Feline immunodeficiency virus: molecular subtyping and evaluation of potential prognostic indicators

Abstract Feline immunodeficiency virus (FIV) is an important infectious agent of domestic cats worldwide. It has been classified into the Lentivirus genus of the Retroviridae family, together with human immunodeficiency virus (HIV). Five FIV subtypes (A, B, C, D and E) have been described based on sequence variation of the V3-V5 region of the envelope (env) gene. There is considerable sequence diversity within and between subtypes, which has been a major obstacle in the development of a successful vaccine. However, an FIV vaccine that incorporates inactivated whole viruses from subtypes A and D is now commercially available. Although the vaccine has been shown to be efficacious in protecting against challenge with homologous and a heterologous (subtype B) subtypes, its effectiveness against other viral variants is unknown. Therefore, identifying the type and diversity of FIV strains in different regions is important to establish the potential efficacy of the vaccine in areas where vaccination is to be implemented. The proviral DNA sequence of the V3-V5 region of the env gene was determined for 102 FIV-infected cats from locations in Australia, New Zealand and South Africa. Subtype A was the predominant subtype in Australia and South Africa, although subtype B and C were also identified in each of these countries, respectively. Both subtypes A and C were also present in New Zealand. Of interest, there were some samples in New Zealand and South Africa that demonstrated subtype assignment discrepancies when different regions of the genome were analysed, suggesting co-infection and/or recombination. Cats infected with FIV exhibit varying degrees of immunological impairment. Currently, prognosis for an FIV-infected cat is based on clinical signs alone, which is a relatively subjective measure. In HIV-infected patients it is recognised that viral RNA load correlates with disease stage and prognosis. This PhD research tested whether viral RNA load may be a useful prognostic marker in FIV infection. A real-time PCR assay was developed to quantify plasma viral RNA load in 42 FIV-infected cats at three different clinical stages (1:healthy, 2:unwell without signs of immunodeficiency, 3:unwell with signs of immunodeficiency). In cats older than 5 years of age, log-transformed viral RNA loads were significantly higher in cats in category 3 compared to cats in category 1. There were no significant differences in the viral RNA load of older cats in category 2 compared to category 1. There were no cats younger than 5 years of age in category 3 and there was no significant difference in viral RNA load between young cats in categories 1 and 2. Of the 15 cats for which follow-up data was available, eight showed no change in clinical signs, and seven showed a worsening of clinical signs with six of these showing a progression of clinical category including death. One of the cats in category 2 that progressed clinically had one of the highest viral RNA loads of cats in that category. Three of four cats from category 3 that were followed had either died or been euthanised. Two of these cats had among the highest viral RNA loads in the whole study, while the remaining cat (for which the definitive cause of death was not confirmed) had a relatively low viral RNA load. In summary, measurement of viral RNA load was found to be a potentially useful clinical and prognostic marker but further work is required to better assess its usefulness to veterinarians. Serum acute phase proteins were investigated as possible candidate markers of FIV disease with the aim of developing a more simplified assay that could be used as a prognostic marker for FIV infection. Blood samples from 43 FIV-infected and 25 FIV-negative cats were assayed for the concentration of four acute phase proteins. Both healthy and sick cats were included in the study. Compared to healthy cats, sick cats had significantly higher concentrations of serum amyloid A (P<0.05). Alpha 1-acid glycoprotein and haptoglobin were also found to be in higher concentrations in sick cats (P<0.1). Other variables such as age and gender were also associated with acute phase protein concentrations. With respect to FIV infection, it was found that in sick cats, serum amyloid A, in combination with the age of the cat, was the best predictor of FIV viral RNA load. Alpha 1-acid glycoprotein and haptoglobin were not significantly associated with FIV viral RNA load. Although health status did not influence albumin levels, they were found to be significantly lower in FIV-positive cats in comparison to FIV-negative cats (P<0.05). The frequent monitoring of viral RNA loads and CD4+ lymphocyte counts that is performed on HIV-infected patients is cost prohibitive in veterinary patients. This study showed that there is potential for the use of acute phase protein concentrations (in particular serum amyloid A) as alternative prognostic tools in FIV-infected cats. Further work, particularly longitudinal studies, is required to more definitively define changes in viral RNA load and acute phase protein concentrations throughout the course of FIV infection.

Análise do líquido endometrial de éguas suscetíveis à endometrite: efeito da corticoterapia / Analysis of the endometrial fluid of mares susceptible to endometritis: effect of corticotherapy

A modulação da resposta inflamatória uterina equina recebe atenção, devido a um distúrbio imunológico, que parece ocorrer em uma população de éguas, classificadas como suscetíveis à endometrite. O objetivo deste estudo foi verificar o efeito da corticoterapia aplicada na presença e na ausência de inflamação uterina sobre a proteômica e a concentração de óxido nítrico no líquido endometrial de éguas suscetíveis à endometrite. Éguas foram sincronizadas com 5 mg de prostaglandina F2α e após a verificação dos sinais de estro foram submetidas a quatro tratamentos. O primeiro foi o Controle, que não recebeu nenhum tipo de tratamento. O segundo foi o GC, onde as éguas receberam aplicação de um glicocorticóide, a cada 12 horas, por três dias consecutivos. O terceiro foi o Infectado, onde as éguas receberam uma infusão intrauterina de Streptococcus zoopepidemicus (1x109/mL) e o quarto foi o GC + Infectado, onde as éguas receberam a aplicação do glicocorticóide e a infusão intrauterina descrita acima. Doze horas após o final de cada tratamento, amostras de líquido endometrial puro e de lavados uterinos foram coletadas para análise proteômica e determinação de óxido nítrico, respectivamente. O primeiro artigo relata os dados preliminares da análise proteômica, com a contagem das bandas protéicas e a observação de 33 bandas protéicas no Controle, 54 no GC, 51 no Infectado e 72 no GC + Infectado. A corticoterapia pode induzir o aparecimento de um número maior de bandas protéicas, pois os géis com as maiores contagens foram nos tratamentos onde ela foi aplicada. No segundo artigo, foi realizada a identificação das bandas protéicas significativas, em relação à densidade óptica relativa e à frequência. A corticoterapia provocou uma alteração na proteômica do líquido endometrial, caracterizada pelo aumento e diminuição na densidade óptica relativa e na frequência de proteínas da fase aguda da inflamação, com as maiores alterações observadas quando a corticoterapia foi aplicada na presença do processo infeccioso. A infusão de Streptococcus zooepidemicus provocou alterações na proteômica do líquido endometrial, caracterizadas pelo aumento e diminuição na densidade óptica relativa e na frequência de proteínas da fase aguda da inflamação. Os resultados do estudo indicam que a corticoterapia provoca alterações imunológicas no endométrio equino, não apenas como depressiva, mas estimuladora da defesa local, através de uma ação imunomoduladora. No terceiro artigo, foi realizada a determinação da concentração de óxido nítrico, não sendo observada diferença significativa nos quatro tratamentos. Portanto, a corticoterapia provoca alterações proteômicas no líquido endometrial de éguas suscetíveis em estro, onde a presença de um estímulo inflamatório causado pela infusão intrauterina bacteriana induz a uma maior alteração, do que a ausência. A infecção do lúmen uterino provoca alterações proteômicas no líquido endometrial e a corticoterapia não influencia a concentração de óxido nítrico de éguas em estro. / The modulation of the equine uterine inflammatory response recieves much attention, due to an immunological disorder, which appears to happen in a population of mares, classified as suscetipble to endometritis. The aim of this study was to verify the effect of corticotherapy applied in the presence and in the absence of uterine inflammation on the proteomics and nitric oxide concentration of the endometrial fluid of mares susceptible to endometritis. Mares were synchronized with 5 mg prostaglandin F2α and after the observation of the signs of estrus were submitted to four treatments. The first was the Control, which did not recieve any kind of treatment. The second was the GC, where mares recieved the administration of a glucocorticoid, each 12 hours, for three consecutive days. The third was the Infected, where mares received an intrauterine infusion of Streptococcus zoopepidemicus (1x109/mL) and the fourth was the GC + Infected, where mares received the administration of glucocorticoid and intrauterine infusion as described above. Twelve hours after the end of each treatment, pure endometrial fluid and uterine flushings were collected for proteomic analysis and nitric oxide determination, respectively. The first article reports the preliminary data of the proteomic analysis, where protein band counts were done, being observed 33 protein bands in Control, 54 in GC, 51 in Infected and 72 in GC + Infected. Corticotherapy can induce the appearance of a higher number of protein bands, because the gels with the highest counts were in the treatments where it was applied. In the second article, the identification of the significative protein bands was done, regarding the relative optic density and frequency. Corticotherapy provoked an alteration in the endometrial proteomics, characterized by an increase and a decrease on the relative optic density and frequency of inflammatory acute phase proteins, with the major alterations observed when corticotherapy was applied in the presence of an infectious process. Streptococcus zooepidemicus infusion provoked alterations in the endometrial fluid proteomics, characterized by an increase and a decrease on the relative optic density and frequency of inflammatory acute phase proteins. Results from this study indicate that corticotherapy provokes immunological alterations in the equine endometrium, not only as depressor, but enhancer of local defense, through an immunomodulatory action. In the third article, the nitric oxide concentration was determined, with no significative diference observed in the four treatments. So, corticotherapy provokes alterations in the proteomics of the endometrial fluid of susceptible mares in estrus, where the presence of an inflammatory stimulus caused by intrauterine bacterial infusion induces a major alteration, than the absence. Uterine lumen infection provokes alterations in the proteomics of the endometrial fluid and corticotherapy does not influence nitric oxide concentration in mares in estrus.

Salvador, BA, Brasil / A confirmação da leptospirose utilizando o Teste de Aglutinação Microscópica (MAT) requer amostras da fase aguda e convalescente para identificar soroconversão ou aumento de quatro vezes nos títulos. A Organização Mundial de Saúde (OMS) recomenda que a coleta da amostra convalescente seja realizada ≥14 dias após a coleta da amostra aguda. No entanto, a dificuldade na coleta de amostras convalescentes impede a confirmação dos casos e é uma das principais causas para sub-notificação da leptospirose. Este estudo investigou a viabilidade da coleta de uma amostra de soro aguda tardia de casos internados com leptospirose e avaliou se a análise sorológica desta amostra pode melhorar a eficiência do protocolo de confirmação diagnóstica de leptospirose. De 2003 a 2009, uma vigilância hospitalar ativa em Salvador-Brasil, identificou prospectivamente pacientes hospitalizados com suspeita clínica da leptospirose. Três amostras de sangue foram coletadas para cada caso: uma amostra aguda precoce, uma amostra aguda tardia e uma amostra convalescente, coletadas respectivamente nas primeiras 24 horas após hospitalização, e 4 e ≥14 dias depois da coleta da primeira amostra. Os pacientes identificados tiveram o diagnóstico de leptospirose confirmado por soroconversão, aumento de quatro vezes de títulos, ou título único ≥1:800 no MAT. O desempenho diagnóstico do MAT e do ELISA IgM na avaliação combinada das amostras aguda precoce e aguda tardia foi comparado ao desempenho da avaliação das amostras aguda precoce e convalescente que segue a recomendação de testagem da OMS. Nós confirmamos 643 (68%) dos 938 casos suspeitos. A coleta de amostra convalescente foi possível para 63% dos pacientes confirmados, e 55% dos pacientes suspeitos. Em contraste, a amostra da fase aguda tardia foi coletada para 77% e 66% dos pacientes confirmados e suspeitos, respectivamente. Para os 302 casos confirmados que tiveram as três amostras de soro coletadas, a sensibilidade do MAT e do IgM-ELISA na análise das amostras aguda precoce e tardia foi de 97% (IC95%, 94-99%) e 96% (93-98%), respectivamente, em comparação aos resultados da análise das amostras aguda precoce e convalescente. Em contraste, considerando apenas as amostras agudas destes 302 pacientes, a sensibilidade do MAT e do IgM-ELISA foi de 44% (38-50%) e 75% (69-79%), respectivamente. Amostra aguda tardia e convalescente foi obtida dos casos suspeitos de leptospirose que evoluíram para óbito de 32% e 6%, respectivamente. Os resultados indicam que a coleta e o teste sorológico da amostra aguda tardia de pacientes hospitalizados por leptospirose é viável e melhora a eficiência dos atuais protocolos de confirmação laboratorial de casos de leptospirose. / Confirmation of leptospirosis with MAT requires evaluating acute and convalescent-phase sera samples to identify seroconversion or fourfold rise in titers. Current World Health Organization (WHO) protocols recommend that convalescent samples are collected with ≥14days after the acute sample collection. However, the difficulty in collecting convalescent samples hampers case confirmation and is a major cause for leptospirosis under-reporting. This study evaluated feasibility of collecting a late acute-sera sample from hospitalized cases of leptospirosis and determined to serological analysis of this sample can improve the efficiency of the protocol to confirm the diagnosis of leptospirosis. From 2003 to 2009, active hospital-based surveillance in Salvador-Brazil prospectively identified hospitalized cases of patients with clinical suspicion of leptospirosis. Three blood samples were collected for each case: an early acute sample, a sample of late acute and convalescent sample collected during the first 24 hours after hospitalization 4 and ≥ 14 days after the first sampling, respectively. The identified patients were diagnosed with leptospirosis by seroconversion, fourfold rise in titers, or a titer ≥1:800 in the MAT. The diagnostic performance of the MAT and IgM ELISA in the combined sample of early acute and late acute sample performance was compared to the early assessment of acute and convalescent samples following a WHO recommendation for testing. We confirmed the leptospirosis diagnosis in 643 (68%) of 938 suspected cases. Convalescent-phase samples were collected from 63% of the confirmed patients, but in only 55% of the suspected cases. In contrast, the late acute phase sample was collected for 77% and 66% of confirmed and suspected patients, respectively. Among the 302 confirmed cases which all three samples were obtained, the sensitivity of MAT and IgM-ELISA was 97% (IC95%, 94-99%) and 96% (93-98%), respectively, when results of early and late acute-phase samples were evaluated in comparison to the results of the early acute and convalescent samples. In contrast, the sensitivity of MAT and IgM-ELISA was 44% (38-50%) and 75% (69-79%), respectively, when only a single early acute-phase sample was evaluated. Late acute-phase and convalescent-phase samples were obtained from 32% and 6% of the suspected leptospirosis and deaths, respectively. These findings indicate that collection and serologic testing of a late-acute-phase sample among hospitalized patients with suspected leptospirosis may significantly increase the efficiency of protocols for laboratory case confirmation.

O objetivo do presente estudo, prospectivo e randomizado, é avaliar comparativamente as respostas hemodinâmicas, laboratoriais, imunomoleculares e clínicas, em dois grupos de pacientes submetidos a cirurgia de revascularização miocárdica com extracorpórea utilizando-se, em um deles infusão de azul de metileno durante o período intraoperatório. As variáveis hemodinâmicas estudadas foram: FC, PAE, PVC, DC, RVS, RVP, PAPM, PASS, PASD e PASM. Os dados laboratoriais avaliados foram: gasometria arterial, creatinina, leucócitos, bastões, neutrófilos, eosinífilos, basófilos, linfócitos, Ht, Hb, paquetas, Na+ e K+. Os marcadores imunomoleculares estudados foram: IL - 1ß, IL-6, IL-8, IL-10, IL-12 p40, TNF alfa e NO. Os dados clínicos avaliados foram: idade, sexo, raça, tabagismo, hipertensão, diabetes, angina estável, angina instável, infarto prévio, tempo de CEC, tempo de pinçamento aórtico, número de pontes realizadas, urina azulada, fezes azuladas, diarréia, náuseas, cefaléia, tontura, disúria, astenia e dispnéia. As variáveis citadas foram obtidas em todos ou em parte dos períodos seguintes: antes da indução, terceira hora, sexta hora, 24ª hora e 48ª hora após o término da CEC. Ocorreram diferenças estatisticamente significativas na resistência vascular sistêmica, na pressão arterial sistêmica diastólica, em parte das citocinas estudadas e no óxido nítrico. A análise dos resultados mostra que no grupo que utilizou azul de metileno houve resistência vascular sistêmica mais alta no período da sexta hora de pós-operátório, menor concentração de TNF alfa no período da terceira e 48ª hora e menores níveis de óxido nítrico no período da sexta hora. As medidas laboratoriais foram semelhantes em ambos os grupos exceto maior número de leucócitos e neutrófilos na 48ª hora após CEC no grupo contrôle. Verifica-se que o azul de metileno levou a melhores condições hemodinâmicas e menores valores e alguns marcadores de reposta inflamatória sistêmica após cirurgia de revascularização miocárdica com circulação extra-corpórea e não apresentou efeitos adversos importantes. / The objective of the present study, prospective and randomized, is to evaluate the hemodynamic, gasometric, immunomolecular and clinical response in two groups of patients submited to coronary artery surgery under cardiopulmonary bypass with the use, in one of them, of methylene blue infusion during the intraoperative period. The hemodynamic variable data evaluated were: heart rate, left atrial pressure, central venous pressure, cardiac output, systemic vascular resistance, pulmonary vascular resistance, mean pulmonary artery pressure, systolic systemic arterial pressure, dyastolic systemic arterial pressure and mean systemic arterial pressure. Laboratoryal data were: arterial gasimetry, urea, creatinine, leucocytes, neutrophils, eosinophils, basophils, lymphocytes, hematocrit, hemoglobin, platelets, sodium and potassium. The immunomolecular data were: IL-1ß, IL-6, IL-8, IL-10, IL-12p40, TNF alfa and NO. The clinical data evaluated were: age, gender, race, smoking, hypertension, diabetes, stable angina, unstable angina, previous infarction, cardiopulmonary bypass duration, aortic cross-clamp time, graft number, bluish urine, bluish feces, diarrhea, nausea, headache, dizziness, dysuria, tiredness and dyspnea. All the data were obtained, in all or part of the following periods: before the anesthesia and at the third, sixth, 24th and 48th hour after the end of the cardiopulmonary bypass. Statistically significant differences occurred in the systemic vascular resistance, diastolic systemic arterial pressure and in part of the evaluated cytokines, as well as with the nitric oxide. The analysis of the results show that, in the methylene blue patients group, the systemic vascular resistance was higher at the sixth hour, lower concentrations of TNF alfa at the third and at the 48th hour and lower levels of NO at the 6th hour. The laboratorial data were similar in the two groups, except for the number of leucocytes and neutrophils were higher in the control group at the 48th hour after the end of the cardiopulmonary bypass. The gasometric data showed no difference between the groups. It was verified that the methylene blue has induced better hemodynamic conditions and lower levels of part of markers of SIRS after coronary artery surgery under cardiopulmonary bypass and no siginifcant deleterious side effects.

Análise do líquido endometrial de éguas suscetíveis à endometrite: efeito da corticoterapia / Analysis of the endometrial fluid of mares susceptible to endometritis: effect of corticotherapy

A modulação da resposta inflamatória uterina equina recebe atenção, devido a um distúrbio imunológico, que parece ocorrer em uma população de éguas, classificadas como suscetíveis à endometrite. O objetivo deste estudo foi verificar o efeito da corticoterapia aplicada na presença e na ausência de inflamação uterina sobre a proteômica e a concentração de óxido nítrico no líquido endometrial de éguas suscetíveis à endometrite. Éguas foram sincronizadas com 5 mg de prostaglandina F2α e após a verificação dos sinais de estro foram submetidas a quatro tratamentos. O primeiro foi o Controle, que não recebeu nenhum tipo de tratamento. O segundo foi o GC, onde as éguas receberam aplicação de um glicocorticóide, a cada 12 horas, por três dias consecutivos. O terceiro foi o Infectado, onde as éguas receberam uma infusão intrauterina de Streptococcus zoopepidemicus (1x109/mL) e o quarto foi o GC + Infectado, onde as éguas receberam a aplicação do glicocorticóide e a infusão intrauterina descrita acima. Doze horas após o final de cada tratamento, amostras de líquido endometrial puro e de lavados uterinos foram coletadas para análise proteômica e determinação de óxido nítrico, respectivamente. O primeiro artigo relata os dados preliminares da análise proteômica, com a contagem das bandas protéicas e a observação de 33 bandas protéicas no Controle, 54 no GC, 51 no Infectado e 72 no GC + Infectado. A corticoterapia pode induzir o aparecimento de um número maior de bandas protéicas, pois os géis com as maiores contagens foram nos tratamentos onde ela foi aplicada. No segundo artigo, foi realizada a identificação das bandas protéicas significativas, em relação à densidade óptica relativa e à frequência. A corticoterapia provocou uma alteração na proteômica do líquido endometrial, caracterizada pelo aumento e diminuição na densidade óptica relativa e na frequência de proteínas da fase aguda da inflamação, com as maiores alterações observadas quando a corticoterapia foi aplicada na presença do processo infeccioso. A infusão de Streptococcus zooepidemicus provocou alterações na proteômica do líquido endometrial, caracterizadas pelo aumento e diminuição na densidade óptica relativa e na frequência de proteínas da fase aguda da inflamação. Os resultados do estudo indicam que a corticoterapia provoca alterações imunológicas no endométrio equino, não apenas como depressiva, mas estimuladora da defesa local, através de uma ação imunomoduladora. No terceiro artigo, foi realizada a determinação da concentração de óxido nítrico, não sendo observada diferença significativa nos quatro tratamentos. Portanto, a corticoterapia provoca alterações proteômicas no líquido endometrial de éguas suscetíveis em estro, onde a presença de um estímulo inflamatório causado pela infusão intrauterina bacteriana induz a uma maior alteração, do que a ausência. A infecção do lúmen uterino provoca alterações proteômicas no líquido endometrial e a corticoterapia não influencia a concentração de óxido nítrico de éguas em estro. / The modulation of the equine uterine inflammatory response recieves much attention, due to an immunological disorder, which appears to happen in a population of mares, classified as suscetipble to endometritis. The aim of this study was to verify the effect of corticotherapy applied in the presence and in the absence of uterine inflammation on the proteomics and nitric oxide concentration of the endometrial fluid of mares susceptible to endometritis. Mares were synchronized with 5 mg prostaglandin F2α and after the observation of the signs of estrus were submitted to four treatments. The first was the Control, which did not recieve any kind of treatment. The second was the GC, where mares recieved the administration of a glucocorticoid, each 12 hours, for three consecutive days. The third was the Infected, where mares received an intrauterine infusion of Streptococcus zoopepidemicus (1x109/mL) and the fourth was the GC + Infected, where mares received the administration of glucocorticoid and intrauterine infusion as described above. Twelve hours after the end of each treatment, pure endometrial fluid and uterine flushings were collected for proteomic analysis and nitric oxide determination, respectively. The first article reports the preliminary data of the proteomic analysis, where protein band counts were done, being observed 33 protein bands in Control, 54 in GC, 51 in Infected and 72 in GC + Infected. Corticotherapy can induce the appearance of a higher number of protein bands, because the gels with the highest counts were in the treatments where it was applied. In the second article, the identification of the significative protein bands was done, regarding the relative optic density and frequency. Corticotherapy provoked an alteration in the endometrial proteomics, characterized by an increase and a decrease on the relative optic density and frequency of inflammatory acute phase proteins, with the major alterations observed when corticotherapy was applied in the presence of an infectious process. Streptococcus zooepidemicus infusion provoked alterations in the endometrial fluid proteomics, characterized by an increase and a decrease on the relative optic density and frequency of inflammatory acute phase proteins. Results from this study indicate that corticotherapy provokes immunological alterations in the equine endometrium, not only as depressor, but enhancer of local defense, through an immunomodulatory action. In the third article, the nitric oxide concentration was determined, with no significative diference observed in the four treatments. So, corticotherapy provokes alterations in the proteomics of the endometrial fluid of susceptible mares in estrus, where the presence of an inflammatory stimulus caused by intrauterine bacterial infusion induces a major alteration, than the absence. Uterine lumen infection provokes alterations in the proteomics of the endometrial fluid and corticotherapy does not influence nitric oxide concentration in mares in estrus.

Em cães as alterações leucocitárias podem ser evidenciadas em 72 horas, enquanto a elevação sérica das proteínas de fase aguda e a velocidade de hemossedimentação ocorrem mais precocemente sendo úteis na monitorização do processo inflamatório/infeccioso. A interleucina-6 é o principal regulador da resposta inflamatória, ocasionando o aumento da síntese hepática das proteínas de fase aguda, como a proteína C reativa e o fibrinogênio. A elevação da velocidade de hemossedimentação, por sua vez, é conseqüente ao aumento das proteínas de fase aguda. O diagnóstico da inflamação/infecção em cães é realizado por meio de técnicas manuais, carentes de precisão e padronização na maioria dos laboratórios veterinários. O objetivo desse estudo foi a investigação de parâmetros e métodos laboratoriais, que possam auxiliar no diagnóstico e monitoramento do processo inflamatório/infeccioso em cães. A avaliação e padronização de tais parâmetros poderão contribuir para o esclarecimento diagnóstico com maior precisão e em um menor intervalo de tempo e poderão ser incluídos na rotina laboratorial veterinária. Materiais e métodos: Foram estudados 117 cães adultos de diferentes raças divididos inicialmente em dois grupos: controle (n=31) e inflamatório/infeccioso (n=86). Posteriormente, os animais doentes foram subdivididos em: inflamatório (n=29) e infeccioso (n=57). O leucograma foi obtido em contador automatizado e a contagem diferencial de leucócitos realizada em esfregaços de sangue corados com May-Grünwald- Giemsa. Os valores de fibrinogênio e velocidade de hemossedimentação foram determinados por métodos automatizados, a proteína C reativa e a interleucina-6 usando kit de ELISA comercial espécie-específicos. Resultados: A velocidade de hemossedimentação automatizada foi o melhor marcador do processo inflamatório/infeccioso, seguida da proteína C reativa. Apresentaram positividade em 82,56% e 65,12% dos casos, respectivamente. Os demais marcadores foram positivos em menos de 50% dos animais. Quando os animais doentes foram subdivididos, o grupo com inflamação apresentou maior freqüência de positividade do que o grupo infeccioso. A concentração de hemoglobina e o número de plaquetas foram superiores no grupo controle, enquanto a contagem total de leucócitos, número de basófilos e presença de desvio à esquerda apresentaram valores elevados no grupo de animais doentes. Níveis superiores de velocidade de hemossedimentação, proteína C reativa e interleucina-6 foram observados no grupo com inflamação/infecção quando comparados aos controles. Quando subdivididos, no grupo de animais com processo inflamatório os níveis de fibrinogênio e proteína C reativa foram superiores aos do grupo infeccioso, enquanto os valores de velocidade de hemossedimentação e interleucina-6 foram semelhantes. Conclusões: A determinação da velocidade de hemossedimentação apresentou a melhor acurácia na detecção do processo inflamatório/infeccioso. Na análise global os níveis de interleucina-6, proteína C reativa e fibrinogênio foram superiores no grupo inflamatório/infeccioso, mas mostraram eficácia limitada na detecção individual de cães doentes. Cães com processo inflamatório mostraram uma resposta mais exacerbada tanto nas determinações sorológicas/plasmáticas como nos parâmetros hematológicos do que os cães com processo predominantemente infeccioso / Abstract: Introduction: Leukocytosis and hyperthermia are the primary indicators of inflammation infection in most animal species. White blood cell count changes are evident after 72 hours in dogs, while the increase of serum acute phase proteins levels and erythrocyte sedimentation rate occur earlier. Thus they are considered as useful and precocious markers in identifying and monitoring inflammatory/infection process. Interleukin-6 is the major inflammatory response regulator and increases hepatic synthesis of acute phase proteins such as C-reactive protein and fibrinogen. Erythrocyte sedimentation rate rises as a consequence of acute phase protein stimulus. The investigation of the presence of inflammation/infection process in dogs is performed in most veterinary laboratories by manuals technics, which show poor precision and fail in standardization of procedures. The aim of this study was to evaluate new parameters and laboratory methods, which may help in the identification of the inflammatory/infection process in dogs. The evaluation and standardization of these parameters may contribute to the diagnosis with greater precision and in a shorter period of time and may be included in the routine veterinary laboratories. Materials and methods: We studied 117 adult dogs of different breeds. They were divided into two groups: normal controls (n = 31) and suspicious of inflammatory / infectious diseases (n = 86). Subsequently, animals were subdivided into animals with inflammatory (n = 29) or infection conditions (n = 57). Hematological parameters were obtained using an automated counter and differential count was performed by microscopic observation in blood smears stained with Giemsa-May-Grünwald. Fibrinogen concentration and erythrocyte sedimentation rate were determined by automated methods and plasma Creactive protein and interleukin-6 were measured by commercial ELISA kits. Results: Erythrocyte sedimentation rate was the most sensitive marker of the inflammatory/infection process, showing significant increase in 82,56% of the cases, followed by C-reactive protein measurement (65,12%). The other markers were positive in less than 50% of the animals. When the animals were subdivided according to inflammatory or infection condition the group with inflammation showed higher frequency of positivity than the infectious group. The concentration of hemoglobin and the number of platelet were higher in the control group, while white blood count, number of basophils and presence of left shift showed high values in the group of sick animals. Erythrocyte sedimentation rate, C-reactive protein and interleukin-6 levels were higher in the group with inflammation/infection when compared to controls. Animals with inflammatory process showed fibrinogen and C-reactive protein levels higher than those with infection, while erythrocyte sedimentation rate and interleukin-6 levels were similar. Conclusions: Results of this study indicate that erythrocyte sedimentation rate showed the best accuracy in identifying inflammatory/infectious process. Interleukin-6, C-reactive protein and fibrinogen levels were higher in inflammatory/infectious group, but showed limited efficacy in detection of individual sick animals.
