New Paracas Architecture Pattern In Lucanas, Southern Highlands Peru / Nuevo patrón arquitectónico paracas en Lucanas, sierra sur del Perú

During our archaeological surveys in the upper reaches of the Palpa valleys, at the western slope of the Andes, we identified and registered a large number of settlements of the Paracas culture, most of them dating to the Middle Paracas(550-350 BC) and Late Paracas (350-200 BC) period. Of special interest are several large sites with stone buildings with circular ground plans, similar to the shape of a flower. The center of these architectural complexes consists of acircular or crescent shaped sunken patio, around which are arranged D-shaped enclosures with small terraces in front of the straight wall.Through our large scale excavations in Cutamalla, one of the largest and best preserved settlements in the area, we were able to document in detail this new type of architecture and to define its formal and constructive features, as well as its cultural affiliation and the activities carried out within the structures.All the data recovered so far indicate that these circular structures have to be considered as a new architectural type of the Late Paracas period. The distribution of this new architectural pattern is limited to a part of the highlands of Lucanas, Ayacucho, and, as far as we know, seems to be unique in the southern highlands of Peru. / Trabajos de prospección arqueológica realizados en la parte alta de los valles de Palpa, en la vertiente occidental de los Andes, nos han permitido identificar y registrar un importante número de asentamientos pertenecientes a la culturaParacas, de manera especial, aquellos pertenecientes a las épocas Paracas Medio (550 a 350 a.C.) y Paracas Tardío (350 a 200 a.C.), entre los que destacan varios sitios bastante grandes que presentan construcciones de piedra que conforman estructuras arquitectónicas en forma de flor. Se trata de construcciones en donde se pueden distinguir patios hundidos de forma de media luna o de forma circular, alrededor de los cuales se disponen recintos en forma de «D» que presentan pequeñas terrazas delante del muro recto.Recientes excavaciones en área realizadas en Cutamalla, uno de los asentamientos más grandes y mejor conservados de la zona, nos han permitido documentar con detalle este tipo de estructuras arquitectónicas, y conocer sus rasgos formales y constructivos, su filiación cultural, así como las actividades que se realizaban en ellas.Todos los datos indican que las estructuras arquitectónicas en forma de flor constituyen un nuevo patrón arquitectónico Paracas Tardío, cuya distribución se limita a una parte de la sierra de Lucanas, Ayacucho, y que, hasta el momento, viene a ser único en toda la sierra sur del Perú.

Estudo experimental das vibrações induzidas pela emissão de vórtices em cilindros flexíveis inclinados em relação à correnteza. / Experimental study on the vortex-induced vibrations of flexible cylinders yawed in relation to the current.

O problema das vibrações induzidas por vórtices em cilindros flexíveis inclinados em relação ao perfil de correnteza incidente é estudado através de uma abordagem experimental. Este tema se mostra de grande importância e aplicação na Engenharia Naval e Oceânica uma vez que são diversas as estruturas que se encaixam nesta descrição, como são o casos dos risers e linhas de amarração. Apesar de extensa, a literatura técnica especializada no assunto é focada em simplificações, tais como os estudos de cilindros rígidos fixos, cilindros rígidos montados em base elástica e cilindros flexíveis verticais ou lançados em catenária livre. A questão da inclinação é abordada em termos do Principio da Independência que diz que as forças e a frequência de emissão de vórtices em um cilindro inclinado são iguais as de um cilindro vertical se considerada apenas a parcela da correnteza normal à direção de inclinação. Estudos anteriores mostraram que este princípio é válido somente em algumas condições específicas e poucos deles apresentam resultados para cilindros flexíveis. Com base nisso, um ensaio em escala reduzida em tanque de reboque foi realizado com um modelo de linha composto por um tubo de silicone preenchido com microesferas de aço e condições de contorno articuladas em suas extremidades, o que conferiu características inerciais e geométricas ideais para a obtenção do fenômeno. Além da configuração vertical (? = 0°), que é o caso de referência, o modelo foi estudado em outras quatro inclinações com a vertical: ? = 10°, 20°, 30° e 45°. Estas inclinações foram ensaiadas para cinco diferentes azimute: ? = 0°, 45°, 90°, 135° e 180°, resultando em vinte e uma configurações de inclinação. Os resultados são analisados em termos da decomposição modal da vibração da linha, ou seja, são apresentadas amplitudes e frequências de oscilação ocorridas em cada um dos modos de vibrar. Além disso são realizadas análises espectrais, gráficos de evolução espaço-temporal, trajetórias de pontos ao longo do modelo flexível, entre outros. Frequências naturais, coeficientes de amortecimento e trações no topo são apresentados, complementarmente, como resultados de caracterização do modelo. Os estudos apontaram para uma validade do PI com algumas ressalvas, principalmente influenciadas pela diferença nas frequências do primeiro e segundo modo que é crescente com o aumento da inclinação. Além disso conclusões importantes sobre a coexistência modal, em regiões de ressonância e/ou de transição da dominância modal, foram obtidas. / The vortex-induced vibration problem in flexible cylinders yawed in relation to the incident current profile is studied through an experimental approach. This topic has great importance and application on Naval and Oceanic Engineering once there are multiple structures that fit into this description, such as risers and mooring lines. Though extensive, technical literature specialized on this topic is focused on simplifications such as the study of fixed rigid cylinders, elastic base rigid cylinders and flexible cylinders assembled vertically or launched in free hanging catenary. The inclination issue is addressed in the terms of the Principle of Independence which states that the forces and frequency shedding in an inclined cylinder are equal to those of a vertical cylinder if only considered the current component normal to the inclination plane. Previous studies showed that this principle is valid only on some specific conditions and few of them showed results to flexible systems. Based on this, a scaled tow tank test was performed with a line model composed of a silicone tube filled with steel microspheres and articulated end conditions, which offered the ideal inertial and geometrical conditions to obtaining the phenomenon. Besides the vertical configuration (? = 0°), which is the reference case, the model was studied in other four inclinations with the vertical: ? = 10°,20°, 30° and 45°. These inclinations were tested for five different azimuth directions: ? = 0°, 45°, 90°, 135° and 180°, resulting in 21 configurations. The results are analyzed on terms of the modal decomposition of the line motions, that is, amplitudes and oscillation frequencies are presented for each one of the vibration modes. Furthermore other analyzes are made, as spectral graphs, spatio-temporal evolution graphs, line points trajectories, among others. Natural frequencies, damping coefficients and tensions at the top are presented, complementarly, as results of the model characterization. The studies pointed to a validity of the IP with some reservations, mainly influenced by the difference in of the first and second mode frequencies that is increasing with the yaw increase. In addition, important conclusions about modal coexistence in regions of resonance and / or transition of modal dominance were obtained.

Entre leões e tigres, entre chacais e raposos: aproximações entre poder e saber em fabulários / Between lions and tigers, between jackals and foxes: relationships between power and knowledge in fable collections

A presente dissertação se propôs a analisar quatro fabulários, tendo como fio condutor dessa escolha o Kalla e Dimna, fabulário árabe do século VIII d.C. Partindo desta obra, os estudos se orientaram tanto às suas origens, que remontam à compilação indiana Pañcatantra, datada do século I d.C. e que teve entrada no mundo árabe, bem como aos seus frutos, o Livro do tigre e do raposo e O leão e o chacal Mergulhador, dois fabulários árabes produzidos, respectivamente, nos séculos IX d.C., XI d.C. e XII d.C. e que tiveram, em relação aos anteriores, menor disseminação. Os dois primeiros fabulários tratam de uma série de temas que podem atender às normas mais gerais de decoro na sociedade, sendo que o Pañcatantra chega até mesmo a ser classificado como um tratado sobre a conduta em manuais de literatura sânscrita. Entre esses temas diversos, é notável o espaço privilegiado dado às relações políticas e aos seus trâmites. Os outros dois livros, embora inseridos na mesma tradição, deixam de lado aspectos mais gerais da convivência e passam a se deter apenas nos temas especificamente políticos. No que se refere a esse assunto em comum, é possível perceber uma série de elementos recorrentes nos quatro fabulários, entre eles, o fato de que todos tratam das vicissitudes das relações entre poder e saber, personificada em personagens-tipo que ora se aproximam, ora se afastam, mas que se mantém atrelados a uma estrutura que os molda conforme os resultados que almeja atingir. Além disso, foram estudadas as estruturas que, no caso, se apoiam na maior parte das vezes em narrativas-quadro ou prólogos-moldura a partir dos quais se desenvolvem subnarrativas e outros elementos que nos possibilitaram, ao término do estudo, apontar o que há de particular na universalidade a que se propõem os fabulários. / This dissertation aims to analyze four collections of fables, choosing as the central work from which further analysis will stem the Kalla and Dimna, an Arab collection of fables dating from the VIII century A.D. From this starting point, these studies were furthered by revisiting its origins in the Panchatantra, an Indian compilation of fables from the I century A.D. that left its marks on the Arab world, as well as its developments in the books that directly follow its legacy, the Livro do tigre e do raposo and O leão e o chacal Mergulhador, two arab collections of fables dating, respectively, to the IX century A.D. and the XI and XII century A.D. which had, however, far less dissemination than the last two works here cited. The first two collections develop around a variety of subjects relating to rules regulating manners in a broader sense, to the point that the Panchatantra has been classified as a treatise on manners in Sanskrit literature manuals. Between the diversity of subjects broached, the privileged attention given to political relations and their formalities is readily noticeable. The two other works, though part of the same literary tradition, set aside more general or day-to-day aspects of good manners to focus exclusively on political relations and political conduct. Regarding this shared thematic, a number of recurring elements can be identified, one of many being the fact that all of them deal with the specifics of the relationship stabilished between power and knowledge, personified in character types that alternate between approaching and distancing themselves from one another, but that are always attached to a structure that moulds them according to the results it desires. Beyond that, the narrative structures of the works were studied they usually make use of frame narratives or frame prologues from which further subnarratives are developed along with other elements that made it possible, at the end of this study, to point towards the particularities of the universality that the collections of fables undertake as their purpose.

Estes modelos, chamados de Níveis de Aproximação (NdA), estão organizados em ordem crescente de precisão da análise, partindo de um nível inicial, com maiores simplificações, até um nível final, com uma modelagem mais realista. No NdA I, a estaca semienterrada é tratada como uma coluna engastada na base e livre na ponta, com base na Técnica do Engastamento Virtual. O cálculo dos esforços resistentes é feito com base no método do pilar-padrão com curvatura aproximada, proposto pela norma brasileira. No NdA II, um programa comercial de análise tridimensional de estruturas é utilizado. Neste programa, o solo é representado de maneira discreta através de uma série de molas de comportamento elástico e linear (baseadas no Modelo de Winkler). A NLF é tratada de maneira aproximada através de um coeficiente, aplicado ao momento de inércia bruto, que simula a diminuição da rigidez do concreto. A NLG é tratada internamente pelo programa através do processo iterativo P-delta. No NdA III, é utilizado um programa elaborado neste trabalho que considera as não-linearidades e a interação solo-estrutura através da equação da viga sobre base elástica, cuja solução é dada a partir do Método das Diferenças Finitas. A NLF, neste modelo, foi considerada através de diagramas momento-curvatura. A NLG e a interação solo-estrutura (Modelo de Winkler) são consideradas diretamente na formulação da equação diferencial. Por fim, o NdA IV utiliza um programa comercial de análise não-linear de estruturas de pontes através de elementos finitos. A NLF é tratada pela integração das curvas de tensãodeformação e a NLG é modelada usando momentos P-delta. A consideração da interação solo-estrutura, neste modelo, é feita através de modelos de curvas p-y. Estes quatro modelos foram aplicados a dois exemplos distintos: um considerando apenas um pilar engastado na base e livre no topo (livre da interação solo-estrutura) e outro considerando uma estaca semienterrada em solo arenoso. Os resultados obtidos pelos quatro métodos foram bastante satisfatórios e se mostraram coerentes entre si, apresentando-se dentro do esperado para cada grau de refinamento da análise. / The present work proposes four different analysis models for the design of partly buried piles of circular section subjected to lateral loads with consideration of the physical nonlinearity of the material (PNL) and the geometric nonlinearity of the structure (GNL). These models, called Levels of Approximation (LoA), are organized in increasing order of accuracy, starting from an initial level, with greater simplifications, to a final level, which is more realistic. In LoA I, the partly buried pile is treated as a column fixed at the base and free at the top, based on the Virtual Clamping Technique. Strength computation is based on the standard column method with approximate curvature, proposed by the Brazilian Code. In LoA II, a commercial three-dimensional structure analysis program is used. In this program, the soil is represented discreetly through a series of elastic and linear springs (based on the Winkler Model). The PNL is approximately treated by a factor, applied to the gross moment of inertia, which simulates the decrease of rigidity of the reinforced concrete due to cracking and nonlinear constitutive properties. The GNL is internally handled by the program through the iterative P-delta process. The third proposed LoA is a program developed in this work that considers the non-linearities and the soil-structure interaction through the equation of the beam on elastic foundation. The equation is solved numerically, with the application of the Finite Differences Method. The PNL, in this model, was considered through moment-curvature diagrams computed internally for the section under analysis, which were used to determine the stiffness of the pile as a function of the lateral displacement. The GNL and the soil-structure interaction (also by Winkler Model) are considered directly in the equation. Finally, LoA IV uses a commercial program of nonlinear analysis of bridge structures through finite elements. The PNL is treated by integrating stress-strain curves into the cross-section analyzed and the GNL is modeled using P-delta moments. Consideration of the soil-structure interaction in this model is made through p-y curves models. These four models were applied to two distinct examples: one considering only one column fixed at the base and free at the top (free of soil-structure interaction) and another one considering a partly buried pile in sand. The results obtained by the four methods were very satisfactory and were shown to be coherent with each other, in line with the expected for each degree of refinement of the analysis.

ExpressÃo de uma quitinase de Chromobacterium Violaceum em Pichia Pastoris: purificaÃÃo e caracterizaÃÃo parcial da proteÃna recombinante / Expression of a chitinase from Chromobacterium Violaceum in Pichia pastoris: purification and partial characterization of recombinant protein

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / A utilizaÃÃo de microrganismos como sistemas heterÃlogos de expressÃo de proteÃnas tem se mostrado uma estratÃgia alternativa e/ou complementar aos passos tradicionais utilizados no processo de purificaÃÃo de proteÃnas. Diante deste contexto, quitinases (EC 3.2.1.14), enzimas hidrolÃticas capazes de degradar quitina, tÃm sido expressas em diferentes sistemas heterÃlogos, incluindo bactÃrias e leveduras. Quitina à um polÃmero linear composto de resÃduos de N-acetil-β-D-glucosamina (GlcNAc), sendo um importante constituinte estrutural da carapaÃa de crustÃceos e membrana peritrÃfica de insetos e, ainda, da parede celular de fungos. Este trabalho teve por objetivo expressar uma quitinase, codificada pela ORF cv3316, de Chromobacterium violaceum ATCC 12472 na levedura metilotrÃfica Pichia pastoris, estirpes GS115 e KM71H, alÃm de purificar e caracterizar a proteÃna recombinante (rCHI3316). A estirpe GS115, portando a construÃÃo (pPICZαA-CV3316), foi selecionada para os experimentos posteriores, pois apresentou um maior nÃvel de expressÃo quando comparada à cepa KM71H. Cromatografia de afinidade em coluna de nÃquel imobilizado foi utilizada para purificar a quitinase recombinante (rCHI3316), que foi eluÃda como um Ãnico pico com imidazol 0,04 M. A proteÃna purificada se mostrou homogÃnea quando submetida à eletroforese em gel de poliacrilamida 15% em presenÃa de SDS e -mercaptoetanol (SDS-PAGE). Nessas condiÃÃes, uma Ãnica banda com massa molecular aparente de aproximadamente 87 kDa foi observada. A quitinase rCHI3316 de C. violaceum foi expressa de forma solÃvel utilizando o sistema de expressÃo P. pastoris e, alÃm disso, sua purificaÃÃo foi realizada de forma satisfatÃria. O conteÃdo de estrutura secundÃria foi estimado por espectroscopia de dicroÃsmo circular (CD), com a proteÃna submetida a diferentes temperaturas (10-90 ÂC). Na temperatura de 24 ÂC, o espectro de CD revelou a predominÃncia do conteÃdo de hÃlice alfa (38%), folha beta (26 %) e estrutura randÃmica (37%). Entre as temperaturas de 10-50 ÂC, rCHI3316 exibiu um espectro caracterÃstico de folha beta. A partir de 60 ÂC atà 90 ÂC, rCHI3316 adquiriu uma conformaÃÃo caracterÃstica de hÃlice alfa. A temperatura mÃdia para essa transiÃÃo conformacional foi calculada como sendo 59,6  1,21 ÂC. Experimentos de espectroscopia de fluorescÃncia, com excitaÃÃo a 280 e 290 nm, produziram espectros de emissÃo com comprimentos de onda mÃximos iguais a 339 e 342 nm, respectivamente. Esses valores sÃo caracterÃsticos de resÃduos de triptofano parcialmente expostos ao solvente. Atividade quitinolÃtica contra vÃrios substratos e dependÃncia de pH da atividade enzimÃtica da proteÃna pura foram avaliados. A rCHI3316 exibiu atividade hidrolÃtica sobre os substratos quitina coloidal (1.189,40 U/mgP), 4-nitrofenil N-Nâ-diacetil-β-D-quitobiosÃdeo (30.411,0 U/mgP) e 4-nitrofenil N-Nâ-Nââ-triacetilquitotriose (13.150,0 U/mgP); entretanto, nenhuma atividade enzimÃtica da rCHI3316 foi detectada frente a 4-nitofenil N-acetil-β-D-glucosaminÃdeo. A enzima exibiu atividade quitinolÃtica Ãtima (100%) em pH 5,0; quando quitina coloidal foi utilizada como substrato. Em adiÃÃo, a atividade antifÃngica contra fungos fitopatogÃnicos, Fusarium solani, Fusarium oxysporum, Rhizoctonia solani e Penicillium herquei foi investigada. rCHI3316 nÃo inibiu a germinaÃÃo e o crescimento micelial dos esporos dos fungos testados, na concentraÃÃo de 0,63 mgP.ml-1. Estudos subseqÃentes deverÃo ser realizados na intenÃÃo de descobrir potenciais aplicaÃÃes desta proteÃna como uma ferramenta biolÃgica no controle de outros fungos fitopatogÃnicos bem como insetos considerados pragas. / Microorganisms are a valuable tool for the expression of proteins from a variety of sources, including plants, animals and other microorganisms. Thus, chitinases, a group of glycosil hydrolases capable to hydrolize chitin, have already been expressed and purified from different systems including bacteria and yeast. Chitin, a linear polymer of N-acetyl-β-D-glucosamine (GlcNAc), is an important structural component found in the crustacean shells, in the peritrophic membrane of insect guts as well as in the fungi cell walls. The aim of this work was to express a chitinase (encoded by the ORF CV3316) from Chromobacterium violaceum ATCC 12472, using the methylotrophic yeast Pichia pastoris strains GS115 and KM71H. Furthermore, purification and partial characterization of the recombinant protein were also achieved. The GS115 strain carrying the expression cassette pPICZαA-CV3316 was selected due to its higher expression level as compared to KM71H strain. Immobilized metal ion affinity chromatography was employed to purify the recombinant chitinase which was eluted as a single peak at 0.04 M imidazol. The homogeneity of the purified protein was confirmed as judged by polyacrylamide gel electrophoresis (SDS-PAGE). In these conditions, the recombinant chitinase migrated as a single protein band with an apparent molecular mass of about 87 kDa. Thus, a chitinase from C. violaceum ATCC 12472 was successfully expressed in P. pastoris and the soluble recombinant protein purified. The content of secondary structure was investigated by circular dichroism (CD) spectroscopy. At 24 oC the CD spectrum revealed secondary structure contents of 37% (alpha helix), 26% (beta sheet) and 38% (random coil). The CD spectra obtained in the temperature range 10-50 oC were characteristic of beta sheet. In contrast, the CD spectra generated in the range 60-90 oC were characteristic of alpha helix. The midpoint temperature of this conformational transition was 59.6 1.2 oC as calculated from the CD experimental data. Fluorescence spectroscopy was carried out with excitation at 280 and 290 nm, producing emission spectra in which the wavelengths of maximum emission were 339 and 342 nm, respectively. This behavior is characteristic of tryptophan residues in limited contact with water. Chitinolytic activity against several substrates and the pH dependency of the enzymatic activity of the pure protein were all accessed. The purified enzyme showed hydrolytic activity on the following substrates: colloidal chitin (1,189.4 U.mgP-1), 4-nitrophenyl N-Nâ-diacetyl--D-chitobioside (30,411.0 U.mgP-1) and 4-nitrophenyl β-D-N-Nâ-Nâ-triacetylchitotriose (13,150.0 U.mgP-1); and, respectively. In contrast, no activity was detected using 4-nitrophenyl N-acetyl-β-D-glucosaminide as substrate. The enzyme presented an optimal chitinolytic activity at pH 5.0 using colloidal chitin as a substrate. Additionally, the antifungal activity against the phytopathogenic fungi, Fusarium solani, Fusarium oxysporum, Rhizoctonia solani and Penicillium herquei was investigated. The recombinant chitinase did not inhibit the spore germination and the mycelium growth of the tested fungi, at the 0.63 mgP.ml-1 concentration. Further studies should be carried out in order to discover potential applications of this protein as a biotechnological tool in the control of other phytopathogenic fungi as well as economically important pests.

Circular Economy in Cities: A Strategic Approach Towards a Sustainable Society?

Increasing environmental and social pressure caused by human activity requires action toward a sustainable society. As our population grows and the proportion living in urban areas increases, cities are in a unique position to affect change. This has led to Circular Economy (CE) gaining momentum in municipalities as a tool for their city. Despite this momentum, there is lack of clarity about what CE is, how it can be most effectively implemented, and its relationship to strategic sustainable development (SSD). This research synthesized definitions of CE used in the field and investigated 21 cities worldwide to see how their municipalities have implemented CE practices. Using the Framework for Strategic Sustainable Development (FSSD), the implemented actions were critically examined for their strategic contribution toward a sustainable society. The findings suggest that CE is defined broadly, and interpreted differently by different users. In some cases, this presents challenges for its strategic use. The CE actions examined are concluded to be within the boundaries of a sustainable society, as defined by the FSSD. Their strategic approach is determined to be largely positive, with some exceptions. Depending on its application, CE is concluded to be a useful, albeit insufficient, tool for municipalities working toward SSD.

The expediency of the contemporary guest worker migration policies that curb mobility : the Arab-Gulf countries and the Indian migrants / La convenance des politiques de migrations temporaires comme frein à la mobilité : les États arabes du Golfe et les migrants indiens

Cette thèse soutient que les politiques de migrations temporaires entravent la mobilité de l'individu, à partir du cas des migrants indiens dans la région du golfe persique. La pratique et le droit international définissent la mobilité comme le droit à la « liberté de mouvement » sans dispositions supplémentaires pour faciliter le choix de s’établir, permettant aux Etats de pratiquer ces politiques de convenance. Ce travail interdisciplinaire qui privilégie l’angle sociologique interroge les implications politiques et le droit international. Il montre que le non-respect prolongé des droits des migrants, surtout par les pays d’accueil, produit des conditions et des résultats restrictifs pour tous les acteurs- des indicateurs montrent l’exclusion systématique du migrant dans la société d’accueil. Cinq chapitres présentent empiriquement le « migrant » dans le golfe qui expérimente la vulnérabilité à différentes échelles : entrepreneurs, marchands-patriarches, travailleurs en col blanc, travailleurs en col bleu, et les femmes migrantes. La politique de migration temporaire des six Etats du Conseil de coopération du Golfe (GCC)- Arabie Saoudite, Oman, Émirats Arabes Unis, Qatar, Bahreïn et Koweït- constitue un exemple heuristique. Sans être un cas unique, il montre une exclusion plus sévère en raison de la nature conservatrice de ces monarchies et de la place de l’Islam comme religion d’Etat. Les politiques de migrations circulaires sont populaires aujourd’hui et les Etats en profitent à leur convenance en privilégiant la flexibilité et la non-intégration pour éviter les responsabilités vis-à-vis des migrants. Le migrant temporaire est donc, le travailleur précaire dans le marché mondial du travail. Le statut d’incertitude structurelle est aussi un des principaux éléments qui séparent « l’Indien du golfe » du reste des Indiens non-résidents (NRI). / The thesis highlights the evidence in the Arab-Gulf region with the Indian migrants to argue that the temporary migration policies hinder the individual’s mobility. International practice and law articulate mobility narrowly as merely the right to “freedom of movement” without also provision to facilitate the choice to settle, which allows states to perpetuate such expedient policies. The work is an inter-disciplinary approach, with mainly a sociological lens and interrogates the implications for policy and international law. It shows that mainly the receiving states’ prolonged non-respect of the migrant’s rights using the “temporary” frame produces limiting conditions and outcomes for all the actors- selected indicators show the systematic exclusion of the migrant in the host society. Five chapters empirically present the “guest worker” in the Gulf who experiences vulnerability at different levels: entrepreneur, trader-patriarch, white-collar worker, blue-collar worker and female migrant. The guest-worker policy practice of the six oil-rich Gulf Cooperation Council (GCC) countries- Saudi Arabia, Oman, the UAE, Qatar, Bahrain and Kuwait- is a heuristic example. The GCC region is not a unique case although it might show more severe exclusion, due to the conservative regimes as monarchies and following Islam as the state-religion. Circular migration policies are popular today and states manifest expediency by privileging flexibility and non-integration to evade responsibility for the migrant. The guest-worker is hence, the precarious worker in the global labour market. The structural uncertainty is a factor that mainly also separates the “Gulf-Indian” from the larger non-resident Indian (NRI) population elsewhere.

La spectroscopie de dichroïsme circulaire (CD) est une des techniques fondamentales en biologie structurale qui permet la détermination du contenu en structures secondaires d'une protéine. Le rayonnement synchrotron a considérablement augmenté l’utilité de la méthode, car il permet de travailler avec une gamme spectrale étendue et à meilleure intensité. Le développement de modèles permettant d’établir une relation entre la structure d’une protéine et son spectre CD d’une manière efficace n’a pourtant pas suivi l’évolution technique et l’analyse de spectres CD de protéines entières reste un défi sur le plan théorique. Dans ce contexte, nous avons développé un modèle "minimaliste" pour la spectroscopie CD des protéines, où chaque atome C-alpha de la chaîne principale porte un oscillateur de Lorentz classique, i.e. une charge mobile qui est tenue par un potentiel quadratique. Les oscillateurs sont couplés par un potentiel coulombien et leurs déplacements suivent les tangentes locales respectives de la courbe spatiale décrite par les atomes C-alpha. Le système d'oscillateurs est couplé à une onde électromagnétique plane décrivant la source de lumière et le phénomène d'absorption est modélisé par des forces de friction. Nous montrons que le modèle reproduit correctement le phénomène CD d'une chaîne polypeptidique hélicoïdale et en particulier son signe en fonction de l'orientation de la chaîne. Comme première application, nous présentons l'ajustement du modèle au spectre CD d'un polypeptide composé de 15 résidus qui se plie sous forme d'une hélice alpha. La transférabilité de ces paramètres est ensuite évaluée pour la myoglobine, une protéine de 153 résidus contenant 8 hélices alpha. / Circular dichroism (CD) spectroscopy is one of the fundamental techniques in structural biology that allows us to investigate the secondary structure of proteins. Synchrotron radiation has considerably increased the usefulness of the method because it allows to work with a wider range of spectrum and much greater signal-to-noise ratios. The development of a theoretical model to establish a relationship between the structure of a protein and its CD spectra in an efficient manner proved to be a complex task. The calculation of the CD spectra of large molecules, such as protein, remains a challenge, due to the size and flexibility of the molecules. In this context, we have developed a “minimal” model to explain the CD spectroscopy of proteins, which associates each C-alpha position on the protein backbone with a classical Lorentz oscillator i.e. a mobile charge attaches to a corresponding atom by a quadratic potential. The coupling between charges is through the Coulomb potential and their displacements follow the direction of the respective local tangents to the Calpha space curve. This system is coupled to a planar electromagnetic wave describing the light source and the absorption phenomenon is modeled by frictional forces. We show that the model correctly reproduces the CD phenomenon of a helical polypeptide chain and in particular its sign depending on the orientation of the chain. At first, we have fitted a model to CD spectra of a polypeptide chain of 15 residues folded into alpha helix. The transferability of these parameters is then evaluated with myoglobin, a protein of 153 residues containing eight alpha helices.

Study of circular code motifs in nucleic acid sequences / Étude des motifs de code circulaire dans les séquences d'acides nucléiques

Le travail effectué dans cette thèse présente une nouvelle approche de la théorie du code circulaire dans les gènes qui a été initiée en 1996. Cette approche consiste à analyser les motifs construits à partir de ce code circulaire, ces motifs particuliers sont appelés motifs de code circulaire. Ainsi, nous avons développé des algorithmes de recherche pour localiser les motifs de code circulaire dans les séquences d'acides nucléiques afin de leur trouver une signification bioinformatique. En effet, le code circulaire X identifie dans les gènes est un ensemble de trinucleotides qui a la propriété de retrouver, synchroniser et maintenir la phase de lecture. Nous avons commencé notre analyse avec le centre de décodage du ribosome (ARNr) qui est une région majeure dans le processus de traduction des gènes aux protéines. Puis, nous avons étendu les résultats obtenus avec le ribosome aux ARN de transfert (ARNt) pour étudier les interactions ARNr-ARNt. Enfin, nous avons généralisé la recherche de motifs de code circulaire X dans l'ADN aux chromosomes d'eucaryotes complets. / The work done in this thesis presents a new direction for circular code identified in 1996 by analysing the motifs constructed from circular code. These particular motifs are called circular code motifs. We applied search algorithms to locate circular code motifs in nucleic acid sequences in order to find biological significance. In fact, the circular code X, which was found in gene sequences, is a set of trinucleotides that have the property of reading frame retrieval, synchronization and maintenance. We started our study in the ribosomal decoding centre (rRNA), an important region involved in the process of translating genes into proteins. Afterwards, we expanded our scope to study the interaction of rRNA through the X circular code. Finally, we search for the X circular code motifs in the complete DNA sequences of chromosomes of the eukaryotic genomes. This study introduced new properties to the circular code theory.
