Influência do peptídeo P34 na expressão gênica em Listeria spp. e estudo da citotoxicidade dos peptídeos P34 e P40 / Influence of peptide P34 in gene expression in listeria spp. and study of cytotoxicity of peptídes P34 and P40

Neste estudo foram realizados inicialmente, experimentos para avaliar a ação sinérgica do peptídeo antimicrobiano P34 com sobrenadantes de culturas de algumas bactérias lácticas selecionadas e isoladas de queijo Minas Frescal. Foi investigada a influência deste peptídeo na expressão de genes em L. monocytogenes e L. seeligeri, sua citotoxicidade em diferentes células eucarióticas e toxicidade “in vivo”. Também foram realizados alguns testes para avaliar a citotoxicidade do peptídeo antimicrobiano P40. A adição do peptídeo P34 no queijo provocou uma diminuição de até 3 ciclos logarítmicos na contagem de células viáveis de L. monocytogenes inoculada artificialmente. Um aumento significativo na expressão dos genes dltA, Imo 1695 e mptA de L. monocytogenes foi observado após 96 h com a presença do peptídeo P34 no queijo. A influência do peptídeo P34 na expressão de genes associados aos componentes do envelope celular de L. monocytogenes e L. seeligeri, promoveu um aumento não significativo nos níveis de transcrição de genes dltA, Imo1695 e mptA observados em L. monocytogenes após inoculação em placas e incubação por 24 h a 37°C ou 240 h a 4°C. Em L. seeligeri uma diminuição significativa na expressão do gene dltA foi observada. Os genes Imo1695 e mptA demonstraram uma diminuição significativa de sua expressão (2000 e 31872 vezes, respectivamente) na presença do peptídeo P34 e incubação por 24 h a 37°C. A inoculação da placa com o peptídeo P34 e incubação por 240 h a 4ºC não promoveu diminuição significativa da expressão do gene mptA. A citotoxicidade dos peptídeos P34 e P40 foi avaliada em células VERO, tratadas com diferentes concentrações (0,02 - 2,5 μg ml-1). Nos ensaios de MTT, NRU e LDH as EC50 para o peptídeo P34 foram 0.60, 1.25, 0.65 μg ml-1 e do peptídeo P40 foram 0,30, 0,51 e 0,57 μg ml-1, respectivamente. A atividade hemolítica em eritrócitos humanos foi de (5,8%) e (19%), respectivamente. Os efeitos sobre a viabilidade, motilidade e exocitose acrossomal de espermatozóides humanos também foram avaliadas para o peptídeo P34. Não houve reações de hipersensibilidade ou aumento significativo de títulos de anticorpos durante os experimentos imunogenicidade ou morte dos animais durante experimentos de toxicidade aguda ou subcrônica. A DL50 foi superior a 332,3 ± 0,76 mg/kg. Não foram observadas alterações significativas nos parâmetros bioquímicos séricos nos animais tratados com o peptídeo P34. Não foram detectados sinais de possível toxicidade nos animais do grupo tratado com 0,825 mg/ kg/dia do peptídeo P34. Neste grupo apenas alterações histológicas no baço com a presença de megacariócitos foram observadas. A partir destes resultados evidencia-se o potencial do peptídeo P34 para ser utilizado como bioconservante em alimentos. / In this study initial experiments were performed to evaluate synergistic action of the antimicrobial peptide P34 and culture supernatants of some selected lactic acid bacteria isolated from Minas Frescal cheese. The influence of this peptide in the expression of genes in L. monocytogenes and L. seeligeri, their cytotoxicity in differents eukaryotic cells and “in vivo” toxicity was investigated. Also, some tests were carried out o evaluate the cytotoxicity of the antimicrobial peptide P40. The peptide P34 caused a decrease of up to 3 log cycles in viable counts of L. monocytogenes artificially inoculated in cheese. A significant increase in expression of genes dltA, Imo1695 mptA of L. monocytogenes was observed after 96 h incubation of the peptide P34 in cheese. The influence of peptide P34 on the expression of genes associated to components of cell envelope of L. monocytogenes and L. seeligeri, promoted a non significant increase in the levels of transcription of genes dltA, Imo1695 and mptA were observed after incubation of L. monocytogenes for 24 hs at 37°C and 240 hs at 4°C in plates. In L. seeligeri a significant decrease was observed in gene expression dltA. The gene Imo1695 showed a significant decrease in its expression (2000-fold) after inoculation with the peptide P34. A significant decrease of expression was also observed for the gene mptA (31872 - times) after inoculation with the peptide P34 and incubation for 24 hours at 37°C. The inoculation of the plate with the P34 peptide and incubated for 240 hrs at 4°C, showed a non-significant decrease of gene expression. The cytotoxicity of the peptide P34 and P40 was assessed in VERO cells treated with different concentrations (0.02 - 2.5 μg ml- 1). In MTT, NRU and LDH assays the EC50 to the peptide P34 were 0.60, 1.25, 0.65 μg ml-1 and the peptide P40 were 0.30, 0.51 and 0.57 μg ml-1, respectively. The hemolytical activity on human erythrocytes was of (5.8%) and (19%), respectively. The effects on viability, motility and acrosomal exocytosis of humam sperm were also evaluated for peptideP34. There were no hypersensitivity reactions or significant increase in antibody titer during the immunogenicity experiment or death of animals during the acute or subchronic toxicity tests. The LD50 was more the 332.3 ± 0.76 mg/kg. No significant changes in the serum biochemical parameters were observed in the animals treated with the peptide P34. Signs of possible toxicity were no detected in animals in the group treated with 0.825 mg/kg day of peptide P34. In this group only histological changes in the spleen with the presence of megakaryocytes were observed. From these results show the potential o peptide P34 to be used in future as biopreservative in foods.

Interação entre um peptídeo antimicrobiano e vesículas de fosfolipídeos / Interaction between an antimicrobial peptide and phosfolipids membranes.

Neste trabalho, estudamos a interação de um peptídeo antimicrobiano com membranas modelo, por meio de dicroísmo circular (CD), fluorescência e microscopia óptica. Tal peptídeo, chamado de híbrido, foi sintetizado como uma mistura das regiões mais ativas de dois outros peptídeos antimicrobianos, chamados de pediocina A e plantaricina 149. Esse peptídeo híbrido possui carga de +8, em pH fisiológico, e as membranas estudadas foram compostas por uma mistura de fosfolipídeos zwiteriônicos (cabeça polar de fosfatidilcolina, PC) e aniônicos (cabeça polar de fosfatidilglicerol, PG), em diferentes razões molares. Os resultados de CD evidenciaram que este peptídeo se apresenta de forma desordenada em solução aquosa, porém adota uma conformação helicoidal na presença de grandes vesículas unilamelares carregadas negativamente (LUVs). A quantidade de componente helicoidal é dependente da quantidade de lipídeo negativo presente na bicamada lipídica. A fluorescência do triptofano revelou um deslocamento para o azul muito significativo, chegando a 20 nm para membranas compostas por 100 mol% de PG. Os dois resultados (CD e fluorescência) indicam que a região dos aminoácidos que contém o triptofano deve estar interagindo muito fortemente com a região hidrofóbica da membrana, numa conformação tipo-helicoidal. Experimentos de vazamento de carboxifluoresceína encapsulada em LUVs, por espectroscopia de fluorescência, demonstraram a ação lítica do peptídeo induzindo a formação de poros nas membranas, independentemente da composição das LUVs. Entretanto, a razão molar peptídeo:lipídeo necessária para induzir vazamento da sonda foi menor para membranas lipídicas compostas por bicamadas contendo altas quantidades de PG. Tal fato coloca em evidência o papel fundamental da interação eletrostática entre os peptídeos carregados positivamente com as membranas carregadas negativamente para o processo de ligação e mecanismo de ação deste peptídeo. Para estudar mais detalhadamente o mecanismo de ação, realizamos experimentos de microscopia óptica em vesículas unilamelares gigantes. Concluímos que o peptídeo provoca total desestabilização das vesículas unilamelares gigantes, com formação de poros, seguidos de ruptura da bicamada lipídica e sua transformação em pequenos e mal definidos complexos de peptídeos e fosfolipídeos. / In this work, we investigated the interaction between an antibacterial peptide with model membranes, by means of circular dichroism (CD), fluorescence and optical microscopy. Such a peptide was synthesized from the most active regions of two others antimicrobial peptides, namely pediocin A and plantaricin 149. The hybrid peptide has a net charge of ~ +8, at physiological pH, and the studied model membranes were composed of a mixture of zwitterionic phospholipids (phosphatidylcholine polar head) and anionic phospholipids (phosphatidylglycerol polar head), at differente molar ratio. The CD results evidenced that the peptide was essentially structureless in aqueous solution, but acquired an helical conformation in the presence of charged large unillamellar vesicles LUVs. The helical content is dependent on the negative charge amount on membrane surface. The tryptophan fluorescence revealed a significant blue shift of the maximium emission wavelength, up to 20 nm for the membranes composed of 100 mol% of PG in respect to the peptide fluorescence in the aqueous solution. This indicates that part of the aminoacid residues, that contains the tryptophan, must be buried into the hydrophobic medium of the lipid membrane. Leakage experiments using fluorescence spectroscopy of carboxyfluorescein encapsulated in LUVs demonstrated the lytic action of the peptide, inducing the pore formation in the membrane, regardless of lipid membrane composition. However, it should be stressed that the peptide:lipid molar ratio necessary to induce probe leakage was smaller for lipid membranes made up of large PG amounts. Such evidence points out the key role of the electrostatic interaction between a positively charged peptide and the negatively charged membrane, mediated by hydrophobic contribution. To gain further insight into the lytic mechanism of the peptide, we performed single vesicle experiments using giant unilamellar vesicles under optical microscopy observations. We conclude that the peptide provokes a total membrane desestabilization, with pore formation, followed by a membrane disruption and its transformation into smaller and not well defined complexes of phospholipids and peptides.

Desenvolvimento de novos peptídeos antimicrobianos a partir de proteínas dos venenos das serpentes peruana Bothrops pictus e Bothriopsis oligolepis / Development of new antimicrobial peptides based on the structures of proteins found in the venoms of the Peruvian snakes B. pictus e B. oligolepis

A resistência aos antibióticos adquirida por micro-organismos patogênicos é um problema de saúde mundial e, por isso, o desenvolvimento de novos agentes antimicrobianos vem sendo amplamente estimulado. Sabendo que muitos peptídeos bioativos correspondem a fragmentos peptídicos de proteínas e/ou seus análogos, este trabalho teve o objetivo de desenvolver novos peptídeos antimicrobianos (AMPs) a partir das sequências aminoacídicas e das estruturas 3D de proteínas possivelmente envolvidas na atividade antimicrobiana de venenos de serpentes pouco estudados. As etapas iniciais seguidas foram: a) escolher uma fosfolipase A2 (PLA2) de veneno de serpente peruana do gênero Bothrops da família Viperidae com sequência de aminoácidos conhecida e modelar por homologia a sua estrutura 3D; b) verificar atividade antimicrobiana em venenos de serpentes peruanas dos gêneros Bothrops e Bothriopsis da família Viperidae, selecionar um veneno ativo, fracioná-lo para isolar proteínas provavelmente envolvidas nessa atividade, tripsinizar as proteínas isoladas, sequenciar os fragmentos trípticos para identificá-las, localizar esses fragmentos em sequências aminoacídicas de proteínas com estruturas 3D disponíveis correlatas às proteínas isoladas/identificadas em classe, função e fonte natural. Em seguida, foram escolhidos fragmentos peptídicos da PLA2 (item a) e das proteínas isoladas do veneno ativo (item b) e/ou desenhados análogos que apresentassem características exibidas por AMPs conhecidos. Os peptídeos desenhados foram sintetizados, purificados, caracterizados e testados em suas atividades antimicrobianas. Os modelos estruturais 3D da PLA2 de Bothrops pictus e quatro peptídeos (PLA2-1 a -4) amidados derivados dela foram obtidos, sendo o PLA2-1 ativo frente a Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Candida krusei e Candida parapsilosis (MICs de 6,25-200 µmol.mL-1). Dos três venenos de serpentes peruanas testados, Bothrops taeniatta, Bothrops barnetti e Bothriopsis oligolepis, os dois últimos inibiram o crescimento de S. aureus (MICs 0,78-50 µmol.mL-1), mas apenas B. oligolepis demonstrou espectro de ação amplo. O seu fracionamento sequencial, acompanhado de ensaios de inibição do crescimento de S. aureus, gerou frações ativas relativamente homogêneas que, tripsinizadas e os fragmentos trípticos sequenciados, continham metalo-peptidases do tipo III, serino-peptidase ou lectinas do tipo C. A verificação de atividade enzimática e de coagulação sanguínea nessas frações confirmaram as naturezas das proteínas isoladas. Dos três peptídeos amidados (Bo-Ser1, Bo-Met1 e Bo-Lec1) desenhados a partir de suas estruturas, um deles foi ativo frente às leveduras C. albicans, C. krusei e C. parapsilosis (Bo-Met1; MIC de 6,25 - 200 µmol.mL-1). Pela primeira vez, foi demonstrado que: a) os venenos das serpentes peruanas B. barnetti e B. oligolepis apresentam ação antimicrobiana, sendo o último de espectro amplo; b) que as proteínas acima citadas, que incluem uma serino-peptidase, estão envolvidas com essa propriedade do veneno de B. oligolepis; c) que as sequências aminoacídicas e modelo 3D de uma PLA2 ácida e de proteínas presentes nos venenos das serpentes peruanas B. pictus e Bothriopsis oligolepis podem funcionar como fontes naturais para o desenvolvimento de novos AMPs de ação potente em micro-organismos de interesse clínico e científico. / Resistance to antibiotics obtained by pathogenic microorganisms is a global health problem, so the search for new antimicrobial agents has been encouraged. Knowing that many protein fragments and analogues are bioactive peptides, the aim of this work was to develop new antimicrobial peptides (AMPs) based on the amino acid sequences and 3D structures of proteins apparently involved in the antimicrobial activity of snake venoms very little or not studied so far. The first steps taken were: a) selection of a phospholipase A2 (PLA2) present in the venom from a Peruvian Bothrops sp. belonging to the family Viperidae, whose amino acid sequence was known, to model by homology its 3D structure; b) detection of antimicrobial activity in venoms from other Peruvian Viperidae Bothrops and Bothriopsis snakes, selection of an active venom, fractionation of it for isolation of proteins possibly involved in the antimicrobial activity, trypsinization of the isolated proteins, sequencing of the tryptic fragments for protein identification, location of such fragments in the amino acid sequences and 3D structures of proteins directly related in class, function and natural source to the isolated proteins. Then, peptide fragments from the chosen PLA2 (item a) and from the isolated proteins (item b) that presented structural features found in the known AMPs were selected and/or their analogues were designed. Finally, synthesis, purification and characterization of the peptides with AMP potential, (viii) verification on whether or not they display antimicrobial activity. The 3D-structure models of Bothrops pictus PLA2 and four amidated peptides (PLA2-1 to -4) derived from it were obtained, being PLA2-1 active against Gram negative bacteria Escherichia coli and Pseudomonas aeruginosa as well as the yeasts Candida albicans, Candida krusei and Candida parapsilosis (MICs de 6.25-200 µmol.mL-1). Among the three Peruvian snake venoms tested Bothrops taeniatta, Bothrops barnetti and Bothriopsis oligolepis, the last two inhibited the growth of S. aureus (MICs 0.78-50 µmol.mL-1) and B. oligolepis presented a wide spectrum of bacterial action. Sequential fractionation followed by S. aureus growth inhibition assays of the main fractions led to active relatively homogeneous ones. Their trypsinization and sequencing of the tryptic fragments indicated that they contained metalloproteinases type III, serine-proteinase or lectins type CTL. Enzymatic activity and blood coagulation assays confirmed the nature of the isolated proteins. From the three amidated peptides (Bo-Ser1, Bo-Met1 e Bo-Lec1) derived from them, Bo-Met1 showed to be active against C. albicans, C. krusei e C. parapsilosis (MIC 6,25 - 200 µmol.mL-1). In summary, for the first time, it was demonstrated that: a) the venoms of the Peruvian snakes B. barnetti and B. oligolepis display antimicrobial activity, being the last of wide spectrum of action, b) the proteins isolated from B. oligolepis snake venom, including a serine-peptidase, are involved in the antimicrobial activity of the B. oligolepis snake venom, c) the amino acid sequences and 3D structures of acidic PLA2 and of other proteins found in the venoms of the Peruvian B. pictus e Bothriopsis oligolepis snakes can be used as safe and natural sources for the development of new AMPs potent against microorganisms of clinical and scientific interest.

Expressão da dermicidina e correlações clínico-patológicas em melanomas malignos / Dermcidin expression and clinicopathological correlations in malignant melanoma

O melanoma cutâneo é a neoplasia de pele de maior mortalidade e grande imprevisibilidade na sua evolução. Na doença disseminada, as opções terapêuticas são pouco eficazes. A pesquisa de novos marcadores tumorais permite a melhor compreensão da patogênese do melanoma e possibilita a descoberta de alvos moleculares. A proteína Dermicidina (DCD) foi identificada entre os 9 genes de uma assinatura gênica de predição de diagnóstico clínico do melanoma humano, porém vários autores divergem sobre o papel desta na doença e os mecanismos moleculares pelos quais a DCD atua nos tumores permanecem incertos. O presente estudo teve como objetivo investigar o papel da DCD na tumorigênese do melanoma maligno e correlacionar sua expressão com dados clínicos, demográficos e patológicos dos pacientes. Através da técnica de imuno-histoquímica em lâminas de TMAs (tissue-microarray), o padrão de expressão de DCD foi analisado em tecido tumoral de duas coortes de pacientes, a primeira com 53 casos tratados no Hospital A.C. Camargo, predominantemente caucasianos, e a segunda com 48 casos, todos asiáticos, obtidos comercialmente da empresa IMGENEX. A análise in situ da Dermicidina mostrou que a proteína está expressa de forma heterogênea nas células tumorais, e pode ocorrer tanto em tumores amelanocíticos como melanocíticos. Em melanomas primários, a expressão de DCD foi mais frequente em tumores localizados nas regiões do tronco e membros superiores, já nas metástases, a proteína foi detectada predominantemente em células em transito nos linfonodos (69,23% dos casos). Analisando os resultados dos 101 pacientes das duas coortes em conjunto, pelo método de Kaplan-Meier, foi confirmado que nos indivíduos com tumores DCD-negativo, a taxa de sobrevida foi de 65,54% em 60 meses, e de 62,86% em 130 meses. Já indivíduos com tumor DCD-positivo tiveram sobrevida de 43,33% em 5 anos, e 28,12% em 130 meses, sendo a diferença significante entre os grupos (p=0,0229). A taxa de óbito dos pacientes com tumor DCD-positivo foi mais elevada, 56%, quando comparada à taxa dos indivíduos com tumor DCDnegativo, 33,33% (p=0,0281). Também foi encontrada uma tendência de tumores expressando DCD se relacionarem a pacientes com idade superior a 50 anos (p=0,1057). Em uma consulta de 4 estudos diferentes que reuniram os dados de sequenciamento de DNA de tumores de 515 pacientes, observamos que o gene DCD, em melanomas, não se encontra predominantemente amplificado, mas sim mutado. A substituição E43K foi a alteração mais frequente, correspondendo a 70% dos casos com mutação no gene. Ao relacionarmos os casos disponíveis de mutação em DCD com os genes BRAF, NRAS, MITF, CDKN2A e ERBB4, encontrou-se uma associação com a mutação BRAF V600E nos casos em que ocorria a mutação DCD E43K. Por ter alta frequência em melanomas (variando entre 45 e 54%), e ser um indicador de pior prognóstico para a neoplasia, a expressão de DCD pode ser considerada um potencial biomarcador / Cutaneous melanoma is the skin neoplasia with the highest mortality rate and great unpredictability in its evolution. In disseminated disease, the treatment options are little effective. The research for new tumor markers allows a better understanding of the pathogenesis of melanoma and enables the discovery of molecular targets. The Dermcidin protein (DCD) was identified among the nine genes of a gene signature predicting clinical diagnosis of human melanoma, although many authors differ on its role in the disease and the molecular mechanisms by which DCD acts in tumors remain unclear. The present study aimed to investigate the role of DCD in the tumorigenesis of malignant melanoma and to correlate its expression with clinical, demographic and pathologic data of patients. Using the technique of immunohistochemistry in TMA slides (tissue microarray), the expression pattern of DCD was analyzed in tumoral tissue of two cohorts of patients, the first with 53 cases treated in hospital AC Camargo, predominantly caucasians, and the second with 48 cases, all Asians, commercially obtained from IMGENEX company. The in situ analysis of Dermcidin showed that the protein is expressed in a heterogeneous manner in tumor cells, and can occur in non-melanocytic as well as in melanocytic tumors. In the primary melanoma, DCD expression was more seen in tumors located in the regions of torso and upper limbs. In the metastases, the protein was found predominantly in cells in transit in the lymph nodes (69.23% of cases). Analyzing the 101 patients of the two cohorts together, by the Kaplan-Meier method, it was confirmed that patients with DCD-negative, the survival rate was 65.54% in 60 months, and 62.86% in 130 months, while the group of patients with DCD-positive tumor had 43.33% in 5 years, and 22.12% in 130 months, knowing that a difference between the groups was significative (p=0.0229). The death rate of patients with DCD-positive tumor was higher, 56%, when compared with the death rate of individuals with DCD-negative tumor, 33.33% (p=0.0281). It was also found a trend of tumors expressing DCD related to patients older than 50 years (p=0.1057). In a search of 4 different studies grouping the DNA sequencing tumor of 515 patients we observed that the DCD gene, in melanoma, is not predominantly amplified, but mutated. The E43K substitution was the most frequent alteration, corresponding to 70% of cases of gene mutation. When comparing the available cases of mutations in DCD with the genes BRAF, NRAS, MITF, CDKN2A, ERBB4, we found an association with the BRAF V600E mutation in cases where occurred the DCD E43K. By having high frequency in melanomas (ranging between 45 and 54%) and being an indicator of poor prognosis for the neoplasia, DCD expression can be considered as a potential biomarker

Inhibition de souches bactériennes par de nouveaux composés photosensibles conjugués à la Polymyxine B / Bacterial inhibition through innovative photosensitizers conjugated to polymyxin B

L’émergence de nouvelles souches bactériennes résistantes a signé la fin de l’« âge d’or » des antibiotiques. L’organisation mondiale de la santé a reconnu l’imminence des problèmes associés à ces nouvelles bactéries, qui engendrent de nouveau une mortalité en augmentation. Afin de palier à ce problème, de nouvelles alternatives aux antibiotiques sont envisagées par les chercheurs à travers le monde. La thérapie photodynamique antimicrobienne est l’une de ces alternatives. Elle a su se démarquer grâce à son incapacité à induire la résistance bactérienne. Alors que les résultats furent initialement très prometteurs contre les bactéries Gram positif, une moins bonne sensibilité fut rapidement observée de la part des bactéries Gram négatif. Afin d’augmenter le potentiel antibactérien de cette technique, diverses stratégies furent élaborées comme l’utilisation de photosensibilisateurs cationiques, l’ajout d’un agent de ciblage, ou la présence d’un agent de perméabilisation membranaire. L’un de ces agents est la polymyxine B, un peptide antimicrobien qui arbore une très bonne affinité avec les bactéries Gram négatif. Dans cette étude, nous avons voulu associer chimiquement différents photosensibilisateurs avec des dérivés de polymyxines B de façon covalente, par l’intermédiaire d’un bras « spacer » ou d’une plateforme. L’association de ces deux familles de molécules a permis d’élaborer de nouveaux composés qui ont démontré une activité photobactéricide accrue contre un large spectre de bactérie. New approaches have already been developed in order to overcome this problem. Photodynamic antimicrobial chemotherapy (PACT) could provide an alternative to fight infectious bacteria. Interesting results have been obtained against Gram-positive bacteria, but it also appeared that Gram-negative strains were less sensitive to PACT. Enhanced efficacy against Gram-negative bacteria had been previously obtained following three differents strategies, which are respectively the use of cationic photosensitizers, photosensitizers bound to antimicrobial peptides, or a membrane disrupting agent. Polymyxin B is an antimicrobial peptide, known as the “last-line” treatment against Gram-negative resistant strains, which has already been used as a disrupting agent in order to improve PACT. In addition of this enhancement, this peptide is known for its strong interaction for Gram-negative bacteria. Thus, in this work, we designed differents coumpounds, consisting of a photosensitizer covalently attached to derivatives of polymyxin B, through a spacer or a chemical platform. These combinations have led to the creation of novel compounds which have shown highly photobactericidal activities against a wide spectrum of bacteria. Moreover, these compounds present enhanced affinity for bacteria, which should significantly reduce side effects on mammalian cells. This study confirmed the importance of using antimicrobial in order to target bacterial strains. Thus, such results may allow the creation of novels PACT-based dermatological treatments efficient against a wide spectrum of bacterial strains.

Plusieurs insectes se développant dans des milieux nutritionnellement déficients vivent en symbiose durable avec des bactéries intracellulaires (endosymbiotes) qui complémentent leur alimentation et améliorent leur pouvoir adaptatif. Alors que ces associations ont été largement étudiées sur les plans physiologiques et évolutifs, peu de travaux se sont consacrés à l’étude des mécanismes impliqués dans la tolérance et le contrôle des endosymbiotes par l’hôte. L’objectif de cette thèse est d’étudier, chez les charançons des céréales du genre Sitophilus, les particularités moléculaires et immunitaires du bactériome, un organe que l’insecte développe pour héberger les symbiotes et les isoler de sa réponse immunitaire systémique. Le bactériome du charançon exprime une réponse immunitaire modulée : des études transcriptomiques ont montré que les effecteurs de l’immunité sont peu exprimés dans cet organe, à l’exception d’un gène codant un peptide antimicrobien, la coléoptéricine A. Cette dernière interagit avec les endosymbiotes et participe à leur confinement intracellulaire. Dans une première partie, j’ai montré avec une approche d’interférence à l’ARN que l’expression du gène colA serait contrôlée par un système original qui impliquerait les gènes relish et tollip. Cette régulation « interne » au bactériome semble assurer le maintien des endosymbiotes et l’homéostasie de l’organe. Afin de comprendre comment le bactériome répond à une infection par les bactéries exogènes, j’ai suivi par RT-qPCR l’expression de gènes effecteurs de l’immunité dans le bactériome après injection systémique de bactéries à Gram positif ou négatif. Ceci a mis en évidence une réponse « externe », induite en cas d’infection, et qui aurait un rôle de protection des endosymbiotes contre les bactéries exogènes. Enfin, je me suis consacré à l’étude des changements de régulation accompagnant le passage du stade larvaire au stade adulte, marqué par une symbiose très dynamique. Le nombre d’endosymbiotes augmente fortement pendant les premiers jours de vie imaginale, puis diminue jusqu’à leur élimination complète par recyclage autophagique. Une analyse RNAseq a permis d’identifier les voies de signalisation dont l’activité accompagne cette dynamique. Une approche de RT-qPCR a également montré que l’immunité du bactériome est maintenue à un faible niveau d’activation pendant tout le processus de recyclage. Ce travail montre qu’au cours de leur évolution, les insectes ont sélectionné plusieurs stratégies pour assurer le maintien et l’ajustement de leur charge endosymbiotique en fonction de leurs besoins physiologiques : une signalisation immunitaire assurerait le confinement intracellulaire des endosymbiotes, et un ensemble de processus cellulaires incluant l’apoptose et l’autophagie semble être en associé aux voies métaboliques pour assurer le contrôle de la dynamique bactérienne et garantir le compromis bénéfice/coût de la symbiose. / Many insect species living on nutritionally unbalanced media depend on intracellular mutualistic bacteria, called obligatory endosymbionts, for their development and reproduction. Endosymbionts are housed in specialized host cells called bacteriocytes, that group together to form the bacteriome organ. Although such associations have been widely investigated on a physiological and evolutionary point of view, little is known about the mechanisms involved in the tolerance and the control of endosymbionts by the host. This work aims at deciphering the molecular and immune specificities of the bacteriome using the model system Sitophilus oryzae, the cereal weevil, and its obligate endosymbiont Sodalis pierantonius. The weevil bacteriome expresses a modulated immune response: transcriptomic studies showed that immune effector genes were lowly expressed despite the massive bacterial presence, with the exception of colA, a gene encoding for Coleoptericin A, an antimicrobial peptide. Coleoptericin A interacts with endosymbionts and participates in their intracellular seclusion. In a first chapter, I used RNA interference to demonstrate that colA gene expression may be controlled by an original system involving the genes relish and tollip. This “internal” regulation for endosymbiont control seems to maintain bacteriome homeostasis. In a second chapter, in order to understand how the bacteriome responds to an infection by exogenous bacteria, I followed up by RT-qPCR the expression of immune effector genes in the bacteriome after injection of Gram positive and Gram negative bacteria. This highlighted an “external” immune response, inducible upon infections, which may enable endosymbiont protection against exogenous intruders. In a third and last chapter, I focused on the regulation changes that accompany the switch from the larval stage to the imaginal stage, the latter being characterized by a very dynamic symbiosis. Endosymbiont load drastically increases during the first days of imaginal life, then rapidly decreases until complete elimination of the bacteria by autophagic recycling. RNAseq analysis allowed the identification of signaling pathways linked to this dynamic. A complementary RT-qPCR approach also showed that bacteriome immunity was laid low during the whole recycling process. This work shows that several strategies have been selected during host-symbiont coevolution to ensure the maintenance of the endosymbionts and the adjustment of their population depending on the insects physiological needs: immunity allows the intracellular seclusion in the bacteriocytes, and cell processes including autophagy and apoptosis are associated to metabolic pathways to control the endosymbiotic dynamics and secure the cost and benefit trade-off of symbiosis.

Estudo da relação estrutura-atividades e de propriedades do Hb40-61a, uma hemocidina sintética / An investigation of the structure-activity relationship and the properties of Hb40-61a, a synthetic hemocidin

A hemoglobina (Hb) é uma fonte reconhecida de peptídeos com funções biológicas diversas. O fragmento 33-61 da cadeia α da Hb, isolado do trato gastrointestinal do carrapato Boophilus microplus, foi o primeiro a ser descrito com ação antimicrobiana. O seu análogo sintético amidado, Hb33-61a, mostrou-se ativo contra bactérias Gram-positivas e fungos [Fogaça et al. (1999) J. Biol. Chem. 274, 25330-4]. O estudo de análogos do Hb33-61 nas formas amidada e com carboxila livre revelou que a amidação provoca aumento significativo da atividade frente a Candida albicans. Por apresentar propriedades biológicas e estruturais idênticas às do Hb33-61a, o Hb40-61a pareceu ser a sua porção mÌnima ativa [Sforça et al. (2005) Biochemistry 44, 6440- 51; Machado et al. (2007) Biopolymers 88, 413-26]. Para comprovar tal sugestão, no presente trabalho, sintetizamos, purificamos e caracterizamos novos an·logos do Hb33-61a, bem como os avaliamos quanto às suas atividades frente a C. albicans e Micrococcus luteus. Os resultados confirmaram a sugestão apenas para a ação antifúngica. O análogo Hb40-61a também se mostrou ativo frente a C. albicans resistente a fluconazol. A sua atividade antifúngica se mostrou fortemente dependente da força iônica do meio. A sua baixa atividade hemolítica foi confirmada mesmo em meio de baixa força iônica. O peptídeo Hb40-61a não apresentou sinergismo com o fluconazol frente a C. albicans. A cinética de morte celular mostrou que ele mata a levedura de forma rápida. Portanto, esta hemocidina sintética pode apresentar valor comercial se a via de administração for tópica ou se o seu uso envolver meios de baixa força iônica. Além disso, ela é um modelo valioso para o estudo do mecanismo de ação de peptídeos antimicrobianos com características estruturais similares e pode servir de base para o desenho de novos agentes antibiôticos. / It is well known that hemoglobin (Hb) is a source of biologically active peptides. The fragment 33-61 of bovine hemoglobin α-chain, isolated from the gut contents of the tick Boophilus microplus, was the first identified with antimicrobial activity . Its amidated analogue, Hb33-61a, showed to be active against Gram-positive bacteria and fungi strains [FogaÁa et al. (1999) J. Biol. Chem. 274, 25330-4]. The study of a series of carboxyl-free and amidated synthetic analogues of Hb33-61 revealed that C-terminus amidation enhances the activity against Candida albicans. Since Hb33-61a and Hb40-61a presented identical biological and structural properties, it seemed that Hb40-61a was Hb33-61a minimal active motif [SforÁa et al. (2005) Biochemistry 44, 6440- To test this suggestion, in the present study 51; Machado et al. (2007) Biopolymers 88, 413-26]. we synthesized, purified and characterized Hb40-61a analogues and assayed them against C. albicans and Micrococcus luteus. The results confirmed the suggestion only for the antifungal activity. When tested against fluconazole-resistant C. albicans, Hb40-61a was also active. Its antifungal activity showed to be dependent on the ionic strength of the medium. Its low hemolytic activity was confirmed even under low ionic strength conditions. Hb40-61a had no synergic effect with fluconazole on C. albicans. In vitro time-kill assays demonstrated that Hb40-61a kills the yeast rapidly. Therefore, this synthetic hemocidin may be of commercial interest for topical application or other uses involving low ionic strength medium. Moreover, it can serve as a template for the study of the mechanism of action of structurally related antimicrobial peptides or for the design of novel antibiotic drugs.

Tritrpticina (TRP3) é um peptídeo antimicrobiano com 13 resíduos de amino ácidos com três Ws sequenciais. Com o objetivo de contribuir para a compreensão de seu mecanismo de ação, realizaram-se estudos funcionais e conformacionais da TRP3 e de dois análogos onde um (WLW) ou dois (LWL) W foram substituídos por L. Os peptídeos foram igualmente ativos contra bactérias Gram positivas e negativas. Sua atividade hemolítica requereu concentrações maiores, diminuindo na ordem TRP3>WLW>LWL. Os peptídeos permeabilizaram membranas modelo de E. coli ou contendo fosfolipídios carregados negativamente. Espectros de CD sugeriram que os peptídeos adquirem diferentes conformações ao se ligarem a bicamadas e micelas. Estudos de fluorescência mostraram que a ligação a membranas decresce na ordem: TRP3>WLW>LWL e que os peptídeos se localizam próximos à interface membrana-água. Espectros de RPE de marcadores de spin lipídicos indicaram que a ligação dos peptídeos altera a organização dos lipídios, aumentando o empacotamento molecular / Tritrpticin (TRP3) is a 13-residue antimicrobial peptide that contains three sequential Ws. With the aim of contributing to the understanding of its mechanism of action, functional and conformational studies were performed with TRP3 and two of its analogues where one (WLW) or two (LWL) of the W were replaced by L. The peptides were equally active against both Gram positive and Gram negative bacteria. Higher concentrations were required for hemolytic activity which varied in the order: TRP3>WLW>LWL. The peptides permeabilized membranes model membranes mimicking E. coli\'s lipid composition or containing different negatively charged phospholipids. CD spectra suggested the peptides acquired different conformations upon binding to bilayers or micelles. Fluorescence studies showed that membrane binding decreases in the order: TRP3>WLW>LWL and that the peptides are located close to the water-membrane interface. Dans notre groupe, nous nous intéressons au développement de peptides contenant des acides γ-aminés. Comme d’autres peptides contenant des acides aminés non naturels, ils ont montré leur capacité à posséder des conformations stables et/ou des propriétés biologiques intéressantes. De plus, ces peptides sont généralement résistant à la protéolyse. Dans l’objectif de synthétiser des acides -diaminés sous la forme d’un seul stéréoisomère, nous avons développé une voie de synthèse reposant sur une réaction de Blaise suivie d’une réduction diastéréosélective. En appliquant cette méthode, nous avons synthétisé des acides β,γ-diaminés dérivés de la D-phénylalanine et de l’acide L-glutamique. Le premier a été utilisé pour concevoir des analogues d’un peptide antimicrobien, la gramicidine S. Comparé à la molécule parent, les analogues ont montré une cytotoxicité beaucoup moins importante pour les cellules hôtes tout en conservant une activité antibactérienne intéressante. Cette étude nous a donné de meilleures connaissances pour développer d’autres analogues de la gramicidine S ainsi que d’autres peptides antimicrobiens. Nous avons également effectué de nombreuses optimisations pour synthétiser de façon efficace des acides β,γ-diaminés cycliques à partir de l’acide L-glutamique. Les oligomères incorporant ces acides β,γ-diaminés et des acides α-aminés ont montré un fort potentiel pour l’adoption de conformations stables. Ces études vont être poursuivies. / In our group, we are interested in developing peptides containing β,γ-diamino acids . Along with many other peptides containing unnatural amino acids, they have shown the ability to possess stable conformations and/or interesting biological activities. Moreover, those peptides are usually more resistant to proteolysis. In order to synthesize stereopure γ-amino acids, we have developed a synthetic route using Blaise reaction and subsequent diastereoselective reduction as key reactions. Through applying this method, we have synthesized β,γ-diamino acids derived from D-phenylalanine and L-glutamic acid. The former β,γ-diamino acid was used for designing antimicrobial peptide gramicidin S analogues. Compared with mother molecule, the analogues exerted much less host cell cytotoxicity while remaining interesting antibacterial activity. Meanwhile, it gave us more knowledge for further developing analogues of gramicidin S as well as other antimicrobial peptides. We also paid lots of effort to efficiently synthesize cyclic β,γ-diamino acids starting from L-glutamic acid. Interestingly, when oligomers incorporating this β,γ-diamino acids and α-amino acids, they have shown the potential to adopt stable conformations. The following studies will be continuously investigated.

Les bactériocines RumC, une nouvelle famille de peptides antimicrobiens comme alternative aux antibiotiques conventionnels / RumC peptides, a new family of bacteriocins as viable alternative to conventional antibiotics

Les antibiotiques sont des médicaments qui ont changé notre manière d’aborder les infections bactériennes et sont devenus l’un des symboles de la médecine moderne. Cependant leur utilisation massive a conduit à l'émergence de souches bactériennes multirésistantes. Ce problème est sans aucun doute un des grands défis que la médecine actuelle doit relever. Sachant que les bactéries évoluent à un rythme plus rapide que la production de nouveaux antibiotiques, il est urgent de trouver des approches alternatives. Il a été mis en évidence que ces mêmes bactéries sont capables de sécréter différents peptides antimicrobiens, ou bactériocines. Ces macromolécules présentent une grande diversité structurale et sont très efficaces pour combattre un grand nombre de souches pathogènes de façon spécifique. Les bactériocines ont un immense potentiel dans les domaines agroalimentaire et pharmaceutique. Notre projet s’intéresse aux bactériocines RumCs produites par une souche dérivée de Ruminococcus gnavus, une bactérie anaérobie stricte, membre dominant du microbiote intestinal humain. Le travail présenté dans ce manuscrit concerne la mise au point d’un système d’expression et de maturation hétérologue chez E. coli de la bactériocine RumC1. La caractérisation biochimique du peptide RumC1 montre que les bactériocines RumCs appartiennent à la famille des sactipeptides pour laquelle l’étape de biosynthèse fait intervenir une enzyme radical-SAM. Les sactipeptides présentent dans leurs séquences peptidiques un ou plusieurs ponts thioéther entre une cystéine et le carbone alpha d’un acide aminé partenaire. RumC1 renferme 4 ponts thioéther ce qui lui confère une structure originale en double épingle à cheveux. L’activité biologique de RumC1 montre que ce peptide est efficace contre un large spectre de bactéries à Gram positif incluant des pathogènes résistants tels que S.aureus et E. faecalis. Dans ces études nous n’avons pas noté de toxicité significative de RumC1 sur différentes lignées cellulaires humaine ni observé de phénomène de résistance. Les travaux en cours visent notamment à définir le mode d’action de RumC1 et à évaluer l’activité biologique de RumC1 dans un contexte d’infection in vivo chez la souris. / Antibiotics are drugs that have changed the way we approach bacterial infections and have become one of the symbols of modern medicine. However, their widespread use has led to the emergence of multiresistant bacterial strains. This problem is undoubtedly one of the major challenges facing today's medicine. Knowing that bacteria evolve at a faster rate than the discovery of new antibiotics, it is urgent to find alternative approaches. It has been shown that these same bacteria are capable of secreting antimicrobial peptides, the bacteriocins. These macromolecules have a high structural diversity and are very effective in combating a large number of pathogenic strains in a specific way. Bacteriocins have immense potential in the agro-food and pharmaceutical sectors. Our project focuses on the bacteriocins RumCs produced by a strain derived from Ruminococcus gnavus, a strict anaerobic bacterium of the human intestinal microbiota. The work presented in this manuscript concerns the development of a heterologous expression and maturation system in E. coli of the bacteriocin RumC1. The biochemical characterization of the RumC1 peptide shows that the RumCs bacteriocins belong to the family of sactipeptides for which the biosynthesis step involves a radical-SAM enzyme. The sactipeptides have in their peptide sequences one or more thioether bridges between a cysteine and the alpha carbon of a partner amino acid. RumC1 contains 4 thioether bridges which gives it an original structure in double hairpin. The biological activity of RumC1 shows that this peptide is effective against a broad spectrum of Gram-positive bacteria including resistant pathogens such as S.aureus and E. faecalis. In these studies, we did not note any significant toxicity of RumC1 on different human cell lines nor observed resistance phenomena. Current work aims to define the mode of action of RumC1 and to evaluate the biological activity of RumC1 in an in vivo context of infection in mice.
