The D3 dopamine receptor (D3R) belongs to D2-class of dopamine receptors (DARs) and is
involved in emotion, movement, and reward. D3R dysfunction has been reported in some
neuropsychiatric disorders such as addiction, cognitive deficits, depression, schizophrenia, and
Parkinson’s disease. Genetic studies have shown two polymorphic variants of the D3R gene
resulting from substitution of serine to glycine at position nine of the amino terminus. Isoform
5 of adenylyl cyclase (AC5) is one of the nine transmembrane bound ACs in the brain and
myocardium. Previous studies in rats have shown that AC5 is expressed in the striatum, nucleus
accumbens and olfactory tubercle and at lower levels in islands of Calleja, where the D3R is
also expressed. Previous studies showed that although D2R and D4R inhibit ACs activity in
different cell types, inhibition of ACs by D3R is weak and often undetectable. It has been
shown that D3R selectively inhibits AC5 activity in human embryonic kidney 293 (HEK293)
cells co-transfected with D3R and AC5. Co-expression of D3R and AC5 in brain regions which
are major coordinators of normal and pathological movement, and the selective inhibition of
AC5 activity by D3R raise the possibility of a functional link between AC5 and D3R in the
modulation of signal transduction and trafficking. I hypothesized that AC5 plays a unique role
in modulation of D3R trafficking and signaling pathways through interaction between D3R
and AC5. Herein, I demonstrated an interaction between D3R and AC5 in vivo and in vitro
using reciprocal co-immunoprecipitation/immunoblotting (co-IP/IB) assays. Interestingly, DA
may facilitate the formation of protein complex between D3R and AC5 in vitro. Radio ligand
binding assays revealed that heterodimerization of D3R polymorphic variants with AC5 does
not change ligand binding affinity and expression of the D3R. Furthermore, taking advantages
of GloSensor assays, selective inhibition of AC5 activity by D3Ser9 and D3Gly9 has been
shown following activation by DA and quinpirole. Using ELISA studies showed that AC5
promotes cell surface expression and total expression of D3Ser9 and D3Gly9. Moreover, ELISA results suggested that AC5 facilitates DA-induced D3Ser9 endocytosis in dynamin and
β-arrestin 2 dependent process, while having no effect on D3Gly9 polymorphic variant. The
results also revealed that AC5 attenuates heterologous (PKC-induced) internalization of
D3Ser9, while it does not have any effect on D3Gly9 heterologous internalization. My results
also displayed a complex formation between D3R, AC5 and, β-arrestin 2 under basal and DA
stimulation conditions, which emphasize the role of β-arrestin 2 in D3R signal transduction.
Overall, a new regulatory mechanism for D3R has been suggested. My results suggested that
complex formation between both D3R polymorphic variants with AC5 can regulate signaling
and trafficking properties of D3R without changing the binding affinity of the receptor. These
data will be meaningful for understanding of diseases and developing treatment strategies.

Rapporten är en sammanställning av gruppens samlade erfarenheter och lärdomar av ett projekt vars syfte var att utveckla ett verktyg som ska visualisera svarstider mellan mobila klienter och datacenter. Genom att använda kontinuerlig prototypning har gruppen kunnat arbeta på ett användarna sätt för att uppfylla kundens verkliga behov. För att uppnå detta användes utvecklingsmetodiken Kanban. Under projektets gång anpassades metodiken för att bättra passa in i arbetet. Projektets användartester har lett till sammanfattande erfarenheter om visualisering av data. Visualiseringar som projektgruppen ansetts tydliga uppfattades inte alltid på samma sätt av användarna. Att visualisera flera parametrar på en världskarta anses vara problematiskt då kartan i sig endast består av länder. För visualisering av flera parametrar måste då även externa användas, exempelvis cirklar eller andra former.

Fibroblast growth factor-23 and Klotho in bone/mineral and parathyroid disordersKrajisnik, Tijana January 2009 (has links)
Fibroblast growth factor-23 (FGF23) is a novel, bone-produced hormone that regulates renal phosphate (Pi) reabsorption and calcitriol metabolism. Disorders of mineral and bone metabolism, such as autosomal dominant hypophosphatemic rickets (ADHR) and hyperostosis-hyperphosphatemia syndrome (HHS), witness the importance of well-balanced serum levels of FGF23. Patients with chronic kidney disease (CKD) are highly morbid due to Pi retention/hyperphosphatemia and calcitriol deficiency, which lead to elevated serum levels of parathyroid hormone (PTH) and secondary hyperparathyroidism (sHPT). As a response to hyperphosphatemia, CKD patients have also remarkably high serum FGF23 levels, which are associated with cardiovascular risk factors and increased mortality in CKD. The overall aim of this dissertation was to discern a possible role of FGF23 in parathyroid biology. Our in vitro experiments on isolated bovine parathyroid cells demonstrate that FGF23 directly and dose-dependently suppresses the PTH production and secretion, while increasing the expression of the 25-hydroxyvitamin D3-activating enzyme 1α-hydroxylase. We investigated possible expressional changes in the FGF23 receptor co-factor Klotho in hyperparathyroid disorders and found that Klotho expression is decreased or absent and inversely correlated to serum calcium (Ca) in adenomas of primary HPT (pHPT). In the hyperplastic parathyroid glands of sHPT, Klotho expression declines in parallel with the kidney function and correlates with the glomerular filtration rate. Moreover, Klotho expression is suppressed by Ca and FGF23, increased by calcitriol, but unaffected by Pi and PTH in vitro. Finally, we identified a novel missense mutation in the gene encoding GALNT3, which is normally involved in the post-translational glycosylation of FGF23, as the cause of aberrant FGF23 processing in a patient with HHS. In summary, we provide evidence for a novel bone/parathyroid axis in which FGF23 functions as a direct, negative regulator of the PTH production. High extracellular Ca is a major determinant of the Klotho expression in pHPT, whereas the Klotho levels in sHPT may be attributed to a combination of the high FGF23 and Ca, and low calcitriol levels associated with CKD. Hence, the decreased Klotho expression in sHPT could explain the concomitantly high FGF23 and PTH levels, as well as the failure of FGF23 to prevent or mitigate the development of sHPT in CKD.

Informal environmental regulation of industrial air pollution: Does neighborhood inequality matter?Moser, Mathias, Zwickl, Klara 11 1900 (has links) (PDF)
This paper analyzes if neighborhood income inequality has an effect on informal regulation of environmental quality, using census tract-level data on industrial air pollution exposure from EPA´s Risk Screening Environmental Indicators and income and demographic variables from the American Community Survey and EPA´s Smart Location Database. Estimating a spatial lag model and controlling for formal regulation at the states level, we find evidence that overall neighborhood inequality - as measured by the ratio between the fourth and the second income quintile or the neighborhood Gini coefficient - increases local air pollution exposure, whereas a concentration of top incomes reduces local exposure. The positive coefficient of the general inequality measure is driven by urban neighborhoods, whereas the negative coefficient of top incomes is stronger in rural areas. We explain these findings by two contradicting effects of inequality: On the one hand, overall inequality reduces collective action and thus the organizing capacities for environmental improvements. On the other hand, a concentration of income at the top enhances the ability of rich residents to negotiate with regulators or polluting plants in their vicinity. <div>Large datasets related to Indiana’s Education and Workforce development are used by various demographics such as stakeholders and decision makers in education and government, parents, teachers and employees of various companies to find trends and patterns in the data to better guide decision-making through statistical analysis. However, most of this data is scattered, textual and available in the form of excel sheets which makes it harder to look at the data from different perspectives, drill down and roll up the data and find trends and patterns in the data. Such data representation does not take into account the inherent characteristics of the user which can affect how well the user understands, perceives and interprets the data.</div><div>Information dashboards used to view and navigate between visualizations of different datasets, provide a coherent, central access to all data, and make it easy to view different aspects of the system. The purpose of this research is to create a new data visualization dashboard for visualizing education and workforce data and find which design principles are applicable while designing such a dashboard for the target demographic in the education and workforce domain. This study also aims at assessing how the introduction of such a data dashboard affects the work processes and decision making of stakeholders involved in education and workforce development in the state of Indiana.</div><div>User studies consisting of usability testing and semi-structured interviews with the stakeholders in education and workforce development in the state of Indiana is conducted to test the effectiveness of the dashboard. Finally, this research proposes how a regional map-based dashboard can be used as an effective method to design a data dashboard for education and workforce data for other states and other domains as well.</div>
