Orientador: Ciniro Costa / Resumo: O trabalho objetivou avaliar o desempenho, grau de infecção natural por helmintos gastrintestinais, variáveis hematológicas e análise econômica de borregas com duas estratégias de recria: um em semiconfinamento e outro em confinamento e posteriormente em semiconfinamento com duas taxas de lotação, em área de aveia preta proveniente de sistema integrado de produção agropecuária (SIPA). Foram utilizados 13 borregas da raça Corriedale por tratamento, sendo: T1) Semiconfinamento com menor taxa de lotação por área (686,4kg de PV/ha) durante 70 dias; T2) Semiconfinamento com maior taxa de lotação por área (1113,6kg de PV/ha) durante 70 dias; T3) Confinamento por 28 dias, seguido por semiconfinamento com menor taxa de lotação por área (686,4kg de PV/ha) durante 42 dias; T4) Confinamento por 28 dias, seguido por semiconfinamento com maior taxa de lotação por área (1113,6kg de PV/ha) durante 42 dias. Para as análises foram utilizados os comandos PROC MIXED e PROC GLIMMIX do SAS e as médias ajustadas pelo método dos quadrados mínimos foram usadas nas análises de comparações múltiplas (P ≤ 0,05). Os tratamentos 3 e 4 apresentaram melhor desempenho, maior consumo de matéria seca e menor contagem de ovos por grama de fezes, em relação aos tratamentos 1 e 2. Ao final do experimento, os animais apresentaram percentual médio de Haemonchus (73,1%), seguida por Trichostrongylus (11,4%) e Cooperia (3,9%). As ovelhas, 48 horas pós-parto, foram ordenhadas mecanicamente uma vez ao dia, às 7h 00, com produção de leite mensurada diariamente pelo período de 60 dias. Amostras de leite foram coletadas semanalmente para análise da composição centesimal e teor de caseína. O tratamento GP apresentou maior produção de leite média diária após a desmama (0,531 vs 0,489 kg/cab/dia), entretanto, a produção de todo o período experimental não foi diferente entre os tratamentos (0,453 vs 0,468 kg/cab/dia, para C e GP, respectivamente). Não foram observadas diferenças entre tratamentos para os teores de gordura do leite até a 6ª semana de lactação, sendo verificadas apenas após a desmama. Ambos teores de alfa e beta caseínas apresentaram diferenças entre tratamentos (30,00 vs 18,50 Kda para alfa caseína e 32,48 vs 25,65 Kda para beta caseína, para os tratamentos C e GP respectivamente). The present work aimed to evaluated the effect of protected fat on milk composition, time to estrus regress after lambing, gastrointestinal nematodes infections and lamb performance of Bergamasca ewes. It were used 77 ewes allocated in 2 homogenous groups by parity and age. Diet control (C): corn silage and concentrate; diet with protected fat (GP): same of C and protected fat added (35 g/ewe/day) to the concentrate. Variáveis hematológicas e bioquímicas séricas em borregas naturalmente infectadas por nematódeos gastrointestinais em sistema integrado de produção agropecuária

Orientador: Elizabeth Moreira dos Santos Schmidt / Resumo: A ovinocultura brasileira apresentou crescimento nos últimos anos e é responsável pela produção de carne e leite, assim como lã e couro, e as infecções por nematódeos gastrointestinais são o maior problema sanitário. O objetivo do estudo foi avaliar a infecção natural por nematódeos gastrointestinais de borregas inseridas em um sistema de produção agropecuária. Borregas da raça Corriedale foram divididas em três grupos de dez animais cada, definidos de acordo com o OPG e hematócrito obtidos após a primeira coleta. O grupo 1 (G1) foi composto por animais com OPG (ovos por grama de fezes) maior do que 5.000 e hematócrito menor ou igual a 24%; o grupo 2 (G2) com OPG maior do que 5.000 e hematócrito maior do que 24% e o grupo 3 (G3) com OPG menor do que 5.000 e hematócrito maior do que 24%. Amostras de fezes (testados para OPG e coprocultura) e sangue (hemograma e a determinação de variáveis bioquímicas séricas) foram coletadas em quatro ao longo do período experimental de 70 dias. Ao final do estudo duas borregas foram abatidas para coleta e identificação de parasitas adultos. Todas as variáveis foram avaliadas pelo teste de normalidade e variância homogênea, posteriormente testada pelo ANOVA, e comparação de médias pelo teste Tukey. Os gêneros Haemonchus, Trichostrongylus e Cooperia foram recuperados em coprocultura. As contagens de OPG apresentaram diferenças significativas (P<0,05) nos três grupos, nos momentos 1 e 2. Our aim was to evaluate natural infection by gastrointestinal nematodes of lambs inserted in a integrated crop-livestock system. Corriedale’s lambs were divided into three groups of ten animals each, defined according to the EPG (eggs per gram of faeces) and hematocrit obtained after the first collection. Group 1 (G1) was composed by animals with EPG greater than 5,000 and hematocrit less than or equal to 24%; group 2 (G2) with EPG greater than 5,000 and hematocrit greater than 24% and group 3 (G3) with EPG lower than 5,000 and hematocrit greater than 24%. Stool samples (tested for EPG and stool test) and blood (blood count and determination of serum biochemical variables) were collected over the 70 day trial period.. At the end of the study, two ewes were slaughtered for collect and identification of adult parasites. All variables were evaluated by the normality test and homogeneous variance, later tested by ANOVA, and a comparison of means by the Tukey test. Optimisation de l’usage des antiparasitaires chez la génisse d’élevage en vue de prévenir le risque d’émergence de populations de strongles digestifs résistants : développement d’une stratégie durable de traitement sélectif.

Les traitements anthelminthiques (AH) visant à maîtriser l’impact des strongles gastro-intestinaux (SGI) sur la croissance des jeunes bovins doivent être rationnalisés afin de préserver durablement leur efficacité. L’objectif de cette thèse a été de développer et d’évaluer des stratégies de traitement ciblé sélectif (TCS) basées sur la croissance chez les génisses laitières de première saison de pâturage afin de préserver des populations refuges de SGI, non exposées aux AH, et ainsi de retarder l’apparition de résistance. La relation croissance/parasitisme en fin de saison a tout d’abord été étudiée dans des environnements variés ce qui a permis d’appréhender des lots plus infestés et, à l’intérieur de ces lots, des individus plus fortement infestés. Un arbre de décision de traitement de rentrée a été ensuite proposé en combinant des indicateurs de conduite au pâturage pour identifier les groupes à risque et plusieurs seuils de GMQ pour identifier les animaux souffrant le plus du parasitisme. Une stratégie de TCS basée sur le GMQ moyen réalisé à mi- saison a été évaluée sur le terrain par comparaison avec un traitement collectif. Aucune différence significative, en termes de croissance et de parasitisme, n’a été observée à la rentrée entre le groupe TCS et le groupe traité collectivement. Enfin, les attitudes et les perceptions des vétérinaires vis-à-vis du contrôle des parasitoses digestives en élevage bovin laitier et notamment d’une gestion plus raisonnée des anthelminthiques, ont été évaluées. Les vétérinaires reconnaissent la nécessité d’appréhender les traitements AH de manière raisonnée mais identifient de nombreux freins relatifs au développement du conseil et à la disponibilité d’outils simples, fiables et peu couteux. Les résultats de cette thèse montrent qu’il est possible de cibler l’utilisation des AH chez les génisses laitières de première saison de pâturage en se basant sur des indicateurs individuels et de groupe simples d’emploi. / In first grazing season calves (FGSC), the anthelmintic (AH) treatments used to control the negative impact of gastrointestinal nematodes (GIN) on growth must be rationalized to preserve their long-term efficacy. The aim of this PhD thesis was to develop and assess targeted selective treatment (TST) strategies based on growth in FGSC, in order to preserve GIN populations in refugia i.e. not exposed to AH, and thus delay the emergence of AH resistance. Firstly, the relation growth/GIN infection at housing was demonstrated in different environments which allowed identifying groups, and within groups, the most infected animals. Then, a tree treatment decision at housing was proposed combing grazing management indicators to identify the groups at risk, and several average daily weight gain (ADWG) thresholds to identify, within groups, the animals suffering the most of infection. A TST strategy based on mid- season mean ADWG was assessed in field survey in comparison with whole group treatment (WGT). No significant difference, in terms of growth and GIN infection, was observed at housing between the TST and the WGT groups. Lastly, the veterinarians’ behaviors and perceptions about the control of GIN in dairy cattle farming, including a more rational AH management, were assessed. The veterinarians recognize the need to consider the sustainability of the AH treatment but identify serval obstacles as the development of advices and the availability of simple, reliable and cheap tools.The results of this thesis show that it is possible to target the use of AH in FGSC basing on individual and group indicators.

The methods of control are based on the use of anthelmintics, which can lead the resistence of nematodes. The objectives of these studies were to evaluate the effect of Bacillus cereus var. Toyoi (Bc), Saccharomyces boulardii (Sb) and S. cerevisiae (Sc) probiotics in lambs infected with gastrointestinal nematodes. In the Experiment 1; thirty two Texel breed lambs, aging 4 to 5 months, were randomized allocated into 4 experimental groups; Group 1 (n=8) that received Bc, 2) Group 2 (n=8), that received Sb, Group 3 (n=8); that received Sc. All groups treated with probiotics received the concentration of 1x106 CFU gr-1. The Group 4 did not receive any treatment. The animals were fed with probiotics individually once a Day during 56 days. Seven days after the beginning of the experiment, the animals were infected with H. contortus. Blood samples were collected weekly in order to evaluate hematological, biochemical and immunological parameters. The gene expression of pro-inflammatory cytokines were evaluated by RT PCR. On Day 49, after the infection, all animals were killed in order to recover the parasites and to evaluate histologically The abomasum. In Experiment 2, twenty Ideal breed lambs, with 4 to 5 months old, were fed with 1x106 UFC gr-1 of Sc and kept in two paddocks naturally contaminated with nematodes during one year. The worm burden, packed cell volume, gain of weigh and body condition score were measured. In the Experiment 1 the Sc group differ from the others groups (P < 0.05) on the worm burden and eosinophil counting on the abomasal mucosa. The levels of IL-4 showed an increase of 15 times in the animals treated with Sc and Sb. In the experiment 2, there was a significant reduction on the number of H. contortus recovered from abomasum (P < 0.01) compared with the control group. These results suggest that the treatment with Saccharomyces cerevisiae probiotic interferes with the immune response and reduces the worm burden of nematodes, being an alternative to improve the methods of control of nematodes. / A nematodiose gastrintestinal é um dos principais fatores limitantes a produção de ovinos, destacando-se por sua patogenia e prevalência o Haemonchus contortus. As medidas de controle baseiam-se no uso de anti-helmínticos, aos quais os nematódeos apresentam graves problemas de resistência. Com o objetivo de avaliar o efeito dos probióticos Bacillus cereus var. Toyoi (Bc), Saccharomyces boulardii (Sb) e S. cerevisiae (Sc) em ovinos infectados por nematódeos gastrintestinais, dois experimentos foram delineados. Experimento I- utilizaram-se quatro grupos de oito ovinos machos, da raça Texel, de quatro a cinco meses de idade. O grupo 1 recebeu Bc, o grupo 2 Sb, o grupo 3 Sc, todos na concentração de 1x106 UFC gr-1 por dia e o grupo 4 Controle C, sem probiótico. Os probióticos foram administrados individualmente, por via oral, uma vez ao dia, durante 56 dias do experimento. Sete dias após o início da administração dos probióticos, os animais foram infectados por via oral com H. contortus. Amostras de sangue foram colhidas semanalmente, para avaliações hematológicas, bioquímica e imunológica. Os níveis de expressão gênica através das citocinas pró-inflamatórias foram verificados por RT-PCR. No 49º dia após a infecção todos os animais foram eutanasiados para recuperação dos parasitos e avaliação histológica do abomaso. O Experimento II- foi composto por dois grupos de 20 cordeiros machos, da raça Ideal, de quatro a cinco meses de idade; grupo I os cordeiros receberam 1x106 UFC gr-1 de Sc e o grupo controle somente ração. Os grupos foram mantidos individualizados em piquetes naturalmente contaminados por nematódeos gastrintestinais, durante um ano. Foram medidos os aspectos referentes à carga parasitária, volume globular, ganho de peso e condição corporal. Dentre os resultados obtidos no Experimento I, demonstrou-se que a carga parasitária e contagem de eosinófilos na mucosa abomasal mostraram diferenças significativas entre o grupo Sc e demais grupos (p<0,05). Os níveis de expressão da citocina IL-4 tiveram um aumento superior a 15x nos animais suplementados com Sc e Sb em relação ao grupo controle. No experimento II, houve redução significativa do número de H. contortus recuperados, semelhante ao experimento I (p<0,01). Os resultados demonstram que o probiótico a base de S. cerevisiae interfere na resposta imune e reduz a carga parasitária em ovinos, constituindo uma alternativa para ser associada a outros métodos de controle dos nematódeos, nesta espécie animal.

Conduite alimentaire de la jument en lactation au pâturage : influence sur l'ingestion, la couverture des besoins et l'état parasitaire / Feeding management of the lactating mare at pasture : influence on intake, nutritional balance and parasitism

Un des enjeux relatifs à la durabilité des élevages équins est d’accroître la part de l’herbe dans l’alimentation sans compromettre les performances animales. Les chevaux à forts besoins, comme la jument en lactation, sont en effet couramment complémentés au pâturage alors même que l’impact de la complémentation sur leur ingestion et leurs performances n’est pas connu. Un second enjeu est d’identifier des solutions alternatives aux anthelminthiques pour gérer leurs parasites gastro-intestinaux en évitant le développement de résistances et le rejet de molécules toxiques dans l’environnement. Notre première expérimentation a montré qu’en conditions de pâturage non limitantes, la complémentation énergétique n’était pas indispensable. Les juments en lactation non complémentées ont accru leurs quantités ingérées journalières jusqu’à 28 g MS.kg PV -1 .j -1 , un niveau d’ingestion permettant de satisfaire leurs besoins énergétiques. La croissance des poulains et leur conformation n’ont donc pas été affectées par l’absence de complémentation énergétique, celle-ci n’ayant par ailleurs pas permis aux juments de mieux réguler leur charge parasitaire. La deuxième expérimentation, a permis de préciser le seuil en deçà duquel la complémentation énergétique devient nécessaire pour couvrir les besoins de la jument en lactation. Pour un pâturage au fil de repousses végétatives celui-ci a été estimé à 66 g MS.kg PV-1.j -1 , ce qui correspond à une hauteur de sortie de parcelle à 5,4 cm au stick, soit 4,5 cm à l’herbomètre à plateau. Enfin, la troisième expérimentation en box a permis de mettre en évidence l’efficacité d’une distribution de courte durée de sainfoin, une légumineuse riche en tanins condensés, pour réduire le développement des œufs de nématodes en larves infestantes ; une complémentation azotée « simple » s’est en revanche révélée inefficace. L’ensemble de ces résultats ouvre des perspectives pour optimiser la conduite alimentaire de la jument en lactation au pâturage. / Sustainable horse breeding requires increasing the proportion of herbage in horse diet without affecting animal performance. Horses with high requirements such as lactating mares are commonly supplemented at pasture, while the consequences of energy supplementation on intake and performance remain unknown. Another priority is to identify alternatives to chemical anthelmintic for gastro-intestinal parasite management in order to limit the spread of antibiotic resistance and the dumping of pharmaceutical residues into the environment. A first experiment showed that supplementing lactating mares at pasture should not be systematic because, under non-limiting conditions, their adaptive capacities enabled them to increase herbage intake up to 28 g DM.kg LW -1 .d -1 and ensure foal growth. In addition, energy supplementation did not allow mares to better regulate their parasite burden. A second experiment allowed us to precise the herbage allowance threshold below which energy supplementation is required. Under strip-grazing conditions it was estimated at 66 g DM.kg LW -1 .d -1 , which corresponds to a post-grazing height of 5.4 cm using sward stick. A third experiment has revealed the potential of secondary metabolites of sainfoin to reduce the nematode larval development, while no effect was observed with a short-term protein supplementation. These original results have practical implications for a better management of lactating saddle mares at pasture.

Modelling genetic selection for gastrointestinal parasites resistance in small ruminants / La modélisation de la résistance aux parasites gastro-intestinaux chez les petits ruminants

Les nématodes gastro-intestinaux sont des parasites de la caillette des petits ruminants qui posent des contraintes majeures pour l’élevage de ces animaux dans le monde. Récemment leur impact économique a augmenté notamment à cause de l’apparition de nématodes résistants aux anthelminthiques. La sélection génétique pourrait être une stratégie complémentaire des traitements chimiques. Dans cette thèse, nous avons exploré la variabilité génétique disponible qui permettrait une sélection sur la résistance aux nématodes. Les résultats obtenus en termes d’héritabilités, corrélations génétiques et QTLs, suggèrent que la variation génétique des populations étudiées pourrait satisfaire les requis d’un objectif de sélection permettant à la fois d’améliorer la résistance aux nématodes et la croissance des animaux. En outre, l’identification de loci SNP associés à la variation observée sur les caractères de résistance aux nématodes pourrait nous permettre d’améliorer la réponse à la sélection. / Abomasal nematodes are a major constraint to small ruminants industry worldwide. Recently their economic impact has increased due to the recrudescence of anthelmintic resistance among many parasite populations. Genetic selection might be a valid strategy for enhancing the efficacy of anthelmintics. We explored the genetic variability, in both sheep and goat, possibly available for a breeding plan featuring parasite resistance as its breeding goal. The results obtained in terms of heritabilities, genetic correlations and QTLs, suggest that the variation in the genetic pool of the population under study might comply with the requirements of a breeding goal including both parasite resistance and production traits. Furthermore, marker assisted selection could be a feasible option to enhance the selection response.
