A flexibilidade da socialidade presente em muitas espécies sugere que muitos dos traços comportamentais e fisiológicos presentes em grupos sociais complexos possam existir em estado latente em indivíduos de espécies solitárias. Hierarquia de dominância e assimetria reprodutiva são traços característicos de espécies sociais. O hamster Sírio é um excelente modelo para o estudo do estresse social e dos seus efeitos fisiológicos e comportamentais. Na presente pesquisa foram enfocados os efeitos do alojamento em grupo antes e no início da gestação na fêmea de hamster Sírio. Investigou-se se o estresse social produz algum grau de assimetria reprodutiva nesta espécie solitária e se o sucesso reprodutivo e o comportamento materno das fêmeas variam em função do seu status social. Dois experimentos foram realizados envolvendo respectivamente 34 fêmeas de hamster criadas em grupo do desmame até o início do estudo e 76 fêmeas criadas isoladamente. Em cada experimento, um terço dos animais foram alojados em gaiolas individuais, enquanto pares de fêmeas desconhecidas umas das outras eram formados com os outros dois terços. O comportamento dos pares foi observado ao longo de 10 dias para determinar o status social de cada fêmea. Todas as fêmeas foram acasaladas neste período. Quatro dias após o parto, os filhotes foram contados, sexados e pesados e as ninhadas padronizadas a seis filhotes, três machos e três fêmeas. As mães e suas ninhadas foram observadas diariamente durante sessões de 40min e 13 categorias comportamentais foram registradas. No intuito de esclarecer os mecanismos fisiológicos subjacentes à relação entre estresse social e fertilidade, os níveis dos hormônios reprodutivos e dos glicocorticóides foram monitorados ao longo da gestação por métodos não-invasivos, previamente validados, de quantificação dos seus metabólitos nas fezes. Foi demonstrada, pela primeira vez, a adequação de um enzimaimunoensaio e de um conjunto diagnóstico comercial de radioimunoensaio para a quantificação respectiva dos metabólitos fecais de glicocorticóides em hamsters Sírios dos dois sexos e de testosterona no hamster Sírio macho, mas não da fêmea. As variações das concentrações de metabólitos fecais de progesterona, estrógenos e glicocorticóides, refletiram os perfis séricos descritos na literatura para hamsters gestantes. Social hierarchy and reproductive skew are typical features of social species. Syrian hamsters (Mesocricetus auratus) are an ideal model for the investigation of social stress and its physiological, neuroendocrine, and behavioral effects. The present research focused on the effects of group-housing prior to mating and on the first days of gestation of the female hamster. We investigated if social stress produces some degree of reproductive skew in this solitary species, and if female reproductive success varies as a function of social rank. Two experiments were carried out. The first one involved 34 females group-raised from weaning until the beginning of the trial and the second one 76 singly raised females. In each case one third of the animals were singly housed. Pairs of unrelative females were formed with the remaining animals. Pair behavior was observed through 10 days for assessment of the social rank of each female. All females were mated during this period. On day 4 post-partum, pups were counted, sexed, and weighed and litters were culled to six, three males and three females. Litters and dams were observed daily through 40-min sessions and 13 behavioral categories recorded. In order to assess the physiological relationship between social stress and fertility, we monitored reproductive hormones and glucocorticoids of solitary and pair-housed females during pregnancy by utilizing recently established non-invasive methods for measuring the respective hormone metabolites in the feces. The suitability of an enzyme immunoassay and of a commercial radioimmunoassay for respective quantification of fecal glucocorticoid metabolites of hamsters of both sexes and fecal testosterone metabolites in the male but not in the female was proven for the first time. The patterns of fecal progesterone, estrogen, and glucocorticoid metabolites were similar to blood profiles reported in the literature for pregnant hamsters. Our results showed that when female hamsters are singly housed from weaning, social stress did not affect their fertility. However, when females are group-raised, both isolation and subordination stress induced a significant reproductive skew, which is a characteristic feature of cooperative breeders. It seems that group-keeping of adult hamsters evokes social physiological and behavioral mechanisms present as latent traits in this solitary species. As in other rodents, sociability seems flexible in the Syrian hamster and our findings support the idea that social interactions may be a function of ecological conditions.

La reproduction chez Oscarella lobularis (Porifera - Homoscleromorpha) : gènes impliqués et effets de l'environnement / The reproduction of Oscarella lobularis (Porifera - Homoscleromorpha) : genes and environmental effects

Les Porifères et les Cténophores sont probablement les deux lignées animales les plus anciennes. Leur étude permet de retracer l’histoire précoce des métazoaires, et d’aborder l'origine de la distinction entre les lignées somatique et germinale. En effet, chez les éponges cette distinction n’existe pas: les archéocytes et choanocytes pouvant donner les cellules somatiques et les gamètes.Après avoir établi la liste des gènes considérés comme impliqués dans la gamétogenèse chez les métazoaires, j’ai caractérisé leurs séquences et retracé leur histoire grâce à des analyses comparatives intégrant les principales lignées animales. Enfin, un suivi in situ d'Oscarella lobularis m’a permis d’affiner son cycle de vie et d’accéder à toutes les étapes du développement pour étudier l’expression de ces gènes et de tester leur implication dans la gamétogenèse.Ainsi, j'ai montré que 18 gènes du GMP (Germline Multipotency Program) sont présents ancestralement chez les animaux. Parmi ceux-ci 11 s’expriment pendant la gamétogenèse, au cours de l’embryogenèse, de la reproduction asexuée et de la régénération. Enfin, le suivi in situ a montré l’influence de la variation de la température et de la matière organique sur le déclenchement de la gamétogenèse.Mon travail suggère d’une part, que la spécification des cellules germinales est régie par des mécanismes génétiques communs à l’échelle de métazoaires, et d’autre part que ces gènes pourraient être impliqués dans la multipotence. Ces résultats renforcent l’hypothèse proposant une origine commune de la lignée germinale et des cellules souches somatiques. / Porifera and Ctenophora are probably the two most ancient animal lineages. Their study therefore allows to trace back the early history of metazoans and to address the origin of the distinction between somatic and germ lines. Indeed, in sponges this distinction does not exist: archeocytes and choanocytes can give rise to both somatic cells and gametes.After establishing the list of genes considered to be involved in gametogenesis in metazoans, I searched for these candidate genes (by local blast) in the transcriptomes of two sponge species (Oscarella lobularis and Oopsacas minuta). I thereby managed to characterize their sequences (phylogenetic and protein domain analyzes) and to trace their evolution through comparative analyzes including all main animal phyla. Finally, the in situ monitoring of O. lobularis enabled me to refine its life cycle and access all key developmental stages in order to study the expression of candidate genes in order to test their possible involvement in gametogenesis in this species.I have shown that 18 GMP (Germline Multipotency Program) genes are present ancestrally in animals. Among them, at least 11 are expressed not only during gametogenesis but also during embryogenesis, asexual reproduction and regeneration. Finally, in situ monitoring showed the influence of temperature variations and organic matter availability on gametogenesis.My work suggests, firstly that the specification of germ cells is controlled by common genetic mechanisms across metazoans, and secondly that these ancestral genes might be involved in pluripotency. These results reinforce the hypothesis suggesting a common origin of the germline and somatic stem cells.

Macroalgae in the Baltic Sea : responses to low salinity and nutrient enrichment in Ceramium and Fucus

<p>The brackish Baltic Sea is a marginal environment for both marine and freshwater species. The rate of ecological differentiation is presumably high due to strong selection pressure from a gradient of decreasing salinity that has been present in its current state for only about 3 000 years. Even more recently, increased nutrient loading due to human activities has affected the growth rate of species, with potential effects on their competitive interactions and responses to other regulating factors. I have investigated the potential effects of low salinity and nutrient enrichment on the distributional ranges of two marine macroalgae with a wide distribution in the Baltic Sea, the red alga <i>Ceramium tenuicorne</i> (Kütz.) Wærn and the brown alga <i>Fucus vesiculosus</i> L. </p><p>A field study in the northern Baltic Sea indicated a strong relationship between the community structure of macroalgae and abiotic factors even on a small, local scale. The abiotic factors are potentially modulated by eutrophication, which may have a strong effect on the depth distribution and abundance of macroalgae. On a regional scale, laboratory experiments suggested that nutrient enrichment is unlikely to affect the distribution of <i>Ceramium</i> and <i>Fucus</i> along the salinity gradient. Growth in <i>Ceramium</i> from the Baltic Proper was enhanced by nitrate and phosphate, but the response did not override growth constraints due to low salinity. <i>Ceramium</i> from the Gulf of Bothnia had an inherently lower growth rate that was not positively affected by nitrate and phosphate increase. In <i>Fucus vesiculosus</i>, reproductive performance was impaired by nitrate and phosphate levels corresponding to ambient levels in eutrophicated areas of the Baltic Sea, when measured by their effect on zygote attachment, germination, and rhizoid development.</p><p>The wide distribution of <i>Ceramium</i> in the inner Baltic Sea is probably related to local adaptation, rather than a generalized tolerance of different salinity levels. Ecotypic differences were observed when comparing strains from the Baltic Proper (salinity 7 psu) and the Gulf of Bothnia (4 psu). A high rate of vegetative reproduction was evident, although sexual reproduction was occasionally observed in salinity 4. In <i>Fucus vesiculosus</i>, genetic and morphological analyses of sympatric and allopatric populations of the common, vesicular, morphotype and a dwarf morphotype, characteristic for the Gulf of Bothnia, showed that the dwarf morphotype represents a separate evolutionary lineage. Also, vegetative reproduction was observed in <i>Fucus</i> for the first time, as supported by genetic and experimental data. </p><p>The results show that the biota of the inner Baltic Sea may have unique adaptive and genetic properties, and that it is highly relevant to consider subspecies diversity in Baltic Sea management.</p>

Modes de perturbation de la stéroïdogenèse testiculaire et de la spermatogenèse chez le poisson zèbre (Danio rerio) par des fongicides azolés

Les azoles sont des fongicides présents dans les milieux aquatiques et connus pour inhiber des activités enzymatiques de cytochromes P450 (CYP). L'objectif de ce travail de thèse est de caractériser le mécanisme d'action d'un fongicide pharmaceutique, le clotrimazole sur la stéroïdogenèse testiculaire chez le poisson zèbre au travers l'étude d'un réseau de gènes fonctionnels le long de l'axe cerveau-hypophyse-gonade, et d'évaluer la capacité du clotrimazole à perturber la spermatogenèse. Nous montrons que le clotrimazole est capable d'affecter la stéroïdogenèse de manière différente in vitro et in vivo (i) des expositions d'explants testiculaires in vitro conduisent à l'inhibition de la synthèse de 11-kétotestostérone (11-KT), montrant une action directe de la molécule sur le testicule et (ii) l'exposition in vivo provoque une augmentation de l'expression de gènes impliqués dans le processus de la stéroïdogenèse. Nous avons ainsi mis en évidence un système de compensation biologique au niveau de l'organisme, avec un rôle prépondérant de la voie Fsh/FshR dans la médiation des effets du clotrimazole. Enfin, des effets sur la spermatogenèse ont été observés in vivo suite à une exposition chronique au clotrimazole, avec notamment une augmentation de la masse gonadique et du nombre de cellules de Leydig. Les effets tissulaires observés sont cohérents avec des effets mesurés au niveau moléculaire. L'ensemble de ces données montre l'intérêt de la démarche expérimentale utilisée pour caractériser le mécanisme d'action du clotrimazole. Ce travail ouvre de nombreuses perspectives, en premier lieu sur l'étude de l'impact fonctionnel du clotrimazole sur la reproduction.

Thesis abstract The influences of energy quantity on the sexual and asexual reproduction and possible trade-offs between them in the sea anemone, Aiptasia pulchella were investigated. Different feeding frequencies, light intensities and presence of zooxanthellae were the treatments of energy quantity. Lacerate numbers, lacerate sizes and total weight of lacerates were used to quantify investment in asexual reproduction; oocyte sizes and gonad weights were used as the measures of sexual reproduction. The correlation between total weight of lacerates and that of gonads provided an opportunity for evidence of trade-offs between sexual and asexual reproduction. Under higher feeding frequencies, A. pulchella produced more and larger lacerates as well as larger oocytes and heavier gonads than under lower feeding frequencies. Under lower light intensities, A. pulchella produced more and heavier lacerates with than without zooxanthellae. Lacerate sizes were influenced by light intensities, presence of zooxanthellae and the interaction of the two factors. At intermediate light intensity (2000 lux), Aiptasia pulchella produced heavier gonads than at 0 lux or 10000 lux. A. pulchella produced heavier gonads with than without zooxanthellae. The oocyte sizes of A. pulchella were also influenced by light intensities, zooxanthellae and the interaction of the two. In both sexual or asexual modes of reproduction, A. pulchella performed better under higher than lower feeding frequencies. There were higher level of asexual reproduction for A. pulchella under lower light intensities with zooxanthellae. A. pulchella achieved the highest levels of fecundity, as an indication of sexual reproduction, under a light intensity of 2000 lux in individuals containing zooxanthellae. There is a positive correlation between sexual and asexual investment under different energy regimes. This is not compatible with the prediction of the trade-off hypothesis.

La présente analyse est ciblée sur la transition entre les phases géomorphologiques, pionnières et biogéomorphologiques décrites par le concept de succession biogeomorphologique fluviale d’une rivière sablo-graveleuse de plaine, la Loire moyenne. L’étude se focalise sur la reproduction sexuée et la survie (dans leur premiers stades, au cours des crues) de deux espèces de Salicacées : Populus nigra L. et Salix alba L. sur une barre sédimentaire et repose sur des approches in- et ex-situ. La dynamique hydro-sédimentaire d’une barre forcée influence le potentiel de survie des semis des ligneux pionniers. Les semis présentent des adaptations morphologiques différentes en fonction des conditions sédimentaires de germination modulant leur potentiel de survie. Trois modèles conceptuels sont proposés : (i) dynamique d’une barre forcée en crue, (ii) évolution longitudinale des processus de mortalité des semis selon le granoclassement amont-aval, (iii) vitesse d’évolution d’une barre forcée vers une île pionnière initiée par l’installation de semis de ligneux pionniers et archivage sédimentaire associé dans une rivière sablo-graveleuse de plaine. / The present study is focused on the three first stages: geomorphic, pioneer and biogeomorphic of the Fluvial Biogeomorphological Succession applied to a sandy-gravel bed lowland river: the Loire River (in its middle reaches). This work is based on both an in and ex situ approach. It analyses the recruitment and survival during their first stages of growth of two Salicaceae trees: Populus nigra L. and Salix alba L. developed on a non-migrating (forced) bar. The dynamics of this bar influences the survival potential of seedlings during floods. The seedlings adapt morphologically according to the sedimentary structure, and as a consequence, modify their survival potential. Three conceptual models detail: (i) the dynamics of a mid-channel forced bar during floods, (ii) the longitudinal evolution of the mortality of seedlings induced by the downstream fining of sediments, (iii) the sediment archiving and time needed to reach a pioneer island state from a non-migrating vegetated bar.
