Modificações epigenéticas da cromatina e sua relação com a reprogramação nuclear de bovinos / Epigenetic modifications of chromatin and their relation with the nuclear reprogramming of bovine

A reprogramação nuclear de uma célula somática a um estado embrionário tem diversas aplicações, como pesquisas básicas na biologia do desenvolvimento, terapia celular, melhoramento genético em animais de produção e conservação de espécies. As principais técnicas utilizadas para a reprogramação nuclear são a transferência nuclear de células somáticas (TNCS) e a geração de células tronco pluripotente induzidas (iPS). Muitos trabalhos têm mostrado uma baixa eficiência no processo de reprogramação nuclear nas duas técnicas, além disso, modificações epigenéticas tem sido apontada como a principal barreira para uma reprogramação nuclear eficiente. Por esse motivo, medidas como a utilização de células menos diferenciadas e/ou alteração do perfil epigenético das células somáticas podem aumentar a eficiência destas técnicas. Por isso, o objetivo deste trabalho foi investigar a influência de marcas epigenéticas em células bovinas utilizadas na reprogramação nuclear mediada por TNCS ou superexpressão de genes relacionados a pluripotêcia (iPS). Para isso, utilizamos 3 abordagens. Primeiro, analisamos marcações epigenéticas relacionadas ao desenvolvimento embrionário e pluripotência (H3K9me2, H3K9me3, H3K9ac, 5mC e 5hmC) em diferentes tipos celulares, analisamos a expressão gênica de genes responsáveis por essas marcações em células de diferentes tecidos (ex. células tronco mesenquimais (MSC) e fibroblastos) e as utilizamos como doadoras de núcleo na TNCS. Na segunda e a terceira abordagem, utilizamos células com menores níveis de H3K9me2 para a geração de iPS e na TNCS, respectivamente. Além disso, por se mostrar eficiente na TNCS, analisamos o efeito da sincronização do ciclo celular por privação de soro fetal bovino (SFB) na geração de células iPS. Com o intuito de diminuir os níveis de H3K9me2, as células foram tratadas com UNC0638, um inibidor especifico das metiltransferases de histona G9a/GLP. Nossos resultados do primeiro experimento mostraram que as MSC podem ser utilizadas como doadoras de núcleo na TNCS, no entanto, mesmo com algumas diferenças na expressão gênica em relação aos fibroblastos, a produção de blastocistos não foi diferente entre as duas células. No segundo experimento, as células privadas de SFB geraram mais colônias que as células controle, enquanto que as células tratadas não apresentaram diferença. Por último, as células tratadas com o UNC0638 apresentaram um menor nível de metilação no DNA em zigotos em relação às células controle. Os resultados encontrados neste trabalho podem contribuir para o melhor entendimento dos mecanismos epigenéticos envolvidos na reprogramação nuclear de bovinos / Nuclear reprogramming of somatic cells to embryonic state has several aplications, such as basic research on developmental biology, cell therapy, genetic improvement in livestock animals and preservation of endangered species. The principal techniques utilized to achieve nuclear reprogramming are Somatic Cell Nuclear Transfer (SCNT) and induced pluripotency. Several works has reported low efficiency rates of nuclear reprogramming when these techniques are used to reprogram somatic cells. Moreover, epigenetic modifications acquired during development act as epigenetic barrier to the complete reprogramming process. For this reason, strategies such as use of less differentiated cells and/or modification of epigenetic profile of somatic cells might increase the efficiency these techniques. The objective of this work was investigate the influence of epigenetic marks in bovine cells utilized on nuclear reprogramming experiments mediated by SCNT or induced pluripotency. To investigate it, we used three approaches. First, we analyzed the epigenetic marks related to the embryonic development and pluripotency (e.g H3K9me2, H3K9me3, H3K9ac, 5mC and 5hmC), gene expression of genes involved in these epigenetic marks in different tissues (i.e. mesenchymal stem cells (MSC) and fibroblasts) and their use as nuclear donor cells on SCNT procedure. Regarding the second and the third approach, we utilized cells with reduced levels of H3K9me2 to generate iPS cells and cloned embryos, respectively. Furthermore, since serum starvation has been demonstrated increase SCNT developmental rates, we assessed the effect of cell cycle synchronization mediated by serum starvation on nuclear reprogramming using iPS cells. Aiming decrease the levels of H3K9me2, cells were treated with UNC0638, a chemical probe that works as a specific inhibitor of the histone methyltransferases G9a and its counterpartner GLP. Our results showed that MSC are suitable to be used as nuclear donors on SCNT procedures, however, in spite of differences on gene expression comparing with fibroblasts, the embryonic developmental rates were not improved. On the second experiment, cells privated of fetal calf serum produced more iPS cells colonies than control cells, whereas cells treated with UNC did not show differences when compared with untreated cells. Lastly, UNC treated donor cells treated produced cloned zygotes with lower levels of DNA methylation compared to zygotes derivated from untreated cells. The results presented here will contribute to the better understanding of the epigenetic mechanisms involved on bovine nuclear reprogramming

Normative Instruction 62 establishes maximum levels of somatic cell count of 500,000 cells / ml of milk, minimum fat content of 3.0%, total solids 11.4%, 2.9% protein, dry extract 8.4%. The somatic cell count reflects the health of the mammary gland of the animal. Of the 108 samples of milk collected from cows participants on the last day of the milk match, after complete milking and weighing of milk, 24% of the animals showed somatic cell counts that do not fit the parameters established by the Normative Instruction 62. The other parameters evaluated as lactose, fat, total solids and nonfat dry extract showed significant differences by comparing the milk with high somatic cell count, milk with low somatic cell count. The level of milk protein with high somatic cell count remained in the Normative Instruction 62 standard, because of the increase of plasma proteins in milk as a result of the inflammatory response. Most milk samples from dairy tournaments did not fit in Instruction 62, as the increase in somatic cells interfered in the chemical physical quality. / Objetivou-se nesta pesquisa avaliar as características físico-químicas do leite de vacas holandesas e mestiças oriundas de torneios leiteiros na região do Sul de Minas Gerais, no período de março a outubro de 2012, sendo utilizados os dados não paramétricos para as variáveis de proteína, gordura, lactose, sólidos totais, extrato seco desengordurado e contagem de células somáticas. A Instrução Normativa 62 estabelece níveis máximo de contagem de células somáticas de 500.000 células/mL de leite, teores mínimos de gordura de 3,0%, sólidos totais de 11,4%, proteína 2,9%, extrato seco desengordurado 8,4%. A contagem de células somáticas reflete na saúde da glândula mamária do animal. Das108 amostras de leite coletadas das vacas participantes no último dia do torneio leiteiro, após a ordenha completa e pesagem do leite, 24% dos animais apresentaram contagem de células somáticas que não se enquadram nos parâmetros estabelecidos pela Instrução Normativa 62. Os demais parâmetros avaliados como lactose, gordura, sólidos totais e extrato seco desengordurado não apresentaram diferenças significativas, comparando o leite com contagem de células somáticas altas, com leite com contagem de células somáticas baixas. O nível de proteína em leite com contagem de células somáticas altas se mantiveram no padrão da Instrução Normativa 62, pois houve um aumento das proteínas plasmáticas no leite em decorrência da resposta inflamatória. Conclui-se que a maioria das amostras de leite coletadas em torneios leiteiro não se enquadraram na Instrução Normativa 62.

Personhood and Cloning: Modern Applications and Ethics of Stem Cell and Cloning Technology

Within many communities and religions, including the LDS community, there is some controversy surrounding the use of stem cells – particularly embryonic stem cells (ESC). Much of this controversy arises from confusion and misconceptions about what stem cells actually are, where they come from , and when life begins. The theology of the Church of Jesus Christ of Latter-day Saints has interesting implications for the last of these considerations, and it becomes less a question of “when does life begin” and more an exploration of “when does personhood begin” or “when does the spirit enter the body.” With no official Church stance, statements from Church leaders vary on this topic, and this first section of the thesis explores the philosophical and practical meaning of personhood with a biological background intended for those not familiar with the origin or uses of stem cells.The second portion of the thesis explores possible cloning technologies. Recent events and advances address the possibility of cloning endangered and extinct species. The ethics of these types of cloning have considerations uniquely different from the type of cloning commonly practiced. Cloning of cheetahs (and other endangered or vulnerable species) may be ethically appropriate, given certain constraints. However, the ethics of cloning extinct species varies; for example, cloning mammoths and Neanderthals is more ethically problematic than conservation cloning, and requires more attention. Cloning Neanderthals in particular is likely unethical and such a project should not be undertaken. It is important to discuss and plan for the constraints necessary to mitigate the harms of conservation and extinct cloning, and it is imperative that scientific and public discourse enlighten and guide actions in the sphere of cloning.

Delivery of CRISPR/Cas9 RNAs into Blood Cells of Zebrafish: Potential for Genome Editing in Somatic Cells

Factor VIII is a clotting factor found on the intrinsic side of the coagulation cascade. A mutation in the factor VIII gene causes the disease Hemophilia A, for which there is no cure. The most common treatment is administration of recombinant factor VIII. However, this can cause an immune response that renders the treatment ineffective in certain hemophilia patients. For this reason a new treatment, or cure, needs to be developed. Gene editing is one solution to correcting the factor VIII mutation. CRISPR/Cas9 mediated gene editing introduces a double stranded break in the genomic DNA. Where this break occurs repair mechanisms cause insertions and deletions, or if a template oligonucleotide can be provided point mutations could be introduced or corrected. However, to accomplish this goal for editing factor VIII mutations, a way to deliver the components of CRISPR/Cas9 into somatic cells is needed. In this study, I confirmed that the CRISPR/Cas9 system was able to create a mutation in the factor VIII gene in zebrafish. I also showed that the components of CRISPR/Cas9 could be piggybacked by vivo morpholino into a variety of blood cells. This study also confirmed that the vivo morpholino did not interfere with the gRNA binding to the DNA, or Cas9 protein inducing the double stranded break.

Facteurs de risque modifiables associés à l’incidence, l’élimination et la prévalence d’infections intra-mammaires chez la vache laitière en lactation

La mammite subclinique est un problème de santé fréquent et coûteux. Les infections intra-mammaires (IIM) sont souvent détectées à l’aide de mesures du comptage des cellules somatiques (CCS). La culture bactériologique du lait est cependant requise afin d’identifier le pathogène en cause. À cause de cette difficulté, pratiquement toutes les recherches sur la mammite subclinique ont été centrées sur la prévalence d’IIM et les facteurs de risque pour l’incidence ou l’élimination des IIM sont peu connus. L’objectif principal de cette thèse était d’identifier les facteurs de risque modifiables associés à l’incidence, l’élimination et la prévalence d’IIM d’importance dans les troupeaux laitiers Canadiens. En premier lieu, une revue systématique de la littérature sur les associations entre pratiques utilisées à la ferme et CCS a été réalisée. Les pratiques de gestion constamment associées au CCS ont été identifiées et différentiées de celles faisant l’objet de rapports anecdotiques. Par la suite, un questionnaire bilingue a été développé, validé, et utilisé afin de mesurer les pratiques de gestion d’un échantillon de 90 troupeaux laitiers canadiens. Afin de valider l’outil, des mesures de répétabilité et de validité des items composant le questionnaire ont été analysées et une évaluation de l’équivalence des versions anglaise et française a été réalisée. Ces analyses ont permis d’identifier des items problématiques qui ont du être recatégorisés, lorsque possible, ou exclus des analyses subséquentes pour assurer une certaine qualité des données. La plupart des troupeaux étudiés utilisaient déjà la désinfection post-traite des trayons et le traitement universel des vaches au tarissement, mais beaucoup des pratiques recommandées n’étaient que peu utilisées. Ensuite, les facteurs de risque modifiables associés à l’incidence, à l’élimination et à la prévalence d’IIM à Staphylococcus aureus ont été investigués de manière longitudinale sur les 90 troupeaux sélectionnés. L’incidence d’IIM semblait être un déterminant plus important de la prévalence d’IIM du troupeau comparativement à l’élimination des IIM. Le port de gants durant la traite, la désinfection pré-traite des trayons, de même qu’une condition adéquate des bouts de trayons démontraient des associations désirables avec les différentes mesures d’IIM. Ces résultats viennent souligner l’importance des procédures de traite pour l’obtention d’une réduction à long-terme de la prévalence d’IIM. Finalement, les facteurs de risque modifiables associés à l’incidence, à l’élimination et à la prévalence d’IIM à staphylocoques coagulase-négatif (SCN) ont été étudiés de manière similaire. Cependant, afin de prendre en considération les limitations de la culture bactériologique du lait pour l’identification des IIM causées par ce groupe de pathogènes, une approche semi-Bayesienne à l’aide de modèles de variable à classe latente a été utilisée. Les estimés non-ajusté de l’incidence, de l’élimination, de la prévalence et des associations avec les expositions apparaissaient tous considérablement biaisés par les imperfections de la procédure diagnostique. Ce biais était en général vers la valeur nulle. Encore une fois, l’incidence d’IIM était le principal déterminant de la prévalence d’IIM des troupeaux. Les litières de sable et de produits du bois, de même que l’accès au pâturage étaient associés à une incidence et une prévalence plus basse de SCN. / Subclinical mastitis is a very frequent and costly health issue that can be detected using somatic cell count (SCC) measurements, but requires the use of milk bacteriological culture for identification of the causal pathogen. Because of this latter difficulty, nearly all subclinical mastitis research has focused on prevalent intramammary infections (IMI) and less is known on risk factors for IMI incidence or elimination. The main objective of this thesis was to identify the manageable risk factors associated with the incidence, elimination, and prevalence of IMI of importance in Canadian dairy herds. First, a systematic review of the literature on the associations between management practices used on farms and SCC was carried out. Management practices consistently associated with SCC were identified and differentiated from other practices for which anecdotic reports were available. Then, a bilingual questionnaire was developed, validated, and employed to measure the practices used on a sample of 90 Canadian dairy herds. To validate this tool, measures of repeatability and of validity of the questionnaire’s items were analyzed and an evaluation of the equivalence of the English and French versions was conducted. These analyses indicated that the questionnaire was, in general, acceptable, but also pinpointed some problematic items. These items were recategorized when possible or otherwise excluded from subsequent analyses to ensure good data quality. Most of the herds studied were already using post-milking teat disinfection and blanket dry cow therapy. Many other frequently recommended practices were not widely adopted. Next, manageable risk factors associated with Staphylococcus aureus IMI incidence, elimination, and prevalence were investigated on the 90 selected herds in a longitudinal fashion. The S. aureus IMI incidence appeared to be a stronger determinant of the herd prevalence than the elimination rate. Among other practices, wearing gloves during milking, using pre-milking teat disinfection, and having an adequate teat end condition showed desirable associations with the outcomes. These results highlight the importance of good milking practices to achieve a long-term reduction of S. aureus IMI prevalence. Finally, manageable risk factors associated with coagulase-negative staphylococci (CNS) IMI incidence, elimination, and prevalence were investigated in a similar manner. Because of the greater limitations of milk bacteriological culture to identify IMI caused by this group of pathogens, a semi-Bayesian latent class model approach was used. Sensitivity and specificity estimates from an internal validation study were used to link the observed IMI milk culture result to the latent true quarter IMI status. Non-adjusted estimates of IMI incidence, elimination, prevalence, and of associations with expositions appeared to be considerably biased by the diagnostic procedure’s imperfections. Most often, estimates were biased toward the null value. Again, IMI incidence was the main determinant of the herd IMI prevalence. Sand and wood products bedding were associated with lower CNS incidence and prevalence. Sending cows to pasture was also associated with lower CNS IMI incidence and prevalence.

Impact d’une infection intra-mammaire causée par Staphylococcus aureus ou un staphylocoque coagulase-négative présente en début de lactation chez les taures laitières

L’objectif de cette étude était de déterminer l’impact d’une infection intra-mammaire (IIM) subclinique causée par staphylocoque coagulase-négative (SCN) ou Staphylococcus aureus diagnostiquée durant le premier mois de lactation chez les taures sur le comptage de cellules somatiques (CCS), la production laitière et le risque de réforme durant la lactation en cours. Des données bactériologiques provenant d’échantillons de lait composites de 2 273 taures Holstein parmi 50 troupeaux ont été interprétées selon les recommandations du National Mastitis Council. Parmi 1 691 taures rencontrant les critères de sélection, 90 (5%) étaient positives à S. aureus, 168 (10%) étaient positives à SCN et 153 (9%) étaient négatives (aucun agent pathogène isolé). Le CCS transformé en logarithme népérien (lnCCS) a été modélisé via une régression linéaire avec le troupeau comme effet aléatoire. Le lnCCS chez les groupes S. aureus et SCN était significativement plus élevé que dans le groupe témoin de 40 à 300 jours en lait (JEL) (P < 0.0001 pour tous les contrastes). La valeur journalière du lnSCC chez les groupes S. aureus et SCN était en moyenne 1.2 et 0.6 plus élevé que le groupe témoin respectivement. Un modèle similaire a été réalisé pour la production laitière avec l’âge au vêlage, le trait génétique lié aux parents pour la production laitière et le logarithme népérien du JEL de la pesée inclus. La production laitière n’était pas statistiquement différente entre les 3 groupes de culture de 40 à 300 JEL (P ≥ 0.12). Les modèles de survie de Cox ont révélé que le risque de réforme n’était pas statistiquement différent entre le groupe S. aureus ou SCN et le groupe témoin (P ≥ 0.16). La prévention des IIM causées par SCN et S. aureus en début de lactation demeure importante étant donné leur association avec le CCS durant la lactation en cours. / The objective of this study was to determine the effect of a subclinical intramammary infection (IMI) caused by coagulase-negative staphylococci (CNS) or Staphylococcus aureus diagnosed during the first month of lactation in heifers on somatic cell count (SCC), milk production and culling risk during the entire first lactation. Bacteriological analysis data of composite milk samples taken from 2,273 Hostein heifers among 50 dairy herds were interpreted according to the National Mastitis Council guidelines. Among the 1,691 heifers meeting the selection criteria, 90 (5%) were diagnosed with S. aureus, 168 (10%) with CNS, and 153 (9%) were negative (no pathogen isolated). Test-day SCC transformed in natural logarithm (lnCCS) was fit in a linear regression model with herd as random effect. The lnSCC in S. aureus and CNS groups were significantly higher than in negative group from 40 to 300 days in milk (DIM) (P < 0.0001 for all contrasts). At test-day level, lnSCC in S. aureus and CNS groups was on average 1.2 and 0.6 higher than the negative group respectively. A similar model was used for milk yield with age at calving, parent average genetic value for milk yield and natural logarithm of tested DIM included. Milk yield was not statistically different between culture groups from 40 to 300 DIM (P ≥ 0.12). Compared with negative heifers, the culling hazard ratio estimated using Cox survival analysis in S. aureus and CNS infected heifers was not significant (P ≥ 0.16). Prevention of CNS or S. aureus IMI in early lactation remains important for its association with SCC during the ensuing lactation.

Controle epigenético do gene imprinted SNRPN durante o desenvolvimento e reprogramação nuclear em equídeos / Epigenetic control of the SNRPN imprinted gene during developmental and nuclear reprogramming in equids

A tranferência nuclear de células somáticas (TNCS) está sendo utilizada para produzir cavalos de elite. No entanto, durante este procedimento pode ocorrer a perfuração da zona pelúcida, levando, ocasionalmente, à secção da massa celular interna, e conseqüente derivação de gêmeos monozigóticos. Além de serem relatadas alterações no processo de imprinting genômico, que conduzem ao desenvolvimento de doenças. Com a descoberta da possibilidade de reprogramar as células somáticas a um estado de pluripotência (iPSCs), estas células passaram a ser muito utilizadas em pesquisas de neurociência. Contudo, também ocorrem modificações epigenéticas durante esta reprogramação celular. Portanto, nossas hipóteses são que os gêmeos eqüinos gerados pela TNCS podem levar às irregularidades no desenvolvimento do sistema nervoso. O padrão de metilação do SNRPN nas estruturas dos fetos muares clonados, e as células iPSCs são diferentes dos padrões encontrados nos muares analisados. A expressão dos genes SNRPN, Necdin e UBE3A são maiores no cérebro, enquanto a expressão do H19 é maior nas membranas extra-embrionárias. Em nosso estudo, obtivemos duas gestações gemelares equinas derivadas da TNCS, que foram interrompidas com 40 e 60 dias de gestação, e comparados com gestações eqüinas únicas de idade similar. Diferenças no comprimento entre os embriões gêmeos foram observadas aos 40 (2.0 e 2.2 cm 10%) e aos 60 (6,5 e 8,5 cm 24%) dias de gestação. Somente o plexo coróide do quarto ventrículo apresentou-se mais desenvolvido nos fetos com maior comprimento. Ao analisarmos fetos muares clonados em diferentes idades gestacionais e compará-los com muares, nos períodos embrionário, fetal e adulto, não foi observada diferença no padrão de metilação do gene SNRPN. No entanto, na décima passagem das células iPSC o padrão de metilação alterou, em relação aos muares estudados e ao padrão observado nos fibroblastos. Ao analisarmos os fetos clonados nas diferentes idades gestacionais observou-se no cérebro menor expressão dos gene H19 e UBE3A, e maior expressão do gene SNRPN. Contudo, a expressão do gene Necdin variou entre as estruturas estudadas. Em conclusão, apesar dos gêmeos eqüinos provenientes de TNCS diferirem quanto ao tamanho, morfologicamente são iguais. Dentre as estruturas cerebrais o plexo coróide se apresentou mais desenvolvido nos fetos de maior comprimento. Os fetos muares clonados não apresentaram diferença no padrão de metilação do gene SNRPN. No entanto, as iPSCs apresentaram alteração no padrão de metilação deste gene na décima passagem. Embora os genes SNRPN, Necdin e UBE3A sejam expressos no cérebro, o SNRPN apresentou-se prevalente nessa estrutura / The nuclear transfer of somatic cells (SCNT) is being used to produce elite horses. With the discovery of the possibility to reprogram somatic cells to a pluripotent state (iPSCs), these cells have become widely used in neuroscience research. However, also occur epigenetic changes during this cellular reprogramming. Therefore, our hypothesis is that equine twins caused by equine ART could lead to developmental irregularities of the nervous system. The patterns of SNRPN methylation in the structures of cloned mule fetuses and in iPSCs are different from the patterns found in the analyzed mules. And the expression of SNRPN, Necdin and UBE3A genes are higher in the brain, while the higher expression of H19 gene occurs in the extraembryonic membranes. In our study we derived two equine twin SCNT pregnancies that were interrupted at 40 and 60 days of gestation and compared to singleton fetuses of similar age. Differences in lengths between twin embryos were observed at both 40 (2.0 and 2.2 cm 10%) and 60 (6.5 and 8.5 cm 24%) days of gestation. Only the choroid plexus in the fourth ventricle more developed in the twins with the greatest length. Analyzing mules cloned fetuses at different gestational ages, and compare them with mules at embryonic, fetal and adult period; there was no difference in the pattern of methylation in SNRPN gene. However, in the tenth passage of the iPSCs the methylation pattern was altered in relation to the studied mules and the pattern observed in fibroblasts. When the cloned fetuses at different gestational ages were analyzed, the brain presented lower expression of H19 and UBE3A genes, and higher expression of SNRPN gene. However, the expression of Necdin gene varied among the structures studied. In conclusion, despite the twin horses from SCNT differ in size, they are morphologically identical. Among the brain structures the choroid plexus performed more developed in the fetuses of greater length. Cloned mules fetuses showed no difference in the pattern of methylation SNRPN gene. However, iPSCs have changes in the pattern of methylation of this gene in the tenth passage. Although SNRPN, Necdin and Ube3A genes are expressed in the brain, SNRPN is prevalent in this structure

Einfluss des Absetzverfahrens und anderer systematischer Effekte auf die Milchleistung und ausgewählte Eutergesundheitsparameter einer Herde Ostfriesischer Milchschafe

In der vorliegenden Arbeit wurde die Bedeutung des Einflusses von Laktationsnummer, Laktationsstadium, Körperkondition und Absetzverfahren auf Merkmale der Milchleistung und Eutergesundheit einer Herde Ostfriesischer Milchschafe untersucht. Die Tiere wurden nach dem Ablammen zufällig einer Früh- bzw. Spätabsetzergruppe zugeordnet (Absetzen der Lämmer 3 bzw. 42 Tage post partum). An insgesamt 40 Terminen wurden über eine vollständige Laktation Vorgemelks-, Hälftenanfangsgemelks- und Einzeltiergemelksproben gewonnen. Das Spektrum der untersuchten Merkmale umfasste die Milchmenge, die Milchinhaltsstoffe (Fett, Eiweiß, Laktose), die klinische Untersuchung des Euters, die somatische Zellzahl, die elektrische Leitfähigkeit und bakteriologische Untersuchungen. Zusätzlich wurden das Körpergewicht und die Körperkondition der Mutterschafe erfasst. Das durchschnittliche Leistungsniveau der Herde betrug 301±101,3 kg Milch, bei mittleren Milchfett-, Milcheiweiß-, und Laktosegehalten von 5,00, 5,14 bzw. 5,00 % (150-Tageleistung). Die Laktationsnummer hatte keinen signifikanten Effekt auf die Milchleistung. Schafe der Spätabsetzer-Gruppe produzierten im Anschluss an die Säugephase eine um 300 g signifikant höhere Testtagsmilchmenge als Tiere der Frühabsetzer-Gruppe. Das Absetzverfahren hatte keinen nachweisbaren Effekt auf die Milchinhaltsstoffe. Die Herde wies eine gute Eutergesundheit auf. Der Anteil bakteriologisch positiver Schafmilchproben betrug 28,5 %. Als dominante Erreger wurden in 96,6 % der bakteriologisch positiven Schafmilchproben Koagulasenegative Staphylokokken nachgewiesen. Der Anteil bakteriologisch positiver Euterhälftenbefunde stieg signifikant mit dem Fortschreiten der Laktation und zunehmender Laktationsnummer. Eine Euterinfektion mit Koagulasenegativen Staphylokokken sowie das Verfahren des Spätabsetzens beeinflussten alle drei untersuchten Eutergesundheitsparameter (logarithmierte Zellzahl, elektrische Leitfähigkeit und Laktosegehalt) negativ (p < 0,001).

Impact d’une infection intra-mammaire causée par Staphylococcus aureus ou un staphylocoque coagulase-négative présente en début de lactation chez les taures laitières

L’objectif de cette étude était de déterminer l’impact d’une infection intra-mammaire (IIM) subclinique causée par staphylocoque coagulase-négative (SCN) ou Staphylococcus aureus diagnostiquée durant le premier mois de lactation chez les taures sur le comptage de cellules somatiques (CCS), la production laitière et le risque de réforme durant la lactation en cours. Des données bactériologiques provenant d’échantillons de lait composites de 2 273 taures Holstein parmi 50 troupeaux ont été interprétées selon les recommandations du National Mastitis Council. Parmi 1 691 taures rencontrant les critères de sélection, 90 (5%) étaient positives à S. aureus, 168 (10%) étaient positives à SCN et 153 (9%) étaient négatives (aucun agent pathogène isolé). Le CCS transformé en logarithme népérien (lnCCS) a été modélisé via une régression linéaire avec le troupeau comme effet aléatoire. Le lnCCS chez les groupes S. aureus et SCN était significativement plus élevé que dans le groupe témoin de 40 à 300 jours en lait (JEL) (P < 0.0001 pour tous les contrastes). La valeur journalière du lnSCC chez les groupes S. aureus et SCN était en moyenne 1.2 et 0.6 plus élevé que le groupe témoin respectivement. Un modèle similaire a été réalisé pour la production laitière avec l’âge au vêlage, le trait génétique lié aux parents pour la production laitière et le logarithme népérien du JEL de la pesée inclus. La production laitière n’était pas statistiquement différente entre les 3 groupes de culture de 40 à 300 JEL (P ≥ 0.12). Les modèles de survie de Cox ont révélé que le risque de réforme n’était pas statistiquement différent entre le groupe S. aureus ou SCN et le groupe témoin (P ≥ 0.16). La prévention des IIM causées par SCN et S. aureus en début de lactation demeure importante étant donné leur association avec le CCS durant la lactation en cours. / The objective of this study was to determine the effect of a subclinical intramammary infection (IMI) caused by coagulase-negative staphylococci (CNS) or Staphylococcus aureus diagnosed during the first month of lactation in heifers on somatic cell count (SCC), milk production and culling risk during the entire first lactation. Bacteriological analysis data of composite milk samples taken from 2,273 Hostein heifers among 50 dairy herds were interpreted according to the National Mastitis Council guidelines. Among the 1,691 heifers meeting the selection criteria, 90 (5%) were diagnosed with S. aureus, 168 (10%) with CNS, and 153 (9%) were negative (no pathogen isolated). Test-day SCC transformed in natural logarithm (lnCCS) was fit in a linear regression model with herd as random effect. The lnSCC in S. aureus and CNS groups were significantly higher than in negative group from 40 to 300 days in milk (DIM) (P < 0.0001 for all contrasts). At test-day level, lnSCC in S. aureus and CNS groups was on average 1.2 and 0.6 higher than the negative group respectively. A similar model was used for milk yield with age at calving, parent average genetic value for milk yield and natural logarithm of tested DIM included. Milk yield was not statistically different between culture groups from 40 to 300 DIM (P ≥ 0.12). Compared with negative heifers, the culling hazard ratio estimated using Cox survival analysis in S. aureus and CNS infected heifers was not significant (P ≥ 0.16). Prevention of CNS or S. aureus IMI in early lactation remains important for its association with SCC during the ensuing lactation.
