Por que os adolecentes comem o que comem? Determinantes do consumo alimentar / Why adolescents eat what they eat? Determinants of food choice

Introdução: Muitos são os estudos que avaliam o consumo alimentar na adolescência entretanto não se sabe quais fatores motivam esses jovens a se alimentarem de forma inadequada. Objetivo: Avaliar, de forma quantitativa e qualitativa, os fatores determinantes do consumo alimentar de adolescentes de escolas técnicas em São Paulo. Métodos: O presente estudo é parte de uma pesquisa matriz, que avaliou 1167 jovens de ambos os gêneros. Os fatores determinantes do consumo alimentar foram avaliados em duas etapas. Na primeira etapa quantitativa foram utilizados dois instrumentos que foram desenvolvidos para a pesquisa matriz, o primeiro um questionário de avaliar atitudes alimentares de adolescentes e o segundo avaliou o consumo alimentar através de um questionário de frequência alimentar baseado na pirâmide dos alimentos. O primeiro instrumento apresenta uma questão onde 15 fatores determinantes do consumo alimentar são apresentados e cada adolescente deveria escolher os 3 mais importantes. O peso e a estatura de todos os indivíduos foi mensurado e posteriormente o Índice de Massa Corporal foi calculado afim de classificar os adolescentes quanto à adequação de seu estado nutricional segundo os critérios da Organização Mundial da Saúde. A avaliação qualitativa foi realizada com uma subamostra do projeto matriz (n=45) e contou com a realização de grupos focais. Os 15 fatores determinantes do consumo foram agrupados em 3 grupos a partir da realização da análise de cluster e esses grupos foram comparados com relação ao gênero, estado nutricional, consumo de alimentos e dos grupos alimentares baseados na pirâmide dos alimentos para adolescentes. Resultados: Os 3 fatores determinantes mais referidos pelos adolescentes na etapa quantitativa foram, em ordem de preferência, o sabor, o fato do adolescente gostar do alimento e tentar ter uma alimentação mais saudável, enquanto que a etapa qualitativa mostrou que os fatores de falta de tempo, conveniência e praticidade foram os mais referidos. Os adolescentes que preferiam os fatores determinantes relacionado às características dos alimentos, entre eles o sabor, consumiam significativamente mais alimentos com alto conteúdo de gorduras e açúcares. Conclusão: Conclui-se que para o grupo de adolescentes avaliados o sabor foi o fator mais importante e que ele pode explicar a inadequação dietética nesse grupo etário. / Introduction: Many studies evaluate food consumption in adolescence however little is known about the factors that influence adolescent\'s food choices. Objective: To evaluate, in a quantitative and qualitative way, the factors that influence adolescent\'s food choice from technical schools from São Paulo. Methods: This study is part of a research which evaluated 1167 adolescents from both gender. Factors that influence adolescent\'s food choice were evaluated in two steps, the first one evaluated in a quantitative way used two surveys that were developed to the research. The questionnaire evaluated adolescent\'s eating attitudes and included a question which included 15 factors which the adolescents should select the 3 more important, and the second survey assessed food consumption through a food frequency questionnaire that was based on the food pyramid. Body weight and height were measured and the Body Mass Index calculated in order to classify their weight status according to the World Health Organization criteria. The second part of this study, the qualitative analyses, used a smaller sample from the main study (n=45) and focus groups were performed. The 15 factors were divided into groups using the cluster analyses method and compared between genders, weight status and food consumption. Results: Taste and liking the food and trying to have a healthier eating were the 3 most referred factors in the quantitative analyses, while in the qualitative research lack of time and convenience were the most reported factors that influence the adolescent\'s food choices. Adolescents who prefer factors related to the food characteristics, such as taste, eat significantly more high fat and sugar foods. Conclusion: In this group the taste of food was the most important factor and can explain the eating inadequacies seen in this population.

Avaliação de eficiência das cooperativas de crédito no Brasil: um estudo com base na intermediação financeira e na prestação de serviços / Efficiency evaluation of credit cooperatives in Brazil: A study based on financial intermediation and on provision of services

As cooperativas de crédito são instituições financeiras que têm como finalidade principal a prestação de serviços bancários e de intermediação financeira voltando-se para a geração de benefícios ao cooperado, através de benefícios econômicos encontrados sob a forma cooperativada. Considerando o objetivo destas organizações, o presente trabalho teve por objetivo avaliar a eficiência das mesmas considerando duas vertentes: 1) a atividade de intermediação financeira; e 2) a atividade de prestação de serviços bancários, a partir de variáveis contábeis. A técnica utilizada para a avaliação da eficiência foi a Análise Envoltória de Dados e foram analisados os determinantes da eficiência por meio de regressões do tipo Tobit. A amostra é composta por 315 cooperativas singulares listadas na base de dados do Banco Central do Brasil e foram utilizados os dados dos balancetes anuais e semestrais (soma dos semestres para as contas de resultado de cada ano) apresentados para período de 2007 a 2014. Os resultados indicaram baixa eficiência das cooperativas na prestação de serviços bancários, com 73% da amostra apresentando ineficiência ao longo de todos os anos do período em análise. Na atividade de intermediação financeira 20 cooperativas foram eficientes ao longo do período completo, com as demais apresentando graus de ineficiência abaixo de 16% em todos os anos. Quanto aos determinantes da eficiência, verificou-se como principais fatores na atividade de intermediação financeira a Imobilização, a Insolvência, o Descasamento Passivo e as Despesas Administrativas, enquanto na atividade de prestação de serviços os principais determinantes foram a Captação por Floating e as Despesas Administrativas. Os resultados da eficiência na intermediação financeira indicaram baixos graus de ineficiência, enquanto os altos graus de ineficiência na prestação de serviços bancários alertaram para a pouca importância dada ao fornecimentos destes serviços e para a importância de se utilizar dois modelos distintos para a avaliação da eficiência em cooperativas de crédito, dado o comportamento desvinculado entre as duas atividades avaliadas. / Credit unions are financial institutions whose main purpose is the provision of banking and financial intermediation turning to the generation of benefits to the cooperative through economic benefits found in the cooperative form. Considering the purpose of these organizations, this study aimed to evaluate the efficiency of these organizations considering two aspects: 1) the financial intermediation activity and 2) the activity of banking services. For this aim it was used accounting variables. The technique used for the assessment of efficiency was the Data Envelopment Analysis and the determinants of efficiency were analyzed using Tobit regressions. The sample is composed of 315 individual cooperatives listed in the Central Bank\'s database of Brazil and it was used the data of the annual and semi-annual balance sheets (sum of semesters for the result accounts of each year) presented for the period 2007 to 2014. The results indicated low efficiency of cooperatives in providing banking services, with 73% of the sample showing inefficiency over each year of the period. In the financial intermediation activity 20 cooperatives were effective over the entire period, and the other cooperatives presented degrees of inefficiency below 16% in every year of the period. As the determinants of efficiency, it was as key determinants in financial intermediation activity: the immobilization, the insolvency, the mismatch liabilities and the administrative expenses, while in activity of providing services the main determinants were the capture by floating and the administrative expenses. The results of efficiency in financial intermediation were considered satisfactory, but the high degree of inefficiency in the provision of banking services warned of the low priority given to the provision of such services and the importance of using two different models for the assessment of efficiency in credit cooperatives, given the unlinked behavior between the two activities evaluated.

Robust determinants of OECD FDI in developing countries: Insights from Bayesian model averaging

In this paper, we examine the determinants of outward FDI from four major OECD investors, namely, the US, Germany, France, and the Netherlands, to 129 developing countries classified under five regions over the period 1995-2008. Our goal is to distinguish whether the motivation for FDI differs among these investors in developing countries. Rather than relying on specific theories of FDI determinants, we examine them all simultaneously by employing Bayesian model averaging (BMA). This approach permits us to select the most appropriate model (or combination of models) that governs FDI allocation and to distinguish robust FDI determinants. We find that no single theory governs the decision of OECD FDI in developing countries but a combination of theories. In particular, OECD investors search for destinations with whom they have established intensive trade relations and that offer a qualified labor force. Low wages and attractive tax rates are robust investment criteria too, and a considerable share of FDI is still resource-driven. Overall, investors show fairly similar strategies in the five developing regions.

Through a Math Educational Didactic and technological work- shop offered in the ambit of activities in the Programa de Estudos Pós-Graduados em Educação Matemática da PUC/SP (grupo TecMEM), where a group of teachers from Estado de São Paulo Public School were under observation in the realization in dynamic activities using the software WinMat as well as in the executions of tasks with traditional medias, all of them related to the theme "matrices and determinants". Through qualitative analysis of data, it was possible to verify several difficulties by these groups of researchers, conceptual and technical, but these difficulties could decrease as soon as this articulation was being reached between the interface technological domain and the recover of mathematics concepts involved. Besides, the teachers who are in this investigation have noticed the importance of pedagogical strategies in which mathematics technological classes should be included, more than the simple use of software or other artifact to the students use / Este trabalho relata uma experiência investigativa relacionada à formação continuada de professores de Matemática do Ensino Médio, mais precisamente no que tange ao preparo dos mesmos em relação ao uso de tecnologias como mediadoras em suas aulas. Através de uma Oficina Didática de Educação Matemática e Tecnologias, oferecida no âmbito das atividades do Programa de Estudos Pós-Graduados em Educação Matemática da PUC/SP (grupo TecMEM), um grupo de professores da rede pública do Estado de São Paulo foi acompanhado na realização de atividades dinâmicas com uso do software Winmat, bem como na consecução de tarefas em com mídias tradicionais, todas relacionadas ao tema matrizes e determinantes . Através de análise qualitativa dos dados, foi possível constatar diversas dificuldades por parte dos sujeitos da pesquisa, tanto conceituais quanto técnicas, que puderam ser minoradas à medida que se alcançava uma articulação entre o desenvolvimento do domínio tecnológico da interface e a recuperação dos conceitos matemáticos envolvidos. Além disso, os docentes envolvidos nesta investigação indicaram perceber a importância das estratégias pedagógicas que incluam tecnologias nas aulas de Matemática, muito além da mera inserção de softwares e ou artefatos para o uso de seus estudantes

Contexte: De récentes études ont explicitement démontré que les communautés d’immigrants doivent affronter des difficultés particulières afin d'accéder aux services de santé et font face à des disparités sanitaires significatives, subissant l'effet de déterminants sociaux sous-jacents de la santé (DSS). Les immigrants représentant un pourcentage grandissant de la population dans de nombreux pays développés, mieux comprendre et supprimer ces barrières est une des grandes priorités. En se fondant sur l'utilisation d'une structure DSS, cette thèse contribue aux recherches actuelles concernant l'accès aux services de santé parmi les populations d'immigrants bangladais dans trois villes internationales : Lisbonne (Portugal), Bruxelles (Belgique) et Boston (Massachusetts, U.S.A.). À cet effet, elle apporte des précisions sur les données qualitatives limitées disponibles concernant les communautés immigrantes bangladaises qui vivent sur les sites de recherche mentionnés ci-dessus.Résultats: Les résultats obtenus ont montré que les immigrants bangladais vivant sur ces trois sites de recherche possèdent une vision généralement holistique de la santé et du bien-être. Les pressions de l'acculturation et l'isolement social constituent deux des DSS pertinents ayant une influence sur la santé et l'accès aux services sanitaires. Parmi les principaux enjeux de santé et besoins en soins de cette communauté, les problèmes de santé mentale et l'isolement social, l'alimentation et les habitudes alimentaires, le manque d'activité physique, des niveaux élevés de diabète et de maladies cardiovasculaires ont été identifiés comme facteurs influençant. Certaines valeurs culturelles associées à des motivations d'ordre économique peuvent conditionner l'importance que les individus accordent à leur santé. A titre d'exemple, l'obligation envers la famille, profondément ancrée dans la culture bangladaise, se traduit souvent chez les immigrants travaillant à l'étranger par un sentiment de pression économique et se manifeste par un stress chronique et un déséquilibre entre vie professionnelle et vie personnelle. La communication et les barrières linguistiques apparaissent comme étant les principaux freins à l'engagement des immigrants bangladais avec les prestataires de soins. Enfin, le type ainsi que le contexte de l'immigration joue un rôle important dans l'utilisation des services de santé par les immigrants bangladais: le sentiment d’être dans une situation transitoire sur le lieu où ils se trouvent tel qu'à Lisbonne et Bruxelles, en opposition à une perspective d'établissement plus permanent tel qu'à Boston, semble influencer l'utilisation active de soins de santé continus.Conclusion: Les problèmes de santé de chaque individu, la situation de leur statut d'immigrant et les circonstances familiales ont joué un rôle sur la manière dont les membres de la communauté bangladaise ont accédé et utilisé les services de santé sur chaque site de recherche. Bien que les liens sociaux au sein des communautés bangladaises vivant à l'étranger soient forts, tous les individus ne se trouvaient pas nécessairement en contact avec les membres de leur communauté. Ceci met en exergue le besoin d'un engagement et d'une sensibilisation auprès des immigrants susceptibles de ne pas bénéficier de la protection du capital social de leur communauté. Cette dissertation a permis de dresser le profil des déterminants sociaux influençant l'accès aux services de santé de la communauté immigrante bangladaise, avec pour objectif d'informer les professionnels travaillant avec cette population. A cet effet, les conclusions de cette recherche seront partagées avec les participants et intervenants de chaque site d'études afin de renforcer la compréhension des communautés immigrantes bangladaises et accroître les ressources mises à leur disposition. / Background: Recent studies clearly identify that immigrant communities experience specific challenges in accessing health care services, and face significant health disparities, that are impacted by underlying social determinants of health (SDH). As immigrants comprise an increasing percentage of the population in many developed nations, a better understanding and eliminating of these barriers is a major priority. Using an SDH framework, this dissertation contributes to the current research regarding access to health services among Bangladeshi immigrant populations in three international cities: Lisbon (Portugal), Brussels (Belgium), and Boston (MA-USA). In doing so, it expands upon the limited qualitative data available that concerns Bangladeshi immigrant communities living in these aforementioned research sites.Methods: This dissertation utilized a qualitative descriptive research design to comprehend issues from the perspective of both immigrant community members and service providers. The findings presented in this study, therefore, focus on data analyzed from 45 original in-depth interviews with Bangladeshi immigrant community members (n=32) and service providers (n=13) across Lisbon, Brussels, and Boston. Data was collected primarily using a semi-structured interview guide.Results: Bangladeshi immigrants living across the three research sites possess an overall holistic view of health and well-being according to results. Relevant SDH affecting health and accessing health services included pressures of acculturation and social isolation. The following were identified as some of the key community health issues and care needs: mental health and social isolation, food and dietary habits, lack of exercise, high levels of diabetes and cardiovascular disease. Certain cultural values coupled with economic motivators may influence the way individuals prioritize their health. For example, the obligation towards family that is deeply embedded in Bangladeshi culture often translated to feelings of economic pressure by immigrants working abroad and manifested as chronic stress and a work-life imbalance. Communication and language barriers emerged as the most prominent issue of engagement between Bangladeshi immigrants and service providers. Finally, the type and context of immigration played in important role in health service utilization by Bangladeshi immigrants: a transitory outlook on their current location such as in Lisbon and Brussels, as opposed to a more permanent outlook in Boston, appeared to influence the active uptake of continuous health care.Conclusion: Individual health priorities, immigration status, and family circumstances affected how Bangladeshi community members accessed and utilized health services at each research site. Although social networks in Bangladeshi communities abroad were strong, not all individuals were necessarily connected with their fellow community members. This underlines the need to initiate engagement and outreach to immigrant individuals who may fall outside of the protection of social capital in their community. This dissertation has established a profile of social determinants impacting access to health services for Bangladeshi immigrant communities, with the purposes of informing professionals working within this population. As such, results will be shared with participants and stakeholders at each of the study sites in order to strengthen the understanding of and resources available to Bangladeshi immigrant communities.

Decolonizing bodies: a First Nations perspective on the determinants of urban indigenous health and wellness in Canada

Through a research partnership with the First Nations Health Authority (FNHA) and using mixed methods participatory action research this Dissertation critically engages with dominant Western-based knowledge systems of well-being from a decolonizing standpoint to better understand the determinants of Indigenous health and well-being. This study specifically asks: what are the main factors effecting different dimensions of well-being for Indigenous peoples living in urban centres, how does engaging in traditional land-based activities and cultural ways of life affect well-being, and to what extent does intergenerational trauma impact well-being? Thirteen key informant interviews were conducted with FNHA members involved in the development of the First Nations Perspective on Health and Wellness (FNPOW) to garner knowledge about the thoughts, feelings, belief systems, values, and knowledge frameworks that are embedded in this perspective. A multi-level statistical model was developed informed by the First Nations Perspective on Health and Wellness, the 2012 Aboriginal Peoples Survey and 2011 National Household Survey, to produce health and wellness outcomes. Using a strength-based approach to well-being this study shows that the FNPOW advocates self-determination, and implementing the perspective in research work offers a pathway to generating measures of health and wellness rooted in Traditional knowledge systems, and a pathway to decolonizing bodies. These outcomes are a form of social capital reflective of Indigenous values that can be utilized as a resource to strengthen community capacity to support Indigenous self-determination. / Graduate

Recherche de nouveaux déterminants génétiques et épigénétiques de susceptibilité à la tumorigenèse intestinale au moyen du modèle murin Apcd14 / Identification of new genetic and epigenetic determinants of intestinal tumorigenesis susceptibility using the Apcd14 mouse model

Le cancer colorectal (CCR) représente un problème majeur de Santé publique. Des facteurs de risque environnementaux, ainsi que l'apparition séquentielle d'altérations génétiques et épigénétiques corrélant avec la progression tumorale ont été extensivement décrites. Cependant, les variations épigénétiques préexistant dans le tissu sain, potentiellement responsables de différences notoires de susceptibilité au CCR, sont restées élusives jusqu'à ce jour. Afin de répondre à cette problématique, la lignée murine Apcd14 (porteuse d'une mutation hétérozygote constitutive au niveau du gène Adenomatous polyposis coli) a servi de modèle au cours de ce projet. Les individus de cette lignée comportent une expressivité très variable, au sens qu'ils développent spontanément un nombre plus ou moins important de tumeurs intestinales, impactant sur leur survie. Cette hétérogénéité est observée en dépit de conditions d'élevage et de génomes considérés identiques. L'analyse exhaustive de cette lignée a conduit à y caractériser deux groupes de souris, présentant une gravité différente de phénotype. La variation d'expression génétique dans le tissu sain (i.e en amont de la tumorigenèse) a ensuite été analysée dans le but de comprendre l'établissement de cette hétérogénéité. Ceci a mené à la découverte d'une signature de gènes différemment exprimés entre les deux groupes, permettant de corréler de façon parfaite données moléculaires et phénotype. La potentielle héritabilité de cette signature a par la suite conduit à remettre en cause le statut considéré syngénique de la lignée. Les approches expérimentales effectuées ont déjà permis de cibler une région chromosomique, ce qui mènera à court terme à la caractérisation d'un nouvel acteur impliqué dans la tumorigenèse intestinale. De manière plus générale, les expériences développées durant ce projet ouvrent la voie à la notion de susceptibilité individuelle face au cancer dépendante de la variation d'expression génétique. / Colorectal cancer (CRC) is a major public health concern. Environmental clues, as well as sequential genetic and epigenetic alterations correlating with tumor progression have been extensively described. Meanwhile, pre-existing epigenetic variations in the healthy intestinal mucosa, potentially leading to differences in CRC suceptibility, have generally been overlooked. In order to answer to this question, we have made use of the Apcd14 mouse model (carrying a heterozygous mutation in the Adenomatous polyposis coli gene). Although all Apcd14 mice apparently share the same genome and are raised in the same environmental conditions, they exhibit a huge phenotypic variability at the individual level, spontaneously developping a few or numerous intestinal tumors, that ultimately results in differences in survival rates. Through detailed analysis of this strain, two groups have been characterized, exhibiting several phenotypic specificities. Gene expression analysis in the healthy intestinal tissue was then performed in order to understand the differences already existing, prior to tumorigenesis. This led to the identification of a group-specific gene expression signature, allowing a correlation between macroscopic phenotype and molecular data. Consideration of the hereditary potential of this signature led to reconsider the syngeneic status of the Apcd14 strain. Several experimental data already targeted a specific genomic region, and this will allow to identify the genetic alteration involved. More generally, this project opens the way to discover a link between individual susceptibility to intestinal tumorigenesis and gene expression variability.

Mudanças nos usos da terra no Brasil entre 1995 e 2006 / Land use changes in Brazil between 1995 and 2006

As mudanças nos usos da terra no Brasil têm sido muito discutidas atualmente, principalmente por causar desmatamento, degradação do solo, redução da biodiversidade e aumento da emissão de gases de efeito estufa, influenciando nas mudanças climáticas. Entretanto, a atividade agropecuária é importante para a economia brasileira por ser grande geradora de emprego e renda. Dessa forma, é necessário que o governo brasileiro, por meio de políticas agrícolas e ambientais, consiga regular a interação entre o meio ambiente e a produção agrícola de modo a gerar os maiores benefícios possíveis para a sociedade tanto do ponto de vista econômico quanto do ambiental. Para contribuir nessa discussão esse trabalho é composto por três artigos. O primeiro discutiu a evolução recente das políticas e instrumentos de políticas públicas que atuam diretamente nos usos das terras agrícolas privadas: o Código Florestal Brasileiro, o Importo Territorial Rural além de políticas públicas mais recentes de pagamentos por serviços ambientais. Concluiuse que a pouca fiscalização prejudica a efetividade das políticas e, consequentemente, não induz ao comportamento esperado dos produtores. Entretanto, as políticas de pagamentos por serviços ambientais e a criação da cota de reserva ambiental tem grande potencial de influenciar os produtores em direção a atitudes mais responsáveis ambientalmente. O segundo artigo discutiu as mudanças no uso do solo e a evolução da produção agrícola abordando o Brasil e cada um dos biomas brasileiros separadamente. Observou-se que as lavouras tem se expandido sobre as áreas de pastagens e florestas. Assim, é necessário aumentar a fiscalização e controle das atividades agrícolas de modo a direcionar a pressão para as áreas de pastagens, estimulando a intensificação da bovinocultura. Além disso, é necessário o desenvolvimento de alternativas específicas para a região da caatinga, visando melhorar a produção agrícola desse bioma. O terceiro e último artigo estudou os principais determinantes do uso do solo no Brasil e nos biomas. Identificou-se grande influência da mecanização em todos os usos de solo e, para o caso da cana-de-açúcar, da presença de usinas no município. Ainda, as elasticidades área preço calculadas identificaram pouca influência dos preços sobre as áreas plantadas, principalmente relacionados às áreas de pastagens e florestas. Concluiu-se que os resultados do modelo estimado podem não refletir adequadamente as pressões para as alterações nos usos da terra e diversas melhorias são sugeridas. Finalmente, conclui-se que é necessária uma abordagem interdisciplinar para compreender e abordar adequadamente as mudanças do uso do solo no Brasil. / Changes in the land use in Brazil have been much discussed today, mainly for causing deforestation, soil degradation, decrease of biodiversity and increased emission of greenhouse gases, influencing the climate change. The agricultural activity, however, is important for the Brazilian economy as it is a great generator of employment and income. Thus, it is necessary that the Brazilian government, through agricultural and environmental policies, gets to regulate the interaction between the environment and agricultural production in order to generate the greatest possible benefits to society not only from the economic but also from the environmental point of view. This work consists of three articles to contribute to this discussion. The first discussed the recent development of the public policies and policy instruments that act directly on the uses of private agricultural land: the Brazilian Forestry Code, the Rural Land Tax besides the latest public policies on payments for environmental services. It was concluded that little oversight undermines the effectiveness of policies and therefore does not lead to the expected behavior of producers. However, the policies of payments for environmental services and the creation of environmental reserve quota has great potential to influence attitudes towards producing more environmental responsibility. The second article discussed the changes in land use and the development of agricultural production in Brazil and each of the Brazilian biomes separately. It was observed that crops have expanded over the grazing areas and forests. Thus, it is necessary to increase the supervision and control of agricultural activities in order to direct the pressure to grazing areas, encouraging intensification of livestock production. Moreover, it is necessary to develop specific alternatives to the caatinga region, to improve agricultural production in this biome. The third and final article studied the main determinants of land use in Brazil and its biomes. A major influence of mechanization was identified in all land use and, for the sugar cane, the presence of industrial plants in the county also influenced its area. Still, in the model for Brazil, the dummies of biomes and the variables of altitude, rainfall and temperature had significant influence. Still, the area price elasticities calculated identified little influence of the prices on the areas planted, mainly related to grazing areas and forests. It was concluded that the results of the estimated model may not accurately reflect the pressures for changes in land uses and several improvements are suggested. Finally, it is concluded that an interdisciplinary approach is required to understand and properly deal with the changes of land use in Brazil.

Etude du rôle lors de l'infection et sur la défense des plantes hôtes des effecteurs de type III RipH1,2,3 et RipAX2 de Ralstonia pseudosolanacearum / Role during infection and on plant defense of the type III effectors RipH1,2,3 and RipAX2 from Ralstonia Pseudosolanacearum

Le système de sécrétion de type III est un des déterminants majeurs de la pathogénicité de Ralstonia pseudosolanacearum qui lui permet d’injecter des effecteurs de type III (les « Rip », « Ralstonia Injected Protein ») directement dans les cellules des plantes hôtes. Les effecteurs RipH1, RipH2 et RipH3 sont des effecteurs de type III conservés dans la plupart des souches séquencées. Au cours de ma thèse, le rôle de ces effecteurs RipH lors de l’infection de différentes plantes a été étudié en prenant comme point d’entrée les protéines de tomates avec lesquelles ces effecteurs interagissent. Un criblage par double hybride dans la levure a permis d’identifier 19 de ces protéines « cibles » de tomate. Des méthodes de génétique inverse ont ensuite été utilisées pour chercher le rôle des orthologues de ces protéines dans différentes plantes modèles lors de l’infection par Ralstonia pseudosolanacearum. Du VIGS chez Nicotiana benthamiana a permis de mettre en évidence l’implication des orthologues de la protéine TOM3 : la multiplication bactérienne est moins importante dans les feuilles lorsque l’expression de ces gènes est diminuée. Dans Arabidopsis thaliana, des mutants d’un gène orthologue de la cible TOM9, décrit comme jouant entre autres un rôle dans la remodélisation de la chromatine, est plus résistant à l’infection par R. pseudosolanacearum. Dans un deuxième chapitre correspondant à un article publié, le rôle de l’effecteur RipAX2 a été étudié dans la résistance des aubergines AG91-25. La présence de cet effecteur dans la souche GMI1000 est nécessaire à l’établissement de la résistance de cette variété dans laquelle le locus de résistance EBWR9 a été mis en évidence. L’ajout de RipAX2 dans la souche PSS4, une souche pathogène de AG91-25 qui ne possède pas cet effecteur naturellement, la rend non pathogène. De plus, le motif protéique putatif « zincbinding » qui est décrit comme nécessaire pour l’induction de réponses de défense chez l’espèce proche de l’aubergine Solanum torvum n’est pas nécessaire pour la résistance de AG91-25. Enfin, la conservation de l’effecteur RipAX2 dans les différentes souches du complexe d’espèces de Ralstonia solanacearum a été étudiée pour évaluer l’efficacité potentielle de cette source de résistance contre différentes souches. / One of the major virulence determinants of plant pathogenic Ralstonia species is the type III secretion system that enables it to inject proteins (also called “Ralstonia Injected Proteins” or Rip) into the host cells. The RipH1,2,3 type III effectors are conserved in different strains of the Ralstonia solanacearum species complex. The role of these effectors during infection has been studied, taking as an entry point the tomato proteins they interact with. Using yeast-two-hybrid screenings we have identified 19 tomato targets of these three RipH. Reverse genetics methods have then been used to study the role of orthologous genes of these targets in other model plants. Virus induced gene silencing in Nicotiana benthamiana showed that the orthologous genes of TOM3 were involved in plant response to Ralstonia pseudosolanacearum, as the bacterial multiplication was diminished in plants silenced for these genes. In Arabidopsis thaliana, mutants of the TOM9 orthologous gene which is described as involved in chromatin remodelisation were more tolerant to infection. In a second chapter corresponding to a published article, the role of RipAX2 has been studied. This effector triggers specific resistance in AG9125 eggplant which carry the major resistance locus EBWR9. This eggplant accession AG9125 is resistant to the wild type R. pseudosolanacearum strain GMI1000, while a ripAX2 defective mutant of this strain can cause wilt. The addition of ripAX2 from GMI1000 to the naturally pathogenic strain PSS4 suppresses its pathogenicity, demonstrating that RipAX2 causes AG9125 resistance. Moreover, a zinc binding motif described as necessary to induce defenses on the eggplant wild relative Solanum torvum upon RipAX2 recognition is not necessary for AG91-25 resistance.
