A influência da salinidade nos processos de tratamento de efluentes por lodos ativados. / The influence of salinity in wastewater treatment processes by activated sludge.

O processo de tratamento de efluentes por lodos ativados é um dos processos de tratamento mais difundido em todo o mundo, devido principalmente a qualidade do efluente obtido. Entretanto, trata-se de um processo biológico dependente da atividade bacteriana para a estabilização da matéria orgânica proveniente dos esgotos e com isso, se faz necessária a manutenção das condições ideais para a sobrevivência e proliferação das bactérias e dos outros microrganismos envolvidos neste processo. Efluentes salinos causam um grande distúrbio na atividade celular dos microrganismos presentes neste processo. A análise biológica do lodo e a taxa de consumo de oxigênio são testes rápidos, práticos, baratos e sem geração de resíduos químicos e que foram adotados neste trabalho para acompanhar a eficiência do processo. Esses dois parâmetros são amplamente utilizados em pesquisas, porém ainda é sub-utilizado para efetivo monitoramento de Estações de Tratamento de Esgotos. Este presente trabalho investigou a influência da salinidade nos processos de lodos ativados através de parâmetros que levam em consideração a atividade metabólica dos organismos aeróbios e através do monitoramento da comunidade de protozoários indicadores biológicos do lodo ativado, os testes de respirometria e os testes da análise biológica do lodo, respectivamente. Os resultados da atividade metabólica são apresentados em forma de gráficos, em termos de taxa de consumo de oxigênio específico e porcentagem de inibição. Os resultados da qualidade biológica são apresentados em números de 0 a 10 de acordo com o Índice de Madoni (1994). Os resultados demonstraram a imediata intoxicação dos lodos ativados em concentrações de sal a partir da concentração mínima utilizada neste trabalho, que foi de 5 gL de NaCl. Para entender melhor o processo de intoxicação foi realizado experimentos de 96 horas de monitoramento após o choque de cloreto de sódio nos reatores com lodos ativados, os resultados não demonstraram melhoria no processo. Os resultados sugerem que os testes de taxa de consumo de oxigênio em consonância com os testes da qualidade biológica são eficientes e complementares para a avaliação da influência da salinidade no processo de lodos ativados. / Effluent treatment process by sludge activated is one of the most used processes of treatment all over the world, mainly because of the quality of the final effluent. However, this process is a biological one, and so, it depends on the bacterial activity to the stabilization of the organic matter of the sewage. Considering this, it is necessary to maintain the ideal conditions to the survival and proliferation of the bacteria responsible for the process. This work considered two parameters to evaluate the influence of the salinity in the sludge activated processes; these parameters are widely used in research, however, little used for the monitoring of the Wastewater Treatment Plants. The objectives of this work are to investigate the influence of the salinity through the respirometry test and the biological quality of the sludge. The results of the metabolic activity are presented in graphics, in terms of Oxygen Uptake Rate and percentage of inhibition. The results of the biological quality are presented in numbers by 0 to 10 according to the Mandonis Index (1994). The results show the intoxication of the sludge activated in saline concentrations higher than 5 g/L of NaCl in the saline shock, and it seems that efficiency do not get better even after 96 hours. The results suggests that the respirometry test and the biological quality test are efficient and complementary to the evaluation of salinity in the sludge activated processes.

Phytoremediation of soil contaminated with petroleum hydrocarbons and trace elements

The rapid urbanization and industrialization has led to an increase of disposal petroleum hydrocarbons (PHC) and trace elements (TE) into the environment. These pollutants are considered as the most toxic contaminants in the world due to their persistence in the environment, and the long range of toxicological effects for living beings when their concentrations exceed critical thresholds. Recent concerns regarding the environmental contamination have initiated the development of several remediation technologies, including physico-chemical, biological and Dig and Dump approaches. In my thesis, gentle soil remediation options (GRO) were investigated at different scales for the reclamation of PHC and TE co-contaminated soil. In the first part of my thesis, laboratory experiments were performed to characterize PHC and TE contaminated soil as well as the indigenous microorganisms (bacteria and fungi) present in these contaminated soils. It was found that the studied aged contaminated soil had a negative effect on earthworm’s development and Lepidium sativum biomass. Moreover, a high respiration of microorganisms attributed to the transformation/ mineralization of organic matter or/and organic pollutants was observed. This presence of viable microorganisms suggested an adaptation of microorganisms to the contaminant. Further results showed that the long-term exposure of soil microorganisms to high PHC concentration and the type of isolation culture media did not influence the ability of isolates to effectively degrade PHC. However, phylogenic affiliation had a strong effect on PHC biodegradation. In the second part of my thesis, preliminary studies in greenhouse trials were performed to investigate the ability of Medicago sativa assisted by compost in the greenhouse aided-phytoremediation of PHC and TE. The results clearly showed that compost amendment into the soil promoted PHC degradation, M. sativa growth and survival, and phytoextraction of TE. Residual risk assessment after the phytoremediation trial also showed a positive effect of compost amendment on plant growth and earthworm development. Pilot-scale ecopile experiment carried out in the third part of this thesis allow a reduction of up to 80% of PHC and 20% of metals after 17 months. My thesis showed that alfalfa (M. sativa) and sunflower (Helianthus annus) plants were suitable for phytodegradation of PHC and phytoextraction of TE. The outcomes of my thesis can be extend to other plants and they bring a new level of understanding that can be helpful for further full-scale phytoremediation studies. / L'urbanisation rapide et les activités industrielles ont abouti à la contamination de l’environnement par les hydrocarbures pétroliers (HP) et les éléments traces (ET). Ces composés sont particulièrement toxiques en raison de leur persistance dans l'environnement, et de leurs effets toxicologiques sur les êtres vivants quand les concentrations de ceux-ci dépassent des seuils critiques. Les préoccupations de plus en plus croissantes sur la contamination de l'environnement ont favorisé le développement de plusieurs technologies de remédiation des sites contaminés par les approches biologiques, physico-chimiques et par l’excavation et l’entreposage. Dans cette thèse, des options douces d'assainissement des sols (ODA) ont été utilisées à différentes échelles pour la remédiation des sols contaminés par des mélanges des HP et des ET. Dans la première partie de ma thèse, des expériences en laboratoire ont été effectuées dans le but de caractériser les sols contaminés et les micro-organismes autochtones (bactéries et champignons) qu’ils contiennent. Malgré la contamination ancienne du sol, les résultats obtenus montrent des effets négatifs des contaminants sur le développement des lombrics et la biomasse de Lepidium sativum. En outre, une respiration élevée de microorganismes, attribuée à la transformation / minéralisation de la matière organique et / ou des polluants organiques a été observée. Cette présence de micro-organismes viables dans les sols contaminés suggère leur adaptation aux contaminants. Toutefois, d'autres résultats ont montré que l'exposition à long terme des microorganismes du sol à de fortes concentrations en HP et le type de milieu de culture utilisé pour l'isolation n'influencent pas la capacité des isolats microbiens à dégrader efficacement les HP. Cette capacité de biodégradation des HP est liée à la phylogénie des microorganismes. Dans la deuxième partie de cette thèse, les études préliminaires en serre ont été réalisées dans le but d’évaluer l’efficacité de phytoremédiation en utilisant Medicago sativa assistée par l’ajout du compost. Les résultats ont montré dans cette expérience que l’ajout du compost dans le sol favorise la dégradation des HP, la croissance et la survie de M. sativa, ainsi que la phytoextraction des ET. L’évaluation des risques résiduels après la phytoremédiation a également montré un effet positif de l'amendement du sol en compost sur la croissance des plantes et le développement des lombrics. L’expérience pilote réalisée sur le terrain dans la troisième partie de ma thèse a permis une réduction de 80% des HP et de 20% des ET après 17 mois. Ma thèse a démontré que la luzerne (M. sativa) et le tournesol (Helianthus annus) sont des choix judicieux de plantes pour la phytodégradation des HP et pour la phytoextraction des ET. Les résultats qui en résultent sont utiles pour d’autres études de phytoremédiation à grande échelle. / Den snabba urbaniseringen och industrialiseringen har lett till en ökning av petroleumkolväten (PHC) och olika spårämnen (TE) i miljön. Dessa föroreningar anses vara de mest giftiga föroreningarna i världen på grund av att de stannar kvar i miljön samt att de har toxikologisk påverkan på levande varelser. På senare tid har oron för dessa miljöföroreningar lett till utvecklingen av flera saneringstekniker, såsom fysiska, kemiska och biologiska metoder. I denna avhandling undersöktes enkla marksaneringsalternativ (GRO) på olika nivåer, för återvinning av PHC och TE från förorenad jord. I den första delen av denna avhandling, utfördes laboratorieförsök för att karakterisera PHC- och TE-förorenad jord samt av de inhemska mikroorganismerna (bakterier och svampar) som förekommer i dessa förorenade jordar. Det konstaterades att den studerade förorenade jorden hade en negativ inverkan på daggmaskars utveckling och biomassan av L. sativum. Dessutom kunde den höga respirationen bland mikroorganismerna tillskrivas omvandlingen och mineraliseringen av organiskt material och/eller de organiska föroreningar som observerades. Denna närvaro av livsdugliga mikroorganismer antydde att mikroorganismerna anpassat sig till föroreningssituationen på platsen. Ytterligare resultat visade dock att den långvariga exponeringen av höga PHC-koncentrationer i isolerade odlingsmedier, för mikroorganismer i jorden, inte påverkade förmågan för dessa att effektivt bryta ned PHC. Dock hade den fylogenetiska tillhörigheten en stark påverkan på bionedbrytning av PHC. I den andra delen av denna avhandling genomfördes preliminära studier i växthus där förmågan hos M. sativa undersöktes, med hjälp av kompost, gällande den växthusstödda fytosaneringen av PHC och TE. Resultaten visade att inblanding av kompost i jorden främjade nedbrytningen av PHC, tillväxten och överlevnadsgraden av M. sativa och fytoextraktion av Pb. Återstående riskbedömning efter fytosaneringen visade också en positiv effekt, när komposten användes, på växternas tillväxt och daggmaskarnas utveckling. Ett experiment med eco-bädd utfördes för den tredje delen av avhandlingen. Denna studie visade på en minskning på upp till 80% av PHC och 20% av metallerna. Denna avhandling visar att M. sativa och H. annus var lämpliga för nedbrytning av PHC och fytoextraktion av Pb och Cu. Resultaten från denna avhandling förväntas vara användbara för ytterligare studier av fytoremediering i fullskala.

Effects of Hypoxia and Exercise on In Vivo Lactate Kinetics and Expression of Monocarboxylate Transporters in Rainbow Trout

The current understanding of lactate metabolism in fish is based almost entirely on interpretation of concentration measurements that cannot be used to infer changes in flux. Moreover, the transporters regulating these fluxes have never been characterized in rainbow trout. My goals were: (1) to quantify lactate fluxes in rainbow trout under normoxic resting conditions, during acute hypoxia, and exercise by continuous infusion of [U-14C] lactate; (2) to determine lactate uptake capacity of trout tissues by infusing exogenous lactate in fish rest and during graded exercise, and (3) to clone monocarboxylate transporters (MCTs) and determine the effects of exhausting exercise on their expression. Such information could prove important to understand the mechanisms underlying the classic “lactate retention” seen in trout white muscle after intense exercise. In normoxic resting fish, the rates of appearance (Ra) and disappearance (Rd) of lactate were always matched (~18 to 13 µmol kg-1 min-1), thereby maintaining a low baseline blood lactate concentration (~0.8 mM). In hypoxic fish, Ra lactate increased from baseline to 36.5 µmol kg-1 min-1, and was accompanied by an unexpected 52% increase in Rd reaching 30.3 µmol kg-1 min-1, accounting for a rise in blood lactate to 8.9 mM. In exercising fish, lactate flux was stimulated > 2.4 body lengths per second (BL s-1). As the fish reached critical swimming speed (Ucrit), Ra lactate was more stimulated (+67% to 40.4 μmol kg-1 min-1) than Rd (+41% to 34.7 μmol kg-1 min-1), causing an increase in blood lactate to 5.1mM. Fish infused with exogenous lactate stimulated Rd lactate by 300% (14 to 56 μmol kg-1 min-1) during graded exercise, whereas the Rd in resting fish increased by only 90% (21 to 40 µmol kg-1 min-1). Four MCT isoforms were partially cloned and characterized in rainbow trout: MCT1b was the most abundant in heart, and red muscle, but poorly expressed in gill and brain where MCT1a and MCT2 were prevalent. MCT4 was more expressed in the heart. Transcript levels of MCT2 (+260%; brain), MCT1a (+90%; heart) and MCT1b (+50%; heart) were stimulated by exhausting exercise. This study shows that: (i) the increase in Rd lactate plays a strategic role in reducing the lactate load imposed on the circulation. Without this response, blood lactate accumulation would double; (ii) a high capacity for lactate disposal in rainbow trout tissues is elicited by the increased blood-to-tissue lactate gradient when extra lactate is administered; and (iii) rainbow trout may be unable to release large lactate loads rapidly from white muscle after exhausting exercise (lactate retention) because they poorly express MCT4 in white muscle and fail to upregulate its expression during exercise.

Vliv biodegradace bioplastů na kvalitu půdy / Influence of bioplastics´ biodegradation on soil quality

V poslední době se pozornost polečnosti obrátila k mikroplastům. Jsou produkovány různými odvětvími a šíří se napříč prostředím. Po dlouhou dobu byly považovány za inertní, bez dalšího vlivu na rostliny a jiné živé organismy, avšak jak zjistily nedávné studie, mohly by představovat vážnou hrozbu. Několik vědců, včetně nás, se proto začalo soustředit na jejich transport a transformace v životním prostředí. Většina se však zaměřuje pouze na jejich přítomnost v mořských a sladkých vodách, a proto jejich chování ve vzduchu a půdě zůstává nejasné. Kromě toho byla pozornost soustředěna i na bioplasty. Jsou prezentována jako ekologická alternativa, která má vyřešit všechny dosud zmíněné problémy (a další). Avšak často se zapomíná, že jejich hlavní výhoda může být zároveň nevýhodou. Z tohoto důvodu se tato diplomová práce zaměřuje na negativní účinky spojené s přítomností mikroplastů (konkrétně bioplastu poly-(R)-3-hydroxybutyrátu) v půdě jako jejich běžný receptor. Kombinovali jsme respirometrii, elementární analýzu, termogravimetrii a enzymatické testy, abychom zkoumali fyzikálně-chemické změny v půdě vyvolané přítomností bioplastu. Naše výsledky ukázaly negativní vliv na půdní organickou hmotu a zadržování vody v půdě. V tomto smyslu byl zkoumán i tzv. "priming effect", jelikož docházelo k urychlení a také zpomalení rozkladu půdní organické hmoty. Zaznamenali jsme rozdílný vliv vybraných koncentrací biopolymeru na půdu a také vliv půdních vlastností na průběh degradace. V neposlední řadě zvýšení enzymatické aktivity jasně naznačovalo vliv přítomnosti biopolymeru na mikrobiální komunitu. Na základě takových zjištění jsme došli k závěru, že přidání biopolymeru vede k dlouhodobému dopadu na řadu funkcí půdního ekosystému.

The Metabolic Physiology of Planarian Flatworms

Using a high throughput closed respirometry method to measure oxygen consumption, I determined metabolic rates in asexual and sexual Schmidtea mediterranea and Girardia dorotocephala, as a function of temperature, taxon, stressors, reproductive mode, age, regeneration, and specific dynamic action. This study has shown that oxygen consumption can reliably be measured in planaria using optode closed respirometry, and also provided a reliable method for measuring wet mass in planaria, which has been a challenge to researchers in the past. This research revealed that oxygen consumption in S. mediterranea is 1.5-2.1X greater in the sexual strain over the asexual strain at 13-18°C. Within the sexual strain, oxygen consumption is 1.5 -2.2X greater in sexually mature adults over the sexually immature groups (hatchlings, juveniles, and regenerating sexuals). Furthermore, I was able to quantify differences in sexual morphology between these groups exhibiting significant differences in oxygen consumption. The results of this research supports a theory of higher metabolic costs with sexual maturity in S. mediterranea. Therefore, this study has established sexual and asexual S. mediterranea as simple, yet attractive models for investigating energetic costs between sexual and asexual phenotypes. This research also provided quantitative values for specific dynamic action in planaria, with a maximum increase in oxygen consumption of 160% induced by feeding, as well as metabolic relationships in planaria involving temperature, age, and regeneration. These values establish planaria as one of the simplest animal models in which common metabolic patterns, such as SDA and poikilothermic temperature sensitivity, have been demonstrated. Therefore, this research has contributed to the overall knowledge of the basic physiology in this animal, providing the framework for future metabolic studies in planaria involving environmental factors, reproduction, regeneration, development, and aging. Information from this study may supplement interpretation and understanding of modern cellular, molecular, and genomic studies in planaria.

Influence of seasonally variable hypoxia on epibenthic communities in a coastal ecosystem, British Columbia, Canada

Natural cycles of environmental variability and long-term deoxygenation in the ocean impose oxygen deficiency (hypoxia) on marine communities. My research exploits a naturally occurring hypoxia cycle in Saanich Inlet, British Columbia, Canada where I combined spatial surveys with remotely operated vehicles, ecological time-series from the subsea cabled observatory VENUS, and lab-based respirometry experiments to examine the influence of seasonally variable oxygen conditions on epibenthic communities. In situ oxygen thresholds established for dozens of fish and invertebrate species in this system show they naturally occur in lower oxygen levels than what general lethal and sublethal thresholds would predict. Expansion of hypoxic waters induced a loss of community structure which was previously characterized by disjunct distributions among species. Communities in variable hypoxia also have scale-dependent structure across a range of time scales but are primarily synchronized to a seasonal oscillation between two phases. Time-series revealed timing of diurnal movement in the slender sole Lyopsetta exilis and reproductive behavior of squat lobster Munida quadrispina in the hypoxia cycle. Hypoxia-induced mortality of sessile species slowed the rate of community recovery after deoxygenation. The 10-year oxygen time-series from VENUS, revealed a significant increase in the annual low-oxygen period in Saanich Inlet and that deoxygenation has occurred in this system since 2006. Differences in the critical oxygen thresholds (O2crit) and standard metabolic rates of key species (spot prawn Pandalus platyceros, slender sole, and squat lobster) determined the lowest in situ oxygen at which populations occurred and explained disproportionate shifts in distributions and community respiration. Finally, a meta-analysis on global O2crit reported for crustaceans showed that hypoxia tolerance differs among major ocean basins. Long-term trends of deoxygenation suggest a future regime shift may occur when the duration at which a system remains below critical oxygen levels exceeds the time needed for communities to recover. Species-specific traits will determine the critical threshold and the nature of the community response in systems influenced by variable states of oxygen deficiency. Réponse métabolique du saumon Atlantique (Salmo salar) aux fluctuations journalières de température : rôles de la température d’acclimatation et de l’historique thermique

En général, le métabolisme des poissons est estimé à des valeurs de température constantes, mais les effets de fluctuations journalières de température similaires à celles retrouvées en milieu naturel semblent peu connus. Les objectifs du présent mémoire sont de quantifier les effets de la température moyenne d’acclimatation et d’évaluer les effets de l’historique thermique des individus, sur les réponses métaboliques de tacons de saumon Atlantique (Salmo salar) aux fluctuations journalières de la température. Des tacons provenant de deux rivières, une fraîche et une chaude, ont été acclimatés à un maximum de quatre régimes thermiques (constant 15 °C ou 20 °C, fluctuant 15 °C ± 2.5 °C ou 20 °C ± 2.5 °C) et leur taux métabolique standard estimés par respirométrie par débit-intermittent. Les fluctuations journalières de température (15 °C ± 2.5 °C) près de l’optimum thermique pour cette espèce (16 °C) n’affectent pas le taux métabolique standard. À l’opposé, les fluctuations journalières de température plus chaudes (20 °C ± 2.5 °C) augmentent de 35.4% le taux métabolique standard des tacons de la rivière plus chaude, mais pas ceux des poissons de la rivière fraîche. Ainsi, la température moyenne à laquelle sont acclimatés les poissons peut affecter leur réponse métabolique aux fluctuations journalières de température, mais cette réponse peut varier entre populations provenant de rivières présentant des régimes thermiques différents. Enfin, grâce aux données de métabolisme précédemment estimées, un modèle de métabolisme standard a été développé pour des tacons de saumon Atlantique soumis à des fluctuations journalières de température. / Usually, fish metabolism is evaluated under constant values of temperature, but the effects of daily temperature fluctuations similar to those found in natural environment on metabolism seems to be much less understood. The goals of this study are to quantify the effects of the mean acclimation temperature and to assess the effects of the thermal history on the metabolic response of Atlantic salmon (Salmo salar) parr to daily fluctuations of water temperature. Atlantic salmon parr originating from two rivers, one cool and one relatively warm, were acclimated to up to four thermal regimes (constant 15 °C or 20 °C, daily fluctuating 15 °C ± 2.5 °C or 20 °C ± 2.5 °C) and their standard metabolic rate was estimated using intermittent-flow respirometry. Daily temperature fluctuations (15 °C ± 2.5 °C) near this species’ thermal optimum (16 °C) do not influence standard metabolic rate. In contrast, daily fluctuations of higher temperature (20 °C ± 2.5 °C) do increase standard metabolic rate by 35.4% for fish from the warmer river, but not for fish from the cooler river. Therefore, the mean temperature to which fish are acclimated may affect their responses to daily fluctuations of water temperature, but this response may vary between populations originating from rivers having different thermal regimes. Using standard metabolic rates previously estimated, a standard metabolic rate model was developed for Atlantic salmon parr exposed to daily fluctuations of temperature.

Intermittent hypoxia elicits a unique physiological coping strategy in Fundulus killifish

Fish encounter daily cycles of hypoxia in the wild, but the physiological strategies for coping with repeated cycles of normoxia and hypoxia (intermittent hypoxia) are poorly understood. Contrastingly, the physiological strategies for coping with continuous (constant) exposure to hypoxia have been studied extensively in fish. The main objective of this thesis was to understand how Fundulus killifish cope with a diurnal cycle of intermittent hypoxia, an ecologically relevant pattern of aquatic hypoxia in the natural environment. To do this, I characterized the effects of intermittent hypoxia on hypoxia tolerance, oxygen transport, metabolism, and the oxidative stress defense system of killifish, and compared these effects to fish exposed to normoxia, a single cycle of hypoxia-normoxia, and constant hypoxia. Specifically, I studied the following topics: (i) how acclimation to intermittent hypoxia modifies hypoxia tolerance, and the hypoxia acclimation response of Fundulus heteroclitus (Chapter 2), (ii) metabolic adjustments occurring during a hypoxia-reoxygenation cycle (Chapter 3), (iii) how acclimation to intermittent hypoxia alters O2 transport capacity and maximal aerobic metabolic rate (Chapter 4), (iv) the effects of hypoxia and reoxygenation on reactive oxygen species and oxidative stress (Chapter 5), and (v) variation in hypoxia tolerance and in the hypoxia acclimation responses across Fundulus fishes (Chapter 6). Killifish rely on a unique and effective physiological strategy to cope with intermittent hypoxia, and that this strategy is distinct from both the response to a single bout of acute hypoxia-reoxygenation (12 h hypoxia followed by 6 h reoxygenation) and to chronic exposure to constant hypoxia (24 h hypoxia per day for 28 d). Key features of the acclimation response to intermittent hypoxia include (i) maintenance of resting O2 consumption rate in hypoxia followed by a substantial increase in O2 consumption rate during recovery in normoxia, (ii) reversible increases in blood O2 carrying capacity during hypoxia bouts, (iii) minimal recruitment of anaerobic metabolism during hypoxia bouts, and (iv) protection of tissues from oxidative damage despite alterations in the homeostasis of reactive oxygen species and cellular redox status. Of these features, (i) is unique to intermittent hypoxia, (ii) also occurs in fish exposed to acute hypoxia-reoxygenation, and (iii) and (iv) are observed in both fish acclimated to intermittent hypoxia as well as those acclimated to constant hypoxia. This is the most extensive investigation to date on how fish cope with the energetic and oxidative stress challenges of intermittent hypoxia, and how these responses differ from constant hypoxia. This thesis adds substantial insight into the general mechanisms by which animals can respond to an ecologically important but poorly understood feature of the aquatic environment. / Dissertation / Doctor of Philosophy (PhD) / Oxygen levels in the aquatic environment are dynamic. Many fishes routinely encounter changes in oxygen content in their environment. However, we have very little understanding of how cycles between periods of low oxygen (hypoxia) and periods of high oxygen (normoxia) affect the physiology of fish. This thesis investigated how Fundulus killifish cope with daily cycles between hypoxia and normoxia (intermittent hypoxia) by modifying oxygen transport, metabolism, and oxidative stress defense systems. I found that killifish rely on a unique and effective physiological strategy to cope with intermittent hypoxia, and that this strategy is distinct from how they respond to a single bout of hypoxia (followed by normoxia) and to a constant pattern of only hypoxia. This is the most extensive investigation to date on how fish respond to the challenges of intermittent hypoxia, an understudied but ecologically important type of aquatic hypoxia.

Behavioral Activity and Hypoxia Tolerance of African Weakly Electric Fish

In dieser Arbeit wurden die Morpho-Physiologie und das Verhalten zweier Arten Afrikanischer schwach elektrischer Fische, Marcusenius victoriae und Petrocephalus degeni, im Labor und in einem ihrer natürlichen Habitate im Lwamunda Sumpf in Uganda untersucht. Die zwei Hauptziele dieser Arbeit waren (i) tageszeitabhängige Verhaltensrhythmen (Aktivität, Habitatnutzung) im Labor und im Freiland zu untersuchen und (ii) die Ausprägung und Plastizität der morpho-physiologischen Merkmale von P. degeni zu untersuchen, die ihnen erlauben bei natürlich vorkommender, geringer Sauerstoffverfügbarkeit (Hypoxie) zu überleben. Tageszeitabhängige Verhaltensrhythmen beider Arten wurden im Labor über 42 Stunden und im natürlichen Habitat dieser Fische für sechs Tage erfasst. In den Laborversuchen verbrachten beide Arten tagsüber annähernd 100% der Zeit in einem bereitgestellten Versteck und schwammen nachts heraus um aktiv ihre Umwelt zu erkunden. Im Habitat wurden die meisten Fische in strukturell komplexen Habitaten unter schwimmenden Pflanzen detektiert. Nachts schwammen die Fische aktiv in die offenen und ungeschützten Bereiche der Lagune, vermutlich um nach Futter zu suchen und zu interagieren. Die Begleitende in-situ Messung der Sauerstoffverfügbarkeit zeigte, dass beide Arten präsent und vermutlich sogar am aktivsten waren während Phasen extremer nächtlicher Hypoxie. Zur Untersuchung der respiratorischen Merkmale von P. degeni wurden Respirometrieversuche mit hypoxie-akklimatisierten Tieren durchgeführt, Hämoglobin- und Laktatkonzentration im Blut gemessen, und morphologische Parameter an den ersten beiden Kiemenbögen erfasst. Die Fische zeigten niedrige Sauerstoffverbrauchsraten, welche sie bis zu einem sehr niedrigem äußeren Sauerstoffpartialdruck aufrechterhielten. Zusätzlich zeigten sie hohe Hämoglobin- und Laktatkonzentrationen im Blut. Bis zu 75 Tage Normoxie-Akklimatisierung führte zu reduzierter Hämoglobinkonzentration und kürzeren Kiemenfilamenten. / In this thesis, I investigated the morpho-physiology and behavior of two species of African mormyrid weakly electric fish, Marcusenius victoriae and Petrocephalus degeni, in the laboratory and in one of their natural habitats, the Lwamunda Swamp in Uganda. The two main objectives of this work were to (i) observe behavioral rhythms and habitat use patterns of both species under natural and laboratory conditions, and (ii) assess expression and plasticity of morpho-physiological traits that might enable P. degeni to survive naturally occurring low oxygen conditions (hypoxia). Behavioral rhythms were recorded in the laboratory over 42 hours and in the habitat on six sampling days. In the laboratory, both species spent close to 100% of the time in their shelter during the day and actively explore their environment at night. In the swamp lagoon, fish were most often encountered in structurally complex habitats under floating vegetation and ventured into open and unsheltered areas of the lagoon at night, presumably to forage and interact. Concomitant in-situ oxygen measurements revealed that these fish were present, and presumably most active during periods of extreme nocturnal hypoxia in their swamp habitat. To investigate respiratory traits of swamp-dwelling P. degeni, I conducted respirometry experiments and measured blood lactate and hemoglobin and gill morphometrics on the first two gill arches. Fish showed low routine oxygen consumption rates, which they maintained until a very low ambient oxygen partial pressure was reached. Additionally, they had high concentrations of hemoglobin and lactate in their blood. Up to 75 days of normoxia exposure reduced blood hemoglobin and gill filament length.
