Skillnader i fysisk prestation inomhus och utomhus : En randomiserad kontrollerad experimentell cross-over studie

Bakgrund: Träning i naturliga miljöer har hälsofrämjande effekter och ökar välbefinnandet i jämförelse med träning som sker i inomhusmiljöer. Individer som utför träning i utomhusmiljöer visar sig ha högre hastighet, hjärtfrekvens och blodlaktatkoncentration, i jämförelse med individer som tränar inomhus vilket påverkar den fysiska prestationen.Syfte: Syftet med denna studie var att undersöka mätbara skillnader i fysisk prestation inomhus och utomhus genom ett maxpulstest.Metod: Studien genomfördes med en randomiserad kontrollerad cross- over design där åtta män och fem kvinnor (medelålder 26 ± 3.67), fysiskt aktiva, genomförde ett beeptest vid två tillfällen. Testet utfördes av samma person vid ett tillfälle inomhus och ett tillfälle utomhus. Analysen utgick från uppskattat VO2 max genom protokoll av beeptest, objektiv mätning och kontroll av puls direkt före tester och direkt efter genomförda tester.Resultat: Resultaten visade en signifikant skillnad i VO2 max (O2ml/kg/min) (p= 0.047), genom en högre fysisk prestation inomhus i jämförelse med utomhus (46.08 ± 8.76 och 44.73 ± 8.84). Puls (slag/min) efter genomförda tester visade ett högre medelvärde inomhus i jämförelse med utomhus (192.36 ± 10.13 och 190.45 ± 9.11), men ingen signifikant skillnad (p=0.11). Puls före tester visade ingen signifikant skillnad (p=0.898) mellan inomhusmiljön och utomhusmiljön (107.16 ± 18.78 och 107. 75 ± 18.49).Slutsats: Resultaten indikerar på att det är skillnader i fysisk prestation i olika miljöer. VO2 max var signifikant högre i inomhusmiljön i jämförelse med utomhusmiljön. Medel-maxpulsen tenderade ha ett högre medelvärde inomhus i jämförelse med utomhus. / Background: Exercise performed in natural environments produces health-promoting effects and increases well-being, in comparison with exercises performed in indoors-environments. Individuals who exercise outdoors has higher velocity, heartrate and blood lactate concentration compared to individuals who exercise indoors, which affects the physical performance.Purpose: The purpose of this study was to investigate measurable differences in physical performance indoors and outdoors through a maximum heartrate test.Method: The study was conducted with a randomized controlled cross over design where five women and eight men, physical active, in the middle age of 26 ± 3.67 years, performed a shuttle run test at two times. The test was repeated by the same individual one time indoors and one time outdoors. The analysis was made by estimated VO2 max using a protocol from the performed shuttle run test, and controlled objective measurements of pulse levels, directly before the test started and directly after finished test.Results: Significant differences were found between indoors and outdoors, where VO2 max (O2ml/lbs/min) (p= 0.047) was higher indoors than outdoors (46.08 ± 8.76 and 44.73 ± 8.84). Pulse levels (beats/min) was higher indoors than outdoors (192.36 ± 10.13 and 190.45 ± 9.11) after finished test, but significant differences were not found (p=0.11). Significant differences were not found in pulse levels before the test (p=0.898), between the different environments (107.16 ± 18.78 and 107. 75 ± 18.49).Conclusion: The results in this study show that there are differences in physical performance between different environments. VO2 max was significantly higher in the indoor environment compared to the outdoor environment. The average maximum pulse tended to have a higher average indoors compared to outdoors.

Controle cooperativo aplicado a sistemas de posicionamento dinâmico. / Cooperative controler applied to dynamic positioning systems

Hoje em dia com o crescente aumento da exploração de petróleo e gás em águas profundas, há um aumento na demanda por operações offshore envolvendo a cooperação entre unidades flutuantes. Tais operações requerem um alto nível de planejamento e coordenação, o que na maioria dos casos é feito com a troca de informação no nível de operação, com cada unidade flutuante comandada independentemente. Exemplos de operações deste tipo vão desde operações de alívio passando por operações de instalação de equipamento submarino, até operações de pesquisa envolvendo múltiplas unidades flutuantes dotadas de sistema de posicionamento dinâmico (DP). As vantagens do controle cooperativo surgem com a redução do erro da distância relativa durante a manutenção do posicionamento ou durante a execução de manobras de posicionamento conjuntas. No presente trabalho, os conceitos de controle de consenso são aplicados de forma combinada com o sistema DP de cada navio. A influência dos ganhos do controlador cooperativo no sistema como um todo será discutida, utilizando-se técnicas de análise da resposta em frequência. Simulações completas no domínio do tempo e experimentos usando modelos em escala serão utilizados para se demonstrar o funcionamento do controle cooperativo. Todas as simulações serão conduzidas no simulador Dynasim e os ensaios experimentais no tanque de provas da Engenharia Naval da Escola Politécnica da Universidade de São Paulo. Além disso, serão feitas comparações entre os experimentos em tanque de provas e simulações numéricas equivalentes, demonstrando-se a validade dos ensaios numéricos. Será também demonstrado que os requisitos de projetos adotados são atendidos pelos ensaios em tanque de provas. . / With the increasing of deep water oil & gas exploration, there is also an increase in the demand by offshore operations involving muti-vessels. Such operations require a high level of planning and coordination, which in most of the cases is made by information exchange at the operation level, being each vessel commanded independently. Examples of such operations are offloading, subsea equipment installation and subsea research operation; all of them involving multiples dynamically positioned (DP) vessels. The advantage of the cooperative control arises with the reduction of the relative positioning error during station keeping or transient maneuvers. In this work, the consensus control concepts are applied combined with the DP System of each ship. The cooperative DP controller will be investigated with the analysis of the coupled dynamics of the vessels. The influence of the cooperative control gains on the whole system will be discussed, using the frequency response of the open loop system. Fully nonlinear time-domain simulations and experimental results will be used to demonstrate the operation of the cooperative control. Besides that, comparisons between the small-scale experiments and equivalent numerical simulations will be carried out, validating the experimental results. It will also be demonstrated that the adopted design requirements are met. All tests will be carried out using the Dynasim numerical simulator and the small-scale experiments will be carried on the academic towing tank in the Naval Architecture and Ocean Engineering Department, Polytechnic School of University de São Paulo.

Methodology for definition of new operating sectors fo DP assisted offloading operations in spread-moored platforms. / Metodologia para definição de novos setores operacionais para operações de offloading com navios DP em plataformas ancoradas em Spread Mooring.

This thesis defines and applies a methodology for analyzing the possibility of extending the operating sector of DP shuttle tankers for offloading operations in Spread Moored FPSO Platforms. Extending the operating sector is beneficial to increase operations\' availability and to reduce DP power demand under certain environmental conditions. This study is important, since several emergency disconnections have occurred during offloading in Santos Basin because the shuttle tanker was pushed out of the green sector by environmental resultants that pointed to the West. However, this proposal has to be carefully analyzed in order to guarantee the operations\' safety and not to increase the risks of collision between FPSO and shuttle tanker, of oil pollution and of personnel safety. The methodology consists in five basic steps: Preliminary Risk Analysis to assess the potential hazards associated with the new sector; evaluation of the uptime gain through static analysis; evaluation of DP power demand inside the original and extended sector; real time simulations to evaluate the operation in a realistic environment; and, finally, field tests to validate the proposal. This thesis presents the contextualization of the problem, a bibliographical research, theoretical concepts, the detailed methodology and results of each step. The results show that the average uptime gain is significant both in Campos and Santos Basins - up to 9% and 13% respectively - and that the additional risks created by the sector extension are well mitigated if some recommendations are put into place. The conclusion of this thesis is that extending the operating sector is not only beneficial but also safe. / Esta dissertação define e aplica uma metodologia para analisar a possibilidade de extensão do setor operacional de navios aliviadores DP para operações de offloading em plataformas FPSO ancoradas em Spread-Mooring. Esta proposta apresenta como vantagens o aumento da disponibilidade das operações e redução na demanda de energia do navio DP em certas condições ambientais. O estudo é importante tendo em vista que várias desconexões de emergência já ocorreram durante operações de alívio na Bacia de Santos, porque o navio-tanque foi empurrado para fora do setor verde por resultantes ambientais que apontavam para oeste. No entanto, a proposta deve ser cuidadosamente analisada para garantir que o novo setor não aumenta os riscos de colisão, de poluição ambiental e de segurança às pessoas. A metodologia consiste em cinco etapas básicas: Análise Preliminar de Riscos (APR); avaliação do ganho de disponibilidade da operação; avaliação da demanda de energia do sistema DP nos setores original e estendido; simulações de manobra em tempo real; e, finalmente, testes em campo para validação da proposta. Esta dissertação apresenta a contextualização do problema, pesquisa bibliográfica, conceitos teóricos, a metodologia detalhada e os resultados de cada etapa. Os resultados mostram que o ganho médio de disponibilidade é significativo nas bacias de Campos e de Santos - até 9% e 13%, respectivamente - e que os riscos adicionais criados pela extensão do setor são devidamente mitigados se as recomendações levantadas na APR forem implementadas. A conclusão desta dissertação é que estender o setor operacional é, não somente benéfico, mas também seguro.

Mockup einer Betriebsleitstelle für automatisierte Shuttlebusse - Konzeption und Design eines universellen, visuellen und auditiven Interfaces

Derzeit sind automatisierte Shuttlebusse beim Einsatz auf einen Operator an Bord angewiesen. Damit sich dies nicht erst beim vollständig autonomen Fahren ändert, bedarf es weiterer Lösungsansätze. Eine Möglichkeit ist die Überwachung der Busse aus einer Betriebsleitstelle. Allerdings können mit den Betriebsleitstellen von Verkehrsgesellschaften nach heutigem technologischem Stand keine Fahrfunktionen übernommen werden. Um diese Forschungslücke zu schließen, wurde im Rahmen dieses Beitrags ein Mockup für eine Betriebsleitstelle von automatisierten Shuttlebussen entwickelt. Das Hauptaugenmerk bei der Konzeption dieses Interfaces liegt in der Anforderungsanalyse. Zu diesem Zweck wurden Experten aus Wirtschaft und Wissenschaft eingebunden. Als Ergebnis ist ein Mockup mit einer interaktiven Karte, einer Ereignisliste und einer Ansicht zur Übernahme der Fahrfunktionen entstanden.

Development of seamless woven node element structures for application in integral constructions

In order to advance consistent lightweight construction principles in automotive and mechanical engineering, support frame construction made from high-performance materials is becoming more commonplace. These consist of complexly structured nodular connection elements. The required connection elements have not yet been produced satisfactorily. The developed node element structures in this paper are produced on a shuttle weaving loom by flattening and weaving them as multi-surface woven fabrics. The development of the woven concept for the realization of node element structures is based on the fragmentation of the individual sub-elements. The goal of this research is development of a flexible technology for weaving fabrics and intended for the integral realization of woven nodular semi-finished products with complex geometries and connections, which are to be used to connect Fiber-reinforced Plastic components in support frame structures.

Effect of Electrolytes on Room-Temperature Sodium-Sulfur Battery Performance

<p>  </p> <p>Room-temperature sodium-sulfur (RT Na-S) batteries are an emergent new technology that are highly attractive due to their low raw materials cost and large theoretical specific energy. However, many fundamental problems still plague RT Na-S batteries that prevent their progression from the research and development phase to the commercial phase. Sulfur and its final discharge product are insulators, and intermediate polysulfide discharge products are soluble in commonly used liquid electrolytes. As a result, RT Na-S cells exhibit large capacity defects, low coulombic efficiencies, and rapid capacity fading. Additionally, the reactive sodium metal anode can form dendrites during cycling, which reduces capacity and shortens cell life. One way to combat these issues is the judicious selection of electrolyte components. In this study, the effects of monoglyme (G1), diglyme (G2), and tetraglyme (G4) glyme ether electrolyte solvents on RT Na-S cell performance are investigated. Galvanostatic cycling of Na/Na symmetric coin cells reveals that the G2 solvent enable stable cycling at low overpotentials over a wide range of current densities. In contrast, the G1-based cells show evidence of dendritic plating, and G4-based cells are not suitable for use at high current densities. Electrochemical impedance spectroscopy during cycling confirms that the G2 solvent facilitates the formation of a strong, stable SEI on the Na electrode surface. Results from galvanostatic cycling of RT Na-S full coin cells demonstrates that G1-based cells deliver the highest initial specific discharge capacities among the three cell types, but G4-based cells are the most reversible. Infinite charging, the indefinite accrual of charge capacity at the high charge voltage plateau, affects all cell types at different cycle numbers and to different extents. This behavior is linked to the strength of the polysulfide shuttle during charge. Optical microscopy experiments show that G2 and G4 facilitate the formation of the S3•- sulfur radical, which reduces capacity. G1 minimizes the radical formation and thus delivers higher initial specific discharge capacity. In order to fully optimize the electrolyte for RT Na-S cells, future work should study glyme solvent blends, additives, and concentrated salts.</p>

DEVELOPMENT OF AN ADVANCED GENETIC TOOLBOX TO ENABLE GENOME SCALE ENGINEERING IN SINORHIZOBIUM MELILOTI

Synthetic biology has ushered in a new age of molecular biology with the aim towards practical developments in disciplines ranging from medicine, agriculture, and industry. Presently, it remains difficult to manipulate the genomes of many organisms due to lack of genetic tools. These problems can be circumvented by cloning large fragments of DNA into strains where many genetic tools are in place, such as Saccharomyces cerevisiae. However, this organism is unable to directly transfer cloned DNA to other organisms and is unable to stably maintain DNA with a G+C content >40%. Many organisms relevant in biotechnology often have G+C content DNA >60%, and therefore are difficult to engineer. Here, the soil bacteria Sinorhizobium meliloti was chosen as a host strain to clone and manipulate large fragments of high G+C content DNA. S. meliloti is a Gram-negativeα-proteobacteria that forms symbiotic relationships with legumes to fix nitrogen. It has a multi-partite genome with a G+C content of 62.7% that includes a chromosome (3.65 Mb), the pSymA (1.35 Mb), and pSymB (1.68 Mb) replicons. A restriction endonuclease hsdR mutant strain lacking pSymA and pSymB was created and used in this study. Multi-host shuttle (MHS) vectors were constructed that allow for direct transfer and maintenance of DNA in E. coli, S. cerevisiae, and P. tricornutum. Characterization of strains was conducted to determine transduction, conjugation, and transformation frequencies, as well as stability of MHS plasmids. Furthermore, a proof-of-concept experiment was conducted to clone large plasmids (70-205 kb) with G+C content >58% via site-specific recombination at a landing pad in the MHS vector, which was then verified using colony PCR. This work demonstrates the usefulness of S. meliloti containing a MHS vector for cloning of large fragments with high G+C content DNA, a technology that may be used for several applications in both applied and basic research. / Thesis / Master of Science (MSc) / Synthetic biology is an emerging field that incorporates principles of molecular biology and engineering for the design and construction of biological systems for application in medicine, agriculture, and industry. Presently, it remains difficult to modify genomes of several organisms due to lack of available techniques. Yeast is currently used for the modification of large DNA pieces, however it is unable to transfer and maintain modified DNA with high G+C content. Here, the bacteria Sinorhizobium meliloti was used as a host organism to conduct genetic engineering due to its ability to maintain large DNA pieces with a high G+C content. Characterization experiments were conducted to assess the efficiency of this organism for this task. Using this strain, a proof-of-concept experiment to demonstrate the uptake and maintenance of large, high G+C DNA pieces was completed. This technology may be useful in biotechnology applications for engineering of large DNA pieces from industrially relevant organisms.

Contribution au développement d’une loi de guidage autonome par platitude : application à une mission de rentrée atmosphérique

Cette thèse porte sur le développement d'une loi de guidage autonome par platitude pour les véhicules de rentrée atmosphérique. La problématique associée au développement d'une loi de guidage autonome porte sur l'organisation globale, l'intégration et la gestion de l'information pertinente jusqu'à la maîtrise du système spatial durant la phase de rentrée. La loi de guidage autonome proposée dans ce mémoire s'appuie sur le concept de platitude, afin d'effectuer un traitement des informations à bord, dans le but double d'attribuer un niveau de responsabilité et d'autonomie au véhicule, déchargeant ainsi le segment sol de tâches opérationnelles "bas niveau", pour lui permettre de mieux assumer son rôle de coordination globale. La première partie de ce mémoire traite de la caractérisation formelle de sorties plates pour les systèmes non linéaires régis par des équations différentielles ordinaires, ainsi que pour les systèmes linéaires à retards. Des algorithmes constructifs sont proposés afin de calculer des sorties plates candidates sous un environnement de calcul formel standard. Dans la seconde partie, une méthodologie complète et générique de replanification de trajectoires de rentrée atmosphérique est proposée, afin de doter la loi de guidage d'un certain niveau de tolérance à des pannes actionneur simple/multiples pouvant survenir lors des phases critiques d'une mission de rentrée atmosphérique. En outre, une méthodologie d'annexation superellipsoidale est proposée afin de convexifier le problème de commande optimale décrit dans l'espace des sorties plates. La loi de guidage proposée est ensuite appliquée étape par étape à une mission de rentrée atmosphérique pour la navette spatiale américaine STS-1. / This thesis deals with the design of an autonomous guidance law based on flatness approach for atmospheric reentry vehicles. The problematic involved by the design of an autonomous guidance law relates to the global organization, the integration and the management of relevant data up to the mastering of the spacecraft during the re-entry mission. The autonomous guidance law proposed in this dissertation is based on flatness concept, in order to perform onboard processing so as to locally assign autonomy and responsibility to the vehicle, thus exempting the ground segment from "low level" operational tasks, so that it can ensure more efficiently its mission of global coordination. The first part of the manuscript deals with the formal characterization of flat outputs for nonlinear systems governed by ordinary differential equations, as well as for linear time-delay systems. Constructive algorithms are proposed in order to compute candidate flat outputs within a standard formal computing environment. In the second part of the manuscript, a global and generic reentry trajectory replanning methodology is proposed in order to provide a fault-tolerance capability to the guidance law, when facing single/multiple control surface failures that could occur during the critical phases of an atmospheric reentry mission. In addition, a superellipsoidal annexion method is proposed so as to convexify the optimal control problem described in the flat outputs space. The proposed guidance law is then applied step by step to an atmospheric reentry mission for the US Space Shuttle orbiter STS-1.

Hur korrelerar GIH:s Pyramidtest med VO2max på rullband för elitorienterare?

Syfte och frågeställningar Syftet med studien var att undersöka för elitorienterare resultaten vid och korrelationen mellan ett så kallat 5 minuters pyramidtest (5MPT) och ett maximalt test för bestämning av maximalt syreupptag (VO2max) på rullband. Frågorna vi ställde oss var: Vilken korrelation föreligger mellan 5MPT och ett bestämt VO2max på rullband hos elitorienterare? Hur ser en jämförelse ut mellan uppmätt samt beräknat VO2max? Vilken är reliabiliteten för 5MPT? Hur ser ekvationen ut för att beskriva sambandet mellan dessa två olika maxtester? Metod I valideringsstudien deltog 16 elitorienterare (6 kvinnor och 10 män) i åldrarna 17 till 37 år. De två testerna som genomfördes var 5MPT och bestämning av VO2max på rullband. 5MPT är ett skytteltest som är fem minuter långt, där testpersonen (tp) tar sig fram och tillbaka mellan två stolpar (sträcka 5.5 m) där en vertikal trappa (formad likt en pyramid) passeras under varje vända. Antalet vändor, skattad ansträngning samt hjärtfrekvens (HF) registrerades. Utvecklad power (effekt) erhölls genom produkten av vikt, gravitation, antal vändor, höjd på högsta plinten (0,62 m) dividerat med total duration i 5MPT. Testet för att bestämma VO2max utfördes med löpning på ett motordrivet rullband. Under den första minuten av testet sprang tp på en hastighet av 8km/h (kvinnor) respektive10km/h (män) utan lutning. För varje minut ökades hastigheten med 1km/h. Lutningen höjdes med en grad efter minut ett och därefter med 0,5 grader/minut. Testet pågick till dess tp nått sin VO2max. Var 15:e sekund registrerades tp:s HF, VO2 (l/min), VCO2 (l/min), VO2 (ml/kg/min) och respiratorisk kvot (RER) med hjälp av ett datoriserat syreupptagningssystem. Resultat Korrelationen var signifikant och hög (r = 0,89, p&lt;0,0001) mellan utvecklad power på 5MPT och VO2max (l/min) och även mellan antalet vändor på 5MPT och VO2max (l/min) (r = 0,89, p&lt;0,0001) på detta material av elitorienterare. Följande formler, baserade på resultaten från elitorienterarna, kan användas för att predicera VO2max efter genomfört 5MPT: VO2max  (l/min) = (5MPTPOWER – 50,914) / 25,795 VO2max (l/min) = (5MPTVÄNDOR – 74,447) / 9,7668 Slutsats 5MPT uppvisar en stark korrelation med bestämd VO2max sett till power och/eller antalet genomförda vändor. Beräkningsformler för prediktion av VO2max har varit möjligt att beskriva. Se resultatdelen för andra resultat och samband mellan 5MPT och VO2max. / Abstract Aim The aim of this study was to investigate for elite orienteers the results in and the correlation between a so called 5-minutes-pyramid test (5MPT) and a specific maximum oxygen uptake test (VO2max) on a treadmill. The questions we asked ourselves were: Which correlation exists between 5MPT and a specific VO2max on the treadmill for elite orienteers? How would a comparison look like between measured and estimated VO2max? What is the reliability for the 5MPT? How would an equation look like that describe the correlation above? Method In the validity the study 16 elite orienteers (6 women and 10 men) aged 17 to 37 years participated. The two tests that have been carried out were the 5MPT and determined VO2max during running on a treadmill. The 5MPT is a 5-minute-shuttle test, in which the participant moves back and forth in a short interval (5.5 m) over boxes (highest height: 0.62) formed like a pyramid. Power in the pyramid test (5MPTPOWER) was calculated as the product of numbers of laps, body weight, gravity and highest box level divided by time. To determine VO2max the method running on a treadmill was performed. During the first minute the running speed was 8km/h (women)/10 km/h (men) without elevation. Each minute the velocity increased with 1 km/h. The elevation increased with 1 degree after the first minute and thereafter with 0,5 degrees/minute.  The test was finished when the participant had reached her VO2max. Every 15 second the participants heart rate, VO2 (l/min), VCO2 (l/min), VO2 (ml/kg/min) and RER, was registered by means of an online system. Results The correlation between the developed power of the 5MPT and measured VO2max (l/min) was high (r = 0.89, p&lt;0,0001) and also between the number of laps in the 5MPT and measured VO2max (l/min) (r = 0.89, p &lt; 0,0001). The following formulas can be used to predict VO2max after completed 5MPT: VO2max  (l/min) = (5MPTPOWER – 50,914) / 25,795 VO2max (l/min) = (5MPTLAPS – 74,447) / 9,7668 Conclusions 5MPT show a strong correlation with VO2max determined in terms of power and/or the number of completed laps.
