Apesar de seu potencial farmacológico, a designação do farmacógeno, como sendo as cascas do tronco, parte da planta que depende de um processo de regeneração após sua retirada, o que de acordo com a demanda pode ser visto como um risco ambiental à espécie, este estudo busca o reconhecimento laboratorial das folhas da espécie como droga, visando sua conservação e utilização racional. Este trabalho teve como objetivo estudar os aspectos farmacognósticos de folhas de S. terebinthifolius Raddi, de três localidades, visando contribuir com informações referentes ao controle de qualidade físico-químico da droga vegetal, bem como avaliar a atividade antimicrobiana e antiproliferativa in vitro de extratos, óleo essencial e frações obtidas de folhas de S. terebinthifolius. As análises do controle de qualidade foram realizadas segundo normas farmacopeicas e não farmacopeicas, a atividade antimicrobiana segundo normas do CLSI para microdiluição em placa e bioautografia e a atividade antiproliferativa pelo ensaio da sulforrodamina B. Microbial Aspects of Plant Invasion: An Analysis of Soil Microbial Communities Associated with the Invasive Plant - (Schinus terebinthifolius) Brazilian Pepper Tree Across South-Eastern Florida.

The Brazilian pepper tree (BP, Schinus terebinthifolius), introduced to the United States in the 1800s, has since become a category one invasive plant in Florida, aggressively spreading to 3000 km2 of prime habitat. There is a serious dearth of knowledge on whether the rhizobiome plays any roles in the displacement of native flora and the range expansion of BP. This thesis discusses the well-established plant invasion mechanisms of the BP and highlights key emerging mechanisms and gaps in (a) the current understanding of the molecular, below-ground processes of BP invasion and (b) studies on the potential role of microbial interactions in the success of BP invasion already established for other select invasive species, and the intervention of soil metagenomic studies to elucidate plant invasive mechanisms. Schinus terebinthifolius, popularly known as pepper tree is a kind of abundance in Pernambuco and due to their biological properties is widely used in folk medicine. This study aimed to determine the chemical constituents of essential oils of ripe fruits (OORF) and unripe (OOUF) mastic and evaluate the potential acaricide and insecticide through lethal and sublethal effects on Tetranychus urticae and Rhyzopherta dominica . Analyzed by GC / MS revealed the limonene as the main constituent in both oils (OOUF = 44,1 ± 1,3%; OORF = 31,8 ± 1,2%). The residual contact and fumigant action of these oils on T. uticae revealed OOUF the most active in fumigation (LC50 = 1,46 μL / L of air), while OEFM was more active in the tests of the contact petri dishes closed and opened petri dishes (LC50 μL/L/cm² = 3,04) and did not differ statiscally from eugenol. These oils exhibited significant repellent activity against T. urticae compared with eugenol. For R. dominica fumigation test showed no statistical difference between the two oils, but both were more effective in the exposure period of 72h. Limonene the main constituent of OOUF and OORF, showed the same level of toxicity compared to the essential oils. Tests with blends prepared with the major and minor constituents of these oils indicated that limonene is the major contributor to toxicity obtained for OOUF and OORF. The OOUF was repellent to 25, 35 and 45 μL/2g) and the oil of OORF proved attractive in the lower concentration tested (15 μL/2g) and repellent at the highest (30 and 50 μL/2g). / A busca por novos compostos biologicamente ativos aos artrópodes tem um papel importante no manejo integrado de pragas. Schinus terebinthifolius, conhecida popularmente como aroeira é uma espécie de grande abundância o estado de Pernambuco e, devido as suas propriedades químicas e biológicas, é amplamente utilizada na medicina popular. Este trabalho tem por objetivo determinar os constituintes químicos de óleos essenciais dos frutos maduros (OEFM) e imaturos (OEFI) de aroeira e indicar o potencial acaricida e inseticida, a partir dos efeitos letais e subletais, sobre Tetranychus urticae Koch (Acari: Tetranychidae) e Rhyzopherta dominica Fabr. (Coleoptera: Bostrichidae). A análise realizada por GC/EM revelou o limoneno como constituinte majoritário em ambos os óleos (OEFI = 44,1 ± 1,3%; OEFM = 31,8 ± 1,2%). A ação fumigante e contato residual desses óleos sobre T. urticae revelou que o OEFI foi mais ativo por fumigação (LC50 = 1,46 μL/L de ar), enquanto que o OEFM foi mais ativo nos testes de contato com placas fechadas do que placas abertas (LC50 = 3,04 μL/L/cm2) e não diferiram estatisticamente do eugenol. Estes óleos exibiram significante atividade repelente a T. urticae comparados com o eugenol. Para R. dominica no teste de fumigação não houve diferença estatística entre os óleos, mas ambos se mostraram mais eficazes no período de exposição de 72h. O limoneno, principal constituinte dos OEFI e OEFM, apresentou o mesmo nível de toxicidade quando comparado aos óleos essenciais. Testes com blendas preparadas com os constituintes principais e minoritários desses óleos indicaram que o limoneno foi o composto que mais contribuiu para a toxicidade obtida para o OEFI e o OEFM. O OEFI foi repelente a 25, 35 e 45 μL/2g) e o OEFM se mostrou atraente na menor concentração (15 μL/2g) e repelente nas mais altas (30 e 50 μL/2g).

Relação entre parâmetros ecofisiológicos e a produção de óleo essencial em espécies arbóreas / Relationship between ecophysiologycal parameters and essential oil production in tree species

The jamun fruit (Syzygium cumini (L.) Skeels) and the rose pepper (Schinus terebinthifolius Raddi) are species of multiple aptitudes with a high oil potential. Its essential oil produced by leaves, fruits and stems show a large variety of pharmacological properties. Evaluation of chlorophyll a fluorescence emission and monitoring of gas exchanges mechanisms may provide valuable data for plant functionality evaluation. Thus, the ecophysiological evaluation of these species, was conducted aiming to relate it with essential oil production. First, the content of chlorophyll a, b and total, chlorophyll a fluorescence, and oil essential productivity of the species was analyzed, extracted in the dry season. Subsequently, the liquid photosynthesis assimilate rate, stomatal conductance, transpiration, vapor pressure deficit, air and leaf temperature, internal carbon and fresh leaves oil productivity, in dry and rainy season were evaluated. For the ecophysiological parameters first studied, jamun fruit had higher values, in companson with rose pepper. As for the essential oil produced by the two species, rose pepper showed the best yield for fresh and dry leaves. It was not verified any direct relation between the photosynthetic apparatus efficiency and essential oil production to the two species studied. The results found in the following study pointed to significant differences in all variables and time influence in the studied species, in both seasons. In the rainy season, a different tendency in the species evaluated according to the dry season was found. In the highest humidity season were verified the highest net photosynthesis rates (A), stomatal conductance (Gs) and transpiration (E), for rose pepper and jamun fruit trees. In this same season, the highest contents of internal carbon (Ci) were verified. The essential oil production in the two species, generally, did not vary seasonally. This variation was only seen for dry rose pepper leaves. with a higher productivity in the rainy season. As the previous study, it was not possible to correlate the ecophysiological parameters studied with the secondary metabolites production / O jamelão (Syzygium cumini (L.) Skeels) e a aroeira (Schinus terebinthifolius Raddi) são espécies de múltiplas aptidões com grande potencial olerífero. Seus óleos essenciais produzidos em folhas, frutos e hastes apresentam propriedades farmacológicas diversas. Técnicas como medição da emissão de fluorescência da clorofila a e o monitoramento dos mecanismos de trocas gasosas podem fornecer dados imprescindíveispara avaliação da funcionalidade das plantas. Portanto, nesse trabalho objetivou-se a avaliação ecofisiológica destas espécies, a fim de relacioná-la com a produção de óleos essenciais. Num primeiro momento, foram avaliados os teores de clorofila a, b e total, fluorescência da clorofila a,e o rendimento de óleo essencial das espécies estudadas, extraído no período de seca. No seguinte, foram avaliados: taxa assimilatória de fotossíntese líquida, condutância estomática, déficit de pressão de vapor, temperaturas do ar e da folha, carbono interno e rendimento de óleo de folhas frescas e secas, nos períodos seco e chuvoso. Para os parâmetros ecofisiológicos primeiramente estudados, as plantas de jamelão apresentaram os maiores valores, enquanto os da aroeira foram estatisticamente menores. Quanto ao óleo essencial produzido pelas duas espécies, a aroeira apresentou o melhor rendimento para folhas secas e frescas. Não foi verificada relação direta entre a eficiência do aparato fotossintético e a produção de óleo essencial para as duas espécies estudadas. Os resultados encontrados no estudo seguinte, apontaram diferenças significativas em todas as variáveis e influência do horário das medições nas respostas das espécies estudadas, nas duas épocas. Pode-se verificar, no período de chuvas, uma tendência diferenciada das espécies em relação ao período seco. No período de maior umidade verificaram-se as maiores taxas de fotossíntese líquida (A), condutância estomática (Gs) e transpiração (E) para aroeira e jamelão. Nesse mesmo período, foram verificados os maiores teores de carbono interno (Ci). A produção de óleo essencial nas duas espécies, de modo geral, não apresentou variações devido a sazonalidade. Essa variação somente foi observada em folhas secas de aroeira, com rendimento muito superior ao observado no período chuvoso. Assim como no estudo anterior, não se pode correlacionar os parâmetros ecofisiológicos estudados com a produção de metabólitos secundários.

No entanto, na literatura encontram-se poucos estudos sobre o seu processamento, em que a secagem é uma etapa indispensável e que ainda é realizada de forma empírica. Nas empresas beneficiadoras, a etapa de secagem tem por finalidade aumentar a vida de prateleira do produto, além de agregar valor aos frutos da aroeira-vermelha. Neste contexto, este trabalho teve por objetivo realizar investigações no processo de secagem de frutos de aroeira-vermelha, focando principalmente no estudo de modelos matemáticos para a simulação do processo. As amostras de frutos de aroeira-vermelha, utilizadas nos ensaios de caracterização e secagem, foram classificadas manualmente e reumidificadas artificialmente. Para a caracterização do sistema particulado, os seguintes parâmetros foram determinados: teor de umidade em base seca; diâmetro médio e massa específica aparente dos frutos; e massa específica e porosidade do leito de frutos. Também foram determinadas, através do método estático, as isotermas de dessorção de umidade dos frutos em temperaturas de 30, 50 e 70 ºC. Um Delineamento Composto Central Rotacional (DCCR) com 11 experimentos foi desenvolvido e foram investigadas as influências da temperatura (40 a 70 ºC) e da velocidade do ar de secagem (0,4 a 0,8 m/s) no processo. Em cada um dos ensaios do DCCR foram analisadas a cinética de secagem e a curva de taxas de secagem dos frutos de aroeira-vermelha. Os modelos de difusão, de Lewis, de Henderson e Pabis, de Page modificado e de Henderson foram ajustados aos dados experimentais da cinética de secagem, sendo realizada a comparação destes modelos com um modelo fundamentado na Inteligência Artificial. Os resultados revelaram que, para as condições operacionais estudadas neste trabalho, a temperatura do ar de secagem apresenta uma influência maior no processo. A cinética de secagem ocorre principalmente no período de taxa decrescente. Isto indica que a secagem ocorre pela remoção da umidade interna do sólido. O modelo via Redes Neurais Artificiais (RNA's) e o modelo de Henderson apresentaram os resultados mais satisfatórios no ajuste aos dados da cinética de secagem dos frutos de aroeira-vermelha. No entanto, observou-se que a principal vantagem do modelo de RNA, em relação aos modelos clássicos, foi a sua generalidade. O modelo de RNA foi capaz de descrever uma série de experiências com precisão, enquanto que a aplicação de modelos clássicos foi limitada a uma experiência específica. Para o modelo neural, esta gama de condições experimentais pode ser expandida através da adição de novos conjuntos de experimentos de treinamento. Deste modo, os resultados satisfatórios da utilização de um modelo baseado em RNA's em processos de secagem indicam a potencialidade da aplicação desta ferramenta na modelagem de processos industriais de secagem / Abstract: The pink pepper, produced from the Brazilian pepper-tree fruits (Schinus terebinthifolius Raddi), is one of the most sophisticated condiments of the international cuisine. The demand for pink pepper is increasing because it adds taste and refinement to international cuisine. Until now, rare are the studies that investigated the drying of Brazilian pepper?tree fruits. In the processing companies, the process is carried out empirically and the drying operation is an important step. The Brazilian pepper?tree fruits are dried to inhibit microbial development and quality decay. Another benefit of drying is a reduction in weight. In this context, this study aimed to perform investigations in the drying process of Brazilian pepper?tree fruits, focusing mainly in the study of mathematical models to simulate drying kinetics. Before each test, the fruits were sorted manually and humidified artificially. In this study, physical characterization of Brazilian pepper?tree fruits was obtained by evaluating the moisture content on dry basis; mean diameter and apparent density; bulk density and porosity. The moisture sorption isotherm, at temperatures of 30, 50 and 70 ºC, were determined by the static method. A Central Composite Rotable Design (CCRD) with 11 experiments was developed. In each experiment, the drying kinetics and the drying rate were investigated, as well as the influence of input variables on the drying process. Classical models were fitted to the drying data, as well as a model based on Artificial Intelligence. In the operating conditions of this study, the results revealed that the air temperature has a greater influence on the thin layer drying process of Brazilian pepper?tree fruits. The drying kinetics occurs mainly in the falling-rate period of drying. This indicates that drying occurs mainly by removal of internal moisture of the solid. The artificial neural network model and the Henderson model fitted to the experimental data more accurately in comparison to the various mathematical models. However, it was observed that the main advantage of the artificial neural network model over classical models was their generality. The ANN model was able to describe a range of experiments accurately, while the application of classical models is limited to a specific experiment. This study aimed to evaluate the antibacterial activity of the hydroalcoholic extract and essential oil from the leaves and fruits of Schinus terebinthifolius Raddi at two growth stages against bacteria related to bovine mastitis, besides testing a plant extract with antibacterial action, to be used antisepsis in milking. The thesis is presented in two papers. The first refers to the in vitro antibacterial activity of hydroalcoholic extract and essential oil from leaves and fruits at two growth stages, tested by serial microdilution plates against bovine mastitis bacteria. The results were expressed as Minimal Bactericidal Concentration. All extracts showed antibacterial activity. The second paper shows results for the in vivo experiment, which tested the hydroalcoholic extract of the leaves of the plant before and after dipping for twelve consecutive weeks, was used as control iodine commercial, weekly data were collected regarding clinical mastitis, subclinical and new intramammary infections, and biweekly health data sphincter and skin from the ceiling. None of the indices analyzed apresentraram difference between treatments, suggesting that the plant extract can be used to teat antisepsis as a replacement of conventional products. In our study we have obtained satisfactory results from the use of this plant, it has antibacterial activity against bacteria in bovine mastitis and teat disinfection has similar results to the conventional product. / O interesse científico pela fitoterapia levou ao desenvolvimento de pesquisas baseadas nas práticas populares. Este trabalho teve como objetivo avaliar a atividade antibacteriana do extrato hidroalcoólico e óleo essencial das folhas e frutos de Schinus terebinthifolius Raddi em dois estádios fenológicos frente a bactérias relacionadas a mastite bovina, além de testar um extrato da planta, com ação antibacteriana, para ser utilizado na antissepsia da ordenha. A dissertação está apresentada em dois artigos científicos. O primeiro, refere-se a atividade antibacteriana in vitro do extrato hidroalcoólico e óleo essencial das folhas e frutos em dois estádios fenológicos, testados pela técnica de microdiluição serial em placas contra bactérias da mastite bovina. Os resultados foram expressos pela Concentração Bactericida Mínima. Todos os extratos apresentaram atividade antibacteriana. O segundo trabalho demonstra resultados referentes ao experimento in vivo, onde foi testado o extrato hidroalcoólico das folhas da planta no pré e pós dipping, durante doze semanas consecutivas, como controle se utilizou iodo comercial, foram coletados dados semanais referentes a mastite clínica, subclínica, e novas infecções intramamárias, e dados quinzenais da saúde do esfíncter e da pele do teto. Nenhum dos índices analisados apresentraram diferença estatísticas entre os tratamentos, sugerindo que o extrato da planta pode ser usado para antissepsia dos tetos como substituinte dos produtos convencionais. Em nosso estudo obtivemos resultados satisfatórios da utilização desta planta, ela apresenta ação antibacteriana frente as bactérias da mastite bovina e na desinfecção dos tetos tem resultados semelhantes aos do produto convencional.
