A lesão por balão cateterismo é um procedimento comumente utilizado para o estudo dos mecanismos de restenose. O estresse mecânico produzido pela passagem do balão promove alterações tanto na artéria lesada quanto na artéria contra-lateral. Observou-se um aumento da densidade de neurônios que contêm neuropeptídeos como a Substância P (SP) e o Peptídeo Relacionado com o Gene da Calcitonina (CGRP) na artéria contra-lateral à lesão, o que evidencia a ocorrência de um processo neurocompensatório. Observou-se ainda um aumento da reatividade desta artéria à fenilefrina (Phe) e à angiotensina II (Ang II) após 4 e 15 dias da lesão, respectivamente. O objetivo do presente trabalho foi estudar as conseqüências dos tratamentos com celecoxibe (inibidor seletivo da enzima COX-2), -tocoferol (Vitamina E, antioxidante natural) e losartan (antagonista dos receptores AT1), sobre a reatividade à Phe, Cloreto de Potássio (KCl), Acetilcolina (ACh), Ang II de artérias ipsilaterais e contra-laterais em relação a artérias controles. Em animais tratados com celecoxibe (10 mg/Kg - 2 vezes ao dia) ou -tocoferol (400 mg/Kg/dia) por 7 dias. Os resultados mostram que os dois tratamentos normalizam os valores de efeito máximo (Emax) da Phe nas artérias contra-laterais com endotélio aos níveis de artérias controle. Em animais tratados com celecoxibe, a artéria ipsilateral não respondeu à Phe, enquanto no tratamento com -tocoferol mostrou valores de Emax reduzidos em relação a animais controle. Os valores de Emax da ACh em artérias de animais controle e tratados com celecoxibe ou -tocoferol, são idênticos. Entretanto, ambos os tratamentos promoveram redução na potência da ACh em artérias controle quando comparadas com as de animais não tratados. A potência da ACh na artéria contra-lateral foi semelhante ao controle em todos os tratamentos. O Emax da Ang II estava aumentado na artéria contra-lateral à lesão. O tratamento com losartan (15 mg/Kg/dia) por 18 dias promoveu redução neste parâmetro na artéria contra-lateral aos níveis do controle. A potência da Ang II em artérias contra-laterais de animais tratados com losartan é igual ao da artéria de animais controle. Nos animais controles tratados com losartan a potência desse peptídeo foi menor do que controle e contra-lateral sem tratamento e contra-lateral tratada. Na artéria contra-lateral o Emax do KCL estava diminuído em relação ao controle e o tratamento com losartan não modificou este parâmetro. O Emax da ACh em artérias controle e contra-laterais não foi alterado pelo tratamento com losartan, entretanto houve aumento da potência deste agonista em artérias controles e contra-laterais em relação a artérias de animais controles. Os resultados obtidos no presente estudo indicam que a produção de espécies reativas de oxigênio, prostanóides vasocontritores e Ang II levam a alterações na reatividade aos agonistas estudados na artéria contra-lateral à lesão por cateter balão. / The injury by balloon catheter is a procedure commonly used to study the mechanisms of restenosis. The mechanical stress produced by the passage of the balloon promotes changes both in the injured artery in the contralateral artery. There was an increased density of neurons containing neuropeptides such as substance P (SP) and the Gene Related Peptide of Calcitonin (CGRP) in the contralateral artery to the lesion, which shows the occurrence of a process neurocompensatory. There was also an increase in the artery reactivity to phenylephrine (Phe) and angiotensin II (Ang II) after 4 and 15 days of injury, respectively. There was also an increase in the artery reactivity to Phe and Ang II after 4 and 15 days of injury, respectively. The objective of this work was to study the consequences of treatment with celecoxib (selective inhibitor of COX-2), -tocopherol (vitamin E, natural antioxidant) and losartan (AT1 receptor antagonist) on the reactivity to Phe, chloride Potassium (KCl), acetylcholine (ACh), Ang II in arteries ipsilateral and contralateral side for control arteries. In animals treated with celecoxib (10 mg / kg - 2 times daily) or -tocopherol (400 mg / kg / day) for 7 days. The results show that both treatments normalize the values of maximum effect (Emax) of Phe in the contralateral arteries with endothelium to the levels of control arteries. In animals treated with celecoxib, the ipsilateral artery did not respond to Phe, whereas the treatment with -tocopherol showed reduced values of Emax for the control animals. The values of Emax of ACh in arteries from control animals and treated with celecoxib or -tocopherol, are identical. However, both treatments promoted reduction in the potency of ACh in control arteries when compared with untreated animals. The potency of ACh in the contralateral artery was similar to control in all treatments. The Emax of Ang II was increased in the contra-lateral artery to the lesion. Treatment with losartan (15 mg / kg / day) for 18 days promoted reduction in this parameter in the contra-lateral artery levels of control. The potency of Ang II in contralateral arteries of animals treated with losartan is equal to the artery of control animals. In control animals treated with losartan the potency of this peptide was lower than control and contra-lateral untreated and control-treated side. Contralateral artery in the Emax of KCL was decreased in the control and treatment with losartan did not modify this parameter. The Emax of ACh in control arteries and contralateral side was not changed by treatment with losartan, however increased the power of this agonist in control arteries and contralateral side on arteries of control animals. The results of this study indicate that the production of reactive oxygen species and vasoconstrictors prostanoids Ang II lead to changes in reactivity to agonists studied in the contra-lateral to the artery by balloon catheter injury.

Estado nutricional relativo ao selênio de pacientes na fase de pós-tratamento da leucemia linfóide aguda e sua relação com o estresse oxidativo / Nutritional status of selenium in patients in post-treatment of acute lymphoblastic leukemia and its relationship with oxidative stress

Este estudo teve como objetivo avaliar o estado nutricional relativo ao selênio (Se) de pacientes na fase de pós-tratamento da leucemia linfóide aguda (LLA) e sua relação com o estresse oxidativo. Foram selecionados 24 pacientes no pós-tratamento da LLA (9,2 ± 1,9 anos) atendidos no Instituto de Oncologia Pediátrica da Universidade Federal de São Paulo e 60 indivíduos saudáveis (9,5 ± 1,3 anos) da Escola de Aplicação da Universidade de São Paulo. Foram coletados 10 mL de sangue venoso para análise de Se plasmático e eritrocitário, glutationa peroxidase (GPx), superóxido dismutase (SOD), α- tocoferol, malondialdeído (MDA) e 8-oxo-desoxiguanosina (8-oxo-dGuo). A urina de 24 horas foi coletada para análise da excreção de Se, e três recordatórios de consumo alimentar de 24 horas para avaliação do Se ingerido. Os resultados obtidos quanto aos parâmetros bioquímicos de avaliação de Se não apresentaram diferença significativa entre os grupos de pacientes e controles, e foram respectivamente: Se plasmático, 44,4 ± 9,0 µg/L e 48,7 ± 12,0 µg/L (p = 0,122); Se eritrocitário, 49,9 ± 15,9 µg/L e 45,0 ± 15,9 µg/L (p = 0,202); Se urinário, 19,6 ± 14,8 µg Se/g de creatinina e 18,6 ± 9,6 µg Se/g de creatinina (p = 0,820). O consumo médio de Se foi de 27,4 ± 8,7 µg/dia e 28,0 ± 1,5 µg/dia (p = 0,756), respectivamente. Os grupos estudados foram considerados deficientes em Se, considerando os pontos de corte adotados. A atividade da GPx foi significativamente menor nos pacientes do que nos controles (33,3 ± 11,1 U/g Hb e 76,9 ± 25,9 U/g Hb) (p = 0,000), e a atividade da SOD não diferiu entre pacientes e controles (1796,9 ± 257,8 U/g Hb e 1915,9 ± 473,9 U/g Hb) (p = 0,145), assim como as concentrações de MDA (1,7 ± 0,3 µmol/L e 1,8 ± 0,4 µmol/L) (p = 0,053). A concentração de α-tocoferol foi estatisticamente maior nos pacientes que nos controles (17,7 ± 4,7 µmol/L e 10,6 ± 3,2 µmol/L) (p =0,000), bem como a concentração de 8-oxo-dGuo (43,6 ± 28,0 8-oxo/106 dG e 21,3 ± 22,9 8-oxo/106 dG) (p = 0,014). Os resultados apresentados apontam que os participantes deste estudo estão deficientes em Se e, em especial os pacientes no pós-tratamento da LLA estão sujeitos a um aumento do estado de estresse oxidativo, pois apesar das concentrações de MDA serem semelhantes entre os pacientes e os controles, a atividade da GPx dos pacientes foi reduzida e a concentração de 8-oxo-dGuo e α-tocoferol estavam aumentadas em relação aos controles. / This study aimed to evaluate the nutritional status of selenium (Se) in patients in post-treatment of acute lymphoblastic leukemia (ALL) and its relationship with oxidative stress. We selected 24 patients in post-treatment of ALL (9.2 ± 1.9 years) at the Pediatric Oncology Institute of Federal University of São Paulo and 60 healthy individuals (9.5 ± 1.3 years) of the School of Application at the University of São Paulo. We collected 10 mL of venous blood for analysis of Se in plasma and erythrocytes, glutathione peroxidase (GPx), superoxide dismutase (SOD), α-tocopherol, malondialdehyde (MDA) and 8-oxo-deoxyguanosine (8-oxo-dGuo). The 24-hour urine was collected for analysis of Se excretion and Se intake was evaluated by using three non-consecutive days of 24- hour recall. The results regarding biochemical evaluation of Se did not differ significantly between patients in post-treatment of ALL and controls, and the results were respectively: Se in plasma, 44.4 ± 9.0 µg/L and 48.7 ± 12.0 µg/L (p = 0.122); Se in erythrocytes, 49.9 ± 15.9 µg/L and 45.0 ± 15.9 µg/L (p = 0.202); Se in urine, 19.6 ± 14.8 µg Se/g creatinine and 18.6 ± 9.6 µg Se/g creatinine (p = 0.820). The average intake of selenium was 27.4 ± 8.7 mg/day and 28.0 ± 1.5 mg/day (p = 0.756), respectively. Both groups were considered deficient in selenium, according to the cut-off points adopted. The GPx activity was significantly lower in patients than in controls (33.3 ± 11.1 U/g Hb and 76.9 ± 25.9 U/g Hb) (p = 0.000), and no difference in SOD activity was observed between groups (1796.9 ± 257.8 U/g Hb and 1915.9 ± 473.9 U/g Hb) (p = 0.145). MDA concentrations were not different between patients and controls (1.7 ± 0.3 µmol/L and 1.8 ± 0.4 µmol/L) (p = 0.053) and α-tocopherol concentration was statistically higher in patients (17.7 ± 4.7 µmol/L and 10.6 ± 3.2 µmol/L) (p = 0.000), as well the concentration of 8-oxo-dGuo (43.6 ± 28.0 8-oxo/106 dG and 21.3 ± 22.9 8-oxo/106 dG) (p = 0.014). These results indicate that the participants in this study are deficient in Se mainly those who are in post-treatment of ALL are exposed to an increased state of oxidative stress, because although the concentrations of MDA were similar between patients and controls, the GPx activity of the patients was reduced and the concentration of 8-oxo-dGuo and α-tocopherol were increased compared to controls.

Alpha-tocopherol acquisition by plasma lipoproteins and changes in lipoprotein profile after cardiac surgery

Alpha-tocopherol, the most abundant form of vitamin E in man, is transported in the circulation by plasma lipoproteins. It plays important roles, not only in preventing lipid peroxidation, but also in modulating several cell functions such as cell signaling and gene expression. While chylomicrons transport dietary alpha-tocopherol after intestinal absorption, LDL and HDL are the major carriers of alpha-tocopherol in fasting plasma and largely contribute to its delivery to cells and tissues. Exchanges of alpha-tocopherol occur between plasma lipoproteins. In addition, alpha-tocopherol transfers have also been observed, in both directions, between plasma lipoproteins and artificial chylomicrons such as intravenous lipid emulsion particles used in parenteral nutrition. In acute conditions, intravenous supply of vitamin E via lipid emulsions, which bypasses the intestinal tract, may offer some advantages over oral administration to rapidly increase alpha-tocopherol plasma concentration. However, many questions remain unanswered regarding kinetics and factors facilitating vitamin E exchanges between lipid emulsions and plasma lipoproteins. The first part of this work aimed at characterizing alpha-tocopherol transfers between alpha-tocopherol rich emulsion particles and plasma lipoproteins as well as the potential for plasma proteins to modulate such transfers. An in vitro model of incubation was used in which emulsion triglyceride concentration was relatively low and lipoprotein levels comparable to those commonly found in the circulation. Results indicate a high capacity for LDL and HDL to acquire extra-amounts of alpha-tocopherol by rapid mass transfers from alpha-tocopherol-rich emulsion particles. Data further shows that, at a fixed alpha-tocopherol concentration provided by emulsion particles, the limiting factor for alpha-tocopherol enrichment is not the capacity of plasma lipoproteins to accommodate extra-amounts of alpha-tocopherol but the facilitating effect of plasma proteins on alpha-tocopherol transfer, the duration of the incubation and possibly the competition between different acceptor particles. Two lipid transfer proteins, PLTP and CETP, appear to largely mediate facilitation of alpha-tocopherol transfer; however, other plasma proteins may be involved. Data further shows that alpha-tocopherol enriched LDL and HDL can readily transfer newly acquired alpha-tocopherol to cells, without any regulation by plasma proteins.<p>Short-term prophylactic vitamin E supplementation has been suggested to be beneficial in some patients in acute conditions who present reduced plasma vitamin E concentrations in association with important changes in plasma lipids and severe oxidative stress. However, it was not clear whether low plasma vitamin E concentration in critically ill patients is related to changes in the composition of plasma lipoproteins or to a decrease in the number of alpha-tocopherol carriers. In the second part of this work, two clinical studies were conducted to analyze changes of lipoprotein concentration and composition in relation to inflammatory reaction and oxidative stress in selected subgroups of critically ill patients, namely patients undergoing cardiac surgery with different procedures. Important changes in LDL and HDL lipid content were observed, some of which contrast with previous observations made in critically ill septic patients. The reduced plasma level of alpha-tocopherol measured after cardiac surgery is entirely due to a reduced number of circulating LDL and HDL particles. Data suggests that such reduced number in alpha-tocopherol carriers post-surgery may impede the delivery of alpha-tocopherol to cells in conditions of increased requirements due to oxidative stress. Avoidance of extracorporeal circulation during cardiac surgery does not reduce inflammation-related changes in plasma lipids but largely prevents oxidative stress. This data on changes occurring in plasma lipoproteins may help to better define strategies against pro-inflammatory changes or oxidative stress. If further studies would confirm a clinical benefit with evidence-based rationale, alpha-tocopherol enriched lipid emulsions may be used to guarantee a sufficient alpha-tocopherol supply in acute conditions associated with fewer alpha-tocopherol transporters and increased requirements due to high risk of oxidative tissue injury.<p> / Doctorat en Sciences biomédicales et pharmaceutiques / info:eu-repo/semantics/nonPublished

The Pathophysiology of Smoking During Pregnancy: A Systems Biology Approach

This article focuses on a systems biology approach to studying the pathophysiology of cigarette smoking during pregnancy. Particular emphasis is given to the damaging role of oxidative stress. Cigarette smoking exerts multiple adverse affects but abundant evidence, mostly in adults, suggests that oxidative stress and free radical damage is a major pathophysiological factor. Smoking during pregnancy is known to contribute to numerous poor birth outcomes, such as low birth weight, preterm birth as well as life-long health and developmental problems. It is clinically important to know the separate contributions that cigarette derived-nicotine and smoking-induced free oxidative stress make to these poor outcomes. Surprisingly, the extent to which smoking dependent oxidative stress contributes to these poor outcomes is not well studied but the application of redox proteomics should be useful. Considerable biochemical evidence suggests that antioxidants, such as tocopherols and ascorbate, could be useful in minimizing oxidative stress induced pathology to the developing fetus in those women who, despite medical advice, continue to smoke. Nevertheless, this suggestion has yet to be tested in well-designed clinical studies.

Effects of Natural Antioxidants on Lipid Oxidation of Menhaden Oil

Preventing oxidative deterioration of fish oil is a significant challenge for the food industry. Natural antioxidants are widely incorporated into foods and oils to prevent oxidation and extend shelf life. The goal of the study is to investigate the activity of novel antioxidants in menhaden oil and to develop optimum formulations containing mixed tocopherols to control oxidation of menhaden oil. Alpha tocopherol, gamma tocopherol, and delta tocopherol in menhaden oil were found at 0.18mg/g, 0.37mg/g, and 0.14mg/g, respectively, using HPLC analysis. Teng Cha extract effectively delayed oxidation of menhaden oil (MO) when stored at 40°C for eight days by measuring primary oxidation products and secondary oxidation products. The combinations of Teng Cha extract and rosemary extract and combinations of ascorbyl palmitate, citric acid, Teng Cha extract and rosemary extract more effectively improved stability of MO containing mixed tocopherols than Teng Cha extract alone at 40°C storage for eight days by measuring primary oxidation products and secondary oxidation products. From this study, Teng Cha extract can be used as a potential natural antioxidant in food industry, especially in combinations with rosemary extract and tocopherols, extending shelf life of menhaden oil. / Master of Science in Life Sciences

Atténuation des oxydations phosphorylantes et induction d'une réponse cellulaire hypoxique : effêt de l'[alpha]-tocophérol-acétate et de miR-210 sur les cellules stromales mesenchymateuses / Attenuation of oxidative phosporylation and induction of hypoxic celle response : [alpha]-tocopherolacetate and miR-210 effects on mesenchymal stromal cells

Dans cette thèse, nous avons combiné les approches des cultures single-cell, de cytométrie en flux,des analyses de métabolisme énergétique et de génétique moléculaire afin d’explorer les effets del’Acétate d’α-Tocopherol (α-TOA) sur les cellules stromales mésenchymateuses (MStroC) et leurs souspopulations fonctionnelles (cellules souches et progénitrices mésenchymateuses). L’autre but était de tester une molécule de miR-210 par rapport à son utilisation potentielle comme « hypoxia mimicking molecule ». Après avoir démontré l’hétérogénéité de la population MStroC et conclu que la population de premier passage est appropriée pour des expérimentations ultérieures, nous avons trouvé que l’α-TOA présentait un effet positif sur le maintien de la capacité proliférative élevée des cellules souches mésenchymateuses. Cet effet est accompagné d’une atténuation de l’activité de la chaîne de transport d’électrons (ETC) qui pourrait d’autre part expliquer l’accroissement modéré du niveau des Reactive Oxygen Species mitochondriales (mtROS) que nous avons détectées. L’augmentation du niveau de mtROS pourrait être associée à une dégradation de protéine HIF-1 dans la population MStroC exposée à l’α-TOA. Bien que nous n’ayons pas détecté d’augmentation compensatoire de la glycolyse, les phénomènes observés représentent en partie la réponse cellulaire complexe au faible niveau d’O2. Il a été établi que ce phénomène était relié au maintien de primitivité des cellules souches. Le mécanisme exact reste à clarifier ainsi que son potentiel translationnel. En outre, nous avons apporté la preuve que miR-210 fait partie intégrante de la réponse des MStroC à la faible concentration en O2. Dans cette étude, nous avons montré que l’augmentation de l’expression de miR-210 sur une période courte (jusqu’à 24 heures) et après une période étendue (jusqu’à 72 heures) d’exposition des MStroC à une faible concentration en O2. De plus, nous avons prouvé que ce micro ARN pouvait être régulé par les deux facteurs transcriptionnels HIF-1 et HIF-2, nous laissant penser que ceci faisait partie intégrante de la réponse des MStroC à une faible concentration en O2. Jusqu’à présent, nos données suggèrent que miR-210 est digne d’intérêt en tant que bonne molécule « hypoxia mimicking ». / In this thesis, we combined approaches of single-cell cultures, flow-cytometry, energetic metabolismanalysis and molecular genetics in order to get insight in the effects of α-Tocopherol-Acetate (α-TOA)on Mesenchymal Stromal Cells (MStroC) and their functional subpopulations (mesenchymal stem and progenitor cells). The other aim was to test a miR-210 molecule with respect to its potential use as hypoxia mimicking molecule. After defining MStroC population heterogeneity and concluding that the first passage population is convenient for further experiments, we demonstrated that α-TOA exhibits a positive effect on the maintenance of high proliferative capacity of mesenchymal stem cells. This effect could be associated with an attenuation of electron transport chain (ETC) activity, which, on the other hand could explain moderate increase in the level of mitochondrial Reactive Oxygen Species (mtROS) we detected. The increase in mtROS level could be associated with a decreased HIF-1 alpha protein degradation in MStroC exposed to α-TOA. Although we did not detect a compensatory increase in glycolysis, the observed phenomena depict part of a complex cellular response to the low O2 that is demonstrated to be related with maintenance of stem cell primitiveness. The exact mechanism remains to be elucidated as well as its translational potential. In addition, we provided new evidences that miR-210 is integral part in MStroC response to low O2. In the study, we showed increased in miR-210 expression in a short-term (up to 24 hours) and after extended (up to 72 hours) MStroC exposed to low O2. Moreover, we demonstrated that this micro- RNA could be regulated by both HIF-1 and HIF-2 transcriptional factors, suggesting it as integral part of MStroC response to low O2. So far, our data suggest that miR-210 is worthy to be considered as good hypoxia mimicking molecule.

Cell Toxicity and Uptake of RRR-Alpha-Tocopheryl Polyethylene Glycol 1000 Succinate (TPGS) by Various Cell ines <em>In Vitro</em>.

This research focused on investigating and comparing the cytotoxicity and cellular uptake of RRR-alpha-tocopheryl polyethylene glycol succinate (TPGS, with that of alpha-tocopheryl succinate (α-TS). Both TPGS and α-TS are water-soluble forms of vitamin E with important clinical applications. Cytotoxicity assays with RAW 264.7 and LNCaP cells incubated overnight with TPGS or α-TS at concentrations ≥ 12.4 μM suggest that α-TS is more cytotoxic than TPGS. Macrophages were found to be more sensitive than LNCaP cells when treated with similar concentrations of α-TS. For both cell lines, most of the TPGS or α-TS taken up remained esterified after 24 hours. Our results suggest that cell death was due to TPGS and/or α-TS and not alpha-tocopherol. A para-hydroxyanilide of α-TS (p-HATS) that could be used to distinguish between cellular TPGS and α-TS was studied. It was found that p-HATS can be detected electrochemically and that it is hydrolyzed to α-TOH.

Объектами исследования являются витамины разной гидрофильности и витаминосодержащие препараты в таблетированной форме. Цель работы: исследование антиоксидантных свойств витаминных препаратов различной гидрофильности. Исследована антиоксидантная емкость модельных растворов и смесей антиоксидантов разной гидрофильности потенциометрическим методом; подобраны оптимальные условия для проведения анализа липофильного и гидрофильного соединений в совместном присутствии; исследованы фармацевтические препараты в таблетированной форме, содержащие витамины С и Е; проведено сравнение результатов с методом Фолина-Чокальтеу. Purpose of the work: study of the antioxidant properties of vitamin preparations of various hydrophilicity. The antioxidant capacity of model solutions and mixtures of antioxidants of different hydrophilicity was investigated by potentiometric method; the optimal conditions were selected for the analysis of lipophilic and hydrophilic compounds in their joint presence; investigated pharmaceutical preparations in tablet form containing vitamins C and E; the results are compared with the Folin-Chocalteu method. The possibility of using the method for determining AOE in a microfluidic device is considered.

Maternal nutrition and the risk of preeclampsia

La prééclampsie est responsable du quart des mortalités maternelles et est la deuxième cause de décès maternels associés à la grossesse au Canada et dans le monde. L’identification d’une stratégie efficace pour la prévention de la prééclampsie est une priorité et un défi primordial dans les milieux de recherche en obstétrique. Le rôle des éléments nutritifs dans le développement de la prééclampsie a récemment reçu davantage d’attention. Plusieurs études cliniques et épidémiologiques ont été menées pour déterminer les facteurs de risque alimentaires potentiels et examiner les effets d’une supplémentation nutritive dans le développement de troubles hypertensifs de la grossesse. Pour déterminer les effets de suppléments antioxydants pris pendant la grossesse sur le risque d’hypertension gestationnelle (HG) et de prééclampsie, un essai multicentrique contrôlé à double insu a été mené au Canada et au Mexique (An International Trial of Antioxidants in the Prevention of Preeclampsia – INTAPP). Les femmes, stratifiées par risque, étaient assignées au traitement expérimental quotidien (1 gramme de vitamine C et 400 UI de vitamine E) ou au placebo. En raison des effets secondaires potentiels, le recrutement pour l’essai a été arrêté avant que l’échantillon complet ait été constitué. Au total, 2640 femmes éligibles ont accepté d’être recrutées, dont 2363 (89.5%) furent incluses dans les analyses finales. Nous n’avons retrouvé aucune évidence qu’une supplémentation prénatale de vitamines C et E réduisait le risque d’HG et de ses effets secondaires (RR 0,99; IC 95% 0,78-1,26), HG (RR 1,04; IC 95% 0,89-1,22) et prééclampsie (RR 1,04; IC 95% 0,75-1,44). Toutefois, une analyse secondaire a révélé que les vitamines C et E augmentaient le risque de « perte fœtale ou de décès périnatal » (une mesure non spécifiée au préalable) ainsi qu’une rupture prématurée des membranes avant terme. Nous avons mené une étude de cohorte prospective chez les femmes enceintes recrutées dans l’INTAPP afin d’évaluer les relations entre le régime alimentaire maternel en début et fin de grossesse et le risque de prééclampsie et d’HG. Un questionnaire de fréquence alimentaire validé était administré deux fois pendant la grossesse (12-18 semaines, 32-34 semaines). Les analyses furent faites séparément pour les 1537 Canadiennes et les 799 Mexicaines en raison de l’hétérogénéité des régimes alimentaires des deux pays. Parmi les canadiennes, après ajustement pour l’indice de masse corporelle (IMC) précédant la grossesse, le groupe de traitement, le niveau de risque (élevé versus faible) et les autres facteurs de base, nous avons constaté une association significative entre un faible apport alimentaire (quartile inférieur) de potassium (OR 1,79; IC 95% 1,03-3,11) et de zinc (OR 1,90; IC 95% 1,07-3,39) et un risque augmenté de prééclampsie. Toujours chez les Canadiennes, le quartile inférieur de consommation d’acides gras polyinsaturés était associé à un risque augmenté d’HG (OR 1,49; IC 95% 1,09-2,02). Aucun des nutriments analysés n’affectait les risques d’HG ou de prééclampsie chez les Mexicaines. Nous avons entrepris une étude cas-témoins à l’intérieur de la cohorte de l’INTAPP pour établir le lien entre la concentration sérique de vitamines antioxydantes et le risque de prééclampsie. Un total de 115 cas de prééclampsie et 229 témoins ont été inclus. Les concentrations de vitamine E ont été mesurées de façon longitudinale à 12-18 semaines (avant la prise de suppléments), à 24-26 semaines et à 32-34 semaines de grossesse en utilisant la chromatographie liquide de haute performance. Lorsqu’examinée en tant que variable continue et après ajustement multivarié, une concentration de base élevée de gamma-tocophérol était associée à un risque augmenté de prééclampsie (quartile supérieur vs quartile inférieur à 24-26 semaines : OR 2,99, IC 95% 1,13-7,89; à 32-34 semaines : OR 4,37, IC 95% 1,35-14,15). Nous n’avons pas trouvé de lien entre les concentrations de alpha-tocophérol et le risque de prééclampsie. En résumé, nous n’avons pas trouvé d’effets de la supplémentation en vitamines C et E sur le risque de prééclampsie dans l’INTAPP. Nous avons toutefois trouvé, dans la cohorte canadienne, qu’une faible prise de potassium et de zinc, tel qu’estimée par les questionnaires de fréquence alimentaire, était associée à un risque augmenté de prééclampsie. Aussi, une plus grande concentration sérique de gamma-tocophérol pendant la grossesse était associée à un risque augmenté de prééclampsie. / Preeclampsia (PE) accounts for about one-quarter of cases of maternal mortality, and ranks second among the causes of pregnancy-associated maternal deaths in Canada and worldwide. The identification of an effective strategy to prevent PE is a priority and fundamental challenge in obstetrics research. The role of nutritional factors in the etiology of PE has recently received increased attention. Many clinical and epidemiological studies have been conducted to investigate potential dietary risk factors for PE and to examine the effects of nutritional supplementation on the development of hypertensive disorders of pregnancy. To investigate the effects of prenatal antioxidant supplementation on the risk of gestational hypertension (GH) and PE, a double blind, multicenter trial (The International Trial of Antioxidants for the Prevention of Preeclampsia – the INTAPP trial) was conducted in Canada and in Mexico. Women were stratified by their risk status and assigned to daily experimental treatment (1 gram vitamin C and 400 IU vitamin E) or to placebo. Due to concerns about potential adverse effects, recruitment for the trial was stopped before the full sample had been achieved. A total of 2640 consenting eligible women had been recruited at that point with 2363 women (89.5%) included in the final analysis. We found no evidence that prenatal supplementation of vitamins C and E reduced the risk of GH and its adverse conditions (RR: 0.99, 95% CI 0.78-1.26), GH (RR 1.04, 95% CI 0.89-1.22), and PE (RR 1.04, 95% CI 0.75-1.44). However, in a secondary analysis, we found that vitamins C and E increased the risk of ‘fetal loss or perinatal death’ (a non-pre-specified outcome) as well as preterm premature rupture of membranes (PPROM). We conducted a prospective cohort study on pregnant women enrolled in the INTAPP trial to investigate the associations between maternal diet in early and late pregnancy and the risk of PE and GH. A validated food frequency questionnaire (FFQ) was administered twice during pregnancy (12-18 weeks, 32-34 weeks). Analyses were conducted separately for 1537 Canadian and 799 Mexican women as there were significant heterogeneities in various nutrient intakes between the two countries. Among Canadian women, after adjusting for pre-pregnancy body mass index (BMI), treatment group, risk stratum (high versus low) and other baseline risk factors, we found that the lowest quartiles of potassium (OR 1.79, 95% CI 1.03-3.11) and zinc (OR 1.90, 95% CI 1.07-3.39) intake were significantly associated with an increased risk of PE. Also in Canadian women, the lowest quartile of polyunsaturated fatty acids was associated with an increased risk of GH (OR 1.49, 95% CI 1.09-2.02). None of the nutrients analyzed were found to be associated with PE and GH risk among Mexican women. We further conducted a case control study ancillary to the INTAPP trial to assess the relationship between plasma concentration of antioxidant vitamins and the risk of PE. A total of 115 PE cases and 229 matched controls were included. Vitamin E concentrations were measured longitudinally at 12-18 weeks (prior to supplementation), 24-26 weeks, and 32-34 weeks of gestation using high-performance liquid chromatography (HPLC). When examined as a continuous variable, and after multivariate adjustment, elevated baseline gamma-tocopherol concentrations were associated with an increased risk of PE (OR 1.35, 95% CI 1.02-1.78). Analyses of repeated measurements indicated that elevated gamma-tocopherol levels were associated with an increased risk of PE (highest vs. lowest quartile at 24-26 weeks: OR 2.99, 95% CI 1.13-7.89; at 32-34 weeks: OR 4.37, 95% CI 1.35-14.15). We found no associations between alpha-tocopherol concentrations and the risk of PE. In summary, we found no effects of vitamins C and E supplementation on the risk of PE in the INTAPP trial. However, in the Canadian cohort we found that lower intakes of potassium and zinc as estimated by the FFQ were associated with an increased risk of PE. Moreover, higher plasma concentration of gamma-tocopherol during pregnancy was associated with an increased risk of PE.

Etude et fonctionnalisation de protéines végétales en vue de leur application en microencapsulation / Study and functionalization of vegetable proteins and their application in microencapsulation

Les protéines extraites des végétaux sont des matériaux relativement peu coûteux, non toxiques, biocompatibles et biodégradables. Elles représentent une bonne alternative aux protéines d’origine animale et aux polymères dérivés du pétrole. Dans le cadre de cette étude, les protéines extraites de graines de soja et de tournesol ont été utilisées en tant que matériaux enrobants pour la microencapsulation de la matière active hydrophobe (α-tocophérol) ou hydrophile (acide ascorbique) par le procédé d’atomisation. Les protéines de soja sont largement utilisées dans les applications alimentaires et non-alimentaires, notamment en microencapsulation. Elles sont donc étudiées dans ce travail comme matériau enrobant de référence. Les protéines de tournesol n’ont quant à elles pas d’application industrielle concrète, si ce n’est sous la forme de tourteaux dans l’alimentation animale. C’est pourquoi il nous semble pertinent de trouver des nouvelles voies de valorisation pour ce coproduit d’origine agricole. Plusieurs modifications des protéines, telles que l’hydrolyse enzymatique, l’acylation, la réticulation enzymatique et la cationisation ont été étudiées dans le but d’améliorer les propriétés encapsulantes du matériau enrobant. Dans le contexte de la chimie verte, toutes les modifications ont été effectuées sans utilisation de solvants organiques ni de catalyseurs chimiques. L’influence des modifications chimiques et enzymatiques des protéines, et des paramètres du procédé (pression d’homogénéisation, ratio matériau enrobant/matière active et concentration en protéines) sur les différentes caractéristiques des préparations liquides et des microparticules (viscosité, taille des gouttelettes dans le cas des émulsions, morphologie et taille des microparticules), ainsi que sur les paramètres liés au procédé d’atomisation (rendement et efficacité de microencapsulation) a été particulièrement étudiée au cours de ce travail. Les résultats obtenus confirment que l’extrait protéique de tournesol est tout à fait pertinent comme matériau enrobant et permet d’obtenir des efficacités de microencapsulation significativement plus élevées par rapport à celles obtenues avec l’extrait protéique de soja. / Proteins extracted from vegetables are relatively low-cost, non-toxic, biocompatible and biodegradable raw materials. They represent a good alternative to animal-based proteins and petroleum-extracted polymers. In this study, proteins derived from soybean and sunflower seeds were used as wall materials for microencapsulation of hydrophobic (-tocopherol) or hydrophilic (ascorbic acid) active material by spray-drying technique. Soybean proteins are widely used in food and non-food applications, especially in microencapsulation. They were studied in this work as wall material of reference. Sunflower proteins are not actually used in industrial application, but only in the form of oil-cake for animal feeding. That’s why new ways of valorization of this agricultural by-product should be investigated. Several proteins’ modifications such as enzymatic hydrolysis, acylation, cross-linking and cationization were studied in order to improve encapsulating properties of wall material. In the context of green chemistry, all the modifications and preparations were performed without use of organic solvents and chemical catalysts. The effect of protein chemical and enzymatic modifications, and process parameters (homogenization pressure, wall/core ratio and protein concentration) on different characteristics of liquid preparations and microparticles (viscosity, emulsion droplet size, microparticle size and morphology) and on parameters related to the spray-drying process (yield and efficiency of microencapsulation) was particularly investigated in this study. The obtained results confirmed that sunflower proteins are quite suitable as encapsulating agent and provide the microencapsulation efficiencies significantly higher compared to those obtained with soy proteins.
