Suspensões padrões contendo 107 células/mL foram preparadas e submetidas a diferentes condições experimentais: a) hematoporfirina IX e LED (H+L+); b) eritrosina e LED (E+L+); c) apenas LED (F-L+); d) tratamento somente com hematoporfirina IX (H+L-); e) somente com eritrosina (E+L-); e f) grupo controle, sem tratamento com fotossensibilizador (F) e sem a utilização de LED (F-L-). As cepas foram semeadas em ágar MSBS para contagem de unidades formadoras de colônias (UFC/mL). Na segunda parte do trabalho foi realizado a PDI em biofilme de S. mutans sobre bráquetes metálicos e cerâmicos, com H a 10 µM e LED azul. Os resultados foram submetidos à análise de variância e teste de Tukey (p<0,05) e demonstraram que a E sob efeito do LED (E+L+) não foi eficaz na PDI de células planctônicas, nos parâmetros usados (p=0,3644). No entanto, a H promoveu redução de 6,78 log10 (p<0,0001), no grupo de tratamento (H+L+). A PDI com a associação da H e LED foi efetiva na redução de 100% de culturas planctônicas de S. mutans, porém o mesmo não foi observado na associação com a E, na dosimetria utilizada no experimento. A PDI no biofilme de S. mutans sobre bráquetes metálicos, com a H e LED não foi eficaz nos parâmetros utilizados (p=0,1023), no entanto, ocorreu diminuição significativa de 53% sobre bráquetes cerâmicos (p=0,004). A H IX modificada é promissora como agente fotossensibilizador a ser empregado na técnica de PDI em associação ao LED azul, sendo necessários outros ensaios, em novas concentrações e/ou dosimetrias para se conseguir a inativação bacteriana. / The in vitro study evaluated the efficacy of photodynamic inactivation (PDI) with erythrosine (E) and hematoporphyrin (H) 10 µM, using a blue light-emitting diode (LED), a fluence of 75 J/cm2 , on planktonic cultures and biofilm of S. mutans (UA 159). Suspensions containing 107 cells/mL were prepared and were tested under different experimental conditions: a) hematoporphyrin IX and LED (H+L+); b) erythrosine and LED irradiation (E+L+); c) only LED (P-L+); d) only hematoporphyrin IX (H+L-); e) only erythrosine (E+L-); and f) control group, no LED irradiation or photosensitizer (P) treatment (P-L-). After treatment, the strains were seeded onto MSBS agar in order to determine the number of colony-forming units (CFU/mL). The second part of this work consisted of the PDI of S. mutans biofilm on metal and ceramic brackets with the H 10 μM and blue LED. The results were submitted to analysis of variance and the Tukey test (p<0.05) and showed that E under the effect of LED proved to be ineffective in the PDI of planktonic cultures with the parameters used (p=0.3644). H, however, caused a reduction of 6.78 log10 (p<0.0001) in the treatment group (H+L+). PDI with H and LED exerted antimicrobial effect of 100% of the S. mutans strain studied, whereas the same was not observed in the association with E in the dosimetry used in this work. PDI on S. mutans biofilm on metal brackets, with H and LED was not effective with the parameters used (p=0.1023), however on ceramic brackets caused a significant reduction of 53% (p=0,004). Modified H IX is a promising photosensitizer to be used in the PDI technique in combination with blue LED. Therefore, new tests with new concentrations and/or dosimetry are needed to achieve bacterial inactivation.

Dissection des fonctions mitotiques de la kinase Aurora B par CALI (Chromophore-Assisted Light Inactivation) / New insights in Aurora B's mitotic functions using chromophore assisted light inactivation.

La kinase Aurora B appartient au complexe des protéines passagères. Ce complexe est impliqué dans la régulation de la condensation, constriction et ségrégation des chromosomes, ainsi que dans la cytokinèse. Son rôle est donc crucial pour prévenir la formation de cellules cancéreuses. Cependant, l'étude de la fonction précise d'Aurora B dans chacune des phases de la mitose est limitée par la durée de celle-ci, et par le manque de spécificité des inhibiteurs existants. Nous avons donc développé une stratégie basée sur la photo-inactivation de la kinase par le chromophore Killer-Red, fusionné à la protéine. L'émission locale de ROS après irradiation, permet alors la photo-inactivation spécifique et temporelle d'Aurora B. La photo-inactivation d'Aurora B avant anaphase aboutie soit à un arrêt de la mitose, soit à la régression du sillon de division, provoquée par l'entrée en anaphase en présence de chromosomes retardés. La photo-inactivation d'Aurora B en début d'anaphase a pour conséquence la régression du sillon de division en cytokinèse ; apportant la première indication directe de l'implication d'Aurora B dans le fuseau mitotique en cytodiérèse. De façon surprenante, la photo-inactivation de la kinase au niveau du corps résiduel, après constriction du sillon de division, n'affecte pas l'abscission. La photo-inactivation d'Aurora B n'affecte pas la localisation des autres membres du complexe des protéines passagères, indiquant que la kinase n'est pas impliquée dans la dynamique du complexe. Les résultats obtenus montrent sans aucun doute l'implication d'Aurora B dans chacune des phases de la mitose, suggérant que la phosphorylation par Aurora B de ses substrats permet le controle de la division cellulaire. / Aurora B is a mitotic kinase involved in chromosome condensation and segregation as well as cytokinesis. Aurora B together with INCENP, Survivin, TD60 and Borealin constitute the chromosome passenger protein complex (CPC), which localizes to the inner centromeres all through metaphase, transfers to the spindle midzone in anaphase and to the midbody in cytokinesis. In order to dissect the mitotic kinase functions of Aurora B as well as its role as an integral part of the CPC in a temporal manner, we have used chromophore assisted light inactivation (CALI) approach. We have combined miRNA ablation of endogenous Aurora B with ectopic expression of miRNA resistant Aurora B fused to the photosensitizer Killer Red (AurB-KR) in HeLa cells. Irradiation at distinct phases of mitosis led to photobleaching of the Killer Red protein, accompanied by emission of reactive oxygen species (ROS) resulting in the photoinactivation of the fused Aurora B. Photoinactivation before anaphase led to either mitotic arrest or cleavage furrow regression due to entry into anaphase with chromosome bridges. CALI at early anaphase also led to cytokinesis failure underlying the role of Aurora B in central spindle function. Consistent with the effects of dominant negative dead-kinase Aurora B, upon CALI the localisation of Incenp, Survivin and Borealin was not affected. Importantly, photoinactivation of Aurora B-KR following cleavage furrow constriction at the midbody had no effect on the completion of abscission. These data, demonstrate unequivocally the distinct roles Aurora B exerts at each phase of mitosis and in particular suggest that Aurora B substrate phosphorylation from metaphase to anaphase is implicated in the spatio-temporal control of cell division and cytokinesis.

Análise do perfil de hipermetilação do gene PTEN e correlação com fatores clínicos anatomopatológicos no carcinoma de células renais / Analysis of hypermethylation profile of PTEN gene and correlation with clinical and pathological findings in renal cell carcinoma

Introdução: Apesar da identificação de fatores prognósticos clínicos e patológicos, muitos pacientes portadores de carcinoma de células renais (CCR) apresentam metástases ao diagnóstico e outros irão desenvolver recorrência local ou à distância durante o seguimento. Novos fatores prognósticos e de origem molecular têm sido avaliados no CCR, destacando-se o PTEN como um dos principais genes envolvidos na carcinogênese renal. Objetivos: Avaliar os fatores clínicos e anatomopatológicos mais significativos nas taxas de sobrevida, identificar a frequência de hipermetilação do gene PTEN através da técnica do pirosequenciamento, o impacto da hipermetilação do gene nas taxas de sobrevida global (SG) e livre de doença (SLD), como também, a associação da presença de hipermetilação com os principais fatores prognóticos. Material e métodos: Foram avaliados 137 pacientes portadores de CCR submetidos a tratamento cirúrgico do tumor primário entre 1997 e 2009. Foram considerados os dados epidemiológicos, clínicos, anatomopatológicos, de estadiamento (TNM 2004) e os obtidos da reação de pirosequenciamento. Resultados: O tempo de seguimento médio foi de 32,3 meses e mediana de 28,8 meses. Considerando o estadimento clínico, foram fatores independentes para a SG: idade (p<0,01), ASA (p=0,02), margens cirúrgicas (p=0,04), grau de Fuhrman (p=0,01), estádio clínico (p<0,001) e subtipo histológico (p<0,01). No modelo múltiplo a SLD foi influenciada únicamente pelo estádio clínico (p<0,001). Dos 137 casos analisados, hipermetilação do gene foi detectada em cinco casos (3,6%). Devido a baixa freqüência detectada optou-se por não realizar a associação da metilação do PTEN com os fatores prognósticos. Em relação às taxas de SG e SLD, de acordo com o perfil de hipermetilação do PTEN, não houve a ocorrência de nenhum evento, ou seja, morte, morte por CCR ou recorrência da doença para os cinco casos que apresentavam hipermetilação. Conclusões: A hipermetilação do xv PTEN foi detectada com baixa frequência, sugerindo a participação de outros genes ou mecanismos moleculares diferentes da metilação na inativação deste gene frequentemente envolvido na carcinogênese renal. As taxas de sobrevida não foram influenciadas pelo perfil de hipermetilação do PTEN, permanecendo o estadiamento clínico do TNM como a principal variável determinante da evolução e do risco de recidiva pelo CCR / Introduction: Despite the identification of clinical and pathological prognostic factors, many patients with renal cell carcinoma (RCC) have metastases at diagnosis and others will develop local or distant recurrence during follow-up. New prognostic factors and of molecular origin have been evaluated in RCC, highlighting PTEN, one of the main genes involved in renal carcinogenesis. Objetives: To assess the most significant clinical and pathological factors in survival rates, and identify the frequency of hypermethylation of the PTEN gene by the pyrosequencing technique, the impact of gene hypermethylation on overall survival (OS) rates and disease free interval (DFS), as well as associating presence of hypermethylation with main prognostic factors. Methods: We evaluated 137 patients with RCC that underwent surgical treatment of primary tumor between 1997 and 2009. We considered the epidemiological, clinical, pathological, staging (TNM 2004) data and those obtained from pyrosequencing. Results: Mean follow-up was of 32.3 months and the median of 28.8 months. Considering the clinical TNM stage, the OS was influenced in the multiple model by age (p < 0.01), ASA (p = 0.02), surgical margins (p = 0.04), Fuhrman´s grade (p = 0,01), clinical stage (p <0.001) and cell subtype (p < 0.01). DFS were influenced in multivariate analysis only by presence of clinical stage (p <0.001). Of the 137 cases examined, gene hypermethylation was detected in five cases (3,6%). Because of this low frequency perceived, we elected not to carry out the association of PTEN methylation with prognostic factors. Regarding OS and DFS rates, according to the hypermethylation of PTEN profile, no event occurred, that is to say death, death from RCC or disease recurrence in the five cases with hypermethylation. Conclusions: Hypermethylation of PTEN was detected with low frequency suggesting involvement of other genes or different molecular mechanisms of methylation upon inactivation of this gene, frequently involved in renal xvii carcinogenesis. Survival rates were not influenced by the hypermethylation of PTEN profile, with clinical TNM staging remaining as the main determinant for development and risk of RCC recurrence

Campylobacter survival under stress conditions encountered between poultry farm and the human intestine

Campylobacter are probably the most important bacterial pathogen related to food-borne illnesses; specifically, gastroenteritis and diarrheal diseases. These bacteria can be isolated from various environments, but always originate from the intestine of warm blooded animals. Particularly, Campylobacter are found in the intestinal tract of poultry, and due to contamination of poultry meat and also further contamination of other food they can cause human infections. Sometimes this results in larger outbreaks, such as during 2016-2017 in Sweden where thousands of persons got infected by a single strain of Campylobacter jejuni sequence type 918 (ST-918). The same strain was also identified amongst a large number of poultry farms and suspicions were directed towards dirty transport cages for poultry as a main route for transmitting the strain between different farms. Similar scenarios with large outbreaks related to one or two single strains (ST-50 and ST-257) had also been observed in previous years and this raised questions about certain strains being especially adapted to survive outside the intestine. The aim here was to examine whether outbreak strains and other strains of C. jejuni have different potential to resist different stress conditions that may be encountered between the poultry farm and the human intestine.

Příprava a charakterizace buněčných modelů lysosomálních dědičných onemocnění - mukopolysacharidos / Preparation and characterization of cell models of lysosomal hereditary diseases - Mucopolysaccharidoses

Mucopolysaccharidoses are a group of diseases that belong to lysosomal storage disorders. A common sign of these monogenic multisystem diseases is a gene mutation leading to a deficiency of the lysosomal enzyme participating in glycosaminoglycan degradation. It results to their accumulation in the tissues and organs, where they cause a progressive damage. There is no efficient treatment available for most mucopolysaccharidoses. Moreover, the research is complicated because of the low prevalence and type of affected tissues. Animal models of these human diseases are used for an evaluation of newly developed therapeutic approaches. However, they also have many limitations due to the different pathogenesis and catabolic pathways of the accumulated substrates between humans and animals. Therefore, animal models are replaced by human cell models. In this thesis, the development of four mucopolysaccharidoses human cell models is reported (MPS IIID, MPS IVA, MPS IVB, MPS VI). Corresponding genes (GNS, GALNS, GLB1, ARSB) were inactivated using CRISPR/Cas9 technology, where plasmids containing specific inserts are delivered to the target human induced pluripotent stem cells (iPSC), using electroporation. Charakterizace genového obsahu chromosomu Z u ptáků. / Characterization of Z chromosome gene content in birds

Theory predicts that sexually antagonistic mutations will be over- or under-represented on the X and Z chromosomes, depending on the average dominance coefficient of the mutations. However, as little is known about the dominance coefficients for new mutations, the effect of sexually antagonistic selection is difficult to predict. To elucidate the role of sexually antagonistic selection in the evolution of Z chromosome gene content in chicken, we analyzed publicly available microarray data from several somatic tissues as well as somatic and germ cells of the ovary. We found that the Z chromosome is enriched for genes showing preferential expression in ovarian somatic cells, but not for genes with preferential expression in primary oocytes or non-sex-specific somatic tissues. Our results suggest that sexual antagonism leads to higher abundance of female-benefit alleles on the Z chromosome. No bias towards Z-linkage of oocyte-enriched genes can be explained by lower intensity of sexually antagonistic selection in ovarian germ cells compared to ovarian somatic cells. An alternative explanation would be that meiotic Z chromosome inactivation hinders accumulation of oocyte-expressed genes on the Z chromosome. Efecto del tratamiento por altas presiones hidrostáticas (HHP) en la calidad de queso fresco y en las proteínas de suero de queserías

Tesis por compendio / [EN] High Hydrostatic Pressure technology emerged several years ago in response to consumer interest for products having greater organoleptic and nutritional quality than those traditionally processed by heat. The food industry initially improved preservation processes developing continuous heat treatment HTST, UHT and aseptic packaging. However, despite these improvements foodstuffs preserved by heat still have quality losses. Although initially the application of High Hydrostatic Pressure in the field of food was focused towards preservation, with the development of technology and research around new applications raised that make it very interesting for the sector. In this context, the aim of this thesis has been the application of High Hydrostatic Pressure on the preservation of fresh cheese and the revalorization of by-products from cheese factories. For this, the effect of treatments by high hydrostatic pressure (HHP) on pathogenic microorganisms (S. aureus, L. monocytogenes) and spoilage microorganisms (Rhodotorula glutinis, aurantiogriseum Penicillium, Pseudomonas fluorescens) that are present in lactic products is evaluated as well as the effect of various factors (sodium chloride (NaCl), initial treatment temperature) on microbial inactivation. To study this effect peptone water at concentrations of 0; 0.1; 0.5; 0.8% NaCl was used as reference medium and fresh cheese as complex matrix containing 0.5% NaCl. Pressure ranges between 200 and 550 MPa (Mega Pascals) were used with short treatment times (in the order of seconds to several minutes), combined with a moderate initial temperature of 20 to 50 °C.The results of microbial inactivation were fitted to the mathematical model that best described the behavior of inactivation curves, allowing predicting the behavior of the microorganisms being treated by HHP under different environmental factors. Treatments by HHP of R. glutinis and P. aurantiogriseum around 350 - 400 MPa generated more than 5 log10 reductions on the initial contamination. The NaCl concentration present in the reference medium for these two microorganisms was an additional control factor. Regarding the effect of the substrate significant differences between the mean values obtained for the reference medium and the complex matrix (cheese) were observed, fresh cheese showed a lower reduction in the initial inoculated load. Due to the production processes of cheesemaking large amount of residual whey generated is a problem under the environmental point of view. In this thesis the use of residual liquid whey was also studied in conjunction with whey protein isolates (WPI) for the purpose of producing gels with a high biological value through various HHP processes. The various mixtures of whey and different percentages of WPI (5, 7, 11, 15% w/v) processed by HHP resulted in gels with different characteristics. The pH values, color (L * a * b *) vary slightly directly depending on the WPI concentration and the storage time. The hardness values and gumminess were modified depending on the liquid medium and the concentration of WPI. The use of cheese whey to gelation favored water retention, significantly reducing the syneresis under refrigeration. This study shows the potential of HHP as control measure, demonstrating the effectiveness of these treatments to prolong the shelf life of fresh cheeses and as unconventional technique to generate new textures that can have great industrial interest at the same time that revalorize some wastes. / [ES] La tecnología de las Altas Presiones Hidrostáticas (HHP) emergió hace varios años como respuesta al interés de los consumidores por disponer de productos de una mayor calidad organoléptica y nutricional que los tradicionalmente procesados por calor. La industria agroalimentaria inicialmente mejoró sus procesos de conservación por calor desarrollando los tratamientos continuos HTST, UHT y el envasado aséptico. Sin embargo, a pesar de estas mejoras los productos alimenticios conservados por calor siguen presentando pérdidas en su calidad final. Aunque al principio la aplicación de las Altas Presiones Hidrostáticas en el campo de la alimentación se enfocó hacia la conservación, con el desarrollo de la tecnología y la investigación alrededor de ella han surgido nuevas aplicaciones que la hacen muy interesante para el sector. En este contexto, el objetivo de la presente tesis doctoral ha sido la aplicación de las Altas Presiones Hidrostáticas en la conservación de queso fresco y en la revalorización de subproductos de queserías. Para ello, se evalúa el efecto de los tratamientos por Altas Presiones Hidrostáticas (HHP) sobre microorganismos patógenos (Staphylococcus aureus, Listeria monocytogenes) y alteradores (Rhodotorula glutinis, Penicillium aurantiogriseum, Pseudomonas fluorescens) de productos lácteos y el efecto de diversos factores presentes en los alimentos (cloruro sódico (NaCl), temperatura inicial de tratamiento) sobre la inactivación microbiana. Para estudiar este efecto se utilizó como medio de referencia agua peptona a concentraciones de 0; 0,1; 0,5; 0,8% de NaCl y como matriz compleja queso fresco con un contenido de 0,5% de NaCl. Se utilizaron intervalos de presión entre 200 y 550 MPa (Mega Pascales) con tiempos de tratamiento cortos (en el orden de segundos a varios minutos), combinados con una temperatura inicial moderada de 20 a 50 °C. Los resultados de inactivación microbiana se ajustaron al modelo matemático que mejor describía el comportamiento de las curvas de inactivación, lo que permite predecir el comportamiento de los microorganismos frente a las HHP y bajo los diferentes factores. En cuanto a las HHP para R. glutinis y P. aurantiogriseum tratamientos en torno a los 350 - 400 MPa generaron más de 5 reducciones log10 de la carga inicial. La concentración de NaCl presente en el medio de referencia para estos dos microorganismos fue un factor adicional de control. Con respecto al efecto del medio de trabajo se observaron diferencias significativas (p<0,05) entre los valores medios obtenidos para el medio de referencia y la matriz compleja (queso fresco): el queso fresco presentó una menor reducción en la carga inicial inoculada.Debido a los procesos productivos de la fabricación del queso se genera gran cantidad de suero lácteo residual que es un problema bajo el punto de vista medioambiental. En esta tesis también se estudió la utilización del suero lácteo líquido residual en conjunto con aislados proteicos del suero (WPI) con la finalidad de producir geles con un elevado valor biológico mediante diversos procesos de HHP. Las diversas mezclas de suero de queso junto con los porcentajes de 5, 7, 11, 15% p/v de WPI procesadas por HHP dieron lugar a geles con diversas características. Los valores de pH, color (L*a*b*) variaron ligeramente dependiendo directamente tanto de la concentración de WPI como del tiempo de almacenamiento. Los valores de dureza y gomosidad sufrieron modificaciones dependiendo del medio líquido y la concentración de WPI. La utilización de suero de queso para la formación del gel favoreció la retención de agua, reduciendo significativamente la sinéresis en refrigeración.Este estudio muestra el potencial de las HHP como una medida de control, demostrándose la efectividad de estos tratamientos para prolongar la vida útil de los quesos frescos y como técnica no convencional para generar nuevas texturas que pueden tener gran inte / [CA] La tecnologia de les altes pressions hidrostàtiques va emergir fa diversos anys com a resposta a l'interés dels consumidors per disposar de productes d'una qualitat organolèptica i nutricional major que la dels processats per calor tradicionalment. En aquest context, l'objectiu de la present tesi doctoral ha sigut l'aplicació de les altes pressions hidrostàtiques en la conservació del formatge fresc i en la revalorització de subproductes de formatgeries. Per a això, s'avalua l'efecte dels tractaments per altes pressions hidrostàtiques (HHP) sobre microorganismes patògens (Staphylococcus aureus, Listeria monocytogenes) i alteradors (Rhodotorula glutinis, Penicillium aurantiogriseum, Pseudomonas fluorescens) de productes lactis i l'efecte de diversos factors presents als aliments (clorur sòdic (NaCl), temperatura inicial de tractament) sobre la inactivació microbiana. Per a estudiar aquest efecte es va utilitzar com a medi de referència aigua peptona a concentracions de 0; 0,1; 0,5; 0,8% de NaCl i com a matriu complexa formatge fresc amb un contingut de 0,5% de NaCl. Es van utilitzar intervals de pressió entre 200 y 550 MPa (Mega Pascals) amb temps de tractament curts (en l'orde de segons a diversos minuts), combinats amb una temperatura inicial moderada de 20 a 50 °C. Els resultats d'inactivació microbiana es van ajustar al model matemàtic que millor descrivia el comportament de les corbes d'inactivació, el que permet predir el comportament dels microorganismes front a les HHP i baix els diferents factors. En referència a les HHP per a R. glutinis i P. aurantiogriseum tractaments al voltant dels 350 - 400 MPa van generar més de 5 reduccions log10 de la càrrega inicial. La concentració de NaCl present en el medi de referència per a aquestos dos microorganismes va ser un factor addicional de control. Respecte a l'efecte del medi de treball es van observar diferències significatives (p<0,05) entre els valors mitjans obtinguts per al medi de referència i la matriu complexa (formatge fresc): el formatge fresc va presentar una menor reducció en la càrrega inicial inoculada. Degut als processos productius de la fabricació del formatge es genera gran quantitat de sèrum lacti residual que és un problema baix el punt de vista mediambiental. En aquesta tesi també es va estudiar la utilització del sèrum lacti líquid residual en conjunt amb aïllats proteics del sèrum (WPI) amb la finalitat de generar gels amb un elevat valor biològic mitjançant diversos processos de HHP. Les diverses mescles de sèrum de formatge juntament amb els percentatges de 5, 7, 11, 15% p/v de WPI processades per HHP van generar gels amb diverses característiques. Els valors de pH, color (L*a*b*) van variar lleugerament depenent directament tant de la concentració de WPI com del temps d'emmagatzematge. Els valors de duresa i gomositat van sofrir modificacions depenent del medi líquid i la concentració de WPI. La utilització del sèrum de formatge per a la formació del gel va afavorir la retenció d'aigua, reduint significativament la sinèresi en refrigeració.Aquest estudi mostra el potencial de les HHP com a una mesura de control, demostrant-s'hi l'efectivitat d'aquestos tractaments per a prolongar la vida útil dels formatges frescos i com a tècnica no convencional per a generar noves textures que poden tindre gran interés / Torres Bello, EF. Modelling the neuropsychopharmacology of obsessive-compulsive disorder in the common marmoset (Callithrix jacchus)

This thesis extends the understanding of the neural and neurochemical contributions to two forms of behavioural adaptation, reversal learning and contingency degradation, in which stimulus/action-reward contingencies are altered. The results are interpreted within the psychological framework of the compulsivity construct, and their implications for the pathological behaviour of obsessive-compulsive-disorder (OCD) are considered. The orbitofrontal cortex (OFC) and striatum are key brain structures involved in reversal learning, as are the neurotransmitters serotonin (5-hydroxytryptamine, 5-HT) and dopamine (DA) within those respective regions. However, there has been little empirical evidence of how these two structures and neurochemical systems interact, especially in the functional context of reversal learning. In Chapter Three, the impact of experimentally-induced reductions of 5-HT in the anterior OFC on monoamine levels in subcortical structures such as the striatum and amygdala was determined, DA being found to be significantly up-regulated in the amygdala. Functionally, 5-HT depletion of the OFC has previously been shown to induce deficits in reversal learning. To determine the possible causal significance of amygdala dopamine up-regulation for said reversal learning deficit, the effects of blocking the upregulation with the infusion of intra amygdala DA receptor antagonists following bilateral OFC 5-HT depletion were investigated in a reversal learning paradigm. In Chapter Four, the differential roles of regions of striatum were examined in visual reversal learning. Two recent investigations in non-human primates highlighted the role of the striatum in reversal learning,but pinpointed the critical region to be either the ventromedial caudate or the putamen. Marmosets were trained on a serial reversal task that allowed multiple acute neural manipulations, and the ventromedial caudate and putamen were then reversibly inactivated using the GABAA agonist muscimol. Results indicated dose-related impairments specifically in reversal learning within the putamen, with sparing of discrimination retention. By contrast, similar reversible inactivation of the caudate nucleus produced marked deficits in visual discrimination performance (retention). In Chapter Five, the neural basis of action-outcome contingency knowledge was investigated by inactivating distinct regions of the PFC, the perigenual ACC (pgACC; area 32) and the anterior OFC, and determining response sensitivity to the degradation of action-outcome contingencies. In previous work, excitotoxic lesions of either the pgACC or the OFC had been found to induce insensitivity to contingency degradation in marmosets. However, the design of that experiment did not allow specification of whether stimulus- or action-outcome associations were disrupted, and a precise neural locus could not be determined for the behavioural effects as the OFC lesions included parts of the lateral and medial OFC. I therefore developed a novel contingency degradation paradigm that distinguished between stimulus- and action-outcome associations to enable the study of acute pharmacological manipulations in both brain regions. The pgACC and OFC were reversibly inactivated using GABAA-GABAB agonists (muscimol-baclofen). Whereas the pgACC inactivation produced selective deficits in sensitivity to action-outcome contingency degradation, OFC inactivation reduced the suppressive effect of noncontingent reward on responding more generally but left intact sensitivity to degradation of the contingencies. These results are discussed in terms of different theories of the functions of the pgACC and OFC. In the final discussion the findings on the neural substrates of reversal learning and contingency degradation are drawn together in terms of their significance for theories of PFC involvement in cognitive control, and for the understanding of OCD and other neuropsychiatric disorders.

Inherited metabolic disorders (IMD) form a diverse group of several hundred different diseases with a relatively high cumulative incidence (stated up to 1:600). They are associated with accumulation of the substrates and lack of the products in specific metabolic pathways, which is caused by deficiency of the enzyme or its activator, or dysfunction of the transport protein. However, the underlying cause is at the DNA level. The grounds for different phenotype manifestation in patients with the same genotype are often not known. During my work at the Institute of Inherited Metabolic Disorders, I designed several new methods for the research of IMD and applied them in the patients and their families. I created procedures for the isolation of lysosomal membranes that are used for the research of lysosomal storage disorders and general properties of lysosomes. Next, I introduced several novel assays for determination of the X-inactivation ratio, which led to a significant increase of informative women. Nowadays, we use these methods in heterozygous women with X-linked diseases in order to study the influence of X-inactivation on the manifestation of the diseases.
