Approche métabolomique pour la compréhension des mécanismes de résistance à Fusarium graminearum et accumulation de trichothécènes chez le maïs / Metabolomic approach to understand resistance mecanisms to Fusarium graminearum and trichothecenes accumulation

Parmi les nombreux pathogènes fongiques susceptibles d’infecter les épis de maïs, les espèces appartenant au genre Fusarium sont particulièrement préoccupantes pour la filière maïsicole. La fusariose est susceptible d’induire des pertes de rendement considérables et est fréquemment associée à une contamination des épis par des mycotoxines. Un des leviers prometteur repose sur la sélection génétique de plantes résistantes à Fusarium et à l’accumulation de mycotoxines. Plusieurs Quantitative Trait Loci (QTL) ont été identifiés d’après la caractérisation moléculaire de la résistance à la fusariose chez le maïs. Cependant malgré les progrès des approches génétiques, les mécanismes moléculaires impliqués restent en grande partie inconnus. L’identification de métabolites majeurs associés à la résistance reste donc indispensable pour la création d’un outil d’aide à la sélection variétale. Une approche métabolomique combinant de la spectrométrie de masse et de la 1H-RMN a été mise au point pour identifier un ensemble de métabolites de défense, constitutifs ou induits par l’infection, susceptibles d’intervenir dans la résistance à Fusarium. Cette approche a été appliquée aux grains à deux stades de développement sur 20 variétés présentant des degrés de résistance contrastés inoculés ou non avec une souche de Fusarium graminearum toxinogène par le canal des soies. Les résultats obtenus mettent en évidence un panel de métabolites liés à la résistance ou la sensibilité des variétés de maïs. / Fusarium graminearum is the main causal agent of maize ear rot or Gibberella ear rot (GER), an important fungal disease affecting maize. GER leads to significant economic loss and serious health issues due to the ability of F. graminearum to produce mycotoxins such as type B trichothecenes. One promising approach to control Giberella Ear Rot and reduce mycotoxins contamination is to promote host-genetic resistance. Several Quantitative Trait Loci (QTLs) have been identified in maize. However molecular basis to resistance to Fusarium infection remains largely unknown and the success of selection for GER resistance is still challenging. Biochemical approaches can provide valuable insights in the mechanisms crops employ against F. graminearum and its production of mycotoxins. A biochemical profiling could actually be an efficient way to decipher plant-pathogen interactions and progress in screening resistant maize lines. This study aims to elucidate the metabolic profiling of F. graminearum resistance and toxin accumulation in kernels toward the combination of high resolution mass spectrometry and 1H NMR to identify a large set of metabolites, preformed, constitutive as well as inducible defense metabolites that could play a key role in GER resistance. This approach was applied to kernels harvested at two developmental stages. Twenty genotypes with contrasting levels of resistance were inoculated, or not, with a toxigenic Fusarium graminearum strain through the silk channel. Through analysis of 1H HR-MAS NMR and multivariate analysis was possible to propose a method to predict the sex of the plant still young, by assessing the metabolic profile of fresh leaves. This analysis led to the identification of regions of spectrum with higher influence on differentiation between males and females, although the corresponding metabolites have not been elucidated. / Araucaria angustifolia é uma planta dióica, que se encontra ameaçada de extinção devido à sua extensiva exploração e às dificuldades de reflorestamento, relacionadas ao seu lento desenvolvimento e à inexistência de métodos para a determinação do sexo da planta antes de sua idade reprodutiva (15 a 20 anos). Através de análises de RMN HR-MAS de 1H e análise multivariada foi possível propor um método para a predição do sexo da planta ainda jovem, através da avaliação do perfil metabólico de folhas in natura. Esta análise levou à identificação de regiões do espectro com maior influência na diferenciação entre machos e fêmeas, embora os metabólitos correspondentes não tenham sido elucidados.

Estudo da magnetoeletrólise durante o acoplamento RMN-Eletroquímica in situ / Magnetoelectrolysis study during the NMR-Electrochemistry in situ coupling

Recentemente foi demonstrado que a técnica de ressonância magnética nuclear no domínio do tempo (RMN-DT) é uma importante ferramenta analítica que é passível de ser acoplada com a eletroquímica (EQ-RMN). No entanto, como foi demonstrado nesse presente trabalho, a técnica de RMN não é passiva, ou seja, ela atua sobre as reações eletroquímicas aumentando a velocidade das reações realizadas in situ quando essas são limitadas por transporte de massas e/ou transferência de cargas. Essa alteração da taxa de reação é ocasionada por um fenômeno conhecido por magnetoeletrólise, que tem como principal resultante a força de Lorentz, que é o produto vetorial entre o campo magnético e o fluxo de íons gerado durante a eletrólise. O efeito do campo magnético sobre diferentes sistemas eletroquímicos foi comprovado na presença de campo magnético de equipamentos de RMN de baixa e alta resolução. Além do foco dado para a magnetoeletrólise durante o acoplamento EQ-RMN, novas células eletroquímicas foram miniaturizadas para a utilização com a RMN a partir de um método simples, rápido e robusto. Também foram feitas medições de velocimetria utilizando a técnica de imagem por spin eco com as quais foi possível visualizar e quantificar o efeito do campo magnético atuando sobre a reação de eletrodeposição de cobre realizada in situ. Esse trabalho foi o primeiro a utilizar o campo magnético do espectrômetro de RMN para estudar a magnetoeletrólise. Além disso, esse trabalho de doutorado foi o primeiro a utilizar um espectrômetro de RMN de bancada de alta resolução para o acoplamento com a eletroquímica. / Recently it was demonstrated that time domain Magnetic Nuclear Resonance (TD-NMR) is an important analytical technique which can be coupled to electrochemistry (EC-NMR). However, it was demonstrated in this work that NMR is not a passive technique, in other words it affects the electrochemical reactions performed in situ by increasing the reaction rate of mass transport and/or charge transfer limited reactions. This change in the reaction rate is caused by a phenomenon known as magnetoeletrolysis, it has as main resultant the Lorentz force, which is the vectorial product between the magnetic field vector and the ion flow produced during the electrolysis. The magnetic field effect on different electrochemical systems has been shown in the presence of magnetic fields of low and high resolution NMR spectrometers. In addition to the focus given to magnetoelectrolysis during the EC-NMR coupling, novel electrochemical cells were miniaturized for coupling with NMR by using a simple, rapid and robust method. Velocimetry measurements were also made using the spin-echo imaging technique with which it was possible to visualize and quantify the effect of the magnetic field acting on the in situ copper electrodeposition reaction. This work was the first to use the magnetic field of the NMR spectrometer to study magnetoelectrolysis. In addition, this doctoral thesis was the first to use a bench-top high resolution NMR spectrometer for coupling with electrochemistry.

Análises de parâmetros físicos e químicos por ressonância magnética nuclear em baixo campo / Analysis of physical and chemical parameters by low field nuclear magnetic resonance

A RMN em baixo campo (RMN-BC) é uma importante ferramenta analítica que vem sendo usada para substituir os métodos clássicos de análise. As principais características da RMN-BC sobre esses métodos são: rapidez da análise, pouca ou nenhuma preparação da amostra, análise não-invasiva e não-destrutiva e não tem descarte de produtos químicos. O presente trabalho avaliou o uso da RMN-BC com sensor (ímã e sonda) unilateral (RMNU). Neste equipamento de baixo custo, a região de análise é projetada para frente do imã, o que permite analisar amostras com grande volume, diferente dos espectrômetros convencionais onde o tamanho da amostra é limitado pelo gap ou bore do ímã. Como o sensor RMNU possui um intenso gradiente de campo magnético é possível investigar o comportamento da amostra em diferentes alturas. Essa propriedade foi explorada para medir a temperatura e a difusividade térmica de sementes de macadâmia em três profundidades. Para isso fez-se uma curva de calibração com o tempo de relaxação transversal (T2) do óleo da semente versus a temperatura. Os resultados obtidos com a RMNU foram coerentes com encontrados na literatura usando sensores invasivos como termopares. Também foram realizadas análises de viscosidade de óleos lubrificantes automotivos dentro das embalagens utilizando a sequência de CPMG. Com o método foi possível discriminar se o óleo base é mineral ou sintético. O sensor RMNU não foi sensível para identificar amostras de diesel com diferentes composição e origem. Para isso usou-se espectrômetro de RMN convencional onde analisou 118 blendas de diesel/biodiesel/óleo vegetal, em que se variou a refinaria, teor de enxofre do diesel (S500 e S10), a origem do biodiesel e do óleo. Os modelos de classificação de análise multivariada apresentaram alta capacidade de distinguir as respectivas blendas. Investigou-se também o uso do acoplamento indutivo para ampliar a região de detecção do sensor a partir da sua superfície. O acoplamento permitiu aumentar da região sensível de 2 mm para 20 mm. Esse sistema foi usado para monitorar a cura de uma amostra de gesso utilizando a sequência de CPMG. / The Low-Field NMR (LF-NMR) is an important analytical tool that has been used to replace the classical methods of analysis. The main features of LF-NMR about these methods are: speed of analysis, little or no sample preparation, non-invasive and non-destructive analysis and no disposal of chemicals. This study evaluated the use of LF-NMR with sensor (magnet and probe) unilateral (UNMR). In this low cost equipment, the region of analysis is projected forward of the magnet, which allows analyzing samples in large volumes, unlike conventional spectrometers where the sample size is limited by the gap or bore of the magnet. As UNMR sensor has an intense magnetic field gradient is possible to investigate the behavior of the sample at different heights. This property has been exploited to measure the temperature and the thermal diffusivity Macadamia seed at three depths. For this became a calibration curve with the transverse relaxation time (T2) of the seed\'s oil versus temperature. The results obtained with the UNMR were consistent with the one found in literature, using invasive sensors such as thermocouples. They were also held viscosity analysis of automotive lubricants in the packaging using the CPMG sequence. With the method it was possible to discriminate whether the base oil is mineral or synthetic. The UNMR sensor was not sensitive enough to diesel samples with different composition and origin. To it was used conventional LF-NMR spectrometer, where 118 blends diesel / biodiesel / vegetable oil, as it the refinery varied, sulfur diesel (S500 and S10), the source of biodiesel and oil were analyzed. The multivariate classification models showed high ability to distinguish their blends. It was also investigated using inductive coupling to expand the sensor detection region from its surface. Coupling allowed to increase the sensitive region of 2 mm to 20 mm. This system was used to monitor the cure of plaster sample using the CPMG sequence.

O trabalho apresentado nesta Tese tem como objetivo a elaboração de estruturas polinucleares baseadas em clusters trinucleares de rutênio. Os oligômeros estudados são opções interessantes para o desenvolvimento futuro de dispositivos supramoleculares, em função de suas propriedades fotofísicas e eletroquímicas. Um trímero e uma série de dímeros mistos foram obtidos pela combinação do cluster [Ru3O(CH3COO)6(py)2(CH3OH)]+ com complexos [Ru(bpy)2(P)(L)]+ (P = ligante de ponte N-heterocíclico; L = P, Cl ou CN). A caracterização por RMN, voltametria cíclica e espectroeletroquímica dos dímeros onde L = Cl foi utilizada como base na interpretação dos resultados obtidos para os demais complexos; os dados refletem uma interação fraca entre os fragmentos, que depende das características eletrônicas da ponte P. Nos dímeros onde L = CN, ocorre a supressão da emissão do cromóforo [Ru(bpy)2] pelo cluster através de um mecanismo de transferência de elétrons fotoinduzida. Para o cluster na forma reduzida, sugere-se também a ocorrência de transferência de energia. Na segunda parte da Tese, são apresentados sistemas policlusters contendo seis e dez unidades ligadas em ponte. As novas supermoléculas apresentam uma série de processos redox multieletrônicos e, neste caso, a comunicação eletrônica entre os centros metálicos é mais pronunciada, sendo dependente do estado de oxidação dos clusters. / The aim of the work reported is to develop polynuclear structures based on trinuclear ruthenium clusters. All the oligomers are seen as interesting options to the future development of supramolecular devices, due to their photophysical and electrochemical properties. A series of mixed dimers and one trimer were obtained by the combination of the [Ru3O(CH3COO)6(py)2(CH3OH)]+ cluster with [Ru(bpy)2(P)(L)]+ complexes (P = N-heterocyclic bridging ligand; L = P, Cl or CN). The NMR, cyclic voltammetry and spectroelectrochemical characterization of the dimers where L = Cl were used as a base to interpret the results collected for the other complexes; the data reflect a weak interaction between the units, depending on the electronic characteristics of the bridging ligand P. In the dimers where L = CN, occurs the quenching of the [Ru(bpy)2] emission by the cluster through a photoinduced electron transfer mechanism. For the cluster in its reduced form, it is suggested that an energy transfer process also occurs. On the second part of the Thesis, polycluster systems containing six and ten bridged cluster units are reported. The novel supermolecules present a series of multielectronic redox processes and, in this case, electronic communication among the metallic centers is more pronounced, depending on the clusters oxidation state.

Avaliação da matéria orgânica de solos tratados com lodo de esgoto: uso de ferramentas quantitativas e espectroscópicas / Evaluation of organic matter from soils treated with sewage sludge: use of quantitative and spectroscopic tools

A reciclagem agrícola do lodo de esgoto devidamente tratado é uma boa opção, pois transforma o resíduo orgânico em um importante insumo agrícola. Entretanto, a aplicação do lodo de esgoto pode alterar a quantidade e a estabilidade da matéria orgânica do solo. Com a presente pesquisa, objetivou-se avaliar os efeitos de doses de lodo de esgoto na matéria orgânica de dois solos (um Latossolo Vermelho distrófico sob floresta de eucalipto e um Latossolo Vermelho distroférrico sob cultivo de milho) por meio de técnicas quantitativas e espectroscópicas. O experimento I, sob cultivo de milho, foi instalado em Jaguariúna-SP, constituiu-se de 5 doses acumuladas de lodo de esgoto da ETE de Barueri, aplicadas de 1999 a 2005: 0; 30; 60; 120 e 240 t ha-1 (em base seca). O experimento II, sob floresta de eucalipto, foi instalado em dezembro de 2004, em área comercial da empresa da Suzano Papel e Celulose, em Angatuba - SP. Aplicou-se, uma única vez, 4 doses de lodo de esgoto da ETE de Jundiaí (0; 7,7; 15,4 e 23,1 t ha-1 em base seca). Nas amostras de solo foram determinados os conteúdos de carbono, por meio de análise elementar (CHN), e então, calculou-se o grau de humificação da matéria orgânica utilizando a Fluorescência Induzida por Laser (FIL). Com base nesses resultados, foram selecionadas algumas amostras para o fracionamento químico da matéria orgânica do solo e, por meio de técnicas espectroscópicas (FTIR, UV-vis, RMN e Fluorescência de UV-vis), foi realizado um estudo aprofundado da influência do lodo de esgoto nas substâncias húmicas do solo, bem como, em seu estágio de humificação. Pode-se verificar um aumento na concentração de carbono no solo com a aplicação do lodo de esgoto em ambos experimentos, bem como um aumento da alifaticidade e, conseqüentemente, diminuição da aromaticidade das substâncias húmicas, resultado da incorporação de matéria orgânica fresca, não compostada / Agricultural recycling of sewage sludge treated properly is a good option, since it transforms organic waste into a major agricultural input. However, the application of sewage sludge can alter the amount and stability of soil organic matter. The present work aimed to evaluate the effects of sewage sludge levels on organic matter from two different soils (an Oxisol under eucalyptus forest and an Oxisol under corn) through quantitative and spectroscopy techniques. The experiment, under corn, was installed in Jaguariuna-SP, consisted of 5 accumulated levels of sewage sludge from ETE Barueri, implemented from 1999 to 2005: 0, 30, 60, 120 and 240 t ha-1. Experiment II, in eucalyptus forest, was installed in December 2004, the commercial company Suzano Papel e Celulose, in Angatuba - SP. Applied only once, 4 doses of sewage sludge from Seawater Sewage Treatment from Jundiai (0, 7.7, 15.4 and 23.1 t ha-1 on a dry basis). Soil samples were determined carbon content by means of elemental analysis (CHN), and then calculated the degree of humification of organic matter using Laser Induced Fluorescence (LIF). Based on these results, we selected some samples for chemical fractionation of soil organic matter, and by means of spectroscopic techniques (FTIR, UV-vis, NMR and Fluorescence UV-vis), was conducted a detailed study of the influence of sludge sewage humic substances on the ground as well as on their stage of humification. One can observe an increase in the amount of carbon in the soil application of sewage sludge in both experiments, as well as an increase in aliphaticity and consequently decreased aromaticity of humic substances, resulting from the incorporation of fresh organic matter, noncomposted

Conventional methods demand a previous treatment which may be time consuming. At present, Nuclear Magnetic Resonance may be employed for the analysis of mixtures by using the DOSY (diffusion ordered spectroscopy) technique, whereby the different components of a mixture are separated by their diffusion coefficients. Pitches, in general, are products which are employed as starting material for the production of a variety of carbon materials, such as anodes for the production of aluminum, graphite electrodes for the steel industry, and carbon fibers. Pitches obtained from coal tar, itself a residue in the production of coke, are the most traditionally used. However, it is a carcinogen and therefore has had its use greatly diminished. Petroleum pitches, which are produced from decanted oil, may be a safer and less expensive alternative. The decanted oil is a heavy fraction obtained from catalytic cracking of petroleum. The difficulties in controlling its processing are derived from its complex composition, which includes paraffins, aromatic and heteroaromatic compounds. The present work investigates the analysis by NMR-DOSY of a mixture of model compounds present in decanted oil. Thus, a mixture of Tetracosane, Phenanthrene, 9-Mehylanthracene, Pyrene, Carbazole, 9-metil Carbazol, Dibenzothiophene, Dibenzofuran, Hexadecane, 1-Methylnaphthalene, Thiophene, Tetrahydrofuran was analyzed by NMR with an Avance III 500 MHz NMR spectrometer equipped with a BBO probe with a gradient on z-axis, at 20?C. To help in the separation by diffusion, the following polymers were used: poly-(ethyleneglycol), poly-(vinylpyrrolidone), poly-(dimethylsiloxan), poly-(butylmethacrylate) e poly-(vinylacetate).The shift reagent Eu(fod)3 was also employed. The results from DOSY-INEPT experiments show that poly-(butylmethacrylate) is the matrix which afforded the best separation of the different components of the mixture, followed by poly-(vinylacetate), poly-(ethyleneglycol) and finally, poly-(vinylpyrrolidone), which showed satisfactory results / Um grande desafio, muitas das vezes at? comum nos laborat?rios, ? a an?lise de misturas. Piches obtidos a partir do alcatr?o da hulha, res?duo da produ??o de coque metal?rgico, s?o os tradicionalmente mais usados, mas apresentam alto potencial carcinog?nico e tem demanda decrescente em fun??o de novas tecnologias empregadas nas coquerias. Piches de petr?leo, produzidos a partir de ?leos decantados, podem potencialmente constituir uma alternativa mais segura e com maior disponibilidade. O ?leo decantado ? uma fra??o pesada obtida nas unidades de craqueamento catal?tico fluido no refino de petr?leo, e o controle de seu processamento ? dificultado pela complexidade de sua composi??o, que inclui compostos paraf?nicos, homoarom?ticos e heteroarom?ticos. O presente trabalho se prop?em a investigar a an?lise por RMN de uma mistura de padr?es de subst?ncias representativas dos tipos de compostos presentes no ?leo decantado. Assim, foram escolhidos padr?es de Tetracosano, Fenantreno, 9-metil Antraceno, Pireno, Carbazol, 9-metil Carbazol, Dibenzotiofeno, Dibenzofurano, Hexadecano, 1-metil Naftaleno, Tiofeno, Tetrahidrofurano. Todos os experimentos foram realizados no laborat?rio de RMN do PPGQ-UFRRJ a 20?C com o espectr?metro de RMN de 500 MHz Bruker Avance III, equipado com sonda BBO, gradiente no eixo z. Para auxiliar na separa??o dos componentes da mistura foram usados o pol?mero poli-(etilenoglicol), poli-(vinilpirrolidona), poli-(dimetilsiloxano), poli-(metacrilato de butila) e poli-(acetato de vinila), al?m do reagente de deslocamento Eu(fod)3. Os resultados obtidos a partir dos experimentos de DOSY-INEPT mostraram que o pol?mero PMAB foi a matriz que mais auxiliou na separa??o dos componentes da mistura em quest?o no presente trabalho, sendo seguido pelas matrizes PAV, PEG e PVP, que tamb?m apresentaram resultados satisfat?rios

Jaburetox is a peptide derived from a urease isoform of the Canavalia ensiformis plant. Structurally, jaburetox is an intrinsically disordered peptide, demonstrated by bioinformatics tools, and experimentally, by circular dichroism (CD) and nuclear magnetic resonance (NMR). Several biological properties of jaburetox have already been described, such as insecticidal activity, ability to interact with lipid bilayers, fungitoxic effect, among others. Soyuretox is a peptide colinear to jaburetox derived from the soybean ubiquitous urease. One of the objectives of this thesis was to carry out structural studies of the soyuretox peptide by CD and NMR, which revealed its intrinsically disordered nature and that its secondary structure content is modified in the presence of SDS micelles. Biological properties of soyuretox were evaluated. The peptide has entomotoxic activity, inducing aggregation of hemocytes from Rhodnius prolixus, the vector of Chagas disease, both in vivo and in vitro. The fungitoxic effect of soyuretox on yeast Candida albicans involves the production of superoxide anions, and the peptide was immunolocalized on the surface of the yeast. Finally, the behavioral and morphological effects of soyuretox on zebrafish larvae (Danio rerio) were evaluated, aiming to deepen the understanding of the mechanism of toxic action and the toxicological profile of this molecule, which presents biotechnological potential as a biopesticide. / Ureases (ureia amido-hidrolases; EC 3.5.1.5) s?o enzimas n?quel dependentes, amplamente distribu?das em bact?rias, fungos e plantas, que catalisam a hidr?lise da ureia ? am?nia e di?xido de carbono. Optimal control of inhomogeneous spin ensembles : applications in NMR and quantum optics / Contrôle optimal d'ensembles inhomogènes de spins : application en RNM et en optique quantique

L’objectif de cette thèse est d’appliquer la théorie du contrôle optimal à la dynamique d’ensembles inhomogènes de spins. La première partie est dévouée au contrôle d’un ensemble de spins couplé à une cavité. La théorie est introduite en détail, et une méthode générale pour contrôler efficacement les spins est présentée. Plusieurs pulses sont déterminés dans les régimes de bonne et de mauvaise cavité. De même, les fonctions non linéaires généralisées sont utilisées afin de déterminer des approximations simples. Dans un second temps, le problème de la maximisation du Signal-sur-Bruit d’un écho de spin est abordé, et des conditions d’optimisations sont établies. Il est montré que les nouveaux pulses sont supérieurs à ceux de l’état de l’art, en termes de fidélité et d’augmentation du Signal-sur-Bruit. Par ailleurs, ils permettent d’explorer de nouvelles situations (e.g. mesure de FID (Free Induction Decay) en CQED avec un taux de perte de cavité plus long que T2∗). La seconde partie est dévouée à des problèmes de RMN/IRM standard. Deux situations de "sélectivité" sont étudiées. La première consiste à déterminer le pulse le plus court qui produit la transformation la plus sélective par rapport aux offsets. Dans le cas ultra-sélectif, la solution optimale est un arc singulier d’amplitude constante. Cependant, si des contraintes de robustesse sont ajoutées, la solution optimale peut-être un arc régulier. La seconde est celle de l’optimisation de base de données pour des expériences de MR-fingerprinting. Dans ce cas, un champ de contrôle est conçu pour générer une base de données "d’empreinte digitale" qui maximise le processus de reconnaissance entre spins de paramètres différents. / The goal of this thesis is to apply optimal control theory to the dynamics ofinhomogeneous spin ensembles. The first part focuses on the control of a spin ensemble coupled to a cavity. The theory is introduced in detail, and a general method to efficiently control spins ispresented. Several pulses are derived in the bad/good cavity regimes using numerical optimal control techniques. Additionally, non-linear generalized functions are used in order to derivesimple approximated solutions. In a second step, the problem of spin echo Signal to Noise Ratio maximization is investigated, and maximization conditions are derived. It is shown that new pulses are superior to state-of-the-art square pulses in terms of fidelity and SNR maximization. Moreover, they allow us to explore new situations (e.g. Free Induction Decay measurementsin cavity-QED with a cavity damping longer than T2∗). The second part focuses on standard NMR/MRI problems. Two distinct situations of selectivity are investigated. The first one consists of determining the time minimum pulse which produces the most offset-selective transformation. In the ultra-selectivity case, the optimal solution is a singular arc of constant amplitude. However,if additional robustness constraints are taken into account, the optimal solution can be a regular arc. The second situation is the optimization of databases for MR-fingerprinting experiments. In this case, a control field is designed so that it generates a fingerprint database which maximizesthe recognition process between several spins with different parameters.

Le lactate est considéré comme un métabolite déchet depuis de très nombreuses années. Cependant, cette vision semble revisitée depuis quelques temps, avec l'apparition de la notion de glycolyse aérobie et de navettes lactate dans différents types cellulaires (muscle, cerveau et sperme). Concernant le cerveau, des études in vitro, ex vivo et in vivo réalisées ces 20 dernières années ont montré, d'une part, que les astrocytes produisent du lactate et d’autre part que le lactate pouvait être un substrat énergétique pour le système nerveux central (SNC), et plus particulièrement les neurones. Cette notion de navette lactate entre astrocyte et neurone a été proposée pour la première fois en 1994 par Pellerin et Magistretti (ANLS, pour astrocyte-neuron lactate shuttle). Malgré de nombreuses recherches depuis, l'existence d'un transfert net de lactate entre les astrocytes et les neurones n'a toujours pas pu être démontrée in vivo. Dans cet optique, la visualisation de la production de lactate in vivo dans le cerveau activé est essentielle. Le rôle des transporteurs au lactate, MCTs (Monocarboxylate Transporters), dans la détection de ce signal est également un point capital. L’objectif de cette thèse a été de développer la spectroscopie de RMN in vivo localisée dans le cortex somato-sensoriel du rat en condition d’activation cérébrale. Dans un premier temps, un travail de développement a été effectué afin de mettre au point le protocole de stimulation neuronale et d’obtenir un rapport signal sur bruit suffisant pour pouvoir quantifier de façon fiable le lactate. Une fois le protocole établi sur des rats contrôles, l’étude a été réalisée sur des rats modifiés génétiquement et réprimés pour le MCT, soit neuronal, soit astrocytaire. Le but était de déterminer si ce partenaire clef de l’ANLS avait une influence sur les fluctuations de lactate lors de l'activation cérébrale. En plus de la spectroscopie proton in vivo et de l’IRM fonctionnelle, des études de RMN du carbone-13 ont été réalisées ex vivo. Le résultat majeur de cette thèse montre qu’en l’absence du transporteur de lactate neuronal, non seulement on perd l’augmentation de lactate lors de la stimulation cérébrale mais on perd également le signal BOLD sur l’IRMf. Ce résultat suggère, et ce pour la première fois, que l’activité neuronale est fortement dépendante du transporteur au lactate. / Lactate has been considered as a waste metabolite for many years. However, this vision has been reconsidered recently, with the appearance of the notion of aerobic glycolysis and lactate shuttles in different cell types (muscle, brain, and sperm). Concerning the brain, in vitro, ex vivo and in vivo studies carried out over the last 20 years have shown, on the one hand, that astrocytes produce lactate and, on the other hand, that lactate can be an energetic substrate for the central nervous system (CNS), and more particularly neurons. This lactate shuttle between astrocyte and neuron was first proposed in 1994 by Pellerin and Magistretti (called ANLS, for astrocyte-neuron lactate shuttle). Despite many studies since then, the existence of a net transfer of lactate between astrocytes and neurons has still not been demonstrated in vivo. In this regard, visualization of lactate production in vivo in the activated brain is essential. The role of lactate transporters, MCTs (Monocarboxylate Transporters), in detecting this signal is also a key issue. The objective of this thesis was to develop in vivo NMR spectroscopy located in the somato-sensory cortex of rats under brain activation conditions. First, experiments were carried out to develop the neural stimulation protocol and to obtain a sufficient signal-to-noise ratio to be able to quantify lactate. Once the protocol was established on control rats, the study was performed on genetically modified rats and down-regulated for MCT, either neuronal or astrocytic. The aim was to determine whether this key partner of the ANLS has an influence on lactate fluctuations during brain activation. In addition to in vivo proton spectroscopy and functional MRI, carbon-13 NMR studies were performed ex vivo. The major result of this thesis shows that in the absence of the neuronal lactate transporter, not only is the increase in lactate lost during brain stimulation but the BOLD signal on the fMRI is also lost. This result suggests, for the first time, that neural activity is highly dependent on the lactate transporter.
