fixation (BNF) and yield of plants. Common bean (Phaseolus vulgaris L.) has been
considered as a species with low capacity of BNF as compared to other grain legumes, but
results demonstrate the potential of BNF to supply the N demand of the crop and to achieve
adequate yields. The objective of this study was to evaluate the effect of seed concentrations
of Mo and P on biomass production, nodulation and grain yield of common bean at different
sources of N supply. The field experiment was conducted between April and July 2009, in the
Santa Monica Farm of Embrapa Gado de Leite in the municipality of Valen?a ? Rio de
Janeiro State. The experimental design was a split-plot 4x4 factorial randomized block with 5
replicates. The main plots consisted of four N sources: control without N, mineral N
fertilization with 20 kg N ha-1 at sowing + 40 kg N ha-1 in cover, inoculation with the
commercial strain of rhizobia, rhizobia inoculation + 40 kg N ha-1 in cover. The subplots
consisted of four combinations between concentrations of P and Mo in the seeds (low P low
Mo, low P high Mo, Mo high P low, high-P high-Mo). The seeds of cultivar Carioca were
originating from a field experiment when 5 kg P ha-1 and 120 g Mo ha-1 were foliar sprayed at
52 and 71 days after emergence (DAE). In each subplot, plant biomass were sampled at 30, 42
and 57 DAE, and root, shoot and nodules dry mass, the number of nodules and the
accumulation of N in the shoot were determined. Grains were harvested at 90 DAE, and grain
yield, yield components, N and Mo concentratios in the grains were measured. Interactions
between the N sources and the concentrations of P and Mo in the seeds were not significant in
the three sampling times, except for shoot dry mass and number of nodules at 57 DAE. Seeds
with low P and high Mo increased shoot dry matter and grain yield of beans. Mineral N
fertilizer increased root mass in the early growth stages. The rhizobia inoculation increased
bean nodulation. There was a small effect of the seeds with high P concentration in the
accumulation of N and mass of shoots at 57 DAE. Mineral N fertilizer increased N
accumulation in the shoots at 57 DAE. The inoculation + 40 kg N ha-1 in cover gave similar
grain yield and N accumulation as the application of 60 kg N ha-1, both treatments superior to
the control. The use of seeds enriched with P and Mo, associated with the inoculation of seeds
and the application of N in cover, could maximize the process of biological nitrogen fixation
in the common bean crop. / A concentra??o de nutrientes nas sementes pode afetar o desenvolvimento inicial, a fixa??o
biol?gica de nitrog?nio (FBN) e a produ??o de gr?os das plantas por elas geradas. O feijoeiro
(Phaseolus vulgaris L.) tem sido considerado uma esp?cie de baixa capacidade de FBN em
compara??o com outras leguminosas de gr?o, mas resultados demonstram o potencial da FBN
em suprir a demanda de N do feijoeiro e obter boas produtividades. O objetivo deste estudo
foi avaliar o efeito dos teores de P e Mo em sementes na produ??o de biomassa, nodula??o e
no rendimento de gr?os de feijoeiro sob diferentes fontes de fornecimento de N. O
experimento foi conduzido em condi??es de campo entre abril e julho de 2009, na Fazenda
Santa M?nica da Embrapa Gado de Leite, no munic?pio de Valen?a ? RJ, em delineamento
em blocos ao acaso em fatorial 4x4 com 5 repeti??es em parcelas subdivididas. As parcelas
foram compostas por quatro fontes de N: testemunha absoluta sem N; aduba??o com N
mineral com 20 kg N ha-1 no plantio + 40 kg N ha-1 em cobertura; inocula??o com a estirpe
comercial de riz?bio recomendada para a cultura; inocula??o com estirpe comercial + 40 kg N
ha-1 em cobertura. As subparcelas foram compostas por quatro combina??es entre teores de P
e de Mo na semente (baixo P baixo Mo, baixo P alto Mo, alto P baixo Mo, alto P alto Mo). As
sementes da cultivar Carioca utilizadas foram oriundas de um experimento de campo onde
foram efetuadas aplica??es foliares com 5 kg P ha-1 e 120 g Mo ha-1 aos 52 e 71 dias ap?s
emerg?ncia (DAE). Em cada subparcela foram efetuadas tr?s amostragens de biomassa, aos
30, 42 e 57 DAE, determinando-se a massa seca de raiz, parte a?rea e n?dulos, o n?mero de
n?dulos e a acumula??o de N na parte a?rea. A colheita de gr?os foi efetuada aos 90 DAE,
determinando-se o rendimento de gr?os, os componentes de produ??o e o teor de N e de Mo
nos gr?os. As intera??es entre as fontes de N e os teores de P e Mo das sementes, nas tr?s
?pocas de coleta, n?o foram significativas, exceto para massa seca de parte a?rea e n?mero de
n?dulos aos 57 DAE. As sementes com baixo P e alto Mo foram as que promoveram maior
massa de parte a?rea e tamb?m a maior produ??o de gr?os de feij?o. A aduba??o com N
mineral promoveu maior massa radicular nos est?gios iniciais de crescimento. A inocula??o
das sementes com riz?bio aumentou a nodula??o do feijoeiro. Houve um pequeno efeito dos
altos teores de P nas sementes na acumula??o de N e de biomassa da parte a?rea aos 57 DAE.
A aplica??o de N mineral promoveu maior ac?mulo da N na parte a?rea aos 57 DAE. A
inocula??o combinada com 40 kg N ha-1 em cobertura forneceu rendimento de gr?os e
acumula??o de N nos gr?os similar ? aplica??o de 60 kg N ha-1, por sua vez superiores ?
testemunha. O uso de sementes enriquecidas com P e Mo, associada com a inocula??o de
sementes e a aplica??o de N em cobertura, podem maximizar o processo de fixa??o biol?gica
de nitrog?nio na cultura do feijoeiro.

Previous issue date: 2016-02-16 / In order to increase agricultural yield and nutrient uptake by plants, diazotrophs as fixing N bacteria from atmospheric air have been used as inoculum for seed treatment since they also promote plant growth. However, the beneficial uses of diazotrophs in agriculture rely on complex interactions, including the sort of fertilizer used. Swine wastewater has been used as biofertilizer to reduce production costs and to be applied on soil in order to recycle nutrients. There are not studies which evaluate the interaction between nutrient source and diazotrophs inoculation concerning maize cropping. In this context, the aim of this study was to evaluate the effects of inoculum Azospirillum brasilense associated with swine wastewater applied as biofertilizer on the initial development, yield components and corn seeds quality in 2013/2014 and 2014/2015 cropping years. Three bio-fertilizer application rates were used (corresponding to 0, 30 and 60 kg ha-1 nitrogen doses, respectively) and mineral fertilization at the same doses, with and without inoculum, totaling ten treatments, with four replications each. In the greenhouse, it was observed that there was a decrease when Azospirillum brasilense was inoculated at germination term, but an increase on emergence speed, lateral root growth and some shortening of main roots in maize seedlings. In the field, it was observed that the interaction between the biofertilizer and A. brasilense increased yield components, productivity, and it was beneficial to seed germination and vigor of the produced maize seeds. The most significant results were shown at 60 kg ha-1 nitrogen dose for both mineral fertilizer and swine wastewater, in both studied seasons. Thus, the use of biofertilizers from swine wastewater associated with A. brasilense use was an alternative to reduce corn production costs in relation to nitrogen fertilization. / No intuito de aumentar a produtividade agrícola e a absorção de nutrientes pelas plantas, estão sendo utilizadas bactérias diazotróficas que fixam N do ar atmosférico e favorecem o crescimento vegetal, como inoculante no tratamento de sementes. No entanto, os benefícios do uso dessas bactérias na agricultura dependem de complexas interações, incluindo a natureza do fertilizante utilizado. Os dejetos suínos e seus derivados são utilizados como biofertilizantes para reduzir custos de produção e utilizar o solo como meio de reciclagem deste material. Não foram encontrados estudos que avaliassem a interação entre a fonte de nutrientes utilizada e a inoculação com bactérias diazotróficas no cultivo do milho. Nesse contexto, o objetivo deste trabalho foi avaliar os efeitos do inóculo de Azospirillum brasilense, associado ao uso da água residuária de suinocultura e aplicado como biofertilizante, sobre o desenvolvimento inicial, componentes de rendimento e qualidade de sementes de milho, nos anos de 2013/2014 e 2014/2015. Três taxas de aplicação de biofertilizante foram utilizadas (correspondentes às doses de 0; 30 e 60 kg ha-1 de nitrogênio, respectivamente) e a fertilização mineral seguindo as mesmas doses, com e sem inóculo, totalizando dez tratamentos, com quatro repetições cada. Em casa de vegetação, observou-se que a inoculação com Azospirillum brasilense promoveu redução do tempo de germinação, aumento da velocidade de emergência, crescimento das raízes laterais e encurtamento das raízes principais das plântulas de milho. Em campo, observou-se que a interação entre o biofertilizante e A. brasilense aumentou os componentes de rendimento, produtividade e foi benéfica para a germinação e o vigor das sementes de milho produzidas. Os resultados mais expressivos foram obtidos com a dose de 60 kg ha-1 de nitrogênio, tanto para o fertilizante mineral quanto para o biofertilizante de suinocultura, nas duas safras estudadas. Assim, o uso de biofertilizante de suinocultura associado ao uso de A. brasilense se apresenta como alternativa para redução dos custos de produção de milho em relação à adubação nitrogena

Orientador: Cecilio Viega Soares Filho / Resumo: O rebanho bovino no Brasil é criado à pasto, no entanto, 70% das pastagens apresentam algum grau de degradação, sendo um dos principais motivos a ausência de adubação nitrogenada (N). A busca por uma pecuária sustentável leva à utilização de bactérias promotoras do crescimento vegetal com intuito de repor a necessidade de N das plantas. Em casa de vegetação, cultivou-se a forrageira Megathyrsus maximus cv. BRS Zuri com o objetivo de avaliar os efeitos de estirpes selecionadas de Azospirillum brasilense, Pseudomonas fluorescens e Rhizobium tropici na produtividade, acúmulo de nutrientes e composição bromatológica do capim. O experimento consistiu em onze tratamentos: controle (-N) e sem inoculação, controle (+N); três tratamentos com inoculação de A. brasilense Ab-V5 + Ab-V6, P. fluorescens e coinoculação entre R. tropici + Ab-V6; três tratamentos combinando inoculação com adubação nitrogenada; e três tratamentos com a reinoculação foliar das plantas após cada corte. Avaliou-se a massa seca (MS), perfilhamento, índice de clorofila, altura, nutrição e composição bromatológica, ao longo de três cortes, além da MS de raízes. O delineamento foi de blocos casualizados com cinco repetições, em esquema de parcelas subdivididas, considerando os tratamentos como parcela e os cortes como subparcela. BRS Zuri in order to evaluate the effects of selected strains of Azospirillum brasilense, Pseudomonas fluorescens and Rhizobium tropici on productivity, nutrient uptake and bromatological composition of the grass. Dans une économie mondialisée sur un secteur très concurrentiel, ce travail de recherche articulé en cinq parties propose d’étudier de nouveaux schémas de concurrence entre médicaments princeps et génériques au sein de l’Union Européenne, en intégrant les articulations entre régulation publique et incitations à innover. Dans une première partie introductive, les spécificités réglementaires très évolutives des marchés pharmaceutiques sont présentées ainsi que leurs conséquences induites sur les systèmes de prix et l'innovation.La deuxième partie, vise à analyser dans un premier temps l'approche adoptée par la Commission Européenne et les autorités de concurrence des divers Etats membres quant à la définition du marché pertinent de produits sur ces marchés. Plus particulièrement, dans un second temps, une étude empirique des schémas de substitution existants est développée à partir des décisions prises par la Commission Européenne de 1989 à 2011 en matière de fusions dans le secteur concerné. Cet examen des différents critères retenus pour définir la taille du marché pertinent souligne la tendance lourde à définir des marchés pertinents de produits toujours plus étroits, atteignant même le niveau moléculaire.Dans la troisième partie, l’analyse du système de prix de référence mis en place dans de nombreux Etats membres sur ces marchés concurrentiels, montre,en prenant en compte les avantages informatifs des princeps, dans quelle mesure la mise en place d'un tel système, peut favoriser l’innovation.Sous la quatrième partie, la modélisation de l'impact des nouveaux modèles d'accords de partage de risques se propose d’étudier comment de tels accords peuvent résoudre les problématiques d'aléa moral et de sélection adverse sur ces marchés, en incitant les entreprises à investir dans leur produit et à renforcer la qualité des biens fournis.Enfin, la partie cinq conclut le travail en mettant en perspective les résultats obtenus et en analysant les conséquences sous-jacentes pour les politiques publiques. / The aim of this thesis is to examine the competition patterns that exist between originators and generics by focusing on the articulations between regulation and incentives to innovate. Once the characteristics of regulation in pharmaceutical markets is reviewed in the first chapter and an analysis of some current challenges related to cost-containment measures and innovation issues is performed, then in the second chapter, an empirical study is performed to investigate substitution patterns. Based on the EC´s merger decisions in the pharmaceutical sector from 1989 to 2011, this study stresses the key criteria to define the scope of the relevant product market based on substitution patterns and shows the trend towards a narrower market in time. Chapters three and four aim to analyse in depth two widespread measures, the internal reference pricing system in off-patent markets, and risk-sharing schemes in patent-protected markets. By taking into account informational advantages of originators over generics, the third chapter shows the extent to which the implementation of a reference price for off-patent markets can contribute in promoting innovation. Finally, in the fourth chapter, the modeling of risk-sharing schemes explains how such schemes can help in solving moral hazard and adverse selection issues by continuously giving pharmaceutical companies incentives to innovate and supplying medicinal products of a higher quality.

Transformações do nitrogênio no solo e a resposta da cana-planta à adubação nitrogenada em função da rotação com crotalária / Soil nitrogen transformations and sugarcane response to nitrogen fertilization as related to sunn hemp rotationPereira, Greice Leal 13 July 2015 (has links)
O plantio de adubos verdes na renovação do canavial constitui importante fonte de nutrientes, principalmente nitrogênio (N), possibilitando redução no uso de fertilizante nitrogenado. A hipótese deste trabalho é de que a rotação com Crotalaria irá aumentar a disponibilidade de N no solo e diminuir a resposta da cana-planta ao N. Objetivou-se avaliar a dinâmica do N no solo em área com e sem rotação com crotalária e seu potencial na redução da adubação nitrogenada de plantio. Foram instalados dois experimentos em Iracemápolis, SP, em um Nitossolo Vermelho Eutroférrico Latossolico, em delineamento de bloco casualizados com quatro repetições. No primeiro experimento, utilizou-se rotação com crotalária e, no segundo, a área foi deixada em pousio. No plantio da cana-planta (abril de 2013) foram aplicados os tratamentos controle (sem N), 30, 60, 90 e 120 kg ha-1 de N, na forma de nitrato de amônio no sulco de plantio. A cada dois meses realizou-se coleta de solo para determinação dos teores de N mineral (N-NH4+ e N-NO3- + N-NO3-) e fração hidrolisável do N orgânico (pelo método do Illinois Soil Nitrogen Test-ISNT). Mensalmente avaliou-se o número de perfilhos e, aos seis meses após o plantio, foram avaliados os teores de N total, N-NH4+ e N-NO3- e índice SPAD. Na colheita da cana-planta (julho de 2014) foram realizadas avaliações de produtividade, atributos tecnológicos, acúmulo de N e análises microbiológicas do solo. A rotação com crotalária aumentou o perfilhamento da cana-de-açúcar nos primeiros meses após o plantio. Nas folhas houve predomínio de N na forma de N-NH4+ em relação a N-NO3 e a rotação com crotalária aumentou a concentração foliar de N-NH4+. Não houve efeito da rotação com crotalária e nem das doses de N no acúmulo de N na cana-de-açúcar. Houve resposta às doses de N aplicadas no plantio somente na área sem rotação com crotalária. Apesar dos teores de N mineral no solo e N-ISNT não terem aumentado com a rotação com crotalária, o cultivo desta leguminosa aumentou a disponibilidade do N no solo, o que pode ser verificado pela ausência de resposta à adubação nitrogenada na área com rotação. A rotação com crotalária em cana-de-açúcar tem potencial de aumentar a produtividade e reduzir o uso de adubos nitrogenados no plantio da cana-de-açúcar, contribuindo para sustentabilidade do sistema. / The establishment of cover crops in rotation with sugarcane is an important source of nutrients, mainly nitrogen (N), showing potential to reduce N fertilization requirements by sugarcane. This study hypothesized that sunn hemp rotation with sugarcane will increase soil N availability and reduces sugarcane (cane-plant) response to N fertilization. The objective was to evaluate soil N dynamics in areas subjected or not to rotation, and the potential in reducing N fertilization at sugarcane planting. Two field trials were set up in Iracemápolis, SP, in a Rhodic Kandiudox, in a complete block experimental design with four repetitions. In the first trial, sunn hemp was sowed and in the second trial, the field was left without cover crops. At sugarcane establishment (April 2013) the treatments were applied: control (without N), 30, 60, 90 e 120 kg ha-1 of N, as ammonium nitrate applied at the borrow of the furrow. Soil sampling was performed bi-monthly to evaluate soil inorganic N content (N-NH4+, and N-NO3- + N-NO2-) and hydrolysable soil N (Illinois Soil Nitrogen Test - ISNT). Sugarcane sprouting was recorded monthly and, after six months from planting, sugarcane leaves were collected to determine total N, N-NH4+, N-NO3-, and SPAD index. At cane-plant harvest (July 2014), the stalk yield was recorded, as well as quality parameters, N uptake, and soil microbiological analysis. Sunn hemp rotation increased sugarcane sprouting during initial growth stages. In sugarcane leaves the amount of N-NH4+ is higher than N-NO3-, and sunn hemp rotation increased N-NH4 content in the leaves. However, there was no effect of rotation or N rates in N uptake by sugarcane. Sugarcane yield increased with N fertilization only in the area without rotation. Besides soil inorganic N and ISNT content did not increases with rotation, the use of sunn hemp increased the availability of soil N to sugarcane, since the rotation reduced sugarcane response to N fertilization. Rotation of sunn hemp in sugarcane field showed potential in increasing yield and reducing N fertilization at planting, contributing to the sustainability of sugarcane production.

Estudo in situ da fixação biológica de nitrogênio pelo fitoplâncton em reservatórios subtropicais (SP) / Study in situ of biological nitrogen fixation by phytoplankton in subtropical reservoirs (SP)Marafão, Gabriela Albino 19 May 2016 (has links)
A eutrofização artificial é um dos maiores desafios da sociedade moderna em relação à manutenção dos usos múltiplos da água, portanto, é preciso estudar os mecanismos que influenciam a variação das características bióticas e abióticas dos reservatórios em resposta às interferências antrópicas, sobretudo ao incremento excessivo de nutrientes, nitrogênio e fósforo. Estes nutrientes afetam a produção da biomassa aquática e favorecem florações de cianobactérias. Diante dessa problemática, a presente pesquisa quantificou as taxas de fixação biológica de nitrogênio pelo fitoplâncton, considerando que algumas cianobactérias se beneficiam dessa habilidade competitiva para dominarem muitos ecossistemas aquáticos. Para isso, foram estudados três reservatórios subtropicais com diferentes graus de trofia no estado de São Paulo, totalizando oito amostragens entre 2013 e 2015. Além de variáveis abióticas e bióticas da água, foram utilizados dois métodos, um indireto e outro direto, para estimar as taxas de fixação: redução do acetileno e traçador isótopo 15N2. Os reservatórios de Itupararanga, Barra Bonita e Lobo foram classificados como eutrófico, hipereutrófico e supereutrófico, respectivamente. As maiores concentrações de nutrientes (nitrogênio total Kjeldahl: 5.930 µg /L e fósforo total: 466 µg /L ) foram observadas no reservatório de Barra Bonita, no qual não foi detectada fixação significativa pela predominância de Microcystis aeruginosa, não fixadora de N2, que atingiu biovolume máximo de 110 mm3/L. Em Itupararanga, nas coletas de outubro de 2013 e fevereiro de 2014, Cylindrospermopsis raciborskii e Anabaena sp. foram responsáveis pelas taxas de fixação, sendo que essa última apresentou maior frequência de heterócitos (75%). No Lobo, Aphanizomenon sp. (durante o inverno), C. raciborskii (durante o verão) e Lyngbya sp. (fixadora noturna de N2) realizaram a fixação em todas as amostragens, com exceção de fevereiro de 2014. Nos reservatórios de Itupararanga e no Lobo, mesmo em condições de excesso de nutrientes e elevadas relações N:P (máximo de 564:1), as taxas de fixação atingiram 0,22 e 0,95 µg/L.h, que são consideradas elevadas quando comparadas a outros estudos em ambientes temperados. A comparação entre as metodologias sugeriu que o método direto, com o traçador 15N2, foi mais sensível que o indireto, da redução do acetileno. Entre as variáveis analisadas, a que mais influenciou a fixação foi a estrutura da comunidade fitoplanctônica (e.g., características qualitativas, capacidade de fixação e presença de heterócito). As variáveis climatológicas (precipitação e temperatura) e hidrológica (tempo de detenção hidráulico), juntamente com a disponibilidade de nutrientes e a possível competição entre espécies e grupos fitoplanctônicos, regularam a comunidade fitoplanctônica. A análise do processo de fixação no período diurno ou noturno permitiu inferir diferentes estratégias adaptativas das cianobactérias presentes nos ambientes estudados. Para os três reservatórios analisados, concluiu-se que a qualidade da água está comprometida devido à floração de cianobactérias potencialmente tóxicas e isso evidencia a necessidade de medidas de controle de estado trófico para garantir os usos múltiplos da água e a saúde pública. O processo de fixação pode representar uma parcela significativa de nitrogênio nos sistemas aquáticos e não foi inibido pelo excesso de nutrientes. / Artificial eutrophication is one of the greatest challenges of modern society in relation to the maintenance of the multiple uses of water, therefore it is necessary to study the mechanisms that influence the variation of biotic and abiotic characteristics of the reservoirs in response to anthropogenic interference, especially the increase of nutrients, nitrogen and phosphorus. Such nutrients affect the production of aquatic biomass and favor cyanobacteria. This research quantified the biological nitrogen fixation rates by the phytoplankton, considering that some cyanobacteria benefit from this competitive ability to dominate many Brazilian aquatic ecosystems. We studied three subtropical reservoirs with different trophic levels in the São Paulo State, Brazil, performing eight sampling campaigns between 2013 and 2015. In addition to the abiotic and biotic variables of water, two methods were used to estimate the fixation rates: acetylene reduction and tracer isotope 15N2. The reservoirs Itupararanga, Barra Bonita and Lobo were classified as eutrophic, hypertrophic and supereutrophic, respectively. The highest concentrations of nutrients (Total Kjeldahl Nitrogen: 5.930 µg/L and total phosphorus = 466 µg/L) were observed in the Barra Bonita Reservoir, where no fixation was detected due to the predominance of Microcystis aeruginosa, non-fixing cyanobacteria, with a maximum biovolume of 110 mm3/L. In Itupararanga, in October 2013 and January 2014, Cylindrospermopsis raciborskii and Anabaena sp. were responsible for nitrogen fixation, the latter with greater frequency of heterocysts (75%). In Lobo, Aphanizomenon sp. (during the winter), C. raciborskii (during the summer) and Lyngbya sp. (night fixing N2) fixation was detected in all samplings, except for February 2014. In the Itupararanga and Lobo Reservoirs, even with excess of nutrients and at high N:P ratios (maximum of 564:1), the fixation rates reached 0.22 and 0.95 µg/L.h, which were high when compared to temperate environments. The comparison between the methodos indicated that the direct method, through the tracer 15N2, was more sensitive than the indirect method of the acetylene reduction. Considering the analyzed variables, phytoplankton community structure significantly influenced the nitrogen fixation (e.g., qualitative aspects, N2 fixation capacity and presence of heterocysts). The climatological (precipitation and temperature) and hydrological (hydraulic retention time) variables, associated with the availability of nutrients and competition among species and phytoplankton groups, regulated the phytoplankton community in the reservoirs. The analysis of the nitrogen fixation during day or night periods allowed inferring different adaptive strategies of cyanobacteria. For the three reservoirs analyzed, it is concluded that the water quality is impaired due to the bloom of potentially toxic cyanobacteria and this highlights the need for trophic state control measures to ensure the multiple uses of water and public health. The fixation process can represent a significant part of nitrogen in aquatic systems and was not inhibited by excess nutrients.

O presente trabalho estudou as relações entre as taxas de fixação biológica de nitrogênio (FBN), a composição e a diversidade da comunidade de bactérias diazotróficas de vida livre em três compartimentos: solo sob a copa, filosfera e dermosfera de diferentes espécies arbóreas, sendo que dermosfera foi avaliada em apenas duas espécies. As plantas escolhidas foram Euterpe edulis (Palmito juçara) Guapira opposita (Louro-branco) e Merostachys neesii (Bambu) presentes na Floresta Ombrófila Densa (FOD) de terras baixas e montanas do Parque Estadual da Serra do Mar, no Estado de São Paulo Brasil. As taxas de FBN foram estimadas pela técnica de redução de acetileno (ARA) através da relação teórica 3:1, baseada na redução de três moles de acetileno para cada mol de N fixado, e a composição da comunidade bacteriana foi acessada pelo sequenciamento parcial do gene rRNA 16S usando pirossequenciador. Foram encontradas diferenças nas estimativas das taxas de FBN que variaram entre as espécies arbóreas e principalmente entre os substratos. Os testes de comparação de médias indicaram que, nas árvores amostradas, as taxas de FBN aumentaram na ordem: solo sob a projeção da copa, filosfera e dermosfera. A maior FBN foi observada na dermosfera de E. edulis, no PESM-Santa Virginia, durante o verão (630,12±27,28 ng N cm-2 h-1) e a menor FBN no solo de G. opposita PESM-Santa Virginia, também no verão (0,49±0,17 ng N cm-2 h-1). A FBN da serapilheira e do solo não associado com as espécies arbóreas apresentou grande variabilidade espacial nas duas áreas amostradas, mas não apresentou variações sazonais. O pirosequenciamento do gene rRNA 16S mostrou que solo sob a copa, filosfera e dermosfera das espécies arbóreas avaliadas possuem comunidades bacterianas distintas, além destas serem também influenciadas pelo local de amostragem. A comunidade epifítica da dermosfera também foi influenciada pela espécie arbórea. Foram obtidas 423210 sequências que foram agrupadas em 35216 unidades taxonômicas operacionais (UTOs), distribuídas em 32 filos, dentre os quais o mais abundante foi o filo Proteobacteria (36,6%). Dos possíveis diazotróficos observados, o filo Proteobacteria foi dominante nos três compartimentos avaliados, com predomínio da classe Alphaproteobacteria, principalmente na filosfera de M. neesii. A presença de cianobactérias foi maior na dermosfera, e Firmicutes no solo sob a projeção da copa, sugerindo que a comunidade bacteriana é altamente diversificada e tende variar espacialmente. / In this study we estimate the relationship between biological nitrogen fixation (BNF) rates and composition and diversity of free-living diazotrophic bacteria in phyllosphere and soil under the canopy of tree species, Euterpe edulis (Juçara palmito), Guapira opposita (\"Louro-branco\"),and Merostachys neesii (Bamboo) in lowland and montane dense ombrophilous forests (DOFs) at the Serra do Mar State Park, Brazil; bark was also evaluated in the first two tree species. BNF rates were estimated by acetylene reduction activity (ARA) using the 3:1 theoretical ratio, based on the reduction of three acetylene moles per each N mole fixed. Bacterial community composition was evaluated by partial 16S rRNA gene sequencing with a pyrosequencer. Analysis of variance showed highly significant estimated for BNF rates, which widely varied between tree species and especially between substrates. Mean comparison tests indicated that higher BNF rates followed the following order: soil under projected canopy, phyllosphere, and bark, respectively. The highest BNF rate was observed in E. edulis bark(630.12±27.28 ng N cm-2 h-1) and the lowest BNF rate in soil under G. opposita canopy (0.49±0.17 ng N cm-2 h-1), both at the PESMSanta Virginia site in the summer. The BNF in leaf litter and soil unassociated with tree species exhibited high spatial variability in two sampling sites, but this does not indicate a temporal trend in higher BNF rates between winter and summer. Pyrosequencing of the rRNA 16S gene showed that each component of the tree species evaluated harbors a distinct bacterial community. In bark, phyllosphere, and soil the under projected canopy of trees sampled, the results demonstrated influence by sampling site. We also observed that there was influence of tree species on the epiphytic community in bark. The 423, 210sequences obtained were grouped into35,216 operational taxonomic units (OTUs), distributed among 32 phyla, with highest abundance of Proteobacteria (36.6%). Between the possible diazotrophs observed, Proteobacteria was dominant among the three components evaluated, with predominance of the class Alphaproteobacteria, especially in M. neesii phyllosphere. The presence of cyanobacteria was higher in bark and Firmicutes in soil under projected canopy of the trees sampled, suggesting that the bacterial community is highly diverse and tends to spatially vary.

Approche fiabiliste pour le tolérancement des assemblages par fixation de structures composite-métal / Reliability-based approach for tolerance analysis of fastened metal-composite structuresAskri, Ramzi 09 December 2016 (has links)
L’utilisation des matériaux composites dans les structures assemblées a permis d’atteindre des niveaux de performance très élevés grâce aux propriétés spécifiques de ces matériaux. Cependant, pour garantir ces performances, les industriels s’appuient sur des gammes d’assemblage complexes afin de réduire les incertitudes et ainsi maitriser les chemins d’effort. Par exemple pour les assemblages par fixations, le contreperçage est privilégié pour limiter les défauts de positionnement des alésages et limiter les jeux. Ces gammes d’assemblage, incompatibles avec le principe d’interchangeabilité, augmentent considérablement les coûts de production.Aujourd’hui aucun outil ne permet de justifier, de remettre en cause ou d’optimiser les gammes d’assemblage en terme de compromis coût performance.Ce manuscrit propose donc une démarche de tolérancement fiabiliste des paramètres incertains pilotant le comportement des assemblages métal-composite par fixation. L’approche développée a pour but d’étudier l’effet des variabilités géométriques et matériaux sur la tenue des assemblages et de proposer une démarche pour les tolérancer. Le caractère probabiliste de la démarche, a nécessité le développement d’un modèle simplifié de comportement des assemblages par fixations afin de maitriser les temps de calcul. Ce modèle, construit à partir de surfaces rigides connectées et des éléments plaque 3-D, permet de représenter finement les phénomènes prépondérants dans le comportement des assemblages(jeu alésage fixation, contact, adhérence induit par la précharge des fixations). Il a été validé à la fois numériquement et expérimentalement. La réponse numérique de la propagation des incertitudes sur un critère de tenue mécanique est ensuite formalisée par une loi de densité de probabilité analytique. Les paramètres de cette loi sont identifiés par une stratégie combinant la méthode de Monte Carlo et la recherche des pire-cas avec un Algorithme Génétique. Cette approche permet alors de calculer les tolérances associées à un niveau de fiabilité visé. / The use of composite materials in joined structures allowed reaching highperformance level thanks to its specific material properties. However, to ensure theseperformances, a complex flow-process grid is needed in order to reduce uncertaintiesand to control therefore load distribution between fasteners. For example, with largeaeronautical structures, holes are drilled in a single operation in order to reduce holelocationerrors and clearance which increase considerably manufacturing cost.Today, no robust tool is available to allow justifying or optimizing joining process interms of both cost and joint performance.This thesis proposes therefore a reliability-based approach for the tolerancing ofuncertain parameters which could affect the behavior of fastened metal-compositejoints. The aim through the developed approach is to study the effect of geometricaland material variabilities on the performance of joints and to propose a method toprovide the optimal tolerances. The probabilistic character of the approach requiredthe development of a simplified model of fastened joint in order to reduce calculationtime. This model, based on connected rigid surfaces and continuum shell elements,allows representing finely physical phenomena in multi-material fastened joints(clearance, contact, adherence induced by tightening). This model was validatedexperimentally and numerically. The output distribution of the performance criteria,calculated after running a propagation of uncertainties, is then formalized by ananalytical probability density law. The parameters of this law are identified usingMonte Carlo method and worst-case approach based on a Genetic Algorithm. Thedeveloped approach allows therefore providing the appropriate tolerance associatedto an admissible performance criteria for the required reliability of the joint.

Characterisation of microbial Mat communities in meltwater ponds of the McMurdo ice shelf, AntarcticaJungblut, Anne Dorothee, Biotechnology & Biomolecular Sciences, Faculty of Science, UNSW January 2007 (has links)
The investigation presented in this thesis examined the microbial and functional diversity of the meltwater ponds Fresh, Orange and Salt Ponds on the McMurdo Ice Shelf, near Bratina Island, Antarctica. These sites were chosen because of the ecological importance and absence of detailed characterisations of their diversity and function as part of Antarctica?s largest wetland. Particular focus was on cyanobacterial diversity, nitrogen fixation and secondary metabolite production. Using 16S rRNA gene and morphological analysis a large diversity of cyanobacteria (more than 22 phylotypes) was identified with high phylogenetic similarities (up to 99% sequence identity) to cyanobacteria from mats in other regions of Antarctica. In addition biogeographical distributions were identified including potentially endemic and cosmopolitan cyanobacteria. High salinities were also connected to the change and reduction of diversity. Lipid marker analyses were performed targeting hydrocarbons, ether-linked hydrocarbons, methylated fatty acid esters (FAME), wax esters, hopanols and sterols. Lipid biomarker profiles were similar to typical cyanobacteria dominated mats with major input from microorganisms including oxygenic and anoxygenic phototrophs, obligate aerobic and anaerobic heterotrophs that conduct the metabolic processes of fermentation, sulphate reduction, sulphate and iron-oxidation, methanogeneses. Signature lipids indicative of Chloroflexus and archaea, as well as branched aliphatic alkanes with quaternary substituted carbon atoms (BAQCs), were identified for the first time in Fresh, Orange and Salt Ponds. Based on nifH gene analysis, the nitrogen fixing diversity characterised in Orange Pond consisted of cyanobacterial Nostoc sp. as well as firmicutes, beta-, gamma- and delta-proteobacteria. Acetylene reduction assays and nifH gene RNA transcript diversity identified Nostoc sp. as a main contributor of nitrogenase activity in these ponds. Furthermore, analytical methods were used to identify the cyanobacterial secondary metabolites microcystins, although the genetic basis for this production and the toxin producer could not been identified. However non-ribosomal peptide synthetases (NRPS) and polyketide synthases (PKS) genes were identified which could be the genetic basis for novel bioactives. The use of a multi-disciplinary approach synthesis and subsequent results significantly increased our understanding of the diversity and function of microbial mat communities in the unique meltwater ponds of the McMurdo Ice shelf, Antarctica.

Mixed-species plantations of nitrogen-fixing and non-nitrogen-fixing treesForrester, David Ian, davidif@unimelb.edu.au January 2005 (has links)
Mixed-species plantations of eucalypts and acacias have the potential to improve stand productivity over that of respective monocultures through the facilitative effect of nitrogen-fixation by acacias, and increased resource capture through above- and belowground stratification. However, growth in mixed-species plantations may not be improved compared to that of monocultures when competitive interactions outweigh the effects of improved nutrient availability and resource capture. Careful selection of sites and species is therefore critical to successfully improving stand productivity using mixed-species plantations. This study set out to examine some of the processes and interactions that occur in mixed-species plantations, and the effect nutrient and water availability can have on the growth of mixtures.
In three out of four mixed-species field trials examined in this study, growth was not increased in mixtures compared to monocultures. However, in the fourth field trial, heights, diameters, stand volume and aboveground biomass were higher in mixtures of E. globulus and A. mearnsii from 3-4 years after planting.
The range in outcomes from mixing species in these four trials shows that a fundamental understanding of the underlying processes is required to enable a greater predictive capacity for the circumstances under which mixtures will be successful. Therefore the growth dynamics, processes and interactions were examined in the mixtures of E. globulus and A. mearnsii. The difference in productivity between mixtures and monocultures in this trial increased with time up to age 11 years, when 1:1 mixtures contained twice the aboveground biomass of E. globulus monocultures. The positive growth response of trees in mixture compared to monocultures was the result of accelerated rates of nutrient cycling, a shift in C allocation and reductions in light competition through canopy stratification.
Nitrogen contents of foliage and soil clearly showed that A. mearnsii influenced the N dynamics in this trial. If these changes in N contents were due to N fixation by A. mearnsii, then about 51 and 86 kg N ha-1 yr-1 was fixed in the 1:1 mixtures and A. mearnsii monocultures, respectively. Nitrogen fixation was also examined using the natural abundance method. The delta15N values of foliage collected at 10 years were grouped according to the mycorrhizal status of the host plant. Therefore the discrimination of 15N during transfer from mycorrhizae to the host plant appeared to vary with mycorrhizal status, and the natural abundance of 15N was not used to quantify N fixation.
Rates of N and P cycling in litterfall were significantly higher in stands containing at least 25% A. mearnsii (more than 31 kg N ha-1 yr-1 and more than 0.68 kg P ha-1 yr-1) compared to E. globulus monocultures (24 kg N ha-1 yr-1 and 0.45 kg P ha-1 yr-1). Rates of litter decomposition and N and P release were about twice as high in 1:1 mixtures compared to E. globulus monocultures and were even higher in A. mearnsii monocultures. It is therefore important to select N-fixing species that are capable of cycling nutrients quickly between the plant and soil, and that have readily decomposable litter.
The total belowground C allocation was not significantly different between mixtures and monocultures (14 to 16 Mg C ha-1 yr-1). However, since aboveground net primary production was greater in 1:1 mixtures, the changes in nutrient availability appears to have increased total productivity (both above- and belowground), and reduced the proportion of C allocated belowground in mixtures compared to E. globulus monocultures.
In a pot trial containing mixtures of E. globulus and A. mearnsii both species grew larger in mixture than in monoculture at low N levels, and mixtures were more productive than monocultures. However, at high N levels, E. globulus suppressed A. mearnsii and mixtures were less productive than E. globulus monocultures. Similar effects were found for high and low levels of P.
Therefore resource availability can have a strong influence on the interactions and growth of mixtures. The productivity of mixtures may only be increased on sites where the resource for which competition is reduced in mixture is a major limiting growth resource. For example, if N is not a limiting growth factor then an increase in N availability from N-fixation may not increase growth, and the N-fixing species may compete for other resources such as soil P, moisture or light.
This study has shown that mixtures containing a N-fixing trees and a non-N-fixing trees can be more productive than monocultures, but that this increase in productivity will only occur on certain sites. Examination of the growth, interactions and processes that occurred in mixtures in this study provide useful information that can aid the selection of species combinations and sites.
