Defesa : Curitiba, 12/08/2016 / Inclui referências : f. 57-64;72-74;82-84;95-97 / Resumo: Introdução: Avaliar a expressão do biomarcador p16INK4a no produto de conização de mulheres com diagnóstico de NIC-AG e correlacionar a sua expressão com a severidade das lesões, a habilidade de prever a recorrência, as diferenças biológicas entre NIC 2 e 3 e a tipagem de HPV. Método: um estudo transversal, conduzido em pacientes que foram submetidas a conização por NIC-AG, com DNA-HPV previamente conhecido, entre janeiro 2009 e agosto 2011, no Hospital Erasto Gaertner/ Brasil. A pesquisa do p16 foi avaliada por meio da técnica de microarranjos teciduais e sua expressão correlacionada com o diagnóstico clínico, índice de recorrência e as diferenças biológicas entre NIC 2 e 3. O teste do X2 foi usado para análise estatística com valor de p?0.05, e a sensibilidade, especificidade, VPP, VPN e acurária dos testes calculadas. Resultados: 192 mulheres, entre 22 e 48 anos (35 ± 6), divididas em 2 grupos: 102 NIC 2 e 90 NIC 3. A média de idade, paridade, hábito de fumar e técnica de conização foi semelhantes nos 2 grupos. O p16 foi positivo em 86% dos NIC 2 e 92% dos NIC 3. Em relação a recorrência o p16 teve VPP de 11%, VPN de 95%, sensibilidade de 95%, especificidade de11% e acurácia de 93%. A habilidade do p16 em predizer as diferenças entre NIC 2 e 3 resultou em VPP de 48% e VPN de 66%, sensibilidade de 92%, especificidade de 13% e acurácia de 89%. Conclusão: A expressão do p16 isoladamente não foi capaz de predizer a recorrência da NIC-AG tratada e nem de demonstrar as diferenças biológicas entre NIC 2 e 3, entretanto sua expressão se relaciona diretamente com a severidade da doença. Palavras-chaves: Neoplasia intraepitelial cervical, recorrência, marcadores biológicos, NIC-AG. / Abstract: Background: The aim of this study is to evaluate the expression of biomarker p16INK4a on the product of conization in women diagnosed with HSIL, to correlate immunostaining with lesion severity, the ability to predict recurrence and the biological differences between CIN 2 and CIN 3. Methods: A cross-sectional study was conducted in patients who had undergone conization due to HSIL and detected HPV DNA, at Erasto Gaertner Cancer Center (EGCC) Hospital, in Brazil. The expression of p16INK4a was evaluated using tissue microarray technique and overexpression was correlated to diagnosis and clinical evolution. A chi-square test was used for statistical analysis with p-value less than or equal to 0.05 and the sensitivity, specificity, PPV, NPV, and accuracy of the tests calculated. Results: The review included 192 women, between 22 and 48 years old (35 ± 6), divided into two groups: 102 CIN 2 and 90 CIN 3 cases. The epidemiological data were similar in the two groups. p16 was positive in 86% of the CIN 2 and 92% of CIN 3. To predict recurrence p16 has PPV of 11%, NPV of 95%, sensitivity of 95%, specificity of 11%, and accuracy 93%. Applied to distinguish between CIN 2 and 3 p16 has VPP of 48%, VPN of 66%, sensitivity of 92%, specificity of 13%, and accuracy 89%. Conclusion: Immunostaging of biomarker p16 alone cannot predict the rate of recurrence of HSIL or can it demonstrate the biological differences between CIN 2 and 3; however, overexpression does increase with the degree of lesion severity. Adicionalmente, foram utilizados valores de Prostaglandina E2 do humor aquoso de cães oftalmologicamente saudáveis (n=7). Para avaliação da pressão intraocular empregou-se tonometria digital de aplanação. A avaliação da densidade e da hexagonalidade das células endoteliais foi realizada por meio de microscopia especular de não contato. Sob anestesia geral, 0,2mL de humor aquoso foram colhidos por meio de paracentese da câmara anterior. As amostras de aquoso foram congeladas e armazenadas à -80 oC para mensuração da concentração de PGE2, por imunoensaio enzimático competitivo. Os valores foram comparados estatisticamente, empregando-se análise de variância de via única, e o teste de múltiplas comparações de Bonferroni. Teste t não pareado foi utilizado para se avaliar os dados obtidos por microscopia especular. Possíveis correlações entre os resultados foram comparadas aplicando-se o teste de Person. Adotou-se nível de significância de p<0,05. Em ambos os grupos observou-se diminuição significativa da pressão intraocular (PIO), entretanto não houve diferença entre eles (p = 0,90). As médias dos valores de densidade celular (CD) e de hexagonalidade (HEX) foram 2253,363 cell/mm² e 59,03% para GSM e 2058,325 cell/mm² e 63,75% para GSH, respectivamente. Entretanto não houve diferença significativa entre ambos, tanto para CD (p=0,201), quanto para HEX (p=0,61). Observou-se incremento nos valores de PGE2 nos grupos com catarata em relação aos cães oftalmologicamente saudáveis (p<0,001), entretanto, não houve diferença significativa entre os grupos com catarata... / Intraocular pressure, endothelial cell density and hexagonality, and aqueous humor prostaglandin E2 (PGE2) were studied and correlated in dogs with mature (n=8) and hypermature (n=8) senil cataratcs. In addition, aqueous humor PGE2 values of ophthalmic healthy dogs (n=7) were used. Non contact specular microscopy was used to evaluate density and hexagonality of endothelial cells. 0.2mL of aqueous humor was collected by means of anterior chamber paracentesis. Aqueous humor samples were frozen at -80 oC and concentration of PGE2 were quantified by means of competitive enzyme immunoassay. PGE2 values were statistically compared with one way analysis of variance and Bonferroni´s multi comparison test. Specular microscopy data were assessed by means of unparied T test. Person´s test was used in order to assess possible correlations among data. Significance was set at value of P < 0.05. Values of endothelial cell density and hexagonality did not change significantly between groups (P = 0.61). PGE2 values were significantly higher in dogs with cataract, when compared with ophthalmic healthy dogs (P < 0.001), however, this parameter did not change significantly between mature and hypermature groups (P > 0.05). Intra - Organizational Mobility: Movers, Incumbents, and Communication Networks

A growing body of research suggests that intra-organizational mobility represents an important source of value creation and retention. Internal hires who are embedded in organizational social networks have greater resources and experience than external workers who are less socially connected. Notwithstanding the great practical and theoretical interest in the benefits of intra-organizational mobility at the organizational level, little is known about how individuals’ intra-organizational careers unfold and the influence of social networks toward that end. This dissertation combines findings from three separate projects to investigate the mechanisms underlying the phenomenon of intra-organizational mobility—the structural factors that explain why people move within an organization, how movers and incumbents do or do not benefit from mobility, and the individual differences in network behavior for mobility. More specifically, in the first chapter, I examine how pre-existing communication contacts affect the mover’s performance upon joining the new group. I expect that movers are more likely to join business units to which they have pre-existing ties. Nonetheless, the ties that facilitate movers’ joining business units are oftentimes not those that help them to perform well subsequently. In the next chapter, I explore gender differences in network behavior as they impact on intra-organizational mobility. I argue that when a mover retains ties to the working unit that is being left, it improves the mover’s post-move performance. And women are more likely to maintain such persistent social ties, whereas men are more likely to establish new ties. In the final chapter, I assess the effects on the receiving group when a mover joins, and I argue that low-ranking incumbents embedded in stable performance hierarchies suffer from the introduction of high-performing newcomers and the induced unfavorable social comparison. I test my predictions using time-series data on the internal inter-branch transfers of retail sales employees at a US-based financial institution between November 2014 and April 2016. The dataset is composed of individual demographic information, monthly performance metrics (in dollars), and meta email communication among all employees. The data permits several methodological advancements: (1) the use of objective and consistent performance measures; (2) analysis of the temporal changes in the networks of the movers and their contacts; (3) analysis of communication network and its impact on performance, and (4) robustness checks that apply instrumental variable techniques. The approach taken in this dissertation adds a new perspective on the relationship between intra-organizational mobility and competitive advantage.
