This manual has been designed for a class of twenty- four students concurrently enrolled in the lecture course. The laboratory aids in the learning process and fosters an interest in the science of genetics. This manual and the experiments contained within are both informative and fun. The manual correlates with and expands upon the genetics course. Each investigation, with the exception of the Drosophila melanogaster project, can be completed in a 3-4 hour timeframe. This manual provides a “hands on” experience of theories simply discussed in the lecture course. This manual is intended to be a one-source manual where each investigation is designed to include an adequate introduction. Special attention has been given for each investigation with both the student and instructor in mind.

The sandbox strategy : the why and how of federal law enforcement integration

CHDS State/Local / This thesis examines the interoperability of federal law enforcement's Big Six investigative agencies, to include the Federal Bureau of Investigation, Immigration and Customs Enforcement, Drug Enforcement Administration, Alcohol Tobacco Firearms and Explosives, Internal Revenue Service-CID, and the United States Secret Service-Investigations. The main issue is whether, in the post-9/11 environment of transnational and terrorist criminal threats, the current administrative and jurisdictional configuration of the Big Six within three executive epartments with overlapping duties marginalizes the nation's investigative work-product. This discussion includes the establishment of metrics used to gauge the functionality of the Big Six and, thus, to determine whether Negative Characteristics are present that materially affect the "total" mission. Ultimately, the conclusion is drawn that the integration of the Big Six into a single agency, namely the FBI, would better serve the nation's federal investigative law enforcement needs. This leads into the next area of discussion, which is how to integrate the Big Six. The processing of the "Siemens case" has not only triggered an unexpected compliance shaft in Germany but has also meant that in the meantime internal investigations are commissioned in nearly all the major public investigations for corruption or other economic crimes by the companies concerned. A critical analysis of "10 years after Siemens" shows that this trend has led to a variety of open legal issues and a different handling in the judicial practice. A legal regulation is likely to be inevitable in the long run.

80 Abstract The aim of this thesis titled Methods of investigation of sexual crimes (selected issues) is to clarify methods of investigation of sexual crimes with special focus on rape. The thesis is divided into nine chapters and sub-chapters. Introduction defines the goal of the thesis. The first chapter describes term of rape and its development from 1852 to the present. The first chapter also includes statistical data for the year 2012. The second chapter describes criminological characteristics of rape relating to typical way of committing, offender and victim. The myth that most of the offenders are sexually deviant is refuted in this chapter. The third chapter analyzes typical traces with traditional distinction between traces in the mind and material traces. Biological traces are emphasized, because they are in the investigation of the crime of rape very common. The fourth chapter is divided into two sub-chapters devoted to typical investigative situations. Following fifth chapter deals with the scope of investigation. Questions which must be clarified are mentioned in order to investigate facts of case beyond all reasonable doubt. The sixth chapter describes impetus to the investigations. The most important part of the work focuses on personal investigation of work rehabilitation. Since the area of investigating sexual crimes is very broad, I have limited the scope of my paper to investigation of the two most frequent types of crime, which are rape and sexual abuse. Regarding the operations usually performed during investigation, I have paid close attention to expert opinions. The purpose of this paper is to stress concrete specifics occuring during investigation, which is discussed throughout the chapters. Chapter One introduces acts of sexual crime (particularly the rape and the sexual abuse) from the criminal-law point of view as it is stated in the contemporary legal regulation in the Czech Republic. This chapter also provides statistical tables showing the evolutionary trend in the number of prosecuted and adjudicated sexual criminal acts of rape and sexual abuse between 2000 and 2010. Syftet med studien är att analysera pedagogers berättelser om hur de upplever arbetet med identifiering och utredning av läs- och skrivsvårigheter hos nyanlända. Vi har samlat empiriskt material i form av intervjuer som vi har analyserat med hjälp av tidigare forskning samt teorier. De nio intervjuerna av speciallärare/specialpedagoger, SVA-lärare och modersmålslärare genomfördes på två skolor med ett stort antal nyanlända elever och på kommunens mottagningsenhet. I studien berättar pedagogerna att de upplever en stor utmaning kring identifiering och utredning av nyanlända elever med läs- och skrivsvårigheter. Det råder osäkerhet kring vem som ansvarar för vad och de beskriver att gränsdragningsproblem leder till att elever faller mellan stolarna. Pedagogerna har svårt att veta om problematiken beror på läs- och skrivsvårigheter eller bristande kunskaper i svenska språket vilket leder till både över- och underidentifiering. För att kunna ge nyanlända elever rätt stödåtgärder efterfrågas förutsättningar och former för samarbete och mer tydliga riktlinjer och rutiner. Enligt pedagogerna skulle det leda till mer likvärdig utbildning. Från intervjuerna framkommer också att identifieringen av läs- och skrivsvårigheter tar lång tid och att det saknas en utvecklad metod och ett heltäckande material för utredning. Bedömningen baseras ofta på pedagogens erfarenhet och i jämförelse med andra elevers utveckling i andraspråket.   Trots flera utmaningar är pedagogerna överlag positiva och har förslag på möjligheter och förbättringar; såsom kompetenshöjning inom andraspråksinlärning och läs- och skrivsvårigheter, mer användning av utredningsmaterial som är oberoende av språkkunskaper, fördjupat samarbete mellan olika professioner, tillgång till extern rådgivning och rekrytering av modersmålslärare bland nyanlända.       Vår förhoppning är att medverka till att skolor ser över sina riktlinjer och rutiner kring hur man identifierar och utreder läs- och skrivsvårigheter hos nyanlända elever / The purpose of the study is to analyze teachers' stories of how they experience the process of identification and investigation of the reading and writing difficulties of new arrivals. We have gathered empirical data in the form of interviews that we have analyzed using previous research and theories. The nine interviews of special education teachers / special educators, teachers teaching Swedish to newly arrived students and native mother- tongue teachers were made at two schools that have a large number of newly arrived pupils and at the municipal reception center. In the study the educators say that they are experiencing a major challenge identifying and investigating newly arrived students´ difficulties in reading and writing. There is some uncertainty about who is responsible for what, and they describe the demarcation problems leading to nobody feeling responsible for the problem. The teachers find it difficult to know if the problems are due to language difficulties or lack of knowledge of the Swedish language. In order to provide newly arrived students with proper support conditions and forms of cooperation as well as clear policies and procedures are requested. According to the teachers that would lead to a more equal education. The identification of reading and writing difficulties is said to take a long time and there is also a lack of a developed methodology and comprehensive materials to detect these difficulties. The assessment is often based on the teacher's experience and in comparison with other students´ learning a second language                                                                                                             Despite the challenges the educators are generally positive and have suggested facilities and improvements; such as increasing skills in how the pupils learn a second language and how to work with reading and writing difficulties, more use of investigative material that is independent of language skills, enhanced cooperation between different professions, access to external advice and recruitment of mother- tongue teachers among new arrivals. Our hope is to contribute to the schools reviewing their policies and procedures on how to identify and investigate the reading and writing difficulties of newly arrived students.

Thermofluidic Transport in Evaporating Droplets: Measurement and Application

<p>Microscale environments provide significant resolution and distortion challenges with respect to measurement techniques; however, with improvements to existing techniques, it is possible to gather relevant data to better understand the thermal and fluidic mechanisms at such small scales in evaporating droplets.</p> <p> </p> <p>Infrared thermography provides several unique challenges at small scales. A primary issue is that the low native resolution of traditional infrared cameras significantly hamper the collection of details of microscale features. Furthermore, surfaces exhibiting vastly different emissivities, results in inaccurate temperature measurements that can only be corrected with irradiance-based emissivity maps of the surface; however, due to the resolution limitations of infrared thermography, these emissivity maps can also display significant errors. These issues are overcome through the use of multi-frame super-resolution. The enhanced resolution allows for better capture of microscale features, therefore, enhancing the emissivity map. A quantitative error analysis of the system is conducted to quantify the feature size resolution improvement as well as the smoothing effect of super-resolution reconstruction. Furthermore, a sensitivity analysis is conducted to quantify the impact of registration uncertainty on the accuracy of the reconstruction. Finally, the improved emissivity map from super-resolution is demonstrated to show the increased accuracy over low-resolution mapping.</p> <p> </p> <p>When applied to water droplets, particularly on nonwetting surfaces, infrared thermography is confounded by the presence of nonuniform reflectivities due to the spherical curvature of the liquid-air interface. Thus, when measuring the temperature along the vertical axis of a water droplet, it is necessary to correct the reflection. Using a controlled background environment, in conjunction with the Fresnel equations, it is possible to correct the reflective effects on the interface and calculate the actual temperature profile. This allows for a better understanding of the governing mechanisms that determine the thermal transport within the droplet. While thermal conduction is the primary transport mechanism along the vertical axis of the droplet, it is determined that the temperature drop is partially dampened by the convective transport from the ambient air to the liquid interface. From this understanding revealed by the measurements, the vapor-diffusion-based model for evaporation was enhanced to better predict evaporation rates.</p> <p> </p> <p>Further exploration into the mechanisms behind droplet evaporation on nonwetting surfaces requires accurate knowledge of the internal flow behavior. In addition, the influence of the working fluid can have a significant impact on the governing mechanisms driving the flow and the magnitude of the flowrate. While water droplet evaporation has been shown to be governed by buoyancy-driven convection on nonwetting substrates, similar studies on organic liquid droplets are lacking. Particle image velocimetry is effective at generating a velocity flow field, but droplets introduce distortion due to the refraction from the spherical interface of the droplet. As such, velocity correction using a ray-tracing approach was conducted to correct the velocity magnitudes and direction. With the velocity measurements, the flow was determined to be surface-tension-driven and showed speeds that are an order of magnitude higher than those seen in buoyancy-driven flow in water droplets. This resulted in the discovery that advection plays a significant role in the transport within the droplet. As such, the vapor-diffusion-governed evaporation model was adjusted to show a dramatic improvement at predicting the temperature gradient along the vertical axis of the droplet.</p> <p> </p> <p>Armed with the knowledge of flow behavior inside droplets, it is expected that droplets with aqueous solutions should exhibit buoyancy-driven convection. The final part of this work, therefore, leverages this phenomenon to enhance mixing during reactions. Colorimetry is a technique that is widely utilized to measure the concentration of a desired sample within some liquid; the sample reacts with a reagent dye the color change is measured, usually through absorbance measurements. In particular, the Bradford assay is used to measure protein concentration by reacting the protein to a Coomassie<sup>TM</sup> Brilliant Blue G-250. The absorbance of the dye increases, most significantly at the 590 nm wavelength, allowing for precise quantitation of the amount of protein in the solution. A droplet-based reaction chamber with buoyancy-enhanced mixing has the potential to speed up the measurement process by removing the need for a separate pre-mixing step. Furthermore, the reduced volume makes the process more efficient in terms of reactant usage. Experimental results of premixed solutions of protein sample and reagent dye show that the absorbance measurement through a droplet tracks strongly with the protein concentration. When the protein sample and dye reagent are mixed <i>in situ</i>, the complex interaction between the reactants, the mixing, and the adsorption of protein onto the substrate creates a unique temporal evolution in the measured absorbance of the droplet. The characteristic peaks and valleys of this evolution track strongly with concentration and provide the framework for measurement of concentration in a droplet-based system.</p> <p> </p> <p>This thesis extends knowledge about droplet thermal and fluidic behavior through enhanced measurement techniques. This knowledge is then leveraged in a novel application to create a simple, buoyancy-driven colorimetric reaction setup. Overall, this study contributes to the field of miniaturized, efficient reaction and measurement devices.</p>

Afin de ne pas prêter le flanc aux railleries et aux critiques, l’Église fait preuve d’une grande prudence à l’égard des déclarations de « guérison miraculeuse ». C’est dans cette perspective qu’elle s’attache à départager l’"authentique" et l’"inauthentique" parmi les manifestations de la vie religieuse et qu’elle considère que « les faux miracles doivent être discernés des vrais ». Ces distinctions ne vont pas de soi. Elles nécessitent des procédures d’enquêtes dont le fonctionnement reste largement inexploré par les sciences humaines. Si, depuis le XIe siècle, le magistère catholique exerce un droit de regard, allant s’élargissant, sur les miracles déclarés par les fidèles, c’est sans doute à la suite des événements de Lourdes (« apparitions » et « guérisons » à partir de 1858) que cette volonté de contrôle s’exprime sous une forme nouvelle. À une époque qui exalte les vertus de la rationalité scientifique, au moment précis où la médecine parisienne atteint le sommet de sa notoriété pour ses travaux sur l’hystérie, redoutant les controverses provoquées par « l’épidémie de guérison » qui suit les visions de Bernadette Soubirous, de nombreux membres de la hiérarchie catholique française souhaitent donner des formes plus respectables à ce que l’on peut considérer comme une « explosion de dévotion populaire » non orthodoxe et difficilement contrôlable. C’est dans le cadre de cette stratégie d’encadrement des événements que s’inscrit, en 1883, au sein même du sanctuaire de Lourdes, la création d’une instance médicale chargée de contrôler les revendications de guérisons miraculeuses. En s’attachant à analyser cette procédure de contrôle, spécifique au sanctuaire de Lourdes, cette thèse voudrait permettre de mieux comprendre la manière dont sont produits les miracles à Lourdes. Notre travail souhaite montrer qu’il est, au contraire, produit collectivement par un ensemble d’acteurs (témoins, médecins, ecclésiastiques…) engagés dans une activité concertée d’enquête complexe au résultat incertain / Not to invite the critics, the Church shows a big caution towards the declarations of "supernatural cure". In a time which excites the virtues of the scientific rationality, at the precise moment when the Parisian medicine reaches the summit of its fame for its works on the hysteria, dreading the caused controversies by " the epidemic of cure " which follows the visions of Bernadette Soubirous, numerous members of the French catholic hierarchy wish to give more respectable forms to what we can consider as a explosion of popular worship not orthodox and with difficulty controllable. It is within the framework of this strategy of frame of the events that joins, in 1883, within the sanctuary of Lourdes, the creation of a medical authority asked to control the claiming of supernatural cures. By attempting to analyze this control procedure, this thesis would like to allow to understand better the way are produced the miracles in Lourdes.
