animais remanescentes continuaram recebendo as mesmas dietas, exceto no grupo B (hipercolesterolemia) que passou a receber o tratamento controle para verificar o efeito. No final da Fase 1, três animais de cada grupo foram eutanasiados com retirada da artéria aorta para determinação de placas lipídicas, histopatológica, colesterol total e imunohistoquímica para verificação da expressão da enzima LDL-receptor e determinação histopatológica do tecido hepático. Em soro foram realizadas determinações de perfil de ácidos graxos, colesterol total, LDL e HDL-colesterol e triacilgliceróis. Células endoteliais aórticas de coelhos foram isoladas para a realização do teste dose resposta com óleo de peixe para identificar a melhor relação do consumo destes ácidos graxos comparando com o experimento in vivo. O objetivo deste trabalho foi monitorar os efeitos do fornecimento de cada dieta sobre o teor de colesterol total, LDLcolesterol, HDL-colesterol e triacilgliceróis em sangue de coelhos. No geral, o perfil de ácidos graxos no soro correspondeu diretamente com a dieta consumida. Os principais efeitos dos ácidos graxos poli-insaturados Omega 3 foi a redução dos lipídios séricos quando os coelhos remanescentes da dieta hipercolesterolêmica (0,5% de colesterol e n-6:n-3 de 2:1) passaram a receber o equilíbrio da relação de ácidos graxos 4:1 de n-6:n-3. O teor de colesterol total no soro, artérias e o crescimento dos ateromas foram influenciados pelo elevado consumo de ácidos graxos n-6 e n-3 associados à adição de colesterol nas dietas. Por outro lado verificamos que o excesso de Omega 3 associado ou não ao colesterol contribuiu para o agravamento das placas ateroscleróticas inclusive deposição de cálcio nas mesmas e paredes endoteliais. Provavelmente, o excesso de n-3 tenha ocasionado efeito contrário às suas funções anti-inflamatória, antiagregatória e anti-trombótica. Dietas hipercolesterolêmicas provocaram esteatose hepática e o elevado consumo de Omega-6 em detrimento do baixo consumo de Omega -3 induziram hepatite crônica. Já o consumo elevado de Omega-3, sem colesterol, regrediu a esteatose hepática nos animais. O consumo equilibrado de n-6:n-3 reduziu a expressão da enzima LDL-receptor no grupo que anteriormente recebeu dieta hipercolesterolêmica. Ressaltando-se a importância do consumo equilibrado destes ácidos graxos já que esta enzima é controlada pelo colesterol livre circulante. No teste dose-resposta em células endoteliais da aorta de coelhos foi evidenciado que a melhor proporção de n-6:n-3 seria de 9 vezes mais do que a concentração de Omega 3 recomendada de 5:1. / The experiment was conducted at the Experimental Biotery by USP in Pirassununga - SP. In the first phase were used 42 male rabbits growing New Zealand breed individually maintained throughout the experiment. The biological assay amounted to 150 days. In phase 1, the animals received diets induced hypercholesterolemia (0.5% cholesterol) with imbalance in the ratio of Omega-6 / Omega-3 (n-6 / n-3) 15:1 and vice versa, and balance the ratio of n-6 / n-3 of 4:1 (control), totaling six tests, which were randomly divided into 7 groups of six rabbits each. Sunflower and fish oils were used as sources of fatty acids polyunsaturated n-6 and n-3, respectively. In Phase 2, the remaining animals continued to receive the same diets except in group B (hypercholesterolemia) who went on to receive the control treatment to check the effect. At the end of Phase 1, three animals from each group were sacrificed with the aorta artery removed for determination of lipid plaques, histopathology, immunohistochemistry and total cholesterol to verify the expression of LDL-receptor enzyme and determination of histopathological liver tissue. In serum profile were made determinations of fatty acids, total cholesterol, LDL and HDL cholesterol and triglycerides. Aortic endothelial cells of rabbits were isolated to perform the dose response test with fish oil to identify the best value for the consumption of these fatty acids compared to the in vivo experiment. The objective of this work was to monitor the effects of provision for each diet on total cholesterol, LDL-cholesterol, HDLcholesterol and triglycerides in the blood of rabbits. In general, the profile of fatty acids in serum corresponded directly with the diet consumed. The main effects of polyunsaturated Omega 3 fatty acids was the reduction of serum lipids when the remaining rabbits with hypercholesterolemic diet (0.5% cholesterol and n-6: n-3 2:1) have received the balance of the relationship fatty acid 4:1 n-6: n-3. The total cholesterol content in serum, the growth of arteries and atheroma were influenced by a high intake of n-6 fatty acids and n-3 associated with the addition of cholesterol in diets. On the other hand we verified that the excess of Omega 3 with or without cholesterol contributed to the aggravation of the atherosclerotic plaques including deposition of calcium in them and endothelial walls. Probably the excess of n-3 has caused the opposite effect to their anti-inflammatory, anti-thrombotic and anti-aggregatory functions. Hypercholesterolemic diets caused hepatic steatosis and high intake of Omega-6 to the detriment of low consumption of Omega -3 induced chronic hepatitis. High consumption of Omega-3, no cholesterol, decreased hepatic steatosis in animals. Highlighting the importance of the balanced intake of this fatty acids, since this enzyme is controlled by circulating free cholesterol. In dose-response test in endothelial cells of the rabbits aorta was evident that the best ratio of n-6: n-3, nine times more than the recommended concentration of Omega 3 to 5:1.

O objetivo foi investigar os principais efeitos das variáveis de processo como a pressão, a temperatura e o diâmetro médio de partícula (0,387, 0,595 e 0,842 mm) no rendimento da extração. O rendimento de extração aumentou com a diminuição do diâmetro de partícula, indicando a importância do efeito da difusão intra-partícula nas partículas maiores (0,842 mm). A extração supercrítica foi realizada a 30 e 40 MPa, temperaturas de 40, 50 e 60°C, utilizando 0,842 mm de diâmetro de partícula a uma vazão de 1,5L de CO2/min. O rendimento máximo de extração para as sementes trituradas foi de 92%, obtido a 40MPa, 60°C e 0,842 mm de diâmetro médio de partícula. Para extração por prensagem a frio, seguida da extração supercrítica a 40Mpa, 60ºC e 0,716 mm de diâmetro médio geométrico da partícula o rendimento total de extração foi de 99%. A solubilidade do óleo da semente de Sacha inchi em dióxido de carbono supercrítico (SC-CO2) foi medida nas temperaturas de 40 e 60 °C e pressões entre 10 e 40 MPa. Os valores de solubilidade mostraram um comportamento retrógrado, abaixo de 35 MPa a solubilidade diminuiu com o aumento da temperatura.A tendência observada acima desta pressão foi um aumento da solubilidade com o aumento da temperatura a pressão constante. O modelo matemático de Chrastil foi usado para correlacionar os valores experimentais de solubilidade. O conteúdo de óleo determinado com hexano em equipamentos soxhlet foi de 54,32% e o rendimento de óleo extraído por prensagem a frio foi de 46,8%. As características físicas e químicas do óleo foram obtidas. As composições em ácidos graxos do óleo obtido por SC-CO2 e do óleo obtido com hexano não apresentaram diferenças significativas. O principal ácido graxo foi o ácido alfa-linolênico (Ômega-3) aproximadamente 50,5%, seguido peloácido linoléico (34,12%) e ácido aléico (14%). O conteúdo de ácidos graxos livres (AGL) e de tocoferóis (tocoferol e tocotrienol) dos óleos obtidos por SC-CO2, solvente e prensagem a frio foram determinados. O conteúdo de tocoferóis so óleo extraído com SC-CO2 variou de 222,8 a 306,8 mg/100g, do óleo extraído por prensagem a frio foi 278,4 mg/100g e do óleo extraído com hexano foi 238,7 mg/100g. O teor de AGL no óleo extraído com hexano foi de 2,98% (medido como % de ác. Linolênico), ligeiramente menor do que aquele extraído por prensagem a frio (3,75%) e SC-CO2 (3,97-4,26%). O óleo de sementes de Sacha inchi pode ter grande potencial debtro do mercado crescente de óleos especiais / Abstract: The supercritical fluid extraction of oil from sacha inchi (plukenetia volubilis L.) seeds, using Co2 as a solvent, it is presented in this study. The objective was to investigate the main process parameters such as extraction pressure and temperature, mean particle size of (0,387, 0,595 and 0,842 mm) on the extraction yield. The extraction yield increased as the particle size decreased, indicating the importance of the effect of the intraparticle diffusion in larger particles (0,84 mm). The extraction was carried out at operating pressures of 30 and 40 MPa, operating temperatures of 40, 50 and 60°C, particle 0,84 mm and flow rate of CO2 1,5L/min. Maximum extraction yield was 92% obtained at 40 MPa, 60°C, 0.84 mm of particle sizes and 1.5 L of CO2/min. For the cold-pressed extraction, followed of supercritical extraction 40 MPa, 60ºC and 0,716 mm of geometric average diameter of the particle, the maximum extraction yield was 99%. The solubility of Sacha inchi seed oil in supercritical carbon dioxide (SC-CO2) were measured in the temperatures from 40 to 60 °C and pressures between 10 and 40 MPa. Under working conditions, the values of solubility showed retrograde behavior. Solubility, increase with pressure and temperature above the crossover pressure, which found between 30 and 40 MPa, and decreased with temperature below the crossover pressure. Mathematical modeling (Chrastil) of the solubility. The oil content was 54,32% and 46.80% determined bu solvent extraction and cold-pressed, respectively. Physical and chemical characteristics of the oil were obtained. The results in terms of fatty acid compositions of oil obtained with SC-CO2 were compared with those obtained when hexano was used as the solvent, and no significant differences between the oils extracted by both methods were found. The main fatty acid was alpha-linolênico acid (Ômega-3) making up 50.5%, followed by linoleic acid (34,12%) and oleic acid (14%). The level of FFA and tocol composition (tocoferol and tocotrienol) contents of the oils obtained by SC-CO2, solvent and cold-pressed extraction was determined. Tocol content of the SC-CO2 - extracted oil varied from 222.85 to 306.82 mg/100g, which was higher than that of hexane-extraced oil (238.72 mg/100g) and cold-pressed oil (278.4 mg/100g). The level of FFA in the solvent-extraced oils (2.98%) (% as ác. Linolenic) was lower than cold-pressed extraced oil (3.75%) and SC-CO2)-extraced oils (3.97-4.26%). EFEITOS DA SUPLEMENTAÇÃO COM ÁCIDOS GRAXOS OMEGA 3 NOS DISTÚRBIOS MOTORES E DISFUNÇÃO COGNITIVA DE PACIENTES TRATADOS COM ANTIPSICÓTICOS TÍPICOS

About 20 to 25 % of psychiatric patients treated with typical antipsychotics may develop an important movement disorder named tardive dyskinesia, with complex etiology and no effective treatment. Cognitive loss is also associated with antipsychotic treatment being harmful and often difficult to identify. Some studies have shown beneficial effects of Omega 3 polyunsaturated fatty acids on tardive dyskinesia and on cognition. Considering the available evidences, we conducted a randomized, double-blind, placebo-controlled, clinical trial, which evaluated the effect of fish oil supplementation rich in n-3 polyunsaturated fatty acids (3 g/dia) or placebo (3 capsules / day), for 12 weeks, on motor and cognitive disturbances of patients on use of neuroleptics. Two evaluations were performed, one before supplementation (baseline) and another at the end of it. For this, were used the following evaluation tools: Abnormal Involuntary Movement Scale for tardive dyskinesia and Mini-Mental State Examination for cognition. Peripheral blood samples were collected before supplementation (baseline) and 4, 8 and 12 weeks after, in order to monitor the supplementation effects on biochemical parameters- triglycerides (TG), fasting glucose, total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and on parameters of blood clotting- prothrombin time and partial thromboplastin time. The biological material collected was also used to run the Comet Assay in leukocytes with the aim of investigate the treatment effect on the DNA damage index. After 3 months of supplementation, we observed that fish oil caused an improvement of 18.5 % in tardive dyskinesia, while cognitive performance did not change when compared with the baseline assessment. Furthermore, lipid profile of the patients was not modified, fasting glucose was reduced in all assessments and prothrombin time increased in the last evaluation. In addition, fish oil did not show genotoxic effect (DNA damage index did not change). Therefore, we have demonstrated the therapeutic potential of fish oil rich in Omega 3 on tardive dyskinesia of patients treated with neuroleptics and its low risk of side effects. The increase of prothrombin time suggests a possible anticoagulant effect, requiring its monitoring during chronic treatment. / Cerca de 20 a 25 % dos pacientes psiquiátricos tratados com antipsicóticos típicos podem desenvolver uma importante síndrome motora denominada discinesia tardia (DT), de etiologia complexa e ainda sem tratamento efetivo. Perdas cognitivas também estão associadas ao uso desses medicamentos, sendo muitas vezes de difícil identificação e igualmente prejudiciais. Alguns estudos têm demonstrado efeitos benéficos dos ácidos graxos poliinsaturados Omega 3 (AGPI n-3) sobre a DT e sobre a cognição. Considerando as evidências disponíveis, realizamos este trabalho que consistiu de um ensaio clínico randomizado, duplo-cego, controlado por placebo. Avaliamos os efeitos da suplementação com de óleo de peixe rico em AGPI n-3 (3 g/dia) ou placebo (3 cápsulas/dia) sobre distúrbios motores e cognitivos de pacientes sob uso de neurolépticos, por um período de 12 semanas. Foram efetuadas duas avaliações, uma antes da suplementação (basal) e outra ao final da mesma. Para isso utilizamos as seguintes ferramentas de avaliação: Escala de Movimentos Involuntários Anormais (EMIA), para a DT e Mini-Exame do Estado Mental (MEEM), para a cognição. Amostras de sangue periférico foram colhidas antes da suplementação (basal) e 4, 8 e 12 semanas após, com o propósito de acompanhar seus os efeitos sobre parâmetros bioquímicos- triglicerídeos (TG), glicemia de jejum, colesterol total, colesterol ligado à lipoproteína de alta densidade (HDL-C) e colesterol ligado à lipoproteína de baixa densidade (LDL-C), bem como sobre parâmetros de coagulação sanguínea- tempo de protrombina (TP) e tempo de tromboplastina parcial (TTP). Com o material biológico colhido, também executamos o Ensaio Cometa em leucócitos, para investigar a influência do tratamento sobre o índice de dano no DNA. Após 3 meses de suplementação, observamos que o óleo de peixe produziu uma melhora de 18,5 % na DT, enquanto o desempenho cognitivo não sofreu alteração em relação à avaliação basal. Por sua vez, o perfil lipídico dos pacientes não foi modificado, a glicemia de jejum foi reduzida em todas as avaliações e o TP aumentou na última avaliação. Ainda, o índice de dano no DNA não foi alterado, mostrando que o óleo de peixe não apresentou efeito genotóxico. Com isso, demonstramos o potencial terapêutico do óleo de peixe rico em Omega 3 sobre a DT de pacientes tratados com neurolépticos e seu baixo risco de efeitos colaterais. O aumento do TP sugere um possível efeito anticoagulante, necessitando de acompanhamento durante tratamentos crônicos.

Le but du présent travail est d’évaluer le potentiel d’oméga-3 à induire la fonction endothélial, et à inhiber la contraction des artères coronaires en réponse aux plaquettes activées.L’induction des relaxations dépendantes de l’endothélium est affecté à la fois par le ratio et la pureté des formulations, avec un effet maximal obtenu pour la formulation EPA:DHA 6:1 à haut degré de pureté via une activation redox-sensible des voies de signalisation PI3-kinase/Akt et/ou MAPKs menant à l’activation de eNOS. La formulation EPA:DHA 6:1 inhibe les contractions induites par les plaquettes impliquant la sérotonine dans les artères coronaires de porc, et les contractions à la sérotonine dans les artères coronaires porcines et mammaires internes humaines, principalement par une augmentation de la formation endothéliale de NO. Ces résultats indiquent que la formulation optimisée EPA:DHA 6:1 exerce un effet bénéfique sur le système cardiovasculaire via l’activation de la fonction endothéliale. / The aim of the present work was to evaluate both the potency of omega-3 formulations to induce the endothelial function and the effect of omega-3 on the platelet-induced coronary artery contraction.The endothelium-dependent relaxations induced by EPA:DHA formulations is dependent on both the ratio and the purity of the formulation, with the maximal effect obtained with a highly purified EPA:DHA 6:1 ratio through a redox-sensitive activation of PI3-kinase/Akt and/or MAPKs pathways and subsequent activation of eNOS. The EPA:DHA 6:1 formulation inhibits the platelet-induced serotonin-mediated contraction of porcine coronary rings and the 5HT-induced contraction in both porcine coronary artery and human internal mammary artery rings, mainly due to an increased endothelial formation of NO. Taken together, the present findings indicate that the optimized EPA:DHA 6:1 formulation is able to exert potent beneficial cardiovascular effects through the activation of the endothelial function.

Omega-3-fettsyror stimulerar protein-anabolism och kan därför vara användbart för att förebygga och behandla sarkopeni. Syfte:Syftet med denna studie var att utvärdera effekten av omega-3-fettsyra-tillskott på graden avmuskelproteinsyntes hos vuxna. Metod: Studien är utförd som en litteraturstudie där relevantavetenskapliga och publicerade studier utvärderats och dess resultat har analyserats för attbesvara studiens syfte. Studiernas relevans bedömdes utifrån artiklarnas titel och abstrakt. 234studier i Onesearch, 5 studier i PubMed gicks igenom till en början. 30 abstrakt lästes, 21studier lästes i fulltext, 7 studier baserat på in- och exklusionskriterierna valdes ut till studienefter läst fulltext. Resultat: De flesta studier har utvärderat effekten av omega-3-tillskott hosyngre vuxna 18-30 år och äldre vuxna 65 år och äldre. Även om det finns en relativ brist påstudier som utvärderar effekten av omega-3-tillskott på muskelproteinsyntes (MPS) hos vuxna30-70 år, verkar det som om omega-3 kan främja anabola ökningar vid styrketräning hos äldreindivider. Slutsats: Ytterligare forskning behövs inom detta område för att dra relevantaslutsatser. Couclusion: Further research is needed in this area to draw relevant conclusions.This literature review concludes that omega-3 fatty acids stimulate the synthesis of muscleprotein in adults and may be useful to prevent and treat sarcopenia.

Monika Kuchařová, Ph.D. Title of Diploma Thesis: The determination of fatty acid levels in the tissues of healthy, suddenly deceased persons and polymorbid patients The aim of the thesis was to determine the levels of 14 fatty acids in tissues of suddenly deceased, otherwise healthy individuals and in the group of polymorbid, chronic patients. It was a comparison of fatty acid levels in seven tissues of the human body: subendocardial left ventricular tissue, liver parenchyma tissue, kidney cortex, adrenal tissue, skeletal muscle, abdominal subcutaneous adipose tissue, and brain tissue. Each group included 10 deceased patients. The theoretical part incudes fatty acids as the main component of lipids. It deals mainly with the group of polyunsaturated fatty acids and their relation to pathologies in the human body. Gas chromatography, thanks to which we analyzed the fatty acid spectrum, is also described. The experimental part consists of the basic characteristics of the research groups, the description of the workflow and the results.
