Élaboration et étude optoélectronique des nanotubes d'oxydes métalliques décorés par des nanoparticules de PbS et/ou CdS pour applications environnementales et photovoltaïques

[ES] En la última década, las crecientes necesidades de protección del medio ambiente han estimulado la investigación en el campo de la fotocatálisis. La contaminación del agua por productos químicos orgánicos, tintes textiles y productos farmacéuticos constituye un peligro cada vez más dañino para el medio ambiente, la salud humana y la vida acuática. Para solucionar este problema y luchar contra la contaminación del agua, varios grupos de investigación han demostrado la utilidad del uso de dióxido de titanio TiO2 como un prometedor fotocatalizador no tóxico y de bajo coste para la fotodegradación de contaminantes. Obviamente, como cualquier otro material, el TiO2 tiene limitaciones en varias áreas de aplicación debido a su ancho de banda prohibida, que limita su rango de absorción a los rayos UV. Esto nos incita a asociarlo a semiconductores de banda prohibida baja como PbS. En este trabajo se asociaron nanopartículas de PbS a nanotubos de TiO2 (NTs) para mejorar su actividad fotocatalítica en el visible. Comenzamos nuestro trabajo optimizando la deposición de micro/nanoestructuras de PbS mediante el método hidrotermal mientras estudiamos la influencia de los parámetros de deposición en las propiedades morfológicas y estructurales de las micro/nanoestructuras de PbS de diferentes formas. Inicialmente, la atención se centró en optimizar los parámetros para sintetizar nanopartículas de PbS a partir de microestructuras mediante un simple cambio del precursor aniónico (fuente de S2-). Posteriormente, se realizó un estudio sobre el efecto de la concentración del precursor en la calidad de las nanopartículas de PbS. A continuación, se estudió la influencia de la decoración de las NP de PbS en las propiedades físicas, ópticas y estructurales de las NT de TiO2. Demostramos la efectividad de la técnica SILAR para la realización de heteroestructuras {NPs PbS}n - NTs TiO2, que son adecuadas para la integración en dispositivos fotocatalíticos. Hemos demostrado que la calidad de la estructura cristalina, el tamaño de los cristales y las propiedades ópticas de los sistemas fabricados dependen del número de ciclos SILAR "n" y de la distribución y tamaño de las NP de PbS depositadas por SILAR. Finalmente, se presentó el estudio cinético de la actividad fotocatalítica de NTs de TiO2 puras y decoradas con NPs de PbS para la fotodegradación de antibióticos (tetraciclina) bajo irradiación UV. Primero, encontramos que el sistema PbS NPs/TiO2 NTs es efectivo para la fotodegradación de tetraciclina (TC), con una degradación óptima para un número de ciclo SILAR de deposición de PbS igual a 5. Segundo, estudiamos la actividad fotocatalítica de TiO2 NTs puros en una solución acuosa de TC bajo luz ultravioleta. Después de optimizar las condiciones experimentales, logramos una fotodegradación de aproximadamente el 99% de la tetraciclina después de 5 horas de irradiación. Además, hemos demostrado que el fotocatalizador es estable y podría usarse con éxito durante al menos cinco ciclos sucesivos sin una pérdida significativa de rendimiento. Estos resultados sugieren que el fotocatalizador PbS NPs/TiO2 NTs es un sistema efectivo y prometedor para la purificación de agua. Además, con base en los resultados de las pruebas de atrapamiento, propusimos un mecanismo de fotodegradación que muestra que la capacidad de las NT de TiO2 para oxidar el antibiótico se debe principalmente a los agujeros fotogenerados, así como a los radicales hidroxilo (OH), que se consideran como agentes oxidantes primarios que actúan no solo en la superficie sino también en solución. / [CA] A la darrera dècada, les creixents necessitats de protecció del medi ambient han estimulat la recerca en el camp de la fotocatàlisi. Comencem el nostre treball optimitzant la deposició de micro/nanoestructures de PbS mitjançant el mètode hidrotermal mentre estudiem la influència dels paràmetres de deposició en les propietats morfològiques i estructurals de les micro/nanoestructures de PbS de diferents formes. Segon, estudiem l'activitat fotocatalítica de TiO2 NTs purs en una solució aquosa de TC sota llum ultraviolada. Després d'optimitzar les condicions experimentals, aconseguim una fotodegradació d'aproximadament el 99% de la tetraciclina després de 5 hores d'irradiació. A més, hem demostrat que el fotocatalitzador és estable i es podria fer servir amb èxit durant almenys cinc cicles successius sense una pèrdua significativa de rendiment. Aquests resultats suggereixen que el fotocatalitzador PbS NPs/TiO2 NT és un sistema efectiu i prometedor per a la purificació d'aigua. A més, amb base en els resultats de les proves de captura, vam proposar un mecanisme de fotodegradació que mostra que la capacitat de les NT de TiO2 per oxidar l'antibiòtic es deu principalment als forats fotogenerats, així com als radicals hidroxil (OH), que es consideren .com a agents oxidants primaris que actuen no només a la superfície sinó també en solució. / [EN] The increasing needs for environmental protection have strongly stimulated research in the field of photocatalysis in recent years. Contamination of water by organic chemicals, textile dyes and pharmaceuticals constitute an increasingly harmful hazard to the environment, human health, and aquatic life. To solve this problem and fight water pollution, several research groups have demonstrated the usefulness of using titanium dioxide TiO2 as a promising low-cost non-toxic photocatalyst for the photodegradation of contaminants. Obviously, like any other material, TiO2 has limitations in several areas of application because of its wide band gap, which limits its absorption range to UV. This incites us to associate it to low band gap semicondors such as PbS. In this work, PbS nanoparticles were associated to TiO2 nanotubes (NTs) to improve their photocatalytic activity in the visible. We started our work by optimizing the deposition of PbS micro-nanostructures by the hydrothermal method while studying the influence of deposition parameters on the morphological and structural properties of the different shaped PbS micro-nanostructures. Initially, the focus was on optimizing parameters to synthesize PbS nanoparticles from microstructures by a simple change of the anionic precursor (source of S2-). Subsequently, a study was conducted on the effect of the precursor concentration on the quality of PbS nanoparticles. Next, we also studied the influence of PbS NPs decoration on the physical, optical and structural properties of TiO2 NTs. We demonstrated the effectiveness of the SILAR technique for the realization of {NPs PbS}n - NTs TiO2 heterostructures, which are suitable for integration into photocatalytic devices. We have shown that the quality, crystal structure, crystallite size and optical properties of the achieved systems depend on the SILAR cycle number ''n'' and the size distribution of the SILAR-deposited PbS NPs. Finally, the kinetic study of the photocatalytic activity of pure TiO2 NTs and decorated with PbS NPs for the photodegradation of antibiotics (tetracycline) under UV irradiation was presented. First, we found that the PbS NPs/TiO2 NTs system is effective for the photodegradation of tetracycline (TC), with an optimum degradation for a SILAR cycle number of PbS deposition equal to 5. Second, we studied the photocatalytic activity of pure TiO2 NTs in an aqueous TC solution under UV light. After optimizing the experimental conditions, we achieved a photodegradation of approximately 99% of tetracycline after 5 hours of irradiation. In addition, we have shown that the photocatalyst is stable and could be used successfully for at least five successive cycles without any significant loss of performance. These results suggest that the PbS NPs/TiO2 NTs photocatalyst is an effective and promising system for water purification. In addition, based on the results of the trapping tests, we proposed a photodegradation mechanism that shows that the ability of the TiO2 NTs to oxidize the antibiotic is mainly due to the photogenerated holes as well as the hydroxyl (OH) radicals, which are considered as primary oxidizing agents acting not only on the surface but also in solution. / Jemai, S. Applicability of a computational design approach for synthetic riboswitches

Riboswitches have gained attention as tools for synthetic biology, since they enable researchers to reprogram cells to sense and respond to exogenous molecules. In vitro evolutionary approaches produced numerous RNA aptamers that bind such small ligands, but their conversion into functional riboswitches remains difficult. We previously developed a computational approach for the design of synthetic theophylline riboswitches based on secondary structure prediction. These riboswitches have been constructed to regulate ligand dependent transcription termination in Escherichia coli. Here, we test the usability of this design strategy by applying the approach to tetracycline and streptomycin aptamers. The resulting tetracycline riboswitches exhibit robust regulatory properties in vivo. Tandem fusions of these riboswitches with theophylline riboswitches represent logic gates responding to two different input signals. In contrast, the conversion of the streptomycin aptamer into functional riboswitches appears to be difficult. Investigations of the underlying aptamer secondary structure revealed differences between in silico prediction and structure probing. We conclude that only aptamers adopting the minimal free energy (MFE) structure are suitable targets for construction of synthetic riboswitches with design approaches based on equilibrium thermodynamics of RNA structures. Further improvements in the design strategy are required to implement aptamer structures not corresponding to the calculated MFE state.

O presente trabalho visa desenvolver métodos que permitam determinar as concentrações de tetraciclina, por Cromatografia Líquida de Alta Eficiência, no plasma e líquido sinovial, além de analisar as concentrações correspondentes em leite de gado leiteiro em lactação submetidos aos tratamentos intravenoso e intravenoso regional. Desta forma, objetivando determinar a depuração da tetracic1ina no organismo dos animais tratados, a concentração do fármaco no sítio de ação e a quantidade residual em leite, as amostras biológicas foram colhidas e quantificadas em diferentes tempos pré e pós-administração do fármaco. Os métodos analíticos validados apresentaram linearidade, limite de detecção, quantificação, exatidão, precisão e recuperação adequados à quantificação do antibiótico nas matrizes biológicas estudadas. As amostras de leite de animais tratados com o medicamento por via intravenosa regional, não apresentaram resíduos após 120h da administração do fármaco. O mesmo ocorreu plasma e líquido sinovial após 48 h. Através da administração via intravenosa do medicamento foram observados resíduos no leite em todos os tempos avaliados, ao passo que no plasma e líquido sinovial, a presença do princípio ativo não foi detectada após 72 horas pós-tratamento. / The purpose of the present work is to develop methods which allow the determination of tetracycline by High Pressure Liquid Chromatography in serum, synovial fluid, as well as analyze the corresponding milk concentrations in milk cows subjected to intravenous and regional intravenous treatment. Therefore, aiming to determine the clearance of tetracycline in the body of the treated animals, the concentration of the active principle in the action site and the residual quantity in milk, biological matrices were collected at different times. The validated analytical methods depicted suitable linearity, detection and quantification limits, accuracy, precision and recovery, allowing the quantification of the antibiotic in the studied biological matrices. In relation to the milk samples from animals treated with the drug by regional intravenous via, they did not present residues of tetracycline after 120 h post-administration. The values were also null for both serum and synovial fluid after 48 h. Through regional intravenous drug administration, milk residues were observed in all the evaluated times whereas for serum and synovial fluid, the presence of the active principle was not detected after 72h post-treatment.

Chitosan comes as a great choice in the field of adsorption, due to their adsorbents properties, low cost and abundance. The presence of amino groups in its chain govern the majority of their properties and define which application a sample of chitosan may be used, so it is essential to determine their average degree of deacetylation. In this work we developed kinetic and equilibrium studies to monitor and characterize the adsorption process of two drugs, tetracycline hydrochloride and sodium cromoglycate, in chitosan particles. Kinetic models and the adsorption isotherms were applied to the experimental data. For both studies, the zeta potential analyzes were also performed. The adsorption of each drug showed distinct aspects. Through the studies developed in this work was possible to describe a kinetic model for the adsorption of tetracycline on chitosan particles, thus demonstrating that it can be described by two kinetics of adsorption, one for protonated tetracycline and another one for unprotonated tetracycline. In the adsorption of sodium cromoglycate on chitosan particles, equilibrium studies were developed at different temperatures, allowing the determination of thermodynamic parameters / Dentre os pol?meros que mais se destacam nas ?ltimas d?cadas, a quitosana, um biopol?mero com propriedades f?sico-qu?micas e biol?gicas promissoras, tem sido alvo de um campo amplo de pesquisa. Many of these properties are associated with the presence of amino groups in its polymer chain. A proper determination of these amino groups is very important, in order to properly specify if a given chitosan sample can be used in a particular application. Thus, in this work, initially, a comparison between the determination of the deacetylation degree by conductometry and elemental analysis was carried out using a detailed analysis of error propagation. It was shown that the conductometric analysis resulted in a simple and safe method for the determining the degree of deacetylation of chitosan. Subsequently, experiments were performed to monitor and characterize the adsorption of tetracycline on chitosan particles through kinetic and equilibrium studies. The main models of kinetics and adsorption isotherms, widely used to describe the adsorption on wastewater treatment systems and the drug loading, were used to treat the experimental data. Firstly, it was shown that an apparent linear t/q(t) ? t relationship did not imply in a pseudo-second-order adsorption kinetics, differently of what has been repeatedly reported in the literature. It was found that this misinterpretation can be avoided by using non-linear regression. Finally, the adsorption of tetracycline on chitosan particles was analyzed using insights obtained from theoretical analysis, and the parameters generated were used to analyze the kinetics of adsorption, the isotherm of adsorption and to ropose a mechanism of adsorption / Devido ?s suas propriedades f?sico-qu?micas e biol?gicas, associadas ? abund?ncia e ao baixo custo da mat?ria-prima, a quitosana tem sido considerada um material de ampla aplica??o em diversos campos, tais como em sistemas de libera??o de f?rmacos. Molecular Mechanisms in Endothelial Cell Differentiation

<p>Angiogenesis is the formation of new blood vessels from the pre-existing blood vessels. Blood vessels are composed of endothelial cells and supporting musculature. Angiogenesis is regulated by numerous soluble ligands and by cell-matrix interactions. We have studied the molecular mechanisms in fibroblast growth factor-2 (FGF-2) and vascular endothelial growth factor-A (VEGF-A)-induced angiogenesis using immortalized endothelial cell lines in different angiogenesis assays.</p><p>The role of the signaling protein H-Ras in FGF-2-induced <i>in vitro</i> angiogenesis was studied by expressing mutated versions of H-Ras in immortalized mouse brain endothelial cells using a tetracycline-regulated expression system. <i>In vitro</i> angiogenesis was analyzed as the ability of cells to invade a fibrin matrix and form branching structures in response to a combination of FGF-2 and tumor necrosis factor-α (TNF-α). Inhibition of H-Ras through the expression of dominant negative (S17N) H-Ras or pharmacological inactivation of H-Ras with a farnesyl transferase inhibitor, did not inhibit growth factor-induced invasion. In contrast, expression of constitutively active (G12V) H-Ras caused cells to adopt a transformed phenotype which inhibited invasive growth and cells formed solid tumors when injected in nude mice. These studies suggest that H-Ras activity is not required for differentiation but its activity must be tightly regulated as aberrant activity impairs endothelial cell differentiation.</p><p>In order to screen for both known and novel genes that regulate angiogenesis we used large scale microarray analysis. In VEGF-A-stimulated telomerase immortalized human microvascular endothelial cells undergoing invasive growth in fibrin gels, or forming cord-like structures on collagen, we identified several genes that were differentially expressed. Some of these are known to be important for endothelial cell functions and angiogenesis while others have no previous connection with endothelial cell function or were transcripts with no assigned function. Further analysis of these proteins will aid in elucidating the molecular mechanisms underlying endothelial cell differentiation. </p>

In contrast, expression of constitutively active (G12V) H-Ras caused cells to adopt a transformed phenotype which inhibited invasive growth and cells formed solid tumors when injected in nude mice. These studies suggest that H-Ras activity is not required for differentiation but its activity must be tightly regulated as aberrant activity impairs endothelial cell differentiation. In order to screen for both known and novel genes that regulate angiogenesis we used large scale microarray analysis. In VEGF-A-stimulated telomerase immortalized human microvascular endothelial cells undergoing invasive growth in fibrin gels, or forming cord-like structures on collagen, we identified several genes that were differentially expressed. Some of these are known to be important for endothelial cell functions and angiogenesis while others have no previous connection with endothelial cell function or were transcripts with no assigned function. Further analysis of these proteins will aid in elucidating the molecular mechanisms underlying endothelial cell differentiation.

Étude de systèmes multicapteurs utilisant des capteurs chimiques, électrochimiques et des biocapteurs pour des applications en agroalimentaire / Development of sensors and biosensors for food analysis

Les capteurs et les biocapteurs sont des moyens d’analyse en plein essor à la fois rapides, sensibles, sélectifs et peu coûteux, applicables à des domaines très variés (agroalimentaire, environnement, biomédical…). Dans ce travail de recherche, nous nous sommes intéressés au développement de trois dispositifs à savoir un nez et une langue électroniques à base de systèmes multicapteurs pour l’analyse des odeurs et des saveurs ainsi que les biocapteurs. Les deux premiers dispositifs ont permis dans un premier temps de caractériser et de détecter les fraudes dans les produits de l’industrie agroalimentaire. Ainsi, nous sommes parvenus à détecter les pratiques frauduleuses dans l’huile d’argan par le nez et par la langue électroniques. Nous avons, par ailleurs, réussi à caractériser des miels de différentes origines géographiques et botaniques et à détecter l’adultération du miel pur par l’utilisation d’une langue électronique voltammétrique. Enfin, nous avons démontré l’efficacité des systèmes de nez et de langue électroniques à discriminer cinq marques de lait pasteurisé marocain. Des limitations du système de nez et de langue électroniques ont été révélées quant à la discrimination du lait pasteurisé (Jawda) en fonction des jours de stockage. Par contre, la fusion des données des deux systèmes moyennant un niveau d'abstraction intermédiaire a permis d’améliorer cette discrimination. Dans une deuxième étape, nous avons développé deux biocapteurs, le premier est basé sur l’utilisation de Polymère à Empreinte Moléculaire (MIP) dédié à la détection de la tétracycline dans le miel. Alors que le second est un immunocapteur conçu pour la détection de l’ochratoxine A. Le MIP, utilisé dans le premier cas, a été synthétisé à la surface d’électrodes en or par électropolymérisation des nanoparticules d'or fonctionnalisées par le p-aminothiophénol en présence de la tétracycline comme molécule empreinte. Dans le deuxième cas, un nouveau biocapteur capacitif basé sur l’utilisation d’un substrat de Nitrure de Silicium (Si3N4) combiné avec une nouvelle structure de nanoparticules magnétiques (MNPs) pour la détection de l’ochratoxine A a été conçu. En effet, Les MNPs possédant une terminaison carboxylique ont été liés de manière covalente à la monocouche auto-assemblée du silane-amine (3-aminopropyl triéthoxysilane APTES). Enfin, les anticorps anti-ochratoxine A ont été immobilisés sur les MNPs par liaison amide. Les performances des deux biocapteurs (limite de détection, sélectivité, reproductibilité) ont ensuite été évaluées / Sensors and Biosensors are rapid, sensitive, selective and low-cost analytical devices of growing interest for a wide range of application fields (e.g. food, environment, health …). This research focused on the development of three devices namely an electronic nose and tongue and electrochemical biosensors with applications in food analysis. The first two devices allowed the characterization and detection of frauds in the food field. Thus, we have been able to detect fraudulent practices in argan oil by using an electronic nose and tongue systems. In addition, the electronic tongue has successfully classified honeys of different geographical and botanical origins and detects the adulteration of pure honey. Finally, we have demonstrated the ability of the electronic nose and tongue systems to classify five brands of Moroccan pasteurized milk and to discriminate against them based on their storage days. In the second stage, we have developed two biosensors. The first one is based on a molecularly imprinted polymer (MIP) for the detection of tetracycline in honey. The second one is based on an immunosensor devoted for the detection of ochratoxin A. For the first biosensor, imprinted gold nanoparticles composites are assembled on Au surfaces by the electropolymerization of p-amino-thiophenol functionalized gold nanoparticles in the presence of the imprint molecule. In the second case, we have developed a novel capacitance electrochemical biosensor based on silicon nitride substrate (Si3N4) combined with a new structure of mangnetic nanoparticles (MNPs). Indeed, The MNPs with terminated carboxylic acid were covalently bonded to Si3N4 through a Self-Assembled Monolayers (SAMs) of the silane-amine (3-Aminopropyl) triethoxysilane (APTES). Finally anti-ochratoxin A antibodies were immobilized on MNPs by amide bonding. The performances of the two biosensors (limit of detection, selectivity, reproducibility) were then evaluated and the results are generally satisfactory

O presente trabalho visa desenvolver métodos que permitam determinar as concentrações de tetraciclina, por Cromatografia Líquida de Alta Eficiência, no plasma e líquido sinovial, além de analisar as concentrações correspondentes em leite de gado leiteiro em lactação submetidos aos tratamentos intravenoso e intravenoso regional. Desta forma, objetivando determinar a depuração da tetracic1ina no organismo dos animais tratados, a concentração do fármaco no sítio de ação e a quantidade residual em leite, as amostras biológicas foram colhidas e quantificadas em diferentes tempos pré e pós-administração do fármaco. Os métodos analíticos validados apresentaram linearidade, limite de detecção, quantificação, exatidão, precisão e recuperação adequados à quantificação do antibiótico nas matrizes biológicas estudadas. As amostras de leite de animais tratados com o medicamento por via intravenosa regional, não apresentaram resíduos após 120h da administração do fármaco. O mesmo ocorreu plasma e líquido sinovial após 48 h. Através da administração via intravenosa do medicamento foram observados resíduos no leite em todos os tempos avaliados, ao passo que no plasma e líquido sinovial, a presença do princípio ativo não foi detectada após 72 horas pós-tratamento. / The purpose of the present work is to develop methods which allow the determination of tetracycline by High Pressure Liquid Chromatography in serum, synovial fluid, as well as analyze the corresponding milk concentrations in milk cows subjected to intravenous and regional intravenous treatment. Therefore, aiming to determine the clearance of tetracycline in the body of the treated animals, the concentration of the active principle in the action site and the residual quantity in milk, biological matrices were collected at different times. The validated analytical methods depicted suitable linearity, detection and quantification limits, accuracy, precision and recovery, allowing the quantification of the antibiotic in the studied biological matrices. In relation to the milk samples from animals treated with the drug by regional intravenous via, they did not present residues of tetracycline after 120 h post-administration. The values were also null for both serum and synovial fluid after 48 h. Through regional intravenous drug administration, milk residues were observed in all the evaluated times whereas for serum and synovial fluid, the presence of the active principle was not detected after 72h post-treatment.
