Influence of Degradable Polar Hydrophobic Ionic Polyurethanes and Cyclic Mechanical Strain on Vascular Smooth Muscle Cell Function and Phenotype

Vascular tissue engineering (VTE) with the use of polymeric scaffolds offers the potential to generate small-diameter (<6 mm) arteries. In this thesis, a degradable polar hydrophobic ionic (D-PHI) polyurethane porous scaffold was synthesized with the objective of demonstrating its potential application for VTE. D-PHI scaffold synthesis was optimized, maximizing isocyanate and methacrylate monomer conversion. Through the incorporation of a lysine-based crosslinker, scaffold mechanical properties and swelling were manipulated. Furthermore, D-PHI scaffolds demonstrated the ability to support the growth and adhesion of A10 vascular smooth muscle cells (VSMCs) during two weeks of culture. This study also investigated the effect of a double porogen approach on D-PHI scaffold properties, demonstrating an increase in the total scaffold porosity and pore interconnectivity. Specifically, it was found that the use of 10 wt% polyethylene glycol and 65 wt% sodium bicarbonate porogens resulted in a porous (79±3%) D-PHI scaffold with the mechanical properties (elastic modulus=0.16±0.03 MPa, elongation-at-yield=31±5%, and tensile strength=0.04±0.01 MPa) required to withstand the physiologically-relevant cyclic mechanical strain (CMS) that is experienced by VSMCs in vivo. Furthermore, the effects of uniaxial CMS (10% strain, 1 Hz, 4 weeks) on human coronary artery smooth muscle cells (hCASMCs), which were cultured in a porous D-PHI scaffold, were studied using a customized bioreactor. Four weeks of CMS was shown to yield greater DNA mass, more cell area coverage, a better distribution of cells within the scaffold, the maintenance of contractile protein expression and the improvement of tensile mechanical properties. The in vitro and in vivo degradation as well as the in vivo biocompatibility of D-PHI scaffolds were also investigated. Following their subcutaneous implantation in rats (100 days), porous D-PHI scaffolds demonstrated more cell/tissue infiltration within their pores and degraded in a controlled manner and at a faster rate when compared to in vitro studies (120 days), retaining the mechanical integrity required during neo-tissue formation. This thesis provides significant insight into the role of the D-PHI scaffold in combination with physiologically-relevant CMS in modulating VSMC proliferation and phenotype. The findings of this work can be used to tailor vascular tissue regeneration by regulating VSMC function in a directed manner.

Scar-free wound healing and regeneration in the leopard gecko (Eublepharis macularius)

Scar-free wound healing and regeneration are uncommon phenomena permitting the near complete restoration of damaged tissues, organs and structures. Although rare in mammals, many lizards are able to undergo scarless healing and regeneration following loss of the tail. This study investigated the spontaneous and intrinsic capacity of the leopard gecko (Eublepharis macularius) tail to undergo scar-free wound healing and regeneration following two different forms of tail loss: autotomy, a voluntary and evolved mechanism of tail shedding at fracture planes; and surgical amputation, involuntary loss of the tail outside the fracture planes. Furthermore, I investigated the ability of the regenerate tail to regenerate by amputating a regenerate tail (previously lost by autotomy). To investigate these phenomena I imaged wound healing and regenereating tails daily (following autotomy and amputation) to document gross morphological changes. I used histochemistry to document tissue structure and immunohistochemistry to determine the tissue/cellular location of my five proteins of interest (PCNA, MMP-9, WE6, α-sma, TGF-β3). Each of these proteins of interest has been previously documented during wound healing and/or regeneration in other wound healing/regeneration model organisms (e.g. mice, urodeles, lizards, zebrafish). Scar-free wound healing and regeneration occurred following autotomy, amputation of the original tail and amputation of the regenerate tail, indicating that the leopard gecko tail has an instrinsic scar-free wound healing and regenerative capacity that is independent of the mode of tail loss (autotomy or amputation). Furthermore immunohistochemistry revealed a conserved sequence and location of the expression of the five proteins of interest following both forms of tail loss. These results provide the basis for further studies investigating scar-free wound healing and regeneration in a novel amniote model, the leopard gecko. / NSERC

Contribution différentielle du tissu adipeux mâle et femelle dans l’établissement du diabète de type 2 et des altérations cardiovasculaires : rôle de l’apport lipidique

Au cours des dernières années, il est devenu évident que les sociétés des pays industrialisés sont à haut risque de maladies métaboliques. Une alimentation riche en énergie (lipide/glucide), combinée à une sédentarité accrue, est un facteur environnemental contribuant à l'augmentation de la prévalence de maladies reliées spécifiquement à des troubles endocriniens comme l'obésité et le diabète. Le traitement de ces désordres métaboliques doit donc passer par la connaissance et la compréhension des mécanismes moléculaires qui contrôlent ces désordres et le développement de traitements ciblés vers les facteurs responsables. Le tissu adipeux est une glande endocrine qui sécrète des substances, regroupées sous le terme d'adipokines, qui contrôlent l'homéostasie énergétique. L'augmentation de la masse adipeuse est responsable du développement de dérégulation hormonale qui mène à des dysfonctions physiologiques et métaboliques. Pour contrecarrer le développement démesuré du tissu adipeux, la signalisation insulinique ainsi que l’apport énergétique, responsables de la différenciation adipocytaire, doivent être inhibés. In vivo, la leptine, adipokine dont la concentration est corrélée à la masse adipeuse, présente des actions pro ou anti-insuliniques dans l’organisme pour réguler ce phénomène. Elle favorise l’effet inhibiteur de l’insuline sur la synthèse hépatique de glucose alors qu’elle s’oppose à son action sur l’expression des enzymes glucokinase et phosphoénol-pyruvate carboxykinase. La leptine influence aussi le taux circulant de triglycérides en diminuant sa concentration plasmatique. D'autre part, l'adiponectine, adipokine insulino- sensibilisante, voit sa sécrétion diminuée avec la prise de poids. La sensibilité à l'insuline est ainsi diminuée au fur et à mesure que le débalancement de ces deux adipokines s'accentue. La résistance à l'insuline s'installe alors pour s'opposer au stockage énergétique et à la prise illimitée de poids et la glycémie augmente. L'augmentation du glucose sanguin stimule la sécrétion d'insuline au niveau des cellules pancréatiques. C'est le diabète caractérisé par une hyperglycémie et une résistance à l'insuline. Le diabète, une des premières causes de mortalité dans le monde, est plus répandu sous sa forme non insulinodépendante (diabète de type 2, DT2) liée à l'obésité. Récemment, différents facteurs de transcription ont été identifiés comme régulateurs de l'expression d'une panoplie de gènes impliqués dans le métabolisme glucidique et lipidique. Parmi eux, les récepteurs des inducteurs de la prolifération des peroxysomes (PPAR, Peroxisome Proliferator-Activated Receptor), appartenant à la famille des récepteurs nucléaires. Les PPAR ont été démontrés comme ayant un rôle central dans le contrôle de la transcription des gènes codants pour des protéines impliquées dans le métabolisme : les adipokines. PPARg, en plus de son implication dans le contrôle de l'homéostasie glucidique et lipidique, est reconnu comme étant un facteur de transcription pivot régulant l'adipogenèse du fait de son expression majeure dans le tissu adipeux. D'autre part, il est bien établi maintenant que l'obésité et le diabète sont des facteurs contribuant au développement du processus inflammatoire vasculaire caractéristique de l’athérosclérose. En effet, les cellules endothéliales et musculaires lisses, principales composantes de la média de l’artère, sont très sensibles aux altérations métaboliques. Une diminution de la sensibilité à l’insuline entraine une réduction de la disponibilité du glucose et l’utilisation des acides gras comme alternatif par ces cellules. Ceci induit l’accumulation des acides gras oxydés dans l’intima et leur filtration dans la média pour former un core lipidique. Bien que l’induction de la dysfonction endothéliale soit impliquée très précocement, certaines études pointent l’accumulation lipidique dans les cellules musculaires lisses vasculaires (CML) et leur dysfonction comme déclencheurs de l’athérosclérose. Ce travail visait donc, dans un premier temps, à développer un modèle d'altérations métaboliques liées à la modulation de l'activité du tissu adipeux via une alimentation riche en lipides. Dans un second temps, cette étude tentait d'évaluer l’impact des adipocytes de souris sur les CML vasculaires et sur la modulation de leurs fonctions dans ce modèle d'altérations métaboliques et DT2 liés à l'alimentation et à l'obésité. Ainsi, par le biais de deux diètes pauvres en cholestérol à profil lipidique différent, nous avons développé un modèle murin présentant divers stades d'altérations du métabolisme allant jusqu'au DT2 en lien avec l'obésité chez les mâles et chez les femelles. D’autre part, des signes de cardiomyopathie ainsi qu’une modulation du taux des adipokines sont reliés à ces mêmes diètes. Parallèlement, l’activité de PPAR!2 est modulée chez les souris sous diètes enrichies en gras. Ensuite, nous avons démontré que les adipocytes, provenant de souris alimentées avec une diète enrichie en gras, modulaient la migration et la prolifération des CML comparativement au groupe contrôle. Ces modulations dépendaient en grande partie de la nature de la diète consommée, mais également du sexe de la souris. Par ailleurs, les altérations fonctionnelles des CML, couplées à des modulations géniques, sont associées aux changements du profil de sécrétion des adipokines mesurées chez les adipocytes. L’ensemble de ces travaux suggère une action directe de la nature de la stimulation du tissu adipeux blanc dans la modulation du profil de sécrétion des adipokines et l'induction du DT2 in vivo. Ces altérations de la physiologie adipocytaire se reflètent in vitro où le tissu adipeux contribue aux altérations physiopathologiques des CML liées au DT2. Ainsi, cette étude est l'une des premières à établir un lien direct entre les modulations adipocytaires et les effets de leurs sécrétions sur la physiologie des CML. Ces observations peuvent être exploitées cliniquement dans un développement futur d’outils thérapeutiques visant à prévenir et à traiter les troubles métaboliques et le DT2, en ciblant le tissu adipeux comme entité métabolique et endocrine. / Obesity is recognized as a risk factor to a variety of chronic diseases linked to the metabolic syndrome like atherosclerosis and type 2 diabetes (T2D), and is a major cause of increased risk of morbidity and mortality worldwide. High fat diets (HFD) coupled with sedentarity in the industrialized societies contribute to the raise of metabolic alterations prevalence specifically linked to endocrine troubles. Treatment of these latter should include the comprehension of the molecular mechanisms underlying these disorders in order to appropriately target factors responsible for the disease establishment. Adipose tissue is no longer considered as a passive organ which only stores lipids, but also works as an active gland that secretes several bioactive substances called adipokines. Among them, there are key factors known to play a pivotal role in the regulation of glucose and lipid homeostasis, lipid storage, adipogenesis. They are also recognized for their control of a wide range of cell type like adipocytes, hepatocytes and skeletal myocytes. Accumulation of adipose tissue in obesity, linked with the type as much as the amount of dietary lipids, is due to hyperplasia and hypertrophy of adipocytes. These changes are associated with modification in their secretion and inflammatory profile. To counteract excessive fat tissue development, insulin signalling known for its role in adipogenesis is inhibited. Thus, leptin is secreted by adipocytes to inhibit insulin action and the insulin sensitizer adipokine, adiponectin, is down regulated. The two factors are correlated to weight gain and their respective secretion profile is upregulated for leptin and down regulated for adiponectin. Insulin resistance is developed to prevent energetic storage and unlimited weight gain but glycemic control fails and glycaemia raises. Hyperglycaemia stimulates more insulin secretion, a characteristic of T2D linked to obesity. An estimated 80% of those who develop T2D are obese. Obesity induces important and complex changes, not only in glycemic homeostasis but also in the adipocytes. Following fatty acids (FA) stimulation, the main ligand-activated transcriptional factor that controls adipose tissue metabolism and adipokine secretion, peroxisome proliferator-activated receptor gamma (PPARg), is activated. This nuclear receptor subtype regroups two isoforms: PPARg1, expressed in many tissues (adipose tissue, muscle, heart and liver) where it controls glucose and lipid homeostasis, and PPARg2, the adipocyte’s specific form, which further governs preadipocyte differentiation, up-regulation of genes involved in lipogenesis and the expression of adipokines. Recent advances showed that increased FA and glycaemia trigger vascular alterations that lead to atherosclerosis. In fact, endothelial cells (EC) and smooth muscle cells (SMC), the main arterial components, are sensitive to metabolic alterations. A lack in insulin sensitivity, leading to lower glucose availability, forces arterial cells to use FA as alternative energy source. Thus, in atheroprone regions susceptible to plaque formation, EC and SMC are subjected to metabolic modifications that lead to oxidized low-density lipoprotein (oxLDL) accumulation in the intima and the progression of vascular disease. Many studies confirmed that the presence of SMC in the atherosclerotic plaque originates from the vascular wall but are showing a distinct phenotype. Even if the role of these cells in atherogenesis is not clear, trans-differentiation of SMC into foam cells has been reported in vitro. Thus, the present study aims at studying a HFD-induced obesity mouse model, developed to evaluate the impact of FA nature on the adipokine secretion profile of adipocytes. We also intended to determine gender-specific impact on modulation of metabolic disorders in response to those diets. On the other hand, we aim to determine the role of adipocytes in the development of obesity-linked atherosclerosis. For that, the second part of this study targeted the effect of adipocytes isolated from mice fed with HFD on SMC physiology. We focused our investigation on the effects of adipocytes regardless of the impact of other cell types in the adipose tissue. To reach our goal, we developed a HFD-fed mouse preparation demonstrating different stages of metabolic disorders leading to T2D. This model allowed us to generate adipocytes with different alteration status, reflected by the modulation of their adipokine secretion profile. Modifications in adipokine secretions were associated with PPAR!2 modulation. These results, reported in both genders, were delayed in female who expressed higher levels of estrogen receptor alpha (ER"). Then, the adipocytes were used to produce conditioned cultured media. To decipher the mechanistic contributions of HFD in adipokines modulation, the potential of adipocytes to induce SMC pathophysiologic disorders was evaluated in SMC stimulated by conditioned cultured media. This protocol enables the transposition of diet-induced fat cell modifications into extended alterations in the physiology of vascular SMC. These results strongly support pro-atherogenic effects of abdominal adipocytes on an important vascular component function through paracrine actions. Thus, adipocytes can be recognized as a link between the pathogenic potential of obesity and the impairments of SMC functions. A better understanding of the pathogenic effects of the adipose tissue on other tissues and organ systems might assist to develop better strategies in treating obesity- induced cardiovascular disease and metabolic syndrome.

Étude de l'impact de combinaisons d'acides gras et de l'insuline sur la fonctionnalité des cellules musculaires lisses vasculaires

L’athérosclérose est étroitement liée au diabète de type 2. De fortes concentrations plasmatiques en acides gras libres (AGL) et en insuline sont des caractéristiques retrouvées chez les patients souffrant de ces deux pathologies. Les AGL, présents dans notre alimentation, font partie de l’environnement auquel les cellules sont exposées. Leurs effets dépendent de leur nature, les acides gras saturés (AGS) étant néfastes et les acides gras monoinsaturés (AGMI) plus protecteurs. Ils ont donc des effets variés sur les cellules musculaires lisses vasculaires (CMLV) impliquées dans la pathogénèse de l’athérosclérose. Ainsi, l’objectif principal de ce projet de maîtrise était d’évaluer l’impact de deux combinaisons d’AGL sur la viabilité des CMLV, en condition hyperinsulinémique ou non. Les deux combinaisons renfermaient les mêmes AGL mais en proportions différentes, l’une étant plus riche en AGS et l’autre en AGMI. Nos résultats ont montré que les combinaisons d’AGL ont un effet pro-apoptotique principalement dû aux AGS. L’acide oléique présent dans les combinaisons atténue cependant cet effet. Il diminue même plus fortement l’apoptose des CMLV lorsqu’associé à un AGS que lorsqu’utilisé seul. Cet impact est significatif uniquement dans certaines proportions de ces AGL et est plus efficace en présence d’insuline. Ces résultats mettent en lumière la présence d’une compétition entre mécanismes anti- et pro-apoptotiques en fonction des proportions d’AGS versus AGMI et de l’insulinémie chez les CMLV. Ils soulignent également l’importance de la présence des AGMI dans les diètes riches en AGS et pourraient être utiles pour l’élaboration de nouvelles diètes adaptées aux patients athérosclérotiques et diabétiques. / Atherosclerosis is closely linked to type 2 diabetes. High plasmatic concentrations of free fatty acids (FFA) and insulin are common features in patients suffering from both diseases. FFA, present in our diet, are part of the environment to which body cells are exposed. Their effects are dependent of their nature, being harmful for saturated fatty acids (SFA) and more protective for monounsaturated fatty acids (MUFA). They can have therefore various effects on vascular smooth muscle cells (VSMC) implicated throughout the development of atherosclerosis. Thus, this study aimed to assess the impact of two FFA combinations on VSMC viability, whether or not in a hyperinsulinemic condition. Both combinations contained the same FFA but in different proportions, one being richer in SFA and the other in MUFA. Our results showed that FFA combinations have a pro-apoptotic impact, mainly due to SFA. However, the presence of oleic acid in the combinations attenuated this effect. Furthermore, oleic acid had the capacity to reduce more strongly VSMC apoptosis when combined with a SFA than when used alone, although only under specific FFA ratios. This impact is even more effective in presence of insulin. These results highlight the presence of a competition between anti- and pro-apoptotic mechanisms dependent of FFA ratios (SFA vs. MUFA) and insulinemia to which are exposed VSMC. They also underline the importance of the presence of MUFA such as oleic acid in diets rich in SFA and could be useful for the development of new diets adapted to atherosclerotic and diabetic patients.

CXCL16 and CD137 in atherosclerosis

Atherosclerosis is a progressive inflammatory disease that is characterized by the accumulation of lipids, infiltrated cells and fibrous elements in large arteries. This thesis focuses on the molecular mechanisms behind foam cell formation and inflammation, two central processes in the development of atherosclerosis. More specific, we studied the effects of proinflammatory cytokines on CXCL16 expression and its role as scavenger receptor on macrophages and smooth muscle cells in atherogenesis. CXCL16 is defined as a chemokine and a scavenger receptor, regulating adhesion and chemoattraction of CXCR6 expressing cells and uptake of oxLDL. We show that the expression of CXCL16 and its receptor CXCR6 are more pronounced in human atherosclerotic lesions compared with non-atherosclerotic vessels. Increased expression of CXCL16 was also seen in atherosclerotic aortas of apoE-/- mice compared with aortas of non-atherosclerotic, age-matched C57BL/6 mice. In vitro, IFN gamma induced CXCL16 expression in primary human monocytes and smooth muscle cells which resulted in an increased uptake of oxLDL. Treatment of mice with IFN gamma also induced CXCL16 expression in atherosclerotic lesions. Thus, we have demonstrated a role for IFN gamma in foam cell formation through upregulation of CXCL16. The expression of CXCR6 was defined to the same regions as for CXCL16 in the lesion, indicating the presence of cells able to respond to CXCL16. Consequently, CXCL16 could serve as a molecular link between lipid metabolism and immune activity in atherosclerotic lesion. CD137 belongs to the TNF family and mediates several important processes in inflammation. CD137 is involved in the activation of T cells, NK cells, B cells and monocytes and regulate cytokine production, proliferation and apoptosis in these cells. A limited number of studies have demonstrated CD137 expression on smooth muscle cells and endothelial cells. Our results show that CD137 mRNA is higher expressed in human atherosclerotic lesions compared with unaffected vessels. We found that endothelial cells express CD137 in atherosclerotic lesions and that cultured endothelial cells and smooth muscle cells express CD137 and CD137 ligand in vitro. CD137 was regulated differentially by proinflammatory cytokines (i.e. IFN gamma, TNF alpha, IL-1 beta) and bacterial lipopolysaccharide depending on cell type. Furthermore, we investigated the effects of CD137 signalling, demonstrating that binding of the CD137 ligand to its receptor increases proliferation and migration of smooth muscle cells. In summary, this thesis has focused on the expression, regulation and role of CXCL16 and CD137, two genes that have not been described earlier in the concept of atherosclerosis. The findings demonstrate some of the molecular mechanisms involved in vascular inflammation and may increase our knowledge about the development of atherosclerosis.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A fibrose hepática é o resultado de uma resposta cicatrizante frente a repetidas lesões no fígado, e é caracterizada pelo acúmulo excessivo de proteínas da matriz extracelular (MEC) no parênquima hepático, incluindo colágeno, fibronectina, elastina, laminina e proteoglicanos, com a participação de diferentes populações celulares do fígado. As principais células responsáveis pela síntese de proteínas da MEC na fibrose hepática são as células estreladas hepáticas ativadas e os miofibroblastos, que surgem após estímulo inflamatório e são caracterizadas pela expressão de alfa-actina de músculo liso (&#945;-SMA). Sabe-se que durante a progressão da fibrose hepática, ocorre a morte de hepatócitos e sua substituição por células fibrogênicas &#945;-SMA+. A apoptose dessas células fibrogênicas é de grande relevância para a regressão da fibrose e regeneração hepática. Nos últimos anos, a terapia com células tronco de medula óssea tem sido utilizada para estimular a regeneração hepática em diferentes modelos experimentais e protocolos clínicos. A fração mononuclear da medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas e células-tronco mesenquimais. O objetivo deste estudo foi analisar a expressão de &#945;-SMA e o processo de apoptose de células hepáticas durante a fibrose hepática induzida por ligadura do ducto biliar (LDB) e após o transplante de células mononucleares de medula óssea (CMMO). Os fígados foram coletados de ratos dos seguintes grupos: normal, 14 dias de LDB, 21 dias de LDB e animais que receberam CMMO após 14 dias de LDB, e foram analisados após 7 dias (totalizando 21 dias de LDB). Para quantificar a expressão de &#945;-SMA por células fibrogênicas nos grupos experimentais, foi realizada imunoperoxidase para &#945;-SMA, seguida de morfometria no programa Image Pro Plus. Para analisar a apoptose nas células hepáticas, foi realizada imunoperoxidase e Western Blotting (WB) para caspase-3 (proteína apoptótica) e imunofluorescência com dupla-marcação para caspase-3 e &#945;-SMA, seguida de observação em microscópio confocal. Os resultados da quantificação de &#945;-SMA por morfometria mostraram que a expressão de &#945;-SMA aumentou significativamente 14 e 21 dias após a LDB. Entretanto, essa expressão diminuiu significativamente no grupo tratado com CMMO, que apresentou parênquima hepático mais preservado em relação ao grupo com 21 dias de LDB. Os resultados de imunoperoxidase, WB e microscopia confocal para expressão de caspase-3 demonstraram que essa proteína diminuiu nos animais fibróticos com 14 e 21 dias de LDB com relação ao grupo normal, e estava significativamente elevada no grupo tratado com CMMO. These proteins include collagen, fibronectin, elastin, laminin and proteoglicans, along with the participation of different cell populations within the liver. The main cells responsible for the synthesis of MEC proteins are activated hepatic stellate cells and myofibroblasts, which appear after inflammatory stimuli and are characterized by the expression of alpha-smooth muscle actin (&#945;-SMA). It is known that hepatic fibrosis progression is accompanied by hepatocyte death and its substitution by &#945;-SMA+ fibrogenic cells. Therefore, apoptosis of these fibrogenic cells is of main relevance to fibrosis regression and hepatic regeneration. In the later years, bone marrow stem cell therapy has been used to stimulate hepatic regeneration in different experimental models and clinical protocols. The adult bone marrow mononuclear fraction contains two stem cell populations particularly important in the treatment of diverse hepatic diseases: hematopoietic stem cells and mesenchymal stem cells. The aim of this study was to analyze &#945;-SMA expression and the apoptotic process in hepatic cells during hepatic fibrosis induced by bile duct ligation (BDL) and after bone marrow mononuclear cell (BMMC) transplantation. Livers were collect from rats of the following groups: normal, 14 days of BDL, 21 days of BDL and rats that received BMMC 14 days after BDL and were analyzed after 7 days (total of 21 days of BDL). To quantify &#945;-SMA expression by fibrogenic cells in the experimental groups, immunoperoxidase to &#945;-SMA followed by morphometry in the Image Pro Plus software was performed. To analyze apoptosis in hepatic cells, immunoperoxidase and western blotting (WB) against caspase-3 (apoptotic protein) were used, along with double immunofluorescence against caspase-3 and &#945;-SMA to confocal microscopy analysis. Results of &#945;-SMA quantification by morphometry showed that &#945;-SMA expression increased significantly 14 and 21 days after BDL. However, this expression was significantly decreased in the BMMC treated group, which presented a more preserved hepatic parenchyma in relation to the group with 21 days of BDL. Immunoperoxidase, WB and confocal microscopy results showed that caspase-3 is decreased in fibrotic livers with 14 and 21 days of BDL in comparison to normal group, and was significantly augmented in the BMMC treated group. Confocal microscopy analysis showed that were cells coexpressing &#945;-SMA and caspase-3 in rats treated with BMMC, suggesting fibrogenic cells death and hepatic remodeling.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A fibrose hepática é o resultado de uma resposta cicatrizante frente a repetidas lesões no fígado, e é caracterizada pelo acúmulo excessivo de proteínas da matriz extracelular (MEC) no parênquima hepático, incluindo colágeno, fibronectina, elastina, laminina e proteoglicanos, com a participação de diferentes populações celulares do fígado. As principais células responsáveis pela síntese de proteínas da MEC na fibrose hepática são as células estreladas hepáticas ativadas e os miofibroblastos, que surgem após estímulo inflamatório e são caracterizadas pela expressão de alfa-actina de músculo liso (&#945;-SMA). Sabe-se que durante a progressão da fibrose hepática, ocorre a morte de hepatócitos e sua substituição por células fibrogênicas &#945;-SMA+. A apoptose dessas células fibrogênicas é de grande relevância para a regressão da fibrose e regeneração hepática. Nos últimos anos, a terapia com células tronco de medula óssea tem sido utilizada para estimular a regeneração hepática em diferentes modelos experimentais e protocolos clínicos. A fração mononuclear da medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas e células-tronco mesenquimais. O objetivo deste estudo foi analisar a expressão de &#945;-SMA e o processo de apoptose de células hepáticas durante a fibrose hepática induzida por ligadura do ducto biliar (LDB) e após o transplante de células mononucleares de medula óssea (CMMO). Os fígados foram coletados de ratos dos seguintes grupos: normal, 14 dias de LDB, 21 dias de LDB e animais que receberam CMMO após 14 dias de LDB, e foram analisados após 7 dias (totalizando 21 dias de LDB). Para quantificar a expressão de &#945;-SMA por células fibrogênicas nos grupos experimentais, foi realizada imunoperoxidase para &#945;-SMA, seguida de morfometria no programa Image Pro Plus. Para analisar a apoptose nas células hepáticas, foi realizada imunoperoxidase e Western Blotting (WB) para caspase-3 (proteína apoptótica) e imunofluorescência com dupla-marcação para caspase-3 e &#945;-SMA, seguida de observação em microscópio confocal. Os resultados da quantificação de &#945;-SMA por morfometria mostraram que a expressão de &#945;-SMA aumentou significativamente 14 e 21 dias após a LDB. Entretanto, essa expressão diminuiu significativamente no grupo tratado com CMMO, que apresentou parênquima hepático mais preservado em relação ao grupo com 21 dias de LDB. Os resultados de imunoperoxidase, WB e microscopia confocal para expressão de caspase-3 demonstraram que essa proteína diminuiu nos animais fibróticos com 14 e 21 dias de LDB com relação ao grupo normal, e estava significativamente elevada no grupo tratado com CMMO. Endothelial TRPV4 dysfunction in a streptozotocin-diabetic Rat Model

Diabetes mellitus is a complex disease characterised by chronic hyperglycaemia due to compromised insulin synthesis and secretion, or decreased tissue sensitivity to insulin, if not all three conditions. Endothelial dysfunction is a common complication in diabetes in which endothelium-dependent vasodilation is impaired. The aim of this study was to examine the involvement of TRPV4 in diabetes endothelial dysfunction. Male Charles River Wistar rats (350-450 g) were injected with 65mg/kg streptozotocin (STZ) intraperitoneally. STZ-injected rats were compared with naïve rats (not injected with STZ) or control rats (injected with 10ml/kg of 20mM citrate buffer, pH 4.0-4.5), if not both. Rats with blood glucose concentrations greater than 16mmol/L were considered to be diabetic. As the results revealed, STZ-diabetic rats showed significant endothelial dysfunction characterised by impaired muscarinic-induced vasodilation, as well as significant impairment in TRPV4-induced vasodilation in aortic rings and mesenteric arteries. Furthermore, STZ-diabetic primary aortic endothelial cells (ECs) showed a significant reduction in TRPV4-induced intracellular calcium ([Ca2+]i) elevation. TRPV4, endothelial nitric oxide synthase (eNOS), and caveolin-1 (CAV-1) were also significantly downregulated in STZ-diabetic primary aortic ECs and were later significantly restored by in vitro insulin treatment. Methylglyoxal (MGO) was significantly elevated in STZ-diabetic rat serum, and nondiabetic aortic rings incubated with MGO (100μM) for 12 hours showed significant endothelial dysfunction. Moreover, nondiabetic primary aortic ECs treated with MGO (100μM) for 5 days showed significant TRPV4 downregulation and significant suppression of 4-α-PDD-induced [Ca2+]i elevation, which was later restored by L-arginine (100μM) co-incubation. Incubating nondiabetic aortic rings with MGO (100μM) for 2 hours induced a spontaneous loss of noradrenaline-induced contractility persistence. Moreover, MGO induced significant [Ca2+]i elevation in Chinese hamster ovary cells expressing rat TRPM8 channels (rTRPM8), which was significantly inhibited by AMTB (1-5μM). Taken together, TRPV4, CAV-1, and eNOS can form a functional complex that is downregulated in STZ-diabetic aortic ECs and restored by insulin treatment. MGO elevation might furthermore contribute to diabetes endothelial dysfunction and TRPV4 downregulation. By contrast, MGO induced the loss of contractility persistence, possibly due to MGO's acting as a TRPM8 agonist.

Alterações renais gênero-dependentes em ratos com insuficiência renal crônica / Gender-dependent renal alterations in rats with chronic renal failure

A insuficiência renal crônica (IRC) é caracterizada por alterações glomerulares secundárias aos mecanismos adaptativos ocasionados por perda de néfrons funcionantes. Alterações na hemodinâmica glomerular, proliferação celular, influxo de células inflamatórias, desequilíbrio na síntese de proteínas da matriz extracelular glomerular (MECG) e perda da seletividade de carga e/ou tamanho da membrana basal glomerular têm sido apontados como mecanismos envolvidos na expansão mesangial e conseqüente glomeruloesclerose. A participação dos hormônios sexuais na função renal e na evolução da insuficiência renal crônica tem sido sugerida. Os glicosaminoglicanos, especialmente o heparan sulfato (HS), têm sido associados à seletividade glomerular de macromoléculas. O remodelamento podocitário precoce e a proteinuria (PTN) se relacionam com a progressão da IRC. Neste contexto, o acúmulo de MECG, proliferação de miofibroblastos e PTN têm sido apontados como mediadores precoces que precedem as lesões glomerulares e túbulo-intersticiais. Neste estudo, avaliamos as alterações renais precoces (30 dias de IRC) gênero-dependentes em ratos (M) e ratas (F) Wistar submetidos à redução de 5/6 da massa renal (IRC) e à castração (c). Os animais foram divididos em 10 grupos: Controles (C) (CM, CF, CMc, CFc) e sham (CM sham, CF sham); e aqueles submetidos à nefrectomia 5/6: IRCM, IRCF, IRCMc, IRCFc. Os animais foram castrados com 5 semanas e submetidos à nefrectomia 5/6 com 7 semanas de idade. Resultados significativos mostraram que os machos com IRC apresentaram maior PTN, acompanhada de maior comprometimento mesangial, imunomarcação positiva para &#945;-actina e maior concentração de heparan sulfato (HS) comparados com as fêmeas IRC (p<0,05). Estas alterações foram reduzidas nos machos castrados. A análise da morfologia podocitária mostrou raras regiões onde ocorreram alterações podocitárias nos grupos IRC. O conjunto de dados sugere que o hormônio masculino pode participar na manutenção do equilíbrio mesangial e que a PTN participa do processo de expansão mesangial. Adicionalmente, a maior concentração de HS nos machos com IRC sugere que durante o processo de remodelação da MEG, tenha ocorrido geração de HS de novo, funcionalmente defeituoso, comprometendo a barreira de filtração glomerular, corroborando com a perda de seletividade da mesma e, contribuindo para maior PTN neste grupo. As fêmeas com IRC apresentaram alterações mais discretas quando comparadas aos machos; apresentaram decréscimo de HS renal associado a PTN e a castração não alterou este perfil. Em resumo, a PTN ocorre precocemente na IRC, contribuindo para o desequilíbrio da MECG. Os mecanismos envolvidos nestes processos parecem sofrer influência dos hormônios sexuais; e os hormônios masculinos parecem agravar estas alterações, contribuindo possivelmente para um pior prognóstico da doença renal nos machos. / Chronic renal failure (CRF) is characterized by adaptive mechanisms secondary to the loss of functioning nephrons. Glomerular hemodynamics alterations, cellular proliferation, inflammatory cells influx, imbalance between synthesis and degradation of the glomerular extracellular matrix (GECM) and loss of charge and/or size selectivity of the glomerular basal membrane are pointed as mechanisms leading to mesangial expansion and glomerulosclerosis. Additionally, participation of gender related hormones on renal function and progression of CRF have been suggested. We evaluated the effect of castration in renal alterations in males (M) and females (F) Wistar rats, after 30 days of 5/6 reduction of renal mass (CRF). The animals were castrated (c) at 5 weeks old and 7 weeks old 5/6 and sham nephrectomy were done. Groups: Control (C) CM, CM sham, CMc, CF, CF sham, CFc, CRFM, CRFMc, CRFF, CRFFc. CRFM group showed higher proteinuria followed by increased mesangial expansion and &#945;-actin immunostaining. Concomitant higher concentration of heparan sulfate (HS) was also observed when compared to CRFF (p<0.05). These alterations were reduced in CRFMc group. Podocyte morphology analysis through electronic microscopy showed few disorders of foot processes in CRF groups Overall, CRFF group showed fewer alterations compared to males, and a reduction of HS was observed in association with PTN. Castration did not change this profile in female rats. Data suggest that male hormones may participate in the maintenance of the mesangial equilibrium and that PTN collaborated with the mesangial expansion process. Additionally, the higher concentration of HS in CRFM suggest that the remodeling process of the GECM, included a synthesis of de novo HS, that presented a functioning defect, compromising the glomerular filtration barrier and, ultimately corroborated with the loss of its selectivity and consequently with a higher PTN. This set of results leads us to conclude that PTN appears early in the course of CRF, may contribute to renal GECM imbalance and, the mechanisms involved in these processes seem to be influenced by gender-related hormones. In addition, male hormones seem to aggravate renal alterations contributing to a poor prognosis of CRF progression in male rats.
