Do Feelings of Defeat and Entrapment Change over Time? (1) Background. Defeat and entrapment have been highlighted as major risk factors of suicidal ideation and behavior. Nevertheless, little is known about their short-term variability and their longitudinal association in real-time. Therefore, this study aims to investigate whether defeat and entrapment change over time and whether defeat predicts entrapment as stated by the integrated motivational–volitional model of suicidal behavior. (2) Methods. Healthy participants (n = 61) underwent a 7-day smartphone-based ecological momentary assessment (EMA) on suicidal ideation/behavior and relevant risk factors, including defeat and entrapment and a comprehensive baseline (T0) and post (T2) assessment. (3) Results. Mean squared successive differences (MSSD) and intraclass correlations (ICC) support the temporal instability as well as within-person variability of defeat and entrapment. Multilevel analyses revealed that during EMA, defeat was positively associated with entrapment at the same measurement. However, defeat could not predict entrapment to the next measurement (approximately two hours later). (4) Conclusion. This study provides evidence on the short-term variability of defeat and entrapment highlighting that repeated measurement of defeat and entrapment—preferably in real time—is necessary in order to adequately capture the actual empirical relations of these variables and not to overlook significant within-person variability. Further research—especially within clinical samples—seems warranted.

Un faible apport alimentaire en acides gras polyinsaturés (AGPI) de la série n-3 a été associé à la prévalence des troubles de l'humeur chez l’Homme. Chez les rongeurs, les approches nutritionnelles visant à modéliser une alimentation pauvre en AGPI n-3 ont largement été développées au siècle dernier. En effet, un régime alimentaire carencé en AGPI n-3 sur une ou plusieurs générations induit chez le rongeur des altérations des comportements émotionnels tels que des comportements de type dépressif ou anxieux. Nous avons montré au laboratoire Nutrineuro que des souris nourries avec un régime déficient en AGPI n-3 présentent des niveaux d’AGPI n-3, en particulier l'acide docosahexaénoïque (DHA, un AGPI n-3) plus faible dans le cortex préfrontal (PFC) et dans le noyau accumbens (NAc) par rapport aux souris contrôle. De plus, nous avons pu mettre en évidence qu’une alimentation déficiente en AGPI n-3 est capable de moduler la plasticité synaptique dépendante du système endocannabinoïde (eCB). De fait, la réduction de DHA dans le CPF et le NAc est accompagnée d'une altération de la dépression à long terme (LTD-eCB) et des voies de signalisation dépendantes du système eCB au niveau du CPF (Lafourcade et al., 2011 ; Larrieu et al, 2012). Nos données indiquent que ces altérations sont dues à un découplage entre le récepteur cannabinoïde 1 (CB1R) et la protéine Gi/o. De plus, les souris déficientes en AGPI n-3 présentent des déficits comportementaux dans plusieurs tests évaluant les comportements émotionnels. Afin de mieux comprendre les mécanismes qui sous-tendent la diminution du DHA dans le CPF et les altérations des comportements émotionnels, nous avons étudié la morphologie neuronale dans le CPF et l’axe hypothalamo-hypophysaire (HPA) chez les souris déficientes en AGPI n-3. Nous avons montré que le régime alimentaire déficient en AGPI n-3 induit une atrophie de l’arborisation dendritique dans les neurones pyramidaux du CPF. L'atrophie dendritique est semblable à celle mesurée chez les souris soumises au régime équilibré en AGPI n-3 et soumises à un stress chronique de défaite sociale (CSDS). Aucun effet additionnel du CSDS sur la morphologie neuronale et le comportement émotionnel n’a été observé chez les souris déficientes en AGPI n-3. Nous avons ensuite étudié le rôle de l’axe HPA dans le développement des altérations comportementales et neurobiologiques chez les souris déficientes en AGPI n-3. Ces souris présentent une diminution de l'expression des récepteurs des glucocorticoïdes (GR) dans le CPF associée à une augmentation des taux circulants de corticostérone. Dans leur ensemble, nos résultats montrent qu’un faible apport alimentaire en AGPI n-3 peut modifier la plasticité synaptique dépendante du système eCB ainsi que l’arborisation dendritique des neurones du CPF. Nous avons également pu montrer que l’élévation des niveaux de corticostérone était impliquée dans l’altération des comportements émotionnels observée chez des souris nourries avec un régime déficient en AGPI n-3. / Low dietary intake of n-3 polyunsaturated fatty acids (PUFAs) has been associated with the prevalence of mood disorders in humans. In rodents, nutritional approaches aiming at modeling poor dietary n-3 PUFAs intake have been extensively developed in the last century. As a result, one- or multi-generation dietary n-3 deficiency induces depressive and anxiety-like behaviors. We have shown in the Nutrineuro lab that mice fed with a diet deficient in n-3 PUFAs exhibit decreased n-3 PUFAs levels, especially docosahexaenoic acid (DHA, a n-3 PUFA) levels in the prefrontal cortex (PFC) and in the nucleus accumbens (NAc). We showed that dietary n-3 PUFA is able to modulate endocannabinoid (eCB) dependent plasticity since DHA reduction in PFC and NAc is accompanied with eCB dependent long term depression (eCB-LTD) and eCB signaling impairment in the PFC (Lafourcade et al., 2011; Larrieu et al., 2012). Our data indicate that LTD alteration results from region-specific uncoupling of CB1 receptor from its effector Gi/o protein. In addition, n-3 deficient mice display behavioral deficits in several tests measuring emotional behavior. To further understand the mechanisms underlying DHA decrease in the PFC and emotional behavior alteration, we thoroughly investigated neuronal morphology and hypothalamic-pituitary-adrenal (HPA) axis in n-3 deficient mice. We showed that n-3 deficient diet induced dendritic atrophy in pyramidal neurons within the PFC. The dendritic atrophy was comparable to the one measured in control diet mice submitted to chronic social defeat stress (CSDS). No additional effect of CSDS on both neuronal morphology and emotional behavior was measured in n-3 deficient mice. We therefore investigated the role of the HPA axis deregulation in the development of behavioral and neurobiological alterations of n-3 deficient mice. We found a decreased expression of glucocorticoid receptor (GR) in the PFC of n-3 deficient mice together with increased circulating levels of corticosterone. Collectively, we unraveled one crucial mechanism underlying n-3 deficiency-induced alterations. Our results show that low dietary n-3 PUFAs can alter eCB-dependent plasticity and neuronal dendritic atrophy within the PFC leading to emotional behavior impairment. Importantly, we further demonstrated that corticosterone elevation in n-3 deficient mice was involved in the n-3 deficiency-induced emotional behavior and dendritic arborization alterations.

Justification for this topic is based on the perception that the primitive man, with his mythic thoughts, and the modern man, straight thoughts formulated, have the same collective ritual dimensional base. Ritual must be read as a group of actions defined by their symbolic importance to a specific society. The actions and symbols aren't freely chosen by individuals, they are imposed by an external source. For the primordial man, the source is the ceremonial priests. For the modern man, the source is the influence of mass media. Showing how media uses the archetypical and sacred aspect of the ritual, manipulating its core for their own needs and satisfaction: fascination, enchantment, infiltration and stereotyping. By the Cultural Semiotic prism, the thesis aspires to contribute to studies and cultural communication, aiming for the rescue of complexity in the means of communication. How primitive rituals formats still survive in modern media, albeit in a saturated way, is the question this study addresses. Adopted methodological procedure was a research of qualitative character, involving analysis and bibliographical survey. As the theorical board of references are the authors B. Cyrulink, D. mídia de massa, como tentativa de suprir o vazio existencial, proporcionado pelo
surgimento da consciência no homem genérico. A justificativa da escolha do tema
repousa na percepção de que tanto o homem primitivo, com sua estrutura de
pensamento mítico, quanto o homem atual, com seu pensamento linearmente
formulado, alimentam-se do ritual de dimensão coletiva. Entende-se por ritual um
conjunto de ações, caracterizadas por seu valor simbólico, para determinado grupo
social. Tanto as ações quanto o simbolismo não são escolhidos livremente pelo
indivíduo, mas ditados por uma fonte externa. No caso do homem primordial, a fonte
relaciona-se aos mestres da cerimônia religiosa. No caso do homem atual, a fonte
refere-se ao poder de influência da mídia de massa. Nesse sentido, a intenção do
trabalho de pesquisa é mostrar como a mídia utiliza o recurso arquetípico e sagrado
do ritual, transportando seu núcleo de sentido para a satisfação dos seus interesses
imediatos: fascínio, encanto, infiltração e estereotipagem. Sob a vertente da Semiótica
da Cultura, a dissertação objetiva contribuir para os estudos de comunicação e
cultura, visando o resgate da complexidade nos meios de comunicação. O problema
de pesquisa diz respeito à questão de como o formato do ritual primitivo sobrevive,
de modo saturado, nos meios de comunicação de massa, e de que modo lhes foi
incorporado. Referente às questões metodológicas, o procedimento adotado foi uma
pesquisa de caráter qualitativo, envolvendo crítica e levantamento bibliográfico. Para
estabelecer a relação desejada entre conteúdo arcaico e mídia de massa foram
selecionadas como referencial teórico a semiótica da cultura (centradas nos autores
I. Bystrina, M. S. Contrera e N. Baitello Jr), a teoria da comunicação (de B. Cyrulink,
K. Lorenz, V. Flusser e V. Romano), as teorias da mídia e do imaginário (baseadas
nos textos de D. Kamper e H. Pross), entre outras áreas do conhecimento e outros
autores, tais como, E. Morin, J. Hillman, M. Berman, N. Sevcenko e Z. Bauman. A
conclusão do estudo foi a de que, apesar de distorcido e banalizado, os princípios de
reorganização simbólica do ritual continuam a exercer notável influência na mídia e
na mentalidade do homem atual

Human genetic data suggests DISC1 (Disrupted-in-schizophrenia 1) is a susceptibility gene for schizophrenia and depression. Disc1 Q31L-/- mutants show depression-like behaviour and Disc1 L100P-/- mutants schizophrenia-like behaviour. Heterozygous mutants show an intermediate phenotype. In a gene-environment interaction study, we exposed heterozygotes to chronic social defeat (CSD) stress and phenotyped behaviour. Disc1, Bdnf(III) and Pde4b mRNA levels were also measured. Moreover, as epigenetic mechanisms may mediate some effects of CSD, we also exposed wildtype mice to CSD concurrently with the histone deacetylase inhibitor valproate. We found that CSD increased anxiety in L100P-/+ mutants, and that levels of Disc1, Bdnf(III) and Pde4b mRNA were higher in this mutant. Valproate treatment did not correct CSD-induced behavioural changes. In conclusion, we have demonstrated an interaction between a strong susceptibility gene for psychiatric disease and an environmental manipulation similar to stressors known to affect mental illness.

[pt] Pesquisa de caráter micro-analítico, utiliza os
pressupostos da Sociolingüística Interacional para
descrever e analisar o discurso oral institucional com o
objetivo de estudar a quebra da expectativa do público no
contexto de um serviço de atendimento telefônico
encarregado da captação de anúncios classificados. Na
construção dos dados, foram realizadas visitas à editora,
entrevistas, consultas à documentos de pesquisa de mercado
de propriedade da empresa e gravações dos atendimentos em
áudio. Os resultados mostram que a quebra da expectativa do
público em relação à empresa ocorre pela dissonância entre
a prática e a publicidade veiculada pela empresa, por
alinhamentos conflitantes entre os interesses da empresa
versus os interesses do público e pelo enquadre assimétrico
que coloca o público em posição secundária. Paralelamente
aos traços lingüísticos, evidenciam-se os aspectos ético-
filosóficos e mercadológicos subjacentes a encontros de
serviço desta natureza que suscitam uma reflexão acerca da
contribuição de posturas responsáveis na gestão de empresas
em favor de uma melhor qualidade de vida. / [en] Case study based on Interactional Sociolinguistics
principles, dealing with a micro-analysis of talk-in-
interaction to investigate the defeat of expectation in the
institutional discourse of a call-center service for
classified ads. Data generating methods include field notes
taken during visits to the company premises and interviews
with some of the staff members; marketing research
documents reading procedures and telephone call-service
recordings and transcriptions. Data analysis has shown that
the defeat of expectation built up by the public is due to
cognitive dissonance between the actual vehicle practice
and its publicity, as well as conflicting footings and
asymmetrical alignments concerning the customers. Alongside
linguistic traits, evidence of ethical and marketing
aspects in this kind of institutional interaction points to
the centrality of business ethics for more responsible
management and better quality of life.
